Full textTumor necrosis factor (TNF) is a cytokine involved in systemic inflammation. Tumor necrosis factor promotes the inflammatory response, which, in turn, causes many of the clinical problems associated with autoimmune disorders such as rheumatoid arthritis, ankylosing spondylitis, Crohn's disease, psoriasis and refractory asthma. These disorders could be treated using a TNF inhibitors. Antibodies against TNF proved to be the most perspective instrument for therapy of increased level of TNF.. Recombinant single chain antibodies (scFv) might be excellent alternative to expensive and complicated monoclonal antibodies. The scFv are recombinant antibody fragments consisting of only two domains: the variable light chain (VL) and variable heavy chain (VH) domains, covalently connected to one another by polypeptide linker. The benefits of scFv over traditional monoclonal antibodies shoul be the small penetrable size and rapid, inexpensive production. In addition, the lack of Fc domains makes them less immunogenic and less capable for binding to Fc receptors distributed on the normal cells. In this work we tried to construct and express scFv antibodies against TNF. Firstly, for this purpose we purified total RNA from hybridoma cells, producing monoclonal anti-TNF antibodies. Full textSUMMARY The Origin and the Incidence of Antibodies against Rhesus Blood System Antigens This research work was carried out seeking to assess origin and incidence of antibodies against antigens of Rhesus Blood Type system as well as their distribution after hemotransfusions. Blood of 115 recipients was explored in patients treated at Vilnius University hospital „Santariškių klinikos”. The age of the persons under research varied between 18 and 82 years. Appearance and spread of antibodies against antigens of Rhesus Blood Type system was established using the direct and indirect Coombs test. We had explored the risk of alloimmunization of antibodies against antigens of Rhesus Blood Type system within the period of four years. The risk of alloimmunization of alloantibodies against antigens of Rhesus Blood Type system, which we found on the first time within the first year of research was 9.56 %. And in the end of research, the risk of alloimmunization of alloantibodies against antigens of Rhesus Blood Type system, which we found on the first time was 19.13 %. On the whole, within the first year of research, alloantibodies against antigens of Rhesus Blood Type system were identified in 92.17 % of the patients, and the autoantibodies against antigens of Rhesus Blood Type – in 7.83 % of the patients.Within the fourth year of research, in total 75.47 % of the patients were identified alloantibodies against antigens of Rhesus Blood Type system, and the autoantibodies against antigens... Full textMelatonin is a hormone produced by the pineal gland that is secreted during the night in response to the darkness. It reaches a maximum level in the middle of the night, then decrease until the morning. The light inhibits the melatonin synthesis: it is the hormone of the circadian rate/rhythm. When people are exposed to frequent artificial light and they do not get enough sleep, melatonin level is suppressed. The production of melatonin decreases with age. Melatonin regulates and controls biological clock: it improves sleep, stimulates the immune system, and protects the central nervous system. This hormone is an extremely powerful antioxidant. The literature data about melatonin effect on immune system are mostly about the T cells and macrophages. Melatonin increases the thymus weight. It stimulates the function of TH1 T cells, increases cytokine production by T cells. Melatonin also activates NK cells and macrophages. However, the role of melatonin on the function of B cells is not investigated very well. We have investigated the role of melatonin on the function of B cells, e.g. production of specific antibodies. For this purpose, BALB/c mice were immunized with T independent and T dependent antigen. We looked for production of specific antibodies in animals living at three different conditions. Full textStreptococcus pneumonine is a gram-positive bacterial organism that colonizes the upper respiratory tract and causes diseases such as pneumonia, meningitis, otitis media, sinusitis and others. Disease rates are particularly high in young children. One of the aims of this study was to assess the prevalence rates of streptococcal infection among the population of healthy children of preschool age. 601 nasopharyngeal samples were tested from 2-7 years age kindergardeners in Vilnius. Streptococcus pneumonine were grown from 280 samples (47 % of collected samples). 