Background: Migraine is a polygenetic disease, considered as a channelopathy. The dysregulation of ion functioning due to genetic changes may activate the trigeminovascular system and induce migraine attack both migraine with aura (MA) and without aura (MO). Objectives: The aim of the study was to analyze the following variants of genes encoding ion channels and associated protein: c.3199G>A SCN1A, c.56G>A SCN2A, c.28A>G and c.328T>C KCNK18, c.3053A>G TRPA1, c.31-1811C>T STX1A in migraine patients. Patients and Methods: The study included 170 migraine patients and 173 controls. HRMA and Sanger sequencing were used for genotyping. Meta-analysis was performed for c.28A>G, c.328T>C KCNK18, and c.31-1811C>T STX1A. Patients and Methods: The study included 170 migraine patients and 173 controls. HRMA and Sanger sequencing were used for genotyping. Meta-analysis was performed for c.28A>G, c.328T>C KCNK18, and c.31-1811C>T STX1A. Results: AA genotype of c.56G>A SCN2A was found only in migraine patients. Patients with c.328T>C KCNK18 mutation had an increased risk of developing migraine before the age of 18. Moreover, individuals with AA/TC haplotype of KCNK18 had higher attack frequency than those with AA/TT (p<0.05). T allele of c.31-1811C>T STX1A was more frequent in MA patients than MO (p<0.05). The c.3053A>G TRPA1 polymorphism was more common in patients with migraine onset before the age of 15 (p<0.05), while c.31-1811C>T STX1A and c.3199G>A SCN1A before the age of 10 (p<0.01). Meta-analysis showed a significant association of c.31-1811C>T STX1A polymorphism with migraine overall (OR=1.22, p=0.0086), MA, and MO. No association was found for c.28A>G KCNK18, c.328T>C KCNK18, and migraine overall. Conclusions: Changes in genes encoding ion channels or proteins regulating their functioning may increase the risk of migraines and correlate with clinical features of disease, e.g. age of onset and attack frequency.

AbstractThe paper presents a novel family of arbitrary high order multioperators approximations for convection, convection-diffusion or the fluid dynamics equations. As particular cases, the 16th- and 32th-order skew-symmetric multioperators for derivatives supplied by the 15th- and 31th-order dissipation multioperators are described. Their spectral properties and the comparative efficiency of the related schemes in the case of smooth solutions are outlined. The ability of the constructed conservative schemes to deal with discontinuous solutions is investigated. Several types of nonlinear hybrid schemes are suggested and tested against benchmark problems.

A 31-year-old Japanese male patient with a history of atrial fibrillation showed elevated serum levels of free thyroxine and triiodothyronine and a normal level of thyrotropin. The same abnormal hormone pattern was also found in his son. These data indicated that the index patient and the son have thyroid hormone resistance syndrome. Exon sequencing using DNA from these two patients revealed that both patients harbored a heterozygous mutation in the THRB gene: G1244C in exon 9, which results in R320P substitution. Therefore, thyroid hormone resistance syndrome caused by THRB mutation (RTHβ) was diagnosed. The mutation of the 320th arginine to proline has not been found to date. In conclusion, herein, we have described the first case of RTHβ that is associated with R320P mutation.

Abstract:
Anaerobic ammonium oxidation (anammox) is a novel nitrogen pathway catalyzed by anammox bacteria which are obligate anaerobic chemoautotrophs. In this study, enrichment culture of marine anammox bacteria (MAAOB) from the samples related to seawater was conducted. Simultaneous removal of ammonium and nitrite was confirmed in continuous culture inoculated with sediment of a sea-based waste disposal site within 50 days. However, no simultaneous nitrogen removal was observed in cultures inoculated with seawater-acclimated denitrifying sludge or with muddy sediment of tideland even during 200 days. Nitrogen removal rate of 0.13 kg/m3/day was achieved at nitrogen loading rate of 0.16 kg/m3/day after 320th days in the culture inoculated with the sediment of waste disposal site. The nitrogen removal ratio between ammonium nitrogen and nitrite nitrogen was 1:1.07. Denaturing gradient gel electrophoresis (DGGE) analysis indicated that an abundance of the bacteria close to MAAOB and coexistence of ammonium oxidizing bacteria and denitrifying bacteria in the culture.

