Abstract:
Mutation of the N-terminal IGF-binding site of IGFBP-5 and its transfection into myoblasts revealed that the functions of IGFBP-5 in myogenesis can be divided in to IGF-dependent inhibition of myogenesis, and an IGF-independent anti-apoptotic role, assessed by decreased caspase-3 and PARP cleavage, caspase-3 and -9 activities, and decreased annexin V staining of cell populations overexpressing wild type (wtIGFBP-5) and non-IGF binding IGFBP-5 (mutIGFBP-5). Maintaining the balance between dynamic DNA methylation and demethylation is crucial to mammalian development and pathogenesis. In vitro methylation at the C-5 position of cytosine enhances cyclobutane pyrimidine dimer (CPD) formation and promotes transition mutations. The dorsal raphe nucleus (DRN) contains the majority of 5-hydroxytryptamine (5-HT)-producing neurons, whose firing is influenced by homeostatic feedback pathways. Dysfunction of 5-HT neurons is implicated in the pathophysiology of diseases such as depression. Thus, pathways regulating 5-HT neuron activity represent potential therapeutic targets. Negative feedback pathways influencing 5-HT neurons include 5-HT1A autoreceptors located in the DRN, and feedback from 5-HT1A, 5-HT2A and 5-HT2C receptors on postsynaptic neurons in forebrain regions. Dans des conditions de synthese maximale, nous avons montre que la production de ltb4 par les neutrophiles d'asthmatiques en intercrise est augmentee par rapport aux neutrophiles temoins. L'augmentation du chimiotactisme induit par le paf et de la fluidite membranaire des neutrophiles des asthmatiques suggerent que ces cellules sont dans un etat pre-active. Nous avons obtenu une correlation inverse entre le chimiotactisme induit par le paf et la presence de 5-hete neo-synthetise a l'interieur de la cellule, suggerant que la generation de 5-hete pouvait etre liee a la migration des neutrophiles

This thesis is focused on the development and utilisation of chemical and biological tools to probe the function of modified bases in DNA with specific exploration of the less well-studied T-modifications: 5-hmU, 5-fU and Base J. LCMS/MS techniques are first utilised to enable the accurate global quantification of T-modifications (5-hmU, 5-fU and Base J) in both trypanosomatids and mammalian DNA. A chemical affinity-enrichment sequencing method for the T-modifications is next described, which allows their chemoselective tagging over their C-modification counterparts.
