Abstract:
The role for protein dynamics in the transition states (TS) of enzyme reactions has been debated over decades. Dihydrofolate reductase (DHFR) catalyses the NADPH-dependent reduction of dihydrofolate to tetrahydrofolate and has long been considered a paradigm of enzymology. Numerous studies on DHFR have provided strong evidence that there is no coupling between the long-range motions on nanosecond to millisecond timescales to the chemical coordinate. However, the role of femtosecond (fs) bond vibrations in the TS has not been fully investigated. This investigation focused on understanding how these fast protein vibrations affect enzyme catalysis. A thermophilic DHFR from Geobacillus stearothermophilus (BsDHFR) was investigated kinetically by complete enzyme isotope substitution. Our studies indicated that, whilst protein vibrations do couple to the reaction coordinate, they do not affect the height or width of barrier crossing. Instead, dynamic coupling enhances the frequency of dynamic recrossing. In line with the other DHFR investigations by enzyme isotope substitution, efficient enzymes tend to reduce dynamic coupling as a mean to maximise enzyme catalytic efficiency. The transition state in DHFR was also characterised by α-secondary hydrogen and heavy atom kinetic isotope effects (KIEs). Secondary KIEs were measured for DHFR isolated from Escherichia coli, Moritella profunda and G. stearothermophilus. The high resemblance in the magnitude and temperature dependence of the measured α-secondary KIE implied that the reaction ready configuration is essentially the same among these enzymes. Carbon isotope effect measurements were measured for EcDHFR. The reacting carbon in NADPH showed a profound isotope effect at low temperature. Further analysis by measuring hydride KIE indicated the hypothetical "promoting" motion is unlikely to act via the C4 atom of NADPH. The carbon isotope effect likely reports on the recrossing events or the reorganisation effect that occur along the transition state dividing surface.

Abstract:
An intermediate temperature solid oxide fuel cell (SOFC) is developed and its performance is investigated experimentally and theoretically. In the experimental program, a gadolinium doped ceria based membrane electrode group is developed with the tape casting and screen printing methodology and characterized. An experimental setup is devised for the performance measurement of SOFCs and the performance of produced cells is investigated over a range of parameters including the electrolyte thickness, the sintering temperature, the operation temperature etc. The experimental setup is then further modified to measure the temperature distribution in the large SOFC single cells. The effects of operating parameters on the temperature distribution are investigated and the parameter spaces leading high efficiency without cracking the ceramic membrane are identified. In theoretical study a mathematical model is developed to represent the fluid flow, the heat transfer, the species transport and the electrochemical reaction in intermediate temperature of solid oxide fuel cells.The differential equations are solved numerically with a commercial CFD code which employs a control volume based approach. The temperature distribution and species distribution during theSOFC operation is analyzed. The effects of operation parameters on critical SOFC characteristics and the performance are numerically investigated over a range of parameter space. The experimental and numerical results are compared to validate the mathematical model. The mathematical model is found to agree reasonable with experimental data.

Foster youth exiting the child welfare system will have poorer outcomes such as, low educational attainment, poor employment, high criminal activity, homelessness, and higher rate of public assistance when compared to non-foster care youth. The purpose of this thesis is to look at the values, theoretical goals, implications, and the outcomes of services outlined in the California Fostering Connections to Success Act of 2010, or Assembly Bill (A.B.) 12. A. B. 12 was designed to help foster care youth who reached the age of majority (18+) and extend their foster care services to age 21. By analyzing A.B. 12, this author will take a take a look at what the lawmakers intended, the results of the last five years, and what we can learn for future legislation.

The following document describes research and development of a cross layer design of two complementary topics: Reliable Multicast over satellite cross-layer design architecture The first topic investigates the capabilities of existing reliable multicast protocols and identifies the drawbacks using a variety of numerical and emulation models. Within this research topic a cross layer approach to designing multicast protocol is investigated and then compared with the existing techniques to show higher performance. Cross-layer security Framework The second topic investigates how network layer security can be provided in a cross layer architecture. This has led to the development of a new framework called Cross Layer IPsec (CL-IPsec). The performance of the new framework is compared with IPsec using emulation and numerical methods in this research topic and evidence of higher performance using CL-IPsec is shown. The two main parts are followed by conclusion and appendix sections. The important points from each chapter are summarised in the conclusion section; which is followed by a future work section that identifies further work that needs to be done in this field. The appendix contains list of publications that were generated as part of this research and schematic diagrams of CL-IPsec functions.

Stemming from the perception of trust and communication in a remote work environment, the experiences of the employees were analyzed based on three factors of self-efficacy, personal engagement, and interpersonal relationships with their leaders. Using Interpretative Phenomenological Analysis (IPA), the study employed semi-structured interviews to gain experiential data from nine qualified remote employee participants who had worked remotely one day per week for a minimum of one year. The data were coded and the four themes that surfaced were: (a) the desire to succeed is key to the feeling of mastery in a remote role, (b) performance in a remote role is the result of feeling independent, support from others, and productivity due to less distractions, (c) communication in a remote work environment is vital for justification of role and understanding perceived distances in relationship development, and (d) trust is an essential element in the perception of relationships in a remote work environment. Findings of this study concluded trust and communication are vital components of the perception of self-efficacy, personal engagement, and interpersonal relationships. This study contributed to understanding the psychological conditions necessary to be successful in a remote work environment for both academic and practical knowledge of remote employment.

