Abstract:
We have engineered a mouse mutation that specifically deletes most of the RXRα N-terminal A/B region, which includes the activation function AF-1 and several phosphorylation sites. The homozygous mutants (RXRαaf1o), as well as compound mutants that further lack RXRβ and RXRγ, are viable and display a subset of the abnormalities previously described in RXRα-null mutants. In contrast, RXRαaf1o/RAR−/−(α, β or γ) compound mutants die in utero and exhibit a large array of malformations that nearly recapitulate the full spectrum of the defects that characterize the fetal vitamin A-deficiency (VAD) syndrome. Altogether, these observations indicate that the RXRα AF-1 region A/B is functionally important, although less so than the ligand-dependent activation function AF-2, for efficiently transducing the retinoid signal through RAR/RXRα heterodimers during embryonic development. Moreover, it has a unique role in retinoic acid-dependent involution of the interdigital mesenchyme. During early placentogenesis, both the AF-1 and AF-2 activities of RXRα, β and γ appear to be dispensable, suggesting that RXRs act as silent heterodimeric partners in this process. However, AF-2 of RXRα, but not AF-1, is required for differentiation of labyrinthine trophoblast cells, a late step in the formation of the placental barrier.

Abstract:
ABSTRACT Estrogenic responses are now known to be mediated by two forms of estrogen receptors (ER), ERα and ERβ, that can function as homodimers or heterodimers. As homodimers the two have been recently shown to exhibit distinct transcriptional responses to estradiol (E2), antiestrogens, and coactivators, suggesting that the ER complexes are not functionally equivalent. However, because the three possible configurations of ER complexes all recognize the same estrogen response element, it has not been possible to evaluate the transcriptional properties of the ER heterodimer complex by transfection assays. Using ER subunits with modified DNA recognition specificity, we were able to measure the transcriptional properties of ERα-ERβ heterodimers in transfected cells without interference from the two ER homodimer complexes. We first demonstrated that the individual activation function 1 (AF-1) domains act in a dominant manner within the ERα-ERβ heterodimer: the mixed agonist-antagonist 4-hydroxytamoxifen acts as an agonist in a promoter- and cell context-dependent manner via the ERα AF-1, while activation of the complex by the mitogen-activated protein kinase (MAPK) pathway requires only the ERα- or ERβ-responsive MAPK site. Using ligand-binding and AF-2-defective mutants, we further demonstrated that while the ERα-ERβ heterodimer can be activated when only one E2-binding competent partner is present per dimer, two functional AF-2 domains are required for transcriptional activity. Taken together, the results of this study of a retinoid X receptor-independent heterodimer complex, the first such study, provide evidence of different stoichiometric requirements for AF-1 and -2 activity and demonstrate that AF-1 receptor-specific properties are maintained within the ERα-ERβ heterodimer.

L'acide retinoique (metabolite actif de la vitamine a) joue un role important au cours du developpement et dans la croissance et la differenciation cellulaire. Deux familles de recepteurs des retinoides ont ete caracterisees : les recepteurs de l'acide retinoique, comprenant trois genes (rar, , ) et les recepteurs du retinoide x, comprenant egalement trois genes (rxr, , ). Les rars et les rxrs comprennent, en n-terminale, une fonction activatrice autonome (af-1) et, en c-terminale, une fonction activatrice induite par le ligand (af-2). Au cours du developpement, rxr est le mediateur principal de la fonction des rxrs. Afin de mieux comprendre les fonctions de rxr, nous avons entrepris, chez la souris, l'inactivation specifique de la fonction af-1, d'une part, et de la fonction af-2, d'autre part, de ce recepteur. Les souris chez lesquelles la fonction af-1 de rxr est mutee sont vivantes et fertiles. Ceci montre que l'af-1 de rxr n'a pas de role essentiel dans les multiples fonctions exercees par le recepteur au cours du developpement. En opposition, les homozygotes pour la mutation af-2 (xaf2#0) meurent a la naissance et presentent, avec une frequence faible, la majorite des malformations rencontrees chez les mutants nuls pour rxr. De plus, l'analyse des mutants composes xaf2#0/rxr#-#/#-/rxr#-#/#-a montre pour la premiere fois, que des mutations dans les rxrs seulement reproduisent la majorite des phenotypes observes dans le syndrome de carence foetale en vitamine a et chez les mutants doubles nuls rars. Finalement, les doubles mutants xaaf2#0/rar(, ou ) presentent avec la meme penetrance et/ou severite l'essentiel des phenotypes observes chez les doubles mutants nuls rxr/rar (, ou ) correspondants. L'ensemble de ces resultats nous a permis de montrer le role important in vivo de la fonction af-2 de rxr dans les differents evenements controles par les heterodimeres rar/rxr au cours du developpement. Ces donnees nous suggerent egalement un role crucial du ligand de rxr dans ces differentes fonctions.

