Abstract:
Since the 19th century, the addax (Addax nasomaculatus) has lost approximately 99% of its former range. Along with its close relatives, the blue antelope (Hippotragus leucophaeus) and the scimitar-horned oryx (Oryx dammah), the addax may be the third large African mammal species to go extinct in the wild in recent times. Despite this, the evolutionary history of this critically endangered species remains virtually unknown. To gain insight into the population history of the addax, we used hybridization capture to generate ten complete mitochondrial genomes from historical samples and assembled a nuclear genome. We found that both mitochondrial and nuclear diversity are low compared to other African bovids. Analysis of mitochondrial genomes revealed a most recent common ancestor ~32 kya (95% CI 11–58 kya) and weak phylogeographic structure, indicating that the addax likely existed as a highly mobile, panmictic population across its Sahelo–Saharan range in the past. PSMC analysis revealed a continuous decline in effective population size since ~2 Ma, with short intermediate increases at ~500 and ~44 kya. Our results suggest that the addax went through a major bottleneck in the Late Pleistocene, remaining at low population size prior to the human disturbances of the last few centuries.

SummaryProtostrongylids, small nematode lungworms, are an integral part of the wild ruminant helminth community, which can damage animals’ health when they are held in captivity or semi-captive conditions. The Sahelo-Saharan antelope species dorcas gazelle (Gazella dorcas), the scimitar-horned oryx (Oryx dammah), and the addax (Addax nasomacculatus), reintroduced to Souss-Massa National Park in Morocco, could be host to many species of Protostrongylids. This study was conducted from January to July 2015 to identify infecting parasite species, and determine their prevalence and abundance in all three antelope species. A total of 180 individual fecal samples were collected, morphologically examined by the Baermann technique, and molecularly identified by PCR amplification and sequencing of the second internal transcribed spacer region of the rDNA (ITS-2).Two parasite species were found in the three antelope populations: Muellerius capillaris and Neostrongylus linearis. The prevalence scores recorded for M. capillaris were 98.40 % in the addax, 96.70 % in dorcas gazelle, and 28.40 % in the oryx. The prevalence rates of N. linearis were 60 % in the addax, 23.40 % in dorcas gazelle, and 90 % in the oryx. Excreted larvae were quantified by LPG (larvae per gram) counting: for M. capillaris, the LPG mean values were 92.94 in the addax, 133.09 in dorcas gazelle, and 1.48 in the oryx; and for N. linearis, the LPG mean values were 6.02 in the addax, 1.37 in dorcas gazelle, and 32.81 in the oryx. These findings indicate that the three species of antelopes are infected with Muellerius capillaris and Neostrongylus linearis to varying degrees in intensity and prevalence.

