Abstract:
In this essay Andrew Anderson proposes that all Lorca’s plays share certain commonalities: his protagonists invariably find themselves in some kind of predicament or dilemma, and it always turns out that there is no favourable solution to the position in which they find themselves. A number of diverse and, on the face of it, conflicting factors play into creating these dramatic situations: family, society, gender, psychology, as well as more metaphysical elements that suggest aspects of existentialism. Here, the characters and plots of four of Lorca’s best-known plays from the 1930s are analyzed to demonstrate the various ways in which these disparate factors combine to produce what are usually tragic outcomes. Andrew Anderson is Professor of Spanish in the Department of Spanish, Italian, and Portuguese at the University of Virginia. He has published extensively on the work of García Lorca and the Spanish historical avant-garde, most recently La recepción de las vanguardias extranjeras en España: cubismo, futurismo, dadá. Estudio y ensayo de bibliografía (Seville: Renacimiento, 2018).

Abstract:
In their article, “Lessons from the pandemic about science education,” Andrew Zucker and Pendred Noyce stress the importance of students making sense of the world around them and use the COVID-19 pandemic as a call to change the Next Generation Science Standards (NGSS) to better address important elements of science literacy. In this response, Eric Brunsell, Kevin Anderson, Kelly Steiner, and Tom Anfinson note that they agree about the importance of science literacy but explain that Zucker and Noyce misrepresent the NGSS.

Abstract:

Abstract:
Abstract Williams, A., Bax, N. J., and Kloser, R. J. 2009. Remarks on “Comment on: Williams et al. (2009) Australia's deep-water reserve network: implications of false homogeneity for classifying abiotic surrogates of biodiversity, ICES Journal of Marine Science, 66: 214–224” by Peter T. Harris, Andrew D. Heap, Tara J. Anderson, and Brendan Brooke. – ICES Journal of Marine Science, 66: 2086–2088. Commitment by many nations at the 2002 World Summit on Sustainable Development to complete national networks of Marine Protected Areas (MPAs) by 2012 focused attention on MPA design and implementation, especially for the deep waters that make up the majority of many national EZs. Further, there is a focus on conservation in the High Seas through the Convention on Biological Biodiversity and the FAO, e.g. for seamounts and other vulnerable benthic habitats. Any manuscript that raises basic issues on one identified area for conservation is therefore a prospectively useful addition to the international debate. The Comment by Harris et al. (2009) on a paper authored by myself and colleagues (Williams et al., 2009) is a potentially useful contribution. The authors provide data, examples, and insights into aspects of the design and planning process for Australia's SE network of reserves. Here, we address the main question of how geological descriptions can be used or modified to provide the most information on marine biodiversity patterns for use in conservation planning—in the context of the major points raised by Harris et al. (2009).

Beal, Jane. "Beauty and the Beast: The Value of Teaching Fairy Tales to University Students in the 21st Century." International Journal of English and Cultural Studies 5, no. 1 (April 28, 2021): 1. http://dx.doi.org/10.11114/ijecs.v5i1.5207.

Abstract:
In this essay, I suggest that fairy tales have particular value for students studying at the university level. Assigning fairy tales allows students to read familiar stories from their childhood and reconsider them from critical perspectives. When teaching a college course on fairy tales, my students and I utilize three essential frameworks for understanding fairy tales, focusing on the psycho-social development and sexual maturation of the human person, feminist critique and the need for gender equality in a patriarchal world, and audience reception and reader responses leading to emotional progress and even spiritual enlightenment. Students primarily familiar with Disney film versions of fairy tales enlarge their understanding of multiple versions of tales, both early modern and contemporary. They become familiar with classic fairy tale writers and collectors, such as Charles Perrault, Madame d’Aulnoy, the Brothers Grimm, Hans Christian Anderson, Oscar Wilde, Andrew Lang, J.R.R. Tolkien, C.S. Lewis, Anne Sexton, Angela Carter, and J.K. Rowling as well as fairy tale scholars like Bruno Bettelheim, Maria Tartar, and Jack Zipes. Their study not only results in a firm grasp of the key aspects of story in general, but in the ability to see connections between the real-world problems of the 21st century – such as poverty, starvation, disease, inequality, child abuse, human trafficking, and abuses of political power, among others – and lessons learned from fairy tales. This essay analyzes “Beauty and the Beast” as a key example of the genre and identifies pedagogical strategies for teaching it.