Les œstrogènes influencent de nombreuses maladies comme le cancer de l'endomètre et les maladies cardiovasculaires. Leurs actions majeures sont relayées par le récepteur aux œstrogènes ERa, régulant la transcription de gènes cibles via deux fonctions activatrices AF-1 et AF-2. Grâce à l'utilisation de souris invalidées pour AF-1, mon équipe a rapporté que l'AF-1 n'était pas nécessaire pour l'action vasculoprotectrice du 17ß-œstradiol (E2), principal œstrogène naturel. Durant ma thèse, nous avons démontré que : i) le tamoxifène, modulateur sélectif des ERs,protège de l'athérosclérose via AF-1, contrairement à l'effet de l'E2 mais n'accélère pas la ré-endothélialisation, ii) l'AF-1 du ERa est nécessaire pour la régulation de la transcription et de la prolifération cellulaire dans l'utérus en réponse à l'E2 et au tamoxifène. Ainsi, ce travail contribue à la dissection moléculaire de l'action du Era in vivo, qui pourrait poser les bases d'une optimisation de la modulation du ERa
Estrogens influence most of the physiological processes in mammals, in particular reproduction and vascular system. Thus, it is not surprising that these steroid hormones influence numerous diseases, including breast and endometrium cancers, as well as cardiovascular diseases. These actions are mediated by two nuclear receptors, estrogen receptors ERa and ERß, which regulate target gene transcription through two independent activation functions AF-1 and AF-2. Using ERa deficient mice or electively targeting AF-1, my team has previously demonstrated that ERa is required for the E2 vasculoprotective actions of 17ß-estradiol (E2), the main natural estrogen. In contrast, activation of AF-1 of ERa is dispensable for atheroprotection, increase of NO relaxation and endothelial healing. During my phD, we have shown that: 1) tamoxifen, a selective ER modulator, used more than 30 years for breast cancer treatment in premenopausal women, prevents atherosclerosis but not accelerates endothelial healing. This atheroprotective effect is ERa-dependant and in contrast to E2, needs its AF-1, 2) AF-1of ERa is necessary to the regulation of gene transcription and cell proliferation in the uterus in response to E2 or tamoxifen. However a residual uterine hypertrophy after chronic E2 treatment persists in mice lacking AF-1 of ERa, potentially due to a stromal edema resulting to the persistence of Vegf-a induction. Furthermore uterine epithelial apoptosis and response to progesterone are largely altered in these mice. Thus, my PhD contributes to the molecular dissection of ERa activation in vivo, which could pave the way of an optimized ERa modulation. Indeed, the selective activation of ERa inducing beneficial effects of E2 without stimulating deleterious effects represents a major therapeutic interest