The survival of E. co/i0157:H7 in Feta cheese and the brine used after 2 months ripening of cheese, was studied during storage at 4°C and 12°C. An inoculum of 5.3 log10CFU/ml of E. coli Ol57:H7 was added in brine, for the contamination with the pathogen of Feta cheese. E. coli Ol 57:H7 was detectable in brine by direct plating on SMAC up to 40th day and 32th day during its storage at 4°C and 12°C respectively, while by enrichment and following plating on SMAC up to 44th day (4°C) and 36th day (12°C). The E. coli 0157:H7 was detectable in Feta by direct plating on SMAC up to 44th day (4°C) and 38th day (12°C), while by enrichment and following plating on SMAC up to 50th day (4°C) and 42th day (12°C). The results showed that E. coli Ol57:H7 survived longer in Feta than in its brine, for both storage temperatures but the difference was very low. The survival of E. coli 0157:H7 in feta and brine was longer during the storage at 4°C than at 12°C.

Introduction The learning curves analysed to date for robot-assisted laparoscopic prostatectomy are based on arbitrary cut-offs of the total cases. Methods We analysed a large dataset of robot-assisted laparoscopic prostatectomies from a single centre between 2008 and 2019 for assessment of the learning curve for perioperative outcomes with respect to time and individual cases. Results A total of 1,406 patients were evaluated, with mean operative time 198.08 minutes and mean console time 161.05 minutes. A plot of operative time and console time showed an initial decline followed by a near-constant phase. The inflection points were detected at 1,398 days (308th case) for operative time and 1,470 days (324th case) for console time, with a declining trend of 8.83 minutes and 7.07 minutes, respectively, per quarter-year (p<0.001). Mean estimated blood loss showed a 70.04% reduction between the start (214.76ml) and end (64.35ml) (p<0.001). The complication rate did not vary with respect to time (p=0.188) or the number of procedures (p=0.354). There was insufficient evidence to claim that the number of operations (p=0.326), D’Amico classification (p=0.114 for intermediate versus low; p=0.158 for high versus low) or time (p=0.114) was associated with the odds of positive surgical margins. Conclusions It takes about 300 cases and nearly 4 years to standardise operative and console times, with a requirement of around 80 cases per annum for a single surgical team in the initial years to optimise the outcomes of robot-assisted laparoscopic prostatectomy.

The present study was conducted at Chaudhary Charan Singh Haryana Agricultural University, Hisar, Regional Research Station, Bawal, Haryana (India). The larvae of castor semilooper remain active from 30th standard weeks to 47th standard weeks. The maximum larval population was recorded on the 32th standard week (6.1 larvae per plant) and 38th standard meteorological weeks (6.2 larvae per plant) due the high rainfall, 158 and 120 mm, during these weeks, respectively. Correlation indicates that minimum temperature, evening relative humidity and rainfall exhibited a positive relationship (r- 0.588, 0.577 and 0.650) with the larval population. Bio-efficacy of four insecticides with different doses was tested against A. janata infesting castor crop. One day after spraying thiodicarb @ 468 g proved best toxicant followed by thiodicarb @ 375 g, deltamethrin @ 11.2 g, deltamethrin @ 10.5 g, quinalphos @ 250 g, quinalphos @ 200 g, novaluron @ 100 g and novaluron @ 75 g. At three days after spraying, it was found that novaluron @ 100 g was the most effective followed by novaluron @ 75 g, thiodicarb @ 468 g, deltamethrin @ 11.2 g, thiodicarb @ 375 g, deltamethrin @ 10.5 g, quinalphos @ 250 g and quinalphos @ 200 g. At seven days after spraying novaluron @ 100 g proved to be best toxicant followed by novaluron @ 75 g, thiodicarb @ 468 g, thiodicarb @ 375 g, deltamethrin @ 11.2 g, deltamethrin @ 10.5 g, quinalphos @ 250 g and quinalphos @ 200 g. Novaluron 100 g was the overall most effective with 75.97 mean per cent reduction in larval population.

SummaryThe impact of maternal height, pre-pregnancy weight status and gestational weight gain on fetal growth patterns and newborn size was analysed using a dataset of 4261 singleton term births taking place at the Viennese Danube Hospital between 2005 and 2013. Fetal growth patterns were reconstructed from three ultrasound examinations carried out at the 11th/12th, 20th/21th and 32th/33th weeks of gestation. Crown–rump length, biparietal diameter, fronto-occipital diameter, head circumference, abdominal transverse diameter, abdominal anterior–posterior diameter, abdominal circumference and femur length were determined. Birth weight, birth length and head circumference were measured immediately after birth. The vast majority of newborns were of normal weight, i.e. between 2500 and 4000 g. Maternal height showed a just-significant but weak positive association (r=0.03: p=0.039) with crown–rump length at the first trimester and with the majority of fetal parameters at the second trimester (r>0.06; p<0.001) and third trimester (r>0.09; p<0.001). Pre-pregnancy weight status was significantly positively associated with nearly all fetal dimensions at the third trimester (r>0.08; p<0.001). Maternal height (r>0.17; p<0.001) and pre-pregnancy weight status (r>0.13; p<0.001), but also gestational weight gain (r>0.13; p<0.001), were significantly positively associated with newborn size. Some of these associations were quite weak and the statistical significance was mainly due to the large sample size. The association patterns between maternal height and pre-pregnancy weight status with fetal growth patterns (p<0.001), as well as newborn size (p<0.001), were independent of maternal age, nicotine consumption and fetal sex. In general, taller and heavier women gave birth to larger infants. This association between maternal size and fetal growth patterns was detectable from the first trimester onwards.

