Abstract:
The analysis of adaptors’ deformation and stress is an important prerequisite for the design of the adapters. This paper proposed the mathematical description and analytical method for the sponge cylinder and rubber cylinder under pressure. The analytical formulas on deformation and stress of interference fit between adapters and launchers are deduced. Also, the correctness of the analytical formula is verified by numerical simulation. Furthermore, the effect of precompression and the sponge’s thickness ratio on adapter’s deformation and stress is studied. The following results can be concluded: the results of numerical simulation are basically consistent with that of analytical formula. After installing the adapter into the launcher, the rubber layer will move inward as a whole, then the sponge layer and the rubber layer are subjected to radial and tangential compressive stress. When the precompression is smaller than 0.0075 or larger than 0.0015, the deformation and stress of adapter are approximately in proportion to precompression. If the precompression keeps constant, the deformation and stress of adapter will have extremum when the thickness ratio of sponge equals to a certain value.

Abstract:
In the background of a container-type vertical launch missile, the simulation method of adapter separation in different wind speeds is researched. Based on force analysis of the adapters during their separation from the missile, the dynamic and kinematics equations of the adapter separation are established. The adapter’s aerodynamic parameters at different attitudes getting from the numerical wind tunnel are chosen to be the input. Through the dynamic simulation of the separation process of the adapters, the simulation results are in good agreement with the experimental data. The trajectory and placement distribution of adapters are obtained during the analysis of force and motion stance at different wind speeds. Then the relative distances between the adapter and missile or launch facility are determined. At the same time, it can be estimated that the combined calculation will save about two-thirds of time compared with dynamic grid method computing, which provides a significant guidance for the simulation method of adapter separation.

Abstract:
Molecular ecologists frequently use genome reduction strategies that rely upon restriction enzyme digestion of genomic DNA to sample consistent portions of the genome from many individuals (e.g., RADseq, GBS). However, researchers often find the existing methods expensive to initiate and/or difficult to implement consistently, especially because it is difficult to multiplex sufficient numbers of samples to fill entire sequencing lanes. Here, we introduce a low-cost and highly robust approach for the construction of dual-digest RADseq libraries that build on adapters and primers designed in Adapterama I. Major features of our method include: (1) minimizing the number of processing steps; (2) focusing on a single strand of sample DNA for library construction, allowing the use of a non-phosphorylated adapter on one end; (3) ligating adapters in the presence of active restriction enzymes, thereby reducing chimeras; (4) including an optional third restriction enzyme to cut apart adapter-dimers formed by the phosphorylated adapter, thus increasing the efficiency of adapter ligation to sample DNA, which is particularly effective when only low quantity/quality DNA samples are available; (5) interchangeable adapter designs; (6) incorporating variable-length internal indexes within the adapters to increase the scope of sample indexing, facilitate pooling, and increase sequence diversity; (7) maintaining compatibility with universal dual-indexed primers and thus, Illumina sequencing reagents and libraries; and, (8) easy modification for the identification of PCR duplicates. We present eight adapter designs that work with 72 restriction enzyme combinations. We demonstrate the efficiency of our approach by comparing it with existing methods, and we validate its utility through the discovery of many variable loci in a variety of non-model organisms. Our 2RAD/3RAD method is easy to perform, has low startup costs, has increased utility with low-concentration input DNA, and produces libraries that can be highly-multiplexed and pooled with other Illumina libraries.

Abstract:
Cell culture medium replacement is necessary to replenish nutrients and remove waste products, and perfusion and batch media exchange methods are available. The former can establish an environment similar to that in vivo, and microfluidic devices are frequently used. However, these methods are hampered by incompatibility with commercially available circular culture dishes and the difficulty in controlling liquid flow. Here, we fabricated a culture dish adapter using polydimethylsiloxane that has a small recess structure for flow control compatible with commercially available culture dishes. We designed U-shaped and I-shaped recess structure adapters and we examined the effects of groove structure on medium flow using simulation. We found that the U-shaped and I-shaped structures allowed a uniform and uneven flow of medium, respectively. We then applied these adaptors to 293T cell culture and examined the effects of recess structures on cell proliferation. As expected, cell proliferation was similar in each area of a dish in the U-shaped structure adapter, whereas in the early flow area in the I-shaped structure adapter, it was significantly higher. In summary, we succeeded in controlling liquid flow in culture dishes with the fabricated adapter, as well as in applying the modulation of culture medium flow to control cell culture.