<p>Detta examensarbete tar upp frågan om kommunikationsproblem mellan de komponenter som ingår i ADDAX systemet, till största del de problem som uppstår mellan router och mätare. ADDAX systemet är uppbyggt på ett hierarkiskt vis med ADDAX server högst upp och mätaren längs ned. Systemet kommunicerar via PLC ”power line comunication”. Uppgiften har varit att kartlägga hur mätsystemet fungerar, vilka geografiska problemområden som finns med avseende på kommunikations problem, lokalisering av störkällor, åtgärdning av dessa störkällor och framtagande av ett flödesschema som skall kunna användas av montörerna vid felsökning i systemet. Arbetet inleddes med att ta fram underlag för vilka områden som påverkas av störningar i elnätskommunikationen. Detta gjordes med hjälp av programmet ”TC Meter Surv SA Edition 0.61” som arbetar mot ADDAX systemets databas för att se vilka mätare som är installerade och vilka som kommunicerar med systemet. Efter vissa problem togs två områden fram, ett på landsbygden och ett i centrala Grästorp. Vid mätning i dessa områden användes till en början två instrument, PL-monitor och HF+. HF+ visade sig vara överflödigt då samma information ges av PL-monnitor båda versioner visar på vilka störningsnivåer som finns i elnätet. HF+ är mer svårtolkat då användaren själv måste ta ställning till informationen som visas medan PL-monnitor enbart visar på den störning som stör ut kommunikationen. Då mätningarna genomförts och analyserats har det visat sig att programmet ej fungerat tillfredställande och att fel information om antal felaktiga mätare i de område som är beläget i centrala Grästorp, detta har resulterat i att en ny slutsats dragits där istället för en störning i LV nu problemet verkar ligga i MV. Kommunikations problemet på landsbygden kan troligtvis lösas med att installera filter hos de kunder vars mätare ej kommunicerar. Andra åtgärder som kan vara aktuella vid framtida kommunikationsproblem kan vara byta av kommunikations fas på mätaren, flytt av mätaren till matningsskåp, installation av repeterare eller att i samråd med kund få denna att stänga av störande utrustning vissa tider på dygnet. En slutsats som dragits av de erfarenheter som ackumulerats under projektets gång är att all mjukvara bör uppdateras och att master-routrar ersätts med en nyare version.</p><br><p>This report handles the issue of communication problems that appears between the components in the ADDAX system, mainly between the router and meter. The system is built around a hierarchy with the ADDAX server at the top and the meter at the bottom. The system uses the power grid as communication medium, this is called PLC “power line communication. The task has bin to se how the system works, witch geographical areas that have problems with communication, localization of source for the disturbing of the communication, attend to the disturbing and to make a flow chart that the electricians can use when they are conducting a measurement. A program called “TC Meter Surv SA Edition 0.61” was used for finding which areas that have problems with the communication. Two areas were chosen, one in the countryside and one in central Grästorp. The program firs showed that some meters where missing but when the measurements where don the program showed that all meters where missing. The reason for this problem is unknown. The problem in the central Grästorp area is not in the LV butt in the MV. The conclusion will be that al of the software must be updated and that the master routers could be replaced with a never versions before any new measurements are to be done. The power quality measurement instruments that has bin used are PL-monitor and HF+. On other conclusion that can bi don is that HF+ is superfluous because it chows the same information that PL-monitor dose but it’s harder to interpret. In the area on the countryside the solution to the communication problem can bi to install a filter. Other solutions to PLC problems can bi to switch the phase the meter uses for communication, move the meter farther from the customer, installing a repeater or in consultation with the customer get him or here to turn of the devise that disturb the communication under some ours in a 24-hour period.</p>

