The Alchian and Allen theorem states that when a common per unit fee is added to the prices of high and low quality goods, the relative price of the high quality good falls, and its relative consumption increases. The theorem has been analyzed in the literature under the assumption that the prices of the goods are exogenous. This thesis presents a spatial equilibrium model that drops this restriction. The comparative statics analysis developed in this thesis does not support the theorem proposition in the general case. The theorem is then tested empirically on U.S. hard wheat exports. Transport costs, represented by ocean grain freight rates, are not found to have a significant influence on the average quality of U.S. wheat exports for the group of importing countries with a high per capita incomes that have not received U.S. export subsidies. However, transport costs are shown to be positively related with the average quality of U.S. wheat exports for the group of low-income importing countries. Additionally, it is shown that there exists an inverse relationship between the relative price of high quality wheat and the average quality of U.S. wheat exports for high-income countries.

This document is focused on three excerpts from a graduate vocal recital, completed in partial fulfillment of the requirements for the Master of Music degree. The recital was held on January 23, 2012 at seven-thirty o’clock in the evening at All Faiths Chapel on the campus of Kansas State University. Selections for this recital were selected upon the theme of Seclusion. The three characters examined further in the document are Alcina in the G. F. Handel opera, Alcina (HWV 34), Gretchen in German Lieder Gretchen am Spinnrade (D. 118) by Franz Schubert, and Lily from the Tony Award-winning Broadway musical, The Secret Garden, by Lucy Simon and Marsha Norman. Each chapter, devoted to a specific work, contains the following: 1) biographical information on the composer, 2) textual analysis, and 3) compositional, stylistic, and technical considerations.

This project analyzes the ways that Louis May Alcott portrays authors in several texts, including Hospital Sketches (1863), "Enigmas" (1864), "Psyche's Art" (1868), Little Women (1868), A Modern Mephistopheles (1877), and Diana and Persis (1878). An examination of prevailing contemporary theories of authorship reveals that Alcott's interest in authorship (as shown through her experiences as a writer and the author figures she depicts within her writing) cannot be adequately analyzed under any of the existing theoretical frameworks because the theories neglect to consider markers of racial, sexual, cultural, and class-based difference. Being a female author in nineteenth-century America was, for Alcott, a preoccupation. Thus much of her writing features representations of authors. For Alcott, as well as many of her female contemporaries, the question "What does it mean to be an author?" cannot be considered without also asking, "What does it mean to be a woman?" and "How can an author be represented in a text?" Alcott's treatment of these questions in her writing was her attempt to create a dialogue between herself, other writers, and her reading public. By studying Alcott's author figures, I advance a model of authorship that highlights issues of gender and multiplicity; in this way my work has applications to other authors who have been excluded by normative definitions of authorship. The concept of "mutable authorship," a model that more accurately incorporates Alcott's treatment of authorship, is the product of several different literary, historical, and feminist theoretical lenses. This dissertation works through the different structuring figures that Alcott uses to represent the author, beginning with the semi-autobiographical first-person narrator and moving to the more metaphorical figures of the artist and the performer. The discussion culminates with the exploration of adaptation and collaboration in the three Hollywood feature films of Alcott's best-known work, Little Women, and several recent texts that respond directly to Alcott's work.

Considering the need for the natural resources protection and conservation, the Water Framework Directive 2000/60/EC Directive was adopted, involving the efforts of all Member States in the task of achieving good status for all waters, in which the aim of achieving good ecological state for natural ecosystems can be included. Within this context, the main objectives of this study are: the characterisation and quantification of the degradation degree in the fluvial ecosystem of the Alcoa river drainage basin, and to that end it was used the Predictive Ecological State methodology; and planning restoration actions for the most degraded stretches of the river system which deteriorate under the effect of human actions. Based on the results obtained, a Map of River Corridor Conservation Status was developed for the Alcoa river catchment area with the intend of revealing the degree of degradation of the river streams, and a Map of River Restoration Planning was developed which summarizes the restoration actions to be implement in the drainage basin.

