Here, we report the resequencing and genotyping ofPXRvariants in 75 Vietnamese individuals previously characterized for CYP3A enzyme activity after artemisinin exposure. We identified a total of 31PXRvariants, including 5 novel single nucleotide polymorphisms (SNPs), and we identified significantly different allele frequencies relative to other ethnic groups. A trend of significance was observed between the level of CYP3A4 induction by artemisinin and twoPXRvariants, the 8118C→T (Y328Y) and 10719A→G variants.

A retrospective analysis was carried out on the basis of epidemiological indicators such as prevalence and incidence for diseases of the musculo-skeletal system and osteoarthritis among residents of Arkhangelsk region and Krasnodar region. The incidence trends to increasing for diseases of bones and joints during the period of 2000-2012 years showed (in the Russian Federation for 13 years of growth was 8%, in the Arkhangelsk Region - 31%, and the Krasnodar Region- 142%). The similar results were obtained in studies for osteoarthritis (in Russia increase by 55% over 13 years for 41% of the Arkhangelsk Region, Krasnodar Region - 46%). Maximum growth indicator prevalence of diseases of the musculo-skeletal system was observed in the Krasnodar region and was 69% and in the Arkhangelsk region - 30%, in Russia on average increase was 26%. For thirteen years also marked increase in the prevalence of osteoarthritis in Russia as a whole and in the Arkhangelsk region and the Krasnodar region. Environmental and climate and geographical features of investigated regions can cause the detected differences.

Abstract:
Tomato (Solanum lycopersicum) is a popular crop among organic growers, particularly open-pollinated varieties; however, there is a lack of information evaluating these varieties in comparison with commercial F1 hybrids. This study was undertaken to compare conventionally produced commercial F1 tomato varieties available in the southeastern United States with open-pollinated varieties popular among organic growers. Nineteen tomato varieties were evaluated in 2011 and 2012 using the U.S. Department of Agriculture National Organic Program guidelines; however, the land used had not been in 3 years of organic production. Staked tomatoes were grown on plastic mulch. Data collected included early and total graded yield. In general, F1 hybrid, determinate or semideterminate varieties, had the best early and total yields compared with open-pollinated varieties. HSX 8115H and ‘Celebrity’ had the highest early total yield. ‘Costoluto Fiorentino’, an open-pollinated variety, also had good early total yield, but significantly less than HSX 8115H or ‘Celebrity’. ‘Costoluto Fiorentino’ was the only open-pollinated variety among the five highest yielding varieties for early total yield. In addition, ‘Celebrity’ had the greatest total yield of all the entries tested. All of the top five varieties for total yield were F1 hybrids and either determinate or semideterminate types. ‘Costoluto Fiorentino’ had total yield of 22,046 lb/acre, which was 14.3% lower than the lowest yielding variety in the top five yielding varieties. ‘Florida Pink’ had the largest average fruit weight, which also had the lowest total yield of all the entries. Although the open-pollinated varieties, popular among organic and local growers, yielded less than F1 hybrids, they may possess characteristics of color, flavor, or texture that are desired and were not evaluated in this study. This may be an opportunity for plant breeders to develop varieties desired by local organic growers that have unique characteristics, in addition to high yields and disease resistance.

