Abstract:
Dans notre travail de recherche nous voulons, d’un côté, valoriser l’actualité éthique et politique de la pensée sophistique et, de l’autre, réhabiliter la pensée de Protagoras et d’Antiphon sur un plan purement philosophique. Nous considérons les sophistes, en général, et Protagoras et Antiphon, en particulier, comme étant les « ancêtres » de l’antidogmatique et du relativisme dans le domaine de la philosophie, de l’anthropologie ou encore dans le domaine historico-politique. Par conséquent, ils étaient aussi les ancêtres du courant laïc, critique et démocratique de la pensée occidentale qui, malgré avoir toujours été « minoritaire », représente l’un des fondements de l’identité culturelle européenne. Cette dernière a une grande « dette » envers eux, desquels elle a repris et réutilisé de nombreux concepts et de multiples inventions philosophiques sans presque jamais leur en attribuer le mérite.Pour reconstruir les philosophies des deux sophistes, nous avons démarré notre recherche par une traduction et une étude des sources à notre disposition, ainsi que par une analyse approfondie des contextes historiques, sociaux, économique, politiques et culturels.Nous avons présenté les discours des deux philosophes comme une sorte d’antilogie : nous apprendons de PRO-tagoras qu’il est possible de créer une société qui correspond à nos valeurs ; et d’ANTI-phon que le sens critique, la remise en question et la lutte politique sont nécessaires afin que le nomos soit toujours respectueux de l’évolution et des changements de notre société. Nous avons enfin opéré une confrontation critique entre les deux philosophies afin d’en ressortir trois perspectives utiles pour notre présent et notre futur : l’antidogmatisme, le relativisme constructiviste et le minoritarisme
In our research work we want, on the one hand, to enhance the ethical and political relevance of sophistic taught and, on the other, to rehabilitate the thinking of Protagoras and Antiphon on a purely philosophical level.In general, we consider the sophists, and particularly Protagoras and Anthiphon, as the “ancestors” of the antidogmatic and relativism in the philosophic, anthropologic or historical-political fields. Consequently, they were also the ancestors of the secular, critical and democratic current of Western thought which, despite having always been a "minority", is one of the foundations of European cultural identity.The latter has a great "debt" towards them, from which it has taken and reused many concepts and numerous philosophical inventions without almost ever granting them the merit.To reconstruct the philosophies of the two sophists, we began our research with a translation and a study of the sources available to us, as well as an in-depth analysis of the historical, social, economic, political and cultural contexts.We have presented the speeches of the two philosophers as a kind of antilogy: we learn from PRO-tagoras that it is possible to create a society that corresponds to our values; and from ANTI-phon that the critical sense, the questioning and the political struggle are necessary so that the nomos is always respectful of the evolution and the changes of our society.We have finally made a critical confrontation between the two philosophies in order to reveal three useful perspectives for our present and our future: antidogmatism, constructivist relativism and minoritarism

Abstract:
In her three major works, Ryder (1928), The Antiphon (1958), and Nightwood (1936), Djuna Barnes explores incest as a mode of being and relating. She focuses on the relationships among the Ryders of Ryder, the Hobbses of The Antiphon, and between Nora Flood and Robin Vote of Nightwood as her three primary instantiations of the incestuous family apparatus. Taken together, Barnes's works present a scale of viability for the incestuous relationship. Through engagement with Barnes's work and her implicit theory of incest, this project seeks to investigate and challenge normative categories of viable relationships that currently do not (nor did they in Barnes's time) include incestuous relationships. Barnes's work suggests that it is the individual instantiations of the incestuous family apparatus that make incest a viable mode of being and relating, just as it is the individual instantiations of the non-incestuous family apparatus that allow the normative definition of the family to thrive. Barnes presents familial relationships as not taboo, and suggests that incest is not necessarily concomitant with trauma, fantasy, or abuse. Through analysis of Barnes's work, this thesis sheds new light on a central dimension of Barnes's work. In addition, through Barnes's conceptions and presentations of incest, the project seeks to open up new understandings of incestuous relationships and their implications, as well as, more generally, consider ways to expand the range of ways-of-life - what I call livelihoods - accepted as valid and valuable (viable) by North American and European culture.
Dans ses trois principaux ouvrages, Ryder (1928), The Antiphon (1958), and Nightwood (1936), Djuna Barnes étudie l'inceste, sous tous ses aspects, en tant que façon d'être et comme moyen relationnel. Elle se concentre sur ces relations à travers les Ryders dans Ryder, les Hobbses dans l'Antiphon ainsi qu'entre Nora Flood et Robin Vote dans Nightwood, comme ses trois principaux cas de l'appareil familial incestueux. L'oeuvre de Barnes mis bout à bout représente l'échelle de viabilité de la relation incestueuse. À travers l'engagement de Barnes dans son travail et sa théorie implicite de l'inceste, ce projet vise à questionner et défier les catégories normatives de rapports viables excluant, de nos jours et du temps de Barnes, la présence de rapports incestueux. L'oeuvre de Barnes propose que dans chaque cas, chaque individu de l'appareil familial incestueux fait de l'inceste une façon d'être viable et un moyen relationnel, de même que dans chaque cas, chaque individu d'un appareil familial non incestueux permet la définition normative de la famille épanouit. Barnes aborde la relation familiale comme sans tabou, et suggère que l'inceste ne coïncide pas nécessairement à traumatisme, fantasme, ou abus. Par l'analyse de son oeuvre, cette thèse éclaire une nouvelle avenue à la dimension centrale de son travail. De plus, dans les conceptions et les présentations de l'inceste de Barnes, ce projet tend à ouvrir de nouveaux horizons des relations incestueuses ainsi que leurs implications, autant que, de façon général, élargir la gamme de modes de vie, que j'appelle moyens de subsistance, acceptée comme valide et valable, dans le sens de viable dans la culture Nord-américaine et européenne.