La période allant de l’invention de Mélusine (c. 1390) à sa réinterprétation dans l’Alector (1560) peut être vue comme un « âge d’or de la féerie », qui s’étend hors de son domaine d’origine (le merveilleux) et fait émerger des figures neuves. Loin de disparaître de l’imaginaire des XVe et XVIe s., les fées en sont une composante essentielle. L’étude chronologiquement délimitée par ces repères et prolongée, pour certaines figures, jusqu’aux romans baroques (d’Urfé et Rosset c. 1612) dresse un tableau de la féerie au féminin. Organisée autour des quatre fées « cardinales », Morgane, Mélusine, Alcine et Urgande, nommées dans le programme des fêtes royales de Bayonne (1566), elle analyse leur évolution du point de vue de l’écriture et de sa réception. Elle cerne aussi des figures qui perdent leur identité féerique (Sibylle, Méridienne) et en recherche les causes. Tout en marquant des continuités et des parentés qui tiennent à des lignages « fictionnels » ou dynastiques, elle analyse des mutations (allégorisation, idéalisation) qui font valoir la richesse de la matière et son rôle dans l’évolution de l’imaginaire et des Lettres, de la fin du Moyen Âge à la RenaissanceFrom Melusine’s first appearance in literature (c. 1390) to its new treatment in the Alector (1560), the late Middle Ages and Renaissance period can be seen as the « golden age of faery », as it expands beyond its original field (the marvellous) and shapes up some new figures. Far from vanishing from the 15th and 16th centuries fiction, faery is a crucial part of it. Within these chronological boundaries and slightly beyond, as some figures are still mentioned in baroque novels (such as d’Urfé’s and Rosset’s c. 1612), this study makes a broad inquiry into feminine faery. Built up around the four « cardinal » faeries – Morgan, Melusine, Alcina and Urganda – still featuring in the royal feasts of Bayonne in 1566, it focuses on their evolution through writing and reading. It also points out Sibyl and Meridiana as figures who tend to lose their faery quality. While showing some continuity among faeries due to fictional kinship or dynastic lineage, it investigates the disruption caused by allegory and idealization, thus bringing into light a huge material and its decisive role in the shaping of imagination and literature, from the late medieval period to the Renaissance

This thesis will explore how social and political conditions in archaic Greece affected the composition of poetry for female choral performance. My primary source material will be the poetry of Alcman and Sappho. I examine the evidence suggesting that poems by both Alcman and Sappho commented on political issues, using this as a basis to argue that women in archaic Greece may have had a more vocal public presence that has previously been imagined. Rather than viewing female performance as a means of discussing purely feminine themes or reinforcing the idea of a disempowered female gender, I argue that the poetry of Alman and Sappho gives parthenoi an authoritative public voice to comment on issues in front of the watching community. Part of this authority is derived from the social value of parthenoi, who can act as economically and socially valuable points of exchange between communities, but I shall also be looking at how traditional elements of female performance genre were used to enhance female authority in archaic Sparta and Lesbos. Once this has been established, this thesis will proceed to examine how public female performance dealt with major political and social issues in the archaic world. I shall argue that the performance of parthenoi did focus on primarily feminine concerns such as marriage, desire, and abduction, but that it could also be an opportunity to discuss much broader political themes that were of major importance to the entire polis. Alcman and Sappho use their poetry as a vehicle to comment on the society in which their poetry was composed, both discussing threats to order and representing solutions for a stable society. The content of female performed poetry was often composed as much for a male audience as for the performers themselves, using traditional female performance as a means of commenting on the current political climate. Through arguing these factors, the intention of this thesis is to suggest a much more prominent public role for archaic Greek females than has previously been recognised.

My thesis investigates the processes of reciprocal, transatlantic literary exchange between Britain and the United States in the nineteenth century. While these specific transnational relations have received much critical attention in recent years, I extend current theoretical frameworks by focusing on how women‘s domestic fiction operates as a currency for literal and ideological interchanges between Britain and the United States. Concentrating primarily upon Elizabeth Gaskell‘s and Louisa May Alcott‘s fictions, I trace how they operate as 'transatlantic domestic narratives‘. I use this term to refer to the mobility of their material texts as they circulate within a transatlantic community, and also to articulate the generic narrative tropes on which their domestic fictions rely. I explore, therefore, how the rhetoric of domesticity – as transmitted through the transatlantic domestic narrative – becomes a shared medium through which specific localised concerns can be articulated and circulated within a transatlantic arena. Focusing on four domestic tropes which were common on both sides of the Atlantic – home, the worker, the nurse, and the witch – I illustrate how both Gaskell and Alcott mobilise these four narrative structures in order to contribute to local and transnational debates in which national, literary and gendered identities are created and contested. Both authors‘ fictions, I demonstrate, exemplify, and have a significant impact upon, a transatlantic literary marketplace.

Previous issue date: 2003-10-03The relation between Alcoa and the regional socioeconomic in Maranhão. We critically analysis the action of a multinational company at a peripheral area. We emphasize the external and internal factors in a capitalist globalization context of Brazilian economy, due to foreign investment in Brazil, specific in Maranhão. We also analysis the public powers participation in Alcoa s implantation process in São Luis and its consequences on Maranhense economy. Finally, we analysis Alcoa s action in Maranhão based on its own socioeconomic indicators.
Relação entre a ALCOA e o desenvolvimento socioeconômico regional no Maranhão. Analisa-se criticamente a atuação de uma empresa multinacional num espaço regional periférico. Destacam-se os condicionantes exógenos e endógenos à economia brasileira que no contexto da globalização capitalista, favorecem a penetração do investimento estrangeiro direto no Brasil e, particularmente, no Maranhão. Analisa-se a participação dos poderes públicos no processo de implantação da ALCOA em São Luís e os efeitos diretos e indiretos decorrentes de sua implantação sobre a economia maranhense. Interpreta-se a atuação da ALCOA no Maranhão a partir da demonstração de alguns indicadores socioeconômicos deste Estado.