Abstract:
Les caracteristiques d'une nouvelle source de plasma pour le depot de revetements par pulverisation sur des grandes surfaces ont ete etudiees. Cette source utilise une onde electromagnetique de frequence 2. 45 ghz pour accelerer les electrons qui ionisent les atomes d'argon par impact electronique. Un champ magnetique permet de confiner les electrons pour d'une part optimiser le couplage avec l'onde en tout lieu ou il est egal a 875 gauss et d'autre part transporter le plasma vers une cible de pulverisation polarisee negativement. Une source produisant une nappe de plasma de section rectangulaire de 75 cm2 a ete realisee. La caracterisation du plasma a ete effectuee en termes de densite electronique par interforemetrie, de temperature electronique par mesure de sonde et de courant d'ions recueillis sur la cible en fonction des parametres puissance micro-onde, pression d'argon, champ magnetique. Les valeurs obtenues ont montre que ce plasma est dense, avec une independance entre les parametres de creation du plasma et ceux de la pulverisation. L'usure des cibles de pulverisation est uniforme et correspond a l'intersection entre les lignes de champ magnetique et la surface de la cible. L'etude des couches de chrome deposees par ce procede a montre que la diminution de la pression permet d'obtenir des couches de densite proche de celle du chrome massique, avec des vitesses de depot elevees, et des contraintes residuelles faibles. Sous pression reduite constante, l'incorporation de carbone dans les couches de chrome par pulverisation reactive a permis d'augmenter la durete de ces couches. La morphologie devient plus dense, avec des contraintes residuelles faibles et une resistivite electrique faible. D'autres revetements (fer, oxyde de chrome, oxyde d'aluminium) ont ete realises pour demontrer la potentialite de ce nouveau dispositif a deposer en tension de polarisation negative continue de la cible des materiaux magnetiques et des oxydes

Ogni giorno nel mondo vengono eseguite migliaia di procedure chirurgiche per sostituire o riparare tessuti che sono stati danneggiati da malattie o traumi. L'ingegneria tissutale rappresenta una strategia alternativa che mira a rigenerare i tessuti danneggiati combinando cellule e biomateriali altamente porosi che fungano da impalcature (scaffolds). In questa tesi compilativa si presenta un approccio recentemente proposto per la rigenerazione del tessuto osseo. Saranno inizialmente descritte caratteristiche e proprietà dell'osso a livello macro e microscopico e il processo riparativo fisiologico. Si illustreranno quindi i principi dell'ingegneria tissutale, evidenziando i biomateriali utilizzabili, le cellule indicate per la rigenerazione e i rapporti funzionali tra di esse e lo scaffold che deve sostenerne la crescita. Successivamente si descriverà il concetto di ‘biomimetica’ dello scaffold e i metodi impiegati per migliorarne la funzionalità, imitando sia l'aspetto meccanico sia quello biologico della reale matrice ossea; verrà trattato infine un caso di scaffold biomimetico realizzato con nanocompositi, che appare un promettente sostitutivo dell'osso.

