The adaptor complex 1 (AP-1) acts in the formation of clathrin-coated vesicles at the transGolgi network of eukaryotic cells. Knowledge about the AP-1 complex in trypanosomatids is scarce, but it has been already demonstrated its importance in the infectivity of Leishmania mexicana in macrophages, as well as the viability of Trypanosoma brucei. Trypanosoma cruzi is a protozoan flagellate that belongs to the family Trypanosomatidae, being the etiologic agent of Chagas disease, which affects millions of people worldwide. In this context, this study aimed to identify the genes encoding the four subunits of the AP-1 complex in the genome of T. cruzi and analyze their expression and subcellular localization in this parasite. Search in genomic database identified gene sequences coding for all subunits of the AP-1 complex, the following accession numbers being obtained: AP1-γ: XP_818958.1; AP1-β: XP_820334.1; AP1-µ: XP_818899 .1; AP1-σ: XP_804127.1. Antibodies were produced in mice against recombinant proteins of all subunits of the AP-1 complex. Analysis of the specificity of the antibodies was performed by western blot and subcellular localization of the proteins by immunofluorescence was done by epifluorescence microscopy and confocal laser microscopy. Negative results were obtained with the antiserum against subunit AP1-σ. Antibodies raised against the AP1-µ (polyclonal) and AP1-β (monoclonal) subunits recognized polypeptides with size compatible to the molecular weight of the endogenous protein in extracts from epimastigotes, but no proteins could be localized by fluorescence microscopy. The antiserum polyclonal obtained against the AP1-γ subunit recognized a polypeptide in extracts of different evolutionary forms of T. cruzi (epimastigotes, trypomastigotes and amastigotes) of molecular weight consistent with that predicted in database (~90 kDa). Immunolocalization showed punctual positive reaction in the region compatible with the Golgi complex of this protozoan: between nucleus and kinetoplast of trypomastigotes and between kinetoplast and flagellar pocket of epimastigotes and amastigotes. Colocalization of AP1-γ with the GTPase Rab7 (a marker of the Golgi complex in T. cruzi) confirmed the location of this subunit in the Golgi apparatus of this parasite. Our results demonstrate that the subunits of the AP-1 complex are conserved and expressed in T. cruzi and that at least the AP1-γ subunit has cellular localization in the Golgi apparatus, similar to that described in other eukaryotic cells.

Full textThis thesis work had the intention to acoording to the processes of Motion Control develop a prototype of a wireless cable replacer for an existing product, USB I/O. USB I/O is an I/O and data collection unit with a number of in- and outputs. It has in some applications the drawback that it is connected through a USB cable. The idea for the work was to create a unit to connect to USB I/O which has the capability to connect to any existing “Bluetooth dongle” on the market.Three investigations were made. The first was a market investigation to see potential system solutions and to find out any direct competitors. It showed that the existing market did not offer any large number of active similar products. After this, two investigations were made to find suitable components for the prototype. Half way in the thesis it came obvious that whichever Bluetooth module that could be chosen, a Bluetooth profile, HID, was needed and had to be modified in order to use the conventional “Bluetooth dongles”. This would make a great cost or effort in development time than the scope of the thesis work allowed. From this, a decision was made to also include development of an additional unit to connect to the computer side, which then would replace the “dongle”.The thesis resulted in a prototype consisting of two subsystems, however in need of further development, offering partly the intended functionality. The USB I/O could be started up on a computer wirelessly using the system.

Full textWe consider open systems being described by an interface and behavior in form of an protocol. Interaction between open systems means exchange of messages. For the interaction to be correct, the interacting open systems must be able to terminate. The interaction of independently developed open systems is not guaranteed to be correct. Although one open system provides functionality a second open system requires, incompatiblities of the interfaces or the behavior may prevent the correct exchange of messages. The idea is to introduce an adapter to overcome the incompatiblities. We present a technique that considers incompatiblities of the interfaces and the behavior separately. With transformation rules we specify how messages of two open systems relate to each other. Using these rules we model the data flow between open systems and overcome incompatiblities of the interfaces. By using existing techniques for controller synthesis we overcome incompatiblities of the behavior. Thus we create correct control flow. We call the composition of transformation rules and controller an adapter. Based on the technique presented in this thesis, we want to answer the following reseach questions: - What are the properties of the proposed technique? When does an adapter exist? How can we apply the technique on open, communicating systems in general? - How do we distribute a generated adapter to different components? - Which information can we provide, if adapter synthesis fails? - How can we discover a formal model of an adapter, when we only have recorded behavior of open systems, but no formal models?