Cílem diplomové práce bylo stanovit hodnotu Sdružení Strojmetal Kamenice, a.s. a Alcan Singen GmbH. Diplomová práce zahrnuje dvě hlavní části ? metodologickou a aplikační část. Metodologická část obsahuje potřebné teoretické poznatky k vypracování části praktické. V rámci aplikační části je nejprve uvedena charakteristika odvětví, ve kterém Sdružení působí, a charakteristika Sdružení. Dále je zde zpracována analýza okolí (makrookolí a mikrookolí) a finanční analýza, na jejímž základě byl následně sestaven finanční plán. Poslední část aplikační části je věnována stanovení hodnoty Sdružení, k čemuž byly využity výnosové metody, konkrétně modely DCF s využitím FCFF a EVA, které určují hodnotu podniku jako celku.

This thesis deals with a summary of individual method for determining of hyaluronan concentration, especially in aqueous solutions. Selected methods were compared to their functionality in university workplace. Any functional methods may be used for many purposes in university workplace in research of materials based on hyaluronan. It has been studied usability to assess residual content of hyaluronan in supernatant of gel formed from hyaluronan and surfuctant, in this thesis. We choosed sptectrophotometric methods, especially with UV spectroscopy and IR spectroscopy. Two precipitation methods using alcian blue were also tested. As the most reliable, the least demanding and specific method seems to be the method using IR spectroscopy, thanks to it we are able to determine the concentration of HA from 0.01 %. With this method, it has been found, that if there will be located some amount of HA in supernatant, it will be below the detection limit of assay. The remanining methods were modified to help determine the concentrations of HA, but works only in aqueous solutions. A possible variation for determination of very low concentrations of HA may be the method using ELISA kits. However their cost is higher than the above-mentioned methods.

O objetivo principal deste trabalho é analisar como se dá a continuidade da consolidação do Mercosul, a ampliação das relações com a União Européia e o momento de formação da ALCA (Área de Livre-Comércio das Américas) no conjunto da política externa brasileira. Isto é, como desenvolve-se o processo decisório baseado em possibilidades de integração a uma economia globalizada que contempla basicamente econômicos deixando em segundo plano aspectos como soberania e legitimidade. A partir do cenário em questão e de conceitos como hegemonia, sistema-mundo e imperialismo, discutimos a possibilidade para que se construa uma nova composição de forças que possibilitem uma outra orientação política fruto da vontade coletiva resultando lutas sociais que gerem ações políticas diferentes das presentes atualmente nos discursos do governo brasileiro. This thesis examines why and how art is collected and used by modern corporations. Primarily, the study is focused on Alcan Aluminium Ltd., but for purposes of comparison and illumination, substantial attention is paid to Martineau Walker, Loto-Quebec and the Banque Nationale du Canada, all Montreal-based companies that have collected art for at least twenty years.An historical introduction outlining the relationship between the arts and business---from the Renaissance in Europe to its place of greatest expansion in the nineteenth and twentieth centuries; in the U.S.A. as well as in Canada---begins the discussion. An attempt is made to position the activities of modern corporate art collecting between two poles---that of the individual collector and that of the institutional art museum.
Motives for collecting include, among many, the search for distinction and the desire to create a perception in the community of humanitarian standards guiding the corporation. The ways in which corporate motives contribute to the creation of a positive external corporate art identity, and thus a productive business environment, are examined and developed, as are the principal objections of their chief public detractor, artist-writer Hans Haacke.
In three of the four corporations studied, original research on the behavioural and emotional reactions of employees to the corporate art surrounding them has allowed an in-depth analysis of the internal effects of art on employees. Foremost among these are feelings of pride, well-being and the enhancement of self-identity.
Finally, a brief discussion of the role of mediators is offered, in particular that of the corporate curator in the modern art world.

