Full textStreptomyces is a genus of Gram+ filamentous soil bacteria well known for their ability to produce antibiotics and other molecules useful as therapeutic or phytosanitary agents in medicine or agriculture. Under specific growth conditions some strains can store an excess of carbon into TriAcylGlycerols (TAGs), a direct Bio-diesel precursor. Streptomyces is thus an interesting canditate to generate bio-oils by fermentation. In this study, our goal is to evaluate, at the subcellular scale, the size/shape and localization of storage lipid inclusions in different Streptomyces strains (such as Streptomyces lividans (TK24), Streptomyces coelicolor (M145)…).In a previous study, the global TAGs content of those strains was easily and precisely quantified using infrared (IR) spectroscopy and thin layer chromatography revealing among other things that S. coelicolor has a very low TAGs content whereas S. lividans has an high TAG content. This was possible since TAG molecules show a specific response in the mid-infrared (IR) region, quite distinct from that of the other cellular constituants.Triacylglycerols posses several absorption bands in IR. In particular we can easily distinguish the band of the C=O stretching of the esters at 1741 cm-1 from the amide I band (protein signature) of the bacterium.For the study of the local repartition of TAG inside the cells, a combination of atomic force microscopy and infrared spectroscopy was employed in order to create sub-cellular chemical maps that allow label-free identification of TAG inclusions in Streptomyces cytoplasm using their specific absorption properties at 1741 cm-1. AFM-IR is a user-friendly benchtop technique that enables infrared spectroscopy with a spatial resolution well below conventional optical diffraction limits. It acquires IR absorption imaging with lateral resolution down to 100 nm.This technique coupled with classical FTIR measurements constitutes a powerful tool to study TAG metabolism in Streptomyces and can be easily used to control the rate of lipid accumulation during the fermentation process. Hence, the AFM-IR technique is likely to provide new insights into the constitution of the fatty inclusions and the role of TAGs in the morphological and metabolic differentiation processes that characterize Streptomyces developmental cell cycle

Full textHfq is a pleiotropic bacterial protein that determines several phenotypic characteristics. Its main function is to facilitate responses to stresses that bacteria may encounter during environmental changes, mainly by using post-transcriptional genetic control. The protein, by its capacity to interact with RNA, in particular small non-coding RNA, enables a rapid regulation of gene expression. In addition, the protein also interacts with DNA and compacts it. From a structural point of view, the protein adopts an Sm-like fold, characterized by a toroidal oligomer formed by a continuous 30-stranded β-sheet. Besides its conserved N-terminal Sm domain, Hfq also possesses a C-terminal region (CTR) that can vary in size and sequence between bacteria. My PhD work focused on the analysis of this CTR region in Escherichia coli bacteria. Indeed, this region has the capacity to form an amyloid structure. This structural dynamic is related to the formation of self-assembled structures in vivo, in the proximity of the inner membrane and in the nucleoid.Using various physicochemical techniques (molecular microscopy, spectroscopy and infrared microscopy, circular dichroism and small angle X-ray scattering), my work consisted in characterizing the assembly of this region of Hfq, as well as the factors influencing its assembly (in particular, the presence of nucleic acids). A part of my work consisted in setting up an innovative correlative–imaging method to analyze the chemical and morphological signature of a single amyloid fibre. Finally, my work focused on the analysis of the effect of compounds that inhibit the aggregation of the amyloid structure, which could constitute a new way to develop a novel class of antibiotics

Google Inc., han llamado la atención de la industria tecnológica, sobre sí el código declarativo y las SSO de las interfaces de aplicación (Application Programming Interface, en sus siglas en inglés APIs) empleadas para el desarrollo del sistema operativo Android, en su condición de elementos distintos al código fuente, también estaban protegidas por el copyright. Una decisión que apoyó esta postura de que sí estaban protegidas hizo que un Juzgado de Distrito conociera sobre el fair use (uso justo en español), o no de las interfaces. Este caso falló a favor de Google declarando que la implementación del código declarativo y sus secuencias, estructura y organización de las 37 APIs de Java, propiedad de Oracle efectivamente fue un uso justo, siendo clave dentro de la interoperabilidad de los programas informáticos y de la innovación.

