This thesis argues that performance can be seen to constitute both a critical discipline and a set of activities entailing an engagement with spatial, temporal, physical and material relations, rather than as a product of linguistic, textual and discursive relations. As such, performance is able to critique the functioning of language, text and discourse in assuming space, time, bodies and matter. Performance also suggests ways of working on and informing writing practices. The social relations of performance pertain to times and spaces which are temporary and processual, to activities which imagine other times, spaces and people, and seek to realise them for a specific time in a specific space for a specific group of people. The social relations realised in this process of contingent realities are able to inform writing, that is, to produce writing which connects theatre with other discourses, and which connects words with bodies in time and space.
It is argued that theatre and performance's process of imagination and realisation and its engagement with spatial, temporal, physical and material relations provides a valuable site for critically examining the ways in which Australia privileges and remembers specific configurations of space, time, bodies and matter, while marginalising others, in producing official representations of the Australian nation. Such representations, reflected ingovernmental programmes such as those concerning citizenship and national security, have a bearing on how Australians view their national past, present and future, and how they perceive their social connections with each other.
Just as specific performances are made subject to the textual and discursive categories of literature and social theory, official enactments of the Australian nation are able to 'contain' Australians who spatially, temporally and physically transgress national boundaries. As a material practice, performance is able to engage with official enactments of the nation in order to 're-open' the spaces, times and encounters concealed within these sites.

This thesis seeks to understand how social capital develops within voluntary sports clubs. It adopts a micro-perspective to examine how social capital, defined as ‘the ability to secure resources by virtue of membership in social networks or larger social structures’ (Portes and Landolt, 2000: 532), emerges from social interaction within voluntary sports club contexts. The thesis is informed by a critical realist perspective that focuses on the underlying social mechanisms involved and how they operate differently for different groups and individuals in different circumstances. The empirical analysis, which represents the main contribution of the thesis, is based on three case studies of voluntary sports clubs in the UK. These case studies were carried out over 18 months and involved a number of in-depth interviews with members and organisers and periods of observation at each of the clubs. The analysis shows that members accessed a range of resources through the social ties they formed at the clubs. Interestingly, the analysis demonstrates that, as well as forming strong and weak ties, as standard network models would predict, members formed ‘compartmentally intimate’ ties at the clubs: strong ties that were domain-specific. The analysis reveals that several core mechanisms – reciprocity exchanges, enforceable trust, value introjection and bounded solidarity – operated at the clubs to develop social capital for members, but that these mechanisms were influenced in multiple ways by various elements of context. In particular, the analysis focuses on the socio-organisational context of the clubs and identifies several key elements, including the nature of the focal activity, the voluntariness of participation, the co-operative nature of engagement, the relative absence of hierarchy and the diversity of membership. Overall, the thesis provides support for an organisationally embedded view of social capital development and offers a rare example of critical realist research on social capital.

This thesis examines the development by local authorities of alternative models of ownership of public housing, focusing on large scale voluntary transfers (LSVTs) to specially created housing associations. I attempt to place stock transfer within the wider policy context of changes in the State’s role in the provision of services; and changes in organisational development, reflected in transfer structures; and the wider shift in responsibility for the delivery of public services to the private sector. I examine theories that explain the development of the State’s role in housing and discuss their relationship to LSVT. As discussions involving ‘the State’ must recognise the different interests and motivations of Central and local government, I examine action taken to promote and achieve stock transfer by local authorities and central government. The research method adopted to achieve my aims includes both analysis of existing data and the collection of new data through a sample survey of twenty LSVT associations. I approached the research in five stages: background research – including reviewing literature and examining the legal and regulatory framework for LSVT; identifying the characteristics of early LSVT associations; selecting a sample of twenty selected associations for in-depth survey; conducting in-depth interviews with the twenty associations; analysing and tabulating the data collected and drawing conclusions. The survey focuses on staff perceptions of the reasons for transfer and why politicians and tenants accepted the change; organisational and management change brought about by the new landlords; and the changes brought about by private sector involvement, including new personnel brought in to help run the organisations and the influence of private funders. I attempt to draw key conclusions about LSVT associations from the evidence presented. Finally, I examine how early LSVTs paved the way for a wider stock transfer programme, including transfers by inner-urban authorities and large city councils. I argue that while LSVT may have been developed in response to the financial pressure upon local authorities from the early 1980s onwards, the process has created a new style of business orientated social landlord.

This thesis studies five mainstream British weekly magazines: 'Time and Tide', the 'Nation and Athenaeum', the 'Spectator', the 'Listener', and the 'New Statesman'. It explores how these magazines reviewed, discussed and analysed modernist literature over an eighteen-year span, 1920-37. Over this period, and in these magazines, the concept of modernism developed. Drawing on work by philosopher Ian Hacking, this research traces how the idea of modernism emerged into the public realm. It focuses largely on the book reviews printed in these magazines, texts that played an important and underappreciated role in negotiations between modernist texts and the audience of these magazines. Chapter 1, on 'Time and Tide', covers a period from the magazine's inception in 1920 to 1926, and draws particularly on Catherine Clay's work on this magazine. It discusses the genre of 'weekly review' that this new magazine attempted to join, and the cultural place of modernism in the early 1920s. Chapter 2, on the 'Nation and Athenaeum', covers Leonard Woolf's literary editorship (1923-30), under the ownership of J. M. Keynes, and makes use of Keynes's archive at King's College, Cambridge, and Woolf's at the University of Sussex. Chapter 3, on the 'Spectator', covers Evelyn Wrench's editorship (1925-32), and explores the relationship between this magazine, ideologies of conservatism, and modernism. Chapter 4, on the 'Listener', focuses on the magazine's publication of new poetry, including an extraordinary 1933 supplement that printed W. H. Auden's 'The Witnesses'. This work revolves around Janet Adam Smith, literary editor in these years, and draws on Smith's archive at the National Library of Scotland as well as the BBC archives at Caversham. Chapter 5, on the 'New Statesman' in the 1930s under new editor Kingsley Martin, explores a period when modernism was more widely recognized, and pays particular attention to a short text by James Joyce printed in 1932, 'From a Banned Writer to a Banned Singer'.

