Das vorliegende dreibändige Werk stellt die Entwicklungsgeschichte von ca. 5300 Siedlungsnamen (einschließlich der Wüstungsnamen) dar, die in den Grenzen des heutigen Freistaates Sachsen überliefert wurden. Hinzu kommt der Namenbestand des thüringischen Kreises Altenburg (Altenburger Land), der siedlungs- und territorialgeschichtlich jahrhundertelang zum ehemaligen Reichsterritorium Pleißenland (in Westsachsen) und zum wettinischen Territorialstaat gehörte.
Die schriftliche Überlieferung der Namen von Siedlungsplätzen im Raum östlich der Saale beginnt – abgesehen von seltenen Ausnahmen der fränkischen Zeit – mit der Errichtung der deutschen Oberherrschaft über das seit etwa 600 von (Alt-)Sorben besiedelte ostsaalisch-ostelbische Gebiet durch König Heinrich I. in den Jahren 929/930. Rund 60% der sächsischen Ortsnamen sind slawischer Herkunft, wobei dieses Sprachgut in der Mehrheit nicht von Slawen, sondern von Deutschen aufgezeichnet wurde.
In den ersten beiden Bänden wird die in einer möglichst lückenlosen Belegkette erfasste Geschichte jedes einzelnen Namens dargestellt: von den ältesten urkundlich tradierten Schreibungen über die Entwicklung seiner weiteren Formen bis zur heutigen Gestalt einschließlich seiner mundartlichen Lautung. Geboten werden Informationen zu Herkunft, Wortbildung, Etymologie und Typologie, zu sprachgeschichtlichen Sachverhalten, Problemen der Integration slawischer (altsorbischer) Toponyme ins Deutsche sowie zu soziolinguistischen Merkmalen der Siedlungsbenennungen. Der dritte Band enthält neben dem Apparat auswertende Register, die das lexikalische Material detailliert erschließen.

This Bachelor thesis is a qualitative study of the public debate regarding the role of Public service in the media converged society today. The purpose of this thesis is to examine how the public debate, that is the Swedish Public service television and people with broad representative knowledge about Public service, motivates a change of the function of the Swedish Public service television. Our issues are: What does the public debate say about the function of Swedish Public service television in our society today and how is a change motivated in this debate? How does the public debate look upon today´s media convergence and the fragmentation of the audience which, according to Denis McQuail, is a consequence of this? The method of this thesis is a qualitative analysis and we have analyzed ten opinion articles and Swedish Public service television magazine “DittSvt”. The theoretical background was obtained from Henry Jenkins and Håkan Hvitfelts theories and thoughts about mediaconvergence. We have supplemented this with Denis McQuails theory regarding the fragmentation of the audience. In this thesis, we see that the public debate is a lot about diversity in Swedish Public service television programming and that the democratic base is still very important. We also see that the audience are not just viewers, as in the beginning of television broadcasting, but have become users of media. The analysis shows that public service is important for our society today and that it is something that should be retained but should be developed and shaped based on our media converged society.

Sammanfattning Det här projektet handlade om att konstruera en effektmätare. Projektet var en del av ett större projekt som heter Wattch som drevs av forskningsinstitutet Interactive Institute. Tanken med projektet Wattch var att elförbrukningen ska jämföras med bensinförbrukningen i en bil. Utgående från en kravspecifikation så skulle en effektmätare konstrueras. Det första som gjordes var en projektplan som beskriver idé, mål och tidplan för projektet. Efter det så gjordes förstudier om hur effekt beräknas, hur fasströmmen kan mätas och om information om trådlöst nätverk. Det studerades också vad för effektmätare som finns på marknaden och hur de fungerar. Efter lite förstudier så bestämdes hur effektmätaren skulle konstrueras för att uppfylla kravspecifikationen. Sedan gjordes en funktionsbeskrivning av en effektmätare. Där beskrevs lite grovt hur effektmätaren kan konstrueras. Därefter beskrevs hela konstruktionen i detalj vilket innefattar hårdvara, mjukvara och mekanik. Sedan gjordes lite olika tester för att jämföra två olika mätare och för att konstatera att konstruktionen fungerade enligt kravspecifikationen. Projektet Wattch blev klart och ett större test utfördes på två hushåll i Hudiksvall. Deras momentana effektförbrukning kunde då följas på en dator som var uppkopplad på Internet. På datorn kördes ett program i flash som var utvecklat av företaget XCOM. På datorn syntes Figur 13 där visarna uppdaterades kontinuerligt från en databas. Rapporten avslutas med ett antal förbättringar som skulle kunna göras med effektmätaren. Mätaren skulle kunna göras mer noggrann och minska effektförbrukningen av effektmätaren.

