Sia nell'attività legislativa che nelle decisioni giudiziarie relative alla valorizzazione dei diritti fondamentali, gli anni 1960-1979 coincidono con una fase di grandi innovazioni. Nonostante le fragili maggioranze parlamentari, in quel periodo vengono emanate una lunga serie di disposizioni legislative di ampio respiro nel campo delle libertà civili e dei diritti sociali: lavoratori, donne, malati mentali e altre minoranze trascurate godono di diritti individuali e beneficiano dell'istituzione di un assetto istituzionale più coerente dei servizi pubblici in molti settori. Il saggio indaga queste evoluzioni muovendo dall'analisi di cinque diversi cluster di diritti (lavoratori, famiglia, procedura penale, diritto alla salute e riforme settoriali) e cerca di evidenziare sia le luci che le ombre di tale processo di riforma.

Nel corso degli ultimi 100 anni l'azione politica del Messico ha attraversato cinque fasi: i primi decenni del ventesimo secolo con l'attuazione di interventi finalizzati a dissuadere i potenziali migranti dal lasciare il Messico; una politica di negoziazione durante e dopo la seconda guerra mondiale; un approccio orientato al "laissez faire" negli anni '70 ed '80, una politica della "limitazione dei danni" negli anni '90; infine le proposte ed i discorsi centrati sulla "responsabilitŕ condivisa". Questo contributo esamina ogni fase e come queste abbiano dato forma alle contemporanee modalitŕ di relazione tra gli Usa ed il Messico in relazione ai processi migratori e alla direzione delle politiche migratorie del Messico.

Propongo di considerare le elaborazioni in atto, da alcuni anni, all'interno della comunitŕ bioenergetica, come parte della tradizione relativa al "cambiamento di paradigma" - tradizione iniziata negli anni '70 del secolo scorso - e, insieme, come "discorso di legittimazione", nella cornice della fase storica attuale denominata: "tarda modernitŕ" e "modernitŕ riflessiva". In questa prospettiva, sviluppo alcune riflessioni sia storiche che epistemologiche; in particolare, suggerisco di utilizzare la dizione "filone corporeo" per individuare la corrente psicoterapeutica di cui l'analisi bioenergetica rappresenta la componente piů importante.

Abstract:
In questo elaborato viene analizzato il Kaidan nel cinema horror giapponese dagli anni ’50 agli anni ‘70. L’analisi viene compiuta attraverso i film più noti del periodo, toccando le varie tematiche affrontate in questo lasso di tempo, che vanno dal teatro classico alle varie critiche sociali. In primis viene citata la figura di Nobuo Nakagawa, regista cardine del movimento cinematografico dello psycho-horror, ai cui si ispireranno successivamente molti altri registi sia in Oriente che in Occidente. Viene messa in luce la presenza della derivazione folkloristica delle storie narrate unita ad elementi psicologici in film come “Kaidan Kasane ga Fuchi”, “Borei Kaibyo Yashiki”, “Tokaido Yotsuya Kaidan” e “Jigoku”. Viene poi affrontata la dicotomia cinema-letteratura, con influenze dal teatro Kabuki e No, dove si assiste a pellicole che grazie al loro ibridismo riescono ad ottenere premi di rilevanza internazionale per la prima volta in Giappone. Di particolare rilevanza sono le opere “Kwaidan” e “Ugetsu Monogatari” nelle quali si fondono egregiamente tradizione e innovazione. In seguito, si approfondisce il cinema horror in relazione agli eventi catastrofici avvenuti a causa delle esplosioni nucleari di Hiroshima e Nagasaki. Viene evidenziato come il cinema horror si unisce agli altri generi filmici nella denuncia e condanna degli avvenimenti, primi su tutti sono “Gojira” e “Onibaba” a farsi portavoce di questi eventi. Infine, viene inserito un capitolo riguardante il ritorno del genere horror classico negli anni ‘90, dimenticato negli anni ’80 per lasciare spazio al gore e allo splatter. Analizzando “Ringu” e “Ju-On” si ripercorrono gli elementi classici ritornati in auge e si sottolineano i nuovi temi affrontati, come la denuncia della condizione della donna nella società.