Esta dissertação traduz e comenta as Tetralogias de Antifonte. Trata-se dos três discursos que lidam com casos hipotéticos de homicídio compostos na segunda metade do século V a.C. e que são caracterizados como exercícios argumentativos. Para isso, tal pesquisa foi dividida em três partes: (i) um capítulo com a exposição de algumas informações acerca do autor e da obra; (ii) a apresentação de uma edição atualizada seguida de uma tradução completa dos três discursos; e (iii) os comentários referentes a cada um deles.
This study aimed to translate and comment the Tetralogies of Antiphon, which consists of three speeches of hypothetical homicide cases. The speeches were written in second half of the fifth century BC. They consist of exercices in argumentation. The study was divided into three parts: (i) the chapter presenting the author and his work; (ii) an up-to-date edition and a complete translation of the three speeches; and (iii) the commentaries concerning each one of them.

Il existe des liens intellectuels significatifs entre la sophistique ancienne et l'ecole d'abdere. Ces echanges, qu'ils soient le reflet d'une communaute de vue ou celui d'une opposition doctrinale, revetent une dimension systematique et revelent une remarquable continuite temporelle. Leur mise en evidence permet de rendre compte des difficultes de la theorie atomistique. L'influence de protagoras sur son concitoyen democrite autorise a justifier le caractere recurrent des sentences pre-sceptiques dans l'oeuvre de l'atomiste. L'infraction du sophiste a l'interdit parmenidien a provoque, en reaction, l'edification par democrite du systeme atomistique, mais a aussi ouvert la voie a l'elaboration de la notion de non-etre relatif : le vide. C'est enfin pour rester fidele aux orientations anthropologiques et politiques de son maitre protagoras que democrite suppose, en contradiction avec le determinisme atomistique, l'existence de la liberte humaine. La these d'une communaute philosophique sophisticoabderitaine permet, en outre, de rendre raison du double heritage -sceptique et atomistique- de l'ecole du nord. Dans les deux cas, un sophiste, par son intransigeance intellectuelle, joue un role central de radicalisation qui induit au sein meme de l'ecole une evolution doctrinale. Cela est vrai de gorgias, qui par son nihilisme absolu inflechit l'ethique du sophiste abderitain anaxarque, aboutissant a la prudente aphasie pyrrhonienne. Cela est egalement vrai d'antiphon l'athenien, democriteen a outrance, qui pousse l'atomisme a ses consequences extremes, soutenant l'existence d'undeterminisme psychique et elaborant une anthropologie strictement atomistique. Representant une radicalisation de la doctrine, dont on trouve une approche parallele chez l'abderitain nausiphane, sophiste lui aussi, une telle orientation conduira epicure a modifier la physique atomistique afin de donner un fondement naturel, contre antiphon, a la liberte de l'homme.

Abstract:
The musical structure of Hildegard von Bingen's "O quam mirabilis est" is directly linked to its notational system. After placing Hildegards's antiphons within an historical context in chapter one, chapter two reviews three previous analyses of "O quam mirabilis est" by Bronarski (1922), Cogan (1990) and Pfau (1990). The first two analyses ignore the syntax and expression of the text by focusing on the motivic level. The third analysis, though it embraces the text, lacks a formalization in its theoretical model. None of the analyses respond to the original notation. In response, chapter three examines the notation found in the sources containing Hildegard's music (the Riesenkodex and the Dendermonde codex) and discusses the structural importance of pitches within the neumatic groupings. The resulting graphic analysis adapt Schenkerian analytic notation to represent a hierarchy of pitch relationships. Chapter four applies this methodology to four other antiphons by Hildegard: "Hodie aperuit," "Nunc gaudeant," "O virtus sapientie," and "O virgo ecclesia."

We report on a study of the invariant mass spectrum of the hadronic system in à - [arrow right] à - à 0 à à à à à ½ à decays. This study was performed using data obtained with the BABAR detector operating at the PEP-II e + e - collider. We present fits to phenomenological models in which resonance parameters associated with the à (770), à '(1450) and à "(1700) mesons are determined. We also discuss the implications of our data with regard to estimates of the hadronic contribution to the muon anomalous magnetic moment.