O Brasil vivencia crise de natureza existencial, refletindo sua inadequação aos signos, códigos e retórica da ideologia mundialmente dominante e desencadeando a busca de uma mais atualizada e consentânea modelagem institucional.O estudo justifica-se, na busca de compreender o conflito entre a soberania dos estados- nacionais e os mandamentos da globalização, quer por um olhar, mesmo ligeiro, sobre o passado, quer à luz de alguns fatos contemporâneos. Desenvolvemos este estudo a partir do método dialético. Tomamos a dialética na concepção de Hegel, que com a sua preocupação com a história, a torna uma descrição prática do progresso cotidiano do mundo. O processo dialético é, por natureza própria, um progresso. Para conhecer realmente a história da globalização no Brasil, é preciso estudá-la em todos os seus aspectos, em todas as suas relações e em todas as suas conexões. Quanto ao delineamento, trata-se de uma pesquisa documental; o material pesquisado foi o conjunto de publicações acerca da globalização, no caso específico da ALCA e suas relações com o Brasil. As publicações foram localizadas nas bibliotecas da Universidade de Pernambuco, nas bibliotecas públicas do Estado, na Internet, em registros estatísticos, documentos e nos meios de comunicação de massa. Esperamos com o estudo mostrar que o conhecimento histórico é perspectivista, ou seja, afasta de si o passado e quer entendê-lo no seu tempo e lugar, mas não assimilá-lo ou reduzi-lo ao presente; é individuante, porque individualizado por dois parâmetros fundamentais o cronológico e o geográfico; pelo material documentário da História e pelos critérios de opção historiográfica na medida em que tende a evidenciar um fato entre os outros, sublinhar-lhe a importância e, portanto, o seu caráter singular e único; é, ainda, seletivo, e, a partir desses resultados, mostrar que a ALCA é apenas um dos cenários possíveis do diálogo da globalização entre brasileiros e norte-americanos cujas agendas bilaterais extensas abrangem interesses conflitantes e metas convergentes

de Louisa May Alcott, para a língua portuguesa.
Tomando como suporte teórico deste trabalho os procedimentos de tradução
preconizados por Jean-Paul Vinay e Jean Darbelnet e os contributos de Itamar Even-
Zohar, Gideon Toury, André Lefevere e Lawrence Venuti no âmbito da tradução
literária, pretendemos salientar a diversidade de fatores que entram em jogo na tradução
literária enquanto processo intercultural, realçando que, neste campo, há sempre uma
negociação entre perdas e ganhos nas apostas interpretativas que deverão ser feitas pelo
tradutor.
Sendo certo que nenhum texto pode ser traduzido em todos os seus aspetos, e não
tomando como adequada apenas uma abordagem, procurámos adotar uma posição de
compromisso e responder ao que é exigido ao “tradutor negociador” (Eco, 2005).
Desenvolvemos, assim, um exercício de equilíbrio que procurou não “apagar” o Outro,

### Abstract:
This project work aims to present a translation of the short stories “M.L.”, “The
Brothers”, “A Hospital Christmas” and “Nelly’s Hospital”, by Louisa May Alcott, into
Portuguese.
Taking as theoretical basis the translation procedures advocated by Jean-Paul
Vinay and Jean Darbelnet, and the contributions of Itamar Even-Zohar, Gideon Toury,
André Lefevere and Lawrence Venuti in the context of literary translation, we intend to
point out the diversity of factors that come into play in literary translation as an
intercultural process and emphasise the fact that, in this field, there is always a
negotiation between losses and gains in the interpretive choices made by the translator.
Certain that no text can be translated in all its aspects, we did not consider only
one approach as adequate, and attempted to adopt a compromise position and respond to
what is required to a " translator as a negotiator" (Eco, 2005).We tried to establish a
balance that would not “erase” the Other, nor jeopardise the fluency and readability of
the translated texto.

No Brasil, até os anos 90, o empresário teve baixo grau de envolvimento nas negociações comerciais internacionais. Com o processo de abertura democrática, a aceleração do processo de globalização e a abertura comercial do Brasil, as negociações comerciais ganharam maior relevância para o empresariado nacional. Este fato levou-o a mobilizar-se e a desenvolver ações para aumentar sua influência nos processos de negociação, disso resultando a criação da Coalizão Empresarial Brasileira, em 1996, que se transformou na principal interlocutora do empresariado com governo nas negociações internacionais. This thesis examines the challenges of managing a global supply chain in a large, well-established organization and outlines certain techniques that can be utilized to achieve more effective supply chain management. The research was conducted at Davenport Works, which is part of the Alcoa Mill Products (AMP) Business Unit, and examined the business unit's global supply chain with its European customers. The presence of inventory can hide many of the root cause problems within a supply chain and the project driver for this work was clearly inventory reduction. However, while excessive inventory is clearly a problem and organizations should strive to reduce unnecessary inventory as much as possible, there is an optimal amount of inventory that should be maintained and that amount is rarely zero. Inventory is held for a variety of reasons and can be utilized as a tool to countermeasure the primary factors that influence inventory requirements: customer demand, demand variability, production yield, production yield variability, lead time, lead time variability, and desired customer service levels. Alcoa utilizes inventory as a countermeasure within their supply chain for a variety of reasons. Customers are demanding increasing levels of service; and their demand patterns are variable. Replenishment lead times are long (on the order of months) and variable. Davenport Works is striving to achieve economies of scale; and their production yields are variable and often times deviate significantly from the customer's forecasted consumption rate. Currently, high levels of inventory are being maintained throughout the supply chain; and desired customer service level targets are not being met. AMP has no formal methodologies to both characterize the reasons why inventory is being maintained and to determine what inventory requirements they need to satisfy each specific customer program. In addition, AMP is driving cost reductions throughout the entire organization. This is forcing the organization to justify the inventory they currently have and also putting pressure on the organization to reduce inventories throughout the supply chain. This thesis has three primary objectives. Firstly, to provide a detailed analysis of the entire AMP supply chain for its European customers and articulate the reasons why AMP is maintaining inventory. This includes a discussion about supply chains, supply chain management, and the role of inventory in the supply chain. Secondly, to describe a methodology, which can be applied to engineer inventory levels for each product. The base stock model was used for this and is an excellent tool to demonstrate how supply chain variables impact inventory requirements, target areas for improvement, and quantify inventory requirements in a systematic manner. Thirdly, to provide recommendations to improve overall supply chain performance and optimize inventories.

