Abstract:
Accumulation of Divine Service is a ceramic installation referencing the sublime attribute of service, and how it relates to our temporal existence. Many aspects of the sublime are implemented into the contemporary art world today. The sublime can refer to ideas from terror to joy, and all across the spectrum of human emotions. The unifying element that seems to tie them together is a quality of awe-inspiring greatness, or the metaphysically divine. These attributescan inspire the mind and often lead one to dwell on the existence of a Supreme Being or Deity,what His purposes are, and how we as mortals work with or for that purpose.This installation encompasses some of my thoughts on the divinity that I believe dwells within all mankind. I do this by incorporating into the installation two repeated elements, the finial and the mug. The finial is an architectural element that is implemented at the apex of most religious meetinghouses. It is used for this purpose because it points toward the heavens and lifts the mind upward toward God. The mug, on the other hand, is one of the most humble and universal ceramic service vessels. It is used around the world as a drinking container whose sole purpose is service, or to give life-sustaining nourishment to mankind. The combination of these two visual elements seeks to encompass my personal art practice, my research, and the element of the sublime that we all possess.

L'exposition «Un temps à part» a été présentée à la Galerie des arts visuels de l'Université Laval. D’une part, de grands dessins blancs créés à partir d'ajouts suivis d'altérations ne laissant à la surface que quelques fragments et des traces d'effacement désormais illisibles. L'autre partie de l'exposition était constituée de sculptures en plâtre réalisées à partir d’un processus d'agglomération, soit par stratification de couches alternées de papiers imprimés et de plâtre créant des blocs, ou par incorporation de débris dans des formes compactes évoquant des phénomènes géologiques. Les oeuvres sculpturales étaient ensuite constituées d'accumulation de ces objets multipliés, les amoncellements suggérant l'état de ruine. Le mémoire décrit l'évolution du processus créatif où l'altération de l'oeuvre génère une esthétique de ce qui reste, proposant une expérience visuelle prenant sa source dans cette ambiguïté entre une oeuvre très présente dans ses caractéristiques matérielles et formelles tout en évoquant un état passé disparu.

Ce mémoire est le fruit d’une réflexion portant sur la pulsion qui m’habite de sans cesse créer de nouveaux objets, ainsi que sur les rapports que j’entretiens avec mes objets d’art et les objets manufacturés qui nous entourent. Elle s’est construite peu à peu, dans un aller-retour entre production à l’atelier et retour critique. Au cours des deux dernières années, j’ai cherché, par l’entremise de sculptures et d’œuvres installatives, à comprendre ma fascination pour le fait de vivre dans une société dont l’activité fondamentale semble être une transformation constante d’énergie et de matière. Dans un premier temps, j’y dresse plusieurs constats relatifs à mon positionnement comme artiste face aux différents discours sur l’art. Essentiellement, je développe l’idée que je n’ai pas besoin de régler le cas de l’art, d’un point de vue théorique, pour faire de l’art. Dans la seconde partie, je réfléchis sur différentes caractéristiques de ma démarche : le fait que je cherche à créer des objets qui s’insèrent et se comportent dans le réel comme des anomalies, que ma démarche est perméable à tout ce qui constitue mon expérience de vie, ou encore que j’active en manipulant de façon intuitive du matériel chargé de significations multiples. Dans un troisième temps, je présente la nature de l’imaginaire qui m’habite et explique la façon dont ce dernier influence ma manière de faire de l’art et la morphologie de mes œuvres. Vient ensuite un chapitre dans lequel je me penche sur mon passé de designer industriel et de musicien, dans lequel j’illustre la façon dont ces deux expériences de création, bien que fort différentes de l’art contemporain, influencent ma pratique actuelle. Finalement, je ferai une représentation schématisée des concepts, des méthodes et des dynamiques à l’œuvre dans mon processus créatif. Cette multitude de regards différents sur ma pratique permettra de faire ressortir, tout au long de ce mémoire, que ma démarche est nourrie par une série de tensions que je cherche à réconcilier, sans vraiment y parvenir, et que c’est cette impossibilité qui me pousse constamment à créer des nouvelles œuvres. Bien que mes œuvres soient souvent interprétées comme des commentaires sur l’actualité, le présent mémoire n’a pas comme point central l’actualité, mais bien mon travail de création, la façon dont il fonctionne, ce à quoi il est relié, etc. Je ne me pose pas la question « pourquoi et comment transformons-nous l’énergie et la matière? », mais plutôt « comment puis-je transformer le monde en transformant autrement l’énergie et la matière? »

Abstract:
Science, technology, engineering, and mathematics (STEM) educational achievement is vital to economic competitiveness in the United States and abroad. Despite a concerted effort, the US lags well behind many similarly developed nations. Research suggests that the integration of fine arts education into traditional STEM curriculum (STEAM) boosts academic achievement in STEM subjects and closes gaps between low- and high-socioeconomic status students. Justifications for STEAM programs are based, however, on the unexamined assumption, for one, that fine arts courses instill creative and critical thinking skills that can be transferred to STEM subjects. The present study explores the impact of taking fine arts courses on mathematics achievement in high school. Using the High School Longitudinal Study of 2009 and multi-level regression modeling, this study provides evidence that credit accumulation in fine arts courses relates positively to advancing past Algebra II in high school. Additionally, this estimated impact is much greater in magnitude for low-SES students than for their high-SES peers.

