Abstract:
ABSTRACT An isopropylalaninyl monoamidate phenyl monoester prodrug of tenofovir (GS 7340) was prepared, and its in vitro antiviral activity, metabolism, and pharmacokinetics in dogs were determined. The 50% effective concentration (EC50) of GS 7340 against human immunodeficiency virus type 1 in MT-2 cells was 0.005 μM compared to an EC50 of 5 μM for the parent drug, tenofovir. The (L)-alaninyl analog (GS 7340) was >1,000-fold more active than the (D)-alaninyl analog. GS 7340 has a half-life of 90 min in human plasma at 37°C and a half-life of 28.3 min in an MT-2 cell extract at 37°C. The antiviral activity (>10× the EC50) and the metabolic stability in MT-2 cell extracts (>35×) and plasma (>2.5×) were also sensitive to the stereochemistry at the phosphorus. After a single oral dose of GS 7340 (10 mg-eq/kg tenofovir) to male beagle dogs, the plasma bioavailability of tenofovir compared to an intravenous dose of tenofovir was 17%. The total intracellular concentration of all tenofovir species in isolated peripheral blood mononuclear cells at 24 h was 63 μg-eq/ml compared to 0.2 μg-eq/ml in plasma. A radiolabeled distribution study with dogs resulted in an increased distribution of tenofovir to tissues of lymphatic origin compared to the commercially available prodrug tenofovir DF (Viread).

Исходя из (R)- и (S)-энантиомеров 2-(6-бензилокси-2,5,7,8-тетраметилхроман-2-ил)этанола, полученных ранее на основе парциального ацилирования соответствующего рацемического спирта в присутствии наиболее доступной липазы дрожжей Candida cylindracea синтезированы в 4 стадии оба энантиомера {2-[6-гидрокси-2,5,7,8-тетраметилхроман-2-ил]этил} триметиламмоний-п-толуолсульфоната (MDL-73404), хиральных аналогов α-токоферола. Сравнительное медико-биологическое тестирование разных по энантиомерному составу образцов соли MDL-73404 в отношении их влияния на генерирование активных форм кислорода перитонеальными макрофагами белых крыс в системах с люминолом или люцигенином показало неодинаковое ингибирование спонтанной и активированной 12-O-миристил-13-O-ацетилфорболом хемилюминесценции макрофагов.

ABSTRACT GS-7340 and GS-9131 {9-[(R)-2-[[(S)-[[(S)-1-(isopropoxycarbonyl)ethyl]amino]phenoxyphosphinyl]methoxy]-propyl]adenine and 9-(R)-4′-(R)-[[[(S)-1-[(ethoxycarbonyl)ethyl]amino]phenoxyphosphinyl]methoxy]-2′-fluoro-1′-furanyladenine, respectively} are novel alkylalaninyl phenyl ester prodrugs of tenofovir {9-R-[(2-phosphonomethoxy)propyl]adenine} (TFV) and a cyclic nucleotide analog, GS-9148 (phosphonomethoxy-2′-fluoro-2′, 3′-dideoxydidehydroadenosine), respectively. Both prodrugs exhibit potent antiretroviral activity against both wild-type and drug-resistant human immunodeficiency virus type 1 strains and excellent in vivo pharmacokinetic properties. In this study, the main enzymatic activity responsible for the initial step in the intracellular activation of GS-7340 and GS-9131 was isolated from human peripheral blood mononuclear cells and identified as lysosomal carboxypeptidase A (cathepsin A [CatA]; EC 3.4.16.5). Biochemical properties of the purified hydrolase (native complex and catalytic subunit molecular masses of 100 and 29 kDa, respectively; isoelectric point [pI] of 5.5) matched those of CatA. Recombinant CatA and the isolated prodrug hydrolase displayed identical susceptibilities to inhibitors and identical substrate preferences towards a panel of tenofovir phosphonoamidate prodrugs. Incubation of both enzymes with 14C-labeled GS-7340 or [3H]difluorophosphonate resulted in the covalent labeling of identical 29-kDa catalytic subunits. Finally, following a 4-h incubation with GS-7340 and GS-9131, the intracellular concentrations of prodrug metabolites detected in CatA-negative fibroblasts were approximately 7.5- and 3-fold lower, respectively, than those detected in normal control fibroblasts. Collectively, these data demonstrate the key role of CatA in the intracellular activation of nucleotide phosphonoamidate prodrugs and open new possibilities for further improvement of this important class of antiviral prodrugs.

