Abstract:
In 1992 and 1993 the Archaeological Resources Management Service (ARMS) of Ball State University in conjunction with the Upper White River Archaeological Society (UWRAS) conducted limited research of Windsor Mound located in Randolph County, Indiana. The project consisted of obtaining accurate profiles of an amateur excavation that began in 1986 and backfilling the excavation. This thesis provides a synthesis of the previous studies on Windsor Mound, an analysis of the materials recovered and the results of the limited UWRAS/ARMS excavations.

Abstract:
Despite incredible sales success, popularity, and a fifty year history, easy readers are one of the most neglected forms of children’s literature. Called everything from “the poor stepchild of the more glamorous picture book or children’s novel” to “literary flotsam,” easy readers are too-often regarded as insubstantial, superficial, sub-par literature. This thesis provides the first comprehensive, theoretically grounded examination of easy readers and endeavors to prove that a surprising complexity lurks beneath the easy reader’s decodable surface. In order to illuminate both extra-textual and textual complexity, easy readers are treated generically and examined using the contemporary genre theories of Amy Devitt and Adena Rosmarin. This thesis ultimately unearths a heretofore unexplored complexity in the easy reader’s publication history and generic emergence, and finds that the easy reader genre has literary potential and can accommodate works of artistic merit.

Aquest estudi tracta de connectar les disciplines de RH i Màrqueting, i examina el model de service-profit chain (SPC) en el sector de Call Centre, caracteritzat per un servei remot i un negoci basat en la reducció de costos. Les dades s'han col·lectat del Projecte "Global Call Center Project". Hem realitzat dos estudis. En el primer estudi (confirmatori) s'ha emprat una mostra internacional (n = 937). En el segon estudi (exploratori) vam utilitzar una mostra espanyola (n = 109). Els resultats revelen que el model SPC té una aplicació diferent en els call centres. Encara que a nivell general podem confirmar la majoria de les relacions del model, els resultats indiquen que la satisfacció del client és un resultat separat, i no un precursor del rendiment de l'empresa. També trobem una alternativa de mesurar el constructe individual de la satisfacció de l'empleat amb dades disponibles de la organització. Addicionalment, vam descobrir certes discrepàncies en la relació entre la productivitat i la satisfacció dels empleats.
El presente estudio es una conexión entre las disciplinas de RH y Marketing, y examina el modelo service-profit chain (SPC) en el sector de Call Centre, caracterizado por un servicio remoto y un negocio basado en la reducción de costes. Los datos se han colectado del Proyecto “Global Call Center Project”. Hemos realizado dos estudios. En el primer estudio (confirmatorio) se ha empleado una muestra internacional (n = 937). En el segundo estudio (exploratorio) utilizamos una muestra española (n = 109). Los resultados revelan que el modelo SPC tiene una aplicación diferente en los call centres. Aunque a nivel general podemos confirmar la mayoría de las relaciones del modelo, los resultados indican que la satisfacción del cliente es un resultado separado, y no un precursor de rendimiento de la empresa. También encontramos una alternativa de medir el constructo individual de satisfacción del empleado con los datos organizacionales disponibles. Adicionalmente, descubrimos ciertas discrepancias en la relación entre la productividad y la satisfacción de los empleados.
This thesis is an interface between HR and Marketing discipline, by examining the Service-Profit Chain (SPC) model in the context of call centre, characterized by remote service and cost cutting business models. Data was gathered from the Global Call Centre Project. We carried out two studies. The first one is a confirmatory study, using an international sample (n=937). In the second study we use a Spanish sample (n=109) and carry out an exploratory study. Findings reveal that the SPC model behaves somewhat differently in call centres. Although there is general support for most of the links in the model, the results indicate that customer satisfaction in the call centre industry is a separate outcome, rather than a precursor to company performance. In addition, we found a way to measure the individual level of employee satisfaction construct with organizational available data. We also discovered some discrepancies in the relationship between employee satisfaction and employee productivity.

