Abstract:
Abstract Metalloproteases of the A Disintegrin And Metalproteases (ADAM) family are important molecular mediators of inflammation. ADAMs participate at all stages of inflammation via the proteolytic cleavage of cell surface molecules. It has been shown that leukocyte expressed ADAM8 and ADAM17 are important during the initial rolling of leukocytes on the endothelium. However, the role of leukocyte expressed ADAMs in transendothelial migration and chemotaxis remains to be investigated and in particular it is still unclear to what extent ADAM10 and ADAM9 contribute to leukocyte migration. In this study we show that treatment of leukocytes with a pharmaceutical ADAM10 inhibitor reduced both the in vitro transendothelial migration and the chemotaxis of the monocytic cell line THP1. The combined pharmaceutical inhibition of both ADAM10 and ADAM17 did not further suppress leukocyte migration in either experimental setting. The subsequent analysis of THP1 cells with a gene silencing of ADAM8, 9, 10 or 17 in transendothelial migration and chemotaxis experiments revealed that ADAM8 and ADAM10 are critically involved in THP1 cell migration. While ADAM9 did not seem to be required ADAM17 was only ofminor importance for the migration of THP1 cells. This study analyses the role of the four proteases ADAM8, 9, 10 and 17 in the migration of leukocytic cells. It shows that ADAM8 and ADAM10 are crucial for the in vitro transendothelial migration and chemotaxis of the monocytic cell line THP1.

Abstract:
Background The functional significance of ADAMs family members in the immune infiltration of pancreatic adenocarcinoma (PAAD) awaits elucidation. Methods ADAMs family members with significant expression were identified among differentially expressed genes of PAAD based on The Cancer Genome Atlas (TCGA) database followed by a verification based on the Oncomine database. The correlation of ADAMs in PAAD was estimated with the Spearman’s rho value. The pathway enrichment of ADAMs was performed by STRING and GSEALite, respectively. The protein–protein interaction and Gene Ontology analyses of ADAMs and their similar genes were exanimated in STRING and visualized by Cytoscape. Subsequently, the Box-Whisker plot was used to show a correlation between ADAMs and different tumor grade 1/2/3/4 with Student’s t-test. TIMER was applied to estimate a correlation of ADAMs expressions with immune infiltrates and immune checkpoint blockade (ICB) immunotherapy-related molecules. Furthermore, the effect of copy number variation (CNV) of ADAMs genes was assessed on the immune infiltration levels. Result ADAM8/9/10/12/15/19/28/TS2/TS12 were over-expressed in PAAD. Most of the nine ADAMs had a significant correlation. ADAM8/12/15/19 expression was remarkably increased in the comparison between grade 1 and grade 2/3 of PAAD. ADAM8/9/10/12/19/28/TS2/TS12 had a positive correlation with almost five immune infiltrates. ADAM12/19/TS2/TS12 dramatically related with ICB immunotherapy-related molecules. CNV of ADAMs genes potentially influenced the immune infiltration levels. Conclusion Knowledge of the expression level of the ADAMs family could provide a reasonable strategy for improved immunotherapies to PAAD.

This study has been designed to analyse the secreted and soluble proteins in bovine seminal plasma by mass spectrometry. The majority of these proteins are produced by accessory glands, and partially by testis, epididymis, ductus deferens and vas deferens of male reproductive tract.
 Seminal plasma of bovine was collected freshly and isolated after centrifuged and removed the sperm. Non boiled and boiled seminal plasma lysate were run to identify and detect the total proteins, as well as the individual single member of ADAMs protein is determined.
 The non-boiled lysate of seminal plasma was displayed diversity of proteins more than boiled sample lysate of seminal plasma.
 However, boiled and non-boiled seminal plasma have distinguished several types of ADAMs protein which are included: ADAM10, ADAM9, ADAM7, ADAM15 in non-boiled samples, while boiled lysate of bovine seminal plasma was displayed ADAM10, ADAM9, ADAM28, and ADAM22.
 Our finding concluded that bovine seminal plasma is very rich in different types of soluble, cleaved and shed proteins such ADAMs protein which could potentially have a biological and physiological role in protection and interactions of sperm during motility inside female reproductive tract, as it might support it to fertilise the ovum. This protection might be via immunosuppression behaviour or by block specific receptors in female reproductive tract for enhancing sperm motility, and avoid sperm the singling response of immunity.