Numerous events during development require the tightly controlled and regulated interaction of cells - from gastrulation in the early embryo to axonal pathfinding and remodeling of synaptic networks. Each of these events is dependent upon signals generated by cell-cell interactions, which are in turn specified by a diverse number of cell adhesion molecules. Many families of cell adhesion molecules have been described, and these fall into the broad categories of cadherins, immunoglobulin superfamily (IgSF) members, selectins, and integrins. Activated Leukocyte Cell Adhesion Molecule (ALCAM) is a member of the IgSF, and controls numerous developmental processes, ranging from hematopoiesis to neuronal targeting. Furthermore, this protein has been implicated in the progression of numerous cancers of diverse origins. Despite the variety of developmental and pathological processes in which ALCAM has been implicated, little is known about how it signals in the cell - few extracellular binding partners have been isolated, and, as of this writing, no cytoplasmic interactors have been identified. The purpose of the work presented in this thesis was to elucidate the mechanisms by which ALCAM influences cell behavior, specifically in uveal melanoma cells, and to determine novel extra- and intracellular ligands. Here, I report the regulation of cadherin-based junctions by ALCAM in uveal melanoma cells, as well as provide evidence for a novel extracellular interaction with L1 cell adhesion molecule, and identify three novel intracellular binding partners.

Resumo: Nesta dissertação fizemos um estudo semântico da designação das palavras integração e progresso a partir de suas enunciações nos documentos que propõem a constituição da ALCA (Área de Livre Comércio das Américas). Trabalhamos numa perspectiva semântico-enunciativa que leva em conta a relação língua, história e sujeito. Nesta medida estabelecemos uma relação com a teoria da análise do discurso francesa. O conceito de enunciação como acontecimento e o conceito de designação foram fundamentais para nossas análises. O modo pelo qual procedemos as nossas análises foi através da leitura dos procedimentos de reescrituração e mais ainda dos procedimentos de articulação para se chegar ao domínio semântico de determinação (DSD) das palavras em questão. Analisamos ainda integração enquanto enunciado nominalizado e com isso pudemos trabalhar um pouco a questão da memória. Saber os sentidos dessas palavras nestes textos é um modo de procurar compreender o que a designação dessas palavras traz a propósito das relações políticas estabelecidas na ALCA. A partir da análise das palavras, tratamos da relação de litígio enunciativo estabelecido entre a ALCA e a ALBA (Alternativa Bolivariana para a América Latina e o Caribe). Abstract: In this master's thesis we have proceeded with a semantic study of the designation (designação, in Portuguese) of the words integration and progress from the enunciations contained in the documents which propose the constitution of the FTAA (Free Trade Area of the Americas). We have worked in the semantic-enunciative perspective that takes the relationship amongst language, history and subject for granted. This way, we could establish a relationship with the theory of the French Discourse Analysis. The concept of enunciation as an event, as well as the concept of designation, was fundamental to our analyses. The mode by which we have conducted our analyses was through the reading of the procedures of rewriting, and, furthermore, through the procedures of articulation in order to get to the semantics domains of determination (domínios semânticos de determinação, in Portuguese) of the word being put into question. We have still analysed the word integration as a nominalised enunciation, and, through this, we were able to worka bit with the issue of the memory. Knowing the senses of such words in these texts represents a mode of trying to understand what the designation of these words brings to the purpose of the public relations settled at the FTAA. From the analysis of these words, we have discussed about the relationship of enunciative litigation between the FTAA and the Bolivarian Alternative for Latin America and Caribbean (ALBA, in Spanish). This is a political litigation characterised as a confrontation and a conflict between a normativity of a statement and another one that fits it as its counterpart