Abstract:
Mound building was a common practice in the Midwestern United States among the Adena and Middle Mississippian tribes from approximately 500 B.C. to 1500 A.D. Though they varied greatly in size, shape, and complexity between cultures, the mounds served a common purpose as means to bury the deceased. Mounds representing both cultures have been found dispersed throughout Indiana, in areas such as Randolph County, Henry County, Madison County, Knox County, and Vanderburgh County.Of particular interest in this investigation is an Adena burial plot, known as Windsor Mound, which is 2000 yrs old and located in neighboring Randolph County. An amateur excavation of the mound from 1986 to 1988 unearthed the fragmented remains of 44 individuals and several artifacts, all of which were loaned to the Ball State University Anthropology Department. Documentation of the excavation was poor and the exact location of artifacts and remains within the mound were not recorded, leaving anthropologists with many unanswered questions that could only be answered through genetic analysis. Pressing questions were the degree to which the 44 individuals were related and which of the five Native American lineages (haplotypes) they belonged to.For this analysis, dentin was recovered from the teeth of three Native American individuals exhumed from Windsor Mound. Two of the individuals (87.17.5 and 87.17.10) were found in the lower mound portion (70 B.C.), while the third individual (87.17.22) was found in the upper mound cap (1180 A.D.). Ancient DNA (aDNA) was extracted from each dentin sample, yielding an average of 0.072 ug/mg of dentin. A highly polymorphic portion of the mitochondria) DNA control region (nt 16,049 - nt 16,221) termed hypervariable region 1 (HVI) was amplified via PCR to generate 172 bp amplicons which were cloned into a plasmid vector. Following a transformation, 10 clones from each individual were sequenced and aligned to identify consistent mutations, as opposed to random post-mortem damage that may have occurred.Sequencing of the HVI region for Individual 87.17.5 revealed a T —~ C base substitution at nt 16,189, a C — T substitution at nt 16,192, and a deletion at nt 16,203. Identical mutations were seen in Individual 87.17.22 from the upper mound cap. The alignment for Individual 87.17.10 revealed a unique T --~ C mutation at nt 16,126 as well as the deletion at nt 16,203. A literature search revealed that the substitution at nt 16,189 is specific to Native American 1-laplogroups B and X. Haplogroup B first appeared in central Asia 60,000 yrs ago while Haplogroup X appeared in western Asia 30,000 yrs ago. The substitution at nt 16,192 is indicative of East Asian origin and specific to Japanese populations. To our knowledge the deletion at nt 16,203 has not been reported before in a Native American, thus its presence was thought to indicate kinship between the individuals. However, an alignment of the ancient consensus sequences to that of the three investigators revealed that the deletion was present in two of the modern samples and was not novel. Nothing is known about the substitution at nt 16,126. Further sequencing downstream of nt 16,221 is needed to identify additional mutations characteristic of Haplogroup B or X and a novel kinship marker.

Abstract:
The scope of the current thesis is the investigation of the seismic behavior of complex structures of soccer stadium in the city of Adana, Turkey. The main structural parts of the building of the stadium are reinforced concrete tribunes, steel façade and cable-stayed roof, resting on the structures of the façade. The behavior of the coupled model in case of seismic excitation is studied. The coupled model is defined as a structural model that contains together all main structural parts: the tribunes, the façade and the roof. In the engineering practice, a large-span roof and supporting substructures (façade, tribunes) are differentiated and analyzed separately. In case when the seismic analysis has to be done, the separated analysis has to be performed with caution and possible amplification effect of the seismic action for the structural model with all main structural blocks introduced has to be taken into account. The amplification effect is defined as the increase of the displacements, total base shear and internal forces in the structural elements. To investigate the extent of the amplification effect, the results obtained from the analysis of coupled and uncoupled models were compared. The results obtained from the modal spectral response analysis and nonlinear dynamic analysis of different structural models show a significant amplification of the displacements of the roof structure and façade bearing columns for a coupled model, with the subsequent increase of the total base shear transferred to the substructures form the roof and internal forces in the elements. Methods of improving the structural behavior for the seismic excitation have been investigated. In particular, the effect of installation of seismic isolators for the roof structures has been studied. For the structural models with the isolated roof, the results of the analysis of both coupled and uncoupled models have been improved and all design requirements have been verified.

En el ámbito médico actual se considera cada vez más importante el incluir la valoración del paciente en la toma de decisiones. Los cuestionarios que evalúan la calidad de vida percibida y directamente expresada por los pacientes (“Patient Rated Outcome Measures” – PROMs), son herramientas muy útiles que permiten al clínico conocer el grado de afectación subjetivo de una patología, así como tener en cuenta al paciente y sus síntomas, y no sólo la enfermedad; para poder hacer más realistas las expectativas del paciente con respecto a los resultados. Hoy en día, la utilización de estos cuestionarios en la práctica clínica diaria es de capital importancia. Su uso juega un valiosísimo papel en la comprensión e identificación de la enfermedad, individualizar y definir los objetivos del tratamiento, priorizar los problemas principales del paciente; e incluso, poder medir la posterior respuesta. También permite la elaboración de guías, indicaciones y protocolos o evaluar actividad asistencial. Utilizar PROMS posibilita y facilita la realización de auditorías comparativas, como el cotejo de la prestación de asistencia sanitaria por diferentes proveedores, la comparación entre diferentes tratamientos, y por lo consiguiente, ayudar a mejorar la futura prestación de asistencia sanitaria. Con todo ello, las PROMs se consideran como elementos cruciales para garantizar en la actualidad y en el futuro, la calidad asistencial. Previo a las PROMs, la mayoría de los estudios sobre indicación de cirugía para las amigdalitis recurrentes, evaluaban sólo la frecuencia de las infecciones como medida de resultado; o en los casos de clínica obstructiva de hipertrofia adeno-amigdalar, evaluaban la mejoría sólo con estudios respiratorios del sueño. La toma de decisiones médicas basándose solamente en estos datos, ha dado lugar a una amplia variabilidad en la indicación quirúrgica entre diferentes comunidades y a cierta controversia. Sin embargo, a día de hoy, existen numerosos cuestionarios de salud específicos de patología adeno-amigdalar que miden distintas variables, aunque ninguno de ellos está validado en lengua española. Este trabajo se ha centrado en la validación y adaptación de dos PROMs, el TAHSI-s (versión española del Tonsil and Adenoid Health Status Instrument) y el T-14-s (versión española del Paediatric Throat Disorders Outcome Test – T-14). Ambos cuestionarios han demostrado ser instrumentos válidos, fiables y sensibles para su aplicación clínica. Son herramientas útiles para su uso en ensayos clínicos y en investigación de servicios de salud, para asegurar la calidad asistencial, y para la atención clínica de rutina, por lo que su uso es recomendado.
In the current medical field, it is considered increasingly important to include the assessment of the patient in decision making. Questionnaires that assess the quality of life perceived and directly expressed by patients (“Patient Rated Outcome Measures” – PROMs), are a useful evaluation tool that allows the clinician to comprehend the subjective affectation of a pathology, as well as having consider the patient and his symptoms, and not only the disease; in order to be able to make the patient’s expectations more realistic with respect to the results. Nowadays, the daily clinical use of these questionnaires is of paramount importance. Its use plays an invaluable role in understanding and identifying the disease, identifying and defining treatment objectives, prioritizing the patient's main problems; and even, to be able to measure the subsequent response. It also allows guidelines, indications and protocols’ development and evaluate care activity. Using PROMS enables and facilitates the performance of comparative audits, such as the comparison of the provision of health care by different providers, the comparison between different treatments, and therefore, help to improve the future provision of health care. With all this, the PROMs are considered crucial elements to guarantee at present and in the future, the quality of care. Prior to PROMs, most of the surgery indication’ studies for recurrent tonsillitis evaluated only the frequency of infections as an outcome measure; or in cases of obstructive symptoms of adeno-tonsillar hypertrophy, they evaluated the improvement only with respiratory sleep studies. Medical decision-making based solely on these data has led to a wide variability in the surgical indication among different communities and to some controversy. However, to date, there are numerous health questionnaires specific to adeno-tonsillar pathology that measure different variables, although none of them is validated in Spanish language. This work has focused on the validation and adaptation of two PROMs, TAHSI-s (Spanish version of the Tonsil and Adenoid Health Status Instrument) and T-14-s (Spanish version of the Pediatric Throat Disorders Outcome Test - T-14). Both questionnaires have proven to be valid, reliable and sensitive instruments for their clinical application. They are useful tools for use in clinical trials and health service research, to ensure quality of care, and for routine clinical care, so their use is recommended.