Foetal growth patterns were reconstructed by the results of three ultrasound examinations carried out at the 11th/12th, 20th/21th and 32th/33thweek of gestation. In detail, crown-rump length, biparietal diameter, fronto-occipital diameter, head circumference, abdominal transverse diameter, abdominal anterior-posterior diameter, abdominal circumference, and femur length were determined. Birth weight, birth length and head circumference were measured immediately after birth. Young teenage mothers (≤15 years), older adolescent mothers (16-19 years), mothers of optimal age range (20-35 years) and mothers of advanced age (>35 years) differed significantly in body height, pre-pregnancy weight status and gestational weight gain. Surprisingly, the foetuses of young adolescent mothers were the largest ones during first trimester. During the second and third trimester however, the foetuses of mothers of optimal age range (20-35 years) and mothers older than 35 years showed larger biometric dimensions than adolescent mothers. According to multiple regression analyses, maternal age was significantly related to Foetal head size (ß =-0.04; 95% CI = -0,08 - 0.01; p=0.034) and abdominal dimensions (ß= 0.03; 95% CI = 0.01-0.05; p=0,011) during the second trimester and to birthweight (ß= -0.03; 95% CI= -4.40 - 0.04; p=0.050). The associations however, are quite weak and the statistical significance is maybe due to the large sample size. At the time of birth, offspring of mothers of optimal age range (20 to 35 years) is significantly larger than that of adolescent mothers and mothers of advanced age. Mothers of advanced age showed the significantly highest (p=<0.0001) prevalence (5.6%) of SGA newborns (<2500g). The small size of newborn among young adolescent mothers may be due to a competition over nutrients between the still growing mothers and the foetuses during the third trimester, while placental ageing may be responsible for smaller size of offspring among mothers of advanced age.

Fetal biometry was reconstructed by the results of three ultrasound examinations carried out at the 11th/12th, 20th/21th and 32th/33thweek of gestation. In detail, crown-rump length, biparietal diameter, fronto-occipital diameter, head circumference, abdominal trans-verse diameter, abdominal sagittal diameter, abdominal circumference, and femur length were determined. Birth weight, birth length and head circumference were measured immediately after birth. Four delivery modes were compared: spontaneous vaginal birth, instrumental vaginal birth, planned cesarean section and emergency cesarean section.The total cesarean section rate was 10.2%. Fetal biometry and newborn size differed significantly between the four delivery modes. From the second trimester onward, head circumferences were significantly larger (p=0.005) among fetuses delivered by instrumental delivery or emergency cesarean section than among fetuses delivered by spontaneous vaginal birth. The fetal abdominal dimensions during the third trimester were significantly largest (p=0.001) among fetuses delivered by emergency cesarean section. In comparison to spontaneous vaginal delivery the risk to require instrumental delivery increased significantly with increasing fetal head dimensions at the second (p=0.019) and third trimester(p=0.032) independent of maternal somatic factors. The risk of emergency CS increased significantly with increasing head dimensions (p=0.030) as well as abdominal dimensions (p=0.001) at the third trimester and newborn size (p=0.002), also independently of maternal somatic factors.In general, larger fetuses are on an increased risk of experiencing instrumental delivery or emergency caesarean section. This association between fetal size and delivery mode is detectable from the second trimester onwards.

All participants will undergo a clinical and functional assessment of the pelvic floor before the beginning of the technique practice, as well as a reassessment of these items and an assessment of perineal integrity between 45 and 60 days after delivery for a blind physiotherapist regarding the interventional procedures. The evaluative procedures will be done by a ‘blinded’ physiotherapist in relation to the intervention procedures.<span lang="EN-IN"> The outcome will be determined by </span>the perineal integrity by the presence or absence of perineal laceration as well as their characteristics observed immediately after delivery<span lang="EN-IN">. </span></p><p class="abstract"><strong>Conclusions:</strong> The knowledge of the effects of antenatal pelvic floor preparation techniques on the perineal integrity and pelvic floor muscles function after delivery, will allow a better choice about which approach is the best to pregnant women to prevent perineal trauma.</p>