The only way to prevent is to vaccinate with BCG, an attenuated Mycobacterium bovis strain. However, the vaccine has variable efficacy in different people and it does not protect adults. The aim of this study was to develop new subunit vaccine formulations, evaluating a new adjuvant: Advax and two recombinant fusion proteins: CMX and ECMX. To formulate the subunit vaccines three adjuvants were used: the Advax3, Advax4 and CpG-DNA. First we addressed the CMX protein. BALB/c mice were separated in experimental groups that received 3 doses of the vaccines, besides the control groups (Saline, BCG, CMX, Advax3 and Advax4). The vaccines induced increase the local inflammatory response and they were able to activate blood and lymphatic endothelial cells on draining lymph nodes. After the first dose, the mice vaccinated with Advax3 + CMX, Advax4 + CMX and CpG-DNA + CMX generate IgG2a humoral immune response. After the second dose, the vaccinated mice also induced an IgG1 response. After challenging with M. tuberculosis and after 45 days, the vaccines BCG, Advax3 + CMX, Advax4 + CMX and CpG-DNA + CMX showed specific Th1 responses. However, the Th17 cellular response was not observed. Evaluating the lung lesions induced by the infection, Advax4 + CMX group presented reduced inflammatory cell infiltrate and maintenance on the pulmonary architecture. The vaccines Advax3 + CMX e Advax4 + CMX were able to extend the animals life, but none, except the BCG, were able to reduce the bacillary load on the lungs of infected mice, forty five or ninety days after the challenge. To improve the vaccine, ECMX was tested with Advax4. This vaccine showed similar humoral and cellular responses and reduced M. tuberculosis bacillary load. The vaccine formulations developed were immunogenic and the formulations containing Advax4 + ECMX were more effective and may be tested as vaccine against tuberculosis.<br>Tuberculose é uma doença infecto-contagiosa causada principalmente pelo Mycobacterium tuberculosis. A única forma de prevenção é a vacina BCG. Entretanto, essa vacina tem eficácia variável em diferentes populações e não protege indivíduos adultos. O objetivo desse trabalho foi desenvolver novas formulações vacinais de subunidade proteica testando um novo adjuvante: Advax e duas proteínas de fusão recombinante: CMX e ECMX. Foram utilizados três adjuvantes: Advax3, Advax4 e CpG-DNA. Primeiro foram testadas formulações vacinais contendo CMX. Camundongos BALB/c foram separados em grupos que receberam 3 doses de vacinas de subunidade proteica, além dos grupos controles (Salina, BCG, CMX, Advax3 e Advax4). Foram avaliadas as lesões induzidas no local da inoculação e os linfonodos drenantes. As vacinas estimularam aumento da celularidade no local da inoculação e conseguiram ativar células endoteliais vasculares e linfáticas nos linfonodos drenantes. Após a primeira dose, os camundongos vacinados com Advax3 + CMX, Advax4 + CMX e CpG-DNA + CMX geraram resposta imune humoral do tipo IgG2a. Após a segunda dose, os camundongos vacinados com as formulações citadas também induziram resposta do tipo IgG1. As vacinas BCG, Advax3 + CMX, Advax4 + CMX e CpG-DNA + CMX induziram respostas do tipo Th1 específicas tanto no baço quanto no pulmão. Entretanto, a resposta celular do tipo Th17 não foi observada. Nos cortes histológicos, foi possível observar que a formulação Advax4+CMX conseguiu reduzir o infiltrado inflamatório e manter a arquitetura tecidual. As formulações Advax3 + CMX e Advax4 + CMX conseguiram prolongar a sobrevida dos animais, mas nenhuma das vacinas, exceto a BCG, conseguiu reduzir a carga bacilar nos pulmões dos camundongos infectados. Utilizando Advax4 + ECMX observou-se o mesmo tipo de resposta imune celular e humoral que culminou com redução da carga bacilar do Mycobacterium tuberculosis. As formulações vacinais desenvolvidas são imunogênicas e a formulação vacinal contendo Advax4 + ECMX apresentou melhor eficácia e poderá ser testada como um candidato a vacina contra tuberculose.

Abstract:
This doctoral dissertation examines the popular Bengali leisure pursuit, adda, which may be defined as a long, informal talking session among friends, interspersing intellectual discussion and debate with gossip, rumour and small talk. Taking its cue from Dipesh Chakrabarty’s acclaimed essay on the Calcutta adda, my thesis analyses how the social practice of adda functions as a cultural signifier of ‘something quintessentially Bengali’. Such a general assumption of adda’s ‘quintessential Bengaliness’, I argue, is problematised by the present geo-political identity of ‘Bengal’ as a region straddling two different nation-states, namely West Bengal in India, and Bangladesh (former East Bengal). I correspondingly deconstruct the discourse of adda’s ‘quintessential Bengaliness’ along two key theoretical axes – adda’s radical capitalism and post-Partition melancholia. Adda’s radical capitalist premise postulates the economic, sociological and political significance of adda’s leisurely work ethic as a challenging alternative to the hegemonic epistemology of a globalised Western capitalism. Adda’s post-Partition melancholia demonstrates how the overarching West Bengali Hindu representation of this ‘quintessential’ talking practice can be traced back to the 1947 Bengal Partition. Employing a range of historiographical, literary and cinematic sources in conjunction with Edward Said’s contrapuntal reading methodology, my thesis interrogates the Hindu bhadralok master-discourse of the Calcutta adda to explore the marginalised adda narratives of various ‘other’ Bengali communities – both in West Bengal and Bangladesh. For this purpose, I revaluate the West Bengali bhadralok hierarchy against the contrapuntal grain of adda’s different subaltern space-times, including those inhabited by the East Bengali refugee, the West Bengali tribal, the bangal woman, and the Bangladeshi modern. I thereby mobilise both the radical capitalist productivity and critical melancholic agency of adda’s diverse representational imaginaries – preeminent and submerged, normative and transgressive, metropolitan and peripheral, to establish their dynamic presence as the heterogeneous constituents of a post-Partition Bengali contemporaneity.

