AbstractA symplectic connection on a symplectic manifold, unlike the Levi-Civita connection on a Riemannian manifold, is not unique. However, some spaces admit a canonical connection (symmetric symplectic spaces, Kähler manifolds, etc.); besides, some ‘preferred’ symplectic connections can be defined in some situations. These facts motivate a study of the symplectic connections, inducing a parallel Ricci tensor. Abstract:
The content of intramuscular fat (IMF) from preadipocytes is proportional to meat quality in livestock. However, the roles of circRNAs in IMF deposition in sheep are not well known. In this study, we show that circRNA-5335/miR-125a-3p/STAT3 play a crucial adjective role in the proliferation and differentiation of sheep preadipocytes. In this study, we characterized the roles of differentially expressed circRNA-5335/miR-125a-3p/STAT3, which were screened from sheep of different months of age and based on sequencing data. Firstly, the expression profiles of circRNA-5335/miR-125a-3p/STAT3 were identified during the differentiation of preadipocytes in vitro by RT-qPCR and WB. Then, the targeting relationship of the circRNA-5335/miR-125a-3p/STAT3 was verified by dual-luciferase reporter assays. The results of RT-qPCR, CCK8, EdU and Oil Red O staining assay showed that miR-125a-3p suppressed the differentiation and raised the proliferation of preadipocytes by targeting STAT3. As a competing endogenous RNA, the downregulation of circRNA-5335 decreased the expression of STAT3 by increasing miR-125a-3p, which inhibited the differentiation of preadipocytes and promoted proliferation. Our present study demonstrates the functional significance of circRNA-5335/miR-125a-3p/STAT3 in the differentiation of sheep preadipocytes, and provides novel insights into exploring the mechanism of IMF.

Abstract The aim of this paper is to provide a new reading of Plato’s precosmos (Ti. 52d2–53c3). More specifically, I shall argue that the precosmos is populated by bodies deriving from random complexes of properties, and that this is the effect of the Receptacle’s full precosmic participation in the Paradigm. This will turn out to be consistent with a robust notion of ‘precosmic generation’ and will reveal why Plato may have sought to refer to this otherwise puzzling scenario: representing the precosmos in this way allows Plato to effectively justify why the Demiurge is responsible only for the goodness and perfection of the universe, and why it is properly the best possible cause.

Abstract CDK2 is a key Cyclin-Dependent Kinase that is responsible for driving cells through the cell cycle. CDK2 is both hyper-activated in a variety of cancers and is a mechanism of resistance to CDK4/6 inhibitors, making it an attractive therapeutic target. Using PF3600 and PF4091 (tagtociclib), we previously showed that upon CDK2 inhibition, cells exhibit a rapid drop in substrate phosphorylation that rebounds within several hours (‘drop-rebound’). We further showed that CDK4/6 activity compensates for inhibition of CDK2 and sustains the proliferative program by maintaining Rb hyper-phosphorylation and E2F transcription, allowing many cell types to continue cycling in drug. With the advent of numerous new CDK2 inhibitors in development and in clinical trials, we sought to compare five CDK2 inhibitors side-by-side. We first show that all five CDK2 inhibitors elicit the drop-rebound in CDK2 activity. Thus, the drop-rebound response to CDK2 inhibition is not drug-specific, but instead reflects how the cell cycle is wired for robustness. Second, we find that GTAI-664 is ten times more potent than other CDK2 inhibitors. This potency advantage paired with improved selectivity may result in greater target coverage at attainable plasma concentrations in preclinical models. In agreement, several xenograft tumor models appear to respond with better tumor growth inhibition when treated with GTAI-664 relative to PF4091. Finally, we test the combinability of CDK2 inhibitors with CDK4/6 inhibitors in vitro and in vivo. We hypothesize that durable cell-cycle arrest requires forcing Rb kinase activity below a specific threshold where the Rb-E2F positive feedback loop is broken. Our results are timely given the increasing interest in CDK2 inhibitors across a variety of cancer types. Citation Format: Lotte P. Program promotif gaya hidup sehat telah banyak