L'oggetto di questo elaborato riguarda i rapporti economici e commerciali instaurati tra la casa automobilistica torinese Fiat e la Cina nel periodo di tempo che va dagli inizi degli anni '70 fino alla fine degli anni '80. Furono proprio quelli gli anni in cui, grazie alle nuove politiche post-maoiste della Cina, venne data una maggiore importanza alla necessità di dare impulso all'industria e alla inevitabile apertura verso gli altri paesi. L'intento, espresso nei tre capitoli, è quello di illustrare la situazione del settore automobilistico in Italia e in Cina tra gli anni '70 e '80 per poi descrivere come si sono svolte le varie missioni avviate reciprocamente dai due paesi per instaurare dei rapporti commerciali.

Abstract:
Gli anni di piombo sono un periodo storico che ha lasciato una traccia indelebile nella società italiana. La presente tesi è un`analisi del modo in cui gli eventi degli anni di piombo vengono rappresentati e riproposti nella cinematografia prodotta nei quarant`anni successivi. Nell`analizzare una così vasta ed eterogenea filmografia si é cercato di individuare l`esistenza di una tendenza di carattere generale che fosse comune alla maggioranza delle pellicole prodotte sull`argomento, in modo da rivelare che tipo di correlazione esiste tra i film analizzati ed il contesto storico, sociale e culturale in cui sono stati prodotti. I film analizzati sono stati raggruppati in tre aree tematiche riguardanti, rispettivamente, i film dedicati al terrorismo di sinistra, quelli dedicati al terrorismo di destra e, infine, la cinematografia che affronta l`eredità lasciata dagli eventi degli anni di piombo sulla società italiana. Nel corso di tale analisi, ciò che è emerso è una produzione cinematografica che risulta estremamente sintomatica del contesto storico e culturale entro il quale è stata prodotta. I film in esame, tutti, seppur in modalità estremamente differenti, alle prese con la ricerca di quel superamento del trauma che gli eventi degli anni di piombo avevano causato, risultano infatti sintomatici delle circostanze storiche e culturali in cui sono emersi: si tratta, quindi, di un corpus cinematografico che rappresenta eventi, circostanze e sentimenti che sono tutti profondamente ancorati nel proprio contesto storico e culturale.

Alla fine delle grandi ed intense stagioni si arriva a un punto nodale in cui si tirano le somme dell’esperienza appena trascorsa. Si tratta di fare i conti col prima, col durante e col dopo: ciò che è stato, ciò che poteva essere, ciò che è e ciò che, alla fine, sarà. Così è stato alla fine degli anni ’70 quando – tra le violenze, le speranze e le tragedie di una tormentata stagione iniziata un ventennio prima – la partita tra la sinistra rivoluzionaria e lo Stato italiano stava per chiudersi mentre altre, altrettanto intense, cominciavano ad aprirsi. In eredità non restavano altro che i nodi irrisolti, le mezze verità, realtà che apparivano sempre più “opache”, gli animi e gli scorni di chi aveva dovuto constatare la propria sconfitta. Molti intellettuali – militanti e non – si trovarono a fare i conti con una stagione intensa e tormentata, intrisa di vitalità ma affossata dalla tragedia della violenza e da un potere percepito come invisibile, all’interno della quale concetti quali “verità”, “ragione” e soprattutto “realtà” vennero messi in crisi. Il post-modernismo, tra la fine degli anni ’70 e l’inizio degli anni ’80, aveva infatti preso piede come corrente culturale che metteva in discussione l’esistenza stessa della realtà constatando la fine delle “grandi narrazioni” delle ideologie. In Italia, però, questa tendenza andò a cozzare con il particolare clima della stagione appena trascorsa: la realtà in sé non venne messa in discussione ma, anzi, da un lato un gruppo di intellettuali “di sinistra” iniziò una intensa discussione per cercare nuovi metodi per descrivere la realtà, dall’altro alcuni militanti – quelli fuggiti, quelli rimasti e quelli incarcerati dopo il 7 aprile – intesero la loro professione di intellettuali come un modo di agire nella realtà.