Abstract:
Despite the considerable advances in diagnostic methods in medicine, central nervous system (CNS) tumors, particularly the most common ones—gliomas—remain incurable, with similar incidence rates and mortality. A growing body of literature has revealed that degradation of the extracellular matrix by matrix metalloproteinases (MMPs) might be involved in the pathogenesis of CNS tumors. However, the subfamily of MMPs, known as disintegrin and metalloproteinase (ADAM) proteins are unique due to both adhesive and proteolytic activities. The objective of our review is to present the role of ADAMs in CNS tumors, particularly their involvement in the development of malignant gliomas. Moreover, we focus on the diagnostic and prognostic significance of selected ADAMs in patients with these neoplasms. It has been proven that ADAM12, ADAMTS4 and 5 are implicated in the proliferation and invasion of glioma cells. In addition, ADAM8 and ADAM19 are correlated with the invasive activity of glioma cells and unfavorable survival, while ADAM9, -10 and -17 are associated with tumor grade and histological type of gliomas and can be used as prognostic factors. In conclusion, several ADAMs might serve as potential diagnostic and prognostic biomarkers as well as therapeutic targets for malignant CNS tumors. However, future research on ADAMs biology should be performed to elucidate new strategies for tumor diagnosis and treatment of patients with these malignancies.

Abstract:
Objectives: ADAMTS (A Disintegrin And Metalloproteinase with ThromboSpondin repeats) proteinases, which are released outside the cell (soluble) have very critical roles in damage and repair of extracellular matrix (ECM) processes. Our aim was to analyse the ADAMTS-1 and ADAMTS-9 which were the member of the ADAMTS gene family of metalloproteinases that might had been involved in the cytokines-mediated etiopathogenesis of endometriosis. Methods: A case-control study was performed in an university hospital. Thirty-four patient with endometriosis which was defined via laparoscopy and thirty-three healthy female volunteers were recruited in the present study. Serum ADAMTS-1 and ADAMTS-9 and IL-beta (IL-1 β) and vascular endothelial growth factor (VEGF) levels were determined by a human enzyme-linked immunoassay (ELISA) in all subjects. Results: The demographic characteristics of the patients were significantly higher than healthy control group. The IL-1 β and VEGF levels were significantly higher; ADAMTS-1 and ADAMTS-9 levels were significantly lower in the endometriosis patients compared to the controls. We also found a negative correlation between ADAMTS-1, ADAMTS-9, and IL-1 β, VEGF. Conclusion: The results of the study might suggest that ADAMS-1 and ADAMTS-9 have a role in the pathogenesis of the endometriosis.

Abstract:
Matrix metalloproteinases (MMPs) are M2 macrophage markers that are modulated by inflammation. A disintegrin and metalloproteinases (ADAMS) and those with thrombospondin motifs (ADAMTS) regulate the shedding of membrane-bound proteins, growth factors, cytokines, ligands, and receptors; MMPs, ADAMS, and ADAMTS may be regulated by tissue inhibitors of metalloproteinases (TIMPs). This study aimed to determine whether these interacting proteins were dysregulated in PCOS. A Somascan proteomic analysis of 12 MMPs, three of their inhibitors (TIMP-1, 2, 3), two ADAMS (9, 12), five ADAMTS (1, 4, 5, 13, 15), insulin-like growth factor binding protein-1 (IGFBP-1), and insulin-like growth factor-1 (IGF-1) was undertaken in a well-validated PCOS database of 143 women with PCOS and 97 controls. Women with PCOS had significantly higher levels of MMP-9 and lower levels of MMP-2, MMP-14, TIMP-2, IGFBP-1, and IGF-1 compared to the controls (p &lt; 0.0001, p &lt; 0.005, p &lt; 0.04, p &lt; 0.05, p &lt; 0.0001, and p &lt; 0.0001, respectively). No differences were observed for any other MMPs. The ADAMS or ADAMTS levels did not differ between groups. Body mass index (BMI) was correlated with MMP-9 (p &lt; 0.01), MMP-1 (p &lt; 0.05), MMP-2 (p &lt; 0.05), MMP-10 (p &lt; 0.005), MMP-12 (p &lt; 0.005), ADAM-9 (p &lt; 0.05), and IGFBP-1 (p &lt; 0.0001), but only MMP-9 still differed after accounting for BMI. MMP-9/TIMP-1, MMP-9/TIMP-2, and MMP-9/TIMP-3 ratios were higher in the PCOS group (p &lt; 0.01), whilst MMP-17/TIMP-1 and MMP-17/TIMP-2 were lower (p = 0.01). MMP-2/TIMP ratios showed no difference between groups. TIMP-2 was positively correlated with CRP (p &lt; 0.01). MMP changes in PCOS are largely driven by BMI, though increased MMP-9 is BMI-independent, suggesting that any deleterious effects of MMP-9 would be potentially exacerbated by a concomitantly increased BMI. The significant increases in the MMP-9/TIMP ratios suggests MMP-9 overactivity in PCOS.