digencarkan pemerintah diantaranya Perilaku Hidup Bersih dan Sehat (PHBS) serta Keluarga Sadar Gizi (Kadarzi). Namun demikian, cakupan penerapan indikatornya seringkali tidak dapat terpenuhi, hanya beberapa atau sebagian keluarga yang menerapkan secara utuh. Dusun Jaranan menjadi salah satu yang belum menerapkan secara utuh. Data terakhir menunjukkan 53,35% untuk PHBS tahun 2018 sedangkan kadarzi sudah 95,44%. Penerapan ini selalu mengalami perubahan naik maupun turun. Oleh karena itu diperlukan penilaian kesehatan dan upaya promotif yang berkesinambungan. Studi ini bertujuan untuk melakukan seluruh tahapan siklus promosi kesehatan di RW 40 Dusun Jaranan. Metodenya penilaian kesehatan menggunakan metode kuantitatif dengan survey. Data yang dihasilkan berupa permasalahan yang prioritas yaitu masih adanya yang merokok di dalam rumah. Oleh karena itu, program yang disepakati bersama masyarakat adalah peningkatan pengetahuan tentang bahaya rook bagi perokok aktif dan pasif dengan metode ceramah dan alat bantu berupa poster, leaflet, dan video. Intervensi ini dapat berjalan sesuai rencana dan memberikan dampak berupa peningkatan pengetahuan sasaran. Upaya ini harus terus ditindaklanjuti oleh puskesmas setempat agar tidak berhenti pada peningkatan pengetahuan saja, namun juga sikap dan perilaku.---The healthy lifestyle promotion program has been intensified by the government including Clean and Healthy Behavior (PHBS) and Nutrition Conscious Families (Kadarzi). However, the scope of the application of the indicators can often not be fulfilled, only a few or some families who apply in full. Jaranan Hamlet is one that has not been implemented in full. The latest data shows 53.35% for PHBS in 2018 while the levels of have been 95.44%. This application always changes up and down. Therefore it is necessary to have an ongoing health assessment and promotive effort. This study aims to conduct all stages of the health promotion cycle in RW 40 of Jaranan Hamlet. The method of health assessment uses quantitative methods with surveys. The data generated in the form of a priority problem that is still the existence of smoking in the house. Therefore, the program agreed with the community is to increase knowledge about the dangers of rooks for active and passive smokers with lecture methods and assistive devices in the form of posters, leaflets, and videos. This intervention can go according to plan and have the effect of increasing target knowledge. This effort must continue to be followed up by local health centres so as not to stop at increasing knowledge, but also attitudes and behavior.

Abstract:
Abstract. The ability to think creatively is one of the 21st century skills that students must possess. This ability can be improved by using a learning model that exposes students to problems directly. This research aims to increase teacher activity, student activities, creative thinking skills, and student academic achievement by implementing Creative Problem Solving (CPS) learning model in class XI IPA 2 SMA PGRI 6 Banjarmasin. The research applied classroom action research (CAR) design with 2 cycles. The subjects were 39 eleventh grade students of natural science class. The data were analyzed by percentage and descriptive quantitative technique. The results showed that the teacher's activity in the implementation of the action increased from 70.15% (good) to 85.46% (very good). Student activity in cycle I 67.27% (quite active) to 85% in cycle II (active). Students' creative thinking ability for fluency indicators was originally 59.25% to 77.42%, flexibility which was originally 37.25% to 55.03%, elaboration which was originally 39.75% to 69.75%. Classical cognitive of students with a percentage of 69.23% increased to 87.17%. Students’ affective increased from 53.35% to 70.15% in cycle II and students' psychomotor learning outcomes were originally 59.69% to 69.4% in cycle II. Keywords: creative thinking ability, academic achievement, creative problem solving Abstrak. Kemampuan berpikir kreatif merupakan salah satu dari keterampilan abad 21 yang harus dimiliki siswa. Kemampuan ini dapat ditingkatkan dengan menggunakan model pembelajaran yang menghadapkan siswa pada masalah secara langsung. Penelitian ini bertujuan meningkatkan aktivitas guru, aktivitas siswa, kemampuan berpikir kreatif, dan hasil belajar siswa di kelas XI IPA 2 SMA