Abstract:

The artificial grammar learning (AGL) paradigm has been intensively researched since the 60-s. In general, these investigations attempt to study the implicit acquisition of structural regularities. Among other things, it has been suggested that the AGL paradigm can serve as a model for the process of acquiring a natural language. Thus it can serve as a well-controlled laboratory task that might be used to understand certain aspects of the process of language acquisition. For example the AGL paradigm has been used in an attempt to isolate the acquisition of syntactic aspects of language. Several experimental studies show that the participants acquire knowledge of the underlying rule system since they are able to differentiate grammatical strings from non-grammatical ones. It has been argued that the traditionally conducted AGL paradigm with grammaticality instructions might make the task explicit, at least during the test phase. In order to imitate the language learning process as close as possible, to rule out the possibility of an explicit component during the testing phase (i.e., keeping the retrieval process implicit) and to rule out explicit rule conformity or rule following, we modified the classical AGL paradigm. In a behavioural study we combined the AGL paradigm with an altered mere exposure paradigm in an attempt to better model aspects of language acquisition. We were able to show that subjects, classifying under mere exposure instructions, categorize grammatical and non-grammatical strings just as well as those solving the classification task with the grammaticality instructions. This indicates that the mere exposure version might serve as a more appropriate model for language acquisition.

Resumo: As doenças de depósito de glicogênio (GSD) incluem aproximadamente 12 tipos, onde existe falta de uma ou de diversas enzimas que metabolizam o glicogênio normal e são também chamadas de glicogenoses. Os acúmulos intracelulares do glicogênio afetam principalmente células hepáticas, renais, músculo cardíaco e esquelético. As diferentes formas de GSD foram classificadas de acordo com a ordem cronológica em que os defeitos enzimáticos foram identificados. A GSDI compreende um grupo de doenças raras, com padrão de herança autossômico recessivo apresentando grande heterogeneidade clínica e bioquímica. Existem dois principais subgrupos, GSDIa e Ib, confirmados em nível molecular. O subtipo Ia se refere à deficiência da enzima glicose-6-fosfatase (G6Pase), enzima responsável pela manutenção da glicose sanguínea; a GSDIb relacionada à deficiência da glicose-6-fosfato translocase (G6PTl) responsável pelo transporte da glicose-6-fosfato para o lúmen do retículo endoplasmático, onde a unidade catalítica da G6Pase está situada. Outros subgrupos menos comuns foram também identificados, tais como o subgrupo Ic que envolve o transporte de fosfato e pirofosfato e, finalmente, Id que envolve a enzima responsável pelo retomo da glicose do lúmen do retículo para o citosol, existindo, no entanto, controvérsia com relação à classificação desses últimos subtipos. A GSDID é causada pela deficiência da enzima bifuncional AGL, uma proteína monomérica com dois sítios catalíticos diferentes: oligo-l, 4-1,4 glucontransferase e amilo-1,6-glucosidase, e para o funcionamento normal, ambas as atividades são requeridas. A enzima AGL combinada à ação da fosforilase é responsável pela completa degradação do glicogênio. Devido à heterogeneidade clínica dos tipos I e m, durante a infância é difícil se diferenciar clinicamente as duas formas, ambas apresentando sintomas similares de hepatomegalia, hipoglicemia, hiperlipidemia,retardo de crescimento entre outros. Neste trabalho foram estudados 14 indivíduos com sinais clínicos sugestivos de GSDI onde não foram encontradas alterações no gene da G6Pase, responsável pelo subtipo Ia, esses pacientes são, portanto classificados como tipo I non-Q. O gene G6PTl foi seqüenciado completamente em todos os pacientes estudados e dois deles foram diagnosticados em nível molecular como sendo na verdade GSDIb. Foram estudados 6 exons do gene AGL, onde estão localizadas a maioria das mutações descritas até o momento no gene, e somente foram detectados polimorfismos em todos os indivíduos estudados. O protocolo molecular desenvolvido no presente trabalho mostrou-se adequado para o rastreamento de mutações, apesar de terem sido diferenciados molecularmente somente dois indivíduos com subtipo Ib