Abstract:
Extracellular matrix (ECM) in the vascular wall is a highly dynamic structure composed of a set of different molecules such as elastins, collagens, fibronectin (Fn), laminins, proteoglycans, and polysaccharides. ECM undergoes remodeling processes to regulate vascular smooth muscle and endothelial cells’ proliferation, differentiation, and adhesion. Abnormalities affecting the ECM can lead to alteration in cellular behavior and from this, this can conduce to the development of pathologies. Metalloproteases play a key role in maintaining the homeostasis of ECM by mediating the cleavage of different ECM components. There are different types of metalloproteases: matrix metalloproteinases (MMPs), disintegrin and metalloproteinases (ADAMs), and ADAMs with thrombospondin motifs (ADAMTSs). ADAMTSs have been found to participate in cardiovascular physiology and diseases and specifically in aortic aneurysms. This review aims to decipher the potential role of ADAMTS proteins in the physiopathologic development of Thoracic Aortic Aneurysms (TAA) and Abdominal Aortic Aneurysms (AAA). This review will focus on what is known on the ADAMTS family involved in human aneurysms from human tissues to mouse models. The recent findings on THSD4 (encoding ADAMTSL6) mutations in TAA give a new insight on the involvement of the ADAMTS family in TAA.

This thesis contains new results about the representability of cohomological functors defined on a subcategory of compact objects (with respect to a fixed cardinal) of a well generated triangulated category. Classical theorems of Adams for the stable homotopy category and Neeman for compactly generated triangulated categories are extended to the first uncountable cardinal. The case of derived categories of rings and the stable motivic category are studied in detail. These results contribute to answering negatively a question raised by Rosický of whether all cohomological functors defined on a subcategory of compact objects with respect to a large enough cardinal are representable. Some of the findings in this thesis are based on new results about abelian categories, the most relevant being a generalization of the Auslander Lemma for non Grothendieck categories.<br><I>TESI "Representabilitat d'Adams en categories triangulades"<br/><br/>TEXT:<br/><br/>En aquesta tesi s'obtenen resultats nous sobre la representabilitat de functors cohomològics definits en subcategories d'objectes compactes (respecte a un cardinal fixat) d'una categoria triangulada ben generada. S'estenen al primer cardinal no numerable teoremes antics d'Adams per a la categoria d'homotopia estable i de Neeman per a categories compactament generades. Abstract:
This dissertation explores the extent to which the political ideology that formed the basis for the American republic shaped American diplomacy, using John Adams, James Madison and John Quincy Adams as case studies. American statesmen drew on a variety of sources for republican principles of diplomacy. The law of nations and the Scottish political economists supplied the ideas of an international balance of power and freedom of trade. English writers of the Opposition Whig school provided concepts such as political separation from Europe, reliance on a navy for defense, abhorrence of a standing army and, indirectly, the belief that the United States could use its economic power to secure its diplomatic goals.;John Adams began his career with a high degree of confidence in the virtue of the American people and the coercive power of American trade. He combined a classical martial ethic with an Opposition whig strategic sense. Adams's experience in Europe disproved these beliefs, and as president he fell back on the republican realpolitik, based on naval power and separation from Europe, suggested by the Opposition Whig school.;James Madison never held out a classical model of virtue and never lost faith in the coercive power of American commerce. His combination of political economy with Opposition thought led him to reject both an army and a navy as monarchical tools of diplomacy. He saw the Constitution as a vehicle for harnessing American economic power. Madison's conception of a republican diplomacy led him, as secretary of state and president, to rely on the Embargo and similar economic measures.;John Quincy Adams combined republican realpolitik with a sense of Christian purpose and saw American government and diplomacy as a vehicle for moral improvement. Adams's republic rested on a continental union and a diplomacy directed against European colonization, as a manifestation of monarchy. Non-colonization included removing Spain as a neighbor in North America, preventing European political encroachment in the Western Hemisphere, and securing a hemisphere-wide consensus on neutral rights. as a congressman and critic of slavery-driven expansion, Adams demonstrated the persistence of Opposition Whig thought in American politics.