PGRI 6 Banjarmasin dengan menerapkan model pembelajaran Creative Problem Solving (CPS). Penelitian menggunakan desain penelitian tindakan kelas (PTK) dengan 2 siklus. Subjek penelitian merupakan siswa kelas XI IPA berjumlah 39 orang. Data dianalisis menggunakan teknik persentase dan deskriptif kuantitatif Hasil penelitian menunjukkan bahwa aktivitas guru dalam pelaksanaan tindakan meningkat dari 70,15% (baik) menjadi 85,46% (sangat baik). Aktivitas siswa pada siklus I 67,27% (cukup aktif) menjadi 85% pada siklus II (aktif). Kemampuan berpikir kreatif siswa untuk indikator fluency, semula 59,25% menjadi 77,42%, flexibility yang semula 37,25 % menjadi 55,03%, elaboration yang awalnya 39,75% menjadi 69,75%. Hasil belajar kognitif siswa secara klasikal dengan persentase 69,23% meningkat menjadi 87,17%. Hasil belajar afektif siswa meningkat dari 53,35% menjadi 70,15% pada siklus II dan hasil belajar psikomotorik siswa yang semula 59,69% menjadi 69,4% pada siklus II. Hasil penelitian menunjukkan bahwa model pembelajaran CPS dapat meningkatkan hasil belajar dan kemampuan berpikir kreatif siswa. Abstract Background Semantic interoperability facilitates the exchange of and access to health data that are being documented in electronic health records (EHRs) with various semantic features. The main goals of semantic interoperability development entail patient data availability and use in diverse EHRs without a loss of meaning. Internationally, current initiatives aim to enhance semantic development of EHR data and, consequently, the availability of patient data. Interoperability between health information systems is among the core goals of the European Health Data Space regulation proposal and the World Health Organization’s Global Strategy on Digital Health 2020-2025. Objective To achieve integrated health data ecosystems, stakeholders need to overcome challenges of implementing semantic interoperability elements. To research the available scientific evidence on semantic interoperability development, we defined the following research questions: What are the key elements of and approaches for building semantic interoperability integrated in EHRs? What kinds of goals are driving the development? and What kinds of clinical benefits are perceived following this development? Methods Our research questions focused on key aspects and approaches for semantic interoperability and on possible clinical and semantic benefits of these choices in the context of EHRs. Therefore, we performed a systematic literature review in PubMed by defining our study framework based on previous research. Results Our analysis consisted of 14 studies where data models, ontologies, terminologies, classifications, and standards were applied for building interoperability. All articles reported clinical benefits of the selected approach to enhancing semantic interoperability. We identified 3 main categories: increasing the availability of data for clinicians (n=6, 43%), increasing the quality of care (n=4, 29%), and enhancing clinical data use and reuse for varied purposes (n=4, 29%). Regarding semantic development goals, data harmonization and developing semantic interoperability between different EHRs was the largest category (n=8, 57%). Enhancing health data quality through standardization (n=5, 36%) and developing EHR-integrated tools based on interoperable data (n=1, 7%) were the other identified categories. The results were closely coupled with the need to build usable and computable data out of heterogeneous medical information that is accessible through various EHRs and databases (eg, registers). Conclusions When heading toward semantic harmonization of clinical data, more experiences and analyses are needed to assess how applicable the chosen solutions are for semantic interoperability of health care data. Instead of promoting a single approach, semantic interoperability should be assessed through several levels of semantic requirements A dual model or multimodel approach is possibly usable to address different semantic interoperability issues during development. The objectives of semantic interoperability are to be achieved in diffuse and disconnected clinical care environments. Therefore, approaches for enhancing clinical data availability should be well prepared, thought out, and justified to meet economically sustainable and long-term outcomes.