Abstract:
Introduction: Atrial fibrillation (AF) is the most common cardiac arrhythmia, with a prevalence of 2% in a general Western population, rising to 22% in patients with heart failure (HF). AF can occur in paroxysms or be present continuously. HF with AF is associated with an increased risk of stroke. Strokes due to AF that occur in HF patients are more severe than those that occur in patients without HF. Oral anticoagulation therapy (OAT) reduces the risk of stroke in patients with AF, although with an increased risk of major bleeding. While the relationship between persistent AF and stroke events in HF patients is well described, there is less clarity about the stroke risk from paroxysmal forms of AF. Paroxysmal AF is commonly described with reference to an ‘AF burden’, but how large a burden of paroxysmal AF warrants therapy with OAT is not known. Remote monitoring of Cardiac Implantable Electronic Devices (CIEDs) can provide a continuous assessment of cardiac rhythm. This offers a new method of describing the burden of paroxysmal AF that occurs in patients with HF. Methods: Remote monitoring data downloads from patients with CIEDs and HF were collected from patients at 2 UK hospitals. 169 patients were analysed in the first centre. Both AF episode data and clinical data were collected. The data from this centre was used to determine the feasibility of the technique, and the burden of AF was described. AF episodes were then collated into ‘AF days’ and the incidence of significant AF days was described with reference to 5 previously used definitions of a significant AF day in previously published work. 327 patients were analysed from a second UK centre with data collected in order to validate the incidence of AF observed in the first centre. Results: In patients from Centre 1, 3,426 episode of AF were detected during remote follow-up in 115 patients over 446 patient years. Paroxysmal AF was detected in 63% of patients who were presumed to have sinus rhythm at the time of implant. The median total AF burden in this group was small (187 seconds; 16 seconds – 7.2 days), but 32% of patients with pAF had a total burden of greater than 1 day. The burden of AF was described in several ways, and exploratory 12 analyses of AF burden was also carried out. The burden of AF was compared to that in the 5 previously published studies of remotely detected AF. One definition (‘≥ 30 seconds of AF in a 24 hour period’) was found to be met by all patients who then also met other definitions, and patients appeared to fulfil this definition of a clinically significant burden of AF sooner. In centre 2, 7,148 episodes of AF occurred in 236 patients over a follow period of 805 patient years. The burden of AF appeared broadly similar to that observed in the first centre, and the comparison with the previously published data was also similar. Conclusions: AF occurs frequently in patients with HF in whom dual chamber CIEDs have been implanted. Many patients spend only a short amount of time in AF, and the clinical significance of this is unclear. A definition of ‘at least 30 seconds in a 24 hour period’ was met by all patients who met other published definitions of a significant burden of AF and was met sooner than the other definitions of significance. Further work is needed to determine if this burden of AF could predict longer burdens and to correlate pAF burden with stroke risk.

This work contains a theoretical analysis of the behavior of plate girders. A plate girder has been calculated based on the criteria of the American standard Manual of Steel Construction Load and Resistance Factor Design (AISC, 1994), and checked by the Brazilian standard NBR 8800: Projeto e execução de estruturas de aço de edifícios (método dos estados limites) (ABNT, 1986), the revision project of the Brazilian standard Projeto de Revisão da NBR 8800: Projeto e execução de estruturas de aço e de estruturas mistas aço-concreto de edifícios (ABNT, 2003), the revision project of the Brazilian standard Projeto de Revisão da NBR 8800: Projeto de estruturas de aço e de estruturas mistas de aço e concreto de edifícios (ABNT, 2007) and the European standard Eurocode 3: Design of steel structures Part 1.1: General rules and rules for buildings (CEN, 1992). PFEIL (1989) has calculated a plate girder by using the Brazilian standard NBR 8800/86. The calculated dimension have been checked by the mentioned standards. NARAYANAN (1992 apud OWENS, KNOWLES E DOWLING, 1992) has dimensioned a plate girder by using the British standard BS 5950: Structural use of steelwork in building. Part 1: Code of practice for design in simple and continuous construction: hot rolled sections (BSI, 1990), which has also been checked by the mentioned standards. Furthermore, theoretical analyses related to the calculus of plate girders presented by many authors have been considered. The main goal is to assess the criteria utilized by the standards for the calculus of plate girders. From the obtained results, for the checked girders, by the standards and related theories, a comparative analysis among the utilized criteria by the standards has been made. The reached conclusion is that the Brazilian standard and its revision projects has identical criteria compared to the American standard for the calculus of plate girders. The European standard presents slightly different criteria compared to the American standard; however, both provide close results one compared to another. The theories utilized have also provided close results from those obtained by the standards.

A partir dos resultados obtidos, para as vigas analisadas, pelas normas e pelas teorias existentes, foi feita uma análise comparativa entre os critérios utilizados pelas normas. A conclusão a que se pôde chegar é que a norma brasileira e seus projetos de revisão apresentam critérios idênticos aos da norma americana para o cálculo de vigas esbeltas. A norma européia apresenta critérios um pouco diferenciados dos critérios da norma americana, mas fornecem resultados aproximados entre si. As teorias utilizadas também forneceram resultados aproximados dos resultados obtidos pelas normas.