In dieser Arbeit wurden 8 und 16 Wochen alte, hypertensive "fawn-hooded" Ratten (FHH8, FHH16) mit genetisch ähnlichen 16 Wochen alten "fawn-hooded" Ratten mit nur geringgradiger Blutdruckerhöhung (FHL16, Kontrollgruppe) hinsichtlich der pathologisch-anatomischen Veränderungen der Nierenmorphologie und hinsichtlich der Expression von NO-Synthase-1 (NOS1), Cyclooxygenase-2 (COX-2) und Renin am juxtaglomerulären Apparat (JGA) verglichen. Die histopathologischen Veränderungen bei FHH16 umfassten die klassischen Schädigungszeichen der fokal segmentalen Glomerulosklerose (FSGS) mit fokaler Überexpression von Kollagen IV und eine moderate Arteriolopathie. Bei FHH8 ließen sich, wie bei FHL16 keine morphologischen Schädigungen nachweisen. Die NOS1-Aktivität an der Macula densa, untersucht mittels der NADPH-Diaphorasereaktion und die NOS1 mRNA Expression waren bei FHH8 (+153 and +88%; P < 0.05) und FHH16 (+93 and +98%; P < 0.05) im Vergleich zu FHL16 signifikant erhöht. Eine gleichgerichtete signifikante Erhöhung zeigte sich für die COX-2-Expression an der Macula densa von FHH8 (+166%; P < 0.05) und FHH16 (+157%; P < 0.05) im Vergleich zu FHL16. Des weiteren ließ sich eine signifikante, ebenfalls gleichgerichtete Überexpression von Renin in der afferenten Arteriole auf Protein- und mRNA-Ebene bei FHH8 (+51 and +166%; P < 0.05) und FHH16 (+105 and +136%; P < 0.05) im Vergleich zu FHL16 nachweisen. Somit konnte gezeigt werden, dass die gleichgerichtete Überexpression von NOS1, COX-2 und Renin am JGA bei der FHH-Ratte der Entwicklung einer fokal segmentalen Glomerulosklerose vorausgeht und damit möglicherweise pathogenetische Bedeutung für die Entstehung der Nierenschädigung bei diesem Rattenstamm hat.This study describes elevated histochemical signals for nitric oxide synthase-1 (NOS1) and cyclooxygenase-2 (COX-2) in juxtaglomerular apparatus (JGA) and adjacent thick ascending limb of the kidney of fawn-hooded hypertensive rats (FHH). Two different age groups of FHH (8 and 16 wk; FHH8 and FHH16, respectively) were compared with genetically related fawn-hooded rats with close to normal blood pressure (FHL) that served as controls. Histopathological changes in FHH16 comprised focal segmental glomerulosclerosis (FSGS), focal matrix overexpression (mainly of collagen IV), and a moderate arteriolopathy with hypertrophy of the media, enhanced immunoreactivity for alpha-smooth muscle actin, and altered distribution of myofibrils. Macula densa NOS activity, as expressed by NADPH-diaphorase staining, and NOS1 mRNA abundance were significantly elevated in FHH8 (+153 and +88%; P < 0.05) and FHH16 (+93 and +98%; P < 0.05), respectively. Even higher elevations were registered for COX-2 immunoreactivity in FHH8 (+166%; P < 0.05) and FHH16 (+157%; P < 0.05). The intensity of renin immunoreactivity and renin mRNA expression in afferent arterioles was also elevated in FHH8 (+51 and +166%; P < 0.05) and FHH16 (+105 and +136%; P < 0.05), respectively. Thus we show that coordinate upregulation of tubular NOS1, COX-2, and renin expression precedes, and continues after, the manifestation of glomerulosclerotic damage in FHH. These observations may have implications in understanding the role of local paracrine mediators in glomerular disease.