Full textStrong electronic correlation and the resultant phenomena are object of interest in the modern solid state physics. Such correlation can be found in totally different systems from insulators and semiconductors to metals. This thesis presents HF-ESR studies of such phenomena in two low dimensional transition metal oxides and two intermetallic compounds. In ESR the electron spin is used as a local probe to measure the interaction between electrons and the magnetic properties nearby. With increasing electron-electron (EE) interaction the resonance becomes broader, so higher frequencies and higher magnetic fields as usual in commercial available ESR devices are needed to study strong EE interactions. With the used HF-ESR device with a frequency range $\nu=\vu{20-700}{GHz}$ special questions can be investigated where the excitation energies are in the order of $h\nu$ or the resonance effects in high magnetic fields can be explored. \textbf{CaCu$_2$O$_3$} have the same crystal structure as \chem{SrCu_2O_3}, a textbook example for a 2-leg spin-\textfrac{1}{2}-ladder with a nonmagnetic groundstate and a spin gap separating the excited state. Surprisingly \chem{CaCu_2O_3} shows an antiferromagnetic (afm) ground state with a relatively high transition temperature. To get a deeper insight in the unexpected afm ordering a combined magnetization and HF-ESR study was performed on a set of Zn-doped \chem{CaCu_2O_3} samples. Contrary to the Sr-compound in \chem{CaCu_2O_3} the \chem{Cu_2O_3}-ladder-layers are buckled due to a reduced rung angle. Furthermore it is a nonstoichiometric compound \chem{Ca_{1- x} Cu_{2+x}O_{3- \delta}}, with an excess of Cu in the order of $x\sim 0.16$ which is in the nonmagnetic \chem{Cu^{1+}}-state, sitting close to Ca-sites and a deficiency of oxygen $\delta\sim 0.07$. With this study one can show that (i) in the undoped compound the extra-spins, responsible for the magnetic Curie-Weiss-behavior, do not sit in the chains, they are sitting on low-symmetry interstitial sites. They recruit themselves from excess \chem{Cu^{1+}}, where the position becomes unstable close to a O-vacancy so they shift to a interstitial site and become \chem{Cu^{2+}}, (ii) the interstitial site of the extra-spins couple n.n. ladders inside a layer with a direct afm exchange path which lifts the frustration of the spin-ladders so that a afm order with such a high ordering temperature can happen and (iii) the regular interstitial site of the extra-spins explains the weak commensurate spin structure additionally found to the incommensurate spin structure of the ladder-spins in the afm ordered state The single layered manganate \textbf{LaSrMnO$_4$} is a two dimensional member of the transition metal oxides. In this compound a strong correlation between the orbital and magnetic degree of freedom can be found, so that the afm ordering below $T_N\sim\vu{125}{K}$ comes along with a ferro-orbital ordering of the \chem{Mn^{3+}} $3d$-Orbitals. With HF-ESR we have measured the temperature dependent mixing of the $3d$-orbitals and proved quantitatively the theory of ferro-orbital ordering. In the afm ordered state below $T_\text{stat}\sim\vu{40}{K}<T_N$ a strong field dependent decrease of the microwave transmission was observed. The frequency dependence of this phenomena could be explained by ferromagnetic polarons resulting from the interaction of additional charge carriers with the afm ordered spins. \textbf{GdNi$_2$B$_2$C} The intermetallic borocarbides $R\chem{Ni_2B_2C}$ ($R$ - rare earth metal) attract much attention due to the mutual interaction of superconductivity and afm ordering with complex phase diagrams. One reason for this complexity is the strong magnetic anisotropy coming from the splitting of the $J$-multiplets of the $R$'s $f$-orbitals in the crystal field. The nonsuperconducting \chem{GdNi_2B_2C} was widely explored because \chem{Gd^{3+}} with a half filled $4f$-shell should show no anisotropic behavior. The HF-ESR study on this system showed, that the assumed pure spin magnet have a uncommonly strong anisotropy which could be ascribed to a highly complex band structure. Involving this new information will help to adjust the model to the reality. YbRh$_2$Si$_2$ In this heavy fermion system where the magnetic Yb ($4f$) built up a regular Kondo-lattice here is a competition between electron-electron- and the Ruderman-Kittel-Kasuya-Yosida-(RKKY) interaction. Thats why in this compound a afm ordered state, a (paramagnetic) heavy fermion (LFL) and a non-Fermi-liquid behavior can be established by changing the magnetic field $B$ and/or the temperature $T$. Below the Kondo-temperature $T^*$ a strong hybridization between the conduction electrons and the $4f$-electrons leads to a strong broadening of the otherwise atomic sharp $4f$-states. Thats why the observation of a small electron spin resonance below $T^*$ was very surprising. Because the yet published ESR-measurements are fully in the NFL-state, we performed HF-ESR measurements to study a $B-T$ area where a NFL-LFL crossover appears to get a deeper inside of the physics behind this resonance. The behavior of the measured $T$- and $B$-dependences indicate that this resonance phenomena in \chem{YbRh_2Si_2} is a resonance of heavy fermions

