We report that Ges (GTPase regulating endothelial cell sprouting), a human RGK protein expressed in the endothelium, functions as a potent morphogenic switch in endothelial cells (ECs). Ges function is sufficient to substitute for angiogenic growth factor/extracellular matrix (ECM) signals in promoting EC sprouting, since overexpression of Ges in ECs cultured on glass leads to the development of long cytoplasmic extensions and reorganization of the actin cytoskeleton. Ges function is also necessary for Matrigel-induced EC sprouting, since this event is blocked by its dominant negative mutant, GesT94N, predicted to prevent the activation of endogenous Ges through sequestration of its guanine nucleotide exchange factor. Thus, Ges appears to be a key transducer linking extracellular signals to cytoskeleton/morphology changes in ECs.

Abstract:
Background:Chronic inflammatory arthritis is a hallmark of Ankylosing Spondylitis (AS), where co-existence of inflammatory bowel disease, such as Crohn’s Disease (CD) is prominent. The clinical overlap of AS and CD has raised the hypothesis that these conditions may have similar pathophysiological mechanisms. Both indications are characterized by an altered extracellular matrix turnover, where particularly collagens are remodeled.Objectives:We investigated the association between biomarkers of collagen degradation in healthy controls and patients with AS, CD and AS/CD overlap, with the aim to investigate the biomarkers’ ability to identify patients with AS/CD overlap.Methods:Patients with AS fulfilling ASAS criteria (n=13), biopsy-proven CD (n=14), subjects with AS and CD overlap (n=10) and healthy controls (n=11) undergoing standard of care colonoscopies were included in the study. The collagen degradation biomarkers measuring type III, IV, VI and X collagen (C3M, C4M, C6M and C10C, respectively) were measured in EDTA plasma samples from all subject groups. Biomarkers were measured by competitive ELISAs. Statistical analysis was performed using an ANCOVA adjusted for age, an AUROC analysis and spearman correlations.Results:The collagen biomarker C4M was significantly higher in patients with AS/CD overlap compared to AS, CD and HCs (all p<0.0001, Figure 1A). The blood levels of C4M in AS patients were significantly lower than HC (p=0.0003), while CD also showed a lower level compared to HC though not significant (p=0.0798). No difference was found between AS and CD alone. In an AUROC analysis, C4M showed a complete separation between the patients with AS/CD overlap compared to HC, AS and CD with an AUC=1.00; p=0.0001. No differences were found between the patient groups for C3M, C6M and C10C (Figure 1, B-D). 91.3 % of patients with AS, 92.8% of patients with CD and 60 % of patients with AS and IBD overlap were actively treated with TNF-α inhibitors, which may explain the suppression of the collagen degradation biomarker levels in AS, CD and AS/IBD overlap compared to healthy controls[1,2]. No correlations were found between the collagen biomarkers and CRP, BASDAI, SCCAI or HBI scores.Conclusion:Degradation of type IV collagen quantified by C4M showed a complete separation of patients with AS/IBD overlap, compared to AS, CD and HC patients, which indicates an excessive collagen degradation and epithelial turnover. Det er receptionsforskningens uomtvistelige fortjeneste, at den har sat focus på modtagernes oplevelser og vurderinger. Brugerne tages tilsyne- ladende alvorligt, i modsætning til den kritiske teori (Frankfurterskolen m.v.), der i debatten om receptionsforskningen er blevet beskyldt for at være formynderisk: Den tager stilling til æstetisk og indholdsmæssig kvalitet på brugernes vegne. I denne artikel forsøger Ib Bondebjerg at fastholde en del næsten glemte, men fundamentale nuancer (ikke mindst mht. tekstens status i forhold til oplevelsen) i både den kritiske teori, semiotikken og dele af receptions- forskningen. Hermed bidrages til den diskussion om hvem der skal fast- lægge den kulturelle og æstetiske værdi i et medieprodukt som tages op af både Jensen, Schrøder og Skovmand i dette nummer. Indirekte viser Bondebjerg, at der sker en afpolitisering af medierne, hvis de alene eksisterer i en uproblematiseret relation mellem afsender og modtager. Populært TV konstrueres jo netop så det fremtræder lyst- og spændings- fyldt og kommer herved til at virke som et passivt spejl for modtagerne. Hvis kunsten forråder det smertefulde og tabuiserede, bliver den ufarlig. Mediernes eneste formål bliver da at igangsætte et på forhånd kalkuleret ping-pong-spil mellem den enkeltes erfaringer, oplevelser og fantasier. Heroverfor stiller Bondebjerg den demokratiske radikalisme hvor æstetik- ken udgør et kampområde. Kunsten er med andre ord en politisk agent, hvor både afsenderen, modtageren og samfundet selv aktivt er med i forandringsprocessen. - "Perspektivet er at gøre modtagerne til kvalifi- cerede producenter i videste forstand".