Abstract:
Through a comparative case study of Trisha Browns choreographies Floor of the Forest (1970) and Accumulation (1971), presented in a visual arts context in the 1970's and in the 2000's, this paper traces how performative aspects of Browns work has changed over time.  Floor of the Forest and Accumulation are analyzed through Erika Fischer-Lichte's theory of performativity in performance and the performing arts. The original choreographies in the 1970's are further contextualized by Browns history and role in the visual arts field in the 1960's and 1970's. Performances of Floor of the Forest and Accumulation in museums in the twentieth century are discussed in relation to Claire Bishops concept dance exhibition.  The findings show how the place/site continued to be a central factor in Brown's choreographing also after her site-specific performances in the 1970's. The exhibition of Floor of the Forest and Accumulation in museums took place through an adaptation of the choreographies to the specific site of the museum, in order to enable new performative spaces. An adaptability to the visual arts institution that, according to Bishop, characterizes the phenomenon dance exhibition.
Genom en komparativ fallstudie av Trisha Browns koreografier Floor of the Forest (1970) och Accumulation (1971), som har tagit plats i bildkonstens rum på 1970-talet och 2000-talet, spåras i uppsatsen hur performativa aspekter av Browns konstnärskap har förändrats över tid. Uppföranden av Floor of the Forest och Accumulation analyseras genom Erika Fischer-Lichte's teori om performativitet i konsthändelsen. De tidiga koreografierna kontextualiseras i relation Browns roll i 1960- och 1970-talens konstfält. Rekonstruktionen av koreografiernas uppföranden på museer på 2000-talet diskuteras vidare utifrån Claire Bishops koncept dansutställningar. Studien visar hur platsen/rummet fortsatte att utgöra en central aspekt i Browns koreograferande även efter hennes platsspecifika koreografier på 1970-talet. Utställningen av Browns koreografier på museer ägde rum genom en anpassning till museets specifika plats, för att således möjliggöra nya performativa rum. En anpassningsförmåga som enligt Bishop präglar fenomenet dansutställningar.

Les virus de la famille des herpesviridae, tels que le cytomégalovirus murin (MCMV), peuvent exprimer leurs propres micro-(mi)ARN, ils sont également capables de déréguler l’expression des miARN cellulaires. Bien que le profil global d’expression des miARN cellulaires ne soit que très modestement perturbé dans les cellules infectées par le MCMV; l’accumulation d’un miARN particulier, miR-27a, est très fortement régulée au cours de l’infection. Il est intéressant de noter que cette régulation semble se dérouler au niveau de la stabilité de la forme mature du miARN. Ainsi, le but de ce travail de thèse fut l’identification de facteurs à la fois viraux et cellulaires impliqués dans la régulation de miR-27. De plus, nous avons cherché à identifier de manière globale de potentielles activités enzymatiques impliquées dans la régulation de la stabilité des miARN. Nous avons à ce titre étudié le rôle de la terminal uridyl transférase TUT1 chez Arabidopsis thaliana ainsi que celui d’un de ses homologues chez les mammifères, Zcchc6. Bien que nos efforts pour établir le rôle de ces protéines dans la modification des miARN ne furent pas complètement concluants, nous avons établi les bases du mécanisme par lequel miR-27 est dégradé via son appariement à un abondant transcrit viral
Viruses from the herpesviridae family, such as the mouse cytomegalovirus (MCMV), are known for expressing their own micro (mi)RNAs, but they can also perturb the expression of cellular miRNAs. Although the cellular miRNA profile is only modestly impacted in MCMV infected cells, one particular miRNA, miR-27a is dramatically down-regulated upon infection. Interestingly, the regulation of this miRNA seems to be at the level of the stability of the mature form. The goal of this thesis was to identify viral and cellular factors involved in miR- 27 regulation. In addition, we also looked more broadly for potential enzymatic activities involved in the regulation of the stability of miRNAs. Therefore, we studied the role of the terminal uridyl transferase TUT1 in Arabidopsis thaliana and of one of its homolog, Zcchc6, in mammalian cells. Although our efforts to establish the role of these proteins in the modification of miRNAs were not fully conclusive, we could get insights into the mechanism by which miR-27 is degraded via pairing to an abundant viral transcript