El carcinoma endometrial (CE) es el tumor infiltrante más común del tracto genital femenino y el cuarto en la mujer en países desarrollados, siendo la tipología endometrioide (CEE) la más frecuente. El 80% de las pacientes son diagnosticadas cuando el tumor sigue localizado en el útero, presentado una supervivencia a los 10 años que alcanza el 80%. Sin embargo, un número pequeño aunque relevante de estas pacientes recurren. Por ello, es de vital importancia encontrar marcadores para predecir la recidiva en esas pacientes para las cuales no se conoce un factor de riesgo suficientemente robusto. El 20% restante de las pacientes son diagnosticadas en una etapa más avanzada y presentan invasión miometrial, siendo este proceso uno de los factores de riesgo más relevantes y significativamente asociado a la recurrencia. La progresión del cáncer combina múltiples procesos que implican a moléculas de adhesión celular. Su expresión aberrante se relaciona con: pérdida de adhesión célula-célula, disgregación de las células tumorales de la lámina propia y adquisición de un fenotipo invasivo y migratorio. ALCAM, miembro de la superfamilia de las inmunoglobulinas, participa en interacciones homotípicas y heterotípicas entre células adyacentes. Su expresión se ha relacionado con la progresión de distintos tipos de cáncer, aunque su función sigue sin conocerse.
Esta tesis se centra en dos objetivos: (i) el estudio de ALCAM como marcador de recurrencia en CEE y su función en la progresión tumoral; (ii) la caracterización de los mecanismos moleculares asociados a ALCAM en el frente superficial e invasivo del tumor y en el proceso de infiltración miometrial.
Un estudio multicéntrico retrospectivo en 174 tumores primarios de CEE evidenció que ALCAM es un marcador independiente de recurrencia en los estadios tempranos de la enfermedad. Además, el silenciamiento de ALCAM in vitro e in vivo mostró una disminución en la migración celular, la invasión y una reducción significativa del tamaño tumoral primario así como de las metástasis loco-regionales. Análisis de expresión génica evidenciaron las funciones más alteradas asociadas a la inhibición de ALCAM: la motilidad, la invasión, y las funciones de ensamblaje y organización celular. FLNB, TXNRD1 y LAMC2, se validaron como genes efectores implicados en las funciones mediadas por ALCAM.
Por otro lado, se estudió la expresión de ALCAM en el frente superficial e invasivo del tumor en 116 tumores primarios de CEE, así como su relación con marcadores epiteliales y mesenquimales relevantes en el CE. Se utilizó un anticuerpo que reconocía la región extracelular de ALCAM, permitiendo la detección de la proteína no-clivada en la superficie celular. Se observó que mientras en el área superficial, ALCAM-extracelular correlaciona con el complejo E-cadherina/β-catenina, en el invasivo, esta correlación se establece con los marcadores mesenquimales COX-2, MMP-9 y ETV5. La correlación negativa significativa entre ALCAM y MMP-9 se observa en tres cohortes específicas, tumores moderadamente-pobremente diferenciados, tumores con invasión miometrial >50% y tumores ETV5-positivos, todos ellos asociados con un peor pronóstico. Además, en el frente invasivo, la negatividad de ALCAM-extracelular resultó un marcador pronóstico independiente de invasión miometrial. Junto a estos hallazgos a nivel tisular, se confirma en aspirados uterinos el papel de ALCAM-soluble como marcador de infiltración miometrial y una correlación significativa positiva de las expresiones de MMP-9 y ALCAM-soluble para estos tumores no bien diferenciados, indicando la existencia de un clivaje de la proteína. Los modelos in vitro e in vivo demostraron, mediante la recuperación de la expresión de la proteína entera, su participación en la adhesión y la migración celular colectiva, sugiriendo que la invasión tumoral requiere la existencia de un “switch” dinámico y adaptativo entre la expresión de ALCAM en la superficie celular y su clivaje por MMP-9.Endometrial carcinoma (EC) is the most common infiltrative tumour of the female genital tract and the fourth most common women cancer in developed western countries. Around 80% of patients are diagnosed when the tumour is still confined to the uterus, which is usually associated with a good prognosis. In fact, the early stages of endometrioid endometrial cancer (EEC) present a 10-year survival of 80%. However, a small but consistent number of these patients relapse. Therefore, it is of vital importance to find markers to predict recurrence for those patients for whom a sufficiently robust risk factor is still not known. The other 20% of patients, diagnosed at a more advanced stage of the disease, present myometrial invasion that is one of the most relevant risk factors, being strongly correlated with an increase in the rate of recurrence. Cancer progression combines multiple processes, which involve cell adhesion molecules. Aberrant expression of these molecules has been related to loss of cell-cell adhesion, disaggregation of tumour cells from the lamina propria, and acquisition of an invasive and migratory phenotype. ALCAM is a member of the immunoglobulin superfamily, which participates in homotypic and heterotypic interactions between adjacent cells. In the last years, ALCAM expression has been related to the tumorigenesis of many cancers, but its function still remain unclear. Under this scenario, this thesis has focused on two specific objectives: (i) The study of ALCAM as a marker of recurrence in EEC and its function in tumour progression; (ii) The characterization of the molecular mechanisms associated with ALCAM in the superficial and the invasive front of the tumour and its relation to myometrial infiltration. A retrospective multicentre study in 174 EEC primary tumours evidenced that ALCAM is an independent marker of recurrence in early stage EEC. Moreover, ALCAM knockdown in vitro and in vivo experiments showed a decreased in cell migration, cell invasion and a reduced primary tumour size and metastatic local spread. Microarray gene expression analysis of Hec1A ALCAM-depleted cells demonstrated that motility, invasiveness, cellular assembly and organization were the most associated deregulated functions. FLNB, TXNRD1 and LAMC2, were validated as downstream effector genes involved in ALCAM-mediated cell migration. In addition, we focused on the role of ALCAM in two tumour areas (superficial and invasive front) in 116 EEC primary tumours and its correlation with a set of important epithelial and mesenchymal EC markers. For that, we used an antibody that specifically recognised its extracellular region, allowing us to detect the presence of full-ALCAM (non-cleaved protein) at the cell surface. While at the superficial tumour extracellular-ALCAM was correlated with the epithelial E-Cadherin/β-catenin complex, at the invasive tumour ALCAM correlated with the mesenchymal markers COX-2, MMP-9 and ETV5 mesenchymal markers. Interestingly, the inverse correlation between ALCAM and MMP-9 was only significant in the moderately-poorly differentiated tumours, in patients presenting a myometrial invasion >50% and in the ETV5-positive tumours cohorts, all the scenarios related to poor prognosis. Moreover, at the invasive front extracellular ALCAM-negativity was an independent prognostic marker of myometrial invasion. Together with these findings at tissue level, we confirmed that in the moderately-poorly differentiated patients, soluble ALCAM detected in uterine aspirates was significantly increased in tumours presenting myometrial invasion and positively correlated to MMP-9 expression, suggesting that ALCAM decrease at the invasive front of primary tumours is caused by protein shedding. In vitro and in vivo models evidenced that ALCAM participates in cell-cell adhesion and collective cell migration, and suggest that to invade in the frontier between tissues-restricted carcinoma and disseminated tumour cells, a dynamic and adaptive switch between ALCAM expression at the cell surface and ALCAM cleavage by MMP-9 might take place.