Abstract Previous studies have shown that the distribution of major immune cell populations differ across mouse strains in peripheral blood and hematopoietic organs at baseline. Additionally, variations in immune responses exist between mouse strains in Leishmania spp. and Melanoma models, but these studies in the context of liver diseases including Non Alcoholic Fatty Liver Disease (NAFLD) and Hepatocellular Carcinoma (HCC) have not been carried out. With the effort to state if these variations exist within the liver, we performed comprehensive spectral flow cytometry analysis on the liver and spleens of three female mouse strains commonly used in cancer research (BALB/c, C57BL/6, and FVBN). NAFLD was induced by providing mice with a methionine choline deficient diet. To promote HCC, a single genotype (myc/p53+/-) was induced by administering transposon-based vectors to overexpress Myc-Luc and CRISPR/Cas-9 vectors to knock down p53 via hydrodynamic tail-vein injection. To confirm vector integration and monitor tumor growth (luciferase activity), in vivo imaging was performed at different time points. As a result, we confirm throughout this study that differences exist within immune cell populations of the liver and spleen at baseline. Moreover, we depict differences in the shift of immune cell populations across mouse strains in the pathologic states included. In NAFLD, there is an increase of dendritic cells (DCs), macrophages, NK cells, ILC1s, T regulatory cells (Tregs), TH1, and TH2, and a decrease of CD4+ T cells, B cells, γδ-T cells, and NKT cells in the liver across all mouse strains, when compared to controls. However, the population of neutrophils in BALB/c decreases, and the population of CD8+ T cells increases in C57BL/6 mice, which differs from the patterns of change seen in the other mouse strains. Moreover, in spleens, NAFLD promotes an increase of TH1 and NK cells, and a decrease of TH2 and NKT Cells across the mouse strains included in the study, whereas the patterns of change differ between mouse strains among the remaining immune cell types. When evaluating liver immune cell population shifts on our HCC model, BALB/c mice showed a decrease in macrophages and ILC1s, C57BL/6 mice showed a decrease in γδ-T cells, prominent shifts across Neutrophils, TH1, and NKT cells, and null changes in TH2 and CD4+ T cell shifts, and FVBN mice had a less prominent decrease of B cells, which differed from the shifts observed in the mouse strains used for comparison. Immune cell populations in the spleens of our HCC mouse model also presented with differences across the mouse strains studied. In conclusion, we highlight that key differences in immune profiles exist across commonly used mouse strains for cancer research in the presence of liver pathology including HCC, suggesting that mouse strains could be possible confounders in the study of immune oncology of HCC. Aim/Purpose: This study explored teachers’ data-driven decision-making processes during routine and emergency remote teaching, as experienced during the COVID-19 pandemic. Background: Decision-making is essential in teaching, with informed decisions promoting student learning and teachers’ professional development most effectively. However, obstacles to the use of data have been identified in many studies. Methodology: Using a qualitative methodology (N=20), we studied how teachers make decisions, what data is available, and what data they would like to have to improve their decision-making. We used an inductive approach (bottom-up), utilizing teachers’ statements related to decision-making as the unit of analysis. Contribution: Our findings shed an important light on teachers’ Data-Driven Decision-Making (DDDM), highlighting the differences between routine and Emergency Remote Teaching (ERT). Findings: Overall, we found that teachers make teaching decisions in three main areas: pedagogy, discipline-related issues, and appearance and behavior. They shift between making decisions based on data and making decisions based on intuition. Academic-related decisions are the most prominent in routine teaching, and during ERT, they were almost the only area in which teachers’ decisions were made. Teachers reported collecting data about students’ academic achievements and emotional state and considered the organizational culture, consultation with colleagues, and parents’ involvement before decision-making. Recommendations for Practitioners: Promote a culture of data-driven decision-making across the education system; Make diverse and rich data of different types accessible to teachers; Increase professional and emotional support for teachers. Recommendation for Researchers: Researchers have the potential to expand the scope of this study by conducting research using other methodologies and in different countries. Impact on Society: This study highlights the importance of teachers’ data-driven decision-making in improving teaching practices and promoting students’ achievement. Future Research: Additional research is required to examine data-driven decision-making in diverse circumstances.

