Abstract:
ABSTRACTPersistentPseudomonas aeruginosainfections are a major cause of morbidity and mortality in cystic fibrosis (CF) patients and are linked to the formation of a biofilm. The development of new biofilm inhibition strategies is thus a major challenge. LL-37 is the only human antimicrobial peptide derived from cathelicidin. The effects on theP. aeruginosaPAO1 strain of synthetic truncated fragments of this peptide were compared with the effects of the original peptide. Fragments of LL-37 composed of 19 residues (LL-19, LL13-31, and LL7-25) inhibited biofilm formation. The strongest antibiofilm activity was observed with the peptides LL7-37 and LL-31, which decreased the percentage of biomass formation at a very low concentration. Some peptides were also active on the bacteria within an established biofilm. LL7-31, LL-31, and LL7-37 increased the uptake of propidium iodide (PI) by sessile bacteria. The peptide LL7-37 decreased the height of the biofilm and partly disrupted it. The peptides active within the biofilm had an infrared spectrum compatible with an α-helix. LL-37, but not the peptides LL7-31 and LL7-37, showed cellular toxicity by permeabilizing the eukaryotic plasma membrane (uptake of ethidium bromide and release of lactate dehydrogenase [LDH]). None of the tested peptides affected mitochondrial activity in eukaryotic cells. In conclusion, a 25-amino-acid peptide (LL7-31) displayed both strong antimicrobial and antibiofilm activities. The peptide was even active on cells within a preformed biofilm and had reduced toxicity toward eukaryotic cells. Our results also suggest the contribution of secondary structures (α-helix) to the activity of the peptides on biofilms.

Abstract:
Background The authors postulated that hypothermic neuroprotection can be attributed to a delayed onset of ischemic depolarization. Methods Halothane-anesthetized rats were prepared for near-complete forebrain ischemia. Direct current (DC) potential microelectrodes were placed in hippocampal CA1. The pericranial temperature was maintained at 31 degrees C, 33 degrees C, 35 degrees C, or 37 degrees C (n = 6 per group). Bilateral carotid occlusion with systemic hypotension was initiated for 10 min. The time to onset of the DC shift was recorded. In a second experiment, rats were assigned to 37 degrees C or 31 degrees C for 10 min of ischemia, or to 31 degrees C for 14 min of ischemia (n = 8 per group). These durations of ischemia were defined to allow 9 min of ischemic depolarization in the 37 degrees C-10 min and 31 degrees C-14 min groups. Neurologic and histologic outcomes were examined 7 days later. Results Hippocampal CA1 time to depolarization increased with decreasing temperature (P < 0.0001). Time to depolarization was increased by approximately 4 min in the rats maintained at 31 degrees C compared with those at 37 degrees C. Time to repolarization during reperfusion was not affected by temperature. Increasing the duration of ischemia from 10 min to 14 min with the pericranial temperature maintained at 31 degrees C resulted in a duration of depolarization that was equivalent in the 37 degrees C-10 min and 31 degrees C-14 min groups. However, hippocampal CA1 damage was not increased (31 degrees C-10 min = 4 +/- 1% dead neurons; 31 degrees C-14 min = 6 +/- 1% dead neurons, 95% CI, -1% to 3%; 37 degrees C-10 min = 90 +/- 17% dead neurons). Conclusions Despite similar durations of DC depolarization, outcome in hypothermic rats was markedly improved compared with normothermic rats. This indicates that hypothermic neuroprotection can be attributed to mechanisms other than the delay in time to onset of ischemic depolarization.

