Abstract:
Cast aluminium composite materials based on AD0 aluminium, reinforced with carbon nanotubes and aluminium oxide nanofibers are obtained. It is shown that the additives of 0.05 wt % of single wall carbon nanotubes (SWCNT) and aluminum oxide nanofibers (AONF) allow increasing ultimate tensile strength of cast metal by 15 % and 16 % respectively. Strengthening of the metal is maintained after cold deformation and annealing. Aluminum AD0 with additives SWCNT or AONF retains high strength after annealing, while aluminum without additives is significantly weakened, this allows to obtain annealed aluminum wire, with a strength limit of 54 – 69 % and yield strength of 53 – 78 % higher than that of metal without additives. The influence of SWCNT and AONF on the size of aluminum grain AD0 in the cast state, after cold deformation and annealing has been studied. Nano-additives have been found to inhibit the growth of metal grains at the stage of collective recrystallization, as well as to influence the process of primary recrystallization of aluminum.

Abstract:
Optimum parameters are defined for friction stir welding modes of aluminum AD0 plates of 28mm thick. It is shown that at friction stir welding with mixing (FSW) aluminum AD0 plates a qualitative joint formation can be ensured at the expense of bilateral welding (alternatively on the right and root side of the joint). At the intensive plastic deformation impact and metal heating at the expense of friction the structure of welded AD0 alloy plates is formed by means of the narrow bands ("onion rings" successive interchange) having an ultra-dispersion grain structure. At heat application increase at the expense of the increase of tool rotation frequency and a tool pressure force a temperature-time mode of welding supporting a partial recrystallization of deformed grains in a joint center is changed. At the application of maximum tensile stresses a welded joint destruction occurs along a division border of media with a different structure in consequence of the incompatibility of their deformation.

We construct a Green’s function for the three-term fractional differential equation −D0+α−1u+aD0+μu+f(t)u=h(t), 0<t<b, where α∈(2,3], μ∈(1,2], and f is continuous, satisfying the boundary conditions u(0)=u′(0)=0, D0+βu(b)=0, where β∈[0,2]. To accomplish this, we first construct a Green’s function for the two-term problem −D0+α−1u+aD0+μu=h(t), 0<t<b, satisfying the same boundary conditions. A lemma from spectral theory is integral to our construction. Some limiting properties of the Green’s function for the two-term problem are also studied. Finally, existence results are given for a nonlinear problem.

Создана интеллектуальная система, реализующая ДСМ-метод автоматизированной поддержки исследований для задачи прогнозирования риска развития пролапса тазовых органов у женщин, перенесших роды через естественные родовые пути. Применяются методы для расширяющихся баз фактов. Впервые реализуется решетка стратегий, формируемая предикатами простого сходства, различия и сходства-различия (a, ad0, ad2), предложен алгоритм оптимизации пополнения базы фактов, использующий значение объясняемости. Описаны три эксперимента, проведенные по одной схеме, во втором при анализе подмассива исходных данных удален шум. Сформулированы новые задачи, позволяющие в дальнейшем улучшить результаты исследований.

SummaryAfter acute myocardial infarction (MI) the damaged heart has to be repaired. Factor XIII (FXIII) is considered a key molecule in promoting heart healing. FXIII deficiency was associated to cardiac rupture and anomalous remodelling in MI. During MI, FXIII contributes firstly to the intracoronary thrombus formation and shortly after to heal the myocardial lesion. To quantify the real contribution of FXIII in this process, and to explore its possible prognostic role, we monitored the FXIII-A subunit levels in 350 acute MI patients during the first six days (d0-d5) plus a control at 30–60 days (d30). A one-year follow-up was performed for all the patients. A transient drop in the FXIII-A mean level was noted in the whole cohort of patients (FXIII-Ad0 99.48 ± 30.5 vs FXIII-Ad5 76.51 ± 27.02; p< 0.0001). Interestingly, those who developed post-MI heart failure showed the highest drop (FXIII-Ad5 52.1 ± 25.2) and they already presented with low levels at recruitment. Similarly, those who died showed the same FXIII-A dynamic (FXIII-Ad5 54.0 ± 22.5). Conversely, patients who remained free of major adverse cardiac events, had lower consuming (FXIII-Ad0 103.6 ± 29.1 vs FXIII-Ad5 84.4 ± 24.5; p< 0.0001). Interestingly, the FXIII-A drop was independent from the amount of injury assessed by TnT and CKMB levels. The survival analysis ascribed an increased probability of early death or heart failure inversely related to FXIII-A quartiles (FXIII-A25th< 59.5 %; hazard ratio 4.25; 2.2–5.1; p< 0.0001). Different FXIII-A dynamics and levels could be utilised as early prognostic indicators during acute MI, revealing the individual potential to heal and suggesting tailored treatments to avoid heart failure or its extreme consequence.Note: This paper was presented in part at the 14th Congress of the International Society on Thrombosis and Haemostasis, Amsterdam, Netherlands, June 29 – July 24, 2013.