Abstract: There are 12 types of glycogen storage disease (GSD). There disease are characterized by the absence of one or many of enzymes called glycogenoses, which are responsible for normal glycogen metabolismo The intracelular increase of glycogen mainly affects hepatic, kidney, cardiac and squeletic muscle cells. GSDs have been classified according to the cronological order in which enzymatic defects were identified. GSDI includes a group of rare disease of autosomal recessive disorders and clinical and biochemical heterogeneity. There are two main subgroups GSDIa and Ib. A diagnosis can only be confirmed by molecular studies. Subtype Ia is caused by a deficiency glucose-6-phosphatase (G6Pase), is responsible for the mantainance of blood glucose homeostasis. Subtype Ib results trom a deficiency of the glucose-6-phosphate translocase enzyme. This enzyme transports glucose-6-phosphato into the lumen ofthe endoplasmic reticulum, where the G6Pase catalitic site situated. Such as Ic and Id have also been identified. Subtype Ic is related to phosphate and pyrophosphate transporter and Id is related to the enzyme responsable for glucose transport trom lumen to cytosol. GSDm is caused deficiency of bifunctional enzyme (AGL) is a monomeric protein, with both amylo-l,6-glucosidase and 0Iigol,4-1,4 glycosyl transferase activity at two separate catalytic sites. The AGL enzyme, together with phosphorylase, is responsible for degradation of glycogen. During infancy, type m disease may be almost indistinguishable from type I disease, as hepatomegaly, hypoglycemia, hyperlipidemia, and growth retardation are similar predominant features. In the present study, we report molecular and clinical finding in 14 patients with suggestive clinical symptoms of GSDI which didn't be found mutations in the G6Pase gene, responsable by subtype Ia, these patients are thus, classifieds as type I non-a. DNA sequencing of the exons ofthe G6PTl in 14 patients mutations in the G6PTl gene were found in two patients. Six exons of AGL gene were studied, which are found the majority of describe mutation, and just be found polymorphism in all studieds individuals. The molecular protocol that was studied, showed suitable to mutation screemng, whowever to be were find only two individuals with subtype Ib

L'arrivée sur le marché des premiers SGBD orientés objet conduit à poser le problème de conception de ces bases de données orientées objet. En effet, plus systèmes d'information sont complexes, plus les outils pour les traiter deviennent sophistiqués, plus le recours à des méthodes et outils de conception deviennent nécessaires. Il convient donc d'explorer de nouvelles voies afin que ces SGBD puissent être pleinement utilisés. Nous proposons quelques concepts pour une approche méthodologique orientée objet. Le but cherché consiste à offrir un ensemble de représentations graphiques permettant de traduire les notions de d'attribut-tuple , d'attributs atomiques, de liens d'agrégation et d'héritage. La finalité est, bien-sûr, la génération de code permettant d'implanter le schéma d'une base de données d'un SGBD orienté objet (cible 02). La méthode se caractérise par une phase d'intégration impliquant des tâches algorithmiques, des décisions humaines (choix de certaines relations , etc. . . ). Parmi ces décisions, certaines ont codées ·et enregistrées comme des règles expérimentales applicables par défaut, d'autres interviennent lors du dialogue entre le système et l'utilisateur. Des processus d'intégration sont proposés afin de fournir une vision un recours aides outils de conception (AGL ). The recent apparition of the Object Oriented DBMSs requires an enhancement of classical information system design. The complexity of the Information System is accompanied by the development of more sophisticated aide tools and by having recourse to design methodologies. Using the traditional design methods are insufficient to abject approach. For example, the methods formed upon the Entity - Association model are not convenient for the design of applications developed on abject oriented DBMSs. New means must be explored to benefit as much as possible from such DBMSs. We propose some concepts for an Object Oriented methodology. The proposed method, folloing an object oriented approach provides a static and dynamic representation of the applications. Our approach considers different aspects of the same object, depending on the viewpoint of each users. We proceed then to integrate all these views in a global conceptual scheme. The views integration, tockled in some classical conceptual methods arises new problems features and highlights the complexity of phenomena. E can mention, for example. Inheritance conflicts, data semantic, synonymy and polysemy. The target DBMS which guides us is 02. We have developed a tool. (CASE)