Multilayered aluminium alloy composites were produced by accumulative roll bonding (ARB) to very high strain to generate sheet materials consisting of either 32 or 64 alternating layers of Al and Al-0.3w.%Sc alloy. Based on the starting heat treatment condition of the Al(Sc) alloy and the roll bonding temperature, several different Al/Al(Sc) combinations were produced: (i) SSSS-ARB (Al(Sc) in the supersaturated condition; Tdef = 200 ???C; 32 layers); (ii) Aged-ARB (Al(Sc) in the artificially aged condition; Tdef = 200 ???C; 32 layers), and (iii) SSSS-ARB-HT (Al(Sc) in the SSSS condition; Tdef = 350 ???C; 64 layers). Regardless of the roll bonding conditions, Al(Sc) in the form of a dispersion of ultrafine Al3Sc particles strongly impedes structural changes during thermomechanical processing whereas Al readily undergoes extensive dynamic and static restoration. The major aim of the thesis is to understand the effect of initial microstructure and processing conditions on microstructural development in these multilayered Al/Al(Sc) composites. The microstructures were investigated mainly by backscatter electron (BSE) and ion channeling contrast (ICC) imaging in the DualBeam Platform and transmission electron microscopy (TEM) whereas the crystallographic nature of the microstructures were investigated by electron backscatter diffraction (EBSD) and the various diffraction techniques available in the TEM. The mechanical properties of the materials were investigated by hardness and tensile testing. The deformation microstructure and texture of these two alloy combinations were strongly influenced by both the initial heat treatment condition of the Al(Sc) alloy whereby large-scale shear bands are generated during rolling when a dispersion of fine Al3Sc particles is present in the Al(Sc) layers. The deformation mechanism of both SSSS-ARB and Aged-ARB was strongly controlled by the relative hardening behaviour of adjacent layers. In Aged-ARB, a higher magnitude of in-plane shear stress, exceeding the flow stress of Al(Sc), was operative at the interfaces between layers; this was shown to cause the shear banding in this material. All materials were annealed for up to 6h at 350 ??C. This extended annealing generated alternating layers of coarse grains (Al layers) and a recovered substructure (Al(Sc) layers) with the substantial waviness of the layers in both Aged-ARB and SSSS-ARB-HT being inherited from the as-deformed material. While the Al(Sc) layers remain unrecrystallized in all materials due to particle pinning effects, the Al layers underwent continuous and discontinuous recrystallization after low and high temperature roll bonding, respectively. Shear banding in Aged-ARB also resulted in a reduction in intensity of the rolling texture components and had a randomizing effect on the recrystallization texture of the Al layers. The Al/A(Sc) multilayered composites were found to conform to the classic inverse strength/ductility relationship and no significant improvement in ductility (for a given strength) was evident. The barriers to achieving an excellent combination of ductility and strength (i.e. toughness) in these materials were identified to be delamination of the layers, which can be largely reduced (or eliminated) by careful control of starting materials (heat treatment condition and thickness) as well as the processing parameters during ARB.

Abstract:
This study examines the occurrence, distribution and storage of selenium in seagrass communities of Lake Macquarie, which is a heavily industrialised area of NSW, where notably high trace metal concentrations have been reported previously. Initially a suite of organisms was collected from a seagrass bed (Zostera capricornia) in the southeastern section of the lake to investigate the bioaccumulation and biomagnification of selenium. All organisms contained selenium, with sediment-dwelling organisms containing the highest Se concentrations. No consistent pattern of significant Se-metal correlations with Cu, Zn and Cd was found. Biomagnification of Se was evident, as concentrations increased from sediments and water to flora (algae and seagrass) to bivalves to Crustacea. However, this trend was not continued to the higher trophic groups of invertebrate predators and teleost fish. The bivalve Anadara trapezia was further studied. Intrinsic and extrinsic factors affecting the accumulation of Se were examined. Gender and mass were assessed in a single day study, followed by a temporal investigation of the effect of gender, reproductive cycle, temperature and salinity on Se accumulation. Gender had no effect on the accumulation of Se in A. trapezia. Se was present in all tissues (adductor, blood, foot, gills, intestine and mantle) and not immobilised or stored in a specific tissue, indicating that it plays a structural role in the tissues of A. trapezia. Se burden increased relative to size (shell length and dry mass) suggesting that Se is metabolically controlled within the organism. Se concentrations were found to fluctuate temporally because of: food availability in response to water temperatures; the reproductive cycle; and associated metabolic activities responding to temperature changes and food availability. Subcellular selenium associations in A. trapezia were examined to assist in the understanding of the fate of Se in marine tissues. Most of the Se was associated with proteins, suggesting that Se has a metabolic role in this marine organism. Proteins are intrinsically associated with the lipid bilayer of the cell membranes. A number of proteins (94, 85, 43, 36.5, 30, 23.4, 17.4 and 15 kDa) were separated by SDS PAGE from ethanol fractions. Determination of the Se concentration within individual proteins was not possible because the ratio of Se to protein was too low for further analysis. These findings indicated that Se plays a metabolic role in the tissues of the marine organism A. trapezia. The biochemical regulatory mechanism responsible for maintaining Se concentrations within the tissues is currently unknown.

