Full textThe artificial grammar learning (AGL) paradigm has been intensively researched since the 60-s. In general, these investigations attempt to study the implicit acquisition of structural regularities. Among other things, it has been suggested that the AGL paradigm can serve as a model for the process of acquiring a natural language. Thus it can serve as a well-controlled laboratory task that might be used to understand certain aspects of the process of language acquisition. For example the AGL paradigm has been used in an attempt to isolate the acquisition of syntactic aspects of language. Several experimental studies show that the participants acquire knowledge of the underlying rule system since they are able to differentiate grammatical strings from non-grammatical ones. It has been argued that the traditionally conducted AGL paradigm with grammaticality instructions might make the task explicit, at least during the test phase. In order to imitate the language learning process as close as possible, to rule out the possibility of an explicit component during the testing phase (i.e., keeping the retrieval process implicit) and to rule out explicit rule conformity or rule following, we modified the classical AGL paradigm. In a behavioural study we combined the AGL paradigm with an altered mere exposure paradigm in an attempt to better model aspects of language acquisition. We were able to show that subjects, classifying under mere exposure instructions, categorize grammatical and non-grammatical strings just as well as those solving the classification task with the grammaticality instructions. This indicates that the mere exposure version might serve as a more appropriate model for language acquisition.

Resumo: As doenças de depósito de glicogênio (GSD) incluem aproximadamente 12 tipos, onde existe falta de uma ou de diversas enzimas que metabolizam o glicogênio normal e são também chamadas de glicogenoses. Os acúmulos intracelulares do glicogênio afetam principalmente células hepáticas, renais, músculo cardíaco e esquelético. As diferentes formas de GSD foram classificadas de acordo com a ordem cronológica em que os defeitos enzimáticos foram identificados. A GSDI compreende um grupo de doenças raras, com padrão de herança autossômico recessivo apresentando grande heterogeneidade clínica e bioquímica. Existem dois principais subgrupos, GSDIa e Ib, confirmados em nível molecular. O subtipo Ia se refere à deficiência da enzima glicose-6-fosfatase (G6Pase), enzima responsável pela manutenção da glicose sanguínea; a GSDIb relacionada à deficiência da glicose-6-fosfato translocase (G6PTl) responsável pelo transporte da glicose-6-fosfato para o lúmen do retículo endoplasmático, onde a unidade catalítica da G6Pase está situada. Outros subgrupos menos comuns foram também identificados, tais como o subgrupo Ic que envolve o transporte de fosfato e pirofosfato e, finalmente, Id que envolve a enzima responsável pelo retomo da glicose do lúmen do retículo para o citosol, existindo, no entanto, controvérsia com relação à classificação desses últimos subtipos. A GSDID é causada pela deficiência da enzima bifuncional AGL, uma proteína monomérica com dois sítios catalíticos diferentes: oligo-l, 4-1,4 glucontransferase e amilo-1,6-glucosidase, e para o funcionamento normal, ambas as atividades são requeridas. A enzima AGL combinada à ação da fosforilase é responsável pela completa degradação do glicogênio. Devido à heterogeneidade clínica dos tipos I e m, durante a infância é difícil se diferenciar clinicamente as duas formas, ambas apresentando sintomas similares de hepatomegalia, hipoglicemia, hiperlipidemia,retardo de crescimento entre outros. Neste trabalho foram estudados 14 indivíduos com sinais clínicos sugestivos de GSDI onde não foram encontradas alterações no gene da G6Pase, responsável pelo subtipo Ia, esses pacientes são, portanto classificados como tipo I non-Q. O gene G6PTl foi seqüenciado completamente em todos os pacientes estudados e dois deles foram diagnosticados em nível molecular como sendo na verdade GSDIb. Foram estudados 6 exons do gene AGL, onde estão localizadas a maioria das mutações descritas até o momento no gene, e somente foram detectados polimorfismos em todos os indivíduos estudados. O protocolo molecular desenvolvido no presente trabalho mostrou-se adequado para o rastreamento de mutações, apesar de terem sido diferenciados molecularmente somente dois indivíduos com subtipo Ib