Abstract:
Precision healthcare, also known as personalised medicine, is based on our understanding of the fundamental building blocks of biological systems, with the ultimate aim to clinically identify the best therapeutic strategy for each individual. Genomics and sequencing technologies have brought this to the foreground by enabling an individual’s entire genome to be mapped for less than a thousand dollar in just one day. Recently, metabolomics, the quantitative measurement of small molecules, has emerged as a field to understand an individual’s molecular profile in terms of both genetics and environmental factors. This is crucial because a genome could only indicate an individual’s susceptibility to a particular disease, whereas a metabolome provides an immediate measurement of body function, enabling a means of diagnosis. However, the current approach of measurements depends on large-scale and expensive equipment such as mass spectroscopy and NMR instrumentation, which does not offer a single analytical platform to detect the entire metabolome. This thesis describes the development of an integrated CMOS sensor array technology as a single platform to quantify different metabolites using specific enzymes. The key stages in the work were: to construct instrumentation systems to perform enzyme assays on the CMOS sensor array; to establish techniques to package the CMOS sensor array for an aqueous environment; to implement and develop a room temperature Ta2O5 sputtering process on CMOS sensor array for hydrogen ion detection; to collaborate with a chemist and investigate an inorganic layer on top of the CMOS ISFET sensor to show an improvement of sensitivity towards potassium ion; to test several different enzyme assays electrochemically and optically and show the functionalities of the sensors; to devise microfluidic channels for segregation of the sensor array into different compartments and perform enzyme immobilisation techniques on CMOS chips; and integrate the packaged chip with microfluidic channels and enzyme immobilisation using 2D inkjet printer into a complete system that has the potential to be used as a multi-enzyme platform for detection of different metabolites. Two CMOS sensor array chips (1) a 256×256-pixel ISFET array chip and (2) a 16×16-pixel Multi-Corder chip were fully understood. Therefore, a high-speed instrumentation system was constructed for the ISFET array chip with a maximum readout speed of 500 frames per second, with 2D and 3D imaging capability, as well as single pixel analysis. Follow by that, a miniaturised measurement platform was implemented for the Multi-Corder chip that has three different sensor arrays, which are ISFET, PD and SPAD. All the sensor arrays can be operated independently or together (ionic sensor and one of the optical sensors). Several post-processing steps were investigated to allow suitable fabrication process on small 4×4 mm2 CMOS chips. Post-processing of the CMOS chips was first established using room temperature sputtering process for Ta2O5 layer, achieving Ta:O ratio of 1:1.77 and a surface roughness of 0.42 nm. This Ta2O5 layer was then fabricated on top of CMOS ISFETs, which improves the ISFET pH sensitivity to 45 mV/pH, with an average drift of 6.5 ± 8.6 mV/hour from chip to chip and a working pH range of 2 to 12. Furthermore, a layer of POMs was drop casted on top of Ta2O5 ISFET to make ISFET sensitive to potassium ions. This was investigated in terms of potassium ions sensitivity, hydrogen ions sensitivity and sodium ions as interfering background ions. The POMs Ta2O5 ISFET was found to have a net potassium sensitivity of 75 mV/pK, with a linear range between pH 1.5 to 3. Moreover, the POMs ISFET has -5 mV/pH in pH sensitivity, showing that it is selectivity towards potassium ions and not hydrogen ions. However, sodium ions were found to produce a large interference towards the pK sensitivity of POMs ISFET and reduced the pK sensitivity of POMs ISFET. Hence, further work is still required to modify POMs layer for better selectivity and sensitivity. Besides that, microfluidic channels were fabricated on top of the CMOS chips that could provide segregation for multiple enzyme assays on a single chip. In addition, a PDMS and a manual dam and fill method were developed to encapsulate the CMOS chips for wet biochemistry measurements. The CMOS sensor array was found to have the ensemble averaging capability to reduce noise as a function of √N , where N is the number of sensors used for averaging. Several enzyme assays that include: hexokinase, lactate dehydrogenase, urease and lipase were tested on the ISFET sensor array. Moreover, using an optical sensor array, namely a PD on the Multi-Corder chip and using LED illumination, quantification of cholesterol levels in human blood serum was demonstrated. Enzyme kinetics calculations were performed for hexokinase and cholesterol oxidase assays and the results were comparable to that obtained from a bench top spectrophotometer. This shows the CMOS sensor array can be used as a low cost portable diagnostic device. Several enzyme immobilisation techniques were explored but were unsuccessful. Alginate enzyme gel immobilisation with a 2D inkjet printer was found to be the best candidate to bio-functionalise the CMOS sensor array. The packaged chip was integrated with microfluidic channels and alginate enzyme gel immobilisation into a complete system, in order to perform an enzyme assay with its control experiments simultaneously on a single chip. As a proof-of-concept, this complete system has the potential to be used as a multiple metabolite quantification platform.

Abstract:
This project has been designed to increase our understanding of the clinical audit process, as it applies to veterinary practice in the UK, and to facilitate its introduction in a manner that brings maximum benefit to all stakeholders. It examines the medical scenario to define the process and glean any relevant information. It then takes the form of an action research project that examines in depth the introduction of the audit process into a small animal practice in outer NW London, including its impact upon the standard of clinical care supplied to its patients, and the sociological effect upon the working environment. The provisional conclusions reached in this way have then been triangulated with the findings of a focus group of veterinarians that are actively involved in the subject, and a broader questionnaire of veterinary practitioners and support staff. The research was able to highlight the key obstacles to introducing clinical audit into a veterinary practice, the benefits that can be achieved when its introduction has been successfully achieved, and how those benefits may ameliorate the time and expense involved. In particular, clinical audit was found to be an effective tool for improving client concordance with the recommended treatment regime for the animals in their care, and thus able to improve both patient welfare and practice income. The sociological changes that are needed to put clinical audit into place successfully, encouraging the development of an integrated team of highly motivated reflective practitioners working within a no-blame practice culture, can bring many additional benefits. This work has taken place at a time when various pressures, such as the RCVS Practice Standards Scheme, and an increased public demand for professional accountability have focused interest in the subject. The author has been leading the way in increasing public awareness of the process, encouraging further research, and ensuring that clinical audit is incorporated into the new modular postgraduate CertAVP designed to develop the learning and skills of the practicing vets of the future.