Abstract:
Mutations in the skeletal muscle ryanodine receptor gene (RYR1) can cause susceptibility to malignant hyperthermia (MH), a potentially lethal genetic condition triggered by volatile anesthetics. MH is associated with hypermetabolism, which has directed research interest into oxidative phosphorylation and muscle bioenergetics. The most common cause of MH in the United Kingdom is the c.7300G>A RYR1 variant, which is present in ∼16% of MH families. Our study focuses on the MH susceptible G2435R-RYR1 knock-in mouse model, which is the murine equivalent of the human c.7300G>A genotype. Using a combination of transcriptomics, protein expression, and functional analysis, we investigated adult muscle fiber bioenergetics in this mouse model. RNA-Seq data showed reduced expression of genes associated with mitochondria and fatty acid oxidation in RYR1 mutants when compared with WT controls. Mitochondrial function was assessed by measuring oxygen consumption rates in permeabilized muscle fibers. Comparisons between WT and homozygous G2435R-RYR1 mitochondria showed a significant increase in complex I–facilitated oxidative phosphorylation in mutant muscle. Furthermore, we observed a gene-dose-specific increase in reactive oxygen species production in G2435R-RYR1 muscle fibers. Collectively, these findings provide evidence of metabolic defects in G2435R-RYR1 knock-in mouse muscle under basal conditions. Differences in metabolic profile could be the result of differential gene expression in metabolic pathways, in conjunction with mitochondrial damage accumulated from chronic exposure to increased oxidative stress.

Summary Feasibility of steam injection for three light-oil reservoirs in different geologic settings has been evaluated. The settings studied were a waterflooded deltaic sandstone, a waterflooded vuggy dolomite, and a deltaic sandstone structural trap with a gas cap. Optimization of steam injection to take advantage of individual reservoir characteristics is demonstrated. Results show that light-oil steamfloods can be designed to take advantage of post-secondary oil-saturation distribution. The resulting project may be carried out in a considerably different fashion from that of conventional heavy-oil steamfloods. We also re-evaluated an unsuccessful light-oil steamflood (LOSF) project carried out in the past. The re-evaluation correctly predicted failure because of early steam breakthrough. The results show that by considering details of geology and displacement process physics, the recent advances in reservoir characterization and modeling tools enable us to predict the performance of these projects more accurately. Introduction Steamflooding in shallow heavy-oil sands is a mature, successful technology with large commercial projects in the U.S., Canada, Indonesia, and Venezuela. Light-oil reservoirs have had fewer steamflood applications even where depth and other factors are favorable because of the generally lower post-secondary oil in place.1,2 Unlike heavy-oil steamflood projects, there are no large-scale commercial light-oil projects to use as analogs for design purposes. There are, however, a number of field trials in the literature, which have been reported both as successes and as failures. A review of the reported cases reveals some common reasons for success or failure. The major successful LOSF field cases are reported in Tables 1 and 2.3–10 Using steam as a heating agent in heterogeneous or extensively fractured reservoirs has given positive results (e.g., Teapot Dome field, Wyoming, U.S.A., and Lacq Supérieur field, France).9,10 Also, injecting steam into thick reservoirs with gas caps has resulted in expanding the gas cap and accelerating the gravity-drainage process (e.g., Shiells Canyon field, California, U.S.A., and Smackover field, Arkansas, U.S.A.).5,7 When steam is used for drive only, field trials have been successful in reservoirs with favorable geology (e.g., Schoonebeek field, The Netherlands, and Brea field, California, U.S.A.).3,4 The major unsuccessful11–15 LOSF field trials are shown in Table 3. One common characteristic of unsuccessful field trials has been steam channeling through thief zones (e.g., East Coalinga, California, U.S.A.; Triumph field, Pennsylvania, U.S.A.; El Dorado field, Kansas, U.S.A.; and Buena Vista field, California, U.S.A.).11–15 Scaling was another reason given for project failure (e.g., Elk Hills, California, U.S.A.).15 The overall screening criteria for the light-oil steamflood are shown in Table 4.1 All the above reservoirs met these screening criteria. Although these criteria are useful preliminary guidelines, the failed projects show that each reservoir should be examined individually. When reservoir geology and oil gravity are considered, the major recovery mechanisms in light-oil steamfloods can change significantly, as compared with conventional heavy-oil steamfloods. Viscosity reduction may increase recovery by accelerating the gravity-drainage process in thick columns for light crudes of 20 to 25°API. The beneficial effect of steam specific to lighter oils is distillation of light hydrocarbons, which results in very low residual oil saturation where steam displacement occurs, as well as enhanced solution gas drive and accelerated depletion from zones that are heated but not displaced. Because of greater initial fluid mobility in light-oil reservoirs, high steam-injection rates can be used. The light-oil steamflood projects can benefit from wider well spacing than conventional heavy-oil steamfloods. All of these can result in effective projects in a variety of geologic settings, despite lower oil in place than would be considered acceptable for heavy oils. The objective of this work was to examine the feasibility of steam-injection processes that are optimized to take maximum advantage of specific reservoir geologic settings. This study was conducted for three Chevron reservoirs:A waterflooded deltaic sandstone reservoir with channel and bar deposits.A waterflooded heterogeneous vuggy carbonate reservoir.A structural trap deltaic sandstone with a gas cap. To help validate the evaluation process, Buena Vista Hills,14,16 a failed light-oil steam field trial reported by Chevron, was also re-evaluated. Deltaic Sandstone Background. The first reservoir considered for steamflood evaluation is Minas in Central Sumatra. The reservoir is in early Miocene sandstones in the Sihapas formation at an average depth of 2,000 ft subsea with a maximum vertical oil column of 480 ft. Average porosity is 26%. The oil is 36°API with an average initial bubblepoint pressure of 235 psig. Current reservoir pressure is approximately 350 psig. Reservoir temperature is 207°F. The original oil in place (OOIP) estimate is 9 billion bbl.17,18 Initial development was on 214-acre spacing. Initial production was by aquifer drive that was augmented by peripheral water injection beginning in 1970. Starting in 1978, infilling reduced spacing to 71 acres. In the early 1990's, phased pattern waterflood development was implemented by use of 71-acre inverted seven-spot patterns. This development is approximately 70% complete and is targeted only in the thickest parts of the field. Ultimate recovery following completion is estimated to be 51% of OOIP. Geologic Setting. The Sihapas group of interbedded sandstones and shales was deposited as part of a large delta complex. Principal sand bodies are channel deposits and subtidal bar deposits. The channel deposits have an erosive base overlain by coarse sand and gravel and become finer-grained and shalier upward. The bar sands have a gradational base and become cleaner and coarser-grained upward. This is shown in Fig. 1. Steamflood Process. Both gravity and permeability contrasts will cause water to underrun in the channel sands, creating high-oil-saturation bypassed zones. Because of the natural tendency of steam to override, oil from these zones should be swept in a steamflood. It was felt that areas of the field with the greatest total thickness of these channel sand tops had the best steamflood potential.