The classical immunoglobulin family of cell adhesion molecules, known as the IgCAMs, have been characterised as having an important adhesive function at areas of cell-cell contact and from cell to extracellular matrix. Members of this family have been shown to participate in signal transduction and increasingly it seems that this could be a general feature of the IgCAM family rather than the exception. We aimed to determine whether IgCAMs that undergo retrograde axonal transport in neurons contribute to neuronal viability and co-operate with neurotrophin signalling. Activated Leukocyte Cell Adhesion Molecule (ALCAM) was identified in cells of immune system as the binding partner of the T-cell glycoprotein CD6, where it contributes to normal T-cell activation. Additionally ALCAM has been shown to have an important role in the nervous system, where it is involved in neuron pathfinding and development of the neuromuscular junction. We identified ALCAM as a protein associated with the axonal retrograde transport compartment containing neurotrophins and their receptors in primary ventral horn neurons. I found that ALCAM is bidirectionally transported in neurons, and predominantly cotransported with the neurotrophin receptor p75NTR toward the cell body. Furthermore, I found ALCAM could specifically potentiate neurotrophic signalling. ALCAM overexpression resulted in extension of longer neuritic processes in PC12 cells treated with nerve growth factor (NGF). The extracellular domain is both necessary and sufficient to potentiate NGF-induced neurite outgrowth, while the transmembrane and cytoplasmic domain have no effect. ALCAM overexpression only potentiated specific growth factor stimuli but had no effect on other pathways leading to differentiation or on undifferentiated PC12 cell morphology. In conclusion, ALCAM appears to synergise with NGF signalling to induce neuronal differentiation and may have a role in the nervous system not only as an adhesion molecule but also in neurotrophin signalling.