Abstract:
Public school teachers have, with few exceptions, invested four or more years in higher education to prepare themselves for the teaching profession. These four years of college education can be viewed as an investment when one realizes that the teacher had to pay for that education and could have otherwise been earning an income during that time. Some teachers make an additional investment in education by earning a master's degree or a doctorate. This study looked at the practice of teachers in Virginia who decide to make the additional investment in graduate education at selected Virginia institutions of higher education. This study used econometric methods to analyze this investment in graduate education. Social benefits and costs of education were not included in the study. Private costs included both direct and indirect acquisition costs. The salary supplements paid by school divisions to teachers who hold an advanced degree were used as the private benefits. Non-pecuniary benefits were not included in the study. Net present valuation, discounted benefits and costs, benefit-cost ratio, and internal rate of return calculations were made. Data from similar studies done in other areas of graduate study and in other areas of the nation and world were reviewed. The purpose of the study was to review the practice of all school divisions in Virginia which provide a salary supplement to teachers who hold a master's degree or a doctorate, and also to analyze the costs involved in the acquisition of such degrees. The cost-benefit analysis of the teachers' investment in graduate education provides information which can be used by teachers who are considering such an investment. The analysis can also be used by those school divisions which are currently spending considerable amounts of money for such salary supplements. The cost-benefit analysis of acquiring a master's degree with a subsequent change from the classroom to an administrative position has implications for school divisions and state level policy makers.

Abstract:
Varicose veins are a type of vascular disease that usually appears in the legs. The failure of the bicuspid valves within the vein means it is unable to function properly to ensure a one-way flow for the blood. This causes problems as the vein wall is not as muscular as those in arteries. This results in the vein wall expanding and swelling due to the reverse flow of the blood. Besides cosmetic issues, patients with varicose veins experience symptoms such as pain and discomfort in the legs, fatigue, itchiness, and swelling of the skin. Various therapies can treat such symptoms, but the current study is to focus on foam sclerotherapy. The foam, consisting of sclerosant liquid and room air bubbles, is injected into the vein. The sclerosant liquid is of similar chemical structure to detergent. The purpose of the foam is to allow its sclerosant liquid content to be in direct contact with the varicose vein wall endothelial cells, and to damage them. The vein wall will collapse due to this damage and the blood is then redirected to other healthy veins and back to the heart. Basic fluid properties data is not available in the literature for the sclerosant foam and hence the study begins with Rheometry and Tensiometry experiments. The bubble size distribution within the bulk foam using microscopy and image analysis is then obtained. An experimental rig is built to resemble the Sapherofemoral Junction (SFJ). The rig is initially filled up with Normal Saline (NaCl 0.9% w/v in H2O) and the sclerosant foam is generated and injected into it to determine the spreading characteristics of the foam. A Computational Fluid Dynamics (CFD) model is also developed to predict the foam behaviour. The CFD model is coupled with a Population Balance Model (PBM) to simulate the distribution of bubble sizes within the vein, which is one of the primary concerns of the Phlebologist (medical specialist treating varicose veins) when performing treatment on the patients as large bubbles are not desirable.

Federal Election Commission decision from the time of the decision, January 21, 2010 until the mid-term elections November 2, 2010. The broadcast transcripts were read for emergent frames to see how the stations framed coverage. The cable channels had the most coverage. MSNBC, ABC, NBC, and CBS framed the decision negatively, Fox News portrayed it positively, and CNN was neutral to negative in coverage.

Abstract:
This mixed methods research is an investigation into the quality of interaction in the provision of Early Childhood Education (ECE) in Ghana. Literature suggests that a reasonable number of ECE teachers from developing countries, including Ghana, lack training and content knowledge in ECE. In addition, there is a general lack of Teaching and Learning Materials (TLMs), play equipment and ECE facilities. Constructivism, a theoretical position which postulates that children create knowledge through their interaction with the environment in which they find themselves, was adopted as the theoretical framework for this study. Data in the form of interviews, observation and questionnaires were collected from 131 participants. These comprised 104 teachers and 23 headteachers, selected from 24 schools in the Greater Accra region of Ghana. Also, four ECE coordinators were interviewed as part of the data collection. The findings from this study suggest that a large proportion of teachers in the study were not appropriately trained. There was a variation with teachers in the remote areas most underequipped. The pedagogical strategies the sampled teachers adopted were found to be what could be described as teacher-dominated which potentially hindered the quality of the interaction with their learners and in their lesson delivery. Based on these findings, it is suggested that teachers endeavour to train in ECE methodologies and also re-orient their pedagogical practices to align with child-centred approaches. This may enable them to interact appropriately and involve their learners in their lesson delivery. Additionally, given the circumstances (e.g. limited TLMs and play items) some of the teachers find themselves in their schools, it is recommended that teachers should encourage their learners to effectively use what the natural environment would afford them.

Abstract:
Image compression aims to reduce the amount of data that is stored or transmitted for images. One technique that may be used to this end is vector quantization. Vectors may be used to represent images. Vector quantization reduces the number of vectors required for an image by representing a cluster of similar vectors by one typical vector that is part of a set of vectors referred to as the code book. For compression, for each image vector, only the closest codebook vector is stored or transmitted. For reconstruction, the image vectors are again replaced by the the closest codebook vectors. Hence vector quantization is a lossy compression technique and the quality of the reconstructed image depends strongly on the quality of the codebook. The design of the codebook is therefore an important part of the process. In this thesis we examine three clustering algorithms which can be used for codebook design in image compression: c-means (CM), fuzzy c-means (FCM) and learning vector quantization (LVQ). We give a description of these algorithms and their application to codebook design. Edges are an important part of the visual information contained in an image. It is essential therefore to use codebooks which allow an accurate representation of the edges. One of the shortcomings of using vector quantization is poor edge representation. We therefore carry out experiments using these algorithms to compare their edge preserving qualities. We also investigate the combination of these algorithms with classified vector quantization (CVQ) and the replication method (RM). Both these methods have been suggested as methods for improving edge representation. We use a cross validation approach to estimate the mean squared error to measure the performance of each of the algorithms and the edge preserving methods. The results reflect that the edges are less accurately represented than the non - edge areas when using CM, FCM and LVQ. The advantage of using CVQ is that the time taken for code book design is reduced particularly for CM and FCM. RM is found to be effective where the codebook is trained using a set that has larger proportions of edges than the test set.

