Abstract:
The angiopoietin (Ang)/Tie2 system is exclusively involved in vasculogenesis and angiogenesis. Ang2 is known to inhibit Ang1-mediated phosphorylation of Tie2 as well as cellular responses during embryonic development. Recent studies have demonstrated that Ang2 has angiogenic activities in adult tissues and cultured endothelial cells. In the present study, we examined the downstream signaling pathways involved in Ang2-mediated cellular responses by murine brain capillary cell line, IBE cells. Tie2 was tyrosine phoshorylated by Ang2. Ang2 showed no effect on proliferation, but stimulated chemotaxis and tube-like structure formation. Phosphoinositide 3-kinase (PI 3-kinase) was activated by Ang2 through c-Fes and was involved in chemotaxis toward Ang2. Ang2 also activated c-Fyn in IBE cells. Cells expressing kinase-inactive c-Fyn attenuated Ang2-induced tube formation, suggesting that c-Fyn was responsible for Ang-2-mediated tube formation. Collecting these data, Ang2 activates c-Fes and c-Fyn, leading to migration and tube formation by murine capillary endothelial cells.

Abstract Communication between endothelial cells (ECs) and mural cells is critical in vascular maturation. Genetic studies suggest that angiopoietin/Tie2 signaling may play a role in the recruitment of pericytes or smooth muscle cells (SMCs) during vascular maturation. However, the molecular mechanism is unclear. We used microarray technology to analyze genes regulated by angiopoietin-1 (Ang1), an agonist ligand for Tie2, in endothelial cells (ECs). We observed that hepatocyte growth factor (HGF), a mediator of mural cell motility, was up-regulated by Ang1 stimulation. We confirmed this finding by Northern blot and Western blot analyses in cultured vascular endothelial cells. Furthermore, stimulation of ECs with Ang1 increased SMC migration toward endothelial cells in a coculture assay. Addition of a neutralizing anti-HGF antibody inhibited Ang1-induced SMC recruitment, indicating that the induction of SMC migration by Ang1 was caused by the increase of HGF. Interestingly, Ang2, an antagonist ligand of Tie2, inhibited Ang1-induced HGF production and Ang1-induced SMC migration. Finally, we showed that deletion of Tie2 in transgenic mouse reduced HGF production. Collectively, our data reveal a novel mechanism of Ang/Tie2 signaling in regulating vascular maturation and suggest that a delicate balance between Ang1 and Ang2 is critical in this process.

Abstract Context: Although the inner fetal zone (FZ) of the midgestation human fetal adrenal (HFA) produces dehydroepiandrosterone sulfate, the function of the outer definitive zone (DZ) remains less clear. We have proposed that the DZ phenotype is that of a pool of progenitor cells, many of which are mitotically active. Recently, we studied HFA expression of a family of vascular endothelial cell-specific angiogenic factors, the angiopoietins (Angs), and demonstrated that Ang2 was localized predominantly in the periphery of the gland. Ang1 stabilizes, whereas Ang2 destabilizes, vessels, increasing responsiveness to angiogenic stimuli such as vascular endothelial growth factor (VEGF)-A and fibroblast growth factor (FGF)-2. Objective: Our objective was to test the hypothesis that the periphery of the HFA is a site of angiogenesis. Design: Studies were conducted involving RNA, frozen sections, and primary cell cultures from midgestation HFAs. Main Outcome Measures: Immunofluorescence, laser capture microdissection, and real-time quantitative RT-PCR were used. Results: Double immunostaining demonstrated that proliferating endothelial cells were limited to the DZ and DZ/FZ border. Ang2 mRNA was primarily expressed in the DZ, whereas Ang1 mRNA was primarily in the FZ. VEGF-A and FGF-2 mRNA levels were higher in the DZ. FGF-2 (10 ng/ml) induced Ang2 mRNA by 4-fold in both zones of cells (P < 0.01, at 24 h), but not Ang1 or VEGF-A mRNA. Conclusion: Data suggest that angiogenesis occurs at the periphery of the HFA. The DZ-predominant expression of Ang2 may be explained, in part, by the parallel pattern of FGF-2 expression.