The storage roots of arracacha have high nutritional value, being rich in carbohydrates, minerals, vitamins A and C, and starch of high digestibility. However, in this crop one of the decisive factors for the production of roots with higher nutritional value is the balanced fertilization. The aim of this study was to evaluate the yield and nutritional quality of storage roots of arracacha, grown at increasing levels of NPK fertilization. The experiment was conducted in the randomized blocks design, with eight replications. The treatments consisted of three NPK levels (AD0 = unfertilized cultivation, AD1 = cultivation with 50% of recommended fertilization, and AD2 = cultivation with 100% ofrecommended fertilization). It was observedthat higher levels of NPK fertilization increase the number, size and marketable yield of storage roots of arracachacrop. NPK fertilization also increases the size and contentsof K, Mg, and Mn of storage roots, without interfering with the pulp pH and contentsof protein, Ca, Cu, and Fe. However, when the increase in root size is not accompanied by an increase in dry matter content, the starch, P, and Zn contents of the roots reduce with increasing levels of NPK fertilization.

Abstract To be able to charge a correct amount of chlorine dioxide in the D0-stage, a pulp mill needs good control of the bleaching process and good sensors to keep the variability of the bleaching result on an acceptable level. It is also important to include the bleaching agent demand from the dissolved matter in the pulp slurry. If this is done correctly, over- or undercharging of bleaching agents can be reduced, which lead to lower bleaching chemical cost, lower polluting emissions and higher pulp quality. Our previous research has shown that the dissolved organic carryover from the O2-stage varies significantly when bleaching softwood kraft pulp. The present study investigated the corresponding impact in the case of bleaching of birch pulp. Different mill configurations and process conditions have been simulated in laboratory trials, including proceeding A-stage treatment, different degrees of washing before and between the stages, and a comparison of the effects of recycled and non-recycled wash filtrates. The results have confirmed the significant impact of the dissolved organic matter, and the knowledge which have been generated can be used to understand how measurement and control concepts can be developed to improve the pulp quality control and to decrease production cost.

During the process of the aluminum ingot manufacturing obtaining of a fine-grained structure has a huge significance for effectiveness of the further rolling process. The reduction of the grain size leads to an increased plasticity and higher grades of deformability. Today the multiple methods of improving the casting process are being developed varying from mechanical to physical or chemical methods, and their choice depends on different factors. However, the effect on the size of grains of the multi-factor impact was not described in literature. This paper investigates methods of casting including piston pressing with simultaneous exposure to the weak pulsed currents from electromagnetic field during crystallization of melted metal for the first time. The fine-grained uniform over the cross-section structure was achieved in the AD0 alloy ingots.

Abstract:
La PARG est une enzyme dégradant un polymère d’ADP riboses, PAR, dont la synthèse est catalysée par les PARPs, formant une famille de 17 protéines codées par 17 gènes différents. La PARG, en revanche, est codée par un seul gène et l’épissage alternatif de l’ARNm conduit à la formation d’au moins 4 isoformes. La PARP la plus décrite est PARP-1 dont l’activité, stimulée par les cassures dans l’ADN, permet la détection et la signalisation des dommages ainsi que le recrutement de facteurs de réparation. Le rôle de PARG dans la réponse aux dommages dans l’ADN est moins connu. Ainsi, afin d’étudier les rôles de PARG dans la réponse aux dommages, nous avons choisi l’approche de l’interférence à l’ARN empêchant l’expression de tous les isoformes de PARG dans une lignée cellulaire tumorale HeLa (PARGkd). L’absence de PARG dans une cellule irradiée induit une persistance de cassures de l'ADN conduisant à la mort cellulaire par catastrophe mitotique. Puis, en utilisant la technique de microirradiation laser, nous avons mis en évidence que le recrutement de PARG aux dommages est dépendant de l’activité de PARP-1 et de PCNA, une protéine de réplication. Nous avons démontré une interaction entre PARG et PCNA et constaté la sensibilité accrue des cellules PARGkd face à l’hydroxyurée, un inhibiteur de la réplication. Ces cellules présentent alors un fort ralentissement de leur progression dans la phase S après leur relâchement ainsi qu’un blocage en phase S précoce, suggérant un rôle de PARG dans la progression de la réplication d’un ADN endommagé. Nos résultats font de la PARG une potentielle cible thérapeutique en thérapie du cancer<br>PARG is an enzyme degrading a polymer composed of ADP-ribose units, called PAR synthesized by PARPs belonging to a family of 17 members all encoded by different genes. PARG is encoded by only one gene and mRNA splicing leads to the expression of at least 4 isoforms. The most described PARP is PARP-1 which activity, stimulated by DNA breaks, allow the detection and signalization of damages and DNA repair protein recruitment. The role of PARG in DNA damage response is lees known. Thus, in order to elucidate the role of PARG in DNA damage response, we chose RNA interference approach to prevent expression of all PARG isoforms in a HeLa cell line (PARGkd). We demonstrated that PARGkd cells present a delay in DNA damage repair leading to cell death by mitotic catastrophe. Moreover, by using laser microirradiation which introduce DNA damages locally in cell nucleus, we showed that he PARG recruitment to DNA damage sites is dependent on PARP-1 activity and on PCNA. We demonstrated an interaction between PARG and PCNA and noted hypersensitivity of PARGkd cells towards hydroxurea (HU), a replication inhibitor. These HU treated cells present a strong delay in S phase progression after their release and a block in early S phase, which indicate a role of PARG in replication process. The proven role of PARG in DNA damage response and its potential implication in replication events, lead to consider PARg as a new interesting therapeutic target in cancer therapy