Abstract:
Questo elaborato affronta il tema delle esperienze di viaggio nella Cina degli anni '70 da parte di studiosi italiani, prendendo in esame i tre viaggi che l’archeologo Vincenzo Tusa compì in quegli anni. Per poter inquadrare e dare il giusto valore a ciò che scrive lo studioso, si è descritto prima il panorama e gli sviluppi storico-politici dei rapporti tra Italia e Cina di quegli anni, procedendo poi col dare uno sguardo alle esperienze di altri studiosi italiani che, dagli anni successivi alla fondazione della RPC in avanti, ebbero l'opportunità di visitare il paese. I tre viaggi di Tusa risultano di particolare interesse perché compiuti in tre grandi momenti diversi, che vanno dalla Rivoluzione Culturale alla denuncia della banda dei quattro, per poi concludersi col terzo viaggio in una Cina che si affaccia agli anni '80. I primi due viaggi, rispettivamente nel 1973 e 1976 sono da inquadrare in un periodo permeato da una forte propaganda socialista, nel quale gli studiosi stranieri subiscono inevitabilmente il fascino della Cina di Mao; il paese del terzo viaggio risente delle vicissitudini trascorse e appare agli occhi dello studioso, che nel 1981 può finalmente recarsi in Cina nelle vesti ufficiali di archeologo, profondamente cambiata. Guardare il paese attraverso la lente critica dell’archeologo ci restituisce tre fotografie molto differenti della Cina, soprattutto perché Tusa riesce a non abbandonare lo spirito curioso tipicamente archeologico di chi è abituato ad investigare sotto la superficie alla ricerca di ciò che è stato.

Il capitolo introduttivo è dedicato al contributo delle donne alla storia della fotografia nel corso degli anni, in modo tale da delineare un'adeguata cornice artistica sulla quale collocare con maggior consapevolezza la mostra in analisi.

Abstract:
The paper deals with the limitation of the entrepreneur’s autonomy of will and freedom of expression in relation to the consumer by the principle of non-discrimination. The subject of the examination is the strict Section 6 of the Consumer Protection Act, which only prohibits the entrepreneur from discriminating against consumers in the sale of goods or services, without defining specific discriminatory grounds. The case-law refers in this respect to the regulation contained in Section 10 of the Anti-Discrimination Act. The paper focuses on the case law of the Czech courts that has dealt with discriminatory conduct in B2C relations and this is critically assessed.

Abstract:
AbstractSocial cognition refers to a wide range of cognitive abilities that allow individuals to understand themselves and others and also communicate in social interaction contexts (Adolphs, Curr Opin Neurobiol 11(2):231–239, 2001). According to Adolphs (Annu Rev Psychol 60(1):693–716, 2009), social cognition deals with psychological processes that allow us to make inferences about what is happening inside other people—their intentions, feelings, and thoughts. Although the term can be defined in many ways, it is clear that it must be safeguarded for the mental operations underlying social interactions. The most investigated cognitive processes of social cognition are emotion recognition and theory of mind (ToM), given that a whole range of socio-affective and interpersonal skills, such as empathy, derive from them (Mitchell RL, Phillips LH, Neuropsychologia, 70:1–10, 2015). Theory of mind is an intuitive ability to attribute thoughts and feelings to other people, and this ability usually matures in children in preschool age (Wellman HM, The child’s theory of mind. Bradford Books/MIT, 1990), whereas emotional recognition refers to an individual’s ability to identify others’ emotions and affective states, usually based on their facial or vocal expressions, it is a critical skill that develops early and supports the development of other social skills (Mitchell RL, Phillips LH, Neuropsychologia, 70:1–10, 2015).