Introduction Acquired and inherited thrombophilias are associated with unfavourable pregnancy outcome. Low-molecular-weight heparin (LMWH) has demonstrated utility in the prevention and treatment of thrombosis during the pregnancy. There is no consensus on the dose of LMWH for prophylaxis, between fixed dose and “adjusted” dose according to anti-Xa levels. We report our experience the management of heparin-therapy during pregnancy in thrombophilic women. Methods We performed a retrospective analysis of patients with high risk of venous thromboembolism (VTE) and obstetric complications during pregnancy. Table 1 shows the risk factors of VTE in pregnancy. The obstetric complications considered were: preeclampsia, fetal growth restriction, fetal death and recurrent miscarriages. All patients were prescribed enoxaparin 70-100 IU /Kg SC once daily. To determine the dose of LMWH we evaluated levels of anti-factor Xa (chromogenic test) and the utero-placental circulation with color Doppler ultrasound. Dose adjustment was performed if the activity of factor Xa was suboptimal (prophylaxis range > 0.4IU/ml and < 0.8 IU/ml anti-Xa value) or were detected alteration in the utero-placental circulation. Outcome Measures Time of delivery, birth weights and Apgar-index. Results 70 pregnancies for 68 women were recruited between January 2008 and December 2012. The median age of patients was 35 years (range 25- 43years) and 7 (10%) were over 40. Twenty-four patients had a previous VTE and thirty-six had an obstetric complications. No one patients reported thrombotic and/or hemorrhagic complications during heparin treatment. In 97 % of cases the time of delivery occurred after 36th weeks of pregnancy, with a birth weight greater than 2600 grams and Apgar score index higher than 7. The dose of LMWH had to be adjusted at least once during the course of the pregnancy and the mean daily dose was increased from 4.000UI to 8.000UI between the 12th and 32th week of gestation. Conclusion In pregnancy LMWH thromboprophylaxis enables the modulation of systemic haemostatic parameters by the inhibition of factor Xa and by increasing TFPI levels. In the literature-in some high risk pregnancy- the weight-based dosing of LMWH could seem to fail achieving a prophylactic anticoagulation. We previously reported that follicular wave synchronization, by removal of the dominant follicle on Day 5 after ovum pickup (OPU), was effective in increasing oocyte quality in the developing follicles (Imai et al. 2006 32th Annual Conference of the IETS, poster presentation no. 277). The current study was designed to examine the effect of superstimulatory treatment to induce subsequent follicular wave synchronization on embryo production by OPU and IVM-IVF-IVC in Holstein dry cows. Cows were reared under the same feeding and environmental conditions, and 2 OPU sessions were conducted in each cow. In the first session, OPU was performed in 8 cows on arbitrary days of the estrous cycle by using a 7.5-MHz linear transducer with needle (Cova needle, Misawa Medical, Tokyo, Japan) connected to an ultrasound scanner (SSD-1200, Aloka, Tokyo, Japan). Follicles larger than 8 mm in diameter were then aspirated and a CIDR was inserted on Day 5 (the day of first OPU session = Day 0). Cows then received 30 mg of FSH (Antrin-R10; Kawasaki Mitaka Pharmaceutical Co., Tokyo, Japan) twice a day from Days 7 to 10 in decreasing doses (6, 6, 4, 4, 3, 3, 2, 2 mg) by i.m. injection. Cloprostenol (PGF; Clopromate C; Sumitomo Pharmaceuticals Co., Tokyo, Japan; 0.75 mg) was administered in the morning of Day 9 (third day of superstimulation). The second OPU session was performed 48 h after PGF administration (Day 11), and only follicles larger than 5 mm in diameter were aspirated. The CIDR was removed from the cows just before OPU. Collected oocytes were evaluated by their cumulus cell morphology, cytoplasmic color, and density. Grades 1 and 2 COC were matured, fertilized, and cultured as described by Imai et al. [2006 J. Reprod. Dev. 52(Suppl.), S19–S29]. Embryo development was assessed by the cleavage rate on Day 2 and by the blastocyst formation rate on Days 7 to 8 (the day of insemination = Day 0). Data were analyzed by Student's t-test. There were no differences in the mean (� SD) number of aspirated follicles or collected oocytes between the first (32.5 � 6.8 and 26.0 � 12.7, respectively) and second (29.3 � 10.4 and 19.0 � 9.4, respectively) OPU sessions (P > 0.1). The percentage of Grade 1 and 2 oocytes for the second OPU session (90.5 � 13.8%) was significantly higher (P < 0.01) than for the first OPU session (63.1 � 6.3%), and significant differences were found for cleavage (79.4 � 14.1, 61.8 � 25.1, P < 0.01) and blastocyst rates (68.1 � 16.7, 24.2 � 22.3, P < 0.001) between sessions. The mean numbers of blastocysts obtained per session were 4.3 � 2.9 and 12.8 � 8.7 in the first and second sessions, respectively (P < 0.01). These results indicate that superstimulatory treatment and subsequent follicular wave synchronization were effective on in vitro embryo production by increasing the oocyte quality.