Abstract:

Instagram (IG) is a social media application that allows users to upload personal images in a public forum by including a searchable link (e.g. hashtag) along with a posted image. An IG community devoted to a pro-recovery (i.e., #anarecovery) perspective on eating disorders (EDs) stands alongside the pro-ana lifestyle IG community (i.e., #ana). Research has yet to examine the nature of visual representations associated with pro-ana and pro-recovery social media imagery and moreover, not much empirical attention has been given to the nature of this content on IG. Thus, the first aim of this qualitative content analysis was to examine how the underlying theoretical constructs of objectified body consciousness (OBC) and positive body image (PBI) were represented among #ana and #anarecovery content. A second aim was to compare the frequency of the themes present for #ana versus #anarecovery images. A coding guide was developed and high levels of interrater reliability were established for the primary codes (kappa ≥ .80). One hundred and fifty IG images each from #ana and #anarecovery were systematically coded. Within- and between-hashtag comparisons of images were performed using frequency counts and chi-square analyses. Within #ana images, the theme of body shame (62.7%) was more frequently depicted relative to body surveillance (22%) and appearance control (22%) themes. Within #anarecovery images, the theme of body protection was most prevalent (67.3%) followed by body functionality (38.7%) and body acceptance (11.3%) themes. All three OBC themes were more likely to be present in #ana images. Conversely, PBI themes of body protection and body functionality were more frequently represented in the #anarecovery images. The frequency of body acceptance themes did not differentiate the images sampled from both hashtags. This research calls for more expanded and distinguished definitions of the constructs that comprise the continuum of PBI. Findings also suggest how the lack of distinction in the frequency of body acceptance observed between the two hashtags may be clinically relevant for exploring potential risk factors that persist in the process of ED recovery.

National Health Service (NHS) statistics show that 4000 people in the United Kingdom are receiving help for gender concerns (DH, 2008). Research has highlighted the importance of romantic relationships during early adulthood and an association with mental health (Fincham and Cui, 2011). The experience of romantic relationships among transgender people in ‘emerging adulthood’ (Arnett, 2000, 2006) is an under-researched area. The decision making process concerning, and construal of, romantic relationships among this group have yet to be investigated using Personal Construct Theory (PCT). This study uses qualitative and quantitative methods of inquiry from PCT to investigate the experiences of transgender people encountering romantic relationships. Overarching themes emerging from the participants in this study were of identity validation through romantic relationships, having a bi-gendered lens of the world, facilitating greater understanding in society, and learning from past relationships. Participants’ experiences are analysed individually, as well as as a group. Clinical significance of this, limitations and future research are also discussed.

ENGLISH ABSTRACT: Although a numerical model of the mirror support truss of the Southern African Large Telescope (SALT) has already been developed during the design thereof, this thesis focuses on the development of the methods and techniques that would result in a more accurate numerical model of the actual structure that could be used as a basis for a numerical control system. This control system will compensate for de ections in the structure by adjusting the positioning of the individual mirror segments of the primary mirror. The two main components from which the support truss is constructed are the steel nodes, and the struts that connect to them. For this project a smaller, simpler laboratory model was designed and built to have geometrical properties similar to that of the support truss. The methods and techniques that were investigated were carried out on this model. By using numerical design optimisation techniques, improved numerical models of the different strut types were obtained. This was done by performing tests on the struts so that the actual responses of the struts could be obtained. Numerical models of the struts were then created and set up so that they could be optimised using structural optimisation software. Once accurate strut models had been obtained, these strut models were used to construct a numerical model of the assembled structure. No additional optimisation was performed on the assembled structure and tests were done on the physical structure to obtain its responses. These served as validation criteria for the numerical models of the struts. Because of unforeseen deformations of the structure, not all of the measured structural responses could be used. The remaining results showed, however, that the predictive accuracy of the top node displacement of the assembled structure improved to below 1.5%, from over 60%. From these results it was concluded that the accuracy of the entire structure's numerical model could be signi ficantly improved by optimising the individual strut types.

Hierdie beheerstelsel sal kan kompenseer vir die ondersteuningsraamwerk se vervormings deur om die individuele spieëlsegmente van die primêre spieël se posisionering te verstel. Die metodes en tegnieke wat ondersoek is, is op hierdie model uitgevoer. Verbeterde numeriese modelle van die verskillende stut-tipes is ontwikkel deur middel van numerieseoptimeringstegnieke. Dit is gedoen deur toetse op die stutte uit te voer sodat hul werklike gedrag bepaal kon word. Numeriese modelle van die stutte is toe geskep en opgestel sodat hulle geoptimeer kon word om dieselfde gedrag as wat gemeet is, te toon. Hierdie geoptimeerde modelle is toe gebruik om numeriese modelle van die toets-struktuur te skep. Geen verdere optimering is op die numeriese model uitgevoer nie en toetse is op die struktuur gedoen om sy werklike gedrag te meet. Data wat deur die toetse verkry is het as validasie kriteria gedien om die akkuraatheid van die numeriese modelle van die stut-tipes te bepaal. Weens die struktuur se onvoorsiene vervorming kon alle gemete struktuurdata nie gebruik word nie. Die oorblywende data het egter getoon dat die akkuraatheid van die finale numeriese modelle van die struktuur verbeter het en dat dit die translasie van die top-node met 'n speling van 1.5% akkuraatheid kon voorspel, teenoor die oorsponlike speling van meer as 60%. Daar is bevind dat die akkuraatheid van die numeriese model van die hele struktuur noemenswaardig verbeter kan word deur die numeriese modelle van die stut-tipes te optimeer.