Abstract:
Background:As well as being an established oncoprotein and a therapeutic target in cancer,Proviral Integration site for murine Moloney leukemia virus-1(pim-1) has been implicated in human autoimmunity. We previously confirmed this serine-threonine protein kinase to be strikingly upregulated in circulating CD4+ T cells of untreated rheumatoid arthritis (RA) patients as a consequence of IL-6 signalling1-2. Evidence for the relevance of pim-1 signalling in the disruption of RA synovial fibroblast (RASF) homeostasis3further supports its candidacy as a therapeutic target. Objectives:To investigate PIM1 and its family members (PIM2 and PIM3) as potential candidates for drug repurposing in RA.Methods:A flow cytometric assay for PIM1 transcript measurement in circulating CD4+ T cells of early arthritis patients was validated against real-time PCR in paired cells isolated by bead selection. Synovial protein expression in tissue from the same cohort of untreated RA patients and disease controls was determined by quantitative multiplex immunofluorescence. The functional consequences of manipulating pim kinase family expression in freshly purified T cell receptor (TCR)-stimulated CD4+ T cells from early RA patients was explored. The impact of pim-1 specific and pim-1-3 (pan-pim) kinase inhibition on progression of the IL-6 dependent collagen-induced arthritis (CIA) model was assessed. Results:The percentage of circulating CD4+ T cells positive forPIM1transcript by flow cytometry proved a faithful surrogate for gene expression in early arthritis (Figure 1A), distinguishing RA from other pathologies (Figure 1B). Pim-1 protein expression was increased in the synovium of untreated RA compared with disease controls, including amongst infiltrating CD4+ T cells (Figure 1C-D).In vitro, exposure of TCR-stimulated early RA CD4+ T cells to pim kinase inhibitors restrained their activation and proliferative capacity; diminished pro-inflammatory cytokine production (IFN-g and IL-17) and an expanded CD25hiFoxP3+ regulatory T cell (Treg) fraction were also observed in treatedversusun-treated cells. Finally, administration of pim inhibitors robustly attenuated clinical scores of arthritis in the CIA model, with reduced cartilage loss observed in animals treated with a pan-PIM inhibitor compared with vehicle control (Figure 2).Conclusion:Our data highlight pim kinases as plausible therapeutic targets for a subgroup of early RA patients that may be identifiable using tractable in vitro assays. Pim kinase inhibitors could simultaneously target immune inflammation and RASF dysregulation; consideration should now be given to their repurposing for this condition.References:[1] Anderson AE et al Annals of the Rheumatic Diseases 2016; 75:466-73.[2] Anderson AE et al Rheumatology 2019; 58:1250-1258[3] Ha YJ et al Rheumatology 2019; 58:154-64Disclosure of Interests:Nicola Maney Consultant of: Current employee of Eli Lilly, Henrique De Paula-Lemos: None declared, Ben Barron-Millar: None declared, Andrew Mellor Shareholder of: NewLink Genetics PLC, and has received patent licensing income from this source., John D Isaacs Consultant of: AbbVie, Bristol-Myers Squibb, Eli Lilly, Gilead, Janssen, Merck, Pfizer, Roche, Amy Anderson: None declared, Arthur Pratt Grant/research support from: Pfizer, GlaxoSmithKlein

Abstract:
The lunatic asylum remains one of the most remarkable institutional monuments of the modern world, dominating the social landscape of Victorian Britain and exercising a powerful attraction for social historians of medicine, an attraction almost as great as the spectre of the madhouse for contemporary novelists. Our image of the Victorian asylum is still pervaded to a surprising degree by the gloomy spectacle of the total institution presented by Michel Foucault, though it has been modified by a whole range of institutional and philosophical accounts undertaken in the past three decades. Pioneering studies by researchers such as Andrew Scull have illuminated not only the power exercised by the new asylum superintendents, armed with medical discourses of moral treatment and the early promise of curability, but also the continuing dominance of the ‘mad doctors’ in the sombre years of neo-Darwinian pessimism and eugenics doctrines. More recent contributions to the now enormous literature on the social history of insanity have shifted the focus of attention from earlier concerns with charting the rise of the asylum and the elaboration of medical discourses under the psychiatric gaze of physicians to a detailed reconstruction of the social environment of the asylum and especially to the interplay between familial circumstances and the way institutions responded to the insane. Such concerns were also clearly evident in important earlier studies by Walton, Scull, Digby and others, which drew on fundamental work by Anderson on the changing role of the family during industrialization. These scholars drew attention to the importance of family and kinship relations in the negotiation of a lunatic's passage to the Victorian asylum, as well as the role of wider forces of economic change, population growth and migration in shaping the environment in which decisions about the care of the mad were made.