This research focuses on how South Korea's development model-namely, the Saemaul Undong, or the New Village Movement-is diffused internationally, in particular, to the developing country of Vietnam. South Korea's successful model has been diffused through various different channels for years, mostly in the form of foreign aid projects. Due to the prevailing view that international diffusion practices take place homogenously (in a near-universally standardized manner) within the recipient communities, and due to the propensity on the part of both donor and recipient governments to highlight only successful cases of diffusion while not publicizing those that have failed, several key questions, such as, how diffusion actually takes place, or how each project is likely to bring about different outcomes based on who initiates or leads the project, and to what extent this particular South Korean model has been viable and sustainable in the recipient country, remain largely unveiled. This research, therefore, aims to analyze the role of each stakeholder and how these stakeholders-either personnel or institutions-make an impact on the degree of diffusion of the Saemaul Undong process. It seeks to differentiate the impact of diffusion between short-term or one-time aid projects and those that have managed to become a sustainable development model in the recipient community. To be specific, the roles of politicians, administrative officials at the local level, non-governmental agents, external factors, and minor actors are investigated at the micro-level.

To meet the labor demands of green economic development, workforce development programs are increasingly training low-income workers for living wage jobs that contribute to environmental services or benefits. Yet, effectively preparing workers for jobs in emergent green economies, which can span many different sectors from energy efficiency to waste management, presents significant challenges for job training programs in practice. This thesis investigates the difficulties of adding a green dimension to job training while still meeting the basic employment objectives of workforce development programs. Through the experiences of the Oakland Green Jobs Corps and the Baltimore Center for Green Careers (BCGC), I analyze the mechanisms through which each program has influenced labor supply and demand to enable jobs in the green economy. Ultimately, the two cases highlight the tradeoffs between achieving green objectives and securing employment for disadvantaged workers at scale. In shedding light on how and why different programmatic decisions have influenced performance outcomes, this thesis aims to inform other cities' decisions in developing green jobs training initiatives.

Public pension funds are increasingly investing in cities. On the one hand, this appears as a positive development, as an organization traditionally based on exclusionary membership shares its benefits with the larger society. On the other hand, that this is occurring in the context of a disintegrating social contract could be a troubling feature, where private citizens are subsidizing their communities in the absence of state support. The Merriam-Webster dictionary defines "social contract" as "an actual or hypothetical agreement among the members of an organized society.. .that defines and limits the rights and duties of each." The past thirty years in U.S. history have seen a drastic deterioration in the tacit social contract binding the State, Capital and Labor. Transformations affecting these three actors have re-shaped their interactions and bargaining power. Through a discussion of the Varieties of Capitalism and Institutional Change literatures, the first objective of this paper is to make the case that community investing by pension funds is an institution (broadly defined) that has emerged in part because of the historic-economic forces driving the disintegration of the social contract: financialization, neoliberalism, and the decline of the labor movement. The second objective of this paper is to address the following research questions at the city level: What is the relationship between community investing by pension funds and the changing roles of Capital, Labor, and the State? How, in turn, does the design and organization of these institutions impact their ability to influence this relationship? This research finds that pension fund capitalism in New York City may both blunt and obscure the impacts of the weakened social contract. The pension funds could possibly strengthen the position of labor and increase benefits to communities if they incorporated opportunities for learning and capacity building into their programs.