The goals of this dissertation work were to assess the bioaccumulation potential of carbamazepine and amiodarone, two widely used ionizable pharmaceutical compounds that possess mid-range and high LogD values, respectively, and to evaluate alternative methods to assess chemical accumulation in bluntnose minnows, catfish, and tilapia. Results indicated that carbamazepine does not appreciably bioaccumulate in fish tissue with BCFk and BAF carbamazepine values < 10. Amiodarone, however, with a log D of 5.87 at pH 7.4, accumulated in fish tissues with kinetic BCF values <2,400. Collectively, the data suggest that full and abbreviated laboratory-derived BCFs, BCFMs derived from S9 loss-of-parent assays, as well as field BAF values are similar for each of the two drugs. In summary, the results from this dissertation indicated: 1) The reduced design BCF test is a good estimate for the traditional OECD 305 test. 2) In vitro S9 metabolism assays provide comparable BCF estimates to the OECD 305 test. 3) Metabolism may play a large role in the accumulation of drugs in fish. 4) Reduced BCF tests and in vitro assays are cost effective and can reduce vertebrate testing.

[Truncated abstract] Zinc deficiency in crops is the most widespread micronutrient deficiency, with about 50% of the cereal-growing areas worldwide containing low levels of plant-available Zn. Zinc plays multiple key roles in different metabolic and physiological processes; its deficiency in crops reduces not only grain yield, but also the nutritional quality of grains. Insufficient micronutrient intake, particularly Zn and Fe, afflicts over 3 billion people in the world, mainly in developing countries. Increasing the amount of Zn in food crops can contribute to improving the Zn status of people. Furthermore, Zn-dense seeds have agronomic benefits, resulting in greater seedling vigour, bigger root system and higher crop yield when sowed to soils with low plant-available Zn. Enhancing nutrient content and nutritional quality of crops for human nutrition is a global challenge currently, but it was mostly ignored during the breeding process in the past. There is a significant genotypic variation for seed Zn accumulation in several crops (including barley) which could be exploited in the breeding programs to produce genotypes with higher seed Zn concentration and content. However, the progress in Zn efficiency until now has mainly relied on conventional plant breeding approaches that have had limited success. Therefore, reliable alternative methods are required. Enhancing mineral nutrition through plant biotechnology may be a sustainable and beneficial approach in developing Zn-dense seeds in the staple crops. ... This DNA band was sequenced and converted into a simple sequence-specific PCR-based marker, which was designated as SZnR1 (seed Zn-regulator1). The developed marker is very easy to score, is inexpensive to run and amenable for a large number of plant samples. The successful development of SZnR1 molecular marker linked to chromosome region associated with seed Zn concentration and content using MFLP in this study illustrates the advantage of this technique over some other DNA fingerprinting methods used for identification of molecular markers for marker-assisted selection (MAS). In conclusion, the greater Zn efficiency of Sahara over Clipper under sufficient Zn supply may be attributed to its higher uptake of Zn. It appears that soil-based pot experiments under controlled condition may offer potential improvements over field experiments in screening for seed Zn accumulation. Shoot and seed Zn concentration and content can be used to diagnose the Zn statues of barley genotypes, and may be a useful selection criterion for Zn efficiency in large populations like doubled-haploid populations aimed at developing molecular markers for Zn efficiency. Identified QTLs influencing seed Zn concentration were repeatable in the field and glasshouse conditions, suggesting their robustness across environments as well as their value in marker-assisted selection. The developed PCR-based marker SZnR1 and other molecular markers associated with the QTLs on the short and long arms of chromosome 2H have the potential to be used for marker-assisted selection in breeding for Zn-dense seed in barley.

Abstract:
The master thesis is a research about the relationship between memorials and light. It first studies the characteristics of cultural memories and tries to find what the advantages of using lighting as a means of commemoration are. The nowadays memorials are very different compared to the traditional monuments and they should include a changing narrative, treating local and universal messages. They should involve the public.A contemporary memorial is ephemeral and continuously changing- the same as light is.A series of contemporary memorials have been selected to understand the tools that makes them successful. Furthermore, it was analyzed how these parameters could be achieved through light. 12 memorials that use light as an eloquent tool have been interpreted according to certain criteria.The second part of the thesis is an applied project related to the events that happened in Timisoara, Romania, in 1989 during the anticommunist Revolution. The process of creating memorials for Timisoara is a key focus of the study. The development is equally important as the end result. It searches for the significant messages and lessons of the event. Testimonials of the participants to the revolution have been studied. Interviews and questionnaires have been developed. Following this, significant places in the city and messages were chosen. The research will conclude with a lighting installations project proposal.

