Abstract:

Die vorliegende Arbeit beschäftigt sich mit der Herstellung und Charakterisierung von dünnen Schichten im ternären System B-C-N. In diesem System sind einerseits die härtesten heute bekannten Materialien (Diamant, c-BN) enthalten, aber andererseits auch Stoffe mit unterschiedlichsten elektrischen Eigenschaften (Graphit, h-BN). Es besteht daher das Bestreben, verschiedene bekannte Materialien zu kombinieren und so deren Eigenschaften in gewünschter Weise einzustellen. Allerdings wird sowohl experimentell als auch theoretisch des öfteren eine Separation in BN- und C/CNx-Phasen beobachtet. Im Rahmen dieser Arbeit wurden Schichten über einen weiten Stöchiometriebereich mittels reaktiver DC-Magnetronzerstäubung hergestellt und untersucht. Schwerpunkt wurde dabei auf die Analyse der Struktur und des Bindungszustandes sowie die mechanischen Eigenschaften Härte und Elastizitätsmodul gelegt. Es wird diskutiert, in welchen Elementezusammensetzungen wirkliche ternäre Verbindungen entstanden sind und ein Zusammenhang zwischen Struktur und mechanischen Eigenschaften in Form einer empirischen Formel abgeleitet.

Abstract:

SP-B and SP-C are two pulmonary surfactant proteins. They were isolated in the tear film in 2006 (Lukovic et al. 2006). The molecular mechanism governing lipid spreading in the tear film is not well understood and, along with their production in yeast (as an alternative to animal derived material) provide the motives for this research. To investigate the involvement of SP-B and SP-C in lipid spreading, SP-B and SP-C have been produced as recombinant proteins in Pichia pastoris yeast. SP-B and SP-C are cleaved from their proproteins to produce mature active proteins and both forms were produced recombinantly. The proteins were extracted with detergents, polymers and organic solvents to determine the best method of protein isolation and purification. Purified proteins were subsequently tested for surface activity using a Langmuir trough; proSP-C10 and SP-B demonstrated surface tension activity. The mechanism of SP-B and SP-C was examined through generation of ab initio structural models. The behaviour of SP-B and SP-C was observed in membranes based on phospholipid compositions surrounding the SP-B and SP-C in the yeast plasma membrane using mass spectrometry analysis. Computational modelling then demonstrated that SP-B induces lipid curvature, whilst SP-C may help to stabilise lipid vesicles by forming a molecular bridge between the membrane and vesicle. SP-B is thought to help remodel the lung lipid layer on compression through the formation of vesicles that are connected to the membrane by SP-C.

Abstract:

Sarains A-C are alkaloids isolated from the Mediterranean sponge Reniera sarai, which possess moderate antibacterial, insecticidal and antitumour properties. These alkaloids are popular targets for total synthesis, thanks to their rather unusual and challenging structure, which consists of a central diazatricyclic cage attached to two peripheral macrocycles. Arguably the most challenging aspect of their synthesis is that of the tricyclic core, and our approach involves a novel rearrangement of a simpler bicyclic aminal system. This thesis describes various attempts to synthesise cis-fused octahydropyrrolo[2,3-b]pyrrole ring systems that bear an endo acetaldehyde-derived substituent in the 3-position. The research into the synthesis of this ring system began with enantioselective approaches, utilising asymmetric Diels-Alder reactions to establish the stereochemistry required for subsequent transformations into the bicyclic aminal. Progress from these reactions was troublesome. In racemic approaches, the use of a Michael reaction was unsuccessful, but successful installation of the required stereochemistry was achieved by use of a thio-Claisen rearrangement. Using this method, the aforementioned functionalised acetaldehyde was produced in good yield in ten steps from simple starting materials. This provided a branching point for the exploration of four different, but related routes to the sarain core involving the generation of diazoketones, a bromoketone and hydroxy-aldehydes. Substrates to test our proposed rearrangement reaction were synthesised, but transformations into the sarain core proved unfruitful. However, a different tricyclic compound has been produced from a hydroxy-aldehyde approach. In a separate investigation instigated by a result from previous research into synthesis of the sarain core, it was found that treatment of tertiary formamides with a silylated diazoester in the presence of rhodium acetate leads to formation of 3-amino-2- silyloxyacrylates in good yields. The scope and limitations of this novel reaction were investigated.

Abstract:

The dynamics of A + B → C reaction fronts is studied under modulated gravitational acceleration by means of a combination of parabolic flight experiments and numerical simulations. During modulated gravity the front position undergoes periodic modulation with an accelerated front propagation under hyper-gravity together with a slowing down under low gravity. The underlying reason for this is an amplification and a decay, respectively, of the buoyancy-driven double vortex associated with the front propagation under standard gravitational acceleration, as explained by reaction–diffusion–convection simulations of convection around an A + B → C front. Deeper insights into the correlation between grey-value changes in the experimental shadowgraph images and characteristic changes in the concentration profiles are obtained by a numerical simulation of the imaging process.

Abstract:

Els sistemes objecte d'estudi en aquest treball són aliatges metàl·lics d'estructura reticular b.c.c. els quals, a baixes temperatures, experimenten una transició de fase estructural de tipus martensític (bàsicament descrita per una deformació de cisallament). Particularment interessants, entre d'altres, són els aliatges: Cu-Zn, Cu-Zn-Al, Ag-Zn, Cu-AI-Ni, ... , a causa de les propietats termomecàniques que posen de manifest i que els fan de gran utilitat en aplicacions tecnològiques (robòtica, aplicacions mèdiques... ) [1]: efecte de memòria de forma i pseudoelasticitat.