Tujuan Pengabdian untuk melakukan deteksi dini masalah kesehatan jiwa dan meningkatkan pengetahuan tentang kesehatan jiwa pada remaja. Metode Pengabdian berupa deteksi dini dengan pengsisian instrumen SRQ-29 berdialek Malamoi (Self Reporting Questionnaire 29) dan edukasi kesehatan jiwa. Jumlah peserta kegiatan sebanyak 60 orang remaja siswa/siswi SMP YPPKK Moria Sorong. Hasil kegiatan pengabdian ini berjalan dengan baik dan lancar. Berdasarkan hasil deteksi dini ditemukan bahwa sebagian besar peserta remaja tidak terindikasi masalah kesehatan jiwa 32 orang (53,35%), kecemasan dan depresi 18 orang (30%), serta PTSD (Post Traumatic Stress Disorder) 10 orang (16,7%). Edukasi kesehatan jiwa yang diberikan juga meningkatkan pengetahuan remaja sebelum dan sesudah pemberian edukasi. Sebelum edukasi yakni 60 remaja (100%) dengan pengetahuan kurang, sesudah edukasi 42 remaja (70%) memiliki pengetahuan baik dan 18 remaja (30%) dengan pengetahuan cukup. Sehingga dapat disimpulkan bahwa gambaran kesehatan remaja di SMP YPPKK Moria Sorong yaitu ada yang tidak terindikasi gangguan, ada yang terindikasi kecemasan dan depresi serta PTSD. Pengetahuan remaja di SMP YPPKK Moria Sorong mengalami peningkatan dari kurang menjadi baik dan cukup setelah pemberian edukasi tentang pentingnya kesehatan jiwa bagi remaja. Mental health also contributes to bodily health and overall well-being. Purpose to carry out early detection of mental health problems and increase knowledge about mental health in adolescents. Method in the form of early detection by completing the SRQ-29 instrument in the Malamoi dialect (Self Reporting Questionnaire 29) and mental health education. The number of participants in the activity was 60 young students at YPPKK Moria Sorong Middle School. Result of this service activity went well and smoothly. Based on the results of early detection, it was found that the majority of adolescent participants had no indication of mental health problems, 32 people (53.35%), anxiety and depression 18 people (30%), and PTSD (Post Traumatic Stress Disorder) 10 people (16.7%). The mental health education provided also increases teenagers' knowledge before and after providing education. Before education, there were 60 teenagers (100%) with poor knowledge, after education 42 teenagers (70%) had good knowledge and 18 teenagers (30%) had sufficient knowledge. The description of the health of teenagers at YPPKK Moria Sorong Middle School is that some have no indication of a disorder, some have indications of anxiety and depression as well as PTSD. The knowledge of teenagers at YPPKK Moria Sorong Middle School has increased from poor to good and sufficient about the importance of mental health for teenagers. Penelitian ini mempelajari pengaruh variasi waktu penguapan terhadap kinerja membran selulosa asetat. Pada proses ultrafiltrasi, selulosa asetat dibuat dengan menambahkan polietilen glikol (PEG) sebagai agen pembentuk pori membran dan meningkatkan nilai fluks membran. Pembuatan membran selulosa asetat dilakukan dengan metode inversi fasa. Variasi waktu penguapan yang digunakan selama 0, 1, 3, dan 5 menit. Membran selulosa asetat yang terbentuk dikarakterisasi meliputi uji porositas, fluks dan rejeksi. Hasil penelitian menunjukkan waktu penguapan selama 5 menit menghasilkan membran yang lebih rapat. Il materiale esaminato proviene dall'archivio storico del «Centro Tumori Ossei» del C.T.O. di Firenze ed è relativo ad una casistica raccolta tra il 1940 ed il 1955. I radiogrammi convenzionali della colonna vertebrale sono stati valutati per determinare la presenza, l'estensione e le caratteristiche radiologiche delle lesioni metastatiche vertebrali. In particolare vengono discussi gli aspetti clinici e radiologici caratteristici delle metastasi vertebrali ed il ruolo che attualmente riveste tale metodica nella diagnostica di questa patologia.

Abstract In recently performed phase I/II clinical studies, we have demonstrated successful delivery of vectorized transgene-encoding mRNA payloads to solids tumors with substantial clinical benefit. In patients, persistent and prolonged transgene expression correlated with positive clinical outcome, and thus temporal expansion of the transgene expression profile is expected to improve the therapeutic benefit even further. Recently, circular RNA (circRNA) has been discovered as a novel class of endogenously expressed RNA. CircRNAs, in contrast to mRNAs, are resistant to exo-nucleolytic decay which results in high intra-cellular stability and persistent expression within cells. Therefore, circRNA-based systems serve as an interesting new technology towards prolonged effects of treatment. Here, we show that our proprietary vector system, circAde, allows high expression of circRNAs through spliceosome-dependent biogenesis. By comparing the protein output from circAde and mRNA-based vectors, we show that the circAde-system out-performs conventional mRNA-based design, both in terms of transgene yield and temporal sustainability of expression. Moreover, using a panel of proof-of-concept-reporters, we show that certain design-features greatly influence the circRNA production and subsequent protein expression from the vector. Without the 5’cap, circRNAs are dependent on IRESs (internal ribosome entry sequences) for effective translation. By conducting two orthogonal IRES screens on 1000+ putative IRES elements, we have identified the most effective IRES sequence in melanoma and lung-cancer cells supporting effective circRNA production and high-yield protein expression, thereby improving our circAde vector system even further. Our results open a new platform towards development of novel, more effective strategies for the treatment of cancer and other settings in which durable transgene expression is desirable. This article provides information on the growth of pike perch (Sander lucioperca) of Lake Tuzkon, which is part of the Aydar-Arnasay system of lakes. It was determined that pike perch in the first year of life grows to an average of 26.40 cm, in the second year - up to 44.45 cm, in the third - up to 53.35 cm, in the fourth - up to 67.37 cm, in the fifth and sixth years, respectively - up to 70.39 and 76.20 cm.