Méconnue, quand elle n'est pas tout simplement ignorée, la monumentale chanson des Enfances Renier est parmi les dernières du Cycle de Guillaume à avoir fait l'objet d'une édition. Il serait hâtif de voir dans la désaffection des éditeurs et des critiques pour ce texte le seul effet de ses dimensions généreuses. Affligées d'une réputation exécrable, les Enfances Renier ont surtout pâti du discrédit qui a longtemps frappé l'épopée tardive. Empruntant son schéma narratif et certains de ses personnages au répertoire folklorique, accumulant sans retenue les rebondissements les plus invraisemblables, usant de l'entrelacement jusqu'à en étourdir le lecteur et accumulant les "éléments aventureux et fantastiques", la chanson n'avoue que trop une jubilatoire propension à succomber "aux charmes frelatés du romanesque". Dès 1852, P. Paris réduit arbitrairement l'oeuvre à de "ridicules inventions". Cent ans plus tard, J. Frappier se contente d'une notice laconique teintée de condescendance : "ce long poème d'aventures, qui a la réputation d'être détestable [. . . ], nous ramène vers la geste de Rainouart. Renier est en effet le fils de Maillefer et de Florentine, par conséquent le petit-fils de Rainouart. Le théâtre de ses exploits est la Sicile et la Grèce. De son mariage sans originalité avec la Sarrasine Ydoine (ce n'est qu'un jeu de mots) naît Tancrède, l'un des héros de la première croisade. Borrowing its narrative structure and certain of its characters from the folk, accumulating without reserve the most incredible bounces, abrasive repertory of interlacing until dazing the reader of it and accumulating the "adventurous and fantastic elements", the song acknowledges only too a jubilatoire propensity to succumb "to the charms adulterated of romance". Since 1852, P. Paris arbitrarily reduces work to "ridiculous inventions". Hundred years later, J. Frappier is satisfied with a tinted laconic note of condescension : "this long poem of adventures, which with the reputation to be hateful [. . . ], brings back for us towards the epic of Rainouart. To disavow is indeed the son of Maillefer and Florentine, consequently the grandson of Rainouart. The theatre of its exploits is Sicily and Greece. From its marriage without originality with Sarrasine Ydoine (it is not a pun) is born Tancrède, one of the heroes of the first crusade. "Forty years later, the song still does not cause a frank passion : M. Tyssens and J. Wathelet-Willem qualify Enfances Renier of "epigone deprived of any originality". L'établissement du texte et de l'apparat critique tient compte des récentes découvertes paléographiques et introduit deux manuscrits négligés, t et v. La traduction essaie de rendre compte du style maniériste de Sénèque, met en valeur les images et les effets rhétoriques. Les notes justifient le choix des leçons, éclairent quelques problèmes mythologiques, historiques, littéraires. L'introduction présente les différents manuscrits utilisés, les éditions anciennes et modernes. Elle étudie la question des sources en insistant sur les sources latines (ennius et accius), puis celle de la dramaturgie en montrant que Sénèque s'attache plus aux sentiments et aux états d'âme de ses personnages qu'à l'intrigue elle-même. Elle suggère ensuite que le style de Sénèque est maniériste et étudie les différentes figures de style utilisées par l'auteur. Elle fait enfin le point sur la versification dans Les TroyennesThe text and the critical apparatus make allowance for the recent paleographic discoveries. The apparatus introduces two disregarded manuscripts, t and v. The translation tries to account for Seneca’s mannerism, enhances the images and the rhetorical figures. The notes justify the choices of the readings and throw light on some mythological, historical and literary problems. The introduction presents first the different manuscripts used for the apparatus, and then the ancient and modern editions. It contains a study of the sources, which underlines the Latin sources, and an analyze of the dramatic problems which shows that Seneca is more interested in his characters' feelings than in the plot itself. Grouting is a common method used to seal rocks around tunnels, successful grouting reduces the duration and cost of the construction, guarantees better working environment and higher safety, minimizes the maintenance and most important decreases the corresponding environmental hazards significantly. Achieving a sufficient grout spread is one of the prerequisites for a successful and efficient sealing, the penetration of a grout is defined as the length of how far grout penetrates in the rock through fractures from a bore hole. Chemical grouts and cement-based grouts are the prevailing ones among the grouting materials. Despite the better penetrability of chemical grouts, they are unfavorable to use due to environmental hazards associated to them, whilst cement-based grouts are more convenient to use because of their low cost and low environmental impact.The major drawback with cement-based grouts is their limited ability to penetrate the very narrow fractures which is directly related to their filtration tendency which is defined as the tendency of cement grains to agglomerate and build an impermeable filter cake during the flow. Many previous studies investigated the factors that affect the filtration tendency. They drew different conclusions and suggested various methods to improve the penetrability of cement-based grouts.The mixing method is one of the factors that have a great influence on the penetrability of the grout. An effective mixing method improves the dispersion of cement particles in the mixture, thus the penetrability of the grout. As it is known from previous studies, the finer the cement particles the harder to disperse. Grouts based on micro-fine cement (< 30 μm) are essential for the development of grouts that can seal very narrow fractures (20-50) μm compared to (70-80) μm at the present.In this study, the dispersion efficiency of three different mixing methods was evaluated, a conventional lab dissolver equipped with 90-mm disk, a conventional lab dissolver equipped with R/S system and an ultrasound UP400St device. Two cement types, INJ30 and UF12, that are similar in chemical composition but differ in degree of milling were tested. Dispersion was tested with filter pump.The results showed that the conventional lab dissolver equipped with 90-mm disk is ineffective method. The conventional lab dissolver equipped with R/S system is a better method compared to the 90-mm disk but still not effective enough especially when it comes to grouts based on ultra-fine cement (UF12). The ultrasound dispersion is not only the best method between the three methods in comparison, but even more stable and reliable. The best result obtained was grout based on UF12 passing through the 54 μm filter. This could mean that fracture aperture down to 55 μm now can be sealed. This is a significant improvement but there is still a marginal for further improvements. In combination with the dispersion efficiency of different dispersion methods, the study investigated the effect of additives on dispersion in particular and penetrability in general. Results showed that additives do not directly contribute to better dispersion, but they are necessary for better spread since they affect the flow properties.