OBJECTIVES/GOALS: Lack of blood vessels remains a major obstacle in tissue regeneration. Angiopoietin 1 and 2 modulate angiogenesis through the Tie2 receptor tyrosine kinase. Ang1 activates pAKT to promote endothelial cell survival while Ang2 antagonizes these effects. We aim to dissect the Ang/Tie2 pathway to uncover the molecular basis for these opposing effects. METHODS/STUDY POPULATION: Ang1 and Ang2 bind Tie2 via nearly identical F-domains (Fd). To investigate the molecular basis regulating the Tie2 pathway, we generated a series of computationally designed self-assembling protein scaffolds presenting F-domains in a wide range of valencies and geometries using Rosette Molecular Modeling Suite. We examined the protein kinase activation, cell migration, and blood vessel formation produced by the designed proteins in human umbilical vein endothelial cells. RESULTS/ANTICIPATED RESULTS: Two phenotypic classes were demonstrated by the number of presented F domains: scaffolds presenting 3 or 4 Fd have Ang2 like activity, upregulating pFAK and pERK but not pAKT and failing to induce cell migration and tube formation; scaffolds presenting more than 6 Fd have Ang1 like activity, upregulating the three signaling branches and enhancing cell migration and tube formation. Scaffolds with 8 or more Fd show superagonist activity, producing significantly stronger phenotypes than Ang1. These results suggest that Fd valency largely determines Ang1 vs Ang2 signaling outcomes, and our designed superagonists can outperform Ang1 in vascularization and wound healing. In in vivo experiments, nanoparticles displaying 60 copies of Fd produce significant revascularization in hemorrhagic brains. DISCUSSION/SIGNIFICANCE OF IMPACT: Targeting the Tie2 pathway is a new paradigm in regenerative medicine. Our designed constructs will enable us to generate high-affinity Tie2 agonists and antagonists as drugs to control angiogenesis, enabling tissue regeneration that recapitulates the biological architecture of the native tissue physiology, improving organ transplant outcome.

ABSTRACT Angiopoietin 2 (Ang2) was originally shown to be a competitive antagonist for Ang1 of the receptor tyrosine kinase Tie2 in endothelial cells (ECs). Since then, reports have conflicted on whether Ang2 is an agonist or antagonist of Tie2. Here we show that Ang2 functions as an agonist when Ang1 is absent but as a dose-dependent antagonist when Ang1 is present. Exogenous Ang2 activates Tie2 and the promigratory, prosurvival PI3K/Akt pathway in ECs but with less potency and lower affinity than exogenous Ang1. ECs produce Ang2 but not Ang1. This endogenous Ang2 maintains Tie2, phosphatidylinositol 3-kinase, and Akt activities, and it promotes EC survival, migration, and tube formation. However, when ECs are stimulated with Ang1 and Ang2, Ang2 dose-dependently inhibits Ang1-induced Tie2 phosphorylation, Akt activation, and EC survival. We conclude that Ang2 is both an agonist and an antagonist of Tie2. Although Ang2 is a weaker agonist than Ang1, endogenous Ang2 maintains a level of Tie2 activation that is critical to a spectrum of EC functions. These findings may reconcile disparate reports of Ang2's effect on Tie2, impact our understanding of endogenous receptor tyrosine kinase signal transduction mechanisms, and affect how Ang2 and Tie2 are targeted under conditions such as sepsis and cancer.

OBJECTIVE: We aimed to investigate the effects of hydroxyapatite bioceramic extract on Ang/Tie2 system and cell proliferation of umbilical vein endothelial cells. METHODS: Human umbilical vein endothelial cells (HUVECs) were used in this research. There are two induvial groups, control group and hydroxyapatite bioceramics extract treatment group. Cell Counting Kit-8 (CCK-8) was used to evaluate cell proliferation. Western blot and real time quantitative PCR (Q-PCR) were used to evaluate the protein and mRNA expression levels of Ang1, Ang2 and Tie2 in Ang/Tie2 system, respectively. All the results were statistically analyzed by Spss19.0. All data were presented as mean ± standard error of mean (SEM). Student’s t-test was performed to determine the differences among grouped data. RESULTS: Hydroxyapatite bioceramics extract showed no effect on the cell morphology and cell proliferation of HUVECs. Interestingly, we found that both Ang2 and Tie2 protein and mRNA level were markedly increased by hydroxyapatite bioceramics extract. CONCLUSIONS: Hydroxyapatite bioceramic extract showed no cytotoxicity to HUVECs, and might regulate vascular remodeling by mediating Ang/Tie2 system.