Baldassare Galuppi (1706--85), during the course of his compositional career, composed a number of settings of the Marian antiphon text " Salve Regina," of which only one choral and five solo settings survive. In this thesis Galuppi's compositional career is reviewed and the history of the Venetian ospedali, or orphanages, addressed, especially as it relates to the composer's tenure at the Ospedale di San Lazzaro dei Mendicanti and the Ospedale del Santissimo Salvatore degli Incurabili. Also, the importance of the Marian text itself is discussed in relation to Galuppi's works at the two institutions. The five solo settings for soprano and orchestra, two of which are securely dated, are examined individually and, based on stylistic considerations, given a tentative chronology. Problems of attribution for one of the settings are discussed in light of the discovery of a second manuscript source attributed to Giovanni Battista Borghi. Critical editions of the five solo settings are included.

Les maladies à prions sont des maladies neurodégénératives touchant l’homme ainsi que l’animal et dont l’issue est toujours fatale. Notre objectif était d’utiliser la 6AP et le GA, deux drogues antiprion isolées au laboratoire comme des hameçons moléculaires pour identifier leurs cibles cellulaires afin de mieux comprendre leur mode d’action ainsi que les phénomènes de prionisation. Deux criblages, l’un génétique et l’autre biochimique ont révélé que les chaperons de protéines et les ribosomes sont impliqués. Les drogues antiprion testées ciblent spécifiquement la fonction de repliement des protéines portée par le grand ARNr 23S/25S/28S. Ceci suggère que l’activité de chaperon de protéines du grand ARNr est nécessaire pour le maintien et la propagation des prions. Ces molécules semblent aussi être actives contre d’autres maladies liées à des problèmes de repliement et d’agrégation des protéines. Nos molécules ont donc un avenir potentiel dans le traitement de ces maladies
Prion-based diseases are fatal neurodegenerative disorders for which no efficient treatment is currently available. We focussed our work on the identification of the intracellular biochemical targets of two drugs 6AP and GA, isolated in the laboratory as antiprion drugs using a yeast-based assay to uncover biochemical pathways controlling prion formation. Using both genetic and biochemical approaches, we show an RNA-dependent interaction of 6AP and GA with ribosomes. 6AP and GA specifically inhibit the protein folding activity of the large ribosomal RNA of the large ribosome subunit. We thus propose that the ribosomal RNA protein folding activity is required for prion maintenance. These drugs are also efficient against other protein misfolding diseases: Huntington disease or cystic fibrosis. 6AP and GA could have much wider applications than in prion diseases

Abstract:
In human bone, 90% of organic bone matrix is composed of type 1 collagen. Cathepsin K is a cysteine protease involved in osteoclast mediated bone absorption and has been identified as a major drug target for the treatment of osteoporosis. Numerous potent inhibitors of cathepsin K have already been identified from natural sources including epoxide inhibitors such as E-64 isolated from the fungi Aspergillus japonicus as well as various peptide aldehydes such as Leupeptin and alpha-MAPI purified from Streptomyces. 350 soil and lichen-associated bacterial strains collected in the rain forests of British Columbia were screened and 22 samples were identified containing significant cathepsin K inhibitory activity. From those active samples, L-91-3 was selected as one of the most potent samples for further characterization of their cathepsin inhibitor content. Three Antipain-related peptide inhibitors were identified from L-91-3 strain of Streptomyces. Antipain (Ki 41nM +/- 37nM) and Vince 2 (cyclized P1 derivative Ki 295nM +/- 123nM) were isolated by traditional purification and subsequent NMR and Mass Spectrometry analysis. The cyclized compound, Vince 2 (phenylalanyl-ureido-arginyl-valinyl- cycloarginal), lacked the aldehyde function and resulted in a lower binding affinity towards cathepsin K. Using Cathepsin K as bait for active site directed inhibitors a third compound, named Lichostatinal was identified by x-ray crystallography where recombinant human cathepsin K was co-crystallized with the semi-crude fermentation broth resulting in 2.0 Å resolution crystal structure. Lichostatinal is a peptide-based aldehyde with the amino acid composition (agmatinyl-ureido-serine-valinyl-arginal). The P1-P4 substrate residues of Lichostatinal interact with the non primed S1-S4 subsites of cathepsin K.