Abstract:
The possibility of increasing the service life of 18-100 three-piece bogies by using taper bearing units with different types of adapters is considered in the article. The main task is the need to increase the longevity of the running parts of freight cars by reducing their dynamic loading. The solution of this problem is possible by improving the axle boxes. To determine possible design solutions, an analysis was made of modern domestic and foreign designs of bogies using bearing assemblies with adapters and three types of adapters with the highest quality indicators, namely the half-box of the 18-7020 bogie, adapter with elastic packing and adapter of the 18-4129 bogie. Three models of adapters were investigated using the finite-elements method, which, in accordance with reported manufacturer’s data, have high reliability index and can provide equal load distribution between bearing systems rollers. The results of mathematical simulation suggest that the forces applied to the bearing systems rollers largely depend on the adapter design. Furthermore, the studied adapter designs are not acceptable and require further designing.

Abstract:
Background:Shear stress at the stump in trans-tibial amputees induced by turning movements may be reduced with the use of torsion adapters in the prosthesis. Objective:Monitoring the motion and kinetic effects of a regular torsion adapter in comparison to a rigid placebo in unplanned spin and step turns.Study design:Single-blinded placebo-controlled cohort study. Methods:In total, 10 trans-tibial amputees underwent three-dimensional gait analysis in level walking and unplanned spin and step turns with a torsion adapter and with a rigid placebo. Results:Kinetic effects varied among participants. No statistically significant reduction of peak internal and peak external transverse plane moments was found for the torsion adapter in any walking condition. However, transverse plane rotation of the adapter was monitored in all participants. Conclusion:Motion between the socket and the residual limb may be reduced during turns due to transverse plane motion of the torsion adapter and shear stress on the residual limb may be reduced. However, there may be good and bad responders to torsion adapters due to differences in coupling between the residual limb and the socket.Clinical relevanceStrong coupling between stump and socket will help the user controlling his prosthesis. Shear stress at the stump may increase in stump–socket interface stiffness and may be the reason for residual limb problems. Torsion adapters therefore may be beneficial for comfort and stump condition in individual cases.

Abstract:
Multiplexed cloning of long DNA sequences is a valuable technique in many biotechnology applications, such as long-read genome sequencing and the creation of open reading frame (ORF) libraries. Long-adapter single-stranded oligonucleotide (LASSO) probes have shown promise as a tool to clone long DNA fragments. LASSO probes are molecular inversion probes (MIP) engineered with an adapter region of user-defined length, flanked between template-specific probe sequences. Herein, we demonstrate that the adapter length is a key feature of LASSO that influences the efficiency of gene capture and cloning. Furthermore, we applied a model based on Monte Carlo molecular simulation in order to study the relationship between the long-adapter length of LASSO and capture enrichment. Our results suggest that the adapter length is a factor that contributes to the free energy of target–probe interaction, thereby determining the efficiency of capture. The results indicate that LASSOs with extremely long adapters cannot capture the targets well. They also suggest that targets of different lengths may prefer adapters of different lengths.