A signal witness?

Abstract:
This paper studies a model widely used in the weak instruments literature and establishes admissibility of the weighted average power likelihood ratio tests recently derived by Andrews, Moreira, and Stock (2004, NBER Technical Working Paper 199). The class of tests covered by this admissibility result contains the Anderson and Rubin (1949, Annals of Mathematical Statistics 20, 46–63) test. Thus, there is no conventional statistical sense in which the Anderson and Rubin (1949) test “wastes degrees of freedom.” In addition, it is shown that the test proposed by Moreira (2003, Econometrica 71, 1027–1048) belongs to the closure of (i.e., can be interpreted as a limiting case of) the class of tests covered by our admissibility result.

Abstract:
ABSTRACT The article derives from the intertextual approach to the notion of literary identity, introduced to the modern literary theory in the 1960’s by Julia Kristeva (born 1941). The main idea behind this approach is that no literary text should be perceived as an isolated unity, but always in relation to other texts, which it refers to. The author applies the intertextual perspective to examine the signs of external literary influences in Lise Andersens minimalist prose collection ”En særlig sommer og andre billeder”, published in 2008. The analysis aims to highlight influences from the American short story genre, which appears as an important source of inspiration not only for Andersen herself, but in fact for most of the Danish writers of minimalist fiction. The analysis provides examples of, how inspiration from the American short story manifests itself in form, narration and structure of the selected stories in the collection. Last but not least, the author draws attention to the general importance of American influences in Danish contemporary short fiction and their role for the critical perception of this literary genre both in Denmark and abroad.

Abstract:
In the linear instrumental variables model with possibly weak instruments we derive the asymptotic size of testing procedures when instruments locally violate the exogeneity assumption. We study the tests by Anderson and Rubin (1949, The Annals of Mathematical Statistics 20, 46–63), Moreira (2003, Econometrica 71, 1027–1048), and Kleibergen (2005, Econometrica 73, 1103–1123) and their generalized empirical likelihood versions. These tests have asymptotic size equal to nominal size when the instruments are exogenous but are size distorted otherwise. While in just-identified models all the tests that we consider are equally size-distorted asymptotically, the Anderson-Rubin type tests are less size-distorted than the tests of Moreira (2003) and Kleibergen in over-identified situations. On the other hand, we also show that there are parameter sequences under which the former test asymptotically overrejects more frequently. Given that strict exogeneity of instruments is often a questionable assumption, our findings should be important to applied researchers who are concerned about the degree of size distortion of their inference procedure. We suggest robustness of asymptotic size under local model violations as a new alternative measure to choose among competing testing procedures. We also investigate the subsampling and hybrid tests introduced in Andrews and Guggenberger (2010a, Journal of Econometrics 158, 285–305) and show that they do not offer any improvement in terms of size-distortion reduction over the Anderson-Rubin type tests.

Abstract:
AbstractThis article proposes that the recent history of the live solo multi-instrumentalist, more conventionally known as the ‘one-man band’, might serve as a useful window onto broader transformations in postmodern culture. If the one-person band has historically been understood as a humorous, culturally devalued phenomenon, with its practitioners occupying marginal race and class positions, the recent visibility of solo multi-instrumentalism in indie and experimental rock attests to its altered status in contemporary culture. The present discussion situates the shifting fortunes of the one-person band in relation to the contemporary hegemony of neoliberal conceptions of subjectivity. In their performance of nimble, flexible and omni-competent self-reliance, artists such as Theresa Andersson, Robert Fripp and Andrew Bird model some of the key qualities idealised by theorists of a free-market ‘entrepreneurial selfhood’.