Den belyser främst hur ett äktenskapsarrangemang går till och huruvida inflyttningen från landsbygd till städer har påverkat dem. Jag använder mig av ett inifrånperspektiv genom att intervjua turkiska kvinnor men gör även kopplingar till artiklar som belyser just moderniseringens effekter på äktenskapsformer.


This essay is about marriages in an urban setting in contemporary Turkey. It exemplifies how a marriage arrangement is done and whether the migration into cities from the country has influenced them. I will mainly give an insider perspective by interviewing Turkish women but the essay also relates to articles about modernization’s effects on marriage forms.

Nos últimos anos a arquitetura, aliada à produção vitivinícola, tem sido constante objeto de renovação e transformação. Existem, já, diversas referências relacionadas com a intervenção de autores consagrados, tanto no panorama nacional como no internacional, associados ao universo da arquitetura contemporânea. É a necessidade de reestruturação e adaptação às novas tecnologias de produção, de divulgação da marca, da aposta no Enoturismo, que tem impulsionado a procura de arquitetos mediáticos. Quando se considera a criação de novos edifícios ou renovação de equipamentos existentes, associados a uma marca, e inseridos num determinado contexto, é fundamental ter como base na conceção do objeto arquitetónico os valores patrimoniais, a capacidade de entender o que se deverá manter ou o que possui capacidade de manutenção pela sua qualidade arquitetónica ou pelo seu valor associado à marca. Há que considerar a sua autoria, a época em que a estrutura foi construída e, também, a sua capacidade de adaptação tecnológica, sendo estes tipos de valências que, surgindo sob diferentes prismas, vão definir a filosofia da intervenção. O cooperativismo é de uma enorme pertinência na relação com os agricultores da região, assim como o desempenho destes para a laboração qualitativa do produto, e consequente contribuição económica. Na análise e compreensão das referências nacionais e internacionais, serão tidos em consideração diversos pontos fundamentais, como a reflexão histórica desde os primórdios, em que alguns dos casos vão primar pela produção artesanal como algo exclusivo, enquanto outros casos vão assentar na produção massiva, associada à exportação e à adaptação polivalente das várias vertentes do mercado, tal como a exploração turística, o alojamento, a restauração, o Enoturismo, num compromisso entre a sustentabilidade e os valores apelativos para a região