Recently, however, some studies have employed macroeconomic variables and dynamic specification in assessing the relationship between market values and accounting numbers (e.g. Bilson et al. 2001, Nissim and Penman, 2003, and Willett, 2003). The objective of this thesis is to investigate the nature of the relationship between share prices and accounting numbers on the Jakarta Stock Exchange for the period 1992-2002, using dynamic modelling principles in addition to the more usual cross sectional analysis. The approach to regression modelling (general-to-specific strategy)incorporated in this thesis relies less heavily than most CMR on prior economic theories of equilibrium behaviour. Apart from these novel aspects of approach and method, the study also provides valuable information about the emerging financial markets of Indonesia. The results of this thesis show that cointegration and the accompanying equilibrium correction relationship between market and book values for firms listed on the Jakarta Stock Exchange (JSX) can often be identified using accounting and macroeconomic regressors. The models are typically more informative, plausible and consistent than cross sectional models and are useful in interpreting the context in which the market to book relationship exists in Indonesia. A possibly surprising result is that in Indonesia, compared to similar models estimated using US data, the book value of net assets seems to have a stronger relationship with market value. This may be a function of the relative importance of financial statements as a source of information on the JSX.

Glucosyltransferases (GTs) are the important group of enzymes which facilitates the incorporation of UDPactivated glucose to a corresponding acceptor molecule through glucosylation. Glucosylation is a common alteration reaction in plant metabolism and is regularly associated with the production of secondary metabolites. Glucosylation serves a number of roles within metabolism including: stabilizing structures, affecting solubility, transport, and regulating the bioavailability of the compounds for other metabolic processes. GTs involved in secondary metabolism share a conserved 44 amino acid residue motif (60–80% identity) known as the plant secondary product glucosyltransferase (PSPG) box, which has been demonstrated to include the UDP-sugar binding moiety. Among the secondary metabolites, flavonoid glycosides affect taste characteristics in citrus making the associated glucosyltransferases particularly interesting targets for biotechnology applications in these species. Custom design of enzymes requires understanding of structure/function of the protein. The present study focuses on creating mutant Flavonol- 3-O- Glucosyltransferases proteins using site-directed mutational analysis and testing the effect of each mutation on substrate specificity and kinetic properties of the enzyme.

Em próteses sobre implantes, o recobrimento oclusal com materiais pouco rígidos como os acrílicos, é por vezes utilizado por supostamente reduzir a carga transmitida ao osso, entretanto não existe um consenso na literatura sobre a sua efetividade. O objetivo deste trabalho foi comparar, por análise fotoelástica e dinâmica, a transmissão de forças após a ação de cargas sobre próteses unitárias sobre implantes em função dos materiais protéticos. Confeccionou-se um modelo em resina fotoelástica com anatomia semelhante à região de pré-molares inferiores, contendo um implante de hexágono interno de 3,75 x 11,5 mm. Foram elaboradas três tipos de próteses em materiais com rigidez decrescente: 1- Metálica (alta rigidez); 2- Resina composta (rigidez moderada), 3- Com um disco de EVA interposto com h=2,5 mm (baixa rigidez). Por meio de um polariscópio circular, observou-se qualitativamente a tensão gerada no modelo durante a aplicação de forças axiais de 100 N, em regime compressivo, sobre cada tipo de prótese. Para a análise dinâmica, realizou-se um modelamento físico de cada um dos três sistemas implante-prótese com modelos de Voigt. A partir daí, uma representação matemática dos sistemas foi deduzida e resolvida, levando em consideração as propriedades mecânicas dos diferentes materiais testados, como o módulo de elasticidade (E), o coeficiente elástico (k) e a constante de amortecimento (c). Para representar uma carga mastigatória fisiológica, foi aplicado um pulso de força senoidal com duração de 0,25 s e intensidade máxima de 100 N, segundo a expressão ()) 8 cos( . 50 50 t t f π −=. Observou-se na análise fotoelástica que após a aplicação de 100N, surgiram tensões na região apical e na cervical dos implantes, com intensidade e localização similares entre os três tipos de prótese. A análise dinâmica mostrou que a força de 100N aplicada na prótese é totalmente transmitida ao implante instantaneamente. Os resultados das análises concordam entre si e mostram que não é possível amortecer as forças mastigatórias por meio dos materiais oclusais.
Less rigid oclusal materials such as acrylic resins are often used in prosthesis over implants because they are supposed to diminish the loads transferred to the bone; however there is no scientific evidence to support this theory. The objective of this work was to compare by photoelastic and dynamic analysis the load transfer after oclusal loads over single prosthesis over implants, varying the oclusal materials. A photoelastic model was created, with an anatomy similar to an inferior premolar region. An internal hexagon implant (3,75 x 11,5 mm) was put in this model. Three different types of prosthesis were constructed with decrescent stiffness: 1- Metallic (higher stiffness); 2- Composite resin (moderate stiffness); 3- With an EVA disc interposed (fewer stiffness). By means of a circular polariscope, the stress generated in the model with a compressive axial load of 100 N, was observed for each prosthesis. For the dynamic analysis, a physical modeling of each implant-prosthesis was carried out with Voigt models. After that, a mathematical representation of the systems was deduced and solved, taking into account the mechanic properties of the different materials, like the elastic modulus (E), the elastic coefficient (k) and the damping constant (c). To represent a physiologic masticatory load, a senoidal force was applied, during 0,25 s and maximum load of 100 N, by the expression ()) 8 cos( . 50 50 t t f π −=. With the photoelastic analysis, it was observed that stresses were generated at the apical and cervical regions of implants, with similar intensity and patterns between the three types of prosthesis. The dynamic analysis, showed that the loads applied over the prosthesis are transferred to the implant instantaneously. The results of both anaylis are in agreement that it is not possible to damp masticatory loads by means of occlusal materials.