Abstract:
En 2017, Statistique Canada a rapporté qu'un Canadien sur quatre mourra d’un cancer. Chaque jour, nous sommes confrontés à des facteurs environnementaux qui imposent à notre ADN un stress génotoxique. Ce stress peut avoir de graves conséquences au point de menacer notre intégrité génomique, comme les cassures d'ADN double-brin (DSBs). Heureusement, nos cellules ont deux voies principales pour combattre ce type de lésions : la recombinaison homologue (HR) et la Classical Non-Homologous End-Joining (CNHEJ). La voie HR, un type de réparation sans erreur utilisé dans la phase-S du cycle cellulaire, assure une réparation fidèle de la zone endommagée et conserve l'intégrité de l'information génétique. Les individus porteurs de mutations dans les protéines de cette voie, telles que BRCA1 et BCRA2, sont plus susceptibles de développer des cancers du sein et de l'ovaire. Récemment, la clinique a connu une percée majeure dans le traitement du cancer de l'ovaire. Une nouvelle classe de médicaments a été autorisée par la US Food and Drug Administration (FDA) pour traiter les cancers de l'ovaire récurrents qui présentent une HR défective. Ces médicaments inhibent un des acteurs les plus précoces dans la réponse aux dommages à l'ADN (DDR): la PARP-1 (Poly(ADP-ribose) polymerase-1). Lors de l'induction de dommages à l'ADN, la PARP-1 devient fortement activée, conduisant à la production massive de polymères de poly(ADP-ribose) (PAR) générés à partir de l'hydrolyse du nicotinamide adénine dinucléotide. Ce polymère agira comme une plateforme pour recruter des facteurs de réparation de l'ADN au site de réparation. L'application clinique réussie des inhibiteurs de la PARP (PARPi) vient des observations où les mutations ou l'extinction de BRCA1/2 entraînent une diminution de l'activité HR. L'inhibition de la PARP-1 combinée à cette déficience en HR favorise la mort cellulaire, un phénomène appelé létalité synthétique. Trois PARPi sont actuellement autorisés par la FDA et sont utilisés pour le traitement du cancer gynécologique. Malgré l'efficacité thérapeutique de ces inhibiteurs, les mécanismes induisant une régression tumorale ne sont pas complètement compris. Ainsi, il devient extrêmement important de déchiffrer davantage ces mécanismes pour atteindre le plein potentiel des PARPi. Pour ce faire, une recherche fondamentale sur les fonctions des PARPs, et de leurs partenaires dans la DDR, est essentielle et constitue l'objectif général de cette thèse. Durant mon doctorat, nous avons étudié l'influence de la PARP-1 dans la voie HR au moment de l'étape initiale de la résection, qui est essentielle pour l'élimination de l'ADN endommagé. Certaines études ont montré l'implication de la PARP-1 dans le recrutement de la protéine de résection MRE11. Ici, nous démontrons que la PARP-1 a une nouvelle fonction dans la résection des DSBs et nous proposons un nouveau modèle pour expliquer la létalité synthétique observée dans les tumeurs avec une HR défective. Pour compléter l'objectif de ce doctorat, nous avons étudié les rôles régulateurs de la PARP-1 au cours du processus HR, mais plus tard dans la résolution des lésions, c'est-à-dire au maximum de la formation des foyers RAD51, une étape cruciale pour la réparation efficace des DSBs via la HR. Nous avons observé que le PAR-interactome (PARylome) est, à ce moment, fortement enrichi en protéines impliquées dans le métabolisme de l'ARN. Plusieurs des protéines les plus abondantes étaient constituées d’hélicases d’ADN et d’ARN, et de facteurs de transcription. Puisque certains de ces gènes sont mutés dans les tumeurs, ils pourraient théoriquement être des cibles prioritaires pour une utilisation conjointe avec des PARPi. Nous avons également étendu notre étude de la PARylation à la chromatine, au niveau des histones. Nous avons constaté que les queues d'histones ne sont pas les seules cibles de la PARP-1 et que les domaines globulaires centraux sont également PARylés. Finalement, le grand intérêt clinique de la PARP-1 méritait une analyse approfondie de son expression systémique. Ainsi, j'ai terminé mes études en décrivant la distribution et l'abondance tissulaire de la PARP-1 dans les organes simiens, avec l'objectif principal de fournir des informations précieuses quant à l'efficacité potentielle des PARPi ou sa résistance, dans un tissu donné et maladies apparentées. En résumé, cette thèse fournit de nouvelles informations importantes sur les mécanismes orchestrés par la PARP-1 lors de la réponse aux DSBs, y compris les réseaux protéiques complexes engagés dans le remodelage des fonctions cellulaires nécessaire au maintien de l'intégrité génomique.<br>In 2017, Statistics Canada reported that one out of four Canadians will die of cancer. Every day, we face environmental factors that burden our DNA with genotoxic stress. This stress can lead to severe types of DNA damage that can threaten our genomic integrity, namely double-strand breaks (DSBs). Fortunately, our cells have evolved with different repair mechanisms to deal with such lesions. There are two primary types of repair against DSBs: Homologous Recombination (HR) and Classical Non-Homologous End-Joining (CNHEJ). The HR pathway is an error-free repair mechanism used in the S-phase of the cell cycle to ensure faithful repair of the damaged area and thus preserve our genetic information. Individuals that bear mutations in proteins involved in this pathway, such as BRCA1 and BCRA2, have been associated with the development of breast and ovarian cancers. Almost 4 years ago, the field went through a major breakthrough in ovarian cancer care. A new class of drugs was accepted by the US Food and Drug Administration (FDA) to manage recurrent ovarian cancers that display HR-deficiencies. These drugs consist of inhibitor molecules against one of the earliest sensors of DNA damage in the cell: PARP-1 (poly(ADP-ribose) polymerase-1). Upon DNA damage induction, PARP-1 becomes highly activated, leading to the massive production of poly(ADP-ribose) (PAR) polymers, from the hydrolysis of nicotinamide adenine dinucleotide, which in turn modify several proteins posttranslationally and act as a scaffold to recruit DNA repair factors to the repair site. The successful application of PARP inhibitors (PARPi) arose from the observations that mutations or silencing of BRCA1/2, resulted in diminished HR activity. In the context of HR deficiency, the concomitant inhibition of PARP resulted in cell-death, an effect called synthetic lethality. Three PARPi are currently accepted by the FDA and are being clinically used for the treatment of gynaecological cancers. Notwithstanding the great promise of these inhibitors for other types of cancers, the mechanism by which these are inducing cancer lethality is not fully understood. Thus, it becomes of extreme importance to further decipher its mechanistic ways, to achieve full potential of PARPi in the clinic. To achieve this, fundamental research on the functions of PARPs and their protein partners in the DNA damage response is indispensable and constitutes the general aim of this thesis. During my doctoral work, we investigated the influence of PARP-1 during the HR pathway, primarily during the initial step of resection, which is essential for the removal of damaged DNA. Early reports of PARP-1 involvement in resection described the recruitment of the resection protein MRE11 to sites of damage in a PARP-1 dependent manner. Here, we demonstrate that PARP-1 has a novel function in DSB resection and we propose a new model for the synthetic lethality observed in HR-deficient tumors. To further complement the general aim of this doctorate, we investigated the regulatory roles of PARP-1 during the HR pathway, however in a later stage of HR resolution, at the peak formation of RAD51 foci, which is a crucial step for the efficient repair of DSBs through HR. We observed that the PAR-interactome (PARylome) at this stage was abundantly enriched with RNA-processing factors. Several of the most abundant proteins consisted of DNA and RNA helicases, as well as transcription factors, some of which were found to be mutated in tumors, and thus can be seen as potentially druggable targets to be used in combination with PARPi. We also extended our PARylome study to the chromatin proteome and investigated the histone PARylome upon DNA damage. Interestingly, we found that histone tails are not the only targets of PARP-1 and that globular domains are also targets of PARylation. Lastly, the high clinical interest of PARP-1 warrants studies addressing PARP-1 organ distribution. Thus, I finalized my studies by extensively describing and reporting PARP-1 tissular and cellular distribution and abundance in monkey organs, with the main objective of providing valuable information to any study assessing PARP inhibition efficacy and resistance in any given tissue and related diseases. In summary, this thesis provides important new information on the mechanisms PARP-1 is regulating during the response to DSBs, including the networks PARP-1 is orchestrating to potentially help reshape the cell environment, to efficiently repair the most lethal lesion our genome faces.