Abstract:
Background The aim of this study was to determine whether progressive levels of hypothermia (37, 34, 31, or 28 degrees C) during cardiopulmonary bypass (CPB) in pigs reduce the physiologic and metabolic consequences of global cerebral ischemia. Methods Sagittal sinus and cortical microdialysis catheters were inserted into anesthetized pigs. Animals were placed on CPB and randomly assigned to 37 degrees C (n = 10), 34 degrees C (n = 10), 31 degrees C (n = 11), or 28 degrees C (n = 10) management. Next 20 min of global cerebral ischemia was produced by temporarily ligating the innominate and left subclavian arteries, followed by reperfusion, rewarming, and termination of CPB. Cerebral oxygen metabolism (CMRO2) was calculated by cerebral blood flow (radioactive microspheres) and arteriovenous oxygen content gradient. Cortical excitatory amino acids (EAA) by microdialysis were measured using high-performance liquid chromatography. Electroencephalographic (EEG) signals were graded by observers blinded to the protocol. After CPB, cerebrospinal fluid was sampled to test for S-100 protein and the cerebral cortex was biopsied. Results Cerebral oxygen metabolism increased after rewarming from 28 degrees C, 31 degrees C, and 34 degrees C CPB but not in the 37 degrees animals; CMRO2 remained lower with 37 degrees C (1.8 +/- 0.2 ml x min[-1] x 100 g[-1]) than with 28 degrees C (3.1 +/- 0.1 ml x min[-1] x 100 g[-1]; P < 0.05). The EEG scores after CPB were depressed in all groups and remained significantly lower in the 37 degrees C animals. With 28 degrees C and 31 degrees C CPB, EAA concentrations did not change. In contrast, glutamate increased by sixfold during ischemia at 37 degrees C and remained significantly greater during reperfusion in the 34 degrees C and 37 degrees C groups. Cortical biopsy specimens showed no intergroup differences in energy metabolites except two to three times greater brain lactate in the 37 degrees C animals. S-100 protein in cerebrospinal fluid was greater in the 37 degrees C (6 +/- 0.9 microg/l) and 34 degrees C (3.5 +/- 0.5 microg/l) groups than the 31 degrees C (1.9 +/- 0.1 microg/l) and 28 degrees C (1.7 +/- 0.2 microg/l) animals. Conclusions Hypothermia to 28 degrees C and 31 degrees C provides significant cerebral recovery from 20 min of global ischemia during CPB in terms of EAA release, EEG and cerebral metabolic recovery, and S-100 protein release without greater advantage from cooling to 28 degrees C compared with 31 degrees C. In contrast, ischemia during 34 degrees C and particularly 37 degrees C CPB showed greater EAA release and evidence of neurologic morbidity. Cooling to 31 degrees C was necessary to improve acute recovery during global cerebral ischemia on CPB.

BACKGROUND Although cohabitation has spread rapidly in Austria during the past decades, it is more a prelude than an alternative to marriage. The individualization thesis serves as a conceptual framework for explaining the rise of cohabiting unions. OBJECTIVE Our aim is to understand what motivates people to cohabit and marry from an individualization perspective. The present study was designed to investigate in which ways key notions of the individualization thesis such as commitment, romantic love and risk are reflected in discourses on cohabitation and marriage. METHODS Research is based on data from eight focus group discussions (71 participants) conducted in Vienna, Austria, in 2012. This data was analyzed with the help of qualitative methods. RESULTS The focus group participants regarded cohabitation and marriage as different life course strategies. They felt that young adulthood is a period characterized by uncertain external circumstances, in which people build up commitment in cohabitation without feeling limited in terms of opportunities. As dissolving a cohabiting union entails lower costs, the risk posed by this type of union was considered low. The respondents associated marriage with security and long-term commitment and saw it as an ideal for a later stage in life. They argued that romantic love and individual satisfaction should prevail throughout the entire marriage. Core terms of the individualization thesis - commitment, romantic love, and risk - were perceived differently between cohabitation and marriage. We conclude that the individualization thesis best fits young adulthood and is less relevant for later life stages.

L'estudi inclou una anàlisi d'eficiència de les universitats del sistema públic espanyol usant la tècnica no paramètrica d'anàlisi de fronteres coneguda com a Data Envelopment Analysis (DEA). Mitjançant una tria d'inputs i outputs vinculats a la docència i a la recerca, s'obté una classificació de les universitats en eficients i no eficients, indicant per aquestes últimes el grau o nivell d'ineficiència. Usant una tècnica desenvolupada a partir del DEA, s'aborda l'estudi de les unitats eficients per tal de determinar-ne aquelles que presenten un comportament atípic i, per a la resta, obtenir-ne una ordenació segons els nivells del que es coneix com eficiència robusta o supereficiència. En una segona part, s'analitza la possible influència de variables o factors ambientals -inputs no controlables- sobre els resultats d'eficiència obtinguts, usant, per tal de comparar resultats, una tècnica paramètrica basada en una análisi de regressió, i una no paramètrica coneguda com a model de valors ajustats.