Abstract: There are 12 types of glycogen storage disease (GSD). There disease are characterized by the absence of one or many of enzymes called glycogenoses, which are responsible for normal glycogen metabolismo The intracelular increase of glycogen mainly affects hepatic, kidney, cardiac and squeletic muscle cells. GSDs have been classified according to the cronological order in which enzymatic defects were identified. GSDI includes a group of rare disease of autosomal recessive disorders and clinical and biochemical heterogeneity. There are two main subgroups GSDIa and Ib. A diagnosis can only be confirmed by molecular studies. Subtype Ia is caused by a deficiency glucose-6-phosphatase (G6Pase), is responsible for the mantainance of blood glucose homeostasis. Subtype Ib results trom a deficiency of the glucose-6-phosphate translocase enzyme. This enzyme transports glucose-6-phosphato into the lumen ofthe endoplasmic reticulum, where the G6Pase catalitic site situated. Such as Ic and Id have also been identified. Subtype Ic is related to phosphate and pyrophosphate transporter and Id is related to the enzyme responsable for glucose transport trom lumen to cytosol. GSDm is caused deficiency of bifunctional enzyme (AGL) is a monomeric protein, with both amylo-l,6-glucosidase and 0Iigol,4-1,4 glycosyl transferase activity at two separate catalytic sites. The AGL enzyme, together with phosphorylase, is responsible for degradation of glycogen. During infancy, type m disease may be almost indistinguishable from type I disease, as hepatomegaly, hypoglycemia, hyperlipidemia, and growth retardation are similar predominant features. In the present study, we report molecular and clinical finding in 14 patients with suggestive clinical symptoms of GSDI which didn't be found mutations in the G6Pase gene, responsable by subtype Ia, these patients are thus, classifieds as type I non-a. DNA sequencing of the exons ofthe G6PTl in 14 patients mutations in the G6PTl gene were found in two patients. Six exons of AGL gene were studied, which are found the majority of describe mutation, and just be found polymorphism in all studieds individuals. The molecular protocol that was studied, showed suitable to mutation screemng, whowever to be were find only two individuals with subtype Ib

Full textThe recent apparition of the Object Oriented DBMSs requires an enhancement of classical information system design. The complexity of the Information System is accompanied by the development of more sophisticated aide tools and by having recourse to design methodologies. Using the traditional design methods are insufficient to abject approach. For example, the methods formed upon the Entity - Association model are not convenient for the design of applications developed on abject oriented DBMSs. New means must be explored to benefit as much as possible from such DBMSs. We propose some concepts for an Object Oriented methodology. The proposed method, folloing an object oriented approach provides a static and dynamic representation of the applications. Our approach considers different aspects of the same object, depending on the viewpoint of each users. We proceed then to integrate all these views in a global conceptual scheme. The views integration, tockled in some classical conceptual methods arises new problems features and highlights the complexity of phenomena. E can mention, for example. Inheritance conflicts, data semantic, synonymy and polysemy. The target DBMS which guides us is 02. We have developed a tool. (CASE)

O tratamento das leucemias agudas continua sendo um grande desafio clínico devido, principalmente à heterogeneidade e alta toxicidade da terapia padrão utilizada. Dessa forma, novos alvos terapêuticos são urgentemente necessários e os anticorpos monoclonais tem surgido como uma das opções terapêuticas mais promissoras. A endoglina, também conhecida como CD105, é um receptor da superfamília do TGF-β, expresso nas células-tronco hematopoiéticas (HSC) de todos os sítios hematopoiéticos, incluindo a medula óssea, onde é descrita como um marcador para HSC de longo prazo. Apesar da expressão de CD105 ter sido relacionada a diversos tipos de tumores sólidos, principalmente devido ao papel desse receptor na angiogênese, relativamente pouco é conhecido em relação a expressão de CD105 e a sua função em neoplasias hematopoiéticas. Este estudo revelou alta expressão de endoglin na maioria dos blastos de pacientes com leucemia mieloide aguda (LMA) e leucemia linfoblástica aguda (LLA). Utilizando um modelo de xenotransplante, verificamos que as blastos CD105+ possuem uma actividade leucemogênica superior em comparação com a população de CD105-. Adicionalmente, investigamos se o bloqueio da endoglina, usando TRC105, poderia resultar em uma opção terapêutica para tratamento das leucemias agudas e descobrimos que na LMA, o TRC105 impediu o engraftment de blastos primários e inibiu a progressão da leucemia após o estabelecimento da doença, mas na LLA, o TRC105 sozinho foi ineficaz devido à uma maior secreção da forma soluvél da endoglina (sENG). No entanto, tanto na LLA quanto na LMA, TRC105 potencializou o efeito terapeutico da quimioterapia padrão e inibiu a progressão da doença, indicando que TRC105 pode representar uma nova opção terapêutica para LLA e LMA.