Legionella pneumophila ist der Erreger der Legionärskrankheit. Die Pathogenität des Bakteriums basiert auf seiner Fähigkeit innerhalb menschlicher Lungenzellen zu überleben und sich zu vermehren. Demzufolge ist L. pneumophila nicht nur interessant als wichtiges Pathogen, sondern kann auch als Sonde verwendet werden, um allgemeine intrazelluläre Ereignisse zu untersuchen. Ein Beispiel hierfür ist die, durch das Pathogen gestörte, intrazelluläre Kommunikation zwischen den Organellen des endoplasmatischen Retikulums (ER) und dem Golgi Apparat (GA). In der vorliegenden Studie schlagen wir ein neues Modell vor, wie das Bakterium erfolgreich seine replikative Nische, die Legionella Vakuole (LV), innerhalb des Zytosols aufbauen könnte, um seine Ausbreitung zu garantieren. Um die Mechanismen für die erfolgreiche Ausbeutung der Wirtszelle gezielt untersuchen zu können, haben wir mit Hilfe von siRNA spezifisch verschiedene Wirtszellproteinen herunterreguliert und den Einfuß der Abwesenheit dieser Proteine auf die Vermehrung von L. pneumophila gemessen. Die Ergebnisse wiesen darauf hin, dass die LV möglicherweise den Golgi Apparat imitiert und auf diese Weise den zellulären Vesikeltransport umleitet. Diese Theorie wurde durch in silico Ergebnisse unterstützt, die in der Proteinsequenz des Legionella Effektor-Proteins LidA, das auf der Vakuole lokalisiert ist, ein SNARE-ähnliches Motiv zeigte. Dies weist auf ein auf der Vakuole lokalisiertes SNARE-Erkennungsmotiv hin, das notwendig sein könnte, um zelluläre Transportvesikel zu koppeln. Aus dem Wissen heraus, dass L. pneumophila in der Lage ist, die Aktivierung der zellulären Proteine Arf1 und Rab1 durch Phosphorylierung und Dephosphorylierung zu regulieren, machten wir uns auf die Suche nach Proteinen, die auf Infektion hin modifiziert werden. Die Kommunikation von Wirt und Pathogen über Phosphorylierung ist bekannt im Bezug auf pathogenspezifische Modifikation des Zytoskeletts und Signalkaskaden in der Anti-Apoptose. Für diese Studie wurde ein Antikörper verwendet, der spezifisch phosphorylierte Tyrosinreste erkennt. Dies resultierte in der Detektion einer Serin-Threonin-Kinase in der Amöbe Acanthamöba castellanii, die an einem Tyrosinrest phosphoryliert ist. Diese Amöben-Kinase wies in silico Homologie zu der humanen GS-Kinase 3 des Wnt-Signalwegs, bekannt aus der Forschung der embronalen Entwicklung bei Drosophila, auf. Der letzte Teil dieser Arbeit konzentrierte sich auf die, durch eine L. pneumophila-Infektion ausgelöste, anti-apoptotische Signalkaskade. Es ist bekannt, dass auf eine Infektion hin NF-kappaB aktiviert wird. Dies führt dazu, dass p65 in den Zellkern wandert und dort als Transkriptionsfaktor aktiv wird. Diese Translokation geschieht in 2 zeitversetzten Phasen. Eine Aktivierungsspitze wird nach dem Kontakt mir bakteriellem Flagellin gemessen, gefolgt, von einer dauerhaften Aktivierung, abhängig von einem funktionierenden Dot/ Icm Typ-IV-Translokationssystem. In dieser Arbeit stießen wir auf eine L. pneumophila Mutante, die den Dot/ Icm-Effektor SdbA nicht bildet, und die daraufhin NF-appaB nicht aktivieren kann. Diese Mutante war ebenfalls nicht in der Lage, sich in Epithelzellen zu vermehren. Dies ist außergewöhnlich, da das L. pneumophila Effektor Repertoire so redundant ist, dass die Abwesenheit eines einzigen Effektors selten einen so starken Einfluss auf die Replikation hat. All diese Ergebnisse zeigen zusammengenommen, auf wie vielen verschiedenen Ebenen L. pneumophila in der Lage ist, seine Wirtszelle zu manipulieren, um einerseits die nötige Nische für seine Vermehrung zu etablieren und andererseits die Zelle am Selbstmord zu hindern. Dies geschieht durch Imitation zellulärer Prozesse.Legionella pneumophila is the causative agent of Legionnaires´ disease. The bacterium’s pathogenicity is based on its ability to survive and multiply efficiently inside human alveolar cells. Therefore, L. pneumophila is not only an important pathogen, but can also be used as a probe to investigate host cell function as for example, in the cellular trafficking pathway. In this study, we establish a new model of how this pathogen efficiently constructs its replicative niche, the Legionella containing vacuole (LCV), inside the host cytosol, enabling its dissemination. To investigate the mechanisms that lead to effective exploitation of the host cell, we down-regulated specific host cellular proteins via siRNA technology and measured the subsequent impact on L. pneumophila replication. The results suggest that the LCV mimicks the Golgi apparatus and via this mechanism hijacks host cellular vesicular trafficking. The L. pneumophila secreted effector protein LidA, located within the LCV, is shown to have a SNARE-like motif, suggesting a SNARE like sole connected to the LCV. Since it is known that cellular signalling proteins are controlled via phosphorylation and dephosphorylation, we went on to search for specifically modulated host cell proteins after L. pneumophila infection. The cross-talk of the pathogen with its host via phosphorylation has been connected to several sub-cellular activities leading to, for instance, cytoskeleton rearrangement and signalling events including anti-apoptosis pathways. Here we used a phosphorylated tyrosine antibody resulting in the detection of an amoeba serine-threonine-kinase, phosphorylated at its tyrosine residue. This kinase shows homologies to the human GSK3 of the wnt-signalling pathway. (“Wnt“ is merged from the names of the homologues genes Wg (Drosophila melanogaster) and Int (mouse) both employed in evolutionary developement.) The final part of this work concentrated on anti-apoptotic signalling events induced upon L. pneumophila infection. It is known that during L. pneumophila infection the activation of NF-kappaB and subsequent translocation of p65 from the cytosol into the nucleus follows a biphasic pattern. One short peak of activation is induced upon contact with bacterial flagellin, succeeded by a permanent Dot/ Icm type IV secretion system-dependent activation. In this study, we found the L. pneumophila mutant lacking the Dot/ Icm effector SdbA to be unable to activate NF-kappaB. This mutant also showed impaired growth in epithelial cells. This is remarkable due to the high redundancy of the L. pneumophila effector system, meaning deletion of a single effector rarely has such a big impact on replication. Taken together this work demonstrates, the manifold ways in which L. pneumophila on the one hand side establishes its niche to ensure replication and on the other hand side to bars its host cell from suicide. All of this is managed by mimicking cellular processes.