The cationic host defence peptide LL-37 is an immunomodulatory agent that plays an important role in epithelial innate immunity. Previously, concentrations of LL-37 thought to represent levels present during inflammation have been shown to elicit the production of cytokines and chemokines by epithelial cells. To investigate the potential of lower concentrations of LL-37 to alter epithelial cell responses, normal primary keratinocytes and bronchial epithelial cells were treated with pro-inflammatory stimuli in the presence or absence of 1 – 3 μg/ml LL-37. Low, physiologically relevant concentrations of LL-37 synergistically increased IL-8 production by both proliferating and differentiated keratinocytes in response to IL-1β and the TLR5 agonist flagellin, and synergistically increased IL-8 production by bronchial epithelial cells in response to IL-1β, flagellin, and the TLR2/1 agonist PAM3CSK4. Treatment of bronchial epithelial cells with LL-37 and the TLR3 agonist poly(I:C) resulted in synergistic increases in IL-8 release and cytotoxicity. The synergistic increase in IL-8 production observed when keratinocytes were co-stimulated with flagellin and LL-37 was suppressed by pretreatment with inhibitors of Src-family kinase signalling and NF-κB translocation. These data suggest that low concentrations of LL-37 may alter epithelial responses to microbes in vivo. Microarray analysis of keratinocyte transcriptional responses after LL-37 treatment suggest that LL-37 may alter the expression of growth factors and a number of genes important to innate immune responses. LL-37 may thus play a more important role than previously suspected in the regulation of epithelial inflammation; an improved understanding of the mechanisms by which LL-37 alters chemokine responses could lead to the development of novel anti-infective and anti-inflammatory therapeutics.

Antimicrobial peptides are key actors in organisms€™ immune systems. They play an important role in phagocytosis, breaking bacteria membranes. They destroy the microbes, keeping them from repairing themselves, and therefore do not promote antimicrobial resistance. LL37 is a peptide produced by the human body. It is a short amino acid chain that is particularly active on the skin and mucous membranes. It has antimicrobial and fungal activity as well as wound healing properties, which makes it a very interesting active substance in wound treatment. However, its fragile and sensitive structure is a challenge to its use. Nowadays, encapsulation in a biocompatible polymer system is a promising technique in drug delivery, and presents a solution to LL37 administration and delivery. LL37 is a hydrophilic active substance, it will be trapped in PLGA (poly (lactic-co-glycolic acid)) by double emulsion and the microspheres will be shaped and stabilized by solvent evaporation. The capsules will be characterized by Dynamic Light Scattering (DLS) and Scanning Electron Microscopy. Their main features, drug loading, encapsulation efficiency and release profile, are determined using the Bradford assay. Since the peptide is expensive and delicate, it is important to optimize its encapsulation. For that reason, we will adapt the process to have the best drug loading as possible using water in oil in oil emulsions. For an external use, the capsules would be used over a few days, so having a fast release is very relevant. The larger the specific surface area, the faster the diffusion. For that reason, we will also study the impact of porosity on the release profile. As a result, different types of capsules will be synthesized, with higher porosity and by two processes: aqueous double emulsion and oil double emulsion. Their characteristic features and impact on bacterial pathogens will be determined and compared in order to determine their optimal synthesis process and formulation in given conditions of use.