Three field sites were chosen to study the environmental assimilative capacity of heavy metals in soil. These sites were the Werribee Farm and the Myome Farm in Australia and Shenyang Zhangshi Irrigation Area in China. The Werribee Farm and the Shenyang Zhangshi Irrigation Area received sewage treatment and application on land for a long time. The Myome farm is an experimental site in which investigations on land application of municipal wastewater on water repellent soils is currently being trailed. Heavy metal contamination, in particular Cr, Cu and Zn, in the Land Filtration soil of Werribee Farm was widespread. More than a century of sewage irrigation has occurred in the Werribee Farm. The temporal distribution pattern of heavy metals (Cd, Cr, Cu, Ni, Pb and Zn) in the soil at this site follow an exponential trend with time and the spatial distribution pattern of accumulation of heavy metals in different paddocks correlates with the number of years of sewage irrigation at that site in the Farm. Extensive sewage irrigation at Shenyang Zhangshi Irrigation Area resulted in significant Cd pollution in soil-plant (rice) system and poses a significant threat to the health of local people. Even after eight years since cessation of sewage irrigation, the bioavailable fractions of Cd in the soil as analyzed by sequential extraction techniques were very high thus illustrating long-term persistence. The simultaneous competitive adsorption of metals in water repellent soils (at Myome Farm in South Australia) was studied. In the competitive situation, Cr, Pb and Cu are the heavy metal cations more strongly adsorbed by the soil, whereas Cd, Ni and Zn are the least adsorbed. The increase in Freundlich adsorption capacity by clay amendment suggested that clayed soils are capable sorption of higher heavy metal loadings compared to the non-clayed water repellent soil, which is more vulnerable to heavy metal inputs. A simple model of environmental assimilative capacity is proposed. The results of comparison of the three field sites shows that the Werribee Farm has a higher environmental assimilative capacity of heavy metals in soil than the soils at Shenyang Zhangshi Irrigation Area and Myome Farm, however heavy metal contamination at Werribee Farm is still a concern. The model of environmental assimilative capacity of heavy metals in soil is an effective tool to assist management of effluent applied land irrigation systems and can be used to better design environmental engineering systems.

Black carbon (BC), a stable form of organic carbon (OC), is a byproduct of the incomplete combustion of biomass, biofuels, and fossil fuel. The main objectives of this research are to examine the spatial distribution of OC and BC in urban soil and determine the influence of tree canopy cover and landscape maintenance on soil carbon accumulation. Soil sampling was conducted at 29 sites throughout the City of Denton, Texas, in May 2019. Samples were collected from underneath post oak canopies and in adjacent open areas and were analyzed for total carbon (TC), total organic carbon (TOC), total N (TN), C:N ratio, and BC. Although maintenance levels had no significant effect, TOC was greater underneath trees (5.47%, 5.30 kg/m2) than lawns (3.58%, 4.84 kg/m2) at the surface 0-10 cm. Total nitrogen concentration was also greater underneath trees (0.43%) than lawns (0.31%) at the surface 0-10 cm. Preliminary results for BC were closely correlated to TOC. The lack of difference in C:N ratio between cover types indicates that leaf litter quality may not be the primary driving factor in soil C and N accumulation. Instead, differences in soil properties may be best explained by manual C inputs and greater atmospheric deposition of C and N to soils with tree canopy cover. Identifying patterns and potential drivers of soil OC and BC accumulation is important because soil carbon sequestration not only reduces atmospheric CO2, but also may provide additional pollution mitigation benefits, thereby contributing to a more sustainable urban environment.

My research examines the three interrelated concepts of crisis; new imperialisms, spatial-temporal fix and accumulation by dispossession (ABD) stemming from the work of David Harvey as a way to understand the contested history of the Pakistan Railways. For the first thirty odd years after Pakistan's inception in 1947, the railways, a state-owned institution, was the primary mode of transport for the public, cargo, and workers. Alongside basic infrastructure, the railways had a vast network of hospitals, schools, workers' colonies and an array of physical infrastructure connected to production, operations and other aspects of the economy. The systematic ransack and decline of the Pakistan Railways reached its peak in 2010. Despite several attempts throughout the 1990s by successive democratic and military-led governments backed by the IMF/World Bank in 2015, it was announced that Pakistan railways would be revived under the banner of the 46 billion dollar China Pakistan Economic Corridor (CPEC) as part of the changing geopolitical context of growing regional connectivity and new Chinese imperialism. By examining the processes that underlie ABD, such as spatial-temporal fix, the following research shows that these processes not only reflect a shift of resources away from the public domain, but in Pakistan also entailed the transformation of the railways from a utilitarian welfare organization to an entity that facilitates looting, unbundling, and dispossession of shared resources and infrastructure.

