Abstract:
Bisphosphonates (BP) are drugs currently administered orally to treat diseases characterised by an excessive bone resorption. Alternative and more efficient delivery routes and more potent compounds are being investigated. Three implantable delivery systems, which allow the controlled release of therapeutic agents from the device core, are examined in this paper. (4- (aminomethyl) benzene) bisphosphonic acid (ABBP) was incorporated on Ca8.8Na0.8(PO4)4.8(CO3)1.2(OH)0.4F1.6 particles by refluxing the powder in a 60 mmol suspension in acetone at 60°C for 5 hours. 4-aminophenyl acetic bisphosphonate monosodium salt (APBP) and 1- H-indole-3-acetic bisphosphonate monosodium (IBP) were loaded on Ca10(PO4)6(OH)1F1 ceramic bodies by stirring the ceramic bodies in 0.04M BP solutions. Injectable acrylic cements based on self-curing formulations of methyl methacrylate (MMA) and vitamin E were loaded with APBP and IBP. The incorporation of ABBP was confirmed by MAS-NMR spectroscopy. Modified powder shows two different phosphorous environments, the first one at 2.91 ppm can be assigned to the apatite base and the second one at 18.0 ppm has to be attributed to the phosphonic group of the ABBP. The IBP addition on ceramic surfaces did not decrease the number of osteoclast colonies and appeared to improve the performance of the HA as a surface for osteoblast culture. A therapeutic dosage of APBP and IBP can be achieved from acrylic cements that showed lack of toxicity and an increased cellular activity and proliferation.

Abstract:
Bisphosphonates (BPs) may play an important role in minimizing osteolysis. In this work two new bisphosphonates pertaining to second and third generations respectively, have been synthesized and incorporated onto a chemically enriched hydroxyapatite. BP synthesis has been performed by adding H3PO3, PCl3 and methanesulfonic acid over 4-aminophenyl acetic acid (APBP) and 1-H-indole-3-acetic acid (IBP) respectively at 65°C in a N2 atmosphere. These compounds bear a primary amine group bonded to an aromatic ring, and a secondary amine group within a heterocyclic ring respectively. A chemically enriched hydroxyapatite with a chemical content corresponding to a 50% fluorided hydroxyapatite has been synthesized. Ceramic bodies manufactured by uniaxial pressure followed by cold isostatic press have a 97% density and submicron grain size. The BP was adsorbed onto the surface by immersion in a stirred solution at 37°C for 48 hours. A 10-fold decrease of the surface energy was observed for bodies modified with the APBP whereas only a 25 % decrease is obtained for bodies loaded with the bisphosphonate loaded with the IBP.

Most bacterial cells are surrounded by an essential cell wall composed of the net-like heteropolymer peptidoglycan (PG). Growth and division of bacteria are intimately linked to the expansion of the PG meshwork and the construction of a cell wall septum that separates the nascent daughter cells. Class A penicillin-binding proteins (aPBPs) are a major family of PG synthases that build the wall matrix. Given their central role in cell wall assembly and importance as drug targets, surprisingly little is known about how the activity of aPBPs is controlled to properly coordinate cell growth and division. Here, we report the identification of MacP (SPD_0876) as a membrane-anchored cofactor of PBP2a, an aPBP synthase of the Gram-positive pathogen Streptococcus pneumoniae. We show that MacP localizes to the division site of S. pneumoniae, forms a complex with PBP2a, and is required for the in vivo activity of the synthase. Importantly, MacP was also found to be a substrate for the kinase StkP, a global cell cycle regulator. Although StkP has been implicated in controlling the balance between the elongation and septation modes of cell wall synthesis, none of its substrates are known to modulate PG synthetic activity. Here we show that a phosphoablative substitution in MacP that blocks StkP-mediated phosphorylation prevents PBP2a activity without affecting the MacP–PBP2a interaction. Our results thus reveal a direct connection between PG synthase function and the control of cell morphogenesis by the StkP regulatory network.