This study aimed to find out how the Adapted Physical Education curriculum subject or similar has appeared in undergraduate and graduate / undergraduate degree in Physical Education and Sport in the three public state universities of São Paulo, and makes interaction with other disciplines the curriculum. The research is of qualitative character and used as techniques to collect the survey data source document via the Internet by consulting the website of the Ministry of Education (MEC): the link: http://emec.mec.gov.br/ and semi structured interview. The documents were collected: curricula and syllabus of the course. In the analysis of documents provided much of the public and private institutions found that, as the nomenclature, the discipline has performed with similar classifications in some private institutions, and differently in public and private institutions within the proposed undergraduate and graduate / undergraduate degree in Physical Education and Sport. Regarding the analysis of interviews with teachers, we could analyze the characterization of the discipline with a nomenclature that identifies and a lack of consensus on the content taught and the mainstream reports about the strategies of education, with emphasis on the university extension. In the matter of the curriculum was found that there is a lack of interaction between the discipline and others. As for training, intervention and preparation for the labor market participants had a concern with the need to prepare students with diverse profiles. Within this context, we consider that there is a need to think of practical content in the absorption of supervised training, training courses and graduate courses similar to medical residency. We found that the discipline does not interact with other disciplines because of the settings in the existing curriculum of physical education courses in Brazil mosaic-like, and what ... Full textThe dissertation presents the work done under the scope of the NP7 Self-Learning project regarding the design and development of the Adapter component as a foundation for the Self-Learning Production Systems (SLPS). This component is responsible to confer additional proprieties to production systems such as lifecycle learning, optimization of process parameters and, above all, adaptation to different production contexts. Therefore, the SLPS will be an evolvable system capable to self-adapt and learn in response to dynamic contextual changes in manufacturing production process in which it operates. The key assumption is that a deeper use of data mining and machine learning techniques to process the huge amount of data generated during the production activities will allow adaptation and enhancement of control and other manufacturing production activities such as energy use optimization and maintenance. In this scenario, the SLPS Adapter acts as a doer and is responsible for dynamically adapting the manufacturing production system parameters according to changing manufacturing production contexts and, most important, according to the history of the manufacturing production process acquired during SLPS run time.To do this, a Learning Module has been also developed and embedded into the SLPS Adapter. The SLPS Learning Module represents the processing unit of the SLPS Adapter and is responsible to deliver Self-learning capabilities relying on data mining and operator’s feedback to up-date the execution of adaptation and context extraction at run time. The designed and implemented SLPS Adapter architecture is assessed and validated into several application scenario provided by three industrial partners to assure industrial relevant self-learning production systems. Experimental results derived by the application of the SLPS prototype into real industrial environment are also presented.

Full textWith the arrival of cloud computing technology, game accessibility and ubiquity havea bright future. Games can be hosted in a centralize server and accessed through theInternet by a thin client on a wide variety of devices with modest capabilities: cloudgaming. Some of the advantages of using cloud computing in game context includes:device ubiquity, computing exibility, affordable cost and lowered set up overheads andcompatibility issues. However, current cloud gaming systems have very strong requirementsin terms of network resources, thus reducing their widespread adoption. In factdevices with little bandwidth and people located in area with limited network capacity, cannot take advantage of these cloud services. In this thesis we present an adaptationtechnique inspired by the level of detail (LoD) approach in 3D graphics. It is based ona cloud gaming paradigm in other to maintain user's quality of experience (QoE) byreducing the impact of poor network parameters (delay, loss, bandwidth) on game interactivity. Our first contribution consist of game models expressing game objects and theircommunications needs represented by their importance in the game. We provided twodifferent ways to manage objects' importance using agents organizations and gameplaycomponents. We then provided a level of detail approach for managing network resourcedistribution based on objects importance in the game scene and network conditions. Weexploited the dynamic objects importance adjustment models presented above to proposeLoD systems adapting to changes during game sessions. The experimental validation ofboth adaptation models showed that the suggested adaptation minimizes the effects oflow and/or unstable network conditions in maintaining game responsiveness and player'sQoE

