The stretch reflex is an involuntary contraction produced in response to muscle stretch. Despite extensive research, its role in the control of movement and posture remains controversial. One reason for this is that the torque output produced by reflex activity has been studied in only a limited number of tasks and behaviours. Previous studies of the human stretch reflex have been limited by difficulties separating the reflex response from those due to passive and intrinsic joint properties. This study has presented and experimentally verified a method for separately determining these contributions to the human ankle response, and used this technique to assess the reflex contribution to joint dynamics during the application of a stochastic position perturbation. A non-linear parallel-cascade system identification technique was recently developed to non-invasively elucidate the ankle reflex dynamics. Muscle fatigue may be defined as any exercise-related reduction in the force-generating capacity of a muscle. Its effect on joint stiffness is not well understood. The purpose of this study was: (1) to characterize triceps surae muscle fatigue, and (2) to determine its effect on the dynamic stiffness of the ankle joint. This thesis developed new pupillometry paradigms to objectively measure melanopsin function in humans with healthy or diseased eyes. It was determined that melanopsin function is independent of age and refractive error. The novel quadrant field pupil protocols detected melanopsin dysfunction in the very early stages of glaucoma (in suspects) when it is difficult to detect visual dysfunction with conventional ophthalmic tests.