Mutations were selected on the basis of sequence similarity between grapefruit F-3-O-GT, an uncharacterized GT gene in blood orange (98%), and grape F3GT (82%). Grapefruit F-3-O-GT prefers flavonol as a substrate whereas the blood orange sequence is annotated to be a flavonoid 3GT and the grape GTs could glucosylate both flavonols and anthocyanidins. Mutants of F-3-O-GT were generated by substituting L41M, N242K, E296K and N242K+E296K and proteins were expressed in Pichia pastoris using the pPICZA vector. Analysis of these mF-3-O-GTs showed that all of them preferred flavonols over flavanone, flavone, isoflavones, or anthocyanidin substrates and showed decrease in enzyme activity of 16 to 51% relative to the wild type F-3-O-GT.

Differential Item Functioning Analyses investigates whether individuals with same ability in different groups also show similar performance on an item. In matching the individuals of the same ability, most of the methodologies use total scores of the tests which are usually constructed to be unidimensional. th purpose of the present study is evaluating the PISA 2003 mathematics literacy items through the use of DIF methodology which uses a multidimensional approach in matching students instead of single total score, improve the matching for DIF analyses. In the study factor structure of the tests will be determeined via both exploratory and confirmatory analyses in a complimentary fashion. then DIF analyses conducted using Logistic regression (LR) and Mantel -Haenszel methods.Analyses showed that the matching criterion improved when multivariate analyses were used. the number of DIF items was decreased when the matching criterion is defined based on multiple criterion scores such as mathematical literacy and problem solving scores or two different mathematical literacy subtest score. In addition, qualitative reviews and examination of the distribution of DIF items by content categories, cognitive demands, item types,item text, visual-spatial factors and linguistic properties of items were analyzed to explain the differential performance. Curriculum, cultural and translation differences were the main criteria for the qualitative analyses of DIF items. The results imply that curriculum and translation differences in items might be causing the DIF across Turkish and English versions of the tests.

La presente investigación explora el fenómeno de los formatos televisivos de ficción desde la perspectiva de la representación audiovisual y la relación de la ficción televisiva con la identidad cultural. En particular, lleva a cabo el estudio de caso de la serie de televisión española Los misterios de Laura y sus adaptaciones en EEUU e Italia. El principal objetivo de la investigación es identificar cuáles son las diferencias y similitudes en el contenido de las tres versiones y comprender los factores que las explican. Para ello, este estudio propone un modelo de análisis cualitativo que analiza el texto con el propósito de caracterizar cada una de las versiones y compararlas. Además, el examen del texto se complementa con el análisis del contexto de producción. Los principales resultados revelan que las similitudes entre las versiones son resultado de un proceso de búsqueda de equivalencias entre los contextos culturales y atienden a los rasgos que caracterizan a la serie como formato de televisión. Por otro lado, las diferencias se explican por un conjunto amplio de factores pertenecientes al enfoque narrativo, al contexto de producción, las convenciones televisivas y al uso del imaginario cultural. En conjunto, esta investigación señala que existe una articulación de lo global y lo local en la adaptación de los formatos televisivos de ficción, donde lo global se relaciona con las prácticas y las lógicas comerciales de la televisión, y lo local se vincula con las desigualdades que existen entre las industrias televisivas y la persistencia en el uso de mecanismos de producción de la identidad cultural en los textos televisivos.
This research explores the phenomenon of television fiction formats from the perspective of audiovisual representation and the relationship between television fiction and cultural identity. In particular, the case study of the Spanish television series Los misterios de Laura and its adaptations in the USA and Italy is carried out. The main objective of the research is to identify the differences and similarities in the content of the three versions and the factors that may explain them. To do so, this study proposes a qualitative analysis model that analyses the text with the purpose of characterizing each of the versions and comparing them. In addition, the examination of the text is complemented by the analysis of the production context. The main results reveal that the similarities between the versions are the result of the process of searching for equivalences between cultural contexts. Moreover, those similarities related to the traits that characterise the series as a television format. On the other hand, the differences are explained by a broad set of factors pertaining to the narrative approach, the context of production, television conventions and the use of the cultural imaginary. To conclude, a connection between the global and the local regarding the adaptation of the television formats is observed, where global is related to the practices and the commercial logic of television, and local is connected to the inequality between the television industries and the persistence in using cultural identity production mechanisms in television texts.

Flavonoids are an important class of secondary metabolites widely distributed in plants. The majority of naturally occurring flavonoids are found in glucosylated form. Glucosyltransferases are enzymes that enable transfer of glucose from an activated donor (UDP-glucose) to the acceptor flavonoid substrates. A flavonol specific glucosyltransferase cloned from Citrus paradisi (Cp3OGT) has strict substrate and regiospecificity. In this study, amino acid residues that could potentially alter the rigidity observed in this enzyme were mutated to position equivalent residues of a putative anthocyanin specific glucosyltransferase from Clitorea ternatea and a GT from Vitis vinifera that can glucosylate both flavonols and anthocyanidins. Using homology modeling followed by site directed mutagenesis to identify candidate regions, three double mutations were made. To test the basis of substrate specificity, biochemical analysis of the three recombinant mutant proteins was carried out. Recombinant protein with mutation S20G+T21S revealed that the enzyme retained activity similar to the wildtype (Cp3OGT) (WT- Km app-104.8 µM; Vmax = 24.6 pmol/min/µg, Mutant- Km app-136.42 µM; Vmax -25pmol/min/µg) but the mutant was more thermostable compared to the WT. The (S290C+S319A) mutant protein retained 40% activity relative to wildtype and has an optimum pH shifted towards the acidic side (pH 6) (Km app-8.27 µM; Vmax-90.9 pmol/min/µg). Mutation of Glutamine87 and Histine154 (H154Y+Q87I) have rendered this recombinant protein inactive with every class of flavonoid tested. Interestingly, the single point mutations H154Y and Q871I had significant activity, slightly greater than that of wildtype enzyme. The two active recombinant proteins will further be analyzed to determine whether the mutations have altered regiospecificity of the original enzyme. Product identification is being conducted using HPLC.