Medidas inéditas de Interferência Coulombiana-Nuclear (ICN) nos núcleos ímpares ANTPOT.99,101 Ru complementam a investigação da coletividade da primeira excitação quadrupolar dos isótopos pares de Ru ao redor de A~100, com projéteis que interagem isoescalamente com núcleo. As distribuições angulares do espalhamento inelástico com dêuterons incidentes de 13 MeV, nos núcleos ímpares estáveis de rutênio, foram obtidas no sistema Acelerador Pelletron - Espectrógrafo Magnético Enge, utilizando emulsões nucleares no plano focal. Foram medidos espectros associados a catorze e dezesseis ângulos de espalhamento nos ANTPOT.99 Ru e ANTPOT.101 Ru, respectivamente, com excelente caracterização do mínimo de interferência na primeira excitação qadrupolar do caroço. O exame da ICN foi realizado na descrição DWBA-DOMP com parâmetros globais de potencial óptico, mantendo tratamento consistente com outras análises de ICN na região. O ajuste das previsões teóricas às distribuições angulares experimentais, através da minimização do Chi2 pelo método iterativo de Gauss-Marquardt, possibilitou a extração simultânea do comprimento de deformação de massa ( POT.N) e da razão entre os comprimentos de deformação de carga ( POT.C) e de massa (C= POT.C/ POT.N). Simulações de Monte Carlo com 5000 novos conjuntos de dados, gerados por sorteio gaussiano, mostraram a adequação estatística do método. Os valores experimentais de C se posicionaram ao redor de 1,25 para o ANTPOT.99 Ru 1,40 para o ANTPOT.101 Ru, com incertezas em torno de 5%, indicando maior contribuição de prótons do que de nêutrons na excitação, na comparação com s razões N/Z. As medidas de POT.N, com incertezas menores que 5%, revelaram valores inferiores aos relatados para os núcleos pares vizinhos. Os valores de C e POT.N extraídos permitiram o cálculo das razões B(E2)/B(IS2) para os estados analisados.
Coulomb-Nuclear Interference measurements not previously reported in the 99,10 Ru odd nuclei complement the investigation of the collectivity of the first quadrupolar excitation in even Ru isotopes around A-I00, with projectiles that interact isoscarlarly with the nucleus. The Inelastic scattering angular distributions with 13 MeV incident deuterons on odd Ru stable nuclei were obtained in the facility Pelletron Accelerator - Enge Magnetic Spectrograph, using nuclear emulsion plates in the focal plane. Spectra associated with fourteen and sixteen scattering angles in 99Ru and 101Ru, respectively, were measured achieving an excellent characterization of the interference minimum of the core first quadrupolar excitation. The DWBADOMP description of the CNI with global optical model parameters employed allows the consistent analysis of the data in comparison with other analysis of the region. The simultaneous extraction of g N, mass deformation length and of C = C/N, ratio of charge (C ) and mass (N ) deformation lengths, was obtained in the fit of the theoretical predictions to the experimental angular distributions through the X2 minimization using the iterative Gauss-Marquardt method. Monte Carlo simulations of 5000 new sets of data, gaussian randomly generated, show the statistical adequacy of the method. The experimental values of C obtained distributed around 1.25 for 99Ru and 1.40 for lOlRu, with uncertainties of approximately 5%, indicating higher contribution of protons than neutrons in the excitations, in comparison with the N/Z ratios. The N measurements, with uncertainties less than 5%, point to lower values than published for even neighbor isotopes. The values of N and C extracted allow for the calculation of B(IS2), isoscalar reduced transition probability, and B(E2)/B(IS2), the ratio between electric (B(E2)) and isoscalar reduced transition probabilities.

Abstract:
Chalcone is an alternative name for an α,β-unsaturated carbonyl compound. The system consists of two aromatic rings linked together by a three aliphatic carbon bridge. In the last few years, chalcones and their derivatives have become one of the interesting compounds that show significant activity in reducing PrPSc levels and for this reason these types of compounds were chosen as lead compounds for this project. Different series’ of chalcones were synthesised via aldol condensation under continuous flow conditions. The first route consists of the reaction between 4-(benzyloxy)-3-methoxybenzaldehyde and different substituted acetophenones. The second route contains the indole moiety, these compounds were synthesised from the reaction between 1H-indol-6-carbaldehyde and different substituted acetophenones. A new chalcone was synthesised from 3-((1H-pyrazol-1-yl)methyl)-4-methoxybenzaldehyde which was treated with acetophenone forming the corresponding chalcone. The synthesised chalcone was then used to synthesise two types of pyrrolones containing the moiety of 5-hydroxy-pyrrolidin-2-one (γ-hydroxy-γ-lactam). Three different types of amines were used to synthesise the fourth type of chalcones. The amines were reacted with 4-flourobenzaldehyde forming the corresponding aldehydes which were treated with a variety of heterocylic ketones to produce the corresponding chalcones. Three types of bischalcones were also synthesised from the reaction between 4-(1H-pyrazol-1-yl)benzaldehyde and different ketones. Chalcones containing the quinoline moiety were synthesised in four steps starting from m-toluidine to form the aldehyde which was treated with a variety of acetophenones forming the corresponding chalcones. The ketothylinic group of the chalcones were cyclized into the corresponding pyrazolines by treating them with hydrazine monohydrate using ionic solvent under continuous flow conditions. Finally, all the synthesised chalcones and their derivatives were screened for their antiprion activity and to investigate the antiprion structure-activity relationship. Amongst 51 chalcones and their derivatives 27 compounds were found to be active as antiprion agents. The Best EC50 values were found in compounds 154 (EC50 = 0.097µM), 155 (EC50 = 0.0103 µM), 163b (EC50 = 0.1 µM), 171 (EC50 = 0.1 µM), 179 (EC50 = 0.14 µM), 180 (EC50 = 0.10 µM), 181 (EC50 = 0.54 µM), 182 (EC50 = 0.01 µM) and 183 (EC50 = 0.03 µM).