Abstract:

The tricyclophosphate ligand, (P₃O₉)³⁻, was investigated as an oxidation resistant ligand for ruthenium chemistry. The silver salt of tricyclophosphate, Ag₃(P₃O₉).H₂O, was found to be a useful synthon in the preparation of organic soluble tricyclophosphate salts. The complex [(P₃O₉)Ru(C₆H₆)]⁻ was prepared and characterized. In water, it was found that the tricyclophosphate ligand dissociated from the [(C₆H₆)Ru]²⁺ dication. Cyclic voltammatry in acetonitrile shows only a small irreversible oxidation at 0.95 V (Vs ferrocene). The rhenium carbonyl complex [(P₃O₉)Re(C0)₃]²⁻ was also investigated as a precursor to high valent tricyclophosphate rhenium complexes. Based on these results as well as related work by Klemperer and coworkers, it is concluded that the tricyclophosphate ligand is an exceptionally hard one that binds only weakly to transition metals.

The preparations of chloro and allyl derivatives of pentamethylcyclopentadienyl aminoborole hafnium complexes are described. The chloro derivative, Cp*[C₄H₄BN(CHMe₂)₂]HfCl•LiCl, is prepared by treatment of Cp*HfCl₃ with Li₂(THF){C₄H₄BN(CHMe₂)₂}. The structures of the chloro derivatives Cp*[C₄H₄BN(CHMe₂)₂]HfCl•LiCl(Et₂O)₂ and {Cp*[C₄H₄BN(CHMe₂)₂]HfCl-LiCl}₂ were determined by single crystal X-ray analysis. Treatment of Cp*[C₄H₄BN(CHMe₂)₂]HfCl•LiCl with allyl magnesium bromide yields Cp*[C₄H₄BN(CHMe₂)₂]Hf(η³-C₃H₅), whose structure was determined by X-ray analysis. The allyl species was active for the polymerization of ethylene, but not for the polymerization of α-olefins. Addition of ligands to the allyl derivative results in the formation of Cp*[C₄H₄BN(CHMe₂)₂]Hf(C₃H₅)(L) (L = PMe₃, pyridine, CO). The structure of Cp*[C₄H₄BN(CHMe₂)₂]Hf(η³-C₃H₅)(CO) was determined. Treatment of Cp*[C₄H₄BN(CHMe₂)₂]Hf(η³-C₃H₅)(CO) with PMe₃ results in the formation of the dieneolate complex Cp*[C₄H₄BN(CHMe₂)₂]Hf(OCHCHCHCH₂) (PMe₃). The electronic spectra of several aminoborole complexes were investigated and the low energy transitions assigned as borole-metal LMCT transitions. The amphoteric complex Cp*{η⁵-C₄H₄BN(CHMe₂)₂}HfCl•LiCl heterolytically cleaves H-X bonds to form Cp*{η⁵-C₄H₄BNH(CHMe₂)₂}HfCl(X) (X = Cl, CCR). Cp*{η⁵-C₄H₄BNH(CHMe₂)₂}Hf(CCTMS)₂ is prepared from Cp*{η⁵-C₄H₄BN(CHMe₂)₂}Hf(η³-C₃H₅) and two equivalents of (trimethylsilyl)acetylene. Control experiments using deuterium labelled substrates show heterolysis occurs with no incorporation of deuterium into the 2,5 positions of the borole heterocycle. The X-ray structure determinations of Cp*{η⁵-C₄H₄BNH(CHMe₂)₂}HfCl₂, Cp*{η⁵-C₄H₄BNH(CHMe₂)₂}HfCl(CCTMS), and Cp*{η⁵-C₄H₃MeBN(CHMe₂)₂}HfClI are reported.