Abstract:
Penelitian ini merupakan penelitian deskriptif yang bertujuan untuk mendeskripsikan kemampuan berpikir kritis matematis siswa pada materi pola bilangan. Penelitan dilaksanakan di SMPN 2 Suwawa pada Tahun Ajaran 2020/2021 denga subjek penelitian adalah siswa kelas VIII. Teknik pengambilan data yang digunakan dalam penelitian ini adalah teknik tes dan wawancara, tes digunakan untuk mengukur kemampuan berpikir kritis matematis siswa, sementara wawancara digunakan untuk melengkapi dan memperkuat informasi tambahan yan diperlukan. Kemampuan berpikir kritis diukur dengan 4 indikator, yaitu: 1. Memberikan penjelasan sederhana, 2. Membangun keterampilan dasar, 3. Menyimpulkan, 4. Mengatur strategi dan taktik. Hasil penelitian menunjukkan bahwa rata-rata tingkat kemampuan berpikir kritis matematis siswa kelas VIII SMPN 2 Suwawa termasuk dalam kategori kurang yaitu sebesar 53,35%.

Abstract:
The purpose of this study is to analyze the relevance of morality with student achievement so that its contribution to learning achievement is known. This research includes the type of verification research conducted through data collection in the field using survey methods. The unit of analysis was taken using a simple random sampling technique. Data were collected through observation, interviews, questionnaires, and documentation. This study consisted of two variables, namely morality as the independent variable, and student learning achievement as the dependent variable. The results of the study show that the correlation between moral reasoning and student achievement is 0.683 (the degree of relationship is strong). This means that students' morality reasoning is 46.65% determined by learning achievement factors, and 53.35% is determined by other factors.

Anisotropic (Sm,Pr)Co5/Fe nanocomposites particles were prepared by electroless plating iron on the surface of (Sm,Pr)Co5nanoflakes after being prepared by ball milling for 4 h. A uniform and continuous coating layer was obtained due to the addition of complexing agent and the particle size of the reduced Fe particles was in the range of 10~20 nm. When the nominal addition of Fe was 15 wt%, the nanocomposites show enhanced remnant and saturation magnetization:Mr=53.35 emu/g,Ms=73.08 emu/g compared to the noncoated nanoflakes withMr=48.52 emu/g,Ms=60.15 emu/g, while the coercivity drops from 10.33 kOe to 8.89 kOe. The effect of Fe content on the magnetic properties of the magnets is also discussed.

Abstract:
Sulphide mineralization was found in relics of mine dumps from medieval mining near Utín, at SE part of the Havlíčkův Brod ore district (Bohemian-Moravian Highlands, Czech Republic). It is represented by pyrite, Fe-rich sphalerite, chalcopyrite, arsenopyrite, galena (with chemical compositions corresponding to other occurrences of this ore district) and more rare jamesonite and boulangerite. The main Ag ore was probably Ag-rich member of tetrahedrite group - kenoargentotetrahedrite-(Fe) with empirical formula (Ag5.45Cu4.48)Σ9.93(Fe1.75Zn0.32Mn0.01)Σ2.08Sb4.00S12.05. Another interesting mineral found in this association is Au-rich silver with Au contents in the range 53.35 - 56.40 wt. % and average empirical formula Ag0.60Au0.39. Schultenite, minerals of the pharmacosiderite group and inhomogeneous Fe-arsenates were detected from the rarely represented supergene mineralization.
