The neuropathogenicity of two retroviruses was investigated. The human immunodeficiency virus, in addition to its profound effect on the immune system, also causes degenerative changes in the brain, the spinal cord and peripheral nerves. The first and rate-limiting step of the kynurenine pathway is the metabolism of tryptophan (Trp) to N-formylkynurenine, which is then rapidly converted to kynurenine. This initial step can be catalysed by three enzymes, tryptophan 2,3-dioxygenase (TDO), indoleamine 2, 3-dioxygenase-1 (IDO1) and the most recently discovered, IDO2. The present study describes a new test of tonic pain in rats which can be used as an animal model of persistent pain. In the first experiment, the response to subcutaneous injection of various doses of bee venom into the hind paw of the rat was quantified.