Patients with Stage 5 chronic kidney disease who are on hemodialysis (HD) remain in a chronic inflammatory state, characterized by the accumulation of uremic toxins that induce endothelial damage and cardiovascular disease (CVD). Our aim was to examine microvesicles (MVs), monocyte subpopulations, and angiopoietins (Ang) to identify prognostic markers in HD patients with or without diabetes mellitus (DM). A total of 160 prevalent HD patients from 10 centers across Spain were obtained from the Biobank of the Nephrology Renal Network (Madrid, Spain): 80 patients with DM and 80 patients without DM who were matched for clinical and demographic criteria. MVs from plasma and several monocyte subpopulations (CD142+/CD16+, CD14+/CD162+) were analyzed by flow cytometry, and the plasma concentrations of Ang1 and Ang2 were quantified by ELISA. Data on CVD were gathered over the 5.5 yr after these samples were obtained. MV level, monocyte subpopulations (CD14+/CD162+ and CD142+/CD16+), and Ang2-to-Ang1 ratios increased in HD patients with DM compared with non-DM patients. Moreover, MV level above the median (264 MVs/µl) was associated independently with greater mortality. MVs, monocyte subpopulations, and Ang2-to-Ang1 ratio can be used as predictors for CVD. In addition, MV level has a potential predictive value in the prevention of CVD in HD patients. These parameters undergo more extensive changes in patients with DM.

Even when patients with nonsmall cell lung cancer undergo surgical resection at an early stage, recurrent disease often impairs the clinical outcome. There are numerous causes potentially responsible for a relapse of the disease, one of them being extensive angiogenesis. The balance of at least two systems, VEGF VEGFR and Ang Tie, regulates vessel formation. The aim of this study was to determine the impact of surgery on the plasma levels of the main angiogenic factors during the first month after surgery in nonsmall cell lung cancer patients. The study group consisted of 37 patients with stage I nonsmall cell lung cancer. Plasma concentrations of Ang1, Ang2, sTie2, VEGF, and sVEGF R1 were evaluated by ELISA three times: before surgical resection and on postoperative days 7 and 30. The median of Ang2 and VEGF concentrations increased on postoperative day 7 and decreased on day 30. On the other hand, the concentration of sTie2 decreased on the 7th day after resection and did not change statistically later on. The concentrations of Ang1 and sVEGF R1 did not change after the surgery. Lung cancer resection results in proangiogenic plasma protein changes that may stimulate tumor recurrences and metastases after early resection.

Colorectal cancer liver metastases (CRCLM) that receive their blood supply via vessel co-option are associated with a poor response to anti-angiogenic therapy. Angiopoietins (Ang1 and Ang2) with their Tyrosine-protein kinase receptor (Tie2) have been shown to support vessel co-option. We demonstrate significantly higher expression of Ang1 in hepatocytes adjacent to the tumor region of human chemonaïve and treated co-opting (replacement histopathological growth patterns: RHGP) tumors. To investigate the role of the host Ang1 expression, Ang1 knockout (KO) mice were injected intra-splenically with metastatic MC-38 colon cancer cells that develop co-opting liver metastases. We observed a reduction in the number of liver metastases and interestingly, for the first time, the development of angiogenic driven desmoplastic (DHGP) liver metastases. In addition, in-vitro, knockout of Ang1 in primary hepatocytes inhibited viability, migration and invasion ability of MC-38 cells. We also demonstrate that Ang 1 alone promotes the migration and growth of both human and mouse colon cancer cell lines These results provide evidence that high expression of Ang1 in the host liver is important to support vessel co-option (RHGP lesions) and when inhibited, favours the formation of angiogenic driven liver metastases (DHGP lesions).