Allt fler skolor satsar idag på att införa digital teknik som ett verktyg i undervisningen, vilket innebär att elevers tillgång till en dator eller surfplatta har ökat. Även om den digitala tekniken har potential att förbättra lärandet, om den används på ett medvetet sätt, sker användandet av den här teknik relativt sparsamt i dagens matematikundervisningen. Den digitala tekniken har således god potential att påverka lärandet positivt men det är viktigt att införandet av tekniken görs med ett tydligt definierat syfte av det som har för avsikt att läras ut och hur det ska ske. Det är en utmaning, inte minst då forskning om hur appar kan användas i matematikundervisningen är begränsad. Syftet med litteraturstudien är att bidra med kunskap om vilken funktion appar kan fylla för matematikundervisningen i de lägre åldrarna. Litteraturstudien har en kvalitativ ansats där innehållet i 9 tidskriftsartiklar och 2 konferensbidrag har granskats och tolkats. Vid materialanalysen arbetades olika teman fram som sedan användes för att presentera resultatet. Resultatet visade att användning av appar kan bidra till att höja nivån av motivation och engagemang hos elever. Det framgick även att appar erbjöd en del unika möjligheter som kan vara svåra att uppnå med traditionell undervisning. Till de här hör möjlighet till omedelbar feedback och vägledning, individanpassning och användning av flera sinnen. Slutsatsen är att användning av appar kan ha potential att stärka elevers lärande. Lärare har en viktig roll i att kunna avgöra när en app är ett lämpligt verktyg för att uppnå lektionsmålet eller när traditionell undervisning lämpar sig bättre.

Chlamydien sind obligat intrazelluläre Bakterien, die sich in einem membranumschlossenen Kompartiment namens Inklusion vermehren. Nach Infektion fragmentiert der Golgi-Apparat der Wirtszelle in kleine Membranstapel. Dies verbessert die Aufnahme von Sphingolipiden und ist deshalb für die chlamydiale Vermehrung essentiell. Die infektionsinduzierte Golgi-Fragmentierung geschieht nach Spaltung des Golgi-Matrix-Proteins Golgin-84. In dieser Arbeit konnte, durch den Vergleich mit bekannten Substraten und Inhibitorstudien, die chlamydiale Protease CPAF (Chlamydia protease-like activity factor) als das Enzym identifiziert werden, das diese Spaltung induziert, abhängig von der Anwesenheit zweier Rab-Proteine, Rab6 und Rab11, die den zellulären Vesikeltransport kontrollieren und zur Inklusion rekrutiert werden. Die Fragmentierung des Golgi-Apparates verhinderte dessen Relokalisierung während der Zellpolarisierung nach Einbringen eines migratorischen Stimulus. Sowohl infizierte als auch Golgin-84-depletierte Zellen migrierten langsamer und randomisiert in einem Motilitätsassay. Die Relokalisierung des Golgi-Apparates konnte durch seine Stabilisierung mittels WEHD oder Rab-Depletion wieder gewonnen werden, was die Zellmotilität teilweise wieder herstellte. Darüber hinaus konnte gezeigt werden, dass die Infektion außer der Golgi-Reorientierung die Signaltransduktion durch GTPasen beeinflusst. Die Aktivität von Cdc42 in infizierten Zellen war erhöht und die Interaktionen mit vielen ihrer Effektoren laut quantitativer Massenspektrometrie stark verändert. Die Ergebnisse dieser Arbeit zeigen, dass CPAF die für Chlamydien lebenswichtige Golgin-84 Prozessierung und Fragmentierung des Golgi-Apparates auslöst. Dies verringert die Mobilität der Wirtszelle, vor allem da der Golgi-Apparat während der Polarisierung nicht mehr ausgerichtet werden kann, des Weiteren durch Modulierung der Protein-Protein-Interaktionen von Cdc42.Chlamydia are obligate intracellular human pathogens that proliferate inside a membrane-bound compartment called the inclusion. In infected cells, the Golgi apparatus is fragmented into small ministacks that are aligned around the inclusion. This facilitates uptake of host cell sphingolipids and is essential for chlamydial development. Infection-induced Golgi fragmentation happens after processing of the Golgi matrix protein golgin-84. This work could, via comparison with well-known substrates and inhibitor studies, identify the chlamydial protease CPAF (Chlamydia protease-like activity factor) as the enzyme accountable for this cleavage. Golgi Fragmentation depended on two Rab proteins, Rab6 and Rab11, which control vesicle transport and are recruited to the Chlamydia inclusion. As a consequence of Golgi fragmentation, cells lost the capacity to reorient the Golgi apparatus during polarization after a migratory stimulus. Both infected and golgin-84 depleted cells with a permanently fragmented Golgi apparatus displayed decelerated and furthermore randomized migration in a motility assay. Relocalization of the Golgi apparatus could be restored via stabilizing WEHD treatment or Rab depletion which partly rescued cell motility. Moreover, it could be shown that migration signaling via small GTPases was influenced by Chlamydia infection. Infected cells exhibited activation of the small polarity GTPase Cdc42. Numerous interactions with downstream effectors were strongly altered in infected cells according to quantitative mass spectrometry. Particularly, the binding of Cdc42 to migration-associated effectors was decreased. The results of this work show that CPAF, by processing of golgin-84, induces Golgi fragmentation which is vitally important for Chlamydia. This disturbs host cell motility because the Golgi apparatus cannot be reoriented during polarization and, additionally, via the modulation of protein-protein-interactions of Cdc42.