Flavonoids are secondary metabolites that are important in plant defense, protection and human health. Most naturally-occurring flavonoids are found in glucosylated form. Glucosyltransferases (GTs) are enzymes that catalyze the transfer of glucose from a high energy sugar donor to an acceptor molecule. A flavonol-specific 3-O-GT enzyme has been identified and cloned from leaf tissues of grapefruit. The enzyme shows rigid substrate specificity and regiospecificity. F3-O-GTs from grape (Vitis vinifera) and grapefruit (Citrus paradisi) were modeled against F7-O-GTs from Crocus sativus and Scrutellaria biacalensis, and several non-conservative amino acid differences were identified that may impact regioselectivity. This research is designed to test the hypothesis that specific amino acid residues impart the regiospecificity of the grapefruit enzyme. Site-directed mutagenesis was performed on three potentially key amino acid residues within the grapefruit F3-O-GT that were identified through homology modeling. Analyses of the enzyme activity of the mutant F3-O-GT proteins revealed that the single point mutations of serine 20 to leucine (S20L) and proline 297 to phenylalanine (P297F) rendered the recombinant enzyme inactive with flavonol substrates. Mutation of glycine 392 to glutamate (G392E) was active at 80% relative to the wild type. The mutant enzyme also did not show broadened acceptor specificity as it also favored flavonols as the preferred acceptor substrate. The glucosylation products of the active mutant enzyme will be analyzed to determine if this resulted in a change in regiospecificity.

Soybean (Glycine max (L.) Merr.) cyst nematode (SCN) (Heterodera glycines Ichinohe), an obligate sedentary endoparasite, is the most economically destructive pathogen in soybean production and causes over $1 billion in annual losses in the United States. Planting resistant cultivars is the primary management method to control SCN for the long-term purpose, but the nature of genetic resistance is little known. The Rhg1 (Resistance to H. glycines) locus on chromosome 18 is found as a major quantitative trait locus (QTL) that contributes resistance to SCN. The chemical mutagen ethylmethane sulfonate (EMS) can be utilized to induce genetic mutations in soybean populations, which screened by an efficient reverse genetic strategy known as Targeting Induced Local Lesions IN Genomes (TILLING) for functional gene analyses. The objective of this study was to analyze the function of SNAP gene (Glyma18g02590) at rhg1 allele from `Forrest' (`Peking'-derived SCN resistant cultivar) using TILLING. Soybean cultivar `Forrest' seeds were mutagenized with EMS and grown to generate M1 plants. M1 plants were self-pollinated to produce approximately 3000 M2 plants. Genomic DNAs were extracted from young leaves of individual M2 plants and quantified to normalize concentration of DNAs. The DNA samples were then pooled eight-fold in 96-well plates for mutations screening by TILLING. Moreover, 12 phenotypic traits including chlorophyll deficiency, leaf shape, branch architecture, seed color, seed weight, fatty acid phenotype were identified in the mutagenized population, analyzed and archived in this study.

The unique international tripartite organization is voluntary but mandates state participants to abide by the Kimberley Process Certification Scheme (KPCS) minimum requirements in order to stem the financing or wars against legitimate governments. This study represents the first to explore the communications within the KP. The linguistic turn relies on Classical rhetoric theory with an emphasis on Aristotle's three appeals of persuasion (“pisteis”): ethos, logos and pathos of elite actors in the KP. As for the precise nature of the contribution to rhetorical analysis, this project is best characterised as an application of Classical principals of rhetorical analysis, rather than as a development of theory. A comprehensive literature review of the KP and KPCS is another distinctive contribution. Furthermore, this academic endeavour offers a unique method as shown in the observation of a KP Intersessional meeting. Supplementing the qualitative inquiry, semi-structured interviews were conducted with all of the KP groups and included a wide sample of civil society international and national non-government organizations, state actors and industry members otherwise not represented in previous empirical efforts on the subject. The data chapters achieve the primary aim to add to the understanding of the KP. Firstly, the civil societies engage in boycott rhetoric using ethos and negative pathos. As for state actors, the KP Chair exhibits charismatic leadership rhetoric, while ‘recognized' established states use logical reasoning, the ‘outlier' states evoke positive pathos. Lastly, the diamond industry experts appeal to negative emotions, the World Diamond Council to logos, and De Beers to positive emotional appeals. Combined, the rhetoric shows (a) how KP rhētors use different rhetorical strategies; (b) which in turn shape distinct discourses; (c) and contain dissimilar claims; (d) points to different motivations; (e) highlight different identities; (f) reveal key characteristics, and; (g) the nature of relationships within the KP. The organization rhetorical analysis also entails how the multiple KP leaders view the KP and KPCS and change. While the rhetoric helps demonstrate the constraints surrounding the KP and KPCS it also underlines the primary human rights and human security in which they all share. This thesis provides an extended critical view of the rhetoric by connecting Aristotelian pisteis with different conceptions of power outlined by French and Raven (1959) and Lukes (2005/1974). Combined, the rhetoric helps to explain the ways the KP attempts to achieve their specific political and economic goals while also building relationships with their stakeholders. Rhetoric is a worthwhile theory and methodological approach in order to explore organizations. The KP, and other international organizations provide an opportune arena for further rhetorical attention.