Abstract:
Over billions of years cells have evolved numerous conserved repair pathways to deal with the continuous onslaught of DNA damage. One particularly toxic lesion is a double-stranded DNA break, which can be faithfully repaired by the ubiquitous homologous recombination pathway. In bacteria, the processing of double-stranded DNA breaks for recombinational repair is carried out by helicase-nuclease enzymes. These are stable multi-protein complexes that coordinate their helicase and nuclease activities to resect DNA ends. This activity generates the 3'-ssDNA overhangs that are required by the homologous pairing enzyme RecA/Rad51. In bacteria, the formation of this ssDNA tail is dependent upon recognition of the recombination hotspot sequence Chi; a specific single-stranded DNA sequence that acts to attenuate nuclease activity on the 3'-strand. Despite their extensive study, the mechanism by which helicase-nuclease enzymes recognise their cognate Chi sequence has remained elusive. This mechanism has been addressed in this thesis using the model helicase-nuclease AddAB, from Bacillus subtilis. Mutagenesis analysis reveals that Chi is recognised by the AddB subunit and interestingly, Chi binds to a domain that resembles a catalytically inactive SFl helicase. Further mutagenesis shows that an "ionic latch" structure regulates a conformational change that is linked to the recognition of Chi. This latch structure is also intimately linked to the specificity of the AddAB:Chi interaction. Using rapid reaction techniques it was also found, somewhat unexpectedly, that Chi attenuates the ATPdependent translocation activity of AddAB. Finally, preliminary data suggest that ATP binding to AddB stabilises the binding of Chi to AddB, but only after Chi has been recognised and AddAB has undergone a Chi-induced conformational change.

The DNA of all living organisms is constantly damaged by a wide variety of endogenous and exogenous agents. Double-stranded DNA breaks are particularly toxic and under certain conditions a single break will result in cell death. To preserve genome integrity, double-stranded breaks are faithfully repaired via homologous recombination. Recombinational repair in prokaryotes is generally initiated by a class of enzymes known as helicase-nucleases. These enzymes bind tightly to broken duplex DNA ends and use ATP driven motors to unwind the DNA.

Abstract:
Abstract Extending the long-range capabilities of unmanned aerial systems (UAS) is paramount to protecting small Soldier teams operating in remote battlefield scenarios. Currently, many small Unmanned Aerial Systems (sUAS) reconnaissance assets have the range necessary to facilitate small-unit missions. The purpose of this paper is to detail the design and testing of a system that fills this capability gap by creating folding-wing drones designed to be aerially deployed from an airdropped dispenser. The dispenser is attached to the Joint Precision Airdrop System (JPADS), which is an autonomously navigated cargo delivery parafoil that can glide several miles, and which can land within 100 meters of its target. To employ the system, the dispenser is launched from a high-altitude aircraft. The system must survive the opening of the parachute in high speed forward flight and provide cushioning to the drones and other components. Once the JPADS navigates the dispenser to a predetermined altitude and distance from the intended reconnaissance area, the dispenser deploys multiple folding-wing drones. The soldiers on the ground can access the drones’ live video feeds through a handheld video transmitter. The system combines the precision navigation and information-transmission capabilities of the fixed wing drone with the long-range capabilities of the JPADS. With a commercial-off-the-shelf drone as the folding-wing aircraft platform, the team designed a wing connection hub that allows for rapid folding and unfolding of the drone’s wings, a separate canister for each drone within the dispenser, and a dispenser capable of interfacing with both the canisters and the JPADS. Though currently in the technology-demonstration phase of the project, the team envisions the system being fully autonomous from launch of dispenser to end of mission.
