Abstract:
Background:Tofacitinib is an oral JAK inhibitor that is being investigated for juvenile idiopathic arthritis (JIA).Objectives:To describe tofacitinib pharmacokinetics (PK) in patients with JIA, identify potential covariates accounting for variability in exposure, assess the formulation effect of oral solution vs tablet and propose a simplified dosing regimen.Methods:This was a pooled analysis of data from 3 tofacitinib clinical studies in patients with JIA aged 2−<18 years: a Phase 1, open-label (OL), non-randomised study (NCT01513902); a Phase 3, randomised, double-blind, placebo-controlled, withdrawal study (NCT02592434); and an OL long-term extension study (NCT01500551). Tofacitinib was dosed at 5 mg twice daily (BID) in patients ≥40 kg or at body weight (BW)-based lower doses BID in patients <40 kg, to achieve average concentrations (Cavg) comparable with those in patients receiving 5 mg BID. A sparse PK sampling scheme was applied, and the plasma samples were assayed using a validated, sensitive and specific high-performance liquid chromatography tandem mass spectrometric method (lower limit of quantification = 0.100 ng/mL). A nonlinear mixed-effects modelling approach was used for the population PK model, and population parameter variability was assumed to be log-normally distributed. Covariates relating to patient demographics, disease characteristics, concomitant medications and formulation (oral solution vs tablet) were selected using a stepwise covariate modelling approach, and parameter-covariate relationships were evaluated using stepwise forward-inclusion (p<0.05) backward-deletion (p<0.001) procedures. The effect of time-varying BW on oral clearance (CL/F) and apparent volume of distribution (V/F) was characterised using an allometric model. Final model quality was assessed by Visual Predictive Checks (VPCs).Results:Of 246 patients in the analysis, 74.0% were female; 87.8% were white, 2% were black, 10.2% were ‘other’ races and no patients were Asian. Median (range) BW was 46.3 (11.1−121.8) kg. Initially, 100 patients received oral solution and 146 patients received tablets; 11 patients switched formulations during the studies. A one compartment disposition model with first-order absorption and a lag time sufficiently described the data. Final estimates for CL/F, V/F and the first-order absorption rate constant (ka) for tablets were 26.1 L/hr, 89.2 L and 2.78 hr-1, respectively. The only statistically significant covariate was a formulation effect on ka. All parameters were estimated adequately. Estimated allometric exponents were 0.310 for CL/F and 0.537 for V/F. Absorption was described with an estimated lag time of 0.186 hr, and the oral solution had a 1.64-fold faster absorption rate vs the tablet. VPCs sufficiently described the observed data over time, across BWs and ages. Given the PK characterisation and variability in patients with JIA, a simplified dosing scheme was proposed, targeting Cavgvalues equivalent to those in patients receiving 5 mg BID: 3.2 mg BID solution in patients 10−<20 kg; 4 mg BID solution in patients 20−<40 kg; and 5 mg BID tablet or solution in patients ≥40 kg.Conclusion:Tofacitinib population PK in patients with JIA were adequately described by a one compartment model parameterised in terms of CL/F, V/F and first-order absorption with a lag time. Drug absorption from the oral solution was faster than from the tablet. Tofacitinib does not require dose modification or restrictions for any covariates, except BW, to account for differences in Cavg. Abstract:
Using the Drucker-Prager elastoplastic constitutive model to describe the nonlinear deformation behavior of soil and simulating step loading on static test piles by ANSYS model which only consider linear elastic compression of pile body. By comparing ANASYS simulation value and the measured values of pile’s axial force and side friction, it shows that the greater the load ratio which transfer through the bottom of the pile, the pile-soil stiffness ratio increased; when vertical load is small, analysis results of elastic theory are fairly accurate. ANASYS simulation results have high accuracy when vertical load is bigger.

The calculation of truss deformation under static loading is studied in this paper. First, an experiment was conducted to get the truss deformation for calibration. Second, the deformation was calculated by the principle of virtual work of structural mechanics and by Ansys. The calculated results agree well with the test result. Finally, some suggestions are presented to the calculation of more complicated truss structures.

Based on WN gear characteristics and considering system error, a multi-degree-freedom (Tangential-Radial-Axial) dynamics analysis model after coupling friction was established. In this article, we established the three-dimensional solid model by using PROE and then imported WN gear’ three-dimensional model into Ansys software through the data interface of Ansys software and PROE software and conducted a three-dimensional simulation anasys of the impact of dynamic contact. By applying load, the stress analysis of WN gear was conducted and the WN gear’s the effective stress clouds was gotten. Meanwhile, basing on ANSYS / LS-DYNA, it established the rigid-flexible body of gear dynamic contact model and analyzed the dynamic simulation anasys of WN gear. The results demonstrated that the tangential and axial vibration of double arc gear was significantly greater than the radial vibration.