Abstract:
Objective.Toll-like receptors (TLR) recognizing endogenous and exogenous danger signals could play a role in rheumatoid arthritis (RA). Our aim was to describe the presence, localization, and extent of expression of TLR and their adapters.Methods.TLR 1, 2, 3, 4, 5, 6, and 9 receptors, and myeloid differentiation primary response protein 88, Toll/interleukin receptor (TIR) domain-containing adapter protein MyD88 adapter-like, and TIR domain-containing adapter-inducing interferon/TIR-containing adapter molecule-1 adapters were analyzed in RA (n = 10) and osteoarthritis (OA; n = 5) samples using real-time polymerase chain reaction (PCR). Their colocalization with cellular markers CD68, CD15, CD3, CD4, CD8, CD20, dendritic cell lysosomal-associated membrane protein (DC-LAMP), CD123, and 5B5 was analyzed in double immunofluorescence staining.Results.In RA, ß-actin standardized messenger RNA of TLR 2, 3, and 9 (p < 0.001) were particularly high. TLR 5 and 6 were also elevated (p < 0.05), but TLR 1 and 4 and adapters did not differ between RA and OA. In double-staining, TLR and adapters were strongly labeled in myeloid and plasmacytoid dendritic cells (DC), moderately in CD68+ type A lining cells/macrophages, and weakly to moderately in 5B5+ type B lining cells/fibroblasts. CD3+/CD4+ and CD3+/CD8+ T cells and CD20+ B cells in perivenular areas and in lymphoid follicles were moderately TLR- and weakly adapter-positive. In OA, TLR and adapters were weakly immunolabeled in vascular, lining, and inflammatory cells.Conclusion.RA synovium showed abundant expression of TLR. RA synovitis tissue seems to be responsive to TLR ligands. DC, type A cells/macrophages, and type B cells/fibroblasts are, in that order from highest to lowest, equipped with TLR, suggesting a hierarchical responsiveness. In RA, danger-associated molecular patterns to TLR interactions may particularly drive DC to autoinflammatory and autoimmune cascades/synovitis.

Abstract:

Abstract:
The purpose of this paper is to study the adaptation process of the TV-show Game of Thrones. This paper focuses mainly on how David Benioff and D.B. Weiss and HBO adapted the TV-show from George R.R. Martins book series A Song of Ice and Fire and what, in the adaptation and in the reception of the TV-show, made it such a global cultural phenomenon. The analysis has been done by analysing interviews and studying behind-the-scenes material that has been available through articles and bonus material provided by the web and on the DVD and Blu-Ray features of the show. Mary H. Snyders Analyzing Literature-to-Film Adaptations provides a ground tool of analysing adaptations where particularly her “A Marriage of Media” theory has significantly helped the study. The paper analyses fantasy as a genre, the book series that the TV-show is based on, and the adaptation process itself and the different components that make up for the TV-show called Game of Thrones. After the adaptation analysis there is a study on how the internet has helped distributing both the show’s popularity and the show itself through the use of illegal downloading. The result is that the adaption is a successful “Marriage between media”, to use Snyders words, where the internet can be seen as product of that metaphorical marriage. The confidence and trust that both the author of the books and the show runners of the TV-series has placed in each other is the binding fundament that made the adaption as successful as it is. HBO as a cable network company also play a significant part where the financial support to the show’s production and the non-existent censorship regarding the adult content otherwise wouldn’t have been provided for, had the show been produced and/or aired by any other network company for example. The previous successes of other fantasy adaptations such as The Lord of the Rings and the Harry Potter-series could also be seen as necessary initiators regarding the industry’s newfound openness towards large scale fantasy projects.

Abstract:

One of the challenges is the way of data collecting from terminal nodes and data processing, including their distribution to the others nodes of Smart-home. The goal of this thesis is to design and to produce Smart-home control node, which will be a gateway between sensoric net and database and will be developed and produced on embedded system with speci c HW boards. Functionality of this gateway should include error states, e.g. should cache sensor messages when the connection between gateway and server interrupts, or take care about time synchronising. The thesis describes theoretical side of gateway design and also practical side of programming and producing SW for the gateway in Smart-home system.

