This priming process can be carried out by treating the soil with strong acid or base and then neutral ising it again. The aim of this study was to investigate the effectiveness of acid and base treatments in reducing inorganic contaminant availability in different soil types. The fir

This dissertation details three studies dealing with the manipulation of nucleicacids. In the first investigation, each of the four natural nucleobases were analyzed for theability to serve as a universal template at the ligation junction of a T4 DNA ligasereaction. This resulted in the first instance of sequence-independent ligation catalyzed byany DNA ligase. Although all of the nucleobases display universal templatingcapabilities, thymidine and guanosine provided the most effective results. In addition, lowered MgCl2 and ATP concentrations, as well as the inclusion of DMSO, also aided inthe

The 64 codons of the genetic code determine which amino acids are linked into a sequence to produce protein synthesis. Some of the codons specify the same amino acid by using only the first two letters of their codon triplet to do so, thus rendering their 3rd base irrelevant. Crick called this the wobble hypothesis, and a more complete understanding of the reading process could someday lead to a drug that can repair a misreading or to the creation of synthetic ribosomes capable of healthy protein synthesis. A step towards this goal is to apply mathematical logic to the 64 codons so that experi