Purified rat CD4+T cells were activatedin vitro, by the polyclonal mitogen Concanavalin A (Con A) or by mixed lymphocyte reaction (MLR), in the presence or absence of the glucocorticoid dexamethasone (DEX). They were then expanded in IL-2 and subsequently restimulated, this time in the absence of the hormone. The results indicate that the exposure of the cells to DEX in the primary stimulation changed the cytokine synthesis induced by the secondary stimulation. IL-4 production was increased by the pretreatment whereas synthesis of IFN-γwas diminished. Addition of DEX in the second activation suppressed all cytokine production. In brief, the transient presence of glucocorticoids in the culture induces a change in the pattern of cytokine production but the continuous presence causes inhibition of cytokine synthesis. Further studies in which IL-4 was used together with DEX showed that the cytokine potentiated the effect of the hormone.The data here presented suggest that glucocorticoids and the neuroendocrine system may be expected to have long-term immunological effects as well as short-lived immunosuppressive ones. High concentration of glucocorticoids suppress cytokine production but when steroids return to basal levels the immune response is directed in a way that favors Th2-type reactions. Possible implications regarding the immune response to pathogens and autoantigens are discussed.

Método: 34 mujeres, víctimas de explotación y violencia sexual, se entrevistaron clínicamente y evaluaron con test psicológicos. 48 sesiones de psicoballet y arteterapia escénica, 8 visitas guiadas a museos de arte y etnología y 24 sesiones de nuevas tecnologías de apoyo a la psicoterapia cognitiva y a la sensibilización artística patrimonial. Resultados: descendió la ansiedad un 40%. Conclusiones: la intervención mediante las artes ha permitido una innovación en la recuperación alternativa a la palabra.

A elevatio animae simboliza o instante em que a alma ascende ao Paraíso. A representação mais comum deste momento mostra a alma como uma criança, nua, envolta num panejamento puxado por dois anjos em sentido ascendente. No entanto, existem variações que serão aqui elencadas e analisadas. Os primeiros exemplares (conhecidos) desta iconografia apareceram patenteados em elementos arquitectónicos, nomeadamente, em capitéis. Mais tarde, a elevatio animae surgiu, sobretudo, em túmulos dos séculos XIII e XIV. Neste estudo, analisaremos algumas tipologias com representações da elevatio animae integradas em diferentes contextos iconográficos.