The aim with this Masters thesis is to examine the professional knowledge of the librarian. The method used is text analysis of a state public investigation, SOU 1969:37, that concerns librarian education. The theoretical starting point is a theory of knowledge thats descended from Aristotle. Three different forms of knowledge are central concepts in the theory: the theoretical, the practical and the ethical. The main issue in this thesis is: In what ways did the investigators look at the librarians knowledge? Was it in a changeable period of time? In that case, what reasons could there be? The conclusion shows that educational political changes were affecting the whole educational sphere at the late 1960:s, and in the beginning of the 1970:s, including library education. A porfiria aguda intermitente é uma doença autossômica dominante, decorrente de um distúrbio na via biossintética do heme, causado pela redução dos níveis da enzima uroporfirinogênio-I-sintetase. As manifestações clínicas envolvem o sistema nervoso periférico e o central. O diagnóstico baseia-se na excreção urinária elevada dos precursores das porfirinas ácido d-aminolevulínico e porfobilinogênio. O objetivo deste estudo foi analisar o quadro clínico apresentado por pacientes do Hospital das Clínicas de São Paulo com porfiria aguda intermitente, atendidos no período de janeiro de 1979 a dezembro de 1999. Foram avaliados 37 pacientes, com idades entre 6 e 48 anos, na proporção de 2,7 mulheres:1 homem. A faixa etária na qual ocorreu o maior número de crises foi a terceira década. Os pacientes apresentaram 63 crises, sendo que 13 deles também tiveram manifestações de caráter crônico. As manifestações clínicas mais freqüentes foram: dor abdominal, alteração da cor da urina, mudança no ritmo intestinal, déficit motor ou sensitivo-motor, vômitos, alteração do nível de consciência ou confusão mental, crises convulsivas, quadros disautonômicos cardio-vasculares e distúrbios psiquiátricos. As crises foram classificadas em leves, moderadas e graves, segundo critérios previamente estabelecidos. Todas as manifestações crônicas foram caracterizadas como leves. A neuropatia periférica motora ou sensitivo-motora nunca foi a manifestação inicial da crise de porfiria aguda intermitente. Houve correlação entre o tipo e o número de fatores precipitantes e a manifestação da neuropatia periférica motora ou sensitivo-motora, verificando-se que as crises nas quais ela está ausente foram em geral desencadeadas por apenas um fator, mais comumente de origem endócrina ou metabólica endógena, como período menstrual e jejum, enquanto que nas crises com neuropatia periférica houve a participação de vários fatores concomitantemente, sendo estes principalmente de origem exógena, como medicamentos. Os tratamentos mais utilizados nos surtos foram a administração de glicose, aumento da ingestão de carboidratos e o uso de fenotiazínicos.Acute intermittent porphyria is an autosomal dominant disease, caused by a disturbance in the heme biosynthetic pathway, secondary to the reduction on the levels of uroporphyrinogen-I-synthetase enzyme. Clinical manifestations involve central and peripheral nervous system. The diagnosis is based on the elevated urinary excretion of porphyrins precursors d-aminolevulinic acid and porphobilinogen. The aim of this study was to analyze the clinical manifestations of acute intermittent porphyria in patients of the Hospital das Clínicas of São Paulo, seen between January 1979 and December 1999. 37 patients were studied, from 6 to 48 years old, with a rate of 2,7 women: 1 man. The age in which most of the crisis occurred was the third decade. The patients presented 63 crisis, and 13 of them presented also with chronic manifestations. The commonest clinical presentations were: abdominal pain, change in urine color, change in bowel habits, motor or sensory-motor deficit, vomiting, alteration of consciousness or mental confusion, convulsions, dysautonomic cardiovascular signs and psychiatric disorders. The crisis were classified as mild, moderate and severe, following criteria previously established. All chronic manifestations were characterized as mild. The peripheral motor or sensory-motor neuropathy was never the initial manifestation. Correlation was found between the kind and the number of precipitating factors, and the absence of peripheral neuropathy was in general related to just one factor, more commonly of endogenous endocrine or metabolic origin, like menstrual period and starvation, while in the crisis with peripheral neuropathy multiple factors were involved at the same time, these being of exogenous origin, like drugs. The most commonly used treatments were glucose administration, elevation of carbohydrate intake, and phenothiazines use.

This dissertation is an effort to contribute to the knowledge base concerning reading instruction for adolescent students who speak African American Vernacular English (AAVE), as well as their acquisition of Standard English (SE), by focusing on language structure during a modified approach to guided reading (MGR). Emphasis was placed on teaching the eight inflectional morphemes in SE, as well as other literacy strategies to AAVE speaking students. This intervention hypothesized that the teaching and learning of inflectional morphemes, in particular, would increase SE literacy acquisition. The intervention utilized formative experiment methodology and a quasi-experimental time series design. Data sources and collection took several forms: a) KTEA II - reading (letter and word recognition and reading comprehension) and written language (written expression) (Kaufman & Kaufman, 2004); b) field reflections; c) student surveys; and d) video-taped MGR lessons. Informal assessments such as the Ekwall/Shanker Reading Inventory, 4th Edition (Shanker & Ekwall, 2000) were used to guide instruction for MGR lessons. The findings supported the hypothesis that instruction of inflectional morphemes in SE has a positive impact on reading, writing, and overall acquisition of SE among AAVE speaking students.

Due to China's important status on the global stage, its language and culture have drawn a great deal of attention in academia. Meanwhile, the United States remains a major power, and English continues to be the most widely spoken language in today's world. Exploring intercultural communication among people who speak Chinese and English continues to be an important research area. This study, with its primarily linguistic concern, was designed to focus on a frequently employed figure of speech, metaphor. Based on a comprehensive review of literature on intercultural communication and cognitive linguistics, this research focused on metaphor's cognitive value in order to explore its universal validity. A sample database was generated utilizing metaphorical expressions in classical poetry from Chinese, British, and American literature. An in-depth content analysis was conducted using grounded theory methodology to investigate the common place understanding between Chinese and English cultures. Similarities were achieved among existing patterns of metaphorical expressions from relevant poems. Three primary types of metaphors were found. The first metaphor included abstract concepts projected to concrete concepts. The second metaphor was objects projected to human beings. The third metaphor was objects projected to objects. Two main implications were found based on this research. The primary implication for intercultural communication was related to common ground understanding, adaptation of Chinese immigrants in the United States, and improved international relations. Metaphor's universally cognitive validity constitutes the secondary implication of this study, which contributes to the development of cognitive linguistic theory.