Studiens syfte är att undersöka hur lärare i årskurs 1–3 använder appar i matematikundervisningen för elever i allmänna matematiksvårigheter samt vilken potential lärarna upplever att appar har för matematikundervisning. Studien bygger på semistrukturerade intervjuer av fem lärare. Det finns begränsat med forskning kring användandet av appar i matematikundervisningen och deras potential för elever i allmänna matematiksvårigheter. Resultatet visar att lärarna i den här studien använder appar på liknande sätt i sin undervisning för elever i allmänna matematiksvårigheter. Resultatet visar också att lärarna upplever att det finns flera fördelar med att använda appar i matematikundervisningen, till exempel bidrar de till ett engagemang hos eleverna.

Heutzutage stellt der Erhalt des posterioren Halteapparates beim Mitralklappenersatz (MKE) ein Routineverfahren dar. Das Ziel dieser Studie war es festzustellen, ob der komplette Erhalt der subvalvulären Strukturen beim Mitralklappenersatz Vor- oder Nachteile hat. Es wird über die Erfahrungen mit 48 Patienten berichtet. Die Gruppe MKEh bestand aus 15 Patienten, bei denen beim MKE der komplette subvalvuläre Halteapparat erhalten wurde. Die Gruppe MKEo, bei denen beim MKE nur der posteriore Halteapparat erhalten wurde, bestand aus 9 Patienten. Die dritte Gruppe (MKR) enthielt 12 Patienten, bei denen die Mitralklappe rekonstruiert worden war und die vierte Gruppe (KG) bestand aus 12 Herzgesunden ohne Klappendefekt, die die Kontrollgruppe darstellte. Die Patienten wurden einer ausführlichen Befragung unterzogen sowie prä- und postoperativ echokardiographisch untersucht und die Ergebnisse verglichen. Bezüglich der Ergebnisse der Befragungen und der präoperativen echokardiographischen Ergebnisse fanden sich bei den drei operierten Gruppen (MKEh, MKEo, MKR) keine signifikanten Unterschiede. Demgegenüber wurden postoperative Unterschiede festgestellt: Bei den Patienten mit MKE, bei denen aber der vordere Halteapparat nicht konserviert wurde (MKEo), fand sich eine signifikant geringere systolische Verkürzung zwischen dem Apex und der Mitralklappenbasis (6,5 mm - 15 mm), die systolische Kontraktion begann bei diesen Patienten außerdem nicht im Bereich des Apex und der posteriore Papillarmuskel führte in diesen Fällen keine aktive Kontraktion aus. Es konnte somit die Schlußfolgerung gezogen werden, daß der komplette subvalvuläre Apparat notwendig ist, damit der linke Ventrikel eine physiologische Kontraktion durchführen kann. Er sollte somit immer beim Mitralklappenersatz komplett erhalten werden, sofern die Papillarmuskeln und die Chorden intakt sind und keine Mitralklappenrekonstruktion durchgeführt werden kann.Today preservation of the chordal attachment to the posterior leaflet in mitral valve replacement (MVR) is a routine, universally accepted procedure. The aim of this study was to show the advantages and disadvantages of the preservation of the entire subvalvular structures. Our institution's experience with 48 patients in four groups is reported. The MKEh group consists of 15 patients who underwent MVR with preservation of the entire subvalvular structures. The MKEo group, treated with MVR and preservation of the posterior subvalvular structures only, consists of 9 patients. The third group (MVR) consists of 12 patients with mitral valve reconstruction and group 4 (CG) of 12 healthy individuals without heart valve dysfunction, as a control group. The patients were assessed by questioning and echocardiography pre- and postoperatively and the results compared. In questioning and in the preoperative echocardiography results no differences were seen between the three surgical groups (MKEh, MKEo, MKR). However, considerable differences were present in the postoperative echocardiography results: in patients without preservation of the anterior subvalvular structures (MKEo group) the shortening between the apex and the mitral valve basis is significantly less (6.5 mm - 15 mm), the systolic contraction does not begin at the apex and the posterior papillary muscle does not actively contract. It is concluded that preservation of the entire subvalvular structures is important to ensure physiological contraction of the left ventricle. They should be preserved in all mitral valve replacement operations when the papillary muscles und chordae are intact and mitral valve reconstruction cannot be performed.