Les virus adéno-associés (AAVs) sont des virus à ADN simple brin, nonenveloppés, considérés comme des candidats de choix pour la thérapie génique. Pour augmenter les chances de succès des thérapies géniques basées sur l’AAV, des efforts sont actuellement faits pour développer de nouvelles capsides virales, qui seraient plus résistantes à l’immunité préexistante, plus spécifiques de certains tissus, et compatibles avec une production à grande échelle. L’un des défis posés par le développement de nouveaux vecteurs consiste à comprendre comment conférer de nouvelles fonctions biologiques aux capsides d’AAVs, sans compromettre leur intégrité structurale. Pour ce faire, il est nécessaire d’améliorer notre compréhension des mécanismes gouvernant la métastabilité des capsides d’AAVs. L’objectif de cette thèse était d’étudier la thermostabilité des AAVs, ses liens avec leurs propriétés biologiques, ainsi que ses applications dans le domaine du contrôle qualité des préparations d’AAVs recombinants Dans un premier temps, nous étendons les travaux existants à l’étude de virus AAVs ancestraux (AncAAVs), reconstruits in silico. Nous montrons que Anc80, l’ancêtre commun prédit d’AAV1, 2, 8 et 9, est plus thermostable que ses descendants (ΔT = 15-20°C). Nous identifions ensuite, par une analyse de type phénotype-phylogénie, 12 acides aminés jouant potentiellement un rôle important dans la stabilisation des capsides virales. Nous montrons ensuite que la thermostabilité des capsides d’AAVs, mesurée par fluorimétrie différentielle à balayage (DSF), est utile pour déterminer, à l’échelle protéique, l’identité des préparations de vecteurs viraux, une opération requise par les agences réglementaires. Pour finir, nous appliquons ce test d’identité à l’étude de l’homogénéité structurale des librairies d’AAVs. Ces travaux de thèse pourraient s’avérer utiles pour développement et le manufacturing de nouveaux AAVs recombinants pour la thérapie génique
Adeno-associated virus (AAV) vectors have emerged as promising gene delivery vehicles for gene therapy. To improve the probability of success of AAV-based therapeutic strategies, efforts are currently being made to engineer novel capsids able to produce and purify well, escape pre-existing immunity, and target specific cell populations more efficiently. One challenge in AAV vector engineering is to understand how to confer new functions to the viral capsid without altering its structural integrity. To do so, there is a critical need to gain further knowledge on the mechanisms steering AAV capsid metastability. The objective of this thesis is to investigate the thermal stability of AAVs, its impact on AAV biology, and applications to quality control of AAV preparations. First, we extend existing thermal stability studies to in silico reconstructed ancestral AAV particles (AncAAVs), and show that, Anc80, the common putative ancestor of AAV1, 2, 8 and 9, is 15-20°C more thermostable than its contemporary homologs. Using phenotype-tophylogeny mapping, we also identify a set of 12 residues potentially playing a key role in capsid metastability. Le voisinage en Turquie tient lieu d’institution. Cette thèse se propose d’étudier les relations de voisinage en Turquie en les abordant suivant l’approche de l’analyse de réseaux sociaux. Elle étudie ainsi le réseau complet formé par les 101 habitants de quatre immeubles d’un quartier d’Adana. L’objectif de la recherche est de voir comment les relations de voisinage se réalisent et s’organisent entre leurs différents acteurs, et, en dégageant la structure du réseau de voisinage, de déterminer ce qui assure sa cohésion. Une typologie des modes de voisiner, élaborée en suivant la distinction de Granovetter entre liens faibles et liens forts, permet de pondérer les relations et est à la base d’une analyse de réseau valué. Les relations s’inscrivent dans différents espaces qu’elles permettent aux habitants de s’approprier. Elles sont de plus fortement influencées par les variations saisonnières. Le temps joue un rôle dans la construction d’une histoire commune des voisins et l’évolution du réseau. Des caractéristiques individuelles déterminent par ailleurs la propension à voisiner. On observe ainsi des différences de modalités ainsi que de structure entre les réseaux de voisinage masculin et féminin. Les femmes au foyer sont au centre du réseau de voisinage. Ce sont notamment leurs réunions régulières qui permettent l’extension et la cohésion du réseau. L’étude d’un réseau complet permet de porter un nouveau regard sur les relations de voisinage qui ne sont pas considérées comme un ensemble de relations interindividuelles mais sont en Turquie à la base d’une communauté de voisinage
Neighbor relationships are an important institution in Turkey. This thesis intends to study neighbor relationships in Turkey by following the social network analysis approach. It studies a complete network composed of 101 inhabitants of four apartment buildings in a neighborhood of Adana. The aim of the research is to see how neighbor relationships take shape and are organized between different actors, and, by bringing out the neighborhood network structure, to determine what maintains the network’s cohesion. A typology of neighbor relationships, elaborated according to Granovetter’s distinction between weak and strong ties, permits valuing the relations and is at the root of a valued network analysis. The network analysis allows determining a prototype of Turkish sociability. Neighbor relationships occur in different spaces and permit the inhabitants to appropriate them. They are furthermore highly influenced by seasonal variations. Time also plays a role in the construction of a neighbors’ common story and in the network evolution. Moreover some individual characteristics determine the propensity to entertain neighbor relationships. We observe thus some differences of modalities and structure between men and women neighbor networks. Housewives are at the core of the network, notably due to their regular meetings which permit the extension and the cohesion of the network. The study of a complete network is a new way to consider neighbor relationships, which more than the whole of inter-individual relations, are at the root of neighborhood communities in Turkey

Pendant la colonisation anglaise (1839-1967) et la période du socialisme (1969-1990) les femmes d' Aden ont acquis de nombreux droits. L'unification des deux Yémen en 1990 a été suivie de l'application de la Charia, comme la base de toutes les lois, et d'un changement brutal (voilement systématique des femmes, repli sur les rôles « traditionnels » de femmes au foyer, séparation des hommes et des femmes dans de nombreux lieux publics, etc. ) Cette thèse analyse l'ensemble de ces mouvements. Tout d'abord, elle retrace l'histoire et les représentations du rôle et du statut des femmes dans les sociétés arabo-musulmanes sur la Péninsule Arabique. Puis, elle analyse l' évolution de la place des femmes adénites (de deux générations) depuis la période dite socialiste en passant par l' unification jusqu' à nos jours. La conformation des attitudes et des comportements des femmes adénites aux attentes des hommes n'apparaît pas seulement comme l'effet d'une contrainte. C'est aussi le produit d'une « conviction réfléchie ». Les femmes participent à ce renouveau «traditionaliste» et elles y trouvent même un certain intérêt personnel, une marge de liberté.