Aquestes propietats tenen el seu origen en la pròpia transformació martensítica (TM) termoelàstica que experimenten aquests aliatges per sota de la temperatura M(s). A més a més, aquests sistemes (aliatges amb efecte de memòria de forma) experimenten una transició ordre-desordre a una temperatura (Tc) normalment molt més alta que M(s). Per sota de Tc presenten estructures d'ordenació B(2), D0(3) o Heusler.

Partint de l'evidència experimental que posa de manifest la forta influència que l'estat d'ordre atòmic de la fase d'alta temperatura [(T)Ms, o fase-beta] té sobre les característiques de la TM [2,3], en aquest treball ens proposem desenvolupar els models pertinents quc permetin de quantificar aquest problema. així com l'associat als efectes d'envelliment observats i que es relacionen amb canvis d'ordre.

En el desenvolupament del treball, primerament es fa una revisió de les característiques generals bàsiques de les TM, així com dels principals models utilitzats en l'estudi de transicions de fase i que són emprats al llarg del treball.

A fi de dur a terme la quantificació esmentada, es fa indispensable una caractcrització prèvia de l'estat d'ordenació atòmic de la fase-beta. Per fer-ho hem desenvolupat un model termodinàmic en l'aproximació de Bragg-Williams per al cas d'aliatges ternaris que presenten estructures d'ordenació DO(3) (a baixes temperatures) i B(2) (a temperatures intermitges). La seva aplicació al sistema Cu-Zn-Al ens permet avaluar les energies d'ordenació tant per a parelles de primers com de segons veïns.

A continuació introduïm un model termodinàmic per a l'estudi concret del nostre problema que ens permet tractar sistemes que experimenten transicions de fase de primer i de segon ordre, amb interacció entre les corresponents formes d'ordenació. Aquesta interacció s'ha introduït a través d'un terme d'acoblament biquadràtic entre els paràmetres d'ordre que descriuen respectivamente les dues transicions de fase. Quan l'apliquem a l'estudi de l'efecte del grau d'ordre atòmic en sistemes que experimenten TM termoelàstiques (aliatges amb efecte de memòria de forma) i transicions ordre-desordre, el model prediu una variació lineal de la constant elàstica C' =(C(11) - C(12))/2 (rellevant del nostre problema) de la fase-beta i el quadrat del paràmetre d'ordre de llarg abast del sistema. Aquest canvi en C' determina els canvis en les característiques termodinàmiques de la TM (T(0), - Delta - H(B->H)). Aquest comportament ha estat justificat comparant els resultats teòrics amb les dades experimentals disponibles per a l'aliatge Cu-Zn-Al.

Posteriorment, hem generalitzat el model anterior per poder tractar sistemes no homogenis i s'ha estudiat, a partir d'un formalisme de Fokker-Planck, com evoluciona, a temperatura constant Tf en fase-beta, la constant elàstica C' durant un procés d'ordenació atòmic que segueix un refredament ràpid des de Ti < Tc. Dins de l'aproximació lineal, el resultat és coherent amb les dades experimentals [4].

Atès el caràcter fenomenològic de l'aproximació utilitzada, que no permet d'extreure una conclusió definitiva en termes físics, pensem que fóra interessant de dur a terme un estudi a nivell microscòpic per assegurar la validesa de la funcionalitat proposada. Per fer-ho s'ha posat a punt un programa de simulació numèrica pel mètode de Monte CarIo per calcular constants elàstiques en sistemes binaris AB que presenten una estructura reticular de tipus b.c.c. Els paràmetres del potencial per a les diferentes interaccions que s'han de considerar AA, BB i AB s'han obtingut mitjançant ajustos a T =OK, de dades conegudes de la constant elàstica C' a T =T(amb) en funció de la composició [5]. No obstant, posteriorment comprovem que aquest comportament es conserva a temperatura finita, quan C' es calcula numèricament pel mètode de Monte Carlo. Finalment, hem calculat a Tf= OK, C' en sistemes parcials o totalment desordenats (després d'un refredament ràpid des de Ti a Tf = OK). Les configuracions corresponents a diferents graus d'ordenació s'han obtingut per la simulació numèrica d'un model d'lsing utilitzant el mètode de Monte Cario. Els resultats que obtenim justifiquen en forma molt satisfactòria la funcionalitat lineal entre C' i el quadrat del paràmetre d'ordre de llarg abast [6] proposada en el marc d'una aproximación de camp mig [7].

Abstract:

The dynamics of A + B → C reaction fronts is studied under modulated gravitational acceleration by means of a combination of parabolic flight experiments and numerical simulations. During modulated gravity the front position undergoes periodic modulation with an accelerated front propagation under hyper-gravity together with a slowing down under low gravity. The underlying reason for this is an amplification and a decay, respectively, of the buoyancy-driven double vortex associated with the front propagation under standard gravitational acceleration, as explained by reaction–diffusion–convection simulations of convection around an A + B → C front. Deeper insights into the correlation between grey-value changes in the experimental shadowgraph images and characteristic changes in the concentration profiles are obtained by a numerical simulation of the imaging process