A recent discovery made at Bar-Ilan University in Israel, demostrated in a scientific setting that the human mind has a neural mechanism actively making the self avoiding thoughts about its own mortality. Curiously enough the research further suggests that an awerness of the selt's death could potently lead to a more meaningful life.There is an inclination around being sick and weak, holding back how we approach end of life care and our inherent mortality; which in return affects presence and expectations regarding our lives. If death and transience were to be viewed as natural occurences, mirroring how we view horticulture and gardens, this could ease anxiety and phobia regarding the end.In my paper I am sharing research by Athul Gawande and Elisabeth Kubler-Ross regarding death-phobia and end of life care. As suggested in some of the literature, death-phobia may occur due to anxiety over not living a meaningful life and I am using Lev Tolstoy's fictional book The Death of Ivan Ilyich to evoke thought about mortality and what a meaningful life could be compromised of.In my conclusion and through my sculpture of enlarged flowers made out of cast glass and high fired stoneware; I modestly argue that love and presence can provide meaning and solace, for conquering death-phobic acculturation. That by acknowledging that transience is a forum for beauty one could enhance the magic of the mundane.

Apparent Motions is an original composition for an ensemble of fifteen musicians consisting of percussion, harp, keyboards, and stringed instruments. This dissertation is comprised of the composition as well as a written analysis of the composition. The analysis presents the most important technical features of the piece, including pitch organization, counterpoint, and motivic elaboration. Techniques are related to the overall objectives of the work, which involve the composer's personal approach to musical time. The particular treatment of time in the piece results from the coexistence of linear/developmental organization along with cyclical organization. The cyclical features intentionally interfere with the implication of progress by the linear/developmental features. The work's temporal strategy is analyzed in relation to theories regarding musical time put forward by Jonathan Kramer. 1 The primary conclusion reached is that the piece makes use of Kramer's "multiply-directed linear time."1Jonathan Kramer, The Time of Music: New Meanings, New Temporalities, New Listening Strategies (New York: Schirmer, 1988).

Compton da solidi, mostrando in seguito i risultati ottenuti. Nel primo capitolo si
introducono alcuni fondamenti sui processi di diffusione di radiazione, affrontati
attraverso un approccio di tipo classico. Nel secondo capitolo si descrive la diffusione
anelastica di tipo Compton, partendo dal caso dell'elettrone stazionario e spostandosi
successivamente sull'elettrone bersaglio in moto; sempre in questo capitolo si
introducono cenni su sezione d'urto differenziale non relativistica, regime Compton e
profili Compton. Nel terzo capitolo sono riportate le proprietà fondamentali dei
rivelatori a semiconduttore, che verranno utilizzati nel corso della prova. Nel quarto
capitolo si ha la descrizione dell'apparato sperimentale: partendo da una sorgente di
bario, viene riportata la progettazione di un apparato atto a realizzare misure di
diffusione Compton a diversi angoli; in particolare, sono descritti i progetti per la
produzione dei collimatori per la sorgente e il rivelatore e il procedimento di
calibrazione dello stesso. Nel quinto capitolo vengono esposti i risultati sperimentali,

In Saccharomyces cerevisiae, experiments with temperature-sensitive mutants of the PI4-kinase Pik1p revealed that the PI4P pool generated by this enzyme is essential for Golgi morphology and normal secretory function and that the PI4P pool at the Golgi represents a regulatory signal on its own. In order to function as a spatial and temporal regulator of membrane traffic, PI4P synthesis and turnover must be tightly regulated. It remains elusive which factors are involved in the targeting and regulation of Pik1p. Little is also known about PI4P binding proteins mediating the effects of this phosphoinositide on Golgi function. Since it has been shown that multiple pathways leave the Golgi towards the plasma membrane one can ask the question whether Pik1p and its product PI4P specifically control one pathway? Here we demonstrate an interaction of Pik1p with the 14-3-3 proteins Bmh1p and Bmh2p. Interestingly, overexpression of Bmh1p and Bmh2p results in multiple genetic interactions with genes involved in late steps of exocytosis and it affects the forward transport of the general amino acid permease Gap1p. The detected interaction depends on the phosphorylation state of Pik1p and Pik1p phosphorylation accompanies its shuttling out of the nucleus into the cytoplasm where presumably the binding to Bmh1/2p occurs. Therefore, we reason that these interactions might serve the sequestration of Pik1p away from the Golgi. This study reveals that Pik1p shows a strong effect on the delivery of Gap1p to the surface whereas the transport of exocytosis markers implicated in the direct Golgi-to-plasma membrane pathway are not significantly disturbed. Cells carrying a deletion of gga2 also show a strong defect in delivery of Gap1p to the surface. In addition, pik1-101 gga2[delta]double mutants display synthetic genetic and membrane transport phenotypes and recruitment of Gga2 to the TGN partially depends on functional Pik1p. Therefore, our results suggest a role of Pik1p in the TGN to endosome pathway.