In the present study, an anaerobic fluidized bioreactor combined with an anaerobic fluidized membrane bioreactor (AFBR-AFMBR) was used to for treat municipal wastewater at ambient temperature. The objectives of this study were to evaluate the start-up time and the performance using zeolite as carriers and the performance of the AFBR-AFMBR. During the start-up period, the reactor system was operated in semi-continuous mode and at a hydraulic retention time (HRT) of 1 d and an organic loading rate (OLR) of 1 g/m3/d with influent chemical oxygen demand (COD) of 1000 mg COD/L (made by synthetic wastewater). The reactor system reached to steady state with a pH level of 7, COD removal efficiency higher than 90% and 1 L/d of the steady biogas production. The microorganisms were successfully attached onto the carrier. Thereafter, the reactor system was changed to continuous mode operation at 3.5 h of total HRT for treating municipal wastewater (73 to 220 mg COD/L) at ambient temperature ranged from 20 to 35oC. The reactor performance reached to 70 to 90% of COD removal, 96% of total suspended solids (TSS) removal and 0.13 L CH4/g CODremoved of specific methane production. The membrane module in AFMBR can be continuously used for 333 d at a flux of 10 LMH without chemical cleaning. Besides, the reactor system only needed 0.015 kWh/m3 of pumping energy requirement and can generate 0.0024 kWh/m3 of methane energy. The above results revealed that the AFBR-AFMBR is a high efficiency energy-saving system for treating municipal wastewater.

CD28, a T-cell transmembrane receptor, is one of key factors to activate the immune responses via the secretion of interleukin-2 from T-cell and can be considered as a possible pathway related to autoimmune diseases. In the study, a pharmaceutical screening method, AFTIR (After Flowing Through Immobilized Receptor) was applied to search blockers from herbal medicines to inhibit the immune response from CD28. The results show that Echinacea and Schisandra possess effective components to block CD28 from binding to its ligand CD80 and the bioassay indicates that the fractions of these herbs have significant immuno-suppressive responses in T-cell. Furthermore, an active compound, cynarin, from Echinacea has been identified and can downregulate CD28-dependent IL-2 expression in T-cells. With the observations above, cynarin of Echinacea and effective fraction of Schisandra can be used in the future as potential agents for curing auto- immune diseases.

The number of diabetes patients is growing fast all over the world. Previous reports showed that lowering blood glucose levels may delay the occurrence of diabetes. Western medicines in clinical treatment of diabetes use oral hypoglycemic agents include α-glucosidase inhibitors which can effectively reduce postprandial blood glucose levels. In this study, AFTIR technology via the immobilization of α-glucosidase on chip was used to find several effective herbal extracts and has been verified as an effective method via finding the epicatechin gallate from green tea. Furthermore, the results show that Taxus blocks epigallocatechin gallate and α-glucosidase with blocking ratio of 35.9% and the activity of inhibiting α-glucosidase is significant. With the observations above, Taxus can therefore be a potential agent used for curing the diabetes patients in the future.