Design of tall and long span structures is governed by the wind forces. Inadequate research in the field of wind dynamics has forced engineers to rely on design codes or wind tunnel tests for sufficient data. The present work uses a computational wind dynamics method to compare the coefficient of pressure (Cp) for the different aerodynamic shapes. ADINA, a finite element package, contains an inbuilt turbulence model which will be used to construct four different shapes for comparison. Results are verified with the experimental and simulation data. The effect of increase in the Reynolds number on the flow has been studied. Graphs for the pressure, velocity and turbulence energy distribution have been developed to assist the engineers in design.

Adeno-associated virus type 2 (AAV-2) integrates preferentially into a defined site on human chromosome 19, and has been developed as a gene therapy vector. We propose to use this unique recombination event for site-specific integration of transgenes in plants. This strategy would alleviate problems associated with current plant transformation methods that integrate transgenes randomly. The AAV-2 gene encoding the enzyme that catalyzes the insertion (rep78) was introduced into Arabidopsis thaliana via Agrobacterium-mediated transformation. PCR and sequence analysis confirmed the presence of rep78 in two plant lines. RT-PCR demonstrated rep78 transcription in one plant line, but protein could not be detected in either line.

O presente trabalho tem como objetivo apresentar as bases para a construção duma adega com preocupações ambientais na região do Algarve. Nesta adega pretende-se produzir vinhos comerciais brancos, tintos e rosés bem como recuperar estilos de vinhos tradicionais como os vinhos de talha, de flor e o moscatel. O edifício projetado teve como base princípios de eficiência energética, poupança de água e de sustentabilidade ambiental. A existência de um grupo de painéis fotovoltaicos com a capacidade de produzir a energia que é consumida (balanço energético nulo), protocolos de vinificação orientados para uma melhor eficiência ao nível do consumo de água e ainda o método de tratamento dos resíduos, são os fatores essenciais para que se possa ter uma adega segundo o conceito de “eco-adega”. O aproveitamento da força gravítica para movimentação de uvas, massas e vinhos foi outro dos pontos de eficiência energética desta adega. Em conjunto, selecionaram-se materiais isolantes (esferovite e caixa de ar) tanto nas estruturas de cimento como nas estruturas metálicas para que a dissipação de calor do interior para o exterior e vice-versa pudesse otimizar o uso de energia. O desenho apresentado foi pensado para ser o mais eficiente e simples possível ao nível da laboração conjugado com a possibilidade de visitas por parte dos turistas da região, para além da venda ao público na loja da adega. Os cálculos de dimensionamento previram uma capacidade total de fermentação de vinhos de 100.000 litros, num investimento inicial de 1.437.813 €. A amortização anual foi de 100.770 €, ao longo de 11 anos. Os custos variáveis foram previstos em 329.333 € o que no final corresponde a uma despesa anual de 430.103,9 €. Relativamente à faturação, espera-se que ao final dos 11 anos atinja o valor de 466.000 € anuais. Os limiares de rendibilidade e encerramento situam-se nas 76.603 garrafas e nas 60.540 garrafas, respetivamente, considerando um valor médio de venda de 5,44 €/garrafa

A globalização quase plena dos mercados, alavancada pelos novos recursos tecnológicos, particularmente ao nível das tecnologias da informação e comunicação, levam à rápida disseminação das alterações económicas, sociais e culturais de forma global, impactando, influenciando e afetando os consumidores e os ambientes contextuais e transacionais das organizações e, dessa forma, as empresas que neles operam. Não sendo a globalização “novidade” para a indústria portuguesa dos vinhos, tendo mesmo sido esta indústria uma das pioneiras nessa vertente ao nível da exportação, a integração na então C.E.E., agora União Europeia, com a Política Agrícola Comum, P.A.C., em conjunto com a abertura dos mercados, representaram grandes alterações para esta indústria e novos desafios para a mesma. Com cerca de 300 associados e uma produção anual em torno dos 8 milhões de litros de vinho que geram um volume de vendas/ano de cerca de 6 milhões de euros, a Adega Cooperativa de Palmela, embora com relativo peso económico no panorama nacional, é uma das maiores empresas vitivinícolas na Península de Setúbal, tendo um enorme peso na economia local, particularmente na agrícola, sendo mesmo vital para muitos viticultores. Today we are living, not only but also in economic terms, perhaps the most turbulent and volatile times in the history of mankind. The almost full globalization of markets, leveraged by new technological resources, particularly at information and communication technologies level, leads to a fast global spread of economic, social and cultural changes, impacting, inducing and affecting consumers and organization's operating and external environments and, therefore, the companies operating in them. The globalization is not "newness" for the Portuguese wine industry as this industry is a globalization’s pioneering industry, at least at the export level. However, the integration in the former EEC, now European Union, with the Common Agricultural Policy, CAP, along with the markets opening, it represents major changes for this industry and brought new challenges to it. Having about 300 partners, an annual wine production of circa 8 million liters generating an annual turnover circa 6 million euros, the Adega Cooperativa de Palmela, although with relative economic weight in the national outlook, is one of the largest wine companies in the Península of Setúbal, having a huge influence in the local economy, particularly in the agricultural sector, being even vital for many winegrowers. Established in 1955, the Adega Cooperativa de Palmela has faced various market changes over time. However, how does Adega Cooperativa de Palmela serves its consumers and their needs? How satisfies them? What marketing practices they have? "The Marketing Practices at the Adega Cooperativa de Palmela" is thus the theme of this Master's Thesis where, using the case study methodology, after a bibliographic review and using documentary analysis, data and observations and interviews with "privileged witnesses", we will analyze the sort of marketing practices the Adega Cooperativa de Palmela is performing.