Abstract:
The use of seismic waves to detect subsurface targets such as landmines is a very promising technology compared to existing methods like Ground Penetrating Radar (GPR) and Electromagnetic Induction (EMI) sensing. The fact that seismic waves induce resonance in man-made targets, and hence more scattering, gives this method a natural ability to discriminate landmines from common types of clutter like rocks, wood, etc. Reflection and resonance from the targets can be used in imaging to detect the location of targets. However, existing methods require a large number of measurements for imaging and detection, which are expensive and time consuming. To reduce the number of measurements and enable faster detections, a new sensing strategy is proposed based on optimally maneuvering sensors. The system would operate in two main modes. In search mode, the goal would be to move on top of a target using the minimum number of measurements. Once the target is found, the system would switch to a detection mode to make its final decision. The seismic sensor system is an active system, where a seismic source generates the probing pulse. The waves reflected from buried targets are collected by an array of sensors placed on the surface, and then an imaging algorithm is used to estimate the target position. The performance bounds for this position estimate are derived in terms of the Fisher information matrix (FIM). This matrix gives the dependence of the target position estimate on the array position. Based on the FIM, the next optimal array position is determined by using the theory of optimal experiments. The next array position will be the one that reduces the uncertainty of the target position estimate the most. The whole array is moved to this new position, where the same steps are repeated. In this way, the target can be localized in a few iterations.

The following master thesis is the product of the work carried out during the Erasmus exchange of the year 2017-2018 that involved the author, exchange student from the University of Bologna, the Universitat Politècnica de Catalunya , TriM, an italian company with a strong knowledge of weather data and forecasting, and Meteocat, the public meteorological company of Catalonia in a collaboration aimed to find new methodologies for the processing of meteorological data. The reason that motivated this work is dictated by the increasing amount of weather data available today, that necessarily drives the weather forecasting in a more automated procedure that reduces the time needed to generate a forecast and the intervention of a human, in the figure of a meteorologist, in the analysis of the data. This allows to process more data and thus having predictions that take advantages of the usage of many information that could result in improved forecasting. The development in the field of machine learning allows today to treat a vast amount of information in an automatic way, leaving the analysis process to the machines, freeing the user of this time-consuming task. And unsupervised learning is the branch that can process data that are not labelled nor preprocessed, speeding up the data mining. The goal of this thesis is to apply unsupervised learning techniques to this scope, taking inspiration from the available literature that experimented in this field and combining different solutions into a new technique that aims to reduce the human decision in the process of the recognition of wind patterns and improve the automationof the whole process.

The interest in modeling non-maturing deposits has skyrocketed ever since thefinancial crisis 2008. Not only from a regulatory and legislative perspective,but also from an investment and funding perspective.Modeling of non-maturing deposits is a very broad subject. In this thesis someof the topics within the subject are investigated, where the greatest focus inon the modeling of the deposit volumes. The main objective is to providethe bank with an analysis of the majority of the topics that needs to be cov-ered when modeling non-maturing deposits. This includes short-rate model-ing using Vasicek’s model, deposit rate modeling using a regression approachand a method proposed by Jarrow and Van Deventer, volume modeling usingSARIMA, SARIMAX and a general additive model, a static replicating port-folio based on Maes and Timmerman’s to model the behaviour of the depositaccounts and finally a liquidity risk model that was suggested by Kalkbrenerand Willing. All of these models have been applied on three different accounttypes: private transaction accounts, savings accounts and corporate savingsaccounts.The results are that, due to the current market, the static replicating portfoliodoes not achieve the desired results. Furthermore, the best volume model forthe data provided is a SARIMA model, meaning the effect of the exogenousvariables are seemingly already embedded in the lagged volume. Finally, theliquidity risk results are plausible and thus deemed satisfactory.
Intresset för att modellera inlåningsvolymer utan en kontrakterad förfallodaghar ökat markant sedan finanskrisen 2008. Inte bara sett utifrån ett perspek-tiv att uppfylla krav som ställs av tillsynsmyndigheter, men också sett utifrånbankens investerings-och finansieringsperspektiv.Målet med det här arbetet är att förse banken med en analys av majoritetenav de olika områdena som man behöver ta hänsyn till när man ska model-lera inlåningar utan förfallodatum, men med ett fokus på volymmodellering.I den här rapporten modelleras räntor (kortränta och kontoränta), kontovoly-merna, kontobeteendet samt likviditetsrisken. Detta görs med hjälp av Vasicekför korträntan, en regressionsmetod samt en metod som föreslagits av Jarrowoch Van Deventer för kontoräntan, SARIMA, SARIMAX och en generell ad-ditiv regressionsmetod för volymerna, en statisk replikeringsportfölj baseradpå Maes och Timmermans modell för att imitera kontona och slutligen så mo-delleras likviditetsrisken med ett ramverk som föreslagits av Kalkbrener ochWilling. Alla dessa nämnda modeller appliceras, där det är möjligt, på de treolika kontotyperna: privatkonton, sparkonton samt företagssparkonto.Resultatet är att räntemodelleringen samt replikeringsportföljen inte ger ade-kvata resultat på grund av den rådande marknaden. Vidare så ger en SARIMA-modell den bästa prediktionen, vilket gör att slutsatsen är att andra exogenavariabler redan är inneslutna i den fördröjda volymvariabeln. Avslutningsvisså ger likviditetsmodellen tillfredsställande resultat och antas vara rimlig.