AbstractWe have emphasised in this chapter that our findings do not fit into any simple political template of thinking that has existed for many years on the topic of “Islam” and “migration”. Our findings show that surveys authoritatively designed by Arab social science clearly measure “political Islam”, but that the phenomenon is less pronounced in the population that says it wants to emigrate to the West than in the Arab population as a whole. We have also clearly pointed out that the RMPE cannot be separated from the climate of lawlessness that many observers unfortunately now see rampant, especially in Western industrialised countries, and secondly, that the drivers of the key variables of the RMPE are rooted in such patterns of thought and values as the demand for a redistributive state, the apolitical young generation, the rejection of the neoliberal market economy, corruption and lawlessness as well as racism and xenophobia. The best blockades against the RMPE are feminism and secularism. An honest examination of the phenomenon of RMPE will also not be able to ignore the fact that especially in Catholic countries, where the decay of traditional values is progressing particularly fast, not only the acceptance of corruption but also of political violence is on the rise again. This problem also arises in countries with a confessional orientation towards Eastern religions. The rejection of free-market competition (competition is harmful) is also clearly linked to a higher acceptance of political violence, according to the World Values Survey. The results of our study on political Islam in the Arab world certainly also have some very shocking aspects that cannot simply be swept under the carpet. Weighted by population, the Arab Barometer data show that more than 70% of Arabs have a (sympathetic) understanding of the anti-American terror that culminated in 9/11 in Manhattan. More than 44% of Arabs favour Sharia with corporal punishment, more than 37% want the rights of non-Muslims in society to be less than those of Muslims, and more than 34% also want Sharia to restrict the rights of women. We finally highlight that following the late Harvard economist Alberto Alesina (1957–2020), social trust is an essential general production factor of any social order, and the institutions of national security of the democratic West would do well to make good use of this capital of trust that also exists among Muslims living in the West.

Come affrontare i processi di riqualificazione quando “il rapporto tra le aree-residenza e elementi primari” non è più alla base della configurazione per tessuti della città e la casa diventa monumentale memoria e gesto rappresentativo dello Stato? Concentrando lo sguardo sul periodo tra gli anni 60' e 70' si rende evidente un intento congiunto delle amministrazioni e degli architetti, attraverso la progettazione di complessi a scala macroscopica, spesso isolati dal contesto classico della città, di creare una visione chiara ed iconica del ruolo assistenzialista dello Stato. Questa stagione, sottolineata da scelte compositive in contrapposizione con la visione “ordinaria” della città e della sua crescita per tessuti, raggiunge la sua massima espressione nel “grande segno” che fa coincidere tipologia architettonica con morfologia urbana e racconta di un’amministrazione che vuole essere il referente diretto per la risoluzione di problematiche sociali e risponde in maniera reattiva e molto rapida alle questioni e alle esigenze poste dalle sue classi più bisognose. Il primo valore da riconoscere ai progetti di questo periodo è di essere stati rappresentativi di vocazioni collettive e di averle riassunte con un gesto progettuale dall'alto valore iconografico. La seconda caratteristica di questa stagione architettonica, continuando a restringere la valutazione ad una constatazione dei fatti scevra di componenti di giudizio, è la sua ampia vocazione ad essere terreno sperimentale sia in ambiti architettonici che urbanistici. La domanda che ci poniamo è se sia possibile ripartire da queste due valori per re-interpretare i progetti dei grandi complessi residenziali pubblici e renderli “abitabili” mantenendone le loro particolari vocazioni. How can we deal with requalification process in parts of the city which are no longer based on the “relation among residential-area and primary elements” and where the house has become the rappresentative memory of programs and ideals of the State? Concentrating on the 60' and 70' period in Italy it is clear a joined intent of the administration and the architects to create a neat and iconographic image of the protective and directive role of the State towards its citizen, through the designing of large, unitary social housing, often off the normal urban scale and isolated by the rest of the city. This season, highlighted by compositive experience in sharp contrapposition with the “ordinary” vision of the city, reach its maximal expression in “great sign design” that tends to an equivalence between architectural typology and urban morphology. The first merit that have to be acknowlegde to those project is to have been a representative of collective vocations through a high valued iconographical design. A second valor to be confered to this period is its vocation on being a laboratory for both architectural and urban experimentation. Question we are facing is, then, if it is possibile to start over on those two intrinsic values, seep through the lens of the european directives for sustainable renewal of the cities, for a reinterpretation of the large housing estates in order to give them back to their peculiar potential and to make the the center of regenerations of the city itself.