Karst regions are composed of soluble rock, often limestone, which leads to the formation of fissures, sinkholes and water flow conduits such as caves. Pollutants in karst waters tend to be quickly directed and concentrated into these subsurface conduits. As a result of this and other factors, water resources are especially sensitive to contamination and pollution in karst areas. Pollutant concentrations going into fluvial systems travelling through the subsurface in karst areas are often very similar to the concentrations arriving at outlets such as springs. Areas connected by karst conduit flows must be distinctly determined and special attention should be given to water quality impacts from land-use practices near conduit inputs. The climate which affects a certain karst area can also have different impacts on water resources considerations. For example, in the temperate climate of Kentucky precipitation is essentially even in distribution throughout the year. In contrast, southwest China is affected by a monsoon climate with high precipitation in the spring to summer and drier conditions in other seasons. In the wet season large storm pulses can effectively transport contaminants to water sources resulting in loads that can be unhealthy for frequent human consumption in drinking water. The dry seasons can be particularly severe in karst areas as water quickly drains to the subsurface, making water access a major hardship. The research for this study focused on the seasonal influences that the climate of southwest China poses for water quality, including differences in pesticide concentrations between agricultural and residential areas hydrologically linked by karst conduits. In late 2007 the fluvial connections in a simple karst system near Chongqing were confirmed using dye tracing techniques. Once these connections were established and the flow of the subterranean stream was assessed, the transport of agricultural runoff in the system was studied. Data loggers were used to record continuous data of the water conditions, including nitrate concentrations. The pesticides in the agricultural runoff entering and exiting the subterranean stream were quantified using ELISA methods. The concentrations were found to be within safe limits for drinking water. The hypothesis that there is a close relationship between concentrations of the pesticides glyphosate, chlorothalonil, and triazines in the input and the output of the system was supported by the results. When considering the hydrology and water chemistry data of the site, along with the water samples tested for pesticides, non-parametric statistical testing showed the correlations between these factors to be significant with p<0.01. The percent difference between the input and the output concentrations of glyphosate, chlorothalonil, and triazines were 31, 43, and 57% respectively. Taking into account the rapid and direct flows in this karst system, the concentrations of the pesticides found in the output were more similar to the input than would be expected in a surface stream. This suggests that there are fewer natural remediation effects reducing contamination in subsurface karst rivers of southwest China than in surface rivers. Therefore, these systems should be handled with extra attention to possible contamination of water resources. The research was conducted in the spring and summer of 2007-2008 and funded by the U.S. Agency for International Development.

Literature uses dance to reveal underlying messages of fantasy through the themes of the central narrative of female characters. Examining the original texts with respect to their varying adaptations for film and stage, performance literature reveals how directors relate a three-dimensional story to an audience from a two-dimensional world. Hans Christian Andersen’s “The Red Shoes” shows an underlying semiotic code where transitioning from the black and white of reality to the red of fantasy is only accomplished through dancing. Examining the physical elements of these works of literature elucidates the use of dance as a lens that lets the performance become speech.

Purpose: The purpose of this study is to evaluate and compare the extent and potential correlates of sexual risk taking behaviors related to condom use and number of sexual partners among Haitians aged 15 to 49 years old living in the urban and rural areas.
Methods: Data were obtained from the 2005-2006 cross-sectional survey conducted by the Demographic Health survey. Our study population (15143) was analyzed based on the Health Belief Model (HBM) theory using a bivariate and multiple logistic regression analysis with SPSS for windows.
Results: In both rural and urban areas dwellers had an accurate knowledge of the disease. However, a small proportion in both areas used condoms during their last sexual intercourse. Perceiving the disease's severity was more likely associated with condom use in the urban (OR 1.36, p ≤ .01) and in the rural areas (OR 1.45, p ≤.05). Strong associations have been found between some variables of the HBM and condom use but none have been found associated with zero or one sex partner. Findings were similar in both areas
Conclusion: Holistic approach should be considered in the prevention strategy conducted in Haiti to tackle other factors that may contribute in delaying responsible sexual behavior in that country.

Bed load transport measurements were made in a small urban stream in Decatur, GA, from which thresholds for motion were calculated using methodologies from the published literature. These methodologies are discussed in terms of their limitations and assumptions. Mobility frequencies were calculated for single grains of each grain size fraction to illustrate the transition from size selective transport to equal mobility. In general, urban streams behave differently than many gravel rivers in non-urban settings because of differences in the availability and character of sediment sources and altered flow hydrographs. This comparison allows for discussion about the way sediment is transported in urban streams versus typical gravel-bed, armored channels.

Utilizing the Northern Ireland case, this study analyzes variance in willingness to integrate between Protestant and Catholic groups. Analysis of the Northern Ireland Life and Times survey data illustrates the shifting relationship between political trust and ingroup/outgroup frustrations on levels of willingness to integrate since the Good Friday Agreement. Statistical analyses indicate confirmation of ingroup attachment and elite political trust hypotheses, and reduced impact of outgroup benefit perceptions on willingness to integrate since the Good Friday Agreement.

There are myriad definitions of the terms race, ethnicity and culture in social sciences literature. Often these terms are used interchangeably with no conceptual rationale. This study aims to contribute to our greater understanding of the similarities and differences between the conceptualization and use of race and culture as they are experienced by African American adolescents. Multiple regression analyses and factor analysis were conducted for 223 African American high school aged students who completed a survey about racial and ethnic identity and a variety of positive youth development outcomes. Results showed preliminary support for race and culture being distinguishable yet intricately related. Results are also presented that compares the relationship of either a racial or cultural orientation on various youth development outcomes.