Resumo: Técnicas de RMN de H foram aplicadas no estudo de metabolômica de Theobroma cacao e interações proteínas ¿ ligantes. Esses dois tópicos são reportados em dois capítulos. O primeiro descreve a análise quantitativa por RMN de H de 8 metabolitos de Theobroma cacao endógenos ou produzidos durante a fermentação de 10 diferentes variedades resistentes ao fungo Moniliophthora perniciosa. Os espectros foram obtidos de extratos aquosos saturando o sinal da água com pressaturação. Os metabólitos monitorados foram: glicose, sacarose, frutose, etanol, cafeínas e os ácidos acético, lático e succínico. O teor dos metabólitos presentes em 7 dessas variedades são similares àqueles de T. cacao comum e análise sensorial selecionou aquelas com maior teor de cafeína como as melhores. O segundo capítulo descreve as interações acetilcolinesterase/alcalóides de Amaryllidaceae e proteína de membrana bacteriana/fosfomicina, com os mapas de epitopo de interação dos ligantes com as macromoléculas determinados por RMN de H ¿ STD (Saturation Transfer Difference). O mapa de epitopo para acetilcolinesterase/fisostigmina foi confirmado por cálculos de ¿doking¿ molecular. A constante de dissociação aparente da fisostigmina com acetilcolinesterase foi obtida através das medidas de T1 seletivo. Interação entre células integras de Escherichia coli CCT 5050, Serratia liquefaciens CCT 7262 e fosfomicina indicaram que microrganismo resistente ao antibiótico (Serratia liquefaciens) não apresentaram sinais no experimento de RMN de H ¿ STD enquanto que o microrganismo não resistente (Escherichia coli) apresenta sinais no mesmo experimento. Esses resultados certamente contribuirão para a elucidação do mecanismo de ação da fosfomicina
Abstract: H NMR tecniques were applied to study Theobroma cacao metabolomics and protein-ligand supramolecular interactions. These two topics are reported in two chapters. First chapter describes the H NMR quantitative analysis of 8 Theobroma cacao secondary metabolites which are endogenous or produced during the fermentation process of 10 different varieties resistent to the Moniliophthora perniciosa fungus. The spectra were obtained from the aqueous extracts saturating the water signal with the PRESAT tecnique. The monitored metabolites were: glucose, sacharose, fructose, ethanol, cafeine and the acids acetic, latic and succinic. The abundance of the metabolites present in 7 of these varieties were similar to those in comon T. Cacao and sensory analysis selected those with high cafeine content as the best. The second chapter describes the supramolecular interactions of acetylcholisterase/Amaryllidaceae alkaloids and bacterial membrane protein /fosfomicine, with the epitope mapping of the interactions of the ligands to the macromolecules obtained by H NMR STD (saturation transfer difference). The epitope mapping of acetylcholisterase/Amaryllidaceae alkaloids was confirmed by molecular docking calculations. The apparent dissociation constant of the fisostigmine which is an acetylcholinesterase inhibitor was obtained by applying selective T1. Interactions of Escherichia coli CCT 5050, Serratia liquefaciens CCT 7262 whole cells and fosfomycin indicated that microorganisms resistent to this antibiotic (Serratia liquefaciens) did not show signal in the H NMR STD experiment while non resistent microorganism (Escherichia coli) showed signals in the same experiment. This data will certainly contribute to the elucidation of fosfomycin action mechanism

This study discusses the use of communication strategies in the dispute for political hegemony in the contemporary society. In order to do so, one starts from the concept of politics in its broad sense, involving, besides parties and parliaments, the groups and organized social movements. The comprehension that the political activity, nowadays, involves the media, made part of this study be focused on the investigation of problems in this relationship. We analyzed, especially, the communication actions promoted by the Grupo Cr?tica Radical, in which participate, among others, Maria Luiza Fontenelle, former mayor of Fortaleza, and Rosa da Fonseca, former concillor. They left their old parties after a process of self-criticism concerning their incumbencies, partly influenced by the books of Robert Kurz about the collapse of capitalism . After this historical contextualization, one starts the analysis of the campaign Greve do Voto promoted by the Group during the municipal elections of 2004. The production of campaign material, the street drama, the small shows and the fact that the press covered the campaign are discussed starting from the theoretical formulations presented on this research, understanding, this way, the challenge of these groups to build an action against the hegemony allied to the universe of the mass media

A measurement of the branching fraction for the rare decay K0L → pi+pi-e +e- was performed using data collected from experiment E871 which ran at the AGS of Brookhaven National Laboratory. Analysis of the data revealed 27.7 +/- 7.4 signal events in the signal region, 0.4905 GeV < Mpipiee < 0.505 GeV together with 13.3 +/- 3.7 background events. The branching fractions of ( 8.5+/-2.3stat +/-1.0sys ) x 10-6 using the phenomenological model acceptance and ( 2.3+/-0.6stat +/-0.3sys ) x 10-6 using the chiral perturbation model represent the first measurements for K0L → pi+pi-e +e- in the dipion invariant mass region 0.475 GeV < Mpipi < 0.497 GeV. These results support the prediction of chiral perturbation theory rather than that of the phenomenological model.

