Abstract:
This article tries to show that the formulaic diction on the level of verse line and formulaic patterning in the composition of scenes are closely related and must be studied together. The analysis is done on the example of Turkic epics. Of the formulaic patterns the most prominent one is the variety of use of the attribute ak (white), which appears to be one of the most common epithets in Turkic epic poetry. It is usually connected with cloth (e.g. caftan, yurt), different parts of body (face, bosom), antelope, the lumps of gold given as bride-price and various kinds of arms (sword, spear) etc. It is usually denoted evaluatively as purity and beauty. In this matter Turkic epics share its position with many national epics of the middle ages including Serbian, Old English, Old German etc. The same role is analyzed for the opposite pattern ?dust of earth?, and for the two themes: preparation of the hero for his journey and council scenes which are also mutual to many medieval epic traditions such as aforementioned Serbian and others.

Abstract:
Previous studies demonstrated that chlorzoxazone or 1-ethyl-2-benzimidazolinone (1-EBIO) enhances transepithelial Cl− secretion by increasing basolateral K+ conductance ( G K) (Singh AK, Devor DC, Gerlach AC, Gondor M, Pilewski JM, and Bridges RJ. J Pharmacol Exp Ther 292: 778–787, 2000). Hence these compounds may be useful to treat cystic fibrosis (CF) airway disease. The goal of the present study was to determine whether chlorzoxazone or 1-EBIO altered ion transport across ΔF508-CF transmembrane conductance regulator homozygous CFT1 airway cells. CFT1 monolayers exhibited a basal short-circuit current that was abolished by apical amiloride (inhibition constant 320 nM) as expected for Na+ absorption. The addition of chlorzoxazone (400 μM) or 1-EBIO (2 mM) increased the amiloride-sensitive I sc ∼2.5-fold. This overlapping specificity may preclude use of these compounds as CF therapeutics. Assaying for changes in the basolateral G K with a K+ gradient plus the pore-forming antibiotic amphotericin B revealed that chlorzoxazone or 1-EBIO evoked an ∼10-fold increase in clotrimazole-sensitive G K. In contrast, chlorzoxazone did not alter epithelial Na+ channel-mediated currents across basolateral-permeabilized monolayers or in Xenopus oocytes. These data further suggest that alterations in basolateral G K alone can modulate epithelial Na+ transport.

AbstractTetraspanins, such as CD81, can form lateral associations with each other and with other transmembrane proteins. These interactions may underlie CD81 functions in multiple cellular processes, such as adhesion, morphology, migration, and differentiation. Since CD81's role in neuronal cells’ migration has not been established, we here evaluated effects of CD81 on the migratory phenotype of SH-SY5Y neuroblastoma cells. CD81 was found enriched at SH-SY5Y cell's membrane, co-localizing with its interactor filamentous-actin (F-actin) in migratory relevant structures of the leading edge (filopodia, stress fibers, and adhesion sites). CD81 overexpression increased the number of cells with a migratory phenotype, in a potentially phosphatidylinositol 3 kinase (PI3K)–Ak strain transforming (AKT) mediated manner. Indeed, CD81 also co-localized with AKT, a CD81-interactor and actin remodeling agent, at the inner leaflet of the plasma membrane. Pharmacologic inhibition of PI3K, the canonical AKT activator, led both to a decrease in the acquisition of a migratory phenotype and to a redistribution of intracellular CD81 and F-actin into cytoplasmic agglomerates. These findings suggest that in neuronal-like cells CD81 bridges active AKT and actin, promoting the actin remodeling that leads to a motile cell morphology. Further studies on this CD81-mediated mechanism will improve our knowledge on important physiological and pathological processes such as cell migration and differentiation, and tumor metastasis.

Hypertonic saline (HS) inhalation therapy benefits cystic fibrosis (CF) patients [Donaldson SH, Bennet WD, Zeman KL, Knowles MR, Tarran R, Boucher RC. N Engl J Med 354: 241–250, 2006; Elkins MR, Robinson M, Rose BR, Harbour C, Moriarty CP, Marks GB, Belousova EG, Xuan W, Bye PT; the National Hypertonic Saline in Cystic Fibrosis (NHSCF) Study Group. N Engl J Med 354: 229–240, 2006]. Surprisingly, these benefits are long-lasting and are diminished by the epithelial Na+ channel blocker amiloride (Donaldson SH, Bennet WD, Zeman KL, Knowles MR, Tarran R, Boucher RC. N Engl J Med 354: 241–250, 2006). Our aim was to explain these effects. Human bronchial epithelial (hBE) cells from CF lungs were grown in inserts and were used in three experimental approaches: 1) Ussing chambers to measure amiloride-sensitive short-circuit currents ( INa); 2) continuous perfusion Ussing chambers; and 3) near “thin-film” conditions in which the airway surface of the inserts was exposed to a small volume (30 μl) of isosmotic or HS solution as the inserts were kept in their incubation tray and were subsequently used to measure INa under isosmotic conditions (near thin-film experiments; Tarran R, Boucher RC. Methods Mol Med 70: 479–492, 2002). HS solutions (660 mosmol/kgH2O) were prepared by adding additional NaCl to the isosmotic buffer. The transepithelial short-circuit current ( ISC), conductance ( GT), and capacitance ( CT) were measured by transepithelial impedance analysis (Danahay H, Atherton HC, Jackson AD, Kreindler JL, Poll CT, Bridges RJ. Am J Physiol Lung Cell Mol Physiol 290: L558–L569, 2006; Singh AK, Singh S, Devor DC, Frizzell RA, van Driessche W, Bridges RJ. Methods Mol Med 70: 129–142, 2002). Exposure to apical HS inhibited INa, GT, and CT. The INa inhibition required 60 min of reexposure to the isosmotic solution to recover 75%. The time of exposure to HS required to inhibit INa was <2.5 min. Under near thin-film conditions, apical exposure to HS inhibited INa, but as osmotically driven water moved to the apical surface, the aqueous apical volume increased, leading to an amiloride-insensitive decrease in its osmolality and to recovery of INa that lagged behind the osmotic recovery. Amiloride significantly accelerated the recovery of INa following exposure to HS. Our conclusions are that exposure to HS inhibits hBE INa and that amiloride diminishes this effect.