I denna uppsats analyseras spelen Howling Dogs och 16 Ways to Kill a Vampire at McDonalds ur ett narratologiskt perspektiv. Huvuddelen av analysen fokuserar på hur hypertextens mediala förhållanden påverkar relationen mellan spelaren och narrativet samt på förhållandet mellan berättare och lyssnarinstans. Analysen baserar sig på metoder med utgångspunkt i possible worlds theory, formulerat av Alice Bell. Uppsatsen exemplifierar hur man går till väga i analys av texter med ett starkt deiktiskt förhållande mellan texten och läsaren. Syftet är att undersöka existerande metoder och att förbättra dem, tillika en, möjligtvis naiv, förhoppning om att komma ett steg närmare ett större litteraturvetenskapligt intresse gällande vad spel kan tillföra litteraturvetenskapen.
This essay contains narratological analyses of the games Howling Dogs and 16 Ways to Kill a Vampire at McDonalds. The analyses are focused on the hypertext’s medial impact on the relations between user and narrative as well as narrator and narratee. The work is based on methods using possible worlds theory, created by theorists such as Alice Bell. A main purpose here is to further exemplify how to analyze texts having a deictic effect between themselves self and their readers. The purpose here is thus to test existing methods and further develop them. This purpose is also motivated by a, possibly naïve, hope of getting one step closer to a wider acknowledgement of how games can contribute to the subject of literature.

El presente trabajo de investigación se centra en el estudio de la competencia interactiva de tres grupos de cuatro aprendices de español como lengua extranjera durante la realización de una tarea compleja, un juego de rol, articulado en un chat. Los participantes son doce estudiantes chinos que cursan estudios superiores en dos universidades en Cataluña. Los alumnos fueron agrupados en función de su competencia lingüística (A2, B2- y B2+) en la lengua meta. Este estudio adopta la metodología cualitativa del análisis de la conversación para describir detallada y secuencialmente la organización de la interacción que cada grupo lleva a cabo para conseguir completar la tarea propuesta. El objetivo de esta investigación es doble. Por un lado, se pretende descubrir y explicar cómo los diferentes grupos de aprendices interpretan una misma tarea e interactúan con su interlocutor para conseguir lograr el objetivo de la tarea. Por otro lado, se quiere obtener una mejor comprensión de cómo los alumnos utilizan los recursos interactivos que poseen para construir y mantener la conversación en el chat. Este segundo objetivo nos permite observar como los aprendices activan su competencia interactiva y, en definitiva, cómo aprenden una lengua extranjera en un contexto, la interacción en un chat, poco estudiado. La consecución del objetivo de la tarea está ligada a la interpretación por parte de los participantes de las instrucciones recibidas y a la identidad que ellos adoptan durante su realización. En algunos casos, los alumnos no conceptualizan la tarea como una actividad comunicativa social y realizan parte de ella tarea de forma individual o se centran en la precisión del producto final. En otros casos, la tarea se resuelve colaborativamente. Estos resultados demuestran que una tarea pedagógica se construye siempre a través de la interacción entre los interlocutores durante su ejecución. El hecho de que la comunicación entre los aprendices se lleve a cabo en un chat, un contexto con su propia organización idiosincrásica de la interacción, plantea a los alumnos retos comunicativos de orden diverso. Al abordarlos ponen en juego y desarrollan su competencia interactiva. Entre los retos comunicativos que aparecen mayoritariamente en nuestros datos cabe citar la necesidad de (1) reparar problemas en la interacción o problemas en el procedimiento empleado para resolver la tarea, (2) mantener la intersubjetividad con el interlocutor a través de actividades de alineamiento, y (3) seleccionar y manejar los temas de la conversación.
This research analyses the interactional competence of Chinese students who are learners of Spanish as foreign language, as it emerges during peer interactions in computer-mediated chat used to complete a task. The students were studying at two universities in Catalonia. Twelve learners were observed, separated into three different groups for the task based on their proficiency in Spanish (A2, B2- and B2+), each group having four learners. The task involved a role-play. The study is qualitative, drawing on conversation analysis to document the participants’ perspectives emerging in their natural talk-in-interaction in the chat. The goals of this investigation were twofold. On the one hand, it is intended to describe and explain how participants in different groups interpreted the same role-play task and interacted with others to resolve it, including the discursive procedures that allowed them to orient to task accomplishment; and on the other hand, we wanted to gain a better understanding of how the participants used these interactional resources to construct, manage and maintain the chat conversations, and activated their interactional competence in this new interactional setting. Task accomplishment is contingent on how learners interpret the instructions and on the identities they adopt in the role-play. Some learners did not orient to the task as a communicative activity; instead, they performed it individually or focused on the accuracy of the final product. The findings of this study show that the task is constructed through the participants’ interaction during task implementation. The communication takes place in a chat, a context with its own idiosyncratic interactional organization, which poses communication challenges for learners. They displayed and developed their interactional competence. Among the communication challenges that appear mostly in our data, learners need to (1) repair emerging problems in relation to the talk and the task, (2) show alignment and maintain intersubjectivity with other interlocutors, and (3) manage and transition between topics.

The governing equations and the associated boundary conditions for three problems of nonuniform Timoshenko beams are derived by using the Hamilton''s principle. They are:(1) the free vibrations of nonuniform Timoshenko beams subjected to axial forces,(2) the nonconservative stability of nonuniform Timoshenko beams subjected to partially tangential follower forces, (3) the pure bending vibrations of rotating nonuniform Timoshenko beams. In each subject, the two coupled differential equations are decoupled into one complete fourth- order differential equations with variable coefficients in the angle of rotation due to bending or in the flexuarl displacement, respectively. The explicit relation between the flexural displacement and the angle of rotation due to bending is established. The frequency equations of the beam are derived and expressed in terms of four normalized fundamental solutions of the associated reduced governing differential equation. Consequently, if the geometric and material properties of the beam are in polynomial forms, then the exact solutions for the problem can be constructed by the method of Frobenius. But its rate of convergence for the solution will be slow for some problem with special variable coefficients. The modified method of Frobenius by which the rate of convergence for the solution is fast,is constructed. However, if the coefficients of the system are not in polynomial form, a semi-closed method is established. Moreover, the mechanism of tensional instability of beam is studied. And the neccesary and sufficient condition at which the phenomenuous of tensional instability happens, is discovered. Finally, the influence of some parameters on the vibrations and the nonconservative stability of nonuniform Timoshenko beams is studied.