Abstract Abstract 4676 INTRODUCTION: Bernard Soulier Syndrome (BSS) is a rare qualitative and quantitative autosomal recessive platelet disorder molecularly characterized by defective GPIb-IX-V membrane glycoprotein that is related to platelet adhesion and interaction with both Von Willebrand factor and endothelium. It is usually expressed by an enlarged bleeding time and decreased yet giant sized platelets, and can cause severe hemorrhagic complications during pregnancy. CASE REPORT: We present two successful pregnancies and labour management on a 33-year-old woman diagnosed in her childhood with BSS. Ristocetin induced platelet agglutination test and membrane glycoprotein study were performed to get the diagnosis. Furthermore, in the last year we have completed the diagnosis with platelet receptor expression flow citometry analysis, finding out depleted antiGPIbα and antiGPIX antibodies. Changes which prevent GPIbα for correct expression and processing were observed. Also integrin αIIbβ3 levels were risen due to the big sized platelets. A PCR product was obtained from genomic DNA using the GPIbα oligonucleotide pair; sense and antisense, and direct DNA sequencing of the amplification was performed in a model ABIprism 377 DNA sequencer (Perkin-Elmer Cetus). The direct sequencing of the sense strand of the 5x fragment shows in the patient the simultaneous presence of G and C nucleotids in 688 position, and the T and A nucleotids in 715 position. These punctual mutations give Ala200Pro substitutions and Cys209Ser in heterozygosis in mature peptide, respectively, Figure 1. The Ala200Pro mutation has not been reported in others cases of Bernard-Soullier, previously. The Cys209Ser has been described in other two Spanish patients: one in homozygosis and the other in heterozygosis associated to the mutation. Our patient presented hemorrhagic symptoms from 18 month old with an average of 40×103/μ l platelet count and increased platelet volume who required multiple platelet transfusions during her childhood. In the menarche main hemorrhagic manifestations consisted on methrorragia and epistaxis which were controlled with tranexamic acid. At the age of 26 a spontaneous ovarian cyst rupture required urgent hospital admission and two platelet pool transfusion. On December 2007 she consulted for epistaxis, and on September 2008 cesarean delivery was planned at 38th week of pregnancy; platelet count was 40×103/μ l, antiplatelet antibodies were negative for membrane glycoproteins and antiHLA-1 was positive against 98% donors tested. As a result, an HLA-compatible donor was searched, and two platelet apheresis were transfused one hour before surgery and every 12 hours after the procedure. The next 4 days one apheresis every 48 hours were transfused with no hemorrhagic symptoms. On January 2010, she presented with her second pregnancy; the patient had severe anemia, with high transfusion requirements; neither hemorrhage or hemolysis was detected, and even the possibility of a Munchausen syndrome was questioned. Unplanned caesarean was performed at 32th week because of the clinical instability of the patient, and this time we could not find on time an HLA-compatible donor, so two random platelet pools were transfused one hour before surgery and one platelet apheresis every day during 4 days after caesarean deliver, without any hemorrhagic complication. The newborn was asymptomatic with normal platelet count. The patientxs haemoglobin level returned to normal after five days of delivery. CONCLUSIONS: We present the successful labour management of two consecutive pregnancies on a young woman diagnosed with BSS. The low prevalence of this syndrome (less than one case per million population) explains the absence of well-established protocols for the management of pregnancy in this disease. Up to date there are only twelve reported cases of pregnancy and delivery in BSS patients, most of them presenting with severe hemorrhagic complications. Correct platelet transfusion is the mainstay of treatment, and screening for anti-platelet antibodies study is mandatory due to its high prevalence in these patients. <p>Anggrek <em>Phalaenopsis</em> memiliki nilai komersial yang tinggi, karena keindahannya dapat dinikmati sepanjang tahun. Hal tersebut berdampak pada kebutuhan benih tanaman yang semakin meningkat. Salah satu cara penyediaan benih secara massal adalah melalui perbanyakan klonal secara in vitro sehingga perlu dilakukan studi kualitas regeneran hasil perbanyakan klonal untuk menjamin ketersediaan benih dengan kualitas baik. Penelitian bertujuan menguji kualitas regeneran yang dihasilkan dari perbanyakan klonal secara in vitro beberapa varietas Phalaenopsis dengan menggunakan eksplan yang berbeda. Penelitian dilaksanakan di Laboratorium Kultur Jaringan, Kebun Percobaan Segunung, Balai Penelitian Tanaman Hias (Balithi) sejak bulan Januari 2014 hingga Mei 2015. Penelitian menggunakan dua faktor, yaitu varietas (Ayu Lestari, Ayu Pratiwi, dan Karindra) dan jenis eksplan (tangkai infloresen, tunas pucuk, dan empulur). Percobaan disusun menggunakan rancangan acak kelompok pola faktorial dan setiap perlakuan diulang tiga kali. Hasil penelitian menunjukan tidak terjadi interaksi yang nyata antara faktor jenis eksplan dan varietas yang diujikan pada semua tahap percobaan. Respon terbaik diperoleh pada eksplan empulur dengan 42,85% eksplan berhasil membentuk kalus pada minggu ke-8 dan hampir 100% kalus tersebut dapat beregenerasi menjadi tunas pada minggu ke-24 dengan tingkat multiplikasi tunas 1,87 kali. Pada minggu ke-32 terbentuk rata-rata 3,13 daun per planlet dengan 2,47 cm panjang daun, 1,36 cm lebar daun, 1,52 akar per planlet, dan panjang akar per planlet mencapai 1,26 cm. Kerapatan stomata memiliki korelasi negatif dengan tingkat abnormalitas planlet. Planlet dengan kerapatan stomata tertinggi dan abnormalitas yang rendah diperoleh pada var. Karindra dan planlet yang berasal dari eksplan empulur dan tunas pucuk. Setelah 8 minggu tahap aklimatisasi, tingkat keberhasilan hidup tertinggi (92%) diperoleh pada tunas yang berasal dari eksplan empulur. This condition gives the impact on the increasing demand of the seeds. One of the ways of providing mass seeds is through in vitro clonal propagation. However, it is necessary to study the quality of regenerants of clonal propagation products to ensure the availability of qualified seeds. The aimed of this study was to test the quality of regenerants obtained from in vitro clonal propagation of Phalaenopsis using inflorescence stalk, shoot tips, and pith explants. This research was conducted at Tissue Culture Laboratory, Segunung Experimental Station, Indonesian Ornamental Crops Research Institute (IOCRI) from January 2014 to May 2015. The study used two treatments, varieties (Ayu Lestari, Ayu Pratiwi, and Karindra) and type of explant (inflorescence stalk, shoot tips, and pith). Experiments were prepared using a randomized complete block design with two factors and each treatment was replicated three times. The results showed there were no significant interaction between types of explants and varieties tested in all experiment stages. The best response was obtained using pith explants with 42.85% callus formation in the week eighth and nearly 100% callus can regenerate into shoots at week 24th with the rate of shoot multiplication up to 1.87 times. At week 32th the cultures formed planlets with an average number of leaves of 3.13 and an average size of 2.47 cm x 1.36 cm (length x width) and an average number of roots of 1.52 with average length reached 1.26 cm. Stomatal density has negative correlation with plantlet abnormality rate. Plantlets with the highest stomatal density and low abnormality were obtained in var. Karindra and plantlet derived from explant pith and shoot buds. After 8 weeks of acclimatization stage, the highest survival rate (92%) was obtained on the shoot originating from pith explant. This study proved that varietal differences did not have a significant effect on regenerant abnormalities, and the highest number of regenerant with good quality (low abnormality rate) was obtained from pith explant.</p>