Abstract:
A distinct difference between forces, especially bending moments, in slabs of slab-and-beam deck of reinforced concrete bridges, determined by the methods of analysis, applied in the former USSR and France, is pointed out in the article. The essence of calculation of forces in the slab due to bridge loads is presented. Deformations of deck as a spatial structure are resolved into local between beams and spatial characteristics for the whole deck. Local bending slab moments due to local deformations are calculated approximately using diagrams of a continuous and with built-in-end beam. It is shown that forces only due to local deformations are evaluated in the method of the former USSR. A more accurate modelling of deck slab behaviour is obtained by using spatial diagrams composed of plate and beam elements or of spatial diagrams composed of plate and beam elements or of even more accurate shell-type elements. Calculations have been performed by the use of three finite-element method programs Lira, Gifts and Staad3. Decks of various length and width are loaded with combinations of permanent and variable, traffic AK and HK-80 loads. More accurately strain and stress states are modelled in non-linear calculations accounting for non- elastic deformation and cracking of slab concrete as well as cracking of beams. The investigation of results has shown that the method of the former USSR, still applied in Lithuania today, erroneously evaluates slab load-effect resulting in unacceptedly reduced positive bending moments in the larger central deck part. Recommendations to calculate these load-effects are proposed. By simplified two-stage- design methods in use to-day when at the first stage basic load effects Mo and Qoare calculated employing diagrams of simple beam (USSR), or that of slab supported on two sides (France), and in the second stage design loads-effects are determined by multiplication of basic values and coefficients. Recommended values of such coefficients for basic bending moments Mo determined by beam method are shown in Fig. 7a, and those by slab method—in Fig. 7b. Other recommendations for evaluation of shear forces and longitudinal moments are also presented.

Abstract:
The article critically assessed temporary vertical standard combined loads on road bridges in Western European countries, the USA and Russia. They are set with constant values of the parameters of their uniformly distributed loads and loads on the axles of trucks without regard to the composition of road traffic and out of communication with the length and shape of the lines of influence of force and deformation factors in the elements of bridge structures and are used in these and in many other countries in road bridge structures engineering and design. It is shown that the bridges constructed with different spans on the same road are designed to have different and unknown for designers, engineers and facility managers load capacity, which decreases when the length of the spans increases more than twice, but that is unacceptable for operational and economic reasons. The author found that the use of regulatory documents of other countries as a model for the modernization of domestic standards is unacceptable due to economic considerations. The article assesses the carrying capacity of such bridges as: the Crimean bridge and the Bugrinsky bridge in Novosibirsk. It is shown that these structures, in case of compliance with the requirements of modern regulatory documents of the Russian Federation, do not allow for the passage of columns of vehicles weighing more than 25–27 tons in a state of congestion. The ways of eliminating the identified deficiencies of these combined regulatory loads are indicated. The results of calculations are presented, indicating that a further increase in the parameter K of the AK load to large values is not an effective way of ensuring the required carrying capacity of bridge structures.

ABSTRACTCostume, dress code, animation, comics, legends, and manga, are inseparable parts of the cosplay costume. Those parts give fantasy and digital world discourse through costume style. Its spiritual domain stands on Japanese culture by being cultured through clothing. One of them, cosplay ideo- logy, reflects the self-imaging through social communities, as an e?ort for group and self-existence. Cosplay entity bridges fantasy and real world, presents designers’ expressions through the costume designs to show. This writing will be analyzed by using the main theories based on Dewitt H. Parker point of view, in The Principles of Aesthetics, which divides principles of aesthetics into three, they are: Principle of Organic Unity, Principle of Dominant Element, and Principle of Balance. Principle of organic unity indicates that cosplay clothing is an accumulation of design elements, to refer and mark a figure. Principle of dominant element, is accentuation, or the center of interest of a cosplay clothing design.