Adeno-associated virus (AAV) infection triggers a DNA damage response in the cell. This response is not induced by viral proteins but by the structure of AAV ssDNA being recognized by the cell as damaged DNA. The consequence of this is the killing of cells lacking p53 activity. We have observed that cells that lack p21 or pRb activity are also sensitive to AAV-induced cell death. We report that cells respond to AAV infection by activating two DNA damage-signaling cascades. The first activates the p84N5 protein, which in turn activates caspase-6, leading to cell death. The second cascade activates the p53-21-pRb pathway, which inhibits activation of the p84N5 protein and thus prevents cell death. The result of the antagonistic interaction between these two pathways is that cells that do not exhibit functional p53-p21-pRb signaling undergo apoptosis as a consequence of AAV infection. Cells with a functional p53- 21-pRb pathway are refractory to AAV-induced cell death. These results show that p53, although a pro-apoptotic protein, together with pRb and p21 proteins, is a member of an anti-apoptotic cellular mechanism. As such, these experiments reveal features that may be exploited to specifically kill cells that lack the p53-p21-pRb pathway, such as cancer cells. We also consider the role of DNA damage signaling in the restriction of AAV replication. We report that helper viruses may inhibit ATM, ATR and DNA-PK signaling and that this may be advantageous for AAV replication. Inhibition of ATM and DNA-PK in tissue culture cell lines led to an increase in AAV replication. We show that ATM, ATR and DNA-PK may be responsible for the hyperphosphorylation of replication protein A (RPA) in response to AAV infection. We speculate that hyperphosphorylated RPA may be a fundamental restriction factor for viral replication and delineate novel experimental tools to test such a hypothesis. The implications of these observations for oncolytic therapy are discussed.

The Gulf of Aden holds one of the strategic chokepoint along the important Eurasian maritime trade route. On both sides of the Gulf of Aden lie countries in need of political stability. In the south lies Somalia, one of the worlds longest failed states and hosts of pirates violently disrupting maritime trade in the region. On its northern shores lies Yemen, one of the poorest countries in the world and home to the terrorist movement Al-Qaeda in the Arabian Peninsula. This thesis aims to look deeper into what effect on piracy another failed state in the region would have and to illustrate that the thesis describes a scenario where Yemen has followed Somalia’s footsteps and developed into a failed state. Applying Lindgren and Bandhold’s method of scenario planning and through a quantitative analysis, recent trends affecting piracy development were identified. Through a scenario cross four different scenarios were developed to illustrate the effects a failed state Yemen would have on piracy leading to which new challenges the EU might be faced with in the region. The overall conclusions are that a failed state Yemen would have a negative effect on the efforts in mitigating piracy and pirates, driven by opportunity and profit, would benefit from further instability in the region. The thesis supports previous research regarding the connection between maritime piracy in the Gulf of Aden and failed states and illustrate the multifaceted challenges the EU could face as a consequence of the new development of piracy.

