Abstract:
Eight cyclic analogues of Substance P were made in order to investigate the conformation of the C-terminal end of the peptide. These analogues were designed to test three literature models describing the active conformation of substance P. Although the potencies of the analogues were low (in the micromolar range), our results support Cotrait's and Hospital's model (1986). Several substance P antagonists were synthesized. These compounds did not demonstrate agonistic activity nor anatagonistic activity. The tryptophan side chain is contributing to the antagonistic activity of these analogues, a

Abstract:
Small molecules, such as oligopeptides can interact with nucleic acid targets and subsequently stimulate or supress biological functions. The purpose of this thesis was to produce nucleic acid targeting oligopeptides which contain non-natural amino acids as part of their sequence. Certain amino acids display selectivity for certain nucleobases. Arginine has a high propensity for being present at the binding interface of both protein-DNA and protein-RNA complexes. This thesis was aimed at the synthesis of two arginine analogues in which the guanidino side chain was locked in a bicyclic framewor

Abstract:
This thesis describes the incorporation of heterocycles as part of an α-amino acid side-chain. Two methods were adopted. The first involved the alkylation of a pseudoephedrine glycinamide enolate using haloalkanes. The method was validated by making the naturally occurring amino acid phenylalanine. Subsequently naphthyl-, pyridyl-, biphenyl- and isoxazolyl- groups were successfully incorporated into the amino acid side chain. The enantiomeric excess of the product amino acids was found to be moderate (10-87%) due to poor diastereoselectivity in the key alkylation step. Moreover, the strict exp

This thesis is divided into two parts: Part One describes synthetic approaches to the naturally occurring antibiotic Lactivicin (1) and its acylamino derivatives, involving the condensation of protected L-cycloserine with 2-oxoglutaric acid. The phenoxyacetyl-amino derivative (2) exhibited antibacterial activity. A method was developed for the selective deprotonation of the endocyclic amide of the protected cycloserine; the resulting anion was then quenched with various electrophiles. In this manner, the Lactivicin analogues (3) and (4) were prepared. Whilst (3) was biologically inactive, (4)

This thesis is concerned with the development of new synthetic routes for the asymmetric syntheses of a range of β- and γ-amino acids. Chapter 1 introduces the various biological activities displayed by cyclic β-amino acid containing compounds together with their occurrence in pharmaceutical molecules and β-peptides. Some of the most commonly used synthetic strategies for the preparation of carbocyclic β-amino acids are briefly described, with the focus on the formation and functionalisation of a five-membered carbocyclic ring. The thermal and metal-catalysed aza-Claisen rearrangement of allylic trichlororacetimidates has found widespread application in the synthesis of nitrogen-containing molecules including alkaloids, antibiotics and unnatural amino acids. We have recently investigated the use of this reaction for the rearrangement of chiral molecules. This has led to the development of a highly diastereoselective, ether-directed palladium(II)-catalysed process which has been used for the synthesis of β-hydroxy-α-amino acids. To expand the scope and understanding of the rearrangement, a number of analogues of 1 hav