Les maladies à prion font partie des maladies neurodégénératives. L’agent responsable est la protéine prion PrPSc. La conversion de la forme cellulaire nativePrPC en forme pathologique PrPSc et son agrégation sous forme des fibres amyloïdeconstituent des éléments clés de la physiopathologie des maladies à prion. Pourtant,les mécanismes contrôlant/favorisant cette conversion sont très mal connus. Chez lalevure Saccharomyces cerevisiae, il n’existe pas d’homologue de la protéine PrP,mais des protéines se comportant comme des prions existent, telle que Sup35p quiest responsable du prion [PSI+] ou encore la protéine Ure2p qui est responsable duprion [URE3]. Lors d’études antérieures à cette thèse, le laboratoire a isolé la 6AP etle GA, des molécules actives contre les prions de levure [PSI+] et [URE3] et contre leprion de mammifère PrPSc dans des tests cellulaires ainsi que in vivo dans unmodèle murin pour les maladies à prion. Ces résultats démontrent au moins certainsdes mécanismes de prionisation sont conservés de la levure aux mammifères.L’équipe a ensuite montré que la 6AP et le GA étaient des inhibiteurs spécifiques etcompétitifs de l’activité chaperon de protéines du ribosome (ou PFAR pour ProteinFolding Activity of the Ribosome). Ces résultats suggéraient donc que l’activité PFARreprésente un nouveau mécanisme de prionisation conservé de la levure auxmammifères. Par ailleurs, la 6AP et le GA s’étant révélées actives dans des modèlespour d’autres maladies neurodégénératives à fibres amyloïdes, l’activité PFARpourrait également être un acteur physiopathologique majeur de ces protéinopathies.Ma thèse avait deux objets : tester l’implication de l’activité PFAR dans l’apparitionet/ou la propagation des prions et enfin identifier de nouvelles molécules antiprion etcomprendre leurs mécanismes d’action. Mes résultats montrent que l’activité PFARjoue bien un rôle dans la propagation des prions de levure. En effet, l’enrichissementen PFAR favorise l’apparition spontanée du prion [PSI+]. Il conduit également à uneinstabilité accrue de ce même prion. Ainsi, l’activité PFAR ressemble à celle duchaperon de protéine Hsp104p, une protéine indispensable au maintien et à lapropagation de tous les prions de levure, mais qui n’a pas d’homologue chez lesmammifères. Mes résultats suggèrent que les activités PFAR et Hsp104p sontpartiellement redondantes pour le maintien des prions chez la levure et que, chez lesmammifères, seule l’activité PFAR jouerait ce rôle. Parallèlement, nous avonsidentifié de nouvelles familles de molécules antiprion, actives tant contre les prionsde levure que de mammifères. Ces molécules inhibent toutes l’activité PFAR. Nosrésultats contribuent ainsi à une meilleure compréhension des mécanismes deprionisation. Ils indiquent également que l’activité PFAR est une cible thérapeutiqueprometteuse pour les maladies à prion, mais aussi probablement pour d’autresprotéinopathies beaucoup plus fréquentes
Prion diseases are considered neurodegenerative diseases. The incriminated agentis the prion protein PrPSc. The conversion of PrP from its native conformation PrPC tothe pathologic form PrPSc is the major element of the pathogenesis of prion diseases.However, the mechanisms involved in this conversion are poorly understood. In theyeast Saccharomyces cerevisiae, there is no counterpart of the PrP protein. Howeverproteins acting as prion do exist in yeast, such as the Sup35 protein responsible forthe prion [PSI+], or the Ure2 protein responsible for the prion [URE3]. In previousstudies, our team isolated two compounds, 6AP and GA, which are active against theyeast prions [PSI+] and [URE3 ] and against the mammalian prion PrPSc in cellbasedassays as well as in vivo in a mouse model for prion diseases. These resultsdemonstrated that the prionisation mechanisms are at least partially conserved fromyeast to mammals. 6AP and GA specific and competitive inhibitors of the ProteinFolding Activity of the Ribosome (PFAR) thereby showing that the PFAR is oneconserved mechanism of the prionisation. Moreover, 6AP and GA have been provenactive against other amyloid diseases thus placing the PFAR as a key player in thepathophysiology of protein folding diseases. My thesis aims were to test theinvolvement of the PFAR in the initiation and / or propagation of prion, to identify newantiprion molecules and to understand their mechanisms of action. My results showthat the PFAR plays a central role in the yeast prion propagation. Indeed, PFARenrichment promotes the spontaneous appearance of the prion [PSI+] and at thesame time leads to an increased instability of the same prion. Thus, PFAR activityresembles the yeast Hsp104p chaperone protein activity in the maintenance andpropagation of all yeast prions. My results suggest that the PFAR and Hsp104pactivity are partially redundant and that only the PFAR should play this role inmammals. Meanwhile, we have identified new antiprion drugs that are active againstboth yeast and mammal’s prions. These compounds are all inhibitors of the PFAR.Our results contribute to a better understanding of the prionisation mechanisms andindicate that the PFAR is a promising therapeutic target for prion diseases andprobably also for common protein folding diseases.Keywords: prion, yeast, ribosome, protein chaperon, Hsp104