Abstract:
Greenhouse and field studies were conducted to determine the effects of shade on photosynthetic, growth and yield responses of "Redchief" strawberries. Plants were exposed to 0 , 30 , 47 and 63 percent shade in the greenhouse. Net photosynthesis (Pn) as measured under the shade treatments demonstrated a curvilinear decrease. Pn as measured under saturating light levels of 790 μ-tmol· m-2 · s·1 began to decrease in plants grown under 30 percent or higher shade levels. Light saturation curves for leaves expanded in full sun and then transferred to shade treatments for seven days showed a decrease in Pn saturation rate and light saturation point at 63 percent shade. Saturation curves for leaves expanded under the shade treatments exhibited a decreasing trend in saturation rate and point at all shade levels. Heavy shade (63%) reduced leaf thickness and depth of the palisade cell layers. Individual leaf area was not affected by shade treatments. As percent shade increased, total plant dry weight decreased linearly as did specific leaf weight. Shade treatments of 0, 30, 63 or 95 percent were applied over field-grown plants either in the fall during flower bud initiation, or in the fall and spring, during flower bud initiation and spring vegetative growth. Berry number was reduced as percent shade increased. Plants shaded only in the fall had higher berry number than plants shaded both in the fall and spring. Berry weight was also reduced as percent shade increased. Sugar concentration, pH and titratable acidity were not affected by shade levels or time of shade application.

O mundo contemporâneo exige agilidade do ser humano na vida cotidiana. Pensar em formas de minimizar o tempo de preparo de alimentos, sem deixar de ter uma alimentação saudável, tem sido a preocupação das pessoas que têm trocado o consumo de frutas e verduras íntegras pelas minimamente processadas. Entre estas se encontra o morango. O morango é um fruto muito valorizado, no entanto é muito delicado, altamente perecível e de alto custo. Destacam-se suas propriedades nutracêuticas, sua ação antioxidante e capacidade de aumentar as defesas imunológicas. Os compostos fenólicos são apontados como principais substâncias que participam destas atividades. Dentre os revestimentos utilizados, aqueles a base de xantana tem se destacado por originar revestimentos com alto grau de proteção. Este estudo tem como objetivo avaliar diversos revestimentos comestíveis a base de goma xantana, gelatina, sorbitol e óleo de canola em morango minimamente processado, em diferentes tempos de armazenamento, como forma de amenizar a perda dos compostos antioxidantes e proporcionar o aumento da vida de prateleira. O experimento foi realizado no período de 15 dias, as amostras foram retiradas para as análises após o 10, 50, 10o e 150 dias de armazenamento. Ao final de cada período foram realizadas as análises de perda de peso, cor, textura, pH, acidez, sólidos solúveis, umidade e sólidos totais. Parte do material relativo a cada período de armazenamento foi triturado e armazenado a – 80 0C, para realização das análises de ácido L- Ascórbico, antocianinas, compostos fenólicos e atividade antioxidante. A conservação das propriedades físico-químicas do morango foi ampliada com o revestimento de gelatina, xantana, sorbitol e óleo de canola a 3% (GXSL 3%) para os parâmetros sólidos solúveis totais e firmeza. Na conservação dos compostos bioativos, a cobertura com xantana, sorbitol e óleo de canola a 3% (XSL 3%) demonstrou ser o melhor tratamento para impedir que o percentual de inibição do DPPH diminuísse. Entretanto, não foram verificados resultados estatisticamente significativos de proteção para os outros fitoquímicos.
The contemporary world requires agility of the human being in everyday life. Think of ways to minimize the preparation time of food, while having a healthy diet, has been the concern of people who have changed the consumption of fruits and vegetables by intact minimally processed. Among these is the strawberry. The strawberry is a highly prized fruit, however is very delicate, highly perishable and expensive. Noteworthy are their nutraceutical properties, its antioxidant action and the ability to increase your immunological defenses. Phenolic compounds are cited as the main substances that participate in these activities. Among the coatings used, those based xanthan has been noted for yield coatings with a high degree of protection. This study aims to evaluate the various edible base xanthan gum, gelatin, sorbitol and canola oil in strawberry minimally processed at different storage times coatings as a way to mitigate the loss of antioxidants and provide increased shelf life. The experiment was performed at 15 days, samples were taken for analysis after 1, 5, 10 and 15 days of storage. At the end of each period analyzes of weight loss, color, texture, pH, acidity, soluble solids, moisture and total solids were performed. Some of the material for each storage period was crushed and stored at - 80 0C, for analyzes of L- ascorbic acid, anthocyanins, phenolic compounds and antioxidant activity. The conservation of the physicochemical properties of the strawberry was expanded with the coating of gelatin, xanthan gum, sorbitol and canola oil 3% ( GXSL 3 % ) for total soluble solids and firmness parameters. Conservation of bioactive compounds, cover with xanthan, sorbitol and canola oil 3% ( XSL 3 % ) proved to be the best treatment to prevent the inhibition of DPPH decreased. However, no statistically significant results for the other protective phytochemicals have been checked.

