Lymphocyte antigen receptor engagement triggers activation of various signaling cascades, initiated by sequential activation of the Src and Syk families of protein tyrosine kinases, and leading to gene transcription. Although they lack both enzymatic and transcriptional activity, adaptor proteins are crucial for this process. The FA pathway is responsible for resolving DNA interstrand crosslinks (ICLs). 3BP2 was originally identified through its interaction with the ABL kinase. Fusion of ABL with the BCR gene forms the BCR-ABL onco-protein, which is causative in Chronic Myeloid Leukemia (CML) and acute lymphoid leukemia (ALL). Due to the ability of 3BP2 to regulate ABL activity in osteoblasts, we hypothesize that 3BP2 modulates BCR-ABL signalling. Two classes of hth cDNA clones were isolated from a Drosophila imaginal discs cDNA library and defined two isoforms that differ by the presence or absence of a 45 bp stretch. The long isoform (L) contains a 48 bp exon 8 and the short isoform (S) contains only the first three bps (ATG) of exon 8.