Os recentes avanços tecnológicos levaram ao desenvolvimento de sistemas de navegação mais precisos e de veículos aéreo não-tripulados (UAV) cada vez mais autónomos que, por sua vez, diminuíram significativamente o número de tarefas a realizar pelos operadores no solo. Com o propósito de reduzir o erro altimétrico do sistema DGPS, utilizado a bordo das plataformas Alfa-Extended da Força Aérea para a realização de aterragens automáticas, optou-se por utilizar um Laser-Altímetro com um erro teoricamente mais baixo. Desta forma, esta dissertação tem por objetivo a análise do desempenho do Sensor AGL, em termos de precisão e exatidão, enquanto sistema auxiliar para aproximação e aterragem automática de UAV. Para a concretização deste objetivo começou-se por desenvolver o driver do Sensor AGL, seguindo-se de todo o software e hardware necessário para os testes e dos scripts para pós-processamento dos dados recolhidos. Posteriormente foram realizados os testes no solo e em voo, permitindo a análise do desempenho deste sensor na realização de medições sobre várias superfícies, a várias distâncias, velocidades e ângulos de incidência. Assim, graças à metodologia seguida nesta dissertação, verificou-se uma precisão e exatidão altimétrica, por parte do Sensor AGL, claramente superior ao do sistema DGPS, registando-se um erro máximo de 0,3 m. Embora a taxa de amostragem obtida se tenha verificado inferior à anunciada pelo fabricante e tenham sido registados alguns erros em função do ângulo de incidência do feixe, estes fatores não foram considerados condicionantes para a realização de aterragens automáticas. Desta forma, concluiu-se que é vantajosa a utilização do Sensor AGL para a redução do erro altimétrico do sistema DGPS, recomendando-se que, no entanto, as medições de baixa resolução não sejam utilizadas para a aterragem e aconselhando cautela na operação sobre superfícies muito molhadas.

Le paradigme « Model Driven Engineering (MDE) » (Ingénierie de modèles) vise à simplifier le développement et de la maintenance des systèmes informatiques. Il se focalise sur l’utilisation de modèles pour spécifier les systèmes et sur les manipulations de modèles avec des outils. Cette thèse a pour but de définir et de réaliser un environnement réparti et ouvert pour réaliser MDE. Cet environnement, nommé ModelBus, vise 1) à supporter les manipulations collaboratives des modèles par multi-développeurs, et 2) à fournir l’interopérabilité entre outils hétérogènes impliqués dans les projets MDE. Pour attendre le premier objectif, ModelBus propose les mécanismes « diff/merge » des modèles, et, pour le deuxième, ModelBus offre les mécanismes d’échange de modèles et d’invocations de services entre outils. Les problèmes majeurs traités par ModelBus concernent le passage à l’échelle face à la taille des spécifications des systèmes complexes. Etant donné qu’un ensemble de modèles décrivant un système est un grand graphe d’objets (à l’ordre de GOs), dont les liens représentent les relations entre les modèles (ex. Traçabilité, relations entre les vues du système), nous soulevons les problèmes suivants: 1) Insuffisance de mémoire pour manipuler ce graphe entier, 2) Incohérences des liens du graphe (liens casés) causées par les manipulations concurrentes des modèles. Pour résoudre ces problèmes, ModelBus partitionne une spécification de système en modèles, et gère la cohérence des liens entre ces modèles

O tratamento das leucemias agudas continua sendo um grande desafio clínico devido, principalmente à heterogeneidade e alta toxicidade da terapia padrão utilizada. Dessa forma, novos alvos terapêuticos são urgentemente necessários e os anticorpos monoclonais tem surgido como uma das opções terapêuticas mais promissoras. A endoglina, também conhecida como CD105, é um receptor da superfamília do TGF-β, expresso nas células-tronco hematopoiéticas (HSC) de todos os sítios hematopoiéticos, incluindo a medula óssea, onde é descrita como um marcador para HSC de longo prazo. Apesar da expressão de CD105 ter sido relacionada a diversos tipos de tumores sólidos, principalmente devido ao papel desse receptor na angiogênese, relativamente pouco é conhecido em relação a expressão de CD105 e a sua função em neoplasias hematopoiéticas. Este estudo revelou alta expressão de endoglin na maioria dos blastos de pacientes com leucemia mieloide aguda (LMA) e leucemia linfoblástica aguda (LLA). Utilizando um modelo de xenotransplante, verificamos que as blastos CD105+ possuem uma actividade leucemogênica superior em comparação com a população de CD105-. Adicionalmente, investigamos se o bloqueio da endoglina, usando TRC105, poderia resultar em uma opção terapêutica para tratamento das leucemias agudas e descobrimos que na LMA, o TRC105 impediu o engraftment de blastos primários e inibiu a progressão da leucemia após o estabelecimento da doença, mas na LLA, o TRC105 sozinho foi ineficaz devido à uma maior secreção da forma soluvél da endoglina (sENG). No entanto, tanto na LLA quanto na LMA, TRC105 potencializou o efeito terapeutico da quimioterapia padrão e inibiu a progressão da doença, indicando que TRC105 pode representar uma nova opção terapêutica para LLA e LMA.