How may a reader who wishes to read the Christian Bible as scripture well today be formed; and how may interpretations of scripture inform such concern? The present work is an exploration of this under-considered question in the field of contemporary biblical scholarship via sustained exegetical engagement with three biblical texts, namely Nehemiah 9:6 – 37, Ezekiel 20:5 – 32 and Acts 7:2 – 60, which offer three different inner-canonical readings of scripture in the form of three distinctive recitals of Israel’s story. The purpose is to consider how these retellings read scriptural traditions in relation to the wider context of the Christian canon; and to reflect on their enduring and formative significance as scripture for readers seeking to appropriate the scripture faithfully today. Chapter one will indicate that the concern of the present work is not a recent one, but rather one that is integral to a Christian practice of reading scripture. This chapter will also consider how such a concern once under-explored in biblical scholarship is now receiving some renewed attention in the field of theological interpretation of scripture. An overview of selected works pertaining to such concern will be considered in chapter two as a means to set a context for articulating the approach and rationale of the present work. In chapters three through to five, each chapter will be devoted to each of the three biblical texts, Nehemiah 9:6 – 37, Ezekiel 20:5 – 31 and Acts 7:2 – 60, to consider how scriptural traditions are interpreted in these three texts in relation to the wider context of the Christian canon. The next step is to reflect on the implications of these three biblical texts as Christian scripture for readers seeking to interpret scripture faithfully today. For such concern, the three texts will be considered individually at the end of chapters three, four and five respectively and then in concert in chapter six.

La culture de cortex ovarien se pose comme une alternative à l’autogreffe après autoconservation ovarienne. Pour améliorer la survie folliculaire après congélation, nous avons évalué chez la brebis l’influence de l’épaisseur de cortex ; 0,5 vs 1mm, du support de culture et du cryoprotecteur DMSO 2M vs PROH 1,5M+sucrose 0,3M. L’épaisseur de tissu de 1mm sur filtre millicell avec le protocole de congélation DMSO 2M a montré les meilleurs résultats avec 39% de survie folliculaire au 4e jour de culture. Sur tissu frais de brebis et humain, nous avons exploré un éventuel déficit d’expression en culture de Gdf9, de Bmp15 et de leurs récepteurs qui expliquerait le blocage de croissance folliculaire in vitro. Dans les deux modèles la présence des ARNm et des protéines de Gdf9 et Bmp15 est maintenue et l’expression protéique de leurs récepteurs BmprIA, BmprIB, BmprII est conservée. Ce travail montre que la survie en culture après cryoconservation peut être optimisée, la cause du blocage de croissance reste à définir
Culture of ovarian cortex can be an alternative to autograft after ovarian cryopreservation. To improve follicular survival after freezing we evaluated the influence of the sheep cortex width; 0. 5 vs 1mm, of the culture support and the cryoprotector DMSO 2M vs PROH 1. 5M + sucrose 0. 3M. The 1mm tissue width on millicell filter with the 2DMSO 2M freezing protocol displayed better results with 39% follicular survival on the 4th day of culture. On fresh human and sheep tissue, we explored a potential expression deficit in culture of Gdf9, Bmp15 and their receptors; this would explain the blocking of follicular growth in vitro. In both cases the level of ARNm and proteins Gdf9 and Bmp15 is maintained and so is the protein formula of their BmprIA, BmprIB, BmprII receptors. This study shows how survival in culture after cryopreservation can be optimised; the reason of the growth blocking remains to be determined