We focus on integrating PhilipsHue personal wireless lighting unto the platform Elis (Mobile services for energye ciency in existing buildings). Within the short timeframe of this study we intendto reach a level of understanding enough to make the systems communicate usingour adapter prototype. Initially we study documentation and prepares a work plan.Further more we try to solve the adaptation needed for the two system to communicate, this involves converting and matching up values. We will do some interviewswith the developers of Elis to get the big picture of how and why they designed theplatform they way it is. The research paradigm design research is a iterative methodologythat creates milestones, develop prototypes and evaluate the work. Thegoal of design research is to create an artifact, in our case an adapterprototype. Wemade a total of four iterations where the work was divided. Step one was to learnhow the platform works and step two was to study Philips Hue. At step three theimplementation of our prototype with the preparatory work from the rst iterationscould begin. The nal step was to interview members of Elis development team to nd out the impact of our work and to discuss the pros and cons of working withtheir platform. We present opinions and ndings of things we have found that canbe improved. We also de ne how our adapter bene ts Elis in a software ecosystemperspective.. . .

Abstract: In Brazil, the bylaws of Universities set forth three specific purposes: Education, Research and Extension Courses. Such universities must create, develop, organize, preserve and foster academic and scientific knowledge and culture, through education and research, in order to train professionals who are able to conduct scientific research, teach, and practice other professions, as well as work with the community, in order to develop academic activities. In recent years, there has been growing interest in the research of Adapted Motor Activity. Another observable fact is that the studies, which discuss the production of scientific knowledge in post graduation and doctorate programs in Physical Education in Brazil focus on several fields and lines of research, but have not prioritized Adapted Motor Activity. Therefore, a relevant research theme would be to survey the knowledge produced in relation to Adapted Motor Activity, within the masters and doctorate programs, in Physical Education, recognized and recommended by Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (Coordination of Improvement of Higher Education Personnel). In order to conduct such a survey, a review of the literature in the field, as well as an analysis of the content, was conducted, whose aim was to analyze a large number of studies and list their basic principles. The data collected were organized into categories: Theory, divided into subcategories - Theme; Area of Concentration, Line of Research; Authors, Advisors and Date of Presentation; Methodological: Data Collection: research techniques, Methodological tools and procedures used; Focus Group. These were further processed by analysis grids. For the purpose of the present work Adapted Motor Activity is defined as the effort to adapt the means to execute a task in the absence of conventional means or inability to use them. 2 In this manner it is related to special groups, as those people with disabilities (physical, motor, physical sensorial and mental). At first, the analysis of dissertations and theses, between 1986 and 2008, of the Courses in Physical Education in Brazil, was conducted based on the abovementioned categories, and later the focus was on the analysis of elements obtained, and the establishment of generalizations based on historical, political and social determinants. The significant changes that have taken place, particularly in the field of Physical Education and Adapted Motor Activities, cannot be ignored, especially since the UN instituted 1981 as the International Year for Disabled Persons, which contributed for the establishment of a profile in this field of study. Studies have been produced mainly in the area of Biological Sciences; there are 123 researchers, who produce work in this area, 35 are professors/thesis advisors, who in turn give orientation to 68 graduate and 20 doctorate students; the themes are focused on groups which mainly involve, in that order, adults, then children, and adolescents. Research in Physical Education in relation to AMA represents a fairly new field, as the first dissertation was presented in 1986 (USP). UNICAMP is the Higher Education Institution with the most prolific production in the area (53 studies), followed by UFRGS (11 studies), USP (6 studies), UCB and UGF (4 studies each), UNESP and UFSC (3 studies each), UDESC and UFPR (with 2 studies each); only two Higher Education Institutions have a specific line of research to deal with issues related to AMA in the field of Physical Education - UNICAMP and UFRGS. This fact and the amount of time dedicated to research in the area, has fostered a greater production in those institutions. Besides the previously mentioned determinants of the manner through which research in Physical Education is structured, are also the following points: the allocation of the field in area number 21, Health; the evaluation criteria for production established by CAPES (Coordination of Improvement of Higher Education Personnel); and the recent discovery of the disabled population as research subjects, in Physical Education; in the listed Bibliography (as can be verified with standardized tests), notably influenced by the North American notion of disability. The research about AMA in Physical Education has been bold as it focuses on individuals who are in disadvantage in relation to standard movements, established during the evolution of Physical Education. However, it is important to emphasize that this profile has been constructed as both a product and a process in a contextual complex that is much more comprehensive and dynamic, with many contributive factors, which have been taken into account in the present study.