Abstract:
How the linear protein chains fold into their three-dimensional active conformation is one of the remaining puzzles of modern science. Other than molecular chaperones, ribosome - the cellular protein synthesis machinery, has also been implicated in protein folding. The active site of protein folding activity of the ribosome (PFAR) is in the domain V of the main RNA component of the large ribosomal subunit, which also constitutes the peptidyl transferase center. We have characterized the mechanism of PFAR using ribosomes or ribosome-borne folding modulators (RFMs) and human carbonic anhydrase I (HCA) as a model system. RFMs from all three kingdoms of life showed PFAR.  By multiple addition of the denatured protein in the refolding assay we demonstrate that the RFMs can recycle efficiently to assist refolding of a new batch of denatured protein. The turnover of the RFMs, which includes release of the protein substrate, takes milliseconds. Furthermore, fast kinetics of HCA refolding suggests that an early folding intermediate is the substrate for PFAR. Our results demonstrate for the first time that PFAR is catalytic. It was shown that two anti-prion drugs 6AP and GA specifically inhibit PFAR by binding to the domain V of the 23S / 25S rRNA. Using UV-crosslinking followed by primer extension we have identified the interaction sites of 6AP on domain V of 23S rRNA, which overlap with the protein binding sites, and are sensitive to mutagenesis. We find that 6AP and GA inhibit PFAR by direct competition with the substrate protein for the binding sites. Also, 6AP derivatives inhibit PFAR in the same order as their antiprion activity, 6AP8CF3 > 6AP8Cl > 6AP > 6APi. These results suggest involvement of PFAR in prion processes. To clarify the role of PFAR in prion processes, we studied HET-s prion aggregation in the presence of domain V/ IV/II of rRNA. The rRNAs, especially domain V rRNA not only reduced HET-s aggregation, but also changed the morphology of the HET-s fibrils, which became shorter and less compact. These results show that PFAR actively prevents large amyloid aggregation and thus, possibly influence prion propagation.

Two of the main organisers were the Swiss geometers Carl Friedrich Geiser (1843-1934) and Ferdinand Rudio (1856-1929). In addition to the congress, they also made valuable contributions to mathematical education, and in Rudio's case, the history of mathematics. Therefore, this thesis focuses primarily on these two mathematicians. As for Geiser, the relationship to his great-uncle Jakob Steiner is explained in more detail. Furthermore, his contributions to the administration of the Swiss Federal Institute of Technology are summarised. Due to the overarching theme of mathematical education and collaborations in this thesis, Geiser's schoolbook "Einleitung in die synthetische Geometrie" is considered in more detail and Geiser's methods are highlighted. A selection of Rudio's contributions to the history of mathematics is studied as well. His book "Archimedes, Huygens, Lambert, Legendre" is analysed and compared to E W Hobson's treatise "Squaring the Circle". Furthermore, Rudio's papers relating to the commentary of Simplicius on quadratures by Antiphon and Hippocrates are considered, focusing on Rudio's translation of the commentary and on "Die Möndchen des Hippokrates". The thesis concludes with an analysis of Rudio's popular lectures "Leonhard Euler" and "Über den Antheil der mathematischen Wissenschaften an der Kultur der Renaissance", which are prime examples of his approach to the history of mathematics.

Abstract:
This study deals with problems of identity of the office antiphons performed in the period after Pentecost. Mentioned antiphons belong to the New Testament canticle Benedictus for laudes and Magnificat for vespers. This part of the office is not so carefully regulated and between the European traditions occur variations. So this material was used for the determination of each tradition. The variability was not only in the selection and order of antiphons, but also in texts and melodies. After my analysis of the individual Post-Pentecost antiphons in several European medieval sources I found out that in certain cases with the same antiphon text appear completely different melodies or significantly different melodic variations. Due to this fact, in my opinion, is necessary to work with the musical part of this repertoire, consequently sort these chants not only according to the text, but also according to the melody. The ability to assign to the antiphons Melody ID based on the melody of chants (except Cantus ID based on the text) allows us to observe links between traditions which used this melody. In this way it is possible to reveal the relationships between the centers, which were performing the office repertoire. Abstract:
This dissertation describes the measurement of asymmetries in neutral B meson decays to two-body final states of charged pions and kaons. The results are obtained from a data sample of 383 million ϒ(4 S) → BB¯ decays collected between 1999 and 2006 with the BABAR detector at the PEP-II asymmetric-energy B factory located at the Stanford Linear Accelerator Center, California. The maximum likelihood fit that incorporates kinematical, event-shape, and particle identification information is used to measure the CP asymmetries in B0 → π +π− and K± π∓ decays. The direct CP-violating asymmetry between decays to K−π + is AKπ = −0.107 ± [special characters omitted]. The time-dependent CP-violating parameters in B0 → π+π− decays are Sππ = −0.60 ± 0.11 ± 0.03, Cππ = −0.21 ± 0.09 ± 0.02. For all the measurements above, the first error is statistical and the second is systematic.