La teórica e historiadora del arte Mieke Bal destaca en sus estudios por encontrarse en un constante viaje, de idas y venidas que aportan frescura y revitalizan el arte contemporáneo. Durante su trayectoria ha conseguido renovar, a través de la reinterpretación, términos como el espacio, la temporalidad o el movimiento, este último entendido como desplazamiento y como tiempo, que se convierten en aspectos claves dentro de sus teorías. Pero si por algo ha destacado es por estar siempre ligada a los estudios visuales. Para ella, las imágenes son algo más que un elemento físico, presentan diferentes formas de mostrarse ante el espectador. Todos estos estudios y todo el trabajo de Mieke Bal se pudo ver reflejado en la exposición Emma & Edvard: Love in the Time of Loneliness que ella misma comisarió en The Munch Museum, del 27 de enero al 17 de abril de 2017. En ella consiguió crear nuevos elementos para el desarrollo de la exposición introduciendo en el mundo del comisariado nuevas formas expositivas.

Abstract:
A lo largo de las últimas décadas, los desarrollos tecnológicos han transformado la experiencia museística, poniendo bajo cuestión varias prácticas relacionadas con ella, como la crítica de arte y la mediación cultural, que se encuentran tanto enfrentadas entre ellas como afectadas por la proliferación de relatos y agentes de opinión. Dicha democratización ha vuelto a plantear el problema de cómo legitimar enunciaciones, usar cuerpos, sensaciones y deseos, y posibilitar el cambio en el interior y exterior de los museos de arte contemporáneo. La presente reflexión intenta conceptualizar la noción de lo intermedio como eje para la discusión sobre dichos temas. Asimismo, realiza un recorrido epistemológico partiendo de la interdisciplinariedad y los estudios CTG con el fin de explorar cómo sesgos y fuerzas inhibidoras dentro de las instituciones dificultan los procesos de habitar los espacios intermedios, mientras que enfoques alternativos, híbridos y situados apoyan la intersubjetividad, el intercambio y la implicación activa.

Abstract:
This study was conducted to determine apparent ileal amino acids digestibility (AIAAD) of diets with different levels of corn distiller’s dried grains with solubles (DDGS) and to determine AIAAD of corn DDGS by difference and regression methods in broiler chickens. One hundred and fifty 30-day-old male broiler chickens were used in the experiment. The corn DDGS were incorporated into basal diets at graded levels (0, 4, 8, 12, and 16%). All birds were killed at the age of 35 days and the contents of the lower half of the ileum were collected. AIAAD was calculated using chromic oxide as the indigestible marker. For AIAAD of DDGS determination, difference and regression methods were used. AIAAD of Lys (78.6%) and Met (91.3%) were the significantly highest in the diet without DDGS in comparison with other diets (P < 0.01). AIAAD of the rest of essential and nonessential amino acids were the lowest in the diet with 8% of DDGS in comparison with the diets with 0 or 16% of DDGS (P < 0.01). The apparent ileal crude protein digestibility was also the lowest in the diet with 8% (61.8%) of DDGS (P < 0.01). The highest differences in AIAAD of DDGS determined by the difference method and in the diet without DDGS (basal diet) determined by the regression methods were found in Lys and Met. There were very low differences between DDGS and basal diet in AIAAD of nonessential amino acids, except Ala and Asp. These results show that higher levels of DDGS decrease AIAAD.

Zwrócono uwagę na liczne przeobrażenia w strukturze społecznej, a w szczególności na zmianę charakteru interakcji społecznych i osłabienie więzi w społeczeństwach. Ponadto wskazano na zjawisko zindywidualizowania społeczeństwa jako przejawu dążenia do realizacji własnych aspiracji.

Abstract:
Pathogens invading the mammary gland are recognized through a range of pattern recognition receptors (PRRs), residing on the plasma membrane of mammary epithelial cells. Toll-like receptor 2 (TLR2) signalling is responsible for recognition of Gram-positive bacteria, which are the most common mastitis-causing pathogens in goats. Somatic cell counts (SCC) in milk are routinely determined in goat dairy flocks and serve as an indicator of milk quality, which is highly correlated to intramammary infections. Recently, a single nucleotide polymorphism of the TLR2 was suggested to be associated with SCC in goat milk. To further test the suggested association, we genotyped 61 Slovenian Alpine goats included in the dataset. The effect of the genotype was analysed using the general linear model (GLM) procedure of SAS/STAT software. We found the TLR2 genotypes significantly (p = 0.0007) associated with milk SCC. Animals with the A/G genotype had significantly (p ≤ 0.05) lower SCC value in milk compared to the G/G genotype. Our data suggest that the A allele is the minor one and is associated with lower milk SCC. In the current study, we provide a validated PCR-RFLP based genotyping assay for the TLR2 SNP (rs650082970) and confirm its association with milk SCC. Further studies to confirm the association on a larger number of animals of different breeds and to explain functional consequences of the polymorphism in relation to SCC are encouraged.