Abstract:
Les poly(ADP-ribose) polymérases (PARPs/ARTDs) sont des senseurs des cassures de l’ADN et utilisent la nicotinamide adénine dinucléotide (NAD+) pour synthétiser un polymère de poly(ADP-ribose) (PAR) long et branché attaché aux résidus accepteurs des protéines nucléaires. Le travail présent montre que les protéines des mammifères PARP-1, PARP-2 et PARP-3 sont capables d’ADPribosyler les extrémités des duplex d’oligonucléotides in vitro. PARP-1 modifie préférentiellement les substrats resequés, alors que PARP-2 et PARP-3 modifient préférentiellement les substrats avec un nick, et sont capables d’ADP-ribosyler les groupements phosphate en 5’ ou 3’ au niveau des extrémités des cassures double brin des duplex d’ADN contenant à proximité un nick ou gap phosphorylé en 5’. Cet étude présente une caractérisationdétaillée des préférences de substrat d’ADN pour les enzymes structuralement proches PARP-2 et PARP-3, et propose un modèle putatif du mécanisme de l’ADP-ribosylation des extrémités de l’ADN catalysée par PARP-3 et PARP-2. L’ADPribosylationefficiente d’un fragment d’ADN d’environ 3kb par PARP-3 et PARP-2, l’ADPribosylationmédiée par les PARPs dans les extrait cellulaires ainsi qu’un signal persistant générée par l’anticorps anti-PAR sur de l’ADN génomique purifié en série à partir des cellules HeLa déplétées de PARG et traitées à la bléomycine suggèrent que certains types de cassures complexes de l’ADN peuvent être efficacement PARylés par les PARPs comme réponse cellulaire aux dommages de l’ADN. Cet nouveau type de modification postréplicative de l’ADN médiée par les PARPs in vitro apporte des nouveaux éléments sur les mécanismes moléculaires sous-jacents aux enzymes qui catalysent l’ADP-ribosylation<br>Poly(ADP-ribose) polymerases (PARPs/ARTDs) act as DNA break sensors and use nicotinamide adenine dinucleotide (NAD+) to catalyze the synthesis of a long branched poly(ADP-ribose) polymer (PAR) attached to the acceptor residues of nuclear proteins. Abstract:
Les mitochondries sont des organites essentiels pour la production d'énergie cellulaire grâce à la synthèse d'ATP au cours des étapes de phosphorylations oxydatives (OXPHOS). Les complexes de la chaine respiratoire sont en partie codés par le génome mitochondrial (ADNmt), dont la structure est très sensible aux facteurs exogènes ou endogènes. De nombreuses mutations de l'ADNmt sont associées à des dysfonctionnements de la chaine respiratoire conduisant à des pathologies. La production d'Espèces Oxygénées Réactives (EOR) mitochondriale est la principale source de dommages à l'ADNmt. Une voie de réparation particulière, le système de réparation par excision de bases (BER) est mis en oeuvre dans ce cas. Nous avons, au cours de notre étude, analysé le système BER mitochondrial chez la drosophile. Dans une première approche, nous avons caractérisé de manière globale par une technologie de puces à ADN un ensemble de glycosylases et endonucléases impliquées dans la voie BER mitochondriale et comparé leur variation au cours du vieillissement. Cette étude a été complétée par une analyse transcriptionnelle sur des modèles de drosophiles mutantes pour des enzymes spécifiques de la voie BER, ceci afin de déterminer les éventuelles interactions transcriptionnelles entre les acteurs de cette voie. L'ARNm de Parp présentait de fortes variations dans les différents contextes mutants testés. C'est une molécule essentielle de la réparation BER. Elle a fait l'objet dans un deuxième temps, d'une étude plus approfondie. Dans le modèle des cellules S2, PARP bien que majoritairement nucléaire est également présent dans la mitochondrie. Le comportement différentiel des deux variants ARNm de Parp a pu être mis en évidence lors de stress cellulaires. Les isoformes protéiques de PARP observées dans nos études apparaissent différentes de celles habituellement décrites dans la littérature. Cet aspect a été discuté.