The adaptor complex 1 (AP-1) acts in the formation of clathrin-coated vesicles at the transGolgi network of eukaryotic cells. Knowledge about the AP-1 complex in trypanosomatids is scarce, but it has been already demonstrated its importance in the infectivity of Leishmania mexicana in macrophages, as well as the viability of Trypanosoma brucei. Trypanosoma cruzi is a protozoan flagellate that belongs to the family Trypanosomatidae, being the etiologic agent of Chagas disease, which affects millions of people worldwide. In this context, this study aimed to identify the genes encoding the four subunits of the AP-1 complex in the genome of T. cruzi and analyze their expression and subcellular localization in this parasite. Search in genomic database identified gene sequences coding for all subunits of the AP-1 complex, the following accession numbers being obtained: AP1-γ: XP_818958.1; AP1-β: XP_820334.1; AP1-µ: XP_818899 .1; AP1-σ: XP_804127.1. Antibodies were produced in mice against recombinant proteins of all subunits of the AP-1 complex. Analysis of the specificity of the antibodies was performed by western blot and subcellular localization of the proteins by immunofluorescence was done by epifluorescence microscopy and confocal laser microscopy. Negative results were obtained with the antiserum against subunit AP1-σ. Antibodies raised against the AP1-µ (polyclonal) and AP1-β (monoclonal) subunits recognized polypeptides with size compatible to the molecular weight of the endogenous protein in extracts from epimastigotes, but no proteins could be localized by fluorescence microscopy. The antiserum polyclonal obtained against the AP1-γ subunit recognized a polypeptide in extracts of different evolutionary forms of T. cruzi (epimastigotes, trypomastigotes and amastigotes) of molecular weight consistent with that predicted in database (~90 kDa). Immunolocalization showed punctual positive reaction in the region compatible with the Golgi complex of this protozoan: between nucleus and kinetoplast of trypomastigotes and between kinetoplast and flagellar pocket of epimastigotes and amastigotes. Colocalization of AP1-γ with the GTPase Rab7 (a marker of the Golgi complex in T. cruzi) confirmed the location of this subunit in the Golgi apparatus of this parasite. Our results demonstrate that the subunits of the AP-1 complex are conserved and expressed in T. cruzi and that at least the AP1-γ subunit has cellular localization in the Golgi apparatus, similar to that described in other eukaryotic cells.

Abstract:
This thesis work had the intention to acoording to the processes of Motion Control develop a prototype of a wireless cable replacer for an existing product, USB I/O. USB I/O is an I/O and data collection unit with a number of in- and outputs. It has in some applications the drawback that it is connected through a USB cable. The idea for the work was to create a unit to connect to USB I/O which has the capability to connect to any existing “Bluetooth dongle” on the market.Three investigations were made. The first was a market investigation to see potential system solutions and to find out any direct competitors. It showed that the existing market did not offer any large number of active similar products. After this, two investigations were made to find suitable components for the prototype. Half way in the thesis it came obvious that whichever Bluetooth module that could be chosen, a Bluetooth profile, HID, was needed and had to be modified in order to use the conventional “Bluetooth dongles”. This would make a great cost or effort in development time than the scope of the thesis work allowed. From this, a decision was made to also include development of an additional unit to connect to the computer side, which then would replace the “dongle”.The thesis resulted in a prototype consisting of two subsystems, however in need of further development, offering partly the intended functionality. The USB I/O could be started up on a computer wirelessly using the system.

Abstract:
We consider open systems being described by an interface and behavior in form of an protocol. Interaction between open systems means exchange of messages. For the interaction to be correct, the interacting open systems must be able to terminate. The interaction of independently developed open systems is not guaranteed to be correct. Although one open system provides functionality a second open system requires, incompatiblities of the interfaces or the behavior may prevent the correct exchange of messages. The idea is to introduce an adapter to overcome the incompatiblities. We present a technique that considers incompatiblities of the interfaces and the behavior separately. With transformation rules we specify how messages of two open systems relate to each other. Using these rules we model the data flow between open systems and overcome incompatiblities of the interfaces. By using existing techniques for controller synthesis we overcome incompatiblities of the behavior. Thus we create correct control flow. We call the composition of transformation rules and controller an adapter. Based on the technique presented in this thesis, we want to answer the following reseach questions: - What are the properties of the proposed technique? When does an adapter exist? How can we apply the technique on open, communicating systems in general? - How do we distribute a generated adapter to different components? - Which information can we provide, if adapter synthesis fails? - How can we discover a formal model of an adapter, when we only have recorded behavior of open systems, but no formal models?

Abstract:
Concretization is one of the most labor-intensive phases of the model-based testing process. This study concentrates on concretization of the abstract tests generated from the test models. The purpose of the study is to design and implement a structure to automate this phase which can reduce the required effort specially in every system update. The structure is completed and discussed as an extension on a model-based testing tool named ModelJUnit using adaptation approach. In this structure, the focus is mainly on bridging the gap in data-level between the SUT and the model.

In 2015 Joint Rail Conference. American Society of Mechanical Engineers, 2015. http://dx.doi.org/10.1115/jrc2015-5790.