Breeding for improved blanchability—the propensity of the testa (skin) to be removed from the kernel following rapid heat treatment—is a priority for improvement in the Australian Peanut Breeding Program (APBP). Easy removal of the testa by blanching is required for processing of peanuts into peanut butter and various other confectionary products. Thus, blanchability is an economically important trait in any newly released cultivar in Australia. A better understanding of the range of genetic variation, nature of inheritance and genotype×environment (G×E) interactions, and the development of a low-cost method to phenotype in early generations, could speed up breeding for this trait. Studies were conducted to develop a low-cost, rapid method utilising minimal amounts of seed to phenotype in early generations, along with an assessment of G×E interactions over a range of years and environments to derive optimal selection protocols. Use of a smaller kernel sample size than standard (50 vs 200g) was effective for accurately assessing blanchability in breeding lines and could allow selection in early generations (e.g. in seed produced from a single F2 plant where seed supply is adequate). G×E interaction for blanchability was shown to be very low. Genotypic variance explained 62–100% of the total variance for blanchability, assessed in two diverse germplasm pools including 107 accessions in the USA mini-core over three environments and multiple APBP breeding lines grown over nine different years–environments. Genotypic correlations between all environments were very high (~0.60–0.96), with heritability for the blanchability trait estimated to be very high (0.74–0.97) across the 13 trials. The results clearly demonstrate that effective selection for improved blanchability can be conducted in early generations and in a limited number of contrasting environments to ensure consistency of results.

The various mechanisms leading to the appearance of horizontal inhomogeneities at the surface of ApBp stars are critically reviewed. The effect of magnetic fields is essential but is it more to control locally, on the surface, the appearance of anomalies that are caused by the vertical transport of particles, or is it to create directly the horizontal inhomogeneities by horizontal transport? The time scales for horizontal and vertical transport will be discussed in this context.The processes discussed include: a) ambipolar diffusion of protons and hydrogen in the presence of magnetic fields; b) the guiding of diffusion by magnetic fields; c) the horizontal component of radiative accelerations; d) mass loss; e) light induced drift.

A cell wall made of the heteropolymer peptidoglycan (PG) surrounds most bacterial cells. This essential surface layer is required to prevent lysis from internal osmotic pressure. The class A penicillin-binding proteins (aPBPs) play key roles in building the PG network. These bifunctional enzymes possess both PG glycosyltransferase (PGT) and transpeptidase (TP) activity to polymerize the wall glycans and cross-link them, respectively. In Escherichia coli and other gram-negative bacteria, aPBP function is dependent on outer membrane lipoproteins. The lipoprotein LpoA activates PBP1a and LpoB promotes PBP1b activity. In a purified system, the major effect of LpoA on PBP1a is TP stimulation. However, the relevance of this activation to the cellular function of LpoA has remained unclear. To better understand why PBP1a requires LpoA for its activity in cells, we identified variants of PBP1a from E. coli and Pseudomonas aeruginosa that function in the absence of the lipoprotein. The changes resulting in LpoA bypass map to the PGT domain and the linker region between the two catalytic domains. Purification of the E. coli variants showed that they are hyperactivated for PGT but not TP activity. Furthermore, in vivo analysis found that LpoA is necessary for the glycan synthesis activity of PBP1a in cells. Thus, our results reveal that LpoA exerts a much greater control over the cellular activity of PBP1a than previously appreciated. It not only modulates PG cross-linking but is also required for its cognate synthase to make PG glycans in the first place.

Abstract:
We provide a formal mathematical definition of the Shortest Paths for All Flows (SP-AF) problem and provide many efficient algorithms. The SP-AF problem combines the well known Shortest Paths (SP) and Bottleneck Paths (BP) problems, and can be solved by utilising matrix multiplication. Thus in our research of the SP-AF problem, we also make a series of contributions to the underlying topics of the SP problem, the BP problem, and matrix multiplication. For the topic of matrix multiplication we show that on an n-by-n two dimensional (2D) square mesh array, two n-by-n matrices can be multiplied in exactly 1.5n ‒ 1 communication steps. This halves the number of communication steps required by the well known Cannon’s algorithm that runs on the same sized mesh array. We provide two contributions for the SP problem. Firstly, we enhance the breakthrough algorithm by Alon, Galil and Margalit (AGM), which was the first algorithm to achieve a deeply sub-cubic time bound for solving the All Pairs Shortest Paths (APSP) problem on dense directed graphs. Our enhancement allows the algorithm by AGM to remain sub-cubic for larger upper bounds on integer edge costs. Secondly, we show that for graphs with n vertices, the APSP problem can be solved in exactly 3n ‒ 2 communication steps on an n-by-n 2D square mesh array. This improves on the previous result of 3.5n communication steps achieved by Takaoka and Umehara. For the BP problem, we show that we can compute the bottleneck of the entire graph without solving the All Pairs Bottleneck Paths (APBP) problem, resulting in a much more efficient time bound. Finally we define an algebraic structure called the distance/flow semi-ring to formally introduce the SP-AF problem, and we provide many algorithms for solving the Single Source SP-AF (SSSP-AF) problem and the All Pairs SP-AF (APSP-AF) problem. For the APSP-AF problem, algebraic algorithms are given that utilise faster matrix multiplication over a ring.