Artistically written recursions of Joseph Smith’s first vision also began to appear in the 1880s, particularly through George Cannon and Andrew Jenson. These stories, while not intended to be historical representations, reflected and contributed to the recursion of collective memory and its transmission to the next generation. Distortion, while inevitable with retellings, occurs as the remembering entity favors coherence over accuracy. Forging collective memory from art, music, and print created by a shared memory and a multitude of individual ones, each inevitably distorted compared to Joseph Smith’s own memories of his first vision. But for all that, the collective memory the saints forged became incredibly strong, resilient, and extremely well known to them.

Cath Moore draws attention to the unique Danish co-writing process that most Danish directors employ, in which a director and screen-writer collaborate on scripts and share writing credit. Bier’s most successful films have been co-written with Anders Thomas Jensen, who also works with actors during the writing process, rewriting lines of story and dialogue after collaborative reading sessions. With a focus on the Bier/Jensen trilogy, Moore delineates the creative divergences between Bier’s solo work and Jensen’s dark satirical and absurdist self-written and directed films. "of Denmark’s most prominent and celebrated screenwriters, Anders Thomas Jensen began his career making short films, receiving three Oscar nominations in the late 1990s in the process. He won on his third try, for the 1999 comedy Election Night, which starred Ulrich Thomsen, who would go on to appear in other Jensen-written projects like Adam’s Apples (2005) and In a Better World (2010). Eventually, Jensen moved on to features, writing and directing the dark comedies Flickering Lights (2000), The Green Butchers (2003), and Adam’s Apples, all of which starred his Danish compatriot Mads Mikkelsen. But perhaps Jensen is best known to worldwide audiences for his ongoing collaboration with director Susanne Bier, who has directed his screenplays for Open Hearts (2002), Brothers (2004), After the Wedding (2006), In a Better World, and Love Is All You Need (2012). As opposed to Jensen’s solo films, his work with Bier has been marked by its emotionally charged, intensely dramatic scenarios, often dealing in themes of violence, reconciliation, and coincidence. Brothers won the Audience Award at the Sundance Film Festival, and In a Better World earned both a Best Foreign Language Film Oscar and a Golden Globe. Jensen, who lives in Copenhagen, was also an early participant in the Dogme film movement, writing the script for both Søren Kragh-Jacobsen’s Mifune (1999) and Open Hearts. His other credits range from the romantic comedy-drama Wilbur Wants to Kill Himself (2002) to the period costume drama The Duchess (2008). Mit Blick auf Deutschland steht außer Frage, dass die im bestehenden föderativen System enthaltene Garantie der kommunalen Selbstverwaltung ein hinreichendes Maß an dezentraler Finanzautonomie voraussetzt. Die Bewältigung der den Kommunen im Rahmen der föderalen Zuständigkeitsverteilung zugewiesenen Aufgaben und das Tätigen der damit verbundenen Ausgaben sind ohne eine entsprechende Ausstattung mit Einnahmen, d.h. der Verfügbarkeit über angemessene finanzielle Mittel zur Aufgabenerfüllung, nicht möglich. Diese grundlegende Einsicht spiegelt