Atrial fibrillation (AF) is the most common sustained cardiac arrhythmia affecting approximately 3 million Americans, and is a prognostic marker for stroke, heart failure and even death [1]. 12-lead electrocardiogram (ECG) is used to monitor normal sinus rhythm (NSR) and also detect AF. Although the persistent form of AF can be detected relatively easy, detecting paroxysmal AF is often a challenge since requiring continuous monitoring, which becomes expensive and cumbersome to collect lot of ECG data [1]. Several researchers have attempted to develop new methods to discriminate NSR and AF which are based on R-R interval analysis, linear methods, filtering, spectral analysis, statistical approaches such as entropy etc. which faces limitation of successfully detecting AF of all types with high sensitivity and specificity using short time ECG data [1–3]. The major issues with these approaches is that they often distort the ECG by several pre-processing steps with filters, do not provide reliable discrimination using short ECG time series data and many of them lack real-time capability that makes it difficult to trust the data for diagnosis and treatment. Both clinical and scientific communities recognize these difficulties and the necessity to develop novel methods that can enable accurate monitoring and detection of AF [2]. In addition, robust detection and classification algorithms are essential for delivering appropriate therapy for implantable cardioverter defibrillators (ICD) to provide lifesaving timely action. In this work, the authors propose and demonstrate the application of a multiscale frequency (MSF) approach [4] for accurate detection and discrimination between AF and NSR ECG traces taken from publically available Physionet database. The MSF approach takes into account the contribution from various frequencies in ECG and thus yield valuable information regarding the chaotic nature of AF. Therefore, we demonstrate that MSF can capture the complexity of AF which is associated with higher MSF value compared with NSR thus enabling robust discrimination e AF manifests itself with numerous chaotic frequencies within the body surface ECG,. We validate the feasibility of this technique to discriminate NSR from AF.

Physiological cyclic loading of the intervertebral disc generally occurs between 1 and 5 Hz [1]. However, most mechanical assessments of disc AF tissue have relied on uniaxial tensile tests under quasi-static conditions [2]. Furthermore, the few studies that have addressed AF viscoelasticity have reported only static viscoelastic properties such as creep or stress-relaxation [3]. As such, there exists almost no data characterizing the dynamic viscoelastic properties of AF tissue in tension. Such data would be critical for several applications: to elucidate the mechanical progression of intervertebral disc degeneration, to develop and validate structural finite element models, and to provide native tissue benchmarks for regenerative approaches that aim to restore mechanical function to diseased or degenerate tissue. Therefore, the objectives of this study are to: (1) quantify human AF tissue frequency-dependent and strain-dependent viscoelastic properties of human AF tissue in circumferential tension, and (2) determine the effects of disc degeneration on these properties.

The annulus fibrosus (AF) is a highly organized structure made up of concentric lamellae of fibers embedded in a hydrated extrafibrillar matrix; the collagen fibers are oriented at alternating angles in each lamella. The AF undergoes multidirectional loading through combinations of compression, bending, torsion and shear of the motion segment. The composition and structure of the AF leads to mechanical stress-strain nonlinearity and anisotropy. Previous tissue-based studies of shear have tested the AF tissue under compressive simple shear and torsion, producing shear modulus on the order of 0.06–0.4 MPa [1, 2]. However, structural testing and mathematical models of the IVD have reported the shear modulus to be between 3–20 MPa [3–6]. We hypothesize that when the fibers of the AF are loaded the shear modulus will be on the same order as structural tests and mathematical models of the IVD. The objectives of this study are to measure the shear mechanical properties of the bovine outer AF and compare the regional variances between anterior and posterior AF.

Atrial Fibrillation (AF) is the cardiac arrhythmia most commonly encountered in clinical practice. Current statistics referring to the US population indicate a prevalence of AF up to 2.2 million people, projected to increase to 2.66 million by the end of 2010. AF has a high impact on society in terms of human costs, with an annual mortality rate of 11,438 patients. AF also increases the risk of ischemic stroke at least by a factor of 4 and it is responsible for at least 15% of all ischemic strokes, which represent the main cause of long-term disability and one of the main contributors to health care costs [1]. AF results from the synergic action of electrophysiological, biochemical and structural remodeling. Ventricular tachypacing (VTP) has been successfully used in animal models to reproduce relevant features observed in patients suffering from AF, such as ion-channel alterations, fibrosis development and atrial dilatation [2] [3].