Abstract:

Abstract:
Background:Rheumatoid arthritis (RA) is a chronic inflammatory disease of immune dysregulation affecting the joints. While T cells play a recognised role in disease pathogenesis, the presence of autoantibodies years before the clinical onset of disease, and the efficacy of the B cell-depleting therapy rituximab, highlight a pathogenic role for these functionally diverse lymphocytes. A novel subset, termed age-associated B cells (ABCs), are described as CD19high CD21- CD11c+, with a high proportion expressing the transcription factor T-bet; they are elevated in murine models of autoimmunity and produce autoantibodies characteristic of autoimmune disease. A detailed characterisation of ABCs in early, drug naïve RA has not yet been conducted. Objectives:We aimed to characterise peripheral blood (PB) and synovial fluid (SF) ABCs in patients suffering from early drug naïve RA. As a secondary objective we sought to determine whether this population differs between RA patients and age-matched early psoriatic arthritis (PsA; disease controls) and healthy controls. Methods:Newly presenting early RA and other inflammatory arthritis patients, naïve to immunomodulatory treatment, were recruited from the Newcastle Early Arthritis Clinic. Patients with established RA (≥1 year duration on treatment and age-matched healthy controls were recruited in parallel. B cell subsets (naïve (CD19+IgD+CD27-), switched memory (CD19+IgD-CD27+) and ABC (CD19+CD21-CD11c+)) in PB and SF were characterised by flow cytometry. FACS sorted PB B cell subset gene expression profiles were assessed using a customised NanoString nCounter Human Immunology v2 Panel. Results:Transcriptionally, ABC differed from both naïve and switched memory B cells. In keeping with published studies ABCs had an activated memory phenotype, exemplified by elevated expression of CD69, CD80, and CD86, as well as Ki67, HLA-DR, IgG and T-bet. Interestingly, ABC had high expression of the Fc Receptor Like (FcRL) family (FcRL1-5), and an inflammatory homing profile (high levels of CXCR3 and low levels of CXCR4 and CXCR5). We found no difference in the proportion of PB ABC between RA patients and control groups, and no association of ABC frequencies with age. Focussing on RA ABCs specifically, we observed elevated ABC frequencies in females, but no association with disease activity. In keeping with their chemokine receptor profile, cross-sectional analysis also demonstrated ABC enrichment in SF compared to PB, with SF frequencies higher in established than early disease. The transcriptome of ABCs from early RA patients differed from both age-matched disease controls and healthy donors (Figure 1 next page). Conclusion:These data demonstrate that ABCs have a unique, activated, class-switched proliferative phenotype that is transcriptionally distinct from switched memory and naïve B cells. Interestingly, at the transcriptome level early RA ABCs differ to their counterparts in health and other inflammatory arthritides, suggesting they may differ functionally and contribute to pathogenesis. Of note, their high expression of MHC class II (HLA-DR) and co-stimulatory (CD80 and CD86) molecules suggests an important antigen-presentation function of ABC, which together with their unique FcRL family expression pattern, warrants further functional characterisation.Figure 1.Differential gene expression in ABCs from early RA patients compared to early PsA patients (A) and healthy controls (B). A NanoString nCounter Technologies chip was used to assess gene expression. Raw counts were normalised to the housekeeping genes. Sample quality was then assessed using the arrayQualityMetrics package. Gene expression profiles between different donor groups was assessed using the DESeq2 R package. In both hierarchical clustering heatmaps gene expression intensities were log2 transformed and their z-scores are displayed as colours ranging from yellow (low expression) to red (high expression) as shown in the key.Disclosure of Interests:Gemma Vidal-Pedrola: None declared, Najib Naamane: None declared, Dagmar Scheel-Toellner: None declared, Arthur Pratt Grant/research support from: GSK, Pfizer and Janssen, Andrew Mellor: None declared, John D Isaacs Speakers bureau: Abbvie, Gilead, Roche, UC, Consultant of: Abbvie, Gilead, Roche, UC, Grant/research support from: Pfizer, GSK and Janssen, Amy E. Abstract:

Abstract:
In the context of continuing political, social, and economic crises, trade unions in most European countries are seen as weakened, protest as having little influence, and the solidarity between European populations as damaged. Under these circumstances, scientists and practitioners have placed hope in the cooperation of social movements and trade unions in order to revitalise trade unions and achieve common goals. However, the conditions for forming alliances between trade unions and social movement organisations have, thus far, been primarily researched, and partially theorised, from the point of view of different disciplines. In doing so, approaches that emphasise the strategic alignment with resources and context diverge from approaches that highlight the relevance and necessity of shared ideologies or identities. This contribution aims to bring existing approaches from movement and industrial relations research into dialogue with each other and calls for a further integration of both perspectives. It contributes to a more holistic understanding of joint movement and trade union action current concepts such as social movement unionism cannot provide. It uses two examples to show that instrumental, strategy-driven modes and identity-based, culturally-driven modes of cooperation are not contradictory. There are situations in which strategic decisions on resources and political influence are more decisive than ideological proximity and vice versa. These findings are of social and scientific relevance for understanding mechanisms of solidarity construction and processes of bridging differences even in increasingly fragmented and unequal societies.