This dissertation follows a three-essay format. Each essay evaluates a different fiscal institution from a public administration perspective. In the first essay I examine whether state-funded property tax exemptions are effective in reducing the property taxes. This class of exemption is characterized by a grant from state to local governments that is intended to replace property tax revenue and lower property tax payments. Two separate theories of local fiscal behavior predict that that price effects and fiscal illusion will reduce the effectiveness of this type of tax relief. I test these predictions using panel regression analysis on county-level data from Georgia. I find that only two thirds of the revenue allocated to this program is actually used for tax relief. In the second essay I test a model of the property tax in which the levy is set to balance the difference between budgeted expenditures and expected receipts from all other revenue sources. This model demonstrates how the property tax can be used to offset unexpected changes to other revenues given a change in personal income. This model is contrasted with an alternative model in which expenditures are budgeted after expected total revenues have been determined. I will estimate both models for local governments in Georgia and test which more accurately describes local fiscal performance. I will also use both to predict changes to the property tax over a period of time and measure which model generated the more accurate forecast. Unlike the first two papers, which are quantitative analyses of fiscal data, this chapter is a case study of the contract city model of governance as implemented in the newly incorporated city of Sandy Springs, Georgia. I investigate whether the scope of outsourcing in contract cities creates additional challenges for city officials that manage contractor performance. I evaluate the incentive structures in the contract agreements that influence the principal-agent relationship using a textual analysis research method. I find that certain combinations of municipal functions in a single public-private partnership creates the potential for negative synergies to arise which would increase the difficulty of monitoring and managing the private partner.

This thesis focuses on the creation and disposal of food waste in the United States. Food waste is a specific yet highly critical issue that implicates the large, incongruous systems of both food production and waste disposal. Waste is created throughout the food supply chain, with producers as well as consumers guilty of throwing away good food. Rather than repurpose food as compost or donate it to those in need, wasted food, although completely biodegradable and often edible, is mixed in with the rest of our garbage and disposed of in a landfill. By evaluating the systems of waste disposal and food production, I illustrate the ways in which both of these industries encourage the creation of food waste and conceal its harmful effects. I argue that it is necessary to prioritize source reduction of wasted food, rather than rely upon infrastructure that keeps waste “out of sight, out of mind.” Despite the factors that shelter it from our critical consideration, it has become necessary to prioritize food waste as a legitimate environmental, social, and economic concern.

Part One. X-ray crystallography has benefited from the synthetic introduction of selenium to different positions within nucleic acids by easing the solving of the phase problem. Interestingly, its addition to the 2' position of the ribose ring also significantly enhances crystal formation. This phenomenon was investigated to describe the effect of selenium-based and other 2' modifications to the ribose ring of nucleosides in solution, as well as the incorporation of the selenium-modified nucleotides into a helical structure. This work correlates the difference in conformation propensity between the selenium containing nucleosides and oligomers towards a rationale behind the enhanced crystal forming behavior. Part Two. Recombinant protein production is a critical tool in laboratories and industries, and inducing extracellular transport of these products to the culture medium shows potential for improving cases where the yields are not sufficient in quality or quantity. This review incorporates current practices and systems with future perspectives.

Bartonella, a group of Gram negative facultative intracellular bacteria, are known to cause diseases, such as Cat Scratch Disease, Trench Fever and Carrion’s Disease, that involve angiogenesis during the infective cycle. B. bacilliformis, the etiological agent of Carrion’s Disease, causes a bi-phasic infection resulting in the formation of blood-filled angiogenic proliferative cutaneous nodules called verruga peruana. The work presented here was undertaken to characterize the mechanism by which these nodules are produced. Previous work in our laboratory suggested that the Bartonella henselae genome contains a homologue to the virB operon, a set of genes coding for a Type IV Secretion System (TFSS) that has been implicated in the pathogenesis of other α-2-proteobacteria. We identified virB operons in two additional Bartonella pathogens, B. quintana and B. clarridgeiae. No corresponding operon sequences were detected in B. bacilliformis DNA, however. This finding suggests that virB gene products are not required for verruga peruana formation. To continue our search for factors involved in B. bacilliformis-induced angiogenesis, we conducted a microarray analysis of differential gene expression in infected and uninfected endothelial cells. The results suggest similarities between later stage (36 hours) B. bacilliformis infection and that of HHV-8, the causative agent of Kaposi’s Sarcoma, particularly in relation to the host immune response. Finally, our research focused on the secreted factors that B. bacilliformis produces during its host infective cycle. Our data suggest that the B. bacilliformis homologue to the molecular chaperone GroEL not only induces angiogenesis in endothelial cells, but also protects endothelial cell tubule from the degradation seen when these cells are in the presence of live B. bacilliformis. In summary, the induction of verruga peruana nodules via B. bacilliformis may be the result of multiple factors over the course of persistent infection. Early infection may cause vascular damage, which induces VEGF and hypoxia factors. As infection persists, bacterial secretion of a unique GroEL may result in continued angiogenesis and the ensuing activation of immune cells, producing a localized environment of continual incomplete angiogenesis in areas of cutaneous infection.