Successful treatment of acute leukemia remains a clinical challenge due to the toxicity and the relatively poor responses to the current standard therapy. Thus, treatments that address novel therapeutic targets are urgently needed and, for this purpose, monoclonal antibodies have been one of the most promising strategy once they are able to deliver their therapeutic effects with minimal toxicity. Endoglin, also known as CD105, is a receptor of the transforming growth factor-beta (TGF-β) superfamily that has been found expressed in hematopoietic stem cells (HSCs) from all hematopoietic sites, including the bone marrow, in which it is described as a marker for long-term HSC. CD105 is also found to be expressed in several cancers. However, because CD105 expression has been studied mostly in the context of solid tumors and angiogenesis, relatively little is known about CD105 expression and role in hematopoietic malignancies. We identified endoglin expression on the majority of blasts from patients with acute myeloid leukemia (AML) and acute B-lymphoblastic leukemia (B-ALL). Using a xenograft model, we find that CD105+ blasts are endowed with superior leukemogenic activity compared to the CD105- population. We test the effect of targeting this receptor using the monoclonal antibody TRC105, and find that in AML, TRC105 prevented the engraftment of primary AML blasts and inhibited leukemia progression following disease establishment, but in B-ALL, TRC105 alone was ineffective due to the shedding of soluble CD105. However, in both B-ALL and AML, TRC105 synergized with reduced intensity myeloablation to inhibit leukemogenesis, indicating that TRC105 may represent a novel therapeutic option for B-ALL and AML.

Abstract:
Defective chromatin remodeling proteins are associated with both germline and acquired human disease. PHF6 is encoded by an X-linked gene that is predominantly expressed in the brain and thymus. Structurally, PHF6 contains nuclear and nucleolar localization sequences as well as two ZaP domains, which bind dsDNA. Germline mutations in PHF6 are the cause of BFLS, an XLID, while somatic PHF6 mutations have been identified in T-ALL, AML, and CML. Indeed, screening of a pediatric cohort of nine T-ALL patients revealed a novel H329Q mutation. In a further clinical analysis, T-ALL onset occurred in a 9-year old male BFLS patient with an R342X mutation, suggesting that BFLS might be a cancer predisposition syndrome. To better understand its protein function, recombinant PHF6 was co-immunoprecipitated for a mass spectrometry based proteomic screen. Notably, PHF6 co-purified with multiple constituents of the NuRD complex, an important transcriptional regulator during embryogenesis and lineage commitment with particularly well characterized responsibilities during lymphogenesis. PHF6-NuRD localization was restricted to the nucleoplasm, however PHF6 also co-purified with several ribosomal and splicing proteins. When examined further, PHF6 was found to be recruited to the nucleolus by an RNA-mediated interaction and co-localized within the subnucleolar FC and DFC compartments. ChIP-qPCR revealed that PHF6 binds to transcribed regions of rDNA, resulting in the repression of rRNA. These data thus present a model of PHF6 acting as a tumour suppressor by mediating both nucleoplasmic and nucleolar transcriptional events.

