Abstract:
The purpose of this study was to undertake detailed molecular and phenotypic characterisation of a MDR isolate of T. circumcincta (MTci5) with particular focus upon the mechanisms underlying benzimidazole (BZ) resistance.  MTci5 was isolated from a farm in Central Scotland, which employed a suppressive anthelmintic dosing regime and was closed in 2002 when control of the parasite population became unsustainable.  Underpinning all of the experiments in this study was an anthelmintic selection process whereby the MTci5 isolate was pressured individually with three broad-spectrum anthelmintics (b

Abstract:
Anthelmintic resistance of parasites infecting livestock animals is a global problem resulting in decreased animal welfare and production losses. Horses are not exempt from this issue as wide-spread anthelmintic resistance exists among the equine cyathostomins and Parascaris spp. Of the three drug classes available for treating equine intestinal helminths anthelmintic resistance, defined as less than 90-95% drug efficacy, exist to all three. New pharmaceutical control regimens and the elucidation of parasite drug response mechanisms are needed. Two studies were carried out evaluating combinati

Abstract:
Anthelmintic treatment of livestock is an important aspect of the control of gastrointestinal parasites. Resistance to anthelmintics is common, and an understanding of resistance requires knowledge of an anthelmintic's mode(s) of action and mechanism(s) of resistance. The parasitic nematode, Haemonchus contortus, has developed resistance to benzimidazoles and avermectins/milbemycins. Proposed mechanisms of resistance are here supported by genetic changes observed in genes whose protein products are believed to interact with these anthelmintics. Statistically significant differences in allele f

Abstract:
Anthelmintic resistance is a major problem in livestock, and while it has been slower to emerge in cattle, there are reports of its occurrence. Three broad-spectrum anthelmintics are available for use, and one mechanism of resistance that is common to all is target site alteration. Glutamate-gated chloride channels (GluCls) are an important target for macrocyclic lactone anthelmintics (MLs), while beta-tubulin represents the benzimidazole (BZ) target. The objectives of this thesis were to determine whether GluCls are involved in ML resistance in the cattle parasite Cooperia oncophora ,

Among common problems affecting human health and veterinary medicines, helmintic infections are major. The pathogens affect 550-750 Million people worldwide, and affect childhood growth, pregnancies, and development of the intellect. Helminths affects the well-being of animals as well including livestock and reduce the animal populations. However, the current anthelmintics are no longer as effective and some strains have developed resistance thus increasing the need for new anthelmintics. Unfortunately, not too much information is available detailing the physiology of helminths. The published

Abstract:
Resistance to anthelmintics used to treat parasitic nematodes of veterinary importance represents a serious welfare and economic problem for the livestock production industry. Research into the mechanisms by which parasites develop resistance is necessary to prolong the life of the available drugs and to minimise development of resistance to new classes. Metabolism of anthelmintic compounds by parasites is a possible mechanism of resistance that has received little research, despite there being precedence in the case of insecticide resistance. The objectives of this study are to estimate the degree to which a glutamate-gated chloride channel gene (HcGluCla) contributes to survival of moxidectin treatment and to study the relative dominance of those alleles. In the absence of moxidectin, homozygous susceptible genotypes fed significantly more than homozygous resistant genotypes. The effect of the susceptible allele was dominant. To date, Parascaris spp. are the only Ascaris worms that have developed resistance to anthelmintic drugs. The mechanisms of resistance in Parascaris spp are incompletely understood, partly due to the absence of robust in-vitro models.
