Abstract:
The aim of the work described in this thesis was to explore the synthesis of glycosidase inhibitors and to evaluate their potential as anti-cancer agents. Glycosidases catalyze the fission of glycosidic bonds and are involved in vital biological functions. With regard to their potential for anti-cancer therapy, two glycosidases were identified: α-N-acetyl-galactosaminidase and β-N-acetyl-hexosaminidase. The former has been implicated in causing immunosuppression in advanced cancer patients by negating the effect of the macrophage activating factor (MAF), while the latter is secreted by invadin

Critical to the recognition of glycosylceramides by NKT cells are antigen presenting cells (APC), such as dendritic cells, which are responsible for binding, processing, and delivery of ligands to these lymphocytes. α-Retaining glycosidases are a class of hydrolytic enzymes that cleave α-glycosidic linkages with net retention of configuration at the anomeric center. The finer details of the mechanisms of two such enzymes, Golgi α-mannosidase II and human pancreatic α-amylase, were investigated in order to gain better understanding of this class of enzymes. α-Retaining mannosidases, like all other retaining glycosidases, employ a double displacement mechanism in which a transient glycosyl-enzyme intermediate is formed. The human gut is home to a significant number of microorganisms including the dominant symbiont Bacteroides thetaiotaomicron. This microbe is predicted to possess an array of glycoside hydrolases, majority of which are involved in starch utilization. Presented here is a comparative functional analysis of two alpha-glucosidases, Family 31 Glycoside Hydrolases from Bacteroides thetaiotaomicron. Enzymatic kinetics revealed these enzymes both preferentially cleave alpha-(1,6) linkage in comparison to the predicted alpha-(1,4) and favour maltose derived substrates of longer length.

The central dogma of molecular biology outlines the process of information transfer from a DNA sequence, to a protein chain. Beyond the step of protein synthesis, there are a variety of post-translational modifications that can take place, one of which is addition of carbohydrate chains to nascent proteins, known as glycosylation. The N-linked glycosylation pathway is responsible for the covalent attachment of multifunctional carbohydrate chains on asparagine residues of nascent proteins at Asn-X-Ser/Thr consensus sequences.