Ring is a composition for orchestra (piccolo [doubling flute], two flutes, two oboes, English horn, two clarinets in B-flat, bass clarinet, two bassoons, contra-bassoon, four horns, four trumpets, two tenor trombones, bass trombone, tuba, three percussion parts [including glockenspiel, vibraphone, tubular bells, snare drum, toms, bass drum, suspended cymbal, drum kit, triangle, and gong], timpani, harp, piano, and standard strings) and spatially separated women's choir (SA right, SA left). This arrangement of media is intended to "ring" the audience with performers. At the notated tempo of two quarter-notes per second, the duration of the piece is exactly 17'40". The title of Ring comes from a poem of the same name written by the composer which is the principal text sung by the choir in the piece. The text of the poem is as follows: Wendy is a ring / A beginning and an end / Connected / The finest gold / Melted by touch / Cooled by breath / She fits my every finger / Without constraint / But permanent / Priceless, Precious, Beautiful / Alone / She clothes me. The poem and composition were written for, and dedicated to, the composer's wife. The ring metaphor ("ring" meaning cyclical, unending, complete) underlies many of the compositional choices in the work. Much of the surface of the music and its deeper structural elements are palindromes, which, because they end as they begin, have a circular nature. Also, the pitch structure of the piece involves the climactic completion of the "cycle" of the twelve available equal-tempered pitch classes. A further organizational element is the Fibonacci series, a mathematical construct which is used to determine small-scale rhythms and the duration of the larger sections of the work. These components, taken together, have resulted in a composition filled with variety and contrasts, which, nonetheless, is quite organically cohesive.

Abstract:
In 1290, members of the de Viane family donated a six-volume set of large, deluxe liturgical manuscripts to the Cistercian nuns of Beaupré in Grimminge, East Flanders. The three extant volumes and a later supplement are now known as the Beaupré Antiphoner (Baltimore, Walters Art Museum, W. 759-762). The nuns used, extensively revised, and supplemented the antiphoner for the next five hundred years until the abbey was suppressed in 1796 during the French Revolution. The manuscript offers a bottom-up perspective on the history of Cistercian chant in a women’s community. It also fills lacunae in the documentary sources related to reform and change in the history of the abbey. Revisions made in the late fifteenth century under the Observant movement suggest a revival of interest in St. Bernard and the “Bernardine” recension of Cistercian chant. Further alterations in the early modern period demonstrate that the nuns did not abandon their medieval chant tradition and adopt post-Tridentine versions until the late eighteenth century, approximately two hundred years after the publication of the Roman breviary of Pope Pius V (1568). The nuns viewed their carefully considered revisions as a necessary condition of continuity, not as a threat to it.

The diploma thesis consists of three parts. The first one describes the historical role of psalms during masses from their origin until presence. It illuminates the new role of psalms in the Cathlolic liturgy after the second Vatican council and outlines how these liturgical changes projected into Czech Catholic enviroments. The second part introduces significant Czech psalters and their authors. The psalters were created for liturgical needs and in reaction to the conclusions of the 2VC for liturgical needs. The third, main part of the thesis consists of detailed analysis of 1992 Karel Sklenička's psalter Responsoriální žalmy pro neděle a významné dny liturgického roku. The analysis of harmonic and melodic procedures of individual tones serves as a basis for the practival part of the thesis which is an extension of existing Sklenička's psalter to all days of liturgical year. The new psalter preserves the music component composed by Karel Sklenička and also preserves the way of work with the liturgical text in new psalms following the pattern of Karel Sklenička. The new psalter keeps strictly to the liturgically prescribed texts for individual liturgical occasions like the psalter from 1992.

Abstract:
Of the three manuscripts that form the basis of this thesis, MS Cape Town, South African Library, Grey 4c7 is, in musicological terms the most important of the three manuscripts. It is a complete Carthusian Antiphonary, of the late 14th century, written for the Charterhouse of Champmol, near Dijon, the mausoleum of the Dukes of Burgundy. It also contains an extensive Tonary, a Hymnary and a Kyriale. The two didactic verses which form part of the Tonary are of particular importance, since MS 4c7is one of the few manuscripts in the world intended for musical performance to contain the Ter terni by William of Hirsau; furthermore it is apparently the only Carthusian manuscript of any kind to contain the Oyapente et dyatessaron by Hucbald. The manuscript is placed in the context of the Carthusian liturgy of the 12th to the 16th centuries and is compared with 33 manuscripts of this period. It is shown that, although a marked textual similarity exists between the manuscripts, there are variant melodies. The conclusion is therefore drawn that the Carthusians did not have a single exemplar for the melodies in their liturgical books. It is shown that MS 4c7 and MS Oijon, Bibliotheque municipale 118, also written for Champmol, were copied from the same exemplar and that they are closely related to MSS Beaune, Bibliotheque municipale 27, 34 and 41, ot the neighbouring Charterhouse of Fontenay. The second manuscript, MS Grey 3c23, an Antiphonary for nuns, for Lauds and Vespers, written for the Charterhouse of Mont-Sainte-Marie, at Gosnay, near Arras, has been dated 1538 by the original scribe. This manuscript is almost identical to MS AGC C II 817. The presence of a Sequence, foreign to the Carthusian tradition, is however unique toMS 3c23. The third manuscript, MS Grey 6b3, is an Evangeliary, signed by the scribe, Amelontius de Ercklems, in 1520. Its provenance is the Charterhouse of Our Lady of the Twelve Apostles at Mont-Cornillon near Liege. Musicological features of the manuscript which are discussed are the Hymn 'Te decet laus', and the accent neumes at the ends of pericopes.