L’esport escolar és un pilar bàsic i fonamental en la pràctica esportiva de la persona. És una porta d’entrada a la vida esportiva i una experiència que marca al futur adult en aquest àmbit. A més dels nombrosos beneficis físics, psicològics i socials, la pràctica esportiva és un excel·lent marc per contribuir a l’educació dels nois i noies però aquest fet no es dona de manera natural, sinó que dependrà de l’orientació que li donin els agents implicats en aquesta pràctica. La família és el més potent agent socialitzador en les primeres edats, i el seu comportament envers la pràctica esportiva dels seus fills influenciarà la percepció que aquests en tinguin. Aquesta Tesi Doctoral té la voluntat de detectar i analitzar com es comporten els acompanyants com a espectadors a les competicions de futbol en edat escolar des del punt de vista dels principals agents implicats (acompanyants, esportistes, tècnics i àrbitres). Per portar a terme la recerca s’han realitzat grups de discussió per extreure categories de comportament que posteriorment s’han inclòs en un qüestionari que s’ha administrat manualment a les 13 delegacions de futbol catalanes, a un total de 4470 persones. Els resultats obtinguts ens ajuden a retratar un estat de la qüestió en el que s’observa que la percepció difereix segons l’agent implicat i la delegació a qui es pregunta, però on tots coincideixen que els comportaments per part dels acompanyants són majoritàriament positius però que tenen encara marge de millora.
El deporte escolar es un pilar básico y fundamental en la pràctica deportiva de la persona. Es una puerta de entrada a la vida deportiva y una experiencia que marca al futuro adulto en este ámbito. Además de los numerosos beneficiós físicos, psicològicos y sociales, la práctica deportiva es un excelente marco para contribuir a la educación de los chicos y chicas, però este hecho no se da de forma natural, sinó que dependerá de la orientación que le den los agentes implicados. La família es el más potente agente socializador en las primeras edades y su comportamiento hacia la práctica deportiva de susu hijos influirá en la percepción que estos tengan. Esta Tesis Doctoral tiene la voluntad de detectar y analizar cómo se comportan los acompañantes como espectadores a las competiciones de futbol en edad escolar desde el punto de vista de los principales agentes implicados (acompañantes, deportistas, técnicos y árbitros). Para llevar a cabo esta investigación se han realizado grupos de discusión para extraer categorias de comportamiento que posteriormente se han incluido en un cuestionario que se ha administrado manualmente a las 13 degelaciones de futbol catalanas, a un total de 4470 personas. Los resultados obtenidos nos ayudan a retratar un estado de la cuestión en el que se observa que la percepción difiere según el agente implicado y la delegación a quien se pregunta, pero todos coinciden en que los comportamientos por parte de los acompañantes son mayoritariamente positivos.
School sport is a basic and fundamental pillar in every person’s sports practice. It’s an entrance door for the sporting life and an experience that will outline the future adult in this area. In addition to numerous physical, psychological and social benefits, sports practice is an excellent framework to contribute to youth education, yet this fact isn’t natural, but it will depend on the orientation that the agents involved give. Family is the most powerful socialising agent at an early age, and its behaviour regarding sports practice of their children will influence their perception. This Doctoral Thesis claims to detect and analyse the behaviour of companions as spectators in school-age football competitions from the point of view of the main agents involved (companions, athletes, technicians and referees). In order to carry out this research, different discussion groups have been performed in order to extract behaviour categories that, subsequently, have been included in a questionnaire managed manually to 13 Catalan football delegations, a total of 4470 people. The results obtained help us to portray a state of the question, where the perception differs according to the agent involved and the delegation which is asked, but everyone agrees that companions behaviours are mainly positives.

AbstractDespite a long history of botanical collecting in the three oceanic islands of the Gulf of Guinea, no recent studies have documented floristic patterns. This chapter summarizes information on the vascular plants of the islands, including inventories conducted on Príncipe and São Tomé since 2017, as well as two recent expeditions to Annobón. An updated database of the vascular flora was compiled, which includes 14,376 records representing 1285 species and infraspecific taxa (1028 native). Príncipe has 445 species and infraspecific taxa (394 native), São Tomé has 1044 (842 native), and Annobón has 344 (274 native). Recent inventory work has generated collections of more than 90% of the endemic woody species. Several very rare taxa were rediscovered, including Balthasaria mannii (Oliv.) Verdc., 1969 (Pentaphylacaceae) and Psychotria exellii R. Alves, Figueiredo and A.P. Davis, 2005 (Rubiaceae), neither of which had been seen for more than 50 years. At least 17 species new to science were also discovered on Príncipe and São Tomé. Of the 1028 indigenous taxa, 164 (16%) are currently considered endemic to the islands. Of the 285 species evaluated according to the IUCN Red List criteria, 2 (0.7%) were Data Deficient, 226 (79.3%) Least Concern or Near Threatened, 55 (19.3%) threatened (including 3 Critically Endangered, 21 Endangered, and 31 Vulnerable), and 2 (0.7%) Extinct. On São Tomé and Príncipe, 325 plant species are used in traditional medicine, 37 of which are endemic. These results should be used to identify new priority sites for conservation, including on Annobón, where priority sites are less well defined.

Introduction: Little is known about the impact of enzyme-inducing antiseizure drugs (EI-ASD) on the reduction of Bone Mineral Density (BMD) in men with epilepsy (MWE). Objectives: To evaluate the BMD in MWE exposed to EI-ASDs (phenytoin, carbamazepine and phenobarbital) and its relationship with the duration of epilepsy. Methods: We evaluated BMD from 74 consecutive MWE (median age (range), 52.5 (25- 74) years) exposed to previous or current EI-ASDs, followed at UNICAMP-Brazil. Individuals were split into two groups (young-group, 31 individuals [25-49 years]; older group, 43 subjects, [50-74 years]). The BMD test evaluated t-score indexes from the femoral neck, whole femur and lumbar spine. Osteopenia was defined with t-score of - 1.0 to -2.4; osteoporosis, with T-scores lower than -2.5. Data were extracted from medical records. We analyzed data with SPSS22, performed chi-square tests for categorical variables and applied a partial correlation test (controlled for age) between BD scores and duration of epilepsy. Results: BMD was reduced in 49/74 men (66.2%). Both groups presented equivalent proportions of BMD abnormalities (p=0.087) (young-group [14/41 normal (45%), 12/31 osteopenia (39%), 5/31 osteoporosis (16%)]; older-group [11/43 normal (26%), 16/43 osteopenia (37%), 16/43 osteoporosis (37%)]. BMD did not correlate with the duration of disease or age of onset. Conclusion: BMD reduction is highly prevalent in MWE exposed to EI-ASD, including young individuals. Data suggest that exposure to EI-ASD may associate with early BMD reduction, which evolve to osteopenia and osteoporosis. BMD evaluation in MWE and appropriate treatment may be necessary to reduce fractures’ risk.