Les cassures double-brin de l'ADN, lorsque incorrectement réparées, peuvent avoir des conséquences fatales telles que des délétions et des réarrangements chromosomiques, favorisant la carcinogenèse. La poly(ADP-ribosyl)ation réalisée par la protéine poly(ADP-ribose) polymérase-1 (PARP-1) est l'une des premières modifications post-traductionnelles qui se produisent en réponse aux dommages à l'ADN. La PARP-1 utilise la nicotinamide pour générer un polymère chargé négativement, nommé poly(ADP-ribose) polymère (PAR), lequel est attaché en majorité à la PARP-1 elle-même ainsi qu'à d'autres protéines cibles. Le PAR a récemment été reconnu comme un signal de recrutement pour certaines protéines de réparation aux sites de dommages à l'ADN, mais un débat est en cours quant au rôle précis de la PARP-1 et du PAR dans la réponse aux dommages de l'ADN. Au cours de mon projet de doctorat, nous avons pu confirmer que les protéines qui se retrouvent en complexe avec le PAR immédiatement après les dommages à l'ADN sont principalement des facteurs de réparation. Étonnamment, les complexes protéiques associés au PAR pendant la période de récupération suite aux dommages sont enrichis en facteurs de liaison à l'ARN. Toutefois, la protéine liant l'ARN la plus abondante que nous avons détectée dans l'interactome du PAR, soit NONO, ne suit pas cette dernière cinétique puisqu'elle est fortement enrichie immédiatement après les dommages à l'ADN. Notre étude subséquente de NONO dans la réponse aux cassures double-brin de l'ADN a étonnamment révélé une implication directe de celle-ci par le mécanismede réparation de jonction des extrémités non-homologues. En plus, nous avons constaté que NONO se lie fortement et spécifiquement au PAR via son motif 1 de la reconnaissance de l'ARN, soulignant la compétition entre les PAR et l'ARN pour le même site de liaison. Fait intéressant, le recrutement in vivo de NONO aux sites de dommages de l'ADN dépend entièrement du PAR et nécessite le motif 1 de la reconnaissance de l'ARN. En conclusion, nos résultats établissent NONO comme une nouvelle protéine impliquée dans la réponse aux cassures double-brin de l'ADN et plus généralement démontrent un autre niveau de complexité supplémentaire dans l'interdépendance de la biologie de l'ARN et la réparation de l'ADN.<br>DNA double-strand breaks are potentially lethal lesions, which if not repaired correctly, can have harmful consequences such as carcinogenesis promoted by chromosome deletions and rearrangements. Poly(ADP-ribosyl)ation carried out by poly(ADP-ribose) polymerase 1 (PARP-1) is one of the first posttranslational modifications occurring in response to DNA damage. In brief, PARP-1 uses nicotinamide to generate a negatively charged polymer called poly(ADP-ribose) polymer (PAR), that can be attached to acceptor proteins, which is to a large extent PARP-1 itself. PAR has recently been recognized as a recruitment signal for key DNA repair proteins to sites of DNA damage but the precise role of PARP-1 and its catalytic product PAR in the DNA damage response are still a matter of ongoing debate. Throughout my doctoral work, we confirmed that the proteins in complex with PAR promptly after DNA damage are mostly DNA repair proteins, whereas during the period of recovery from DNA damage, the PAR interactome is highly enriched with RNA processing factors. Interestingly, one of the most abundant RNA-binding proteins detected in the PAR interactome, namely NONO, did not follow these kinetics as it was highly enriched immediately after DNA damage in the DNA repair protein complexes centered on PAR. Our subsequent investigation of NONO in the DNA damage response to double-strand breaks strikingly revealed a direct implication for NONO in repair by nonhomologous end joining (NHEJ). Moreover, we found that NONO strongly and specifically binds to PAR through its RNA-recognition motif 1 (RRM1), highlighting competition between PAR and RNA for the same binding site. Remarkably, the in vivo recruitment of NONO to DNA damage sites completely depends on PAR and requires the RRM1 motif. In conclusion, our results establish NONO as a new protein implicated in the DNA damage response to double-strand break and in broader terms add another layer of complexity to the cross-talk between RNA-biology and DNA repair.