AbstractThis dissertation aims to investigate the use made of music apps for tablets in the preschool environment in Sweden, and the potential it offers.The background of this subject is of double nature. On one hand, the preschool curriculum stresses the implementation of information technology and musical experiences into everyday activity. On the other hand, the implementation of information technologies in the shape of mobile devices in Swedish preschools is a relatively recent phenomenon which implies the need of a deeper focus on its didactic potential.The perspective chosen for the investigation and analysis of the subject is music didactics. On this basis, the research questions are formulated as follows: 1. Which music apps are used in preschools? 2: Which musical dimensions emerge? 3. How are the music apps used? 4. Who uses them?The study is of qualitative type and is conducted through an online enquiry for preschool educators, the results of which are compared to the analysis of the music apps that the enquiry’s respondents report.The findings show how the majority of the music apps used focuses on production and music, in contrast to the singing reproduction of traditional music teaching. In addition, they reveal that music apps are used to a higher extent with the older preschool children than with the younger ones.These findings mean that the use of music apps can be a valid complement to music learning for children whose educators do not have sufficient musical knowledge. Moreover, they suggest that a knowledge of music and music apps by educators can support their use not only with older preschool children, but with the younger ones too. Implementation of this subject into university programs is a possible way to improve knowledge and make everyday preschool activity comply the curriculum’s prescriptions.

Barn får tidigt i livet komma i kontakt med surfplattor och de används numera i de flesta förskolor. Forskning har dock visat att förskollärare har varierande åsikter kring om och hur surfplattor bör användas i förskolans verksamhet. Syftet med detta arbete är därför att undersöka vilka erfarenheter och uppfattningar förskollärare har av att arbeta med surfplattor i förskolan och vad surfplattorna i sådana fall används till. Jag har som metod valt att föra intervjuer. Fyra intervjuer har gjorts med förskollärare som berättar kring sina erfarenheter och uppfattningar om hur det är att arbeta med surfplattor inom deras verksamhet. Resultatet visar att förskollärarna i relativt liten utsträckning använder surfplattor i verksamheten. Vid tillfällen de ändå användes, användes de främst till pedagogisk dokumentation och som ett verktyg för att få ett lugnare klimat i barngruppen. Vad som också framkom i resultatet var att förskollärarna var intresserade av fortbildning i att använda surfplattor, för att på så vis kunna använda surfplattorna mer tillsammans med barnen.

Lee [Parte II]<br>En una industria que mueve alrededor de 22 millones de dólares al año y que crece más del 6% anual, existe una necesidad latente en el mercado de licores en Panamá que actualmente no está siendo cubierta, pues el 71% de las personas entre 18 y 35 años de edad, residentes de las zonas metropolitanas de Panamá de estratos medio-altos, recurren a los supermercados, para ir en busca de licor u otros artículos en medio de una fiesta, arriesgándose a tener accidentes de tránsito, si se encuentran en estado de embriaguez. Este hecho se hace aún más evidente cuando cerca del 82% de las personas manifiesta que no conoce un servicio que le brinde al mismo tiempo comodidad, seguridad y rapidez en la venta de productos a la hora de realizar celebraciones sociales en sus casas u oficinas, y que cerca del 80% estaría dispuesto a pagar por un servicio que se lo brinde. Investigaciones realizadas para este proyecto resaltan la necesidad latente de un mercado que busca un servicio el que le pueda brindar en la comodidad de sus casas u oficinas, bebidas alcohólicas, comestibles y artículos complementarios para fiestas. El mercado objetivo se centra en residentes del área metropolitana de la ciudad de Panamá que tengan entre 18 a 35 años y que tengan una vida social activa. La industria en la que se está apostando muestra ser solida, con un crecimiento anual en ventas del 6%. Es considerada dinámica y estable como también siendo la de mayor consumo per cápita a nivel latinoamericano. Con este servicio se esperan ingresos cerca del millón de dólares anuales, un VAN positivo de $408.190, una TIR de 75%, un Payback en el año 3, y un margen neto promedio de 17%. Concluyendo, información proporcionada por los estudios realizados cerca del 80% del mercado objetivo utilizaría PartyAPP como una solución a las necesidades de este mercado. Se destacan como atributos la seguridad, rapidez y comodidad, siendo los antes mencionados objetos diferenciadores de este servicio. De esta manera, se comprueba la viabilidad de este proyecto dentro del mercado.