Nesta tese é realizada a análise dos discursos da mídia face às políticas e ações orientadas pela ideologia desenvolvimentista, além da análise de documentos particulares relativos ao processo de implantação da ALCOA MINERAÇÃO S.A. A pesquisa teve como objetivos: descrever os percursos e discursos sobre o processo de industrialização e desenvolvimento do Brasil a partir dos anos 1950, examinando os impactos à luz do contexto da Região Amazônica; relacionar o quadro da economia maranhense em 1980 com o projeto de implantação da ALCOA MINERAÇÃO S.A.; examinar os discursos veiculados pela mídia impressa maranhense no sentido de informar a população de São Luís sobre os impactos ambientais quando da implantação da ALCOA MINERAÇÃO S.A; identificar as ações e reações da ALCOA MINERAÇÃO S.A. em resposta ao discurso da mídia impressa durante a implantação da fábrica de alumina/alumínio. Resumo: Para atingir tal fim, apresenta-se um conjunto histórico-teórico sobre a ideologia desenvolvimentista via processo de industrialização dos Governos Getúlio Vargas, Juscelino Kubitschek, e a estratégia de ocupação da Amazônia durante o regime militar. O Governo Getúlio Vargas defendia um projeto de desenvolvimento nacional autônomo. A retórica governista era a de construção de uma nação desenvolvida. O Governo Juscelino Kubitschek buscava o desenvolvimento pela atração e estímulo aos investimentos estrangeiros em setores produtivos. Já o Governo Militar intensificou a ocupação da Amazônia com a implantação de planos de desenvolvimento regional. Mostra-se que a implantação do projeto ALCOA MINERAÇÃO S.A. desencadeou impactos na economia, no âmbito social e ambiental maranhense O modelo de pesquisa usado é o qualitativo/interpretativo, passando também pela pesquisa exploratória, descritiva, bibliográfica e documental. O suporte documental está respaldado na mídia impressa local, no período da implantação da ALCOA MINERAÇÃO S.A. nos anos 1980 a 1984. A técnica adotada para análise e interpretação dos resultados é a análise do discurso, tendo como suporte a escola francesa. Na análise do discurso da mídia impressa foi possível verificar que ela age em nível da ideologia e que seus discursos têm origem em vários lugares de enunciação: governo, políticos e atores sociais diversos.
In this research it is done the analysis of the discourse of the media faced to the politics and actions oriented by the development ideology and also the private documents in the process of implantation of ALCOA MINERAÇÃO S.A. This research aimed on describing the routes and discourses about the process of industrializing and development of Brazil from the year of 1950, examining the impacts toward the Amazon region context; relate the economy scenario of Maranhão State in1980 with implantation of the ALCOA MINERAÇÃO S.A Project; examine the discourses of the press media from Maranhão when informing the population of São Luís about the environmental impacts of the implantation of the ALCOA MINERAÇÃO S.A Project.; identify the actions and reactions of ALCOA MINERAÇÃO S.A Project about the discourses of the press media during the implantation of the alumini and aluminium plant. To guarantee this aim, it is presented a set of theories about the development ideology, via the process of industrialization of the government of Getulio Vargas and Juscelino Kubitscheck and the strategy of occupation of the Amazon Rain Forest during the military government. The government of Getulio Vargas defended a project of autonomous national development. The governmental rhetoric was the construction of a developed nation. The government of Juscelino Kubitschek aimed on the development through the attraction and stimulus of foreign investments in productive sectors. The Military Government though, intensified the occupation of Amazon Rain Forest, through the implantation of regional development plans. It is shown the impacts of the implantation of the ALCOA MINERAÇÃO S.A Project on the economy, on the social and on the environmental aspects in the State of Maranhão. The model of the research used is quality and interpretation based, also going through the exploring, descriptive, bibliographic and documental research. The documental background is based on the local press media in the period of implantation of the ALCOA MINERAÇÃO S.A Project, from the years of 1980 to 1984. The techniques used to analyze and interpret the results were the analysis of the discourse underlined by the French School. Along the analysis of the discourse of the press media, it was possible to verify its action on an ideological basis and that its discourses, originally come from several enunciations: govern, politics and other civil actors.

Previous issue date: 2004-12-22The evolution of production organization models was observed with more intensity since the second half of the last century. Its purpose is to make organizations more competitive. A core element that accompanies this change is a performance measurement system with a more multidimensional framework in most organizations. The actors in this scenario use the information from these performance measurement systems for issues of planning, control and improvement. In this context, the relationship between performance measures has been providing information to each of these actors. The mental models of these actors are unique. Usually the formal model of relationship between the performance measures in use is not shared. The goal of this work is to research this problem and to evaluate how the sharing process occurs for the use of maps of relationships between performance measures. The main contribution is the creation and sharing of a model of relationships between performance measures among users of a performance measurement system. The evaluation of this sharing process took place for the application of some research tools in a quasi-experimental project.
A evolução dos modelos de organização da produção percebida com mais intensidade a partir da segunda metade do século passado acontece no sentido de tornar as organizações cada vez mais competitivas. Um elemento central que acompanha essa mudança é o sistema de medição de desempenho, que se torna cada vez mais multidimensional, em várias dessas organizações. Os atores desse cenário utilizam as informações geradas por esses sistemas de medição de desempenho para fins de planejamento, controle e melhoria. O relacionamento entre medidas de desempenho está inserido nesse contexto provendo informações a cada um dos atores envolvidos. Os modelos mentais desses atores são individuais, pois, normalmente não há um compartilhamento de um modelo formal de relacionamento entre as medidas de desempenho em uso. Esta dissertação propõe pesquisar sobre esse problema e avaliar como esse compartilhamento acontece pelo uso de mapas de relacionamentos entre medidas de desempenho. A contribuição principal está na criação e no compartilhamento de um modelo de relacionamento entre medidas de desempenho em conjunto com usuários de um sistema de medição de desempenho. A avaliação desse compartilhamento aconteceu pela aplicação de alguns instrumentos de pesquisa no projeto de um quasi-experimento.