Cette etude est basee sur la correspondance de michel-ange a partir de l'edition posthume de g. Poggi qui comporte mille quatre cent quatre lettres reparties sur cinq volumes. La premiere partie de l'etude s'attache a examiner en details le corpus, en etablissant un historique de l'edition de cette correspondance, en se penchant sur les manuscrits pour distinguer les originaux des brouillons et copies de lettres. Il s'agit ensuite d'examiner les criteres de retranscription des editeurs et d'etablir une chronologie de la correspondance, d'examiner sa repartition et les differents correspondants de l'artiste. La deuxieme partie de cette etude analyse les lettres en degageant les informations qui permettent d'avoir une vision de l'homme michel-ange. Sont etudiees tout d'abord sa vie quotidienne et sa sante, puis comment la correspondance eclaire l'etude des relations qu'il entretenait avec sa famille, pour l'essentiel avec son neveu leonardo, puis avec son pere lodovico et avec ses freres. D'autres elements dans les lettres apportent des renseignements sur l'entourage de michel-ange : les relations qu'il avait avec ses anis, ainsi que sa conception du monde : ses idees sur la religion, sur la vie culturelle et politico-sociale de son temps. La troisieme partie analyse les informations apportees sur l'artiste. This study is based on g. Poggi's posthumous edition of the correspondence of michelangelo, which includes one thousand four hundred letters in five volumes. The first part of the study is a detailed examination of the corpus, and presents a historical account of the publication of this correspondence while giving particular consideration to the manuscripts in order to distinguish originals from drafts and copies. Then the criteria used by publishers to decide what was to be printed are studied, the correspondence is sorted chronologically, the way the letters are spread over the years is examined, and the artist's different correspondents are reviewed. The second part of this study is an analysis of the information contained in the letters about the sort of man michelangelo was. First of all his everyday life and his health are investigated, and then what his correspondence tells us about his relations with his family, essentially with his nephew, leonardo, and with his father, lodovico, and his brothers. Other elements contained in the letters tell us about the people michelangelo was acquainted with, such as his relations with his friends, or throw light on the way he viewed the world, such as his ideas on religion, and on the cultural and politico-social life of his day. The third part analyzes information concerning the artist. First the relations he had with the various people who commissioned his works are examined, then the way michelangelo worked with other artists, as well as with his different collaborators - such as stonecutters - and with his assistants

SANTRAUKA Serologinių sifilio ir Laimo boreliozės reakcijų kryžminis reaktyvumas Darbo autorė: Tatjana Subočiūtė Darbo vadovas: med. m. dr. Andrius Vagoras Darbo tikslas: nustatyti VULSK diagnostikos centre rutiniškai naudojamų serologinių sifilio ir Laimo boreliozės testų galimai klaidingai teigiamų rezultatų dažnumą dėl kryžminio viena iš šių infekcijų užsikrėtusiųjų pacientų kraujo serumų reaktyvumo. Darbo metodai: Tyrimai buvo atlikti Vilniaus universiteto ligoninės Santariškių klinikos (VULSK) Laboratorinės diagnostikos centro Mikrobiologijos laboratorijos Infekcijų žymenų tyrimų padalinyje. Serumai buvo surinkti ir ištirti nuo 2005 m. vasario iki 2006 m. kovo mėn. Tyrimo medžiagą sudarė 40 serumų. 23 serumai dėl sifilio tyrimų buvo gauti iš VULSK Dermatovenerologijos centro. 8 serumai dėl Laimo ligos tyrimų buvo gauti iš VULSK Dermatovenerologijos centro. UAB „Endemik” atrinkti 9 serumai ištirti dėl Laimo ligos. Visi į tyrimą įtraukti serumai buvo ištirti šiomis reakcijomis: 1) RPR su titru, 2) TPHA, 3) Treponema pallidum ELISA, 4) Treponema pallidum imunofluorescencija, 5) Borrelia burgdorferi imunofluorescencija. Tyrimo rezultatai: Ištyrus penkiomis reakcijomis serumus (RPR, TPHA, Treponema pallidum IFR, Treponema pallidum IFA bei Borrelia burgdorferi IFR), buvo pastebėtas kryžminis serologinis reaktyvumas tarp Borrelia burgdorferi IFR ir Treponema pallidum IFR reakcijų. Mūsų darbo duomenimis, tiriant 17 serumų (teigiamus dėl Laimo ligos) kryžminis reaktyvumas... [toliau žr. visą tekstą]
SUMMARY Cross - reactivity in serological test for Lyme disease and syphilis Author: Tatjana Subočiūtė Supervisor: Andrius Vagoras, MD Work purpose: to determine the incidence of possibly false positive results of serological syphilis and Lyme borreliosis tests, routinely used in VULSK Diagnostics centre, occurring because of cross reactivity of patients, having one of these disease, blood. Work methods: the research was carried out in Vilnius University Hospital Santariškių Klinikos (VULSK) Infection Markers Research Department of Microbiological Laboratory in Laboratory Diagnostics Centre. Serum samples were collected and tested from February, 2005, till March, 2006. The investigation material was composed of 40 serum samples. 23 serum samples for syphilis test came from VULSK Centre of Dermatovenereology. 8 serum samples for Lyme borreliosis test were received from VULSK Centre of Dermatovenereology. 9 serum samples for Lyme borreliosis test were picked by UAB “Endemik”. All the samples, included into the research, were tested by following reactions: 1) RPR with titer, 2) TPHA, 3) Treponema pallidum ELISA, 4) Treponema pallidum immunofluorescence, 5) Borrelia burgdorferi immunofluorescence. Abstract:
The recent discovery of angiotensin converting enzyme 2 (ACE2) as a functional peptidase within the renin-angiotensin system (RAS) has added a new layer of complexity to the enzymatic cascade of this hormonal system. ACE2 is highly expressed in the proximal tubules of the kidney, an important tissue site involved in blood pressure regulation. Therefore, we derived a model for the processing of Ang I which is the immediate precursor to the biologically active peptides Ang II and Ang-(1-7) based on metabolism data in isolated proximal tubules of the sheep kidney (1). Given the individual experimental velocities for several peptidases expressed in the proximal tubules including ACE, ACE2 and neprilysin, rate constants were calculated to describe the conservation equations for the processing of Ang I, Ang II and Ang-(1-7) We modeled the system with Ang I as the initial substrate and peptide concentrations for the downstream products were calculated using Euler’s method.