sich bereits in Art. 28 Abs. 2 Grundgesetz (GG), wonach den Gemeinden und Gemeindeverbänden das Recht gewährleistet sein muss, „alle Angelegenheiten der örtlichen Gemeinschaft im Rahmen der Gesetze in eigener Verantwortung zu regeln“. Und weiter heißt es dort: „Die Gewährleistung der Selbstverwaltung umfasst auch die Grundlagen der finanziellen Eigenverantwortung“. Dieser im Rahmen der Grundgesetzanpassung von 1994 hinzugefügte Satz stellt nicht allein eine verfassungsrechtliche Grundlage für die Zuweisung eigener Steuerhoheiten an die kommunale Ebene durch den Bundesgesetzgeber dar. Er ist zugleich auch die grundgesetzliche Basis einer garantierten (vertikalen) Zuweisung von finanziellen Mitteln eines jeweiligen Landes an seine Kommunen und bildet damit den rechtlichen Bezugspunkt für den auf Landesebene bestehenden kommunalen Finanzausgleich, dessen Durchführung in den jeweiligen Landesverfassungen geregelt ist. Vor diesem Hintergrund hat auch der Verfassungsgerichtshof Rheinland-Pfalz in seinem Urteil vom 14. Februar 2012 im Rahmen des Normenkontrollverfahrens zur Änderung des Landesfinanzausgleichsgesetzes vom 12. Juni 2007 zum einen auf die Verpflichtung des Landes aus Art. 49 Abs. 6 LV hingewiesen, über den kommunalen Finanzausgleich eine „angemessene Finanzausstattung“ der Kommunen zur Erfüllung pflichtiger wie freiwilliger Selbstverwaltungsaufgaben zu gewährleisten. Zugleich wird vom Verfassungsgerichtshof zum anderen aber auch darauf verwiesen, dass als wesentliche Grundlage für einen funktionsfähigen Finanzausgleich „die Kommunen ihre eigenen Einnahmequellen angemessen auszuschöpfen und Einsparpotentiale bei der Aufgabenwahrnehmung zu verwirklichen“ haben. Als unmittelbare Folge aus dieser vom Verfassungsgerichtshof formulierten Anforderung an die kommunale Einnahmenpolitik kann das Land „im Gegenzug für seinen Beitrag zur Bewältigung der kommunalen Finanzkrise verlangen, dass auch die Kommunen ihre Kräfte größtmöglich anspannen“ (ebenda). Auch wenn die zurückliegend formulierten Anforderungen auf sämtliche Kommunen in Deutschland zutreffen, soll im vorliegenden Beitrag beispielhaft die aktuelle Klage eines Teils der Kommunen des Landes Rheinland-Pfalz, der zufolge die Regelungen des Landesfinanzausgleichsgesetzes für das Jahr 2014 als nicht ausreichend gelten, um die Vorgaben aus dem Urteil des Verfassungsgerichtshofs vom 14. Februar 2012 zu erfüllen, zum Anlass für eine entsprechende Überprüfung der Einnahmenpolitik der Städte und Gemeinden des Landes genommen werden. Hierbei wird sich auf eine Analyse der bestehenden Finanzierungsspielräume im Bereich der sogenannten Realsteuern (Gewerbesteuer, Grundsteuer B, Grundsteuer A) als jenen kommunalen Einnahmequellen konzentriert, die bekanntermaßen mit Abstand zu den bedeutendsten Einnahmequellen von Städten und Gemeinden zählen, die von diesen eigenverantwortlich gestaltet werden können. Ziel der nachfolgenden Untersuchung ist folglich die Fragestellung, ob die rheinland-pfälzischen Kommunen die ihnen im Rahmen der Realsteuerpolitik zur Verfügung stehenden Möglichkeiten zur Finanzierung ihrer Haushalte in angemessenem Umfang in den zurückliegenden Jahren ausgeschöpft haben. Als Betrachtungszeitraum für die empirische Analyse werden dabei die Jahre 2007 bis 2013 zugrunde gelegt, wobei immer dann, wenn Daten der amtlichen Statistik auch für 2014 bereits verfügbar sind, diese in die Betrachtung mit einbezogen werden.