The objective of this research was to analyze the principles behind the collaborative work between the teachers of Physical Education and Specialized Educational Assistants involved in the planning process of scholarly inclusion. This qualitative descriptive study was based on the underlying assumptions of collaborative research. Two Physical Education teachers and two Specialized Educational Assistants of the municipal elementary schools in the Central-West of the state of São Paulo participated in the study. A survey of national and international literature helped identify the theoretical principles that encompass a proposal for collaborative work. A questionnaire was used with the teachers who worked in the Specialized Educational Assistance, to identify whether the principles of the collaborative work outlined in the literature are applicable in practice. Data collection occurred in two different environments. Physical Education classes were filmed, focusing on the actions of the Physical Education teacher with the student, in order to identify the principles that make up the collaborative work. The collection also occurred in the meetings for planning with the teacher of the Specialized Educational Assistance and Physical Education, where the audios were recorded, with the objective of identifying in the teachers' speech the principles that make up the collaborative work and at the end of the work carried out A semi-structured interview, based on a script, with six open questions, related to the teachers' experience in working collaboratively. For the treatment and analysis of the information, the categories identified in the bibliographic survey were separated and regrouped by similarities, and later classified in three broad categories. These categories were used to identify, in the questionnaire, in the filming, in the field notes and in the recordings of the speeches, indications of the first fruits of the collaborative work. The results revealed contents about: 1) characteristics of teachers; 2) conditions for carrying out the collaborative work; And, 3) possibilities based on collaborative work. It is concluded that the principles that make up a proposal for collaborative work are based on questions related to the profile of teachers and their willingness to participate in this proposal, the availability of time for dialogues at school, the need to elaborate a collaborative work plan for That the actions and strategies systematized together and that from the collaborative work teachers have the opportunity to know the student and risk new strategies.

Full textSystems development projects and their documents are more or less standardized and can mainly be applied on systems that are supposed to be built from scratch, or updated. In pace with the number of IT-systems are increasing worldwide there is no need for every organization to build their own IT-system. Nowadays it is also possible to purchase licenses which allow the purchaser to modify or add functions to the system. Along with those changes, there have been an increased amount of “rapid development methods” such as Agile and “Quick and Dirty” solutions, but these methods and perspectives are mainly focusing on entire systems development processes, as the old ones, but quicker.
If a company purchases an off-the-shelf system with source code available, there is no real need to go through a proper systems development process. During interviews with a small company that has acquired a system as mentioned above, the researcher realized that only one single document is needed, the requirements specification. Today’s requirements specifications can be either well detailed or less, but a project still needs the details specified. Combining a known agile development process with IEEE’s standardized requirements specification, a new way to proceed with projects based on one single document (the requirements specification) has been made. This document also has a focus on simplicity for the inexperienced readers, but with the depth that every developer has got a use for.

Quantum error correction (QEC) is an essential concept for any quantum information processing device. Typically, QEC is designed with minimal assumptions about the noise process; this generic assumption exacts a high cost in efficiency and performance. We examine QEC methods that are adapted to the physical noise model. In physical systems, errors are not likely to be arbitrary; rather we will have reasonable models for the structure of quantum decoherence. We may choose quantum error correcting codes and recovery operations that specifically target the most likely errors. This can increase QEC performance and also reduce the required overhead. We present a convex optimization method to determine the optimal (in terms of average entanglement fidelity) recovery operation for a given channel, encoding, and information source. This is solvable via a semidefinite program (SDP). We derive an analytic solution to the optimal recovery for the case of stabilizer codes, the completely mixed input source, and channels characterized by Pauli group errors. We present computational algorithms to generate near-optimal recovery operations structured to begin with a projective syndrome measurement.
(cont.) These structured operations are more computationally scalable than the SDP required for computing the optimal; we can thus numerically analyze longer codes. Using Lagrange duality, we bound the performance of the structured recovery operations and show that they are nearly optimal in many relevant cases. We present two classes of channel-adapted quantum error correcting codes specifically designed for the amplitude damping channel. These have significantly higher rates with shorter block lengths than corresponding generic quantum error correcting codes. Both classes are stabilizer codes, and have good fidelity performance with stabilizer recovery operations. The encoding, syndrome measurement, and syndrome recovery operations can all be implemented with Clifford group operations.

Full textThe aim of this research is bouble : 1) On the one hand, to prove that successfull adaptations go through a precise knowledge of the pupils they are bound to benefit, as well as pedagogic strategies they perform. It is a complex process which must be present at any stage of the institution. A sample of fifteen data sheets has been elaborated, covering the main situations a team is supposed to deal with. 2) On the other hand, to show the adapting process, based on the political will of national unification, has got to face the strong resistance of certain minorities, especially the gipsy minority. This reject is a problem to European school authorities because it generates doubtful compromises with the laws concerning school obligation