The enzymatic oxidation of choline to glycine betaine is of interest because organisms accumulate glycine betaine intracellularly in response to stress conditions, as such it is of potential interest for the genetic engineering of crops that do not naturally possess efficient pathways for the synthesis of glycine betaine, and for the potential development of drugs that target the glycine betaine biosynthetic pathway in human pathogens. To date, one of the best characterized enzymes belonging to this pathway is the flavin-dependent choline oxidase from Arthrobacter globiformis. In this enzyme, choline oxidation proceeds through two reductive half-reactions and two oxidative half-reactions. In each of the reductive half-reactions the FAD cofactor is reduced to the anionic hydroquinone form (2 e- reduced) which is followed by an oxidative half-reaction where the reduced FAD cofactor is reoxidized by molecular oxygen with formation and release of hydrogen peroxide. In this dissertation the roles of selected residues, namely histidine at position 310, valine at position 464 and serine at position 101, that do not directly participate in catalysis in the reaction catalyzed by choline oxidase have been elucidated. The effects on the overall reaction kinetics of these residues in the protein matrix were investigated by a combination of steady state kinetics, rapid kinetics, pH, mutagenesis, substrate deuterium and solvent isotope effects, viscosity effects as well as X-ray crystallography. A comparison of the kinetic data obtained for the variant enzymes to previous data obtained for wild-type choline oxidase are consistent with the valine residue at position 464 being important for the oxidative half-reaction as well as the positioning of the catalytic groups in the active site of the enzyme. The kinetic data obtained for the serine at position 101 shows that serine 101 is important for both the reductive and oxidative half-reactions. Finally, the kinetic data for histidine at position 310 suggest that this residue is essential for both the reductive and oxidative half-reactions.

The goal of this project was to determine if there was a relationship between a company’s perceived reputation for social responsibility and attitudes and behaviors that would favorably impact the company. Specifically, the project uses a case study of UPS and its support of Zoo Atlanta to further test these relationships in a true-to-life scenario. The findings reveal implications for corporate communication efforts in two ways: first, by serving as a framework to evaluate future corporate giving programs and to better understand company reputation; and second, by understanding the importance of strategically positioning one’s company as a good corporate citizen.

The QoS Steiner Tree Problem asks for the most cost efficient way to multicast multimedia to a heterogeneous collection of users with different data consumption rates. We assume that the cost of using a link is not constant but rather depends on the maximum bandwidth routed through the link. Formally, given a graph with costs on the edges, a source node and a set of terminal nodes, each one with a bandwidth requirement, the goal is to find a Steiner tree containing the source, and the cheapest assignment of bandwidth to each of its edges so that each source-to-terminal path in the tree has bandwidth at least as large as the bandwidth required by the terminal. Our main contributions are: (1) New flow-based integer linear program formulation for the problem; (2) First implementation of 4.311 primal-dual constant factor approximation algorithm; (3) an extensive experimental study of the new heuristics and of several previously proposed algorithms.

The research study examines the influence of an African American religious preacher in organizing his congregation into a socioeconomic community. The research question is as fol-lows: How does the leadership performance influence (a) the social interaction of his congrega-tion, (b) the mobilization of congregates into members of a social group, and (c) the development of his congregation into an economic system? This study examines the Pastor, his role, influence, and management of power. Weber argued that there are three pure types of authority: rational, traditional, and charismatic. For an African American religious preacher, the authority would be considered to be charismatic. However, this study discovered that the Pastor of Washington Missionary Baptist Church, the field site for this ethnography, encompasses characteristics of each from of authority. Such characteristics are necessary in order to successfully manage a nonprofit organization, which requires a decentralization of power and shared leadership.

For furthering these issues, a multi-paper dissertation is adopted and studies distinct aspects of user interaction with IT related with adaptation. Thus, in order to better understand how system users adapt to IT disruptions this study examines (1) how system users who become disrupted by IS that provide them with too much information interact with these systems, (2) the influence of espoused cultural values (Srite et al. 2006) on user coping strategies of adaptation to IS, and (3) middle managers responses to the implementation of disruptive IT in public administration. These dissertation studies together help improve our knowledge on individual adaptive responses to IT disruptions.

Wireless sensor networks (WSNs) have recently attracted a great deal of attention due to their numerous attractive applications in many different fields. Sensors and WSNs possess a number of special characteristics that make them very promising in a wide range of applications, but they also put on them lots of constraints that make issues in sensor network particularly challenging. These issues may include topology control, routing, coverage, security, data management and many others. Among them, coverage problem is one of the most fundamental ones for which a WSN has to watch over the environment such as a forest (area coverage) or set of subjects such as collection of precious renaissance paintings (target of point coverage) in order for the network to be able to collect environment parameters, and maybe further monitor the environment. In this dissertation, we highly focus on the area coverage problem. With no assumption of sensors’ locations (i.e., the sensor network is randomly deployed), we only consider distributed and parallel scheduling methods with the ultimate objective of maximizing network lifetime. Additionally, the proposed solutions (including algorithms, a scheme, and a framework) have to be energy-efficient. Generally, we investigate numerous generalizations and variants of the basic coverage problem. Those problems of interest include k-coverage, composite event detection, partial coverage, and coverage for adjustable sensing range network. Various proposed algorithms. In addition, a scheme and a framework are also suggested to solve those problems. The scheme, which is designed for emergency alarming applications, specifies the guidelines for data and communication patterns that significantly reduce the energy consumption and guarantee very low notification delay. For partial coverage problem, we propose a universal framework (consisting of four strategies) which can take almost any complete-coverage algorithm as an input to generate an algorithm for partial coverage. Among the four strategies, two pairs of strategies are trade-off in terms of network lifetime and coverage uniformity. Extensive simulations are conducted to validate the efficiency of each of our proposed solutions.