Abstract:
Artificial Neural Networks (ANNs) have been used to predict assembly time and market value from assembly models. This was done by converting the assembly models into bipartite graphs and extracting 29 graph complexity metrics which were used to train the ANN prediction models. This paper presents the use of sub-assembly models instead of the entire assembly model to predict assembly quality defects at an automotive OEM. The size of the training set, order of the bipartite graph, selection of training set, and defect type were experimentally studied. With a training size of 28 parts, an interpolation focused training set selection, and second order graph seeding, over 70% of the predictions were within 100% of the target value. The study shows that with an increase in training size and careful selection of training sets, assembly defects can be predicted reliably from sub-assemblies complexity data.

Abstract:
The paper examines the potential of a novel adaptive neural network compensator (ANNC) for the position control of a pneumatic gantry robot. Previousl experimental results were disappointing, with only a 20% improvement in performance when ANNC was employed with a PID controller. The conclusion was that the level of improvement with ANNC did not warrant the extra effort required for implementation. However, when the tests were repeated after the system had been reconfigured, improvements on the order of 45% to 70% were achieved. This paper presents a tuning procedure for ANNC, confirms the adaptive nature and provides results that support the conclusion that ANNC can indeed provide a significant improvement in tracking performance.

Abstract:
Transportation has an enormous impact on the U.S. economy and on the lives of all Americans. Many modes of transportation rely on pavement, but pavement conditions deteriorate over time because of the combined effects of traffic and climate. Exposure to moisture often causes premature failure of asphalt pavements as it reduces the stiffness of the asphalt and enables stripping of the asphalt from the aggregate. This research evaluates the effectiveness of clay nanomaterials (i.e., nanoclays) in improving the resistance of hot mix asphalt (HMA) to moisture damage and compares the enhancement results to anti-stripping additives commonly used in pavement construction. Two types of surface-modified nanoclay, lime-treated aggregate, and two amine-based liquid antistripping agents (HP Plus and LOF 6500) were evaluated for improving HMA’s moisture resistance. All additives tested for reducing moisture damage resulted in dry and wet tensile strength of the modified mixes higher than the minimum specified by Caltrans 2018 Standard Specifications (100 psi for dry tensile strength and 70 psi for wet tensile strength). The Tensile Strength Ratio (TSR) of all HMA modified mixes was higher than 0.80, which is the minimum specified by the Superpave mix design method (Asphalt Mix Design Methods MS-2, Asphalt Institute) and exceeded the TSR of the control mix. The TSR for HMA mixes modified using nanoclays were comparable to those for HMA mixes modified using liquid antistripping and lime slurry treated aggregate. Liquid antistripping agents tested herein were the least costly additive at an approximately $2.0/ton added cost. This research can be used to better understand pavement deterioration to enable the most efficient and cost-effective construction and preservation of our nation's critical transportation infrastructure.