22 different serotypes were identified; the most frequent serotypes were determined 19F, 23F, 6B, 6A, 3, and 18C. Frequently S.pneumonine colonizes mucosal epithelium at nasopharynx without causing any symptoms. In that case the immunity of an organism is of great significance. The second aim of this study is to evaluate the immune response to pneumococcal infection. The amount of separate immunoglobuline classes (IgG, IgA, and IgM) were assessed in saliva samples of infected children. Monoclonal antibodies developed by hybridoma technology were applied. The amounts of classes IgA, sIgA, IgG, IgM antibodies were determined parallel to amount of specific antibodies to S.pneumonine. Correlation between data groups (total amount of actual immunoglobulin class, concentration of specific antibodies) was weak or moderate. Full textTBE is the most common and severe CNS affecting zoonosis. No specific treatment for TBE exists. Tick-borne encephalitis can be prevented by active immunization. Due to governmental financial shortages this vaccination is not included in the national program of imunoprophylaxis for the years 2014-2018. It is very important to find out groups for priority vaccination according to living area, age, leisure activities and occupational risk. Lyme borreliosis diagnosis should be based primarily on the clinical presentation and an assessment of tick exposure risk. In most cases laboratory diagnosis is only supporting clinical picture. Antibody assays still fall short of being completely sensitive and specific to distinguish acute and chronic infection. The diagnostic specificity of serum antibody tests is low because seropositivity in normal population ranges from 1.8% to 26% and LB IgM and IgG may persist for years. The interpretation of serological tests may also lead to misdiagnosis not knowing the seroprevalence of LB in the area. This is the first study of seroprevalence of LB and incidence of TBE and LB during a three year period in Lithuania. The study was conducted in high endemic areas and random samples stratified by gender and age groups were made. Only a few studies perform neutralization tests for detecting TBEV antibodies. Assumed risk factors for the prevalence of TBE and LB were established.

Full textDisertacijoje aprašomi monokloniniai antikūnai, sukurti prieš rekombinantinius mielėse susintetintus antigenus: žmogaus paragripo treciojo tipo viruso, Menangle viruso, hantavirusų bei pasiutligės viruso nukleokapsidės (N) baltymus. Sukurtieji antikūnai buvo visapusiškai charakterizuoti įvairiais imunocheminės analizės metodais, įvertintas jų specifiškumas, afiniškumas, sugebėjimas atpažinti natyvius virusus infekuotų ląstelių kultūrose. Buvo nustatyta, kad antikūnai, sukurti prieš rekombinantinius mielėse susintetintus virusų baltymus, tinka virusų nustatymui infekuotose ląstelėse. Šie tyrimai patvirtino, kad rekombinantiniai mielėse susintetinti virusu N baltymai turi panašias antigenines savybes, kaip natyvūs virusų N baltymai, formuojantys nukleokapsides. Sukurtieji monokloniniai antikūnai taip pat buvo panaudoti išsamiems minėtų virusų N baltymų antigeninės struktūros tyrimams bei imunodominuojančių sekų nustatymui. Disertaciniame darbe gauti duomenys svarbūs, kuriant naujas imunodiagnostikos sistemas, skirtas virusų infekcijoms nustatyti. Disertacija sudaro įvadas, trys skyriai, naudotos literatūros sąrašas ir autorės publikacijų sąrašas. Full textThe aim of this study was to generate monoclonal antibodies against Hendra and Nipah virus nucleocapsid proteins. There is high homology between nucleocapsid proteins of Henipavirus genus members, therefore it is difficult to generate monoclonal antibodies that do not show any cross-reactivity with both antigens. This problem was solved by using recombinant chimeric proteins designed by insertion of non-homological segments of Hendra and Nipah virus nucleocapsid proteins into the mouse polyomavirus capsid protein VP1. Mice were immunized with these chimeric proteins and it was determined that they induce a strong immune response. Monoclonal antibodies against Nipah virus nucleocapsid protein as well as carrier protein – mouse polyomavirus capsid protein VP1 – were generated. The specificities of newly developed monoclonal antibodies were confirmed by ELISA and immunoblot. The generation of specific monoclonal antibodies against Hendra virus nucleocapsid protein failed.