Šiuolaikiniai skaitmeniniai fotoaparatai, išmanieji telefonai ir vaizdo apdorojimo programinė įranga leidžia sukurti aukštos kokybės nuotraukas lengviau nei bet kada. Tokios nuotraukų dalijimosi paslaugos, kaip „Imgur“, „Instagram“, „Flicker“, „Google Photos“ ir t. t., žymiai supaprastino apsikeitimą vaizdine informacija. Neretai viešai prieinama vaizdo medžiaga naudojama žiniasklaidoje, dėl to vis aktualesnė tampa rodomo vaizdo autentiškumo problema. Ar visi vaizdai, kuriuos mes matome kiekvieną dieną, yra nemodifikuoti? Vienas iš labiau paplitusių vaizdo informacijos saugojimo formatų yra JPEG formatas. JPEG (angl. Joint Photographic Experts Group) glaudinimo algoritmas skirtas statinių spalvotų vaizdų byloms glaudinti. Formato charakteristikų ir savybių supratimas gali būti naudingas atliekant skaitmeninio vaizdo ekspertizę. Šiame straipsnyje bus aptartas JPEG skaitmeninių vaizdų autentiškumo nustatymo metodas taikant klaidų lygių analizę (angl. ELA – Error Level Analysis).

Abstract:
Adsorbed natural gas (ANG) has been emerging as an attractive alternative to compressed natural gas and liquefied natural gas, on various circumstances. In spite of the advantages associated with ANG over other storage modes, there are some issues that need be properly addressed in order to ensure a viable employment of such alternative. One major problem is that the thermal effects associated with the sorption phenomena tend to diminish the storage capacity, thereby resulting in poorer performance. Hence, in order to design commercially viable storage vessels, the heat and mass transfer mechanisms that occur in these devices must be carefully understood and controlled. In this context, this work presents different mathematical models for simulating ANG charge and discharge processes. For the sake of improving the understanding of mass and energy transport within ANG vessels, dimensionless groups associated with this problem are also discussed. By using the proposed formulations, simulation results were numerically calculated and the process of charging ANG vessels was analyzed for different combinations of the presented dimensionless groups. Pressure, temperature and stored mass histories are compared, showing that the unwanted heating effects can be minimized for certain values of the dimensionless groups. Finally, a comparison between the results obtained using the two different formulations suggests that there may be cases in which the simpler model could be employed.

V prispevku smo se osredotočili na časovne razpo-rejevalnike (ang. cron job schedulers) v brezstre-žniškem (ang. serverless) okolju in njihovo zane-sljivo uporabo. Primerjali smo razporejevalnike, implementirane s pomočjo zabojnikov s tistimi, ki so gostovani v oblaku z uporabo pristopa funk-cije kot storitve. Ugotavljali smo, katere so po-sebnosti časovnih razporejevalnikov v brezstrežni-škem okolju in kdaj je le-te sploh smiselno upo-rabiti. Na praktičnem primeru smo predstavili, kako jih lahko vključimo v večji sistem in na ka-kšen način najlažje rešimo morebitne težave, ki jih ob izbiri brezstrežniškega okolja zavestno pre-vzamemo. Ugotovili smo, da so razporejevalniki v FaaS (ang. Function as a Service) okolju naj-primernejši zaradi enostavnega in hitrega razvoja ter nizkih stroškov obratovanja.