Abstract:
This study is the latest in a series monitoring the evolution of poverty, based on data gathered by The ESRI in the Living in Ireland Surveys since 1994. These have allowed progress towards achieving the targets set out in the National Anti Poverty Strategy since 1997 to be assessed. The present study provides an updated picture using results from the 2000 round of the Living in Ireland survey. The numbers interviewed in the 2000 Living in Ireland survey were enhanced substantially, to compensate for attrition in the panel survey since it commenced in 1994. Individual interviews were conducted with 8,056 respondents. Relative income poverty lines do not on their own provide a satisfactory measure of exclusion due to lack of resources, but do nonetheless produce important key indicators of medium to long-term background trends. The numbers falling below relative income poverty lines were most often higher in 2000 than in 1997 or 1994. The income gap for those falling below these thresholds also increased. By contrast, the percentage of persons falling below income lines indexed only to prices (rather than average income) since 1994 or 1997 fell sharply, reflecting the pronounced real income growth throughout the distribution between then and 2000. This contrast points to the fundamental factors at work over this highly unusual period: unemployment fell very sharply and substantial real income growth was seen throughout the distribution, including social welfare payments, but these lagged behind income from work and property so social welfare recipients were more likely to fall below thresholds linked to average income. The study shows an increasing probability of falling below key relative income thresholds for single person households, those affected by illness or disability, and for those who are aged 65 or over - many of whom rely on social welfare support. Those in households where the reference person is unemployed still face a relatively high risk of falling below the income thresholds but continue to decline as a proportion of all those below the lines. Women face a higher risk of falling below those lines than men, but this gap was marked among the elderly. The study shows a marked decline in deprivation levels across different household types. As a result consistent poverty, that is the numbers both below relative income poverty lines and experiencing basic deprivation, also declined sharply. Those living in households comprising one adult with children continue to face a particularly high risk of consistent poverty, followed by those in families with two adults and four or more children. The percentage of adults in households below 70 per cent of median income and experiencing basic deprivation was seen to have fallen from 9 per cent in 1997 to about 4 per cent, while the percentage of children in such households fell from 15 per cent to 8 per cent. Women aged 65 or over faced a significantly higher risk of consistent poverty than men of that age. Up to 2000, the set of eight basic deprivation items included in the measure of consistent poverty were unchanged, so it was important to assess whether they were still capturing what would be widely seen as generalised deprivation. Factor analysis suggested that the structuring of deprivation items into the different dimensions has remained remarkably stable over time. Combining low income with the original set of basic deprivation indicators did still appear to identify a set of households experiencing generalised deprivation as a result of prolonged constraints in terms of command over resources, and distinguished from those experiencing other types of deprivation. However, on its own this does not tell the whole story - like purely relative income measures - nor does it necessarily remain the most appropriate set of indicators looking forward. Finally, it is argued that it would now be appropriate to expand the range of monitoring tools to include alternative poverty measures incorporating income and deprivation. Levels of deprivation for some of the items included in the original basic set were so low by 2000 that further progress will be difficult to capture empirically. This represents a remarkable achievement in a short space of time, but poverty is invariably reconstituted in terms of new and emerging social needs in a context of higher societal living standards and expectations. An alternative set of basic deprivation indicators and measure of consistent poverty is presented, which would be more likely to capture key trends over the next number of years. This has implications for the approach adopted in monitoring the National Anti-Poverty Strategy. Monitoring over the period to 2007 should take a broader focus than the consistent poverty measure as constructed to date, with attention also paid to both relative income and to consistent poverty with the amended set of indicators identified here.