Function of the annulus fibrosus (AF) of the intervertebral disc is predicated on a high degree of structural organization over multiple length scales. Recently, we have employed aligned electrospun scaffolds to engineer nanofibrous biologic laminates that replicate the form and function of the AF [1]. Further, we determined that interlamellar shearing — a direct consequence of the +/−30° angle-ply architecture — plays an important role in reinforcing the tensile response of these materials (Fig. 1). Although we have utilized fiber-reinforced continuum models to characterize the evolving mechanics of single-lamellar AF constructs with in vitro culture [2, 3], these models are not capable of capturing the interlamellar interactions observed in bi-lamellar constructs. Indeed, continuum models of the native AF typically do not account for the organization of fiber populations into discrete, alternating planes of alignment, and so these models, too, do not account for inter-lamellar shearing interactions [4–6]. Therefore, in the present work we propose a novel constitutive model for the reinforcing role of interlamellar shearing during uniaxial extension of angle-ply biologic laminates and employ this model to evaluate the functional evolution of bilayers for AF tissue engineering.

We propose and study, theoretically and empirically, a new random model for the abstract argumentation framework (AF). Our model overcomes some intrinsic difficulties of the only random model of directed graphs in the literature that is relevant to AFs, and makes it possible to study the typical-case complexity of AF instances in terms of threshold behaviours and phase transitions. We proved that the probability for a random AF instance to have a stable/preferred extension goes through a sudden change (from 1 to 0) at the threshold of the parameters of the new model D(n, p, q), satisfying the equation 4q/((1 + q)(1+q)) = p. We showed, empirically, that in this new model, there is a clear easy-hard-easy pattern of hardness (for a typical backtracking-style exact solvers) associated with the phase transition. Our empirical studies indicated that instances from the new model at phase transitions are much harder than those from an Erdos-Renyi-style model with equal edge density. In addition to being an analytically tractable model for understanding the interplay between problems structures and effectiveness of (branching) heuristics used in practical argumentation solvers, the model can also be used to generate, in a systematic way, non-trivial AF instances with controlled features to evaluate the performance of other AF solvers.

Collagen fibers in the annulus fibrosus (AF) play a significant role in maintaining the disc mechanical behavior. In healthy discs, AF fibers are concentrically arranged in a zigzag fashion. Tensile loads are transmitted to these fibers when the disc is loaded in compression. As a result, disc bulging occurs. Initiation and progression of mild cervical disc degeneration (DD) has been the subject of clinical concern [1]. The mild DD is associated with structural interruption in laminar organization of fibers in partial AF regions and incomplete length of fibers is a major form of structural interruption in AF fibers [2]. Moreover, a loss of lamellar fiber structure contributes to the progression of spinal deformity [3]. Past research have documented the biomechanical role of complete length of fibers in governing a healthy disc behavior [4]; however, there is a lack of clear understanding as to how incomplete length of fibers in different AF regions affect overall disc biomechanics and how they may play a role in initiating the propagation of DD. The focus of the current study is to investigate the disc biomechanical response due to incomplete length of fibers in outer, middle, and inner AF regions.

Atrial Fibrillation (AF) is a common disease that may occur in the heart, especially as we age. AF is due to non-normal myocardial ectopic foci that then causes an uncoordinated atrium contraction. This effectively reduces the atrial kick to the ventricles, which can account for up to 20% of ventricular filling. While not an immediately fatal disease, it can cause reduced quality of life for patients and also puts them at increased risk for stroke. AF as a disease, is expected to affect over 50 million people in the United States alone by 2050 [1].

Det danske sprog anses af mange lørnere som svært at lære, og især anses det som svært at blive flydende i. Dette bliver ofte tilskrevet den svære danske udtale. Artiklen her viser, at sprogskolernes håndtering af udtaleundervisning kan bidrage til voksne andetsprogstaleres udtaleproblemer. Her undersøges voksne lørneres og underviseres iagttagelser omkring det svære danske sprog, og undervisningen af det. I denne undersøgelse inddrages en analyse af undervisningsmateriale brugt ved voksenundervisning i dansk som andetsprog, og en diskussion af danskeksamener for andetsprogstalere. Vores resultater indikerer at udtale, og især vokaler, anses af både lørnere og undervisere som særligt svære at mestre. Samtidig undervises der ifølge vores informanter ikke nok i vokaludtale på de danske sprogskoler. Selvom de to informantgrupper generelt er relativt enige er især mængden af undervisning i udtale et kontrastpunkt mellem lørnere og undervisere. Vi diskuterer denne forskel, samt betydningen af meget lidt undervisningsmateriale der fokuserer på udtale efter DU-modul 1, og et manglende fokus på udtale i danskeksamenerne.