La littérature décrit des différences fondamentales entre l’AAV2 sauvage et ses pendants recombinants. La forme sauvage serait plus efficiente en terme de production, d’encapsidation et d’infectieusité, allant de facteurs de deux à cents en fonction de la propriété étudiée. A cause de ces différences, la quantité de rAAV nécessaire pour traiter un patient atteint d’une maladie implique une injection systémique estimée à 1.1015 particules par kilogramme de tissue à traiter. C’est dans cette optique que s’inscrivent mes travaux de thèse. Essayer de comprendre qu’elles sont les différences entre l’AAV sauvageet les rAAV qui peuvent engendrer tant de différences en terme d’efficacité. L’étude du comportement du génome de l’AAV2 sauvage dans le système baculovirus/cellules Sf9, a permis de découvrir que la régulation de l’AAV2 sauvage était similaire en baculovirus. Nous avons aussi découvert, grâce à notre analyse transcriptomique que les promoteurs naturels del’AAV2 étaient actifs dans notre système, ce qui nous a permis d’imaginer de nouvelles constructions génétiques afin d’améliorer la quantité et la qualité des particules rAAV. Nous avons aussi réaliser des études structurales sur différentesparticules AAV afin d’améliorer notre connaissance de ces particules
The literature describes several fundamental differences between WT AAV and rAAP properties. WT form obtains an one hundred higher production yield compared to rAAV, with the possibility to obtain only full particles, and most importantly, all the WT AAV particles are infectious, compared to only 1% for the rAAV. These lower values for rAAV, implied to inject up to 1.1015 particles per kilogram of tissue. These quantities induce a non-negligible cost for rAAV based gene therapies, even with the productions techniques improvements or the development of baculoviruses based techniques. It is with these ideas in mind that my PhD works were developed. Trying to understand differences between WT and rAAV, trying to produce WT AAV in baculovirus has bring important knowledges about AAV comportment in baculovirus. Our RNA-Seq results have demonstrated than the WT AAV natural promoters were all active, allowing us to design to genetic constructs in order to improve rAAV quantity and quality. We have also tried to solve the AAV crystal structure, to improve our knowledges about these particles

Resumo: Neste trabalho preparamos e utilizamos nanopartículas de PLGA-DOTAP encapsulando o peptídeo P17, que bloqueia a ação da citocina imunossupressora TGF-b, no tratamento de camundongos BALB/c fêmea, inoculados via subcutânea com células de hepatocarcinoma (CHC) murino BNL ou de câncer colo-retal (CCR) murino CT26. No modelo de CHC, nanopartículas foram administradas em conjunto com vírus adenoassociados recombinantes, cujo DNA viral codificava a citocina imunoestimuladora IL-12. No modelo de CCR, nanopartículas foram testadas como potencializadoras de uma vacina contra câncer colo-retal. Além disso, a biodistribuição de nanopartículas de PLGA-DOTAP, funcionalizadas ou não com a glicoproteína assialofetuína, foi verificada pelos métodos de bioluminescência em câmara escura com detector CCD e por PCR quantitativa em tempo real. Como resultado, observou-se que nanopartículas de PLGA-DOTAP encapsulando o fármaco P17 são capazes de potencializar a vacina contra CCR e que a glicoproteína assialofetuína é capaz de direcioná-las ao fígado
Abstract: In this research we prepared and used PLGA-DOTAP nanoparticles loaded with the peptide P17, which blocks the action of the imunossupressive cytokine TGF-b, in the treatment of female BALB/c mice inoculated subcutaneously with BNL (murine hepatocellular carcinoma (HCC) cells) or CT26 (murine colorectal cancer (CRC) cells). In the HCC model, nanoparticles were administered together with recombinant adeno-associated virus whose viral DNA encodes the immunostimulatory cytokine IL-12. In the CRC model, nanoparticles were tested as vaccine booster against colorectal cancer. In addition, the biodistribution of PLGA-DOTAP nanoparticles functionalized or not with the asialofetuin glycoprotein was verified by bioluminescence and real-time quantitative PCR. As a result, it was observed that P17 containing DOTAP-PLGA nanoparticles are able to increase vaccine efficiency and that asialofetuin glycoprotein is able to direct them to the liver

Desafortunadamente la noción de ética –así como los problemas que ésta analiza– ha sido distorsionada por la sociedad de nuestros días, ello ha provocado que el individuo común confunda ética con moral y viceversa. Por lo anterior, considero que es relevante que la filosofía moral sea estudiada en todos los niveles educativos, ya que únicamente un estudio minucioso y constante sobre la ética y la moral nos ayudará a percatarnos de que éstas no son impedimento para forjar un carácter individual y, por otro lado, entenderíamos por qué éstas son indispensables para desarrollar aptitudes que nos permitan desenvolvernos armoniosamente dentro de la sociedad.

The general subject of this thesis is about farm labor intermediaries who are important not only because they are transferring and organizing seasonal agricultural work but also they have social relations and interactions with other actors of seasonal agricultural work. This analysis elaborates multi dimensional relations of farm labor intermediaries and construction and reproduction of these relations. These social relations which can be accepted also as social capital and their nature are important for both understanding farm labor intermediaries and seasonal agricultural work in Turkey. Adana is determined as research field because of its long history about seasonal agricultural work. Moreover, changes and transformations in terms of social and agricultural structure taking place in Adana for 20 years are very important factors with regard to understand how farm labour intermediaries reproduce social relations. Labor intermediaries are the people who are providing and organizing seasonal labor force demanded by employers in labor intense agricultural production in different regions of Turkey. Delegate, backer, or sarge is the some local names of intermediaries using in Turkey. As it can be understood from the function of labor intermediary, it is arbiter between employer and worker. Family, relatives, friends and neighbors are seen as the labor force resources of intermediaries. From the eyes of the seasonal agricultural workers, intermediaries are not only the people who are finding job for them but also they are the people who meet their basic needs. This is very crucial point, since seasonal agricultural labor force is supplied from very poor regions in Turkey. At the same time intermediaries are important actors for employers due to their controlling functions of agricultural work process. However, the relationship between intermediary and employer is simply defined as business where mutual interests are important. In this context labor intermediation in Turkey has lots of meaning other than finding job for workers and worker for employers. Labor intermediation is a legal job which is controlled and regulated by law. However, both pervious researches and this study demonstrate that this job is mostly executed as informal. The main determinant factor for this informal execution of the job can be related with working style of intermediaries, namely, face to face social relations that has been institutionalized within time.

