Abstract:
ABSTRACTArtemisinins induce drug metabolism through the activation of the pregnane X receptor (PXR)in vitro. Here, we report the resequencing and genotyping ofPXRvariants in 75 Vietnamese individuals previously characterized for CYP3A enzyme activity after artemisinin exposure. We identified a total of 31PXRvariants, including 5 novel single nucleotide polymorphisms (SNPs), and we identified significantly different allele frequencies relative to other ethnic groups. A trend of significance was observed between the level of CYP3A4 induction by artemisinin and twoPXRvariants, the 8118C→T (Y328Y) and 10719A→G variants.

Abstract:
A retrospective analysis was carried out on the basis of epidemiological indicators such as prevalence and incidence for diseases of the musculo-skeletal system and osteoarthritis among residents of Arkhangelsk region and Krasnodar region. The incidence trends to increasing for diseases of bones and joints during the period of 2000-2012 years showed (in the Russian Federation for 13 years of growth was 8%, in the Arkhangelsk Region - 31%, and the Krasnodar Region- 142%). The similar results were obtained in studies for osteoarthritis (in Russia increase by 55% over 13 years for 41% of the Arkhangelsk Region, Krasnodar Region - 46%). Maximum growth indicator prevalence of diseases of the musculo-skeletal system was observed in the Krasnodar region and was 69% and in the Arkhangelsk region - 30%, in Russia on average increase was 26%. For thirteen years also marked increase in the prevalence of osteoarthritis in Russia as a whole and in the Arkhangelsk region and the Krasnodar region. Environmental and climate and geographical features of investigated regions can cause the detected differences.

Boyhan, George E., Suzzanne Tate, Ryan McNeill, and Jeffrey McConnaughey. "Evaluation of Tomato Varieties under Organic Production Practices in Georgia." HortTechnology 24, no. 2 (April 2014): 252–58. http://dx.doi.org/10.21273/horttech.24.2.252.

Full text
Abstract:
Tomato (Solanum lycopersicum) is a popular crop among organic growers, particularly open-pollinated varieties; however, there is a lack of information evaluating these varieties in comparison with commercial F1 hybrids. This study was undertaken to compare conventionally produced commercial F1 tomato varieties available in the southeastern United States with open-pollinated varieties popular among organic growers. Nineteen tomato varieties were evaluated in 2011 and 2012 using the U.S. Department of Agriculture National Organic Program guidelines; however, the land used had not been in 3 years of organic production. Staked tomatoes were grown on plastic mulch. Data collected included early and total graded yield. In general, F1 hybrid, determinate or semideterminate varieties, had the best early and total yields compared with open-pollinated varieties. HSX 8115H and ‘Celebrity’ had the highest early total yield. ‘Costoluto Fiorentino’, an open-pollinated variety, also had good early total yield, but significantly less than HSX 8115H or ‘Celebrity’. ‘Costoluto Fiorentino’ was the only open-pollinated variety among the five highest yielding varieties for early total yield. In addition, ‘Celebrity’ had the greatest total yield of all the entries tested. All of the top five varieties for total yield were F1 hybrids and either determinate or semideterminate types. ‘Costoluto Fiorentino’ had total yield of 22,046 lb/acre, which was 14.3% lower than the lowest yielding variety in the top five yielding varieties. ‘Florida Pink’ had the largest average fruit weight, which also had the lowest total yield of all the entries. Although the open-pollinated varieties, popular among organic and local growers, yielded less than F1 hybrids, they may possess characteristics of color, flavor, or texture that are desired and were not evaluated in this study. This may be an opportunity for plant breeders to develop varieties desired by local organic growers that have unique characteristics, in addition to high yields and disease resistance.

"Comparative RNA-sequencing profiled the differential gene expression of liver in response to acetyl-CoA carboxylase inhibitor GS-0976 in a mouse model of NASH." PeerJ 7 (December 20, 2019): e8115. http://dx.doi.org/10.7717/peerj.8115.

Full text
Abstract:
Background Non-alcoholic steatohepatitis (NASH) is a progressive liver disease characterized by hepatic steatosis, lobular inflammation and fibrosis. Acetyl-CoA carboxylase (ACC) isoform 1 and 2 involved in de novo lipogenesis (DNL) and fatty acid oxidation have been identified as a therapeutic target in NASH. GS-0976, the inhibitor of ACC1 and ACC2, has achieved favorable therapeutic effects in clinical trials with NASH. The purpose of this study was to explore the transcriptional alterations regulated by GS-0976 in NASH. Methods C57BL/6 mice were fed on a choline-deficient, L-amino acid-defined, high-fat diet (CDAHFD) or normal diet for 12 weeks. Mice were treated with or without GS-0976 (3 mg/kg per day) in the last 8 weeks. Oil Red O, Haematoxylin-eosin (H & E), and Sirius Red were used to evaluate hepatic steatosis, inflammation and fibrosis. The comparative RNA-sequencing was conducted to analyse the hepatic gene expression profiles in mice. Reverse transcription–polymerase chain reaction analysis was performed to validate the differential expression of representative genes. Results GS-0976 attenuated the steatosis, inflammation, and fibrosis of NASH in CDAHFD mouse model. High-throughput sequencing and differential gene expression analysis showed that there were 516 up-regulated genes and 525 down-regulated genes after GS-0976 treatment. Genes involved in the metabolic process, extracellular matrix formation, immune response, and angiogenesis were significantly enriched. The “Metabolic pathways” and “ECM-receptor interaction” pathways were the most significantly enriched KEGG pathways in the up-regulated and down-regulated differentially expressed genes (DEGs), respectively. Conclusions Transcriptome analysis showed that GS-0976 could regulate the expression of genes related to metabolism, inflammation and fibrosis in NASH. The global transcriptomic changes in gene expression promote the further understanding for the inhibition mechanisms of GS-0976 in NASH.

Abstract:
The aim of this study was to analyze the relationships between cybervictimization, family functioning, and self-concept in adolescents, while taking the gender perspective into account. A study was conducted with a sample of 8115 adolescents, aged between 11 and 16 years (M = 13.34; SD = 1.04) from the State of Nuevo Leon, Mexico. A MANOVA 3x2 was performed to analyze the data. The results showed that family functioning, family self-concept, and academic self-concept were higher when cybervictimization was low. It was also observed that, in situations of cybervictimization, the girls had lower family self-concept, lower academic self-concept, and lower family functioning than the boys. The results that were obtained and their implications are discussed in the final section.

Abstract:
The aim of the present study was to analyze the relationship between family communication problems and cyberbullying, through psychosocial adjustment—psychological distress, attitude towards institutional authority, and problematic use of social networking sites—in adolescents. Random sampling by conglomerates was performed. A total of 8115 adolescents participated in the study (51.5% boys, 49.5% girls), and were aged between 11 and 16 years old (M = 13.34, SD = 1.04) and enrolled in the State of Nuevo León (Mexico). A structural equations model was developed using the Structural Equation Modeling Software (EQS). The results showed that problematic family communication is directly associated with cyberbullying, and also indirectly through the relationships of psychological distress and attitude towards transgression of social norms with the problematic use of social networking sites. The multi-group analyses also revealed gender differences in these relationships. Finally, the obtained results were discussed and their practical implications were shown.