Abstract:
Background:Patients (pts) with psoriatic arthritis (PsA) and axial involvment exhibit greater disease activity and quality of life impairments compared with those without axial involvment. Objectives:To characterize PsA pts with and without axial involvement and compare efficacy of UPA vs placebo (PBO) in PsA pts with axial involvement. Methods:In SELECT-PsA 1 (NCT03104400; N=1705, non-biologic DMARD IR) and SELECT-PsA 2 (NCT03104374; N=642, biologic DMARD IR), pts with active PsA (≥3 swollen and ≥3 tender joints), active or historical psoriasis, and on ≤2 non-biologic DMARDs were randomized to once daily UPA 15 mg, UPA 30 mg, adalimumab 40 mg every other week (SELECT-PsA 1 only), or PBO. Efficacy was assessed in pts with axial involvement (diagnosed by investigators based on totality of information) pooled from the 2 studies. Assessments included change from BL in BASDAI, BASDAI Q2 (neck/back/hip pain) and Q3 (joint swelling/pain), and the AS Disease Activity Score (ASDAS-CRP), and percentage with BASDAI 50 response, ASDAS inactive disease (ID), ASDAS low disease activity (LDA), ASDAS major improvement (MI), and ASDAS clinically important improvement (CII). Uveitis and inflammatory bowel disease (IBD) adverse events were reviewed. Data on 24-week PBO-controlled period are presented. Results:Prevalence of axial involvment was 31.3% in SELECT-PsA 1 and 34.2% in SELECT-PsA 2 (Table). Treatment with UPA 15 mg and 30 mg resulted in significantly greater improvements from BL in the BASDAI, BASDAI Q2 (neck/back/hip pain) and Q3 (joint swelling/pain) and ASDAS-CRP at weeks 12 and 24 vs PBO (Figure). Similarly, significantly higher percentages of pts on UPA 15 mg and 30 mg achieved BASDAI 50, ASDAS ID, LDA, MI, and CII at weeks 12 and 24 vs PBO (Figure). One pt on UPA 30 mg had incident uveitis, and no IBD was reported on UPA.Table 1.Demographics and Baseline CharacteristicsSELECT-PsA 1SELECT-PsA 2Parameter, mean (SD)With Psoriatic Spondylitis(n=534)Without Psoriatic Spondylitis(n=1170)Pvalue*With Psoriatic Spondylitis(n=219)Without Psoriatic Spondylitis(n=421)Pvalue*BMI (kg/m2)29.9 (6.5)30.5 (6.9).081031.6 (8.0)31.3 (6.9).6226TJC6821.6 (15.1)19.2 (13.5).002227.5 (18.0)23.3 (16.2).0027SJC6611.7 (9.4)11.0 (7.9).118412.9 (9.2)11.7 (8.7).0804Physician’s Global Assessment (NRS 0–10)6.7 (1.6)6.5 (1.7).04376.6 (1.8)6.5 (1.7).1897HAQ-DI1.2 (0.6)1.1 (0.6).01701.2 (0.6)1.2 (0.7).2049n=531n=1164n=218n=416Presence of dactylitis, n (%)†188 (35.2)328 (28.0).002869 (31.5)100 (23.8).0348Presence of enthesitis, n(%)‡432 (80.9)884 (75.6).0147189 (86.3)337 (80.0).0125ASDAS–CRP3.4 (0.9)3.1 (1.0)<.00013.3 (1.0)3.2 (1.1).1032n=530n=1161n=217n=416BASDAI5.8 (2.0)5.3 (2.2)<.00016.2 (2.2)5.8 (2.2).0673n=530n=1161n=217n=416Morning Stiffness Duration (NRS 0–10; BASDAI Q6)5.0 (3.0)4.7 (3.0).03685.6 (3.2)5.1 (3.0).0454n=530n=1161n=217n=416Patient’s Assessment of Inflammatory Neck, Back, or Hip Pain (NRS 0–10; BASDAI Q2)5.8 (2.7)4.6 (3.2)<.00016.4 (2.8)5.4 (3.1).0001n=530n=1161n=217n=416*Calculated by t-test for continuous variables and chi-square test for categorical values.