Purpose Electromagnetic noise of permanent magnet synchronous motor (PMSM) seriously affects the sound quality of electric vehicles (EVs). This paper aims to present a comprehensive process for the electromagnetic noise analysis and optimization of a water-cooled PMSM. Design/methodology/approach First, the noises of an eight-pole 48-slot PMSM in at speeds up to 10,000 rpm are measured. Furthermore, an electromagnetic-structural-acoustic model of the PMSM is established for multi-field coupling simulations of electromagnetic noises. Finally, the electromagnetic noise of the PMSM is optimized by using the multi-objective genetic algorithm, where a multi-objective function related to the slot width of PMSM stator is defined for radial electromagnetic force (REF) optimization. Findings The experimental results show that main electromagnetic noises are the 8n-order (n = 1, 2, 3, …) and 12-order noises. The simulated results show that the REFs are mainly generated by the 8n-order (n = 1, 2, 3, 4, 5, 6) vibrations, especially those of the 8th, 16th, 24th and 32th orders. The 12-order noise is a mechanical noise, which might be caused by the bearings and other structures of the PMSM. Comparing the simulated results before and after optimization, both the REFs and electromagnetic noises are effectively reduced, which suggests that an appropriate design of stator slot is important for reducing electromagnetic noise of the PMSM. Originality/value In view of applications, the methods proposed in this paper can be applied to other types of PMSM for generation mechanism analysis of electromagnetic noise, optimal design of PMSM and thereby noise improvement of EVs.