Successful treatment of acute leukemia remains a clinical challenge due to the toxicity and the relatively poor responses to the current standard therapy. Thus, treatments that address novel therapeutic targets are urgently needed and, for this purpose, monoclonal antibodies have been one of the most promising strategy once they are able to deliver their therapeutic effects with minimal toxicity. Endoglin, also known as CD105, is a receptor of the transforming growth factor-beta (TGF-β) superfamily that has been found expressed in hematopoietic stem cells (HSCs) from all hematopoietic sites, including the bone marrow, in which it is described as a marker for long-term HSC. CD105 is also found to be expressed in several cancers. However, because CD105 expression has been studied mostly in the context of solid tumors and angiogenesis, relatively little is known about CD105 expression and role in hematopoietic malignancies. We identified endoglin expression on the majority of blasts from patients with acute myeloid leukemia (AML) and acute B-lymphoblastic leukemia (B-ALL). Using a xenograft model, we find that CD105+ blasts are endowed with superior leukemogenic activity compared to the CD105- population. We test the effect of targeting this receptor using the monoclonal antibody TRC105, and find that in AML, TRC105 prevented the engraftment of primary AML blasts and inhibited leukemia progression following disease establishment, but in B-ALL, TRC105 alone was ineffective due to the shedding of soluble CD105. However, in both B-ALL and AML, TRC105 synergized with reduced intensity myeloablation to inhibit leukemogenesis, indicating that TRC105 may represent a novel therapeutic option for B-ALL and AML.

Full textThe antisecretory factor (AF) is a protein secreted in plasma and other tissue fluids in mammalians which was shown to be a potent inhibitor of intestinal fluid secretion and inflammation. After the AGP ban, several nutritional approaches aimed to the optimization of the weaning transition and reduction of gut diseases have been proposed, but the success rates are really low compared to antimicrobials. AF-inducing diets appear to be suitable alternatives to AGP. Indeed AF content in sows’ colostrum and milk appears to be a protection factor against diarrhoea in suckling piglets. Increased AF level in plasma by dietary means, such as feeding hydro-thermally processed cereals (HPC), has been demonstrated in human and animals. We tested the effect of two different HPC level of inclusion in a wheat-barley based diet on weaned piglets growth performance, reared in an experimental farm. The results confirmed the efficacy of HPC as growth promoters in piglet nutrition: HPC supplementation improved ADG and FCR. In vitro test were made to study the antisecretory and anti-inflammatory properties of the AF protein and its mechanism of action by using AF-16, the active region of the protein. The Ussing chamber experiments performed on polarized IPEC-J2 cells confirmed the neuronal involvement in the antisecretory activity of AF. In fact, AF-16 did not inhibit CT-induced Isc. A slower rate of Isc increase was observed during the simultaneous administration with AF-16 and CT. High dosages of AF-16 were found to reduce the LPS-stimulated NO production in RAW264.7 cell, thus supporting the hypothesis of an anti-inflammatory action. On the contrary, no significant results were obtained on PMA-stimulated ROS generation in pig alveolar macrophages.