Abstract:
This thesis examines inter-organisational or collaborative change. The research commenced with an investigation of organisational change in policing and then broadened to encompass other organisations in the public and private sectors, thereby enabling comparison and contrast. The intention was to explore first principles of best practice for organisations embarking upon collaborative change. The researcher was motivated to learn and develop his thinking as a consequence of professional exposure to this area of strategic management. The exponential growth in the number and complexity of collaborative schemes, coupled with a reported pan sector failure rate of 60% - 80%, underlines the topicality and relevance of this research. In the public sector, there is evidence of a new collaborative paradigm emerging, driven by ideology and austerity. Available evidence indicates that the police have been slow to embrace these changes and achieve the benefits desired. The methodological approach commenced with systematic and targeted reviews of existing literature. Gaps and contradictions were explored and used to inform the primary research, which consisted of forty-three semi-structured interviews and two electronic surveys, producing two hundred and fifty-six responses. The findings suggest that shortcomings in delivering interorganisational change were indicative of wider management failings encompassing other categories of major change. Poor leadership, inadequate resourcing, weak planning, unrealistic timescales and an over emphasis on structure and process, while ignoring the people issues, all contributed to a suboptimal outcome. Opinions were polarised about the performance of the police, where a strong organisational culture, dominant leaderships styles and poor business acumen appear to be conspiring to inhibit ambition and delivery. Ten generic key research findings are discussed, with another dedicated to policing. Collectively, they provide an evidence based foundation for developing best practice for organisations embarking on major inter-organisational change.

Demolt angefertigt in der Universitätsklinik und Poliklinik für Kinder und Jugendliche in Leipzig, Neonatologische Intensivstation betreut von Frau Prof. Dr. med. Eva Robel-Tillig Juli 2012 Chronische intrauterine Hypoxie bedingt durch uterine, feto-maternale und fetale Perfusionsstörung führt zur fetalen Wachstumsrestriktion und Erhöhung der fetalen und neonatalen Morbidität und Mortalität. Die pränatale Kreislaufzentralisation stellt einen pathophysiologischen Kompensationsmechanismus dar, der durch Umverteilung des Blutflusses eine Versorgung lebenswichtiger Organe des Feten sichert (Rizzo et al. 2008), (Robel-Tillig 2003), (Robel 1994), (Saling 1966). In den letzten Jahren haben sich Studien mit der postnatalen Adaptation der wachstumsretardierten Neonaten beschäftigt und wesentliche Risiken im Verlauf der ersten Lebenstage definiert. Wenige validierte Aussagen existieren jedoch zur Kreislaufsituation der betroffenen Kinder und der kardialen Leistungsfähigkeit auch über die erste Lebenswoche hinaus. Die vorliegende Studie vergleicht unter dieser Fragestellung eine Gruppe von 43 Frühgeborenen mit intrauteriner Wachstumsrestriktion auf der Grundlage einer Perfusionsstörung und 33 Frühgeborene mit appropriatem Wachstum und ungestörter pränataler Perfusion während der ersten 42 Lebenstage hinsichtlich der unmittelbaren postnatalen pulmonalen und kardialen Adaptationsparameter und des weiteren klinischen Verlaufs. Besonderer Schwerpunkt wird dabei auf die dopplersonographisch erfasste kardiale Funktion der Kinder gelegt. Als wesentlichstes Ergebnis der dopplersonographischen Messungen lässt sich bei den Frühgeborenen mit pränataler Perfusionsstörung ein signifikant erhöhtes Herzminutenvolumen rechts- und linksventrikulär im Vergleich zur Gruppe der Frühgeborenen mit ungestörter Perfusion darstellen (Robel-Tillig 2003), (Leipälä et al. 2003), Martinussen 1997}, (Guajardo, Mandelbaum & Linderkamp 1994), (Lindner et al. 1990). Die unmittelbar postnatale Adaptation zeigt hinsichtlich des arteriellen Nabelschnur-pH-Wertes eine schlechtere Anpassung der Frühgeborenen mit pränataler Perfusionsstörung auf. Im weiteren Verlauf der ersten Tage benötigen diese Frühgeborenen seltener eine maschinelle Beatmung oder CPAP-Atemhilfe als die Frühgeborenen ohne Wachstumsrestriktion. Bis zum 42. Lebenstag kehrt sich dieser Befund jedoch um. Die Frühgeborenen mit Wachstumsrestriktion bedürfen nun signifikant länger und häufiger einer Atemhilfe und zusätzlicher Sauerstoffsupplementierung. Als Komplikation trat bei den wachstumsretardierten Frühgeborenen eine höhere Rate an bronchopulmonaler Dysplasie auf. Ein weiterer Unterschied wird hinsichtlich der Transfusionshäufigkeit dargestellt. Frühgeborene mit Wachstumsrestriktion erhielten häufiger eine Erythrozytentransfusion und länger eine Transfusion von zusätzlichem Volumen. Zusammenfassend weisen die ermittelten Befunde auf ein Persistieren der intrauterin bestehenden Kompensation der beeinträchtigten Kreislaufsituation hin. Eine genaue Kenntnis der speziellen Probleme dieser Patientengruppe ist zur Vermeidung postnatal anhaltender hämodynamischer Störungen erforderlich.