Abstract:
This paper presents a study of the fatigue life (i.e. number of stress cycles before failure) of Class K cast iron conventional and modified railroad bearing adapters for onboard monitoring applications under different operational conditions based on experimentally validated Finite Element Analysis (FEA) stress results. Currently, freight railcars rely heavily on wayside hot-box detectors (HBDs) at strategic intervals to record bearing cup temperatures as the train passes at specified velocities. Hence, most temperature measurements are limited to certain physical railroad locations. This limitation gave way for an optimized sensor that could potentially deliver significant insight on continuous bearing temperature conditions. Bearing adapter modifications (i.e. cut-outs) were required to house the developed temperature sensor which will be used for onboard monitoring applications. Therefore, it is necessary to determine the reliability of the modified railroad bearing adapter. Previous work done at the University Transportation Center for Railway Safety (UTCRS) led to the development of finite element model with experimentally validated boundary conditions which was utilized to obtain stress distribution maps of conventional and modified railroad bearing adapters under different service conditions. These maps were useful for identifying areas of interest for an eventual inspection of railroad bearing adapters in the field. Upon further examination of the previously acquired results, it was determined that one possible mode of adapter failure would be by fatigue due to the cyclic loading and the range of stresses in the railroad bearing adapters. In this study, the authors experimentally validate the FEA stress results and investigate the fatigue life of the adapters under different extreme case scenarios for the bearing adapters including the effect of a railroad flat wheel. In this case, the flat wheel translates into a periodic impact load on the bearing adapter. The Stress-Life approach is used to calculate the life of the railroad bearing adapters made out of cast iron and subjected to cyclic loading. From the known material properties of the adapter (cast iron), the operational life is estimated with a mathematical relationship. The Goodman correction factor is used in these life prediction calculations in order to take into account the mean stresses experienced by these adapters. The work shows that the adapters have infinite life in all studied cases.

Abstract:
This paper presents a detailed study of the structural integrity of conventional and modified railroad bearing adapters for onboard monitoring applications. Freight railcars rely heavily on weigh bridges and stations to determine cargo load. As a consequence, most load measurements are limited to certain physical railroad locations. This limitation provided an opportunity for an optimized sensor that could potentially deliver significant insight on bearing condition monitoring as well as load information. Bearing adapter modifications (e.g. cut-outs) were necessary to house the sensor and, thus, it is imperative to determine the reliability of the modified railroad bearing adapter, which will be used for onboard health monitoring applications. To this end, this study quantifies the impact of the proposed modifications on the adapter structural integrity through a series of experiments and finite element analyses. The commercial software Algor 20.3TM is used to conduct the stress finite element analyses. Different loading scenarios are simulated with the purpose of obtaining the conventional and modified bearing adapter stresses during normal and abnormal operating conditions. This information is then used to estimate the lifetime of these bearing adapters. Furthermore, this paper presents an experimentally validated finite element model which can be used to attain stress distribution maps of these bearing adapters in different service conditions. The maps are also useful for identifying areas of interest for an eventual inspection of conventional or modified railroad bearing adapters in the field.

Abstract:
Currently, railroad bearing temperatures are monitored using wayside infrared devices known as hot-box detectors (HBDs). HBDs take a snapshot of the bearing temperature at designated wayside detection sites which, depending on the track, may be spaced as far apart as 65 km (∼40 mi). Even though these devices have significantly reduced the number of derailments since their implementation, their discrete nature and limited accuracy prevents them from being utilized as a bearing health monitoring system. Future technologies are focusing on continuous temperature tracking of bearings. Since placing sensors directly on the bearing cup is not feasible due to cup indexing during service, the next logical location for such sensors is the bearing adapter. Understanding the thermal behavior of bearing adapters during operation is essential for sensor selection and placement within the adapter (e.g., typical temperature sensors have operating ranges of up to 125°C). To this end, this paper quantifies the steady-state heat transfer to the bearing adapter through a series of experiments and finite element analyses. The commercial software package ALGOR 20.3™ is used to conduct the thermal finite element analyses. Different heating scenarios are simulated with the purpose of obtaining the bearing adapter temperature distribution during normal and abnormal operating conditions. This paper presents an experimentally validated finite element thermal model which can be used to attain temperature distribution maps of bearing adapters in service conditions. These maps are useful for identifying ideal locations for sensor placement.