Abstract Background Tetrapyrroles play indispensable roles in various biological processes. In higher plants, glutamate 1-semialdehyde 2,1-aminomutase (GSAM) converts glutamate 1-semialdehyde (GSA) to 5-aminolevulinic acid (ALA), which is the rate-limiting step of tetrapyrrole biosynthesis. Up to now, GSAM genes have been successively identified from many species. Besides, it was found that GSAM could form a dimeric protein with itself by x-ray crystallography. However, no mutant of GSAM has been identified in monocotyledonous plants, and no experiment on interaction of GSAM protein with itself has been reported so far. Result We isolated a yellow leaf mutant, ys53, in rice (Oryza sativa). The mutant showed decreased photosynthetic pigment contents, suppressed chloroplast development, and reduced photosynthetic capacity. In consequence, its major agronomic traits were significantly affected. Map-based cloning revealed that the candidate gene was LOC_Os08g41990 encoding GSAM protein. In ys53 mutant, a single nucleotide substitution in this gene caused an amino acid change in the encoded protein, so its ALA-synthesis ability was significantly reduced and GSA was massively accumulated. Complementation assays suggested the mutant phenotype of ys53 could be rescued by introducing wild-type OsGSAM gene, confirming that the point mutation in OsGSAM is the cause of the mutant phenotype. OsGSAM is mainly expressed in green tissues, and its encoded protein is localized to chloroplast. qRT-PCR analysis indicated that the mutation of OsGSAM not only affected the expressions of tetrapyrrole biosynthetic genes, but also influenced those of photosynthetic genes in rice. In addition, the yeast two-hybrid experiment showed that OsGSAM protein could interact with itself, which could largely depend on the two specific regions containing the 81th–160th and the 321th–400th amino acid residues at its N- and C-terminals, respectively. Conclusions We successfully characterized rice GSAM gene by a yellow leaf mutant and map-based cloning approach. Meanwhile, we verified that OsGSAM protein could interact with itself mainly by means of the two specific regions of amino acid residues at its N- and C-terminals, respectively.

AbstractThe influence of a probiotic additive and aromatic oils from medicinal and spicy plants on the basic production parameters and on the incidence of intestinal disorders of weaned pigs (weaning at the age of 30–32 days) was compared in two trials in a commercial pig fattening farm (Michalovce, Slovakia). In the first trial, an experimental (n ex = 38) and a control group (n co = 32) were used. A commercial probiotic additive containing Enterococcus faecium M-74 (6 × 108 CFU/g) and IgY antibodies (920 mg/g dried activated egg matter) against pig Rotavirus and different strains of Escherichia coli (K88, K99, 987P, F18) were added to feed mixture of the experimental group from 30th to 40th day (12 × 108 CFU/g) and subsequently from 40th to 50th day (6 × 108 CFU/g) of age. The second trial was performed with three groups of pigs (n ex1 = 28, n ex2 = 24, n co = 28). Aromatic oils isolated from sage (Salvia officinalis L., Labiateae) or oregano (Origanum vulgare L., Lamiaceae) were added to feed mixtures of the 1st (90 mL/100 kg) and 2nd experimental group (35 mL/100 kg), respectively, from 32th to 53th day of age. A statisticaly non-significant decrease of the incidence of diarrhoeal diseases was observed in the experimental groups of both trials as follows: in the 1st trial by 43.41%, whereas in the 2nd trial by 20.03% (1st exp. group) and 6.72% (2nd exp. group) in comparison with the control groups. The daily weight gains increased in the 1st trial by 42.95 g/day, while in the 2nd trial by 69.52 g/day (1st exp. group) and 105.23 g/day (2nd exp. group; p < 0.05) compared with controls.