This dissertation examines a curricular approach at an institution that claims to maintain a liberal arts focus – that of the canon of Great Books as implemented as a formal curriculum at St. John’s College. My research question is: what enabled the Great Books program at St. John’s College to survive for over seventy years? The significance of this question can be seen by noticing that St. John’s College is the only college in the United States to have exclusively adopted reading the Great Books as its four-year curriculum. Other institutions that have experimented with a Great Books program prior to and since its introduction at St. John’s College have continued their existing programs as well, but many have limited their Great Books efforts to an honors course or general core requirement, if their Great Books effort survives at all. My dissertation is historical starting with the influencing factors leading to this curriculum’s introduction at St. John’s College in 1937. I then outline the implementation and document the changes to the list of Great Books comprising the program as it was updated over the subsequent seventy years as documented in St. John’s College’s academic catalogs from 1937 through 2008. I show that the list of Great Books required to be read by every student over the years has contained a consistent core while making slight adjustments.

Board of Education, the bus boycott, school integration court cases, re-segregation of schools in city and suburban districts, and federal open-housing policies, the volatile transformation appears to shows how, after fifty years, Montgomery has moved from a segregated dual society to a partially integrated society in spite of the massive resistance to integration.

The analysis of longitudinal data has been a popular subject for the recent years. The growth of the Generalized Estimating Equation (GEE) Liang & Zeger, 1986) is one of the most influential recent developments in statistical practice for this practice. GEE methods are attractive both from a theoretical and a practical standpoint. In this paper, we are interested in the influence of different "working" correlation structures for modeling the longitudinal data. Furthermore, we propose a new AIC-like method for the model assessment which generalized AIC from the point of view of the data generating. By comparing the difference of the log-likelihood functions between different correlation models, we define the exact value to create an interval for our model selection. In this thesis, we combine the GEE method and a new generalized AIC Index for the longitudinal data with different correlation structures.

Melanocortins (MCs) play an important role in feeding, metabolism, and energy expenditure. While melanocortin receptor (MCR) mRNA has been found in the mesolimbic dopamine (DA) pathway, the ability of melanocortins to regulate feeding and other behaviors through actions on the mesolimbic DA system have not been examined. Short-hairpin RNAs (shRNAs) were created targeting MC3R and MC4R and were tested via in vitro studies for their ability to knockdown their target receptor. A total of three shRNAs were created targeting each receptor, and each shRNA caused successful knockdown. These shRNAs are tools that can be used for future in vivo studies to examine the various behavioral effects of melanocortins on the mesolimbic DA pathway.

Background: Determinants of vitamin D status are of interest when studying the epidemiology of disease in population groups because vitamin D is now recognized to decrease the risk of diseases such as osteoporosis, cancer, and cardiovascular disease. Understanding modifiable determinants of vitamin D status are important for managing vitamin D deficiency at the individual level and for addressing this issue at population level.
Objective: The objective of this study was to evaluate the associations between serum vitamin D status (deficiency and insufficiency) and distinct demographic, dietary, and lifestyle characteristics of adults in the United States using a large, nationally representative sample survey, the National Health and Nutrition Examination Survey (NHANES) 2005-2006.
Methods: The study sample consisted of 2340 adults aged 20-59 who had serum 25(OH)D measured and who had completed various questionnaires concerning dietary intake of vitamin D and other lifestyle factors. Multivariate logistic regression was used to estimate the odds ratio (OR) of vitamin D deficiency, insufficiency, and sufficiency in adults based on distinct demographic, dietary, and lifestyle characteristics. Statistical significance was set at α < 0.05.
Results: The prevalence of vitamin D deficiency was higher in obese adults than in underweight to normal weight adults (50.9% ± 4.57 vs. 29.3% ± 3.57), higher in adults who reported no sunburns than in adults who reported ≥ 3 sunburns (49.9% ± 3.82 vs. 18.0% ± 3.07), and higher in adults who use sun protective measures regularly than in adults who do not (48.4% ± 3.93 vs. 27.0% ± 3.75). The prevalence of vitamin D deficiency increased as dietary intake of vitamin D decreased. Non-Hispanic black adults were significantly more likely to be vitamin D deficient (OR = 45.27, 95% CI = 17.27-118.64) and insufficient (OR = 9.37, 95% CI = 3.43-25.61) than non-Hispanic white adults. Significant positive associations were found between vitamin D deficiency and several characteristics, namely obesity (OR = 7.43, 95% CI = 4.33-12.77), physical inactivity (OR = 1.63, 95% CI = 1.03-2.58) poor dietary vitamin D intake (OR = 2.34, 95% CI = 1.44-3.81), non-supplement use or supplement use with a low amount of vitamin D (OR = 1.75, 95% CI = 1.05-2.89), and activities that decrease exposure to sunlight (from OR = 2.97, 95% CI = 2.14-4.13 to OR = 5.30, 95% CI = 3.17-8.85).
Conclusion: The results of this nationally representative study demonstrate that obesity, physical inactivity, poor dietary intake of vitamin D, and low sunlight exposure increases the risk for vitamin D deficiency in U.S adults. Future studies are needed to investigate whether vitamin D supplementation, sunlight exposure, and vitamin D-fortified foods are efficient in correcting vitamin D deficiency and insufficiency among these groups.