Abstract:
A quick opening closure using a new rubber sealing ring was proposed in this paper, which can be used in natural gas transmission filters. An innovative saddle-shaped sealing ring with the feature of floatability was designed for substituting of traditional rubber C-ring. A lager gap could be allowed between the door and the top flange which made the assembling and disassembling easier than before. The theory of strength calculation and check for the whole steel structure were also presented. For an actual engineering case, a device with internal diameter of DN1550 (62inches) and design pressure of 12.6MPa (1827.5psi) was designed by rule according to Chinese national standard GB150 and analyzed by FEA (ANASYS). In addition, the sealing performance was simulated by FEM analysis (ABAQUS). Finally, the prototype was produced and the experiments for its sealing and load-bearing performance using hydrostatic test and stress test were carried out successfully.

Abstract:
The runback water flow and heat transfer on the surface of aircraft components has an important influence on the design of anti-icing system. The aim of this paper is to investigate the water flow characteristics on anti-icing surface using numerical method. The runback water flow on the anti-icing surface, which is caused by the impinging supercooled droplets from the clouds, is driven by the aerodynamic shear forces and the pressure gradient around the components. This is a complex model of flow and heat transfer that considers flow field, super-cooled droplets impingement and runback water flow simultaneously. In this case of gas-liquid two phase flow, the Volume-of-Fluid (VOF) method is very suitable for the solution of thin liquid film flow so that it is applied to simulate the runback water flow on anti-icing surfaces in this paper. Meanwhile, the heat and mass transfer of the runback water flow are considered in the calculation using the User-Defined Functions (UDFs) in ANASYS FLUENT. The verification is conducted by the comparison with the results of the experimental measurement and the mathematical model calculation. The effect of the airflow velocity and contact angle on the water flow are also considered in the numerical simulation.

The displacements of nuclear island buildings occur because of earthquake, thermal expansion, creep shrinkage etc. Because there are penetrations and some supports that connect to the wall of reactor building, the displacement loads must be considered when the piping and the supports are verified, the displacement loads have great effect on the piping and supports stress analysis. Studying the effect and looking for that how to reduce this effect are helpful to find a way to reduce the piping stress and the loads on the supports, and it is also helpful to make the plant safe. In this paper some piping calculation units of EPR project are showed as examples, the calculation units are done with a new software ANASYS PIPE, and finite element method is used. there are penetrations and supports that connects to the wall of reactor building in the calculation units, the building displacements due to earthquake, thermal expansion and creep shrinkage are considered, and the effect of displacement loads and the combinations of all kinds of loads under different category conditions on the piping and supports are studied, the result shows that the stress of piping and the loads on the supports caused by building displacement loads are prominent, but the piping and the supports can meet the requirement by optimizing the support concept.

Abstract:
Axial fans are widely used in modern industry and new regulations and stringent environmental concerns are prompting manufacturer to design efficient low-noise axial fans. This paper is focused on improving the aerodynamic performances and reducing the tonal noise at BPF and its harmonics by the optimum choice of lean-swept blade and the stacking line for the low-speed axial fan. The aerodynamic characteristics of the axial fan with a shroud are explored by CFD with ANASYS CFX. A hybrid method, SST turbulence model for flow and FW-H equation for acoustics, is chosen to predict the radiated noise. The accuracy and reliability of predicted aerodynamic and aeroacoustics results are verified by comparing both computation and experimental data. A number of modified blades with different leaned angle, swept angle and the stacking lines are modeled and analyzed, and the investigation into the optimum choice of lean-swept blade and the stacking line is conducted according to aerodynamic performances and tonal noise. Q-criterion which can visualize the major flow disturbances is applied for the purpose of identification of acoustic sources. The turbulent flow structures on the leading edge, tip and suction side of the blade are main noise sources. An optimal modification is determined through the analysis of the aerodynamic performances and noise, which is to achieve the desired performances by blade sweep and lean and adjusting the stacking line. The results show that aerodynamic and acoustic performances of the optimized fan are better than that of the original fan and the improvement is more obvious to change the stacking line with centre of gravity compare to blade sweep and lean for the low-speed axial fan.
