Abstract:
The 2nd Symposium of the Canadian Society for Virology (CSV2018) was held in June 2018 in Halifax, Nova Scotia, Canada, as a featured event marking the 200th anniversary of Dalhousie University. CSV2018 attracted 175 attendees from across Canada and around the world, more than double the number that attended the first CSV symposium two years earlier. CSV2018 provided a forum to discuss a wide range of topics in virology including human, veterinary, plant, and microbial pathogens. Invited keynote speakers included David Kelvin (Dalhousie University and Shantou University Medical College) who provided a historical perspective on influenza on the 100th anniversary of the 1918 pandemic; Sylvain Moineau (Université Laval) who described CRISPR-Cas systems and anti-CRISPR proteins in warfare between bacteriophages and their host microbes; and Kate O’Brien (then from Johns Hopkins University, now relocated to the World Health Organization where she is Director of Immunization, Vaccines and Biologicals), who discussed the underlying viral etiology for pneumonia in the developing world, and the evidence for respiratory syncytial virus (RSV) as a primary cause. Reflecting a strong commitment of Canadian virologists to science communication, CSV2018 featured the launch of Halifax’s first annual Soapbox Science event to enable public engagement with female scientists, and the live-taping of the 499th episode of the This Week in Virology (TWIV) podcast, hosted by Vincent Racaniello (Columbia University) and science writer Alan Dove. TWIV featured interviews of CSV co-founders Nathalie Grandvaux (Université de Montréal) and Craig McCormick (Dalhousie University), who discussed the origins and objectives of the new society; Ryan Noyce (University of Alberta), who discussed technical and ethical considerations of synthetic virology; and Kate O’Brien, who discussed vaccines and global health. Finally, because CSV seeks to provide a better future for the next generation of Canadian virologists, the symposium featured a large number of oral and poster presentations from trainees and closed with the awarding of presentation prizes to trainees, followed by a tour of the Halifax Citadel National Historic Site and an evening of entertainment at the historic Alexander Keith’s Brewery.

Full text
Abstract:
Congenital cataract is the most common cause of the visual disability and blindness in childhood. This study aimed to identify gene mutations responsible for autosomal dominant congenital cataract (ADCC) in a Chinese family using next-generation sequencing technology. This family included eight unaffected and five affected individuals. After complete ophthalmic examinations, the blood samples of the proband and two available family members were collected. Then the whole exome sequencing was performed on the proband and Sanger sequencing was applied to validate the causal mutation in the two family members and control samples. After the whole exome sequencing data were filtered through a series of existing variation databases, a heterozygous mutation c.499T<G (p.E167X) in CRYBB2 gene was found. And the results showed that the mutation cosegregated with the disease phenotype in the family and was absolutely absent in 1000 ethnicity-matched control samples. Thus, the heterozygous mutation c.499T<G (p.E167X) in CRYBB2 was the causal mutation responsible for this ADCC family. In conclusion, our findings revealed a novel stopgain mutation c.499T<G (p.E167X) in the exon 6 of CRYBB2 which expanded the mutation spectrum of CRYBB2 in Chinese congenital cataract population and illustrated the important role of CRYBB2 in the genetics research of congenital cataract.

Full text
Abstract:
The purpose of the study was to study the effect of chronic toxoplasmosis on changes in the levels of pre- and post-implantation mortality in the experiment. Materials and methods. In the experiment there were 90 female Wistar rats with a body weight of 180-200 g. For the development of pregnancy, females of the control and experimental groups were coupled with males for 3 days. After the onset of pregnancy, females of intact controls were orally injected with 2 ml of 0.2 % starch gel. The females of the experimental groups were infected with an invasive culture of Toxoplasma gondii at a dose of 25 tachyzoites per 1 g of body weight (5,000 tachyzoites per rat) and 50 tachyzoites per 1 g of body weight (10,000 tachyzoites per rat). On the 35th day after infection, females of the experimental groups were coupled with males for 3 days before infection. The effect of Toxoplasma on changes in the levels of pre- and post-implantation death was assessed after killing female rats on the 42nd, 49th and 56th (7th, 14th and 21st days after pregnancy) days after infection. Results and discussion. In animals infected at a dose of 25 tachyzoites per 1 g of body weight, there is a decrease in the number of implantation sites in the uterus, the total number of embryos and the number of living embryos at all stages of the development of Toxoplasma by 1.8-2.1 times compared with the control. In females infected at a dose of 50 tachyzoites per 1 g of body weight, a decrease in the number of implantation sites in the uterus and the total number of embryos at all stages of the development of the parasite was recorded by 2.2-2.5 times compared with the control values. There is a decrease in the number of living embryos by the 42nd day after infection by 4.3 times, by the 49th day – by 3.8 times and by the 56th day – by 5.1 times compared to the control. When compared with the results obtained from females with a lower dose of infection, a decrease in this indicator was revealed on the 42nd day after the development of Toxoplasma by 2.1 times, on the 49th day – by 1.7 times and on the 56th day – by 2.5 times. An increase in the number of dead embryos by 0.2-0.4 times in comparison with intact indicators and the results of experimental animals infected at a dose of 5,000 tachyzoites per rat were recorded. There was an increase in the level of resorptions on the 49th and 56th days by 1.4 and 1.6 times in comparison with the control and animals infected at a dose of 25 tachyzoites per 1 g of body weight. Conclusion. Experimental chronic toxoplasmosis causes an increase in pre-implantation and post-implantation embryo death. The recorded effect of Toxoplasma invasion depends on the dose of infection and the duration of the disease

Abstract:
The 49th Parallel divides the western Prairies between Canada and the United States, crossing the central part of the Williston basin and marking the International boundary that separates Saskatchewan from the States of Montana and North Dakota. Discoveries of oil in this area during the 1950s triggered widespread geological activity, and revealed significant differences of stratigraphical understanding on each side of the border. Problems seemed to arise from contrasts between ‘American' and ‘English' interpretations of stratigraphical method, particularly for the oil-producing zones of the Mississippian. This study analyzes the differing points of view, and presents historical reasons for them. Difficulties with stratigraphical method and nomenclature in the 1950s were quite real, becoming the subject in 1959 of a special AAPG-SEPM conference at Dallas. Had the delegates attending that meeting (including the present author) possessed a little more history, they would have known that an ‘American' or mineral-focused view of stratigraphy had originated in a German hard-rock mining terrane, principally through the teaching of Abraham Werner. From there, during the first years of the 19th century, it traveled via Scotland to the State of New York, where, from Amos Eaton's Rensselaer School at Troy, it spread to most of the newly-formed State geological surveys. Some years later, on the other hand, an ‘English' or ‘stratum-focused' view of stratified formations migrated across the Atlantic from the pastoral landscapes and gently inclined rock-formations of southern England, where mapping had tended to discount their mineral content in favor of their observable order and continuity.