Abstract The Hal Adams Bridge is the only suspension bridge in the state of Florida. During the 2001 routine bridge inspection the bridge was identified as a candidate to be repainted. The authors Don Buwalda, Stephen Haney, and Greg Richards participated in the assessment, plan development, and project execution. This paper identifies areas that made the Hal Adams Bridge: “A Challenging Repainting Project on a Unique Florida Bridge.” A history of actual experiences and lessons learned will be presented from the point of view of the owner and third party inspection firms.

Review question / Objective: 1. In Lance-Adams syndrome, what is the effect of current therapeutic management on improving patients' condition compared with the control group? 2. Are EEG, Brain CT, MRI, and brain SPECT more accurate in diagnosing Lance-Adams syndrome? 3. Does Early diagnosis and treatment influence the quality of life in patients with Lance-Adams syndrome? 4. Are patients with abnormal cortical discharge or cerebellum brain stem and thalamus cortical circuit or neurotransmitter imbalance at higher risk for/of Lance-Adams syndrome compared with patients without these symptoms? Condition being studied: LAS is a group of clinical symptoms; The primary manifestation is action myoclonus which can occur as generalized, focal, or multifocal repeated myoclonic motor movement myoclonus. In some patients, sensory stimuli can trigger myoclonus. Furthermore, negative myoclonus can impair posture and cause falls in the lower extremities.

ในปัจจุบันงานวิจัยที่เกี่ยวข้องกับการวิเคราะห์พลศาสตร์ของข้อต่อของร่างกายมนุษย์ขณะที่กำลังทำกิจกรรมอยู่นั้นต้องการข้อมูลการเคลื่อนไหวของร่างกาย ซึ่งระบบการเก็บข้อมูลการเคลื่อนไหวที่มีขายในเชิงการค้ามีราคาสูงมาก งานวิจัยนี้จึงศึกษาและวิเคราะห์พลศาสตร์ของข้อต่อจากการเคลื่อนไหวของร่างกายท่อนล่างโดยใช้เทคนิคการประมวลผลภาพเข้าช่วย โดยการเก็บภาพด้วยกล้องวิดีโอพร้อมทั้งติดมาร์กเกอร์ซึ่งเป็นวิธีที่สะดวกและมีราคาไม่แพง ทำการเก็บข้อมูลการเคลื่อนไหวของร่างกายท่อนล่างของคนในขณะกำลังเดิน ด้วยกล้องวิดีโอจำนวน 2 กล้อง โดยติดมาร์กเกอร์จำนวน 14 จุดที่ผิวหนังเพื่อใช้ระบุตำแหน่งพิกัด ภาพที่ได้จากกล้องวิดีโอจะเป็นภาพของร่างกายฝั่งซ้ายและขวาใน 2 มิติตามแนว Sagittal Plane ภาพที่ได้จะถูกนำไปประมวลผลภาพด้วยโปรแกรม MATLAB เพื่อหาตำแหน่งพิกัดต่างๆ ของมาร์กเกอร์ที่ติดบนร่างกาย ซึ่งจำเป็นต่อการนำไปจำลองการเคลื่อนไหวด้วยโปรแกรม Adams-LifeMOD ซึ่งเป็นโปรแกรมที่ใช้สำหรับวิเคราะห์ระบบทางกลศาสตร์ของร่างกายมนุษย์ โมเดลที่จำลองนั้นได้จำลองให้มีลักษณะใกล้คียงกับความเป็นจริงโดยสามารถกำหนดพารามิเตอร์ เช่น น้ำหนักและส่วนสูง ค่าตำแหน่งของมาร์กเกอร์ที่ได้จากการประมวลผลภาพจะเป็นตัวกำหนดการเคลื่อนไหวของโมเดล เมื่อนำข้อมูลการเคลื่อนที่และพารามิเตอร์ของร่างกายมาทำการวิเคราะห์ด้วยโปรแกรม Adams-LifeMOD แล้วพบว่าสามารถหาค่าพารามิเตอร์ทางพลศาสตร์ที่ข้อเท้า ข้อเข่า และข้อสะโพกในขณะเวลาต่างๆ เช่น ความเร็วแรงบิด หรือโมเมนต์ได้ จากงานวิจัยนี้ทำให้ได้ข้อมูลที่จำเป็นและเป็นประโยชน์อย่างยิ่งต่อการออกแบบข้อเทียม รวมถึงการพัฒนาศักยภาพในการรักษากายภาพบำบัด และวงการวิทยาศาสตร์กีฬาในอนาคต