The current dietary recommendations for optimal health are designed to increase our intake of two bioactive omega-3 (n-3) fatty acids, eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), abundant naturally in fatty fish such as salmon. Health authorities recommend that the general population consume two to three fatty fish meals per week (1) for optimal health and for the prevention of cardiovascular disease. However, some modern Western societies consume only modest amounts of fish and seafood (2;3). Land based vegetable oils may provide an alternative to meet these needs. Linseed and canola oils are rich in alpha-linolenic acid (ALA, 18:3n-3) (4). ALA can be converted endogenously to EPA and DHA and suggests that increasing the dietary intake of ALA may increase the conversion and accumulation of DHA in tissues and plasma. However, elevated dietary intakes of ALA in animals and humans results in an increased level of EPA in tissues yet there is little or no change in the level of DHA (5-7). The current consensus is that the synthesis of DHA from ALA in humans is limited yet the mechanisms involved in regulating the accumulation of DHA in tissues are poorly understood. The reputed rate-limiting enzyme in the conversion of fatty acids is delta 6 desaturase (D6D). ALA is a substrate for D6D and undergoes a series of desaturation and elongation reactions to yield n-3 long chain polyunsaturated fatty acids (LCPUFA). The final step in the synthesis of DHA from ALA involves translocation of its immediate fatty acid precursor, 24:6n-3 from the endoplasmic reticulum to the peroxisome to be partially beta-oxidised to yield DHA. The involvement of multiple enzymes in the desaturation-elongation pathway, and the integration of other pathways, such as phospholipid biosynthesis, suggests there are various steps that may regulate the accumulation of DHA in cell membranes. This thesis aimed to examine the possible regulatory steps in the conversion of fatty acids to LCPUFA, particularly in the synthesis of DHA from n-3 fatty acid precursors. The human hepatoma cell line, HepG2, was used as an in vitro cell system to examine the accumulation of individual fatty acids and their metabolites in isolation from other competing fatty acid substrates. The accumulation of linoleic acid (LA, 18:2n-6) and ALA in HepG2 cell phospholipids following supplementation with increasing concentrations of each respective fatty acid correlated with that described in vivo, as was the accumulation of their conversion products. The accumulation of DHA in cells supplemented with ALA reached a plateau at concentrations above 5 micro g/ml and paralleled the accumulation of 24:6n-3 in cell phospholipids, suggesting that the delta 6 desaturation of 24:6n-3 was prevented by increasing concentrations of ALA, thereby limiting the accumulation of DHA. The accumulation of DHA in cells supplemented with eicosapentaenoic acid (EPA, 20:5n-3) or docosapentaenoic acid (DPA, 22:5n-3) was significantly greater than the level of DHA that accumulated in cells supplemented with ALA. However, regardless of substrate, the level of DHA in cell membranes reached a plateau at substrate concentrations above 5 micro g/ml. This thesis further aimed to examine the effect of fatty acid supplementation on the mRNA expression of D6D in HepG2 cells. The expression and activity of D6D mRNA is subject to nutritional and hormonal regulation. The mRNA expression of D6D in HepG2 cells following supplementation with oleic acid (OA, 18:1n-9), LA, ALA, arachidonic acid (AA, 20:4n-6) or EPA was examined by real time RT PCR. The expression of D6D mRNA was reduced by up to 50% in cells supplemented with OA, LA, ALA , AA or EPA compared with control cells and suggests that fatty acids modulate the expression of the key enzyme involved in the conversion of fatty acids. The effect of fatty acid co-supplementation on the fatty acid composition of HepG2 cell phospholipids was also examined in an attempt to gain insights into the role of D6D and the enzymes involved in peroxisomal beta-oxidation on the accumulation of DHA from n-3 fatty acid precursors. The reduction in the accumulation of DHA in cells co-supplemented with DPA and docosatetraenoic acid (DTA, 22:4n-6) was greater than in cells co-supplemented with DPA and LA, suggesting that peroxisomal beta-oxidation may have a greater role in determining the accumulation of DHA from DPA than the activity of D6D. Further investigation should be directed towards understanding the role that peroxisomal beta-oxidation may play in the synthesis of DHA from precursor fatty acids. The fatty acid composition of cell membranes in vivo is a result of several physiological processes including dietary intake, phospholipids biosynthesis and fatty acid conversion as well as catabolic processes. This thesis demonstrates that a greater understanding of the regulation of the conversion of fatty acids will help to define dietary approaches that enhance the synthesis of n-3 LCPUFA from n-3 fatty acid precursors to lead to improved outcomes for health.

