The human immunodeficiency virus, in addition to its profound effect on the immune system, also causes degenerative changes in the brain, the spinal cord and peripheral nerves. The first and rate-limiting step of the kynurenine pathway is the metabolism of tryptophan (Trp) to N-formylkynurenine, which is then rapidly converted to kynurenine. This initial step can be catalysed by three enzymes, tryptophan 2,3-dioxygenase (TDO), indoleamine 2, 3-dioxygenase-1 (IDO1) and the most recently discovered, IDO2. In the first experiment, the response to subcutaneous injection of various doses of bee venom into the hind paw of the rat was quantified. We evaluated the suitability of three different experimental animal models for studying changes in the nociceptin system; lipopolysaccharide (LPS)-induced sepsis, faecal slurry-induced sepsis and caecal ligation and puncture-induced sepsis.