Abstract:
Background: Mung bean is important pulse crop in India due to its nutritional value. Its grain contains protein, fat, carbohydrate, vitamin and minerals therefore human uses it in various ways in food. Mung bean crop cultivated in kharif, rabi and summer season in India. Various species of insect pests are infested to mung bean crop and cause very harmful effect to crop and farmer. These pests decrease productivity and quality of mung bean. Methods: Field experiment was carried out for the study of seasonal incidence of insect pests on RMG-62 variety of green gram during kharif season 2019. Mung bean was grown on a plot size of 25 m × 25m with 50 cm row to row and 20 cm plant to plant spacing. Randomly selected 20 plants from weekly interval to record population of insect pests. The population of jassid and whitefly was recorded by counting number of pests on 2 upper, 2 middle and 2 lower leaves of a plant whereas population of aphid was recorded by counting number of aphid on 10 cm twig/plant. Result: The mean population of aphid, jassid and whitefly were recorded. Incidence of aphid started during 33th standard week with 0.9 aphid/10cm twig/plant, jassid and whitefly started during 32th standard week with 0.3 jassid/6 leaves/plant and 2.1 whitefly/6 leaves/plant, respectively. Peak mean population of aphid, jassid and whitefly reached during 36th standard week with 10.2 aphid/10cm twig/plant, 10.1 jassid/6 leaves/plant and 14.1 whitefly/6 leaves/plant, respectively. Simultaneously six species of insect pests also infested green gram, these were Mylabris pustulata, Helicoverpa armigera, Trichoplusia ni., Lampides boeticus, Spoladea recurvalis and Diaphania indica. The population of aphid, jassid and whitefly positively correlated with temperature. Population of aphid and whitefly had positive correlation with humidity, whereas jassid population had positive correlation with minimum humidity and negative correlation with maximum and average humidity. All three pests expressed negative correlation with rainfall.

Background: Gestational diabetes mellitus (GDM) is a glucose intolerance that occurs or is identified for the first time all through pregnancy. Perinatal & Neonatal morbidity mortality is significant in pregnant women in GDM with extra hazard of growing diabetes later in life. Uttar Pradesh is a largest state of India with one of the highest rate of the infant as well as maternal mortality which might be, at least partially due to GDM. Thus, Careful evaluation, administration & Training of HCPs for GDM can improve the outcomes in National health Mission supported Govt funded Program, supported by World Diabetes foundation, Denmark. Aims & Objectives: Primary objective of this study to be determine the Maternal-Fetal outcomes of GDM and management of Hyperglycemia in Pregnancy HIP reduces Neonatal & Perinatal Mortality as per the NHM, GOI Guidelines for GDM, As this will go long way help us in reduction of Perinatal & infant mortality. Thus, this study was once undertaken to recognize the extent of the burden on the healthcare and formulating further policy for Implementation of Gestational Diabetes Program in the largest state of Uttar Pradesh. Materials and Methods: A prospective cohort study was done for 2 year from October 1, 2016, to September 31, 2018, at 828 GDM screening units as a part of the Gestational Diabetes Prevention and Control Project, Uttar Pradesh approved by the Indian Government in the state of Uttar Pradesh, India, largest state with second Highest MMR & IMR, A total of 515,532 pregnant women were screened during their 16–32th weeks of pregnancy by impaired oral glucose test (OGTT) as per NHM Guidelines for GDM, 12784 GDM & 7287 Non GDM maternal and perinatal outcomes were followed up in both GDM and non‑GDM categories in the 2 year (2016-2018) after blood sugar management (September 2016-October 2018) was executed at 828 (DHs, CHCs & PHCs healthcare) facilities, 515532 Pregnant Women have been screened at 16-20 Weeks & 24th-28 weeks of pregnancy as per Guidelines of National health Mission, GOI Guideline. Results: Perinatal mortality increased significantly from 2.6% to 9.1% when blood sugar levels increased from 120 mg/dl to 199 mg/dl and above. Perinatal mortality in GDM cases were significantly to the control of blood sugar levels (P < 0.0001). Relative Risk of Stillbirth, Perinatal & neonatal mortality have been respectively 2.5, 2.3 & 2.5 times greater in GDM compare Non GDM (Table 1). Most of the GDM used to be identified in primigravida (52%). It was also found in our study those GDM who were strictly controlled with Hyperglycemia in pregnancy (HIP) to <120 mg/dl, Post Prandial blood sugar, have lowest risk for perinatal and neonatal mortality compare to those GDM pregnant women Blood sugars were not controlled, Risk for Perinatal mortality increases steadily and reaches 9.1% beyond blood sugar> 200 mg/dl. Conclusion: All the Pregnant women need screening in Public health facilities & Implementation of National health Mission, GOI Guidelines for GDM has to be followed to improve outcome for Mother and Newborn, As the lack of information about GDM amongst pregnant women is high, to decrease the risk, increase awareness & full Implementation of NHM GDM Guidelines is key to Perinatal and neonatal mortality reduction in Public health care facilities where large number of ANC visit for Maternal and fetal health care.