La poly(adp-ribos) polymerase (parp) est une proteine nucleaire qui utilise la nad+ comme substrat. Pleinement activee par les coupures de l'adn, elle peut provoquer la mort cellulaire, par privation d'energie. Les mecanismes sous-jacents a la neurodegenerescence ne sont pas totalement compris et pourraient inclure l'activation de la parp. Resultats : la presence de parp et de son arnm dans les populations neuronales selectivement vulnerables aux atteintes du cerveau, suggere son implication dans la neurodegenerescence. Soumises a une concentration toxique de glutamate (glu) les cellules granulaires du cervelet in vitro ont montre une activation rapide de la parp. Des inhibiteurs de la parp ont prevenu la mort des neurones dans une proportion de 60 a 100%. L'afflux de ca#2#+ dans les cellules, provoque par le glu n'a pas ete modifie par la benzamide. Les traitements avec 5 inhibiteurs de la parp, dont la benzamide, ont empeche les depletions de dopamine striatale et de noradrenaline corticale, provoquees par la mptp chez la souris c57bi/6n, 7 jours apres le traitement. La benzamide, est presente, a basses concentrations millimolaires, dans le striatum 30 et 120 minutes apres une unique injection i. P. Chez les animaux naifs, les inhibiteurs de la parp n'ont pas, d'eux-memes, modifie les niveaux de metabolites de la dopamine a 7 jours apres l'injection du produit, la benzamide et ses derives amines ne montrent pas d'effet sur le metabolisme de la dopamine. L'administration de benzamide, a attenue partiellement les depletions de dopamine induites par methamphetamine (meth) chez la souris c57bi/6n. La benzamide n'a pas fait baisser la temperature corporelle. Ces resultats indiquent un role de la parp dans les neurotoxicites du glu, de la mptp de la meth. Et une activite neuroprotectrice de ses inhibiteurs dans differents modeles de neurodegenerescence, permettant d'envisager une nouvelle cible pour la decouverte de medicaments potentiels pour le traitement des maladies neurodegeneratives.

The study aims to analyze how customer engagement can be increased in digital marketing campaigns to improve connections with customers. To achieve this aim, empirical research was conducted to get valuable insights into factors that contribute the most to increasing engagement in digital marketing campaigns. The study employed a qualitative research method, and 10 semi-structured interviews were conducted with potential digital marketing campaigns customers dividing them into two age groups of A20-30 and A40-50. The research showed that engagement tactics enhance the performance of digital marketing campaigns and substantially influence users' willingness to interact with digital ads actively. Relevance, clear message, aesthetic design, personalized content, and informativeness have the greatest impact on shaping user behavior and starting active engagement. Taking research results into consideration, managerial recommendations were provided using the RACE framework regarding each phase of the engagement: plan, reach, act, convert, and engage.

Abstract Viruses and the hosts they infect are involved in an evolutionary battle. The host has physical barriers that protect itself from viruses. Infected cells can commit suicide to preclude virus replication. Virus-infected cells are destroyed by the innate and adaptive arms of the immune system, and virions are eliminated by specific antibodies. Viruses, in tum, have mechanisms that allow the virus to infect cells and survive attack by the host. Virus proteins inhibit intrinsic cellular apoptosis, and block killing of infected cells by cytokines and cells of the immune system. These viral proteins are interesting tools to understand virus pathogenesis, cellular apoptosis, and the immune response. They also provide novel insights into many key regulatory aspects of molecular and cellular biology. In this article we describe human adenovirus (Ad) proteins that counteract host antiviral responses. Aspects of this topic have been reviewed elsewhere (1–5). There are 51 serotypes of human Ads, which fall into six subgroups, A–F. We will limit our discussion to the commonly studied serotypes 5 (Ad5) and Ad2, members of subgroup C.