This project compares the career of the early 20th century ballet dancer, Vaslav Nijinsky, to Friedrich Nietzsche’s theory of the tragic arts. In The Birth of Tragedy (1872) and elsewhere, Nietzsche argues that artists play the central role in communal mythmaking and religious renewal; he prescribes the healing work of the “tragic artist” to save modernity from the decadence and nihilism he identifies in scientism, historicism, and Christianity. As a dancer, and especially as a choreographer for the Ballets Russes (1912-1913), Nijinsky staged a kinetic response to modern culture that not only displayed shared concerns with Nietzsche, but also, as I argue, allow him to be interpreted as Nietzsche’s archetypical tragic artist. By juxtaposing the philologist-philosopher and dancer-choreographer as artists, I situate the emergence of Modern Art as a nascent movement still bound to Romanticism even while rebelling against it, and as an attempt to reinterpret art in a mythic (and thoroughly modern) context.

In recent years, film, art, new media, and music video works created by black makers have demonstrated an increasingly “post-black” impulse. The term “post-black” was originally coined in response to innovative practices and works created by a generation of black artists who were shaped by hip-hop culture and Afro-modernist thinking. I use the term as a theoretical tool to discuss what lies beyond the racial character of a work, image, or body. Using a post-black theoretical methodology I examine a range of works by black filmmakers Kathleen Collins Prettyman and Lee Daniels, visual artists Wangechi Mutu and Jean-Michel Basquiat, new media artist Nettrice Gaskins, and music video works of hip-hop artists and performer Erykah Badu.
I discuss how black artists and filmmakers have moved through Darby English’s notion of “black representational space” as a sphere where bodies and works are beholden to specific historical and aesthetic expectations and limitations. I posit that black representational space has been challenged by what I describe as “metaphysical space” where bodies produce a new set of possibilities as procreative, fluid, liberated, and otherworldly forces. These bodies are neither positive nor negative; instead they occupy the in-between spaces between life and death, time and space, digital and analog, interiority and exteriority, vulnerability and empowerment. Post-black visual culture displays the capacities of black bodies as creative forces that shape how we see and experience visual culture.
My methodology employs textual analysis of visual objects that articulate a post-black impulse, paying close attention to how these works compel viewers to see other dimensions of experience. In three chapters I draw from theoretical work in race and visuality, affect theory, phenomenology, and interiority from the likes of Charles Johnson, Frantz Fanon, Elena del Río, Sara Ahmed, Saidiya Hartman, and Elizabeth Alexander. This study aims to create an interdisciplinary analysis that charts new directions for exploring and re-imaging black bodies as subjects and objects of endless knowledge and creative potential.

This educational study on building art education relationships between art educators and local community art agencies was conducted in early February of 2009. Data was collected by means of an art teacher survey, mailed to the homes of metro Atlanta art educators and by conducting face-to-face interviews with professionals working in the education departments of high profile metro Atlanta art agencies. The data analysis provides insight into the goals of local K-12 art educators, and they are compared to the goals of community art agencies. The findings of this study reveal that art teachers and art agencies share many common goals and face similar challenges. And together, through networking and close communication, they may better serve the needs of students K-12 as they become lifetime participants and supports of the visual arts.

The Perkins IV Act was passed in 2006 to promote high school graduation for youth while introducing skills they need to be prepared for in the labor market. Yearly progress reports measure if CTE programs are successful in reducing dropout rates but fall short for a number of reasons. Using state-level data from the National Association of State Directors of Career Technical Consortium (NASDCTEc) website for the 2009-2010 school year, state-level data from the National Center for Education Statistics (NCES) Common Core of Data (CCD) database for 2009-2010, and dropout data from the U.S. Department of Education for the 2010-2011 school year to establish causality, my research seeks to address the extent to which Perkins IV funding reduces drop out rates at the state-level.

Research on racial border patrolling has demonstrated how people police racial borders in order to maintain socially constructed differences and reinforce divisions between racial groups and their members. Existing literature on border patrolling has primarily focused on white/black couples and multiracial families, with discussions contrasting “white border patrolling” and “black border patrolling,” in terms of differential motivations, intentions, and goals (Dalmage 2000). In my dissertation research, I examined a different type of policing racial categories and the spaces in-between these shifting boundaries. I offer up “multiracial interracial border patrolling” as a means of understanding how borderism impacts the lives of “multiracial” individuals in “interracial” relationships. In taking a look at how both identities and relationships involve racial negotiations, I conducted 60 in-depth, face-to-face qualitative interviews with people who indicated having racially mixed parentage or heritage. Respondents shared their experiences of publicly and privately managing their sometimes shifting preferred racial identities; often racially ambiguous appearance; and situationally in/visible “interracial” relationships in an era of colorblind racism. This management included encounters with border patrolling from strangers, significant others, and self. Not only did border patrolling originate from these three sources, but also manifested itself in a variety of forms, including benevolent (positive, supportive); beneficiary (socially and sometimes economically or materially beneficial); protective, and malevolent (negative, malicious, conflictive). Throughout, I discussed the border patrolling variations that “multiracial” individuals in “interracial” relationships face. I also worked to show how people’s participation in border patrolling encouraged their production of colorblind discourses as a strategy for masking their racial attitudes and ideologies about “multiracial” individuals in “interracial” relationships.