In this thesis, I look at the political economy of the Urban Land (Ceiling and Regulation) Act (ULCRA) in Mumbai, India. Enacted in 1976, the stated aim of this legislation was to prevent speculation in the urban land market, and to make land available for affordable housing. The legislation imposed a "ceiling" on the amount of vacant land individuals could hold in urban areas. During the years it was in effect (1976-2007), almost all excess vacant land in the Mumbai agglomeration was exempted under the Act. Current literature posits that ULCRA failed to achieve its objective because politicians and bureaucrats were self-motivated rent-seekers, who were not interested in socially just redistribution of urban land. In 2007, using a conditional intergovernmental transfer scheme, the Central Government forced the State Government of Maharashtra to repeal ULCRA. I argue that current literature offers at best broad generalizations of the reasons ULCRA failed to achieve its objective in Mumbai. Through an in-depth analysis of the working of ULCRA in Mumbai, I show that it was never implemented as originally intended. Also, I show that ULCRA was frustrated by a number of deficiencies in institutions such as the lack of political will to take proactive action, capacity and cohesiveness in the bureaucracy, and amendments in other enabling statutes, to name a few. Seen from this perspective, it is incorrect to assume, as the current national urban development policy does, that a turn to market-led development in urban land markets will yield better results in delivering affording housing for the urban poor. Further, by analyzing the political economy of ULCRA's repeal, I show that under certain conditions, Central Government intervention may be a necessary step to protect interests of the poor. However, Central Government intervention needs to be designed keeping in mind the "why" and the "how", to ensure that the intervention's does not become dead letter, or redundant. I conclude with the dilemmas development planners are likely to face when designing laws and regulations in developing countries that feature a thinly institutionalized state, and a weak democracy.

After a century of economic and planning trends that sent industry overseas and to the suburbs, manufacturing is stabilizing, if not growing, in American cities. This is good news for many urbanists eager to attract the economic benefits of industry. However, while economic arguments for urban manufacturing are mature, the spatial strategies for supporting it are scattered or nonexistent. Planning codes and urban design ideals remain set in a 20th century mindset, while today's manufacturing has changed dramatically, becoming smaller and more networked than its previous iteration. Outdated perceptions of manufacturing block progressive policy reform at the highest level. Two thriving manufacturing centers, the Greenpoint Manufacturing and Design Center in New York and the American Industrial Center in San Francisco, provide clues for how 21st century manufacturing is spatializing in cities. These facilities are cultural beacons in their mixed-use neighborhoods and help create a framework for thinking about why urban manufacturing matters in our re-industrializing metropolises. This thesis describes each facility in detail, drawing conclusions about their key characteristics at four spatial scales. Few urban design ideals adequately describe the industrial activity occurring in cities today. Therefore, this thesis builds a system of meaning that values history, economics, and the lived experience of cities as a way to approach urban manufacturing. The emerging lens of Creative Placemaking is invoked as a way to unify these theories, suggesting that urban manufacturing is a superlative form of Creative Placemaking. In conclusion, this thesis provides recommendations and tools for cities looking to cultivate industrial urbanism by offering lessons, developing a framework for a new type of land use classification, and outlining a research brief. The thesis ends with a call for action: as industry continues to change its character, becoming leaner and more technologically driven, cities have great competitive advantage. Planners have a critical opportunity to bring manufacturing back into cities through sensitive, mixed-use zoning that connects people to the process of making and awakens policy professionals to manufacturing as a vital element of the contemporary creative city.

Abstract:
A free turbine is intended to produce the effect of a rotating flow field to be fed into a strutted annular diffuser for simulating a 3D fluid mechanic interaction and effect. This turbine is driven by the flow from a flow driver fan, upstream of the turbine. As sufficient pressure is required for the diffuser air flow, the concept of maximum work generation by a turbine is bypassed in the design intent. An inlet guide vane is designed for the turbine such that the rotational speed of the turbine matches the design intended speed and also produces an axial exit flow (no swirl) at design condition, and a rotating wake for off design conditions. The consideration of using of the inlet guide vanes (rather than usual stator-nozzles) is to avoid a mismatch between the absolute angle of the flow coming from the flow driver fan and the absolute design intent flow angle requirement, to run the turbine. Also, due to the 3D flow effects, some uncertainty exists in determining the exact entry air flow angle to the guide vane; this uncertainty is provided for with a rounded leading edge to the inlet guide vanes. The above design considerations when embedded in the conventional turbine design procedure resulted in an unusual turbine design with very low blade cambers.