According to the participants in this study, Google Ads campaign profitability depends, largely, on the ability to choose a keyword pool that achieves the company's goals. Moreover, the complexity of these pay-per-click decisions, the costs involved, and its business implications demand more reasoned, quantified, and, if possible, optimized solutions. The chapter develops linear programming optimization models based on impressions, clicks, conversions, and billing. The models are tested on a real example using Excel optimization add-ins.

Some assistive devices (ADs) aim to ease elders performing daily activities by changing postural and strength requirements. Elders usually have cognitive lacks too, making AD usability perception an important issue. Usability perceptions arise from the affordances conveyed by the AD: design features should act as signifiers transmitting opportunities of how to use the AD. This study assesses the perception of jar openers usability. Eye-tracking (ET) data from 56 subjects were used. Rendered images of 6 ADs were shown, in 2 versions: with and without rubber on the grip area. Each slide showed the 6 ADs, each AD image being an area of interest (AoI) to be tracked. The participants ranked the ADs in the next usability affordances: comfort, effort level, easiness to grip, easiness to use, lid slippery and robustness. For each affordance, they were eye-tracked while deciding the best AD, and afterwards they ranked the other ADs. The ranks were transformed into scores, and their correlation with ET parameters (decision time and number of fixations on each AoI) were studied, along with the effect of the rubber on the scores. Heat maps were also analyzed to identify the signifiers of the ADs that attract attention on usability. The results showed the potentiality of ET to study the perception of ADs usability affordances, and that the addition of rubber on the grip area of the jar openers or the material and shape of the lid area have an effect on their understanding and use.

The thermooptic coefficients α n = n−1dn/dT and the thermal expansion coefficients α L = L−1dL/dT were measured for thirteen of these isomorphs (KDP, KD*P, ADP, AD*P, CDA, CD*A, ADA, AD*A, RDP, RD*P, RDA, RD*A, KDA). A double interferometric technique using Fizeau and Twyman- Green interferometers was developed to measure these thermal coefficients. As a sample was heated, fringes from each interferometer shifted as a result of the changing optical path length in the sample. Even though both fringe patterns were functions of α L and α n , the two patterns changed with temperature at different rates. α L and α n were determined from the measurement of these two fringe rates by the simultaneous solution of the two equations that describe the phase shifts for the two interferometers. The dispersion in α n was measured for all thirteen isomorphs between 441 and 1152 nm. Data for the KDP crystal were fit to a theoretical prediction based on a two-resonance Sellmeier fit of the refractive index.

Abstract:
Forskolin stimulates adenylate cyclase by interacting with the catalytic subunit and inhibits platelet aggregation. This inhibition is greatly potentiated by adenosine (Ado) which stimulates adenylate cyclase through membrane-bound Ado receptors. Forskolin is 2-4 fold more potent as an inhibitor of collagen-induced rat platelet aggregation as compared to human platelets (IC50 values, in rat PRP, 0.5-0.8 μM; in human PRP, 1.5-2 μM). However, if the blood is pretreated with adenosine deaminase (ADA), an enzyme that degrades Ado to inosine, the inhibitory action of forskolin is greatly reduced producing similar effects both in human and rat PRPs (IC50, 2-3 μM) and whole blood (IC50, 4.6 μM). Both 5’-methylthioadenosine (MTA, 50-100 μM), an antagonist of Ado receptors, and 2’,5’-dideoxyadenosine (DDA, 100 μM), an inhibitor of adenylate cyclase, reverse the inhibition of platelet aggregation in rat PRP, whereas, no reversal is seen in human PRP. When Ado in the rat plasma is degraded by ADA pretreatment, DDA or MTA shows no reversal as seen in human PRP. The inhibitory action of forskolin (1-2 μM), which is only weakly inhibitory alone (&lt;20%) in human whole blood, can be greatly potentiated (100% inhibition) by the inhibitors of nucleoside transport, dipyridamole (10 μM) or dilazep (2 μM). Only slight potentiation is seen in rat whole blood suggesting that rat plasma Ado levels are not affected significantly perhaps due to weakly active erythrocytic nucleoside transport system. Sato and Ui (In: Physiology and Pharmacology of Adenosine, Daly et al, Eds. Raven Press, 1983, 1-11), have shown that rat plasma contains much higher Ado levels (7.55 ± 0.51 pM) than human plasma (0.29 ± 0.08 μM). These studies demonstrate that plasma adenosine plays an important role in the forskolin antiplatelet activity which can be greatly potentiated in human whole blood by the clinically used drugs, dipyridamole and dilazep. We studied the biochemical composition and nutritive value of the green mass and prepared hay from local ecotipe of common knotgrass Polygonum aviculare grown in monoculture on the experimental plot of the National Botanical Garden (Institute) “Alexandru Ciubotaru”, Chisinau, R. Moldova. Results revealed that dry matter of whole plant contained 173 g/kg CP, 293 g/kg CF, 84g/kg ash, 327g/kg ADF, 509 g/kg NDF, 55 g/kg ADL, 272g/kg Cel, 182 g/kg HC, 77 g/kg TSS, with nutritive and energy value 59.2% DMD, 51.7% ODM, RFV=116, 10.25 MJ/kg ME and 6.27MJ/kg NEl. The biochemical composition, nutritive and energy value of prepared hay from common knotgrass: 163 g/kg CP, 314 g/kg CF, 93 g/kg ash, 335 g/kg ADF, 491g/kg NDF, 52 g/kg ADL, 283g/kg Cel, 156 g/kg HC, 76 g/kg TSS, RFV=119, 10.16 MJ/kg ME and 6.18-6.54 MJ/kg NEl.

