In this study, the suitability of rodents for the detection and assay of microbial pyrogens was investigated and the pathophysiology of the 'yeast rat' model, currently used for screening antipyretic drugs, was documented. Mice do not consistently develop pyrexia following systemic injections of bacterial endotoxin (BE). The occupational chemicals 4-vinylcyclohexene (VCH) and butadiene diepoxide (BDE) have shown an ovarian effect in small pre-antral follicles in mice and rats (Flaws et al., 1994b; Doerr et al. , 1995). VCH exists as two enantiomers, S-VCH and R-VCH. Daily dosing with the enantiomers affected small primary follicles and wet tissue weights in B6C3F₁ mice. The S-enantiomer decreased small primary follicle numbers (P < 0.05) and both enantiomers reduced the weights of the ovary and uterus (p < 0.05). Qualitative and quantitative ultrastructural observations have been made on myelinated fibres in the tibial and medial plantar nerves of genetically diabetic C57/BL/Ks(db/db) and age-matched nondiabetic (m/m) control mice at 9 and 15 months of age. The cross sectional area of myelinated fibres and their axons in both nerves was found to be significantly less in diabetic mice when compared with age-matched controls. 4-Vinyl-1-cyclohexene diepoxide (VCD) is a known carcinogen, a direct acting mutagen, and an ovarian toxicant. Female B6C3F₁ mice are more susceptible to the ovarian toxicity of VCD than female Fischer 344 rats, a difference which may be explained by the disposition/metabolism of VCD. Following a single dose ¹⁴C-VCD was rapidly metabolized and excreted predominantly in the urine of both species. However, the excretion rate was higher in the female rats compared to female mice. Current lifestyle with high-energy diets and characterized by sedentary is triggering an alarming growth in obesity. Obesity along with metabolic syndrome- related co-morbidities (i.e. insulin resistance, atherosclerosis, sleep apnea, depression, asthma, hypertension and the alteration of blood lipid transport) are the most apparent consequence of the excess energy. Type 2 diabetes is caused by defect pancreatic islet β-cells together with peripheral insulin resistance. The disease is often accompanied by obesity with associated low-grade visceral adipose tissue inflammation, which contributes to insulin resistance. The organization and transcription of the gene for vasoactive intestinal polypeptide (VIP) in rats and mice was investigated using northern- and Southern-blot hybridizations, selective genomic cloning, Sl-nuclease protection assays, oligonucleotide-directed RNase H digestions, and genomic cloning by standard methods. The center of the rat VIP gene and the entire mouse gene were cloned and sequenced. It is a biopolymer containing primarily membrane glycoproteins and polar lipids which contribute to its properties as a possible neutraceutical.