The central theme of this study is to identify strength-stiffness correlations for chemically treated subgrade soils in Indiana. This was done by conducting Unconfined Compression (UC) Tests and Resilient Modulus Tests for soils collected at three different sites—US-31, SR-37, and I-65. At each site, soil samples were obtained from 11 locations at 30 ft spacing. The soils were treated in the laboratory with cement, using the same proportions used for construction, and cured for 7 and 28 days before testing. Results from the UC tests were compared with the resilient modulus results that were available. No direct correlation was found between resilient modulus and UCS parameters for the soils investigated in this study. A brief statistical analysis of the results was conducted, and a simple linear regression model involving the soil characteristics (plasticity index, optimum moisture content and maximum dry density) along with UCS and resilient modulus parameters was proposed.

Abstract:
The Chilean pension system was hit hard during 2020-2021 by the withdrawal of 25 per cent of the individual pensions funds accumulated by 2019, an amount equivalent to 20 per cent of Chiles GDP. We estimate here the impact of those withdrawals on new pension allowances, using a combination of official data sets and the IDB model for the actuarial projection of pensions, including its heterogeneity matrix, to simulate the distribution of pension impacts. The withdrawal impact decreases in new retirees of future years until disappearing around the year 2065. We estimate respective impacts of about 31 percent and 37 percent for males and females new self-financed pensions around the year 2022, which goes to about 56 percent among the third of the affiliates with the lowest savings. However, we found that the recent increase in non-contributory pensions more than counteracted this impact for roughly 90 percent of 2022 new retirees. Regarding labor markets shocks, we found only a moderate role for them in the long-term evolution of the pension system, as we also found to be the case of seven Caribbean countries (Cuba, Haiti, Dominican Republic, Jamaica, Trinidad and Tobago, Bahamas and Barbados). As an overall conclusion, we recommend studying contribution rates, because low-salary workers attain a substantial replacement rate with just the non-contributory pension, which casts doubt on whether a mandatory contribution is appropriate for them.

Purpose: The purpose of this study was to validate the second version of the Prostate Imaging Reporting and Data System (PI-RADSv2) scores in predicting positive in-bore MRI-guided targeted prostate biopsy results across different non-university related institutions. The study focuses on PI-RADS v2 scoring because during the study period, PI-RADS v2.1 had not been released. Materials and Methods: This was a retrospective review of 147 patients who underwent multiparametric magnetic resonance imaging (mpMRI) of the pelvis followed by in-bore MRI-guided targeted prostate biopsy from December 2014 to May 2018. All lesions on mpMRI were rated according to PI-RADS v2 criteria. PI-RADS v2 scores were then compared to MR-guided biopsy results and pre-biopsy PSA values. Results: Prostate Cancer (PCa) was detected in 54% (80/147) of patients, with more prostate cancer being detected with each subsequent increase in PI-RADS scores. Specifically, biopsy results in patients with PI-RADS 3, 4, and 5 lesions resulted in PCa in 25.6% (10/39), 58.1% (33/55), and 86.0% (37/43) respectively. Clinically significant PCa (Gleason score ≥7) was detected in 17.9% (7/39), 52.7% (29/55), and 72% (31/43) of cases for PI-RADS 3, 4, and 5 lesions respectively. When the PI-RADS scoring and biopsy results were compared across different institutions, there was no difference in the PI-RADS scoring of lesions or in the positive biopsy rates of the lesions. The sensitivity, specificity, PPV, and NPV for PI-RADS 3-4 lesions were also not statistically different across the institutions for detecting Gleason 7 or greater lesions. Conclusion: Our results agree with prior studies that higher PI-RADS scores are associated with the presence of clinically significant PCa and suggest prostate lesions with PI-RADS scores 3-5 have sufficient evidence to warrant targeted biopsy. The comparison of PI-RADS score across different types of non-university practices revealed no difference in scoring and biopsy outcome, suggesting that PI-RADS v2 can be easily applied outside of the university medical center setting. Clinical Relevance: PI-RADS v2 can be applied homogeneously in the non-university setting without significant difference in outcome.