Il fattore antisecretivo (AF) è una proteina secreta nel plasma e nei tessuti dei mammiferi, che ha dimostrato di essere un potente inibitore dell’ipersecrezione intestinale e dell'infiammazione. Dopo il bando degli AGP, diversi approcci nutrizionali finalizzati all’ottimizzazione della fase di transizione dello svezzamento e alla riduzione delle malattie intestinali sono stati proposti, ma con scarsi risultati rispetto agli antibiotici. Le diete in grado di indurre la secrezione endogena di AF potrebbero essere una valida alternativa agli AGP. Si ritiene che AF contenuto nel colostro e nel latte delle scrofe possa essere un fattore di protezione contro la diarrea nei suinetti. E’ stato dimostrato che è possibile incrementare il livello di AF nel plasma attraverso la dieta, sia nell’uomo che negli animali, con l’ingestione di cereali sottoposti ad un particolare processo idro-termico (HPC). In questa tesi è stato studiato l’effetto dell’aggiunta di HPC alla dieta di suinetti svezzati, sulle performance e sullo stato infiammatorio dell’epitelio intestinale. La supplementazione della dieta con HPC ha migliorato ADG e FCR durante tutto il periodo sperimentale. I valori I-FABP, considerato un parametro del danneggiamento della mucosa intestinale, nel plasma dei suinetti sono risultati bassi in tutti i gruppi e non influenzati dalla dieta. L'attività antisecretiva di AF-16, il dominio attivo della proteina, è stata valutata mediate su colture cellulari IPEC-J2 trattate con tossina colerica (CT) in Ussing chamber. AF-16 non ha inibito l’incremento di Isc indotto da CT. E’ stato osservato un ridotto incremento di Isc durante la somministrazione contemporanea di AF-16 e CT. L'attività anti-infiammatoria di AF-16 è stata studiata in colture di macrofagi RAW 264,7, trattati con LPS, e su macrofagi alveolari di suino stimolati con PMA. AF-16 ha ridotto la produzione di NO nei macrofagi stimolati con LPS in funzione del dosaggio, con maggiori effetti a dosaggi più elevati. L’effetto antiinfiammatorio di AF-16 non è stata confermata dai ROS test, neanche usando dosi elevate di peptide. I risultati confermano l’efficacia dei cereali HPC come promotori della crescita nei suinetti. Ulteriori studi in vitro sono necessari per capire il meccanismo d’azione di AF.
The antisecretory factor (AF) is a protein secreted in plasma and other tissue fluids in mammalians which was shown to be a potent inhibitor of intestinal fluid secretion and inflammation. After the AGP ban, several nutritional approaches aimed to the optimization of the weaning transition and reduction of gut diseases have been proposed, but the success rates are really low compared to antimicrobials. AF-inducing diets appear to be suitable alternatives to AGP. Indeed AF content in sows’ colostrum and milk appears to be a protection factor against diarrhoea in suckling piglets. Increased AF level in plasma by dietary means, such as feeding hydro-thermally processed cereals (HPC), has been demonstrated in human and animals. We tested the effect of two different HPC level of inclusion in a wheat-barley based diet on weaned piglets growth performance, reared in an experimental farm. The results confirmed the efficacy of HPC as growth promoters in piglet nutrition: HPC supplementation improved ADG and FCR. In vitro test were made to study the antisecretory and anti-inflammatory properties of the AF protein and its mechanism of action by using AF-16, the active region of the protein. The Ussing chamber experiments performed on polarized IPEC-J2 cells confirmed the neuronal involvement in the antisecretory activity of AF. In fact, AF-16 did not inhibit CT-induced Isc. A slower rate of Isc increase was observed during the simultaneous administration with AF-16 and CT. High dosages of AF-16 were found to reduce the LPS-stimulated NO production in RAW264.7 cell, thus supporting the hypothesis of an anti-inflammatory action. On the contrary, no significant results were obtained on PMA-stimulated ROS generation in pig alveolar macrophages.