The general objective of this study was to develop rationally designed mimetic antagonists (and agonists) of the PK/PBAN Np class with enhanced bio-stability and bioavailability as prototypes for effective and environmentally friendly pest insect management agents. The PK/PBAN family is a multifunctional group of Nps that mediates key functions in insects (sex pheromone biosynthesis, cuticular melanization, myotropic activity, diapause and pupal development) and is, therefore, of high scientific and applied interest. The objectives of the current study were: (i) to identify an antagonist biophores (ii) to develop an arsenal of amphiphilic topically active PK/PBAN antagonists with an array of different time-release profiles based on the previously developed prototype analog; (iii) to develop rationally designed non-peptide SMLs based on the antagonist biophore determined in (i) and evaluate them in cloned receptor microplate binding assays and by pheromonotropic, melanotropic and pupariation in vivo assays. (iv) to clone PK/PBAN receptors (PK/PBAN-Rs) for further understanding of receptor-ligand interactions; (v) to develop microplate binding assays for screening the above SMLs. In the course of the granting period A series of amphiphilic PK/PBAN analogs based on a linear lead antagonist from the previous BARD grant was synthesized that incorporated a diverse array of hydrophobic groups (HR-Suc-A[dF]PRLa). Others were synthesized via the attachment of polyethylene glycol (PEG) polymers. A hydrophobic, biostablePK/PBAN/DH analog DH-2Abf-K prevented the onset of the protective state of diapause in H. zea pupae [EC50=7 pmol/larva] following injection into the preceding larval stage. It effectively induces the crop pest to commit a form of ‘ecological suicide’. Evaluation of a set of amphiphilic PK analogs with a diverse array of hydrophobic groups of the formula HR-Suc-FTPRLa led to the identification of analog T-63 (HR=Decyl) that increased the extent of diapause termination by a factor of 70% when applied topically to newly emerged pupae. Another biostablePK analog PK-Oic-1 featured anti-feedant and aphicidal properties that matched the potency of some commercial aphicides. Native PK showed no significant activity. The aphicidal effects were blocked by a new PEGylated PK antagonist analog PK-dF-PEG4, suggesting that the activity is mediated by a PK/PBAN receptor and therefore indicative of a novel and selective mode-of-action. Using a novel transPro mimetic motif (dihydroimidazole; ‘Jones’) developed in previous BARD-sponsored work, the first antagonist for the diapause hormone (DH), DH-Jo, was developed and shown to block over 50% of H. zea pupal diapause termination activity of native DH. This novel antagonist development strategy may be applicable to other invertebrate and vertebrate hormones that feature a transPro in the active core. The research identifies a critical component of the antagonist biophore for this PK/PBAN receptor subtype, i.e. a trans-oriented Pro. Additional work led to the molecular cloning and functional characterization of the DH receptor from H. zea, allowing for the discovery of three other DH antagonist analogs: Drosophila ETH, a β-AA analog, and a dF analog. The receptor experiments identified an agonist (DH-2Abf-dA) with a maximal response greater than native DH. ‘Deconvolution’ of a rationally-designed nonpeptide heterocyclic combinatorial library with a cyclic bis-guanidino (BG) scaffold led to discovery of several members that elicited activity in a pupariation acceleration assay, and one that also showed activity in an H. zea diapause termination assay, eliciting a maximal response of 90%. Molecular cloning and functional characterization of a CAP2b antidiuretic receptor from the kissing bug (R. prolixus) as well as the first CAP2b and PK receptors from a tick was also achieved. Notably, the PK/PBAN-like receptor from the cattle fever tick is unique among known PK/PBAN and CAP2b receptors in that it can interact with both ligand types, providing further evidence for an evolutionary relationship between these two NP families. In the course of the granting period we also managed to clone the PK/PBAN-R of H. peltigera, to express it and the S. littoralis-R Sf-9 cells and to evaluate their interaction with a variety of PK/PBAN ligands. In addition, three functional microplate assays in a HTS format have been developed: a cell-membrane competitive ligand binding assay; a Ca flux assay and a whole cell cAMP ELISA. The Ca flux assay has been used for receptor characterization due to its extremely high sensitivity. Computer homology studies were carried out to predict both receptor’s SAR and based on this analysis 8 mutants have been generated. The bioavailability of small linear antagonistic peptides has been evaluated and was found to be highly effective as sex pheromone biosynthesis inhibitors. The activity of 11 new amphiphilic analogs has also been evaluated. Unfortunately, due to a problem with the Heliothis moth colony we were unable to select those with pheromonotropic antagonistic activity and further check their bioavailability. Six peptides exhibited some melanotropic antagonistic activity but due to the low inhibitory effect the peptides were not further tested for bioavailability in S. littoralis larvae. Despite the fact that no new antagonistic peptides were discovered in the course of this granting period the results contribute to a better understanding of the interaction of the PK/PBAN family of Nps with their receptors, provided several HT assays for screening of libraries of various origin for presence of PK/PBAN-Ragonists and antagonists and provided important practical information for the further design of new, peptide-based insecticide prototypes aimed at the disruption of key neuroendocrine physiological functions in pest insects.
