Tight, yet dynamic, control of translation is critical in order for the cell to respond appropriately to intracellular and extracellular cues. This translational control is achieved by modulating the rate of translation in an mRNA-specific or global manner, often by targeting the protein factors that constitute the translational machinery itself. The function of one such translation initiation factor in mammals, eIF4E Homologous Protein (4EHP), has remained elusive. This is despite evidence from 4EHP's homolog in Drosophila (d4EHP) showing it to act as a translational repressor during embryoni

In general, translation Initiation is considered the rate-limiting step of protein synthesis for most mRNAs. Therefore, great interest and effort has been directed at defining protein factors involved in the regulation of initiation. The role of one particular initiation factor, eIF4H, has yet to be conclusively defined. Therefore, we utilized a protein fragment complementation assay (PCA) method to detect possible interacting partners of eIF4H. We identified a novel interaction between eIF4H and 4E-BP1. The Programme for International Student Assessment, or PISA, administered by the Organisation for Economic Cooperation and Development, surveyed fifteen-year-olds for the first time in 2000. PISA focuses on mathematics, science, and reading literacy, and intends to undertake a new study every three years. The administration in 2003 added a section that measured problem-solving skills. The results from all three surveys thus far have placed Finland as the highest achieving country in PISA. Finland’s top performances in PISA astonished the educational world. Abstract:
Meningococcal disease, caused by the bacterium Neisseria meningitidis, is an important cause of morbidity and mortality in young children and adolescents worldwide. There are 12 serogroups with most disease due to meningococci expressing one of five capsular polysaccharide antigens corresponding to serogroups A, B, C, Y and W135. In Europe, the majority of disease-causing strains are of serogroups B and C. No comprehensive vaccine is available against the bacterium due to the difficulty in producing serogroup B vaccines. A number of countries, e.g. UK and the Republic of Ireland have implement

An analysis of relevant aspects of the history of science shows that theology's loss of credibility in an increasingly science-oriented age can be attributed to unresolved disputes from the past over metaphysical, epistemological and methodological issues. In Chapters 1 and 2, an attempt is made to show that the basic disagreements between science and theology can be traced to the ongoing quest for the principles of knowing shared by all disciplines. In Chapters 3 to 6, an attempt is made to identify these principles. Firstly, the processes and principles by which science acquires its knowledg

The optimisation of ligand-macromolecule interactions is fundamental to the design of therapeutic agents. The GRID method is a procedure for determining energetically favourable ligand binding sites on molecules of known structure using an empirical energy potential. In this thesis, it has been extended, tested, and then applied to the design of anti-influenza agents. In the GRID method, the energy of a hydrogen-bond is determined by a function which is dependent on the length of the hydrogen-bond, its orientation at the hydrogen-bond donor and acceptor atoms, and the chemical nature of these

This thesis examines the experiences of people living with dystonia1 and the ways these are represented by people living with dystonia, the Dystonia Society, neurologists, quality of life (QOL) researchers and pharmaceutical companies. Drawing on ethnographic fieldwork conducted within the Dystonia Society and on projects developing a disease-specific QOL measure, and investigating the impact of dystonia on people’s QOL, the thesis explores a series of questions about the conceptual and practical problems inherent in such measures. It asks: • How dystonia is defined and represented and its imp

The formulation of rigid supersymmetric field theories on curved space leads to a number of results on their strongly-interacting regime, crucial from both the mathematical and physical point of view, starting from Witten's topological twist of four-dimensional Yang-Mills theory. At the same time, strongly-coupled field theories may also be studied holographically via the AdS/CFT correspondence. The aim of this thesis is to study aspects of the holographic dictionary for supersymmetric theories on curved manifolds. A key aspect of the correspondence is the renormalization of gravity observable

In the broadest sense, the focus of this thesis concerns the role of entropy in macroscopic systems related to liquid crystals in equilibrium. We be- gin by defining a broad generalisation of the Ball-Majumdar potential, applicable to a wide variety of order parameter models relevant to liquid crystals. First we provide a geometric characterisation of the set of ad- missible order parameters for general systems. We demonstrate the key properties of convexity, regularity, and blow up at the boundary of the singular potential. This is then followed by a discussion of applications to Onsager type

Immunoglobulins play a key role in humoral immunity and have emerged as an important class of biotherapeutics. While the IgG1 backbone tends to be the conventional choice in these recombinant therapies, several therapeutics that focus on other immunoglobulin isotypes are being developed. With the exception of IgA Fc, the conserved glycan of IgD, IgM, IgG and IgE projects across the surface of an immunoglobulin domain with the two opposing glycans packing within the homodimeric Fc structure. This apparent similarity in structure is largely reflected in shared glycosylation status with most clas

The physiological ketosis of starvation makes sound evolutionary sense, as ketone bodies have several thermodynamic advantages over other nutritional substrates, in addition to their actions to conserve protein and glucose stores. Utilising the body’s metabolic responses to ketosis by delivering a novel nutritional source of ketone bodies, the work in this thesis explored the metabolic effects of ketosis on physical performance in humans. First, the pharmacokinetics and dosing requirements for ketone containing drink preparations were characterised in a population of athletes and healthy contr