CGI-58 is the defective gene in the human neutral lipid storage disease called Chanarin-Dorfman syndrome. This disorder causes intracellular lipid droplets to accumulate in nonadipose tissues, such as skin and blood cells. Here, disruption of the homologous CGI-58 gene in Arabidopsis thaliana resulted in the accumulation of neutral lipid droplets in mature leaves. Mass spectroscopy of isolated lipid droplets from cgi-58 loss-of-function mutants showed they contain triacylglycerols with common leaf specific fatty acids. Leaves of mature cgi-58 plants exhibited a marked increase in absolute triacylglycerol levels, more than 10-fold higher than in wild-type plants. Lipid levels in the oil-storing seeds of cgi-58 loss-of-function plants were unchanged, and unlike mutations in beta-oxidation, the cgi-58 seeds germinated and grew normally, requiring no rescue with sucrose. We conclude that the participation of CGI-58 in neutral lipid homeostasis of nonfat-storing tissues is similar, although not identical, between plant and animal species. This unique insight may have implications for designing a new generation of technologies that enhance the neutral lipid content and composition of corp plants.

This thesis draws on David Harvey’s concept of “accumulation by dispossession” and an international political economy (IPE) approach centred on the institutional arrangements and power structures that privilege certain actors and values, in order to critique current capitalist practices of primitive accumulation by the global corporate extractive industry. The thesis examines how accumulation by dispossession by the global extractive industry is facilitated by the “free entry” or “free mining” principle. It does so by focusing on Canada as a leader in the global extractive industry and the spread of this country’s mining laws to other countries – in other words, the transnationalisation of norms in the global extractive industry – so as to maintain a consistent and familiar operating environment for Canadian extractive companies. The transnationalisation of norms is further promoted by key international institutions such as the World Bank, which is also the world’s largest development lender and also plays a key role in shaping the regulations that govern natural resource extraction. The thesis briefly investigates some Canadian examples of resource extraction projects, in order to demonstrate the weaknesses of Canadian mining laws, particularly the lack of protection of landowners’ rights under the free entry system and the subsequent need for “free, prior and informed consent” (FPIC). The thesis also considers some of the challenges to the adoption and implementation of the right to FPIC. These challenges include embedded institutional structures like the free entry mining system, international political economy (IPE) as shaped by international institutions and powerful corporations, as well as concerns regarding ‘local’ power structures or the legitimacy of representatives of communities affected by extractive projects. The thesis concludes that in order for Canada to be truly recognized as a leader in the global extractive industry, it must establish legal norms domestically to ensure that Canadian mining companies and residents can be held accountable when there is evidence of environmental and/or human rights violations associated with the activities of Canadian mining companies abroad. The thesis also concludes that Canada needs to address underlying structural issues such as the free entry mining system and implement FPIC, in order to curb “accumulation by dispossession” by the extractive industry, both domestically and abroad.

All practical text search systems use inverted indexes to quickly resolve user queries. Offline index construction algorithms, where queries are not accepted during construction, have been the subject of much prior research. As a result, current techniques can invert virtually unlimited amounts of text in limited main memory, making efficient use of both time and disk space. However, these algorithms assume that the collection does not change during the use of the index. This thesis examines the task of index maintenance, the problem of adapting an inverted index to reflect changes in the collection it describes. Existing approaches to index maintenance are discussed, including proposed optimisations. We present analysis and empirical evidence suggesting that existing maintenance algorithms either scale poorly to large collections, or significantly degrade query resolution speed. In addition, we propose a new strategy for index maintenance that trades a strictly controlled amount of querying efficiency for greatly increased maintenance speed and scalability. Analysis and empirical results are presented that show that this new algorithm is a useful trade-off between indexing and querying efficiency. In scenarios described in Chapter 7, the use of the new maintenance algorithm reduces the time required to construct an index to under one sixth of the time taken by algorithms that maintain contiguous inverted lists. In addition to work on index maintenance, we present a new technique for accumulator pruning during ranked query evaluation, as well as providing evidence that existing approaches are unsatisfactory for collections of large size. Accumulator pruning is a key problem in both querying efficiency and overall text search system efficiency. Existing approaches either fail to bound the memory footprint required for query evaluation, or suffer loss of retrieval accuracy. In contrast, the new pruning algorithm can be used to limit the memory footprint of ranked query evaluation, and in our experiments gives retrieval accuracy not worse than previous alternatives. The results presented in this thesis are validated with robust experiments, which utilise collections of significant size, containing real data, and tested using appropriate numbers of real queries. The techniques presented in this thesis allow information retrieval applications to efficiently index and search changing collections, a task that has been historically problematic.

In 1991, India adopted neoliberalism, a system of political economic practices that promotes private property and free trade, as its political and economic system to promote development in their country. India's neoliberal reform has created issues surrounding human development, resource accumulation, and power struggles. Eleven years later, in 2002, Bt cotton was introduced to the Indian agricultural sector. This research examines how the genetically modified organism Bt cotton is being used to commodify nature in the context of agriculture under neoliberalism. The research focuses on the dispossession of the rural farmers through the commodification of agriculture using Bt cotton. Dispossession of the rural farmers happen through the implications that arise from the commodification of nature. Through Marxist theory of primitive accumulation, this research analyzes accumulation by dispossession and how it neglects the working class and its struggle in rural India. Through this examination, the research will argue alternatives to the dispossession of the working class and the commodification of nature through Bt cotton. Dispossession, in this research, is examined both through working class, but also through the dispossession of biodiversity. Through the loss of biodiversity, the rural farmers are becoming dispossessed from a more sustainable environment. Along with these goals, the research will also incorporate themes of food security through changing landscape of agriculture due to the incorporation of Bt cotton. This research argues the contradictions that are presented through the commodification of agriculture under neoliberalism and provide a contribution to social justice literature, and our understanding of the relationship between technology and the commodification of nature.