Full textSUMMARY The prevalence of antibodies agaisnt nuclear antigens in patients in systemic connective tissue diseases The aim of this study was to assess the diagnostic value and to determine the frequency of ENA/ANA in systemic connective tissue diseases (SCTD) and to compare the association of these antibodies with following inflammatory markers ESR, CRP and rheumatoid factor (RF). The analysis involved 50 patiens with seven different diagnosis of connective tissue diseases: mixed connective tissue disease (MITD), systemic lupus eritromatosus (SLE), Sjogren syndrome, systemic sclerodermia, nondifferential connective tissue disease, dermatomiositis/ polimiositis and arthropaties. Sera from each patient was tested for ANA by indirect immunofluoresce (NIF), we assigned, that speckled pattern is appropriate for 60% and homogenous pattern is appropriate for 36% SCTD patients. The result was confirmed by immunoblot method, we estimated, that the prevalence of ENA is as follows anti-Ro-52 (56%), anti-SS-A (44%), anti-SS-B (26%), ds-DNR and nukleosomal antibodies (24%), nRNP/Sm, Scl-70 and hystonic antibodies (14%). Anti-Jo-1 (10%), anti-CENP-B (8%) anti-PCNA (4%). Antibodies to ribosomal and P- Prot antigens (2%). Antibodies to Ku antigens were not detected. Full textObject. Coeliac disease is one of the most prevalent gastrointestinal disorder. It‘s prevalence in Europe and North America is 1 of 150-300 adults and children. In Lithuania there are no data on prevalence of coeliac disease but it‘s seems that there is hypodiagnosis [29]. IgA-deficient individuals have a tenfold risk for coeliac disease compared with the general population. Patients with IgA deficiency have a tendency to develop recurrent sinopulmonary and gastrointestinal infections, allergies, autoimmune diseases and malignancies [148-154]. There are no data on the prevalence of IgA deficiency in Lithuania [29]. The aim. To determine the prevalence of coeliac disease and IgA deficiency among 11-13 years children in Vilnius. Material and methods. From January 2009 to March 2010 we investigated 1000 children (11-13 years of age) for coeliac disease and immunoglobulin A (IgA) deficiency from six Vilnius secondary schools. We examined capillary blood for tissue transglutaminase IgA class antibodies (tTG IgA) and total IgA with Biocard TM test (Ani Biotech OY, Finland) for children whose parents agreed to do this test. The results were evaluated after 5 min. Children with positive test results (tTG IgA positive or total IgA deficiency) underwent small bowel endoscopy with biopsies at Vilnius University Children Hospital to confirm or deny the diagnosis of coeliac disease. Full textThe objective of the present doctoral dissertation was to evaluate the epidemiological situation of porcine reproductive and respiratory syndrome (PRRSV) infection in the wild boar and pig populations between 2008 and 2011 in Lithuania by means of immunological and molecular methods. The serological studies of PRRSV carried out on the pig-breeding farms in Lithuania showed that the changed pig-breeding conditions, technologies and the adopted biosafety measures, enabled PRRSV prevalence in the pig population in Lithuania in which as much as 4.29 per cent of pigs under investigation had PRRSV-specific antibodies in 12 out of 30 Lithuanian regions, to be reduced tenfold. Validation of enzyme-linked immunosorbent assay (ELISA) method demonstrated, that commercial INGEZIM PRRS EU and IDEXX PRRS X3, IDEXX PRRS Herd Chek IFA kits variations in the results were not statistically significant and all the kits are suitable for serological diagnostics in Lithuania when investigating samples of blood serum of both pigs and wild boars. First time in Eastern Europe, investigations into PRRSV infection carried out a relatively large number, 6.36 per cent, of wild boars from 23 regions of Lithuania that had antibodies specific to this virus, which was even larger than the number and prevalence of serologically positive pigs determined in the pig population on the Lithuanian pig–breeding farms.