Background: Alzheimer’s disease (AD) is characterized by a deposition of β-amyloid peptide and the neurofibrillary tangles of tau protein. Resveratrol is a neuroprotective agent, acting in the prevention of redox impairment, and could reduce neuronal damage in patients with AD. Objectives: Systematic review and meta-analysis about the effects of resveratrol supplementation on cognitive and functional performance in patients with AD. Methods: Databases were searched for primary studies that reported cognitive and functional performance based on ADAS-cog, ADCS-ADL or MMSE instruments in AD patients treated with resveratrol. Primary studies published up to May 2021 and without language and publication date restrictions were included. The measure of effect of the meta-analysis was presented as weighted mean difference (WMD). decrease in ADAS-cog scores [WMD: -3.69 points], and significant increases in ADCS-ADL [WMD: 5.65 points] and MMSE scores [WMD: 2.03 points] in the resveratrol intervention group, when compared to the placebo group. Conclusions: Resveratrol supplementation may result in improving cognitive and functional performance in AD patients.

Abstract:
People with visual impairments suffer from the incapacity to understand contextual information in videos, such as the place where characters are, or any other non-spoken actions in general. Some content creators address this issue by providing a secondary audio to describe such information, called Audio Descriptions (ADs). How- ever, some works in the literature have highlighted that people with visual impairment are usually not able to completely understand scene changes based only on characters’ voices or traditional ADs. Moreover, traditional ADs do not completely describe some of the important visual information, such as the background scenery (e.g. colors, furniture) and characters’ details (e.g. blond woman using a red dress). In this work, we propose incrementing the traditional AD techniques with the usage of interactive video features present in TV systems. More precisely, the proposed interactivity enables users to access specialized AD for different visual information (e.g., scene, scenario, character). To support the development of such interactive content, we present an application template, which helps to create the final interactive-enhanced video application. Asa proof of concept for our approach, we created an interactive AD for an independent video mainly composed of visual information, with only a few talks.

"Social advertisements with threat appeals are widely used to reduce drunk driving. However, research on the effectiveness of such advertising is limited. This study aimed to evaluate, what emotions cause threat appeal ads targeting drunk driving and whether these ads change risky driving attitudes. 41 students (17.1 percent males; mean age 20.9 years; 53.7 percent had a driving license) voluntarily participated in the experimental study. Every participant was randomly assigned to one of three groups: two experimental (watched one of two TV ads with threat appeals) or control group (watched car wash ad with no threatening stimuli). After watching one of three ads, all participants were asked to evaluate seven emotions and to fill in Driving Attitude Questionnaire (DAQ). Results revealed that both road safety threat appeal ads targeting drunk driving did not arouse any stronger fear emotions or differences in driving attitudes compared to control group. Both experimental groups didn’t differ in emotions or attitudes as well. No difference in emotions and attitudes was found when comparing the reaction of participants who have seen the specific ad before the experiment and those who haven’t. The fact of being a licensed driver was also not related to the level of reported emotions or attitude differences in both experimental groups. The study results reveal that the possible effectiveness of threat appeal ads from ongoing social marketing campaigns on reducing drunk driving is questionable and further studies are needed."

We used eye tracking to measure visual attention to tobacco products and pro- and anti-tobacco advertisements (pro-ads and anti-ads) during a shopping task in a three-dimensional virtual convenience store. We used eye-tracking hardware to track the percentage of fixations (number of times the eye was essentially stationary; F) and dwell time (time spent looking at an object; DT) for several categories of objects and ads for 30 adult current cigarette smokers. We used Wald F-tests to compare fixations and dwell time across categories, adjusting comparisons of ads by the number of each type of ad. Overall, unadjusted for the number of each object, participants focused significantly greater attention on snacks and drinks and tobacco products than ads (all P&lt;0.005). Adjusting for the number of each type of ad viewed, participants devoted significantly greater visual attention to pro-ads than anti-ads or ads unrelated to tobacco (P&lt;0.001). Visual attention for anti-ads was significantly greater when the ads were placed on the store’s external walls or hung from the ceiling than when placed on the gas pump or floor (P&lt;0.005). In a cluttered convenience store environment, anti-ads at the point of sale have to compete with many other stimuli. Restrictions on tobacco product displays and advertisements at the point of sale could reduce the stimuli that attract smokers’ attention away from anti-ads.