Abstract:
Chez hevea brasiliensis mull. Arg. , du materiel embryogene peut etre maintenu a long terme en culture in vitro sur un milieu depourvu d'hormone. Le maintien des potentialites embryogenes necessite un repiquage selectif des amas proembryonnaires tous les 10 jours. Des repiquages mensuels et une selection des amas les plus hydriques ont permis d'obtenir une lignee non-embryogene. L'etude de la nutrition minerale et carbonee, des -carboxylations, de la respiration et des teneurs en substances de reserve a ainsi ete realisee sur une souche embryogene et une non-embryogene issues du meme materiel vegetal initial. Ces differents parametres ont ete evalues au cours des phases de multiplication des proembryons et de developpement des embryons somatiques. Au cours de la phase de multiplication, la suspension embryogene est caracterisee par une consommation en azote et sulfate et une activite enzymatique pepc plus elevees que la suspension non-embryogene qui pourraient etre a l'origine de leur teneur en proteines totales superieure. L'utilisation des sucres du milieu de culture est differents entre les deux suspensions. Les cellules embryogenes absorbent des sucres lors des premiers jours de culture ; ensuite elles peuvent subvenir elles-memes a leurs besoins en carbone. De plus, l'activite respiratoire indique que le metabolisme de la suspension embryogene est plus economique en energie que celui de la suspension non-embryogene pour la meme production de matiere seche. Lorsque les proembryons sont places sur le milieu de developpement mh0p2, les embryons somatiques obtenus sont peu nombreux de qualite mediocre ; ils forment de nouveaux embryons par autoembryonie. Les differents parametres etudies indiquent que le metabolisme de ces embryons somatiques est plus proche de celui des cellules en phase multiplication qu'en phase de developpement. L'apport de peg et d'aba dans le milieu de culture permet l'arret du phenomene d'autoembryonie et une production d'un plus grand nombre d'embryons de qualite plus satisfaisante. La presence de l'osmoticum entraine, chez la souche embryogene, une consommation en ions et en sucres et une activite respiratoire plus importantes pour lui permettre de mieux resister a la contrainte hydrique. La culture sur le milieu pega5 permet d'orienter ce metabolisme dans le sens du developpement en embryons. Les cellules embryogenes absorbent plus d'elements indispensables a leur developpement comme le phosphate, l'azote, le sulfate, le calcium et le magnesium ; elles presentent egalement une activite enzymatique pepc elevee que sur le milieu mh0p2. De plus, les embryons somatiques obtenus sont capables d'accumuler des reserves amylacees, proteiques et lipidiques. Cependant, elles sont plus faibles que chez les embryons zygotiques immatures. Le peg et l'aba ameliorent donc l'histodifferenciation des embryons mais ne permet pas leur maturation.

Abstract:
Useless things. I normally use trash things like pattern in painting still life. My father suffers from obsessive compulsive disorder – accumulation of things. In addition to painting, I also work with objects themselves. My interest also closely related to how the concepts of the subject, object and artifact are appliing in contemporary art. There are the tradition of imitation fact, principle of ready-made, the issue of technical reproducibility and virtual reality of things in history of art. As a central theme of my thesis, I selected the part of bodywork Tatra 613, which carries all the testimony about ,,useless and hopeless "situation in which it is located. It is also an object monumental and visually very aesthetic. Yet it is not my intention to settle for the principle of redy-made. Asked about artwork as intermediaries art and its seemingly useless producing and reproducing, I want to try to create another such usless situation. That's why I make a cardboard copy (1: 1) of this object. I also make video with motive 3D copy of the same object. I like to create some voltage between that useless object and valuable art practice. I think, it does not just mean such futility when these objects will be viewed as an art.

Abstract:
This thesis investigates how hegemonic white masculinity adopts a new mode of material accumulation by entering into an ambivalent existence as a historical agent and metahistory at the same time and continues to function as a performative identity that offers a point of identification for the working class white man suggesting that bourgeois identity is obtainable through the performance of bourgeois ethics. The thesis postulates that the phenomenal transitions brought on by industrialization and deindustrialization of 50's through 90's coincide with the representational changes of white masculinity from paradigmatic cowboy incarnations to the postmodern action heroes, specifically as embodied by Bruce Willis. The thesis also examines how postmodern heroes' "intero-alterity" is further problematized by antiheroes in Tim Burton's films.