This thesis aims at assessing the innovation performance of the firms and the factors affecting their performance by comparing agglomerated and non-agglomerated firms in the TR62 (Adana, Mersin) Region. Adana Organised Industrial Zone (AOSB) and Mersin-Tarsus Organised Industrial Zone (MTOSB) are selected as agglomeration examples in the region to study the effects of spatial proximity on innovativeness. It is found that local specific policies such as the establishment of an integrated system for cooperation and competition are needed to improve the innovation and competition capacity of the region.

The Adeno-Associated Virus(AAV) is a small, single stranded DNA virus that has been developed as a gene transfer vector. Early clinical trials using recombinant AAV vectors (rAAV) have identified the following concerns that need to be addressed in order to increase efficiency of these vectors. It has since been determined that AAV vector efficiency decreases due to the following mechanisms: ineffective endocytosis, endosomal degradation, inefficient trafficking, and the need to convert from a single-stranded AAV genome to a transcriptional active double-stranded form. The purpose of this study is to elucidate mechanisms that help stabilize the AAV genome in order to make it a more efficient vector for gene therapy. Previously, there have been studies using fluorescent labeling to track the movement of AAV into the nucleus. An integral part of AAV genomic stability may be the obstacles it encounters in the cytoplasm prior to entering the nucleus. Previous studies on improving AAV transduction have focused primarily on the nucleus. The study will hopefully shed light on the hurdles AAV encounters as it moves through the cytoplasm. Thus, this project has designed and utilized a new system for specifically tracking the status of AAV genomes in the cytoplasm as well as in the nucleus. This project utilizes a novel dual luciferase reporter system to track the movement of AAV particles from the cytoplasm into the nucleus to elucidate mechanisms that could contribute to the stabilization of the AAV genome. The novel dual reporter system is comprised of a single-stranded vector containing two different types of secreted luciferases: Cypridina Luciferase and Gaussia Luciferase. Cypridina Luciferase is placed under the control of a nuclear promoter and therefore it is expressed only in the nucleus. The second, Gaussia Luciferase, is under the control of a cytoplasmic promoter that will only be expressed in the cytoplasm upon the presence of T7 RNA polymerase. Using this dual reporter luciferase system along with RT qPCR quantification in a Hek293 cell line expressing the T7 RNA polymerase, demonstrated that genomes are present in the cytoplasm at 18 and 24 hours post infection. The second part of this study is using the dual reporter system to understand the trafficking patterns of rAAV and looking at ways to enhance transduction. One method could be administering rAAV vectors in conjunction with a drug; this approach may help overcome some of these cellular barriers encountered during infection as well as help stabilize the genome. Cidofovir (CDV) is a monophosphate nucleotide analogue that competitively inhibits the incorporation of deoxycytidine triphosphate into viral DNA via viral DNA polymerase. In vitro, CDV actively inhibits a number of DNA viruses including herpes viruses, adenovirus, polyomavirus, papillomavirus, and poxviruses. Cidofovir has already been approved by the Food and Drug Administration (FDA) for the treatment of cytomegalovirus (CMV) retinitis in patients with acquired immunodeficiency syndrome (AIDS). The effects of CDV on small DNA viruses that lack their own viral DNA polymerase, like AAV, has not been documented. Results have demonstrated that CDV is able to increase single-stranded rAAV transgene expression in the nucleus by 2 to 5 fold, depending on the cell type and concentration, in vitro, using both rAAV2 and rAAV8 luciferase reporter vectors. These results have been able to replicated using other reporter vectors: rAAV2-LacZ reporter, rAAV2-GFP, rAAV2-hAAT, and a rAAV8-GFP in vitro. Results have shown a dose-dependent increase in rAAV genomes with CDV pretreatment in HelaS3 cells via Southern Blot. Also southern blot analysis of cells pretreated with CDV then infected with rAAV revealed no difference in the amount of vector present between 0 and 2 hours post infection, suggesting CDV does not enhance viral entry but that CDV may enhance at steps downstream of viral entry. Using RNA sequencing and Ingenuity software analysis of HelaS3 cells pretreated with CDV, an increase in several genes of interest including those involved in the mechanism of viral exit were observed. These genes include Actin and vacuolar proteins, these molecules are involved in and associated with endosomal sorting complexes as well as required for transport inside the cell. This finding along with southern blot data supports the theory that CDV may enhance rAAV trafficking since we observed that CDV pretreatment enhances viral accumulation of rAAV vectors in both the cytoplasm and nucleus 24 hours post-infection. Also utilizing the dual luciferase reporter system an increase in transgene expression present with CDV pretreatment compared to PBS in both the cytoplasm and nucleus was observed suggesting that CDV may enhance with rAAV trafficking. These results taken together demonstrate that this dual reporter system is a powerful tool for understanding and improving rAAV trafficking. Also drugs like CDV can greatly contribute to the understanding of rAAV trafficking, and eventually lead to the development of novel strategies to increase overall efficiency of AAV transduction.