The general aim of this thesis was to evaluate a newly designed and constructed miniaturized rotating disk apparatus for in vitro dissolution rate measurements of different drug substances from all of the classes in the Biopharmaceutical Classification System (BCS). The new equipment is based on a low volume flow-through cell of Plexiglas, a gold plated magnetic bar and a special designed press. The disk of drug substance (approx. 5 mg) is placed eccentrically in the bar. Rotation speeds were set with a graded magnetic stirrer. An external HPLC pump delivered a continuous flow of aqueous medium to the flow-through cell during dissolution testing. A reversed phase high-performance liquid chromatography system using diode array detection (RP-HPLC-DAD) was coupled online to the new equipment. The injections from the miniaturized rotating disk outlet into the quantifying HPLC system were controlled by a six-position switching valve. The injection volumes from the valve and the autosampler, used for the external standards, were statistically evaluated to match each other volumetrically. No analyses were longer than three minutes, using isocratic mode. A traditional USP rotating disk apparatus was used as a reference system and the two instruments were shown to be statistically dissimilar in the numerical dissolution rate values probably due to different hydrodynamics, but had approximately the same precision/repeatability. When correlating the logarithmic values of the in vitro dissolution rate (G) with the apparent solubility (S), using shake-flask methodology in the solubility studies, the two apparatuses gave the same correlation patterns. Further correlation studies were done where the media components were altered by the use of different buffer species or additives into the buffers, such as inorganic salts. Chemometric tools, e.g. orthogonal partial least squares (OPLS), were used to better evaluate the most influential factors for G and S in different media. The most significant factor for a model basic drug substance (terfenadine) was pH, followed by the ionic strength (I) and added sodium chloride in one of the media. However, the surfactants in the Fasted State Simulated Intestinal Fluid (FaSSIF-V2) were found to be insignificant for G and S in this study (using a 95% confidence interval). The new miniaturized apparatus is a promising prototype for in vitro dissolution rate measurements both for early screening purposes and in dissolution testing during drug development, but needs further instrumental improvements.

Esto ha permitido que se pueda desarrollar un amplio análisis cualitativo y cuantitativo para determinar la viabilidad del proyecto.
En el presente curso, se ha planteado una idea innovadora de servir a los usuarios y brindar la facilidad para buscar, ubicar y reservar estacionamientos disponibles en las zonas estratégicas de mayor demanda. Para el desarrollo del proyecto, ha sido necesario trabajar los puntos de aspectos generales del negocio, planeamiento estratégico, investigación de mercado, plan de operaciones, plan de marketing, estructura organizacional, y finalmente plan económico financiero.
Además, se consideran factores relevantes como, la demanda según el distrito, los precios y los espacios de estacionamientos. This has allowed the development of a broad qualitative and quantitative analysis to determine the feasibility of the project. In this course, an innovative idea has been proposed to serve users and provide the facility to search, locate and reserve available parking spaces in strategic areas of greatest demand. For the development of the project, it has been necessary to work on the general aspects of the business, strategic planning, market research, operations plan, marketing plan, organizational structure, and finally economic and financial plan. In addition, relevant factors such as demand according to the district, prices and parking spaces are considered. Likewise, assets, liabilities, equity and other administrative expenses are projected to obtain a correct planning according to the established objectives, in order to meet the expectations of the project, generate income to achieve profitability and solidity of the company. These points have allowed validating the viability of the project and demonstrating that currently technology-focused businesses are highly profitable.

Utifrån utvalda videosekvenser där ett- och tvååriga barn samhandlar med avancerade appar i en förskolepraktik analyseras sekvenserna utifrån posthumanistiska begrepp och ANT-analys. Resultatet belyser mycket täta, intensiva och komplexa möten där varje intra-aktion förändrar och transformerar de innefattande aktörerna i nätverk. Fokus ligger på kommunikativa relationer av transdisciplinära tillblivelseprocesser av barns och avancerade appars möten.

Purpose: Finding which resources and capabilities are deployed among Istanbul-based apparel manufacturers, evaluating to what extent they are tangible, intangible as well as capabilities as well as lastly analysing them according to the valuable, rare, inimitable and non-substitutable framework. Design/methodology: The research design consists of a case study held at several firms. Empirical data was collected using semi-structured interviews. Empirical data: Empirical data was primarily collected at nine different apparel manufacturers within the greater Istanbul region. Conclusion: Conclusions show that within the bundles of resources and capabilities, a large variation was noted with high international competitiveness. However there are areas such as design and quality which could be continuously developed among firms. In the end, whatever bundle of resources and capabilities a firm has, it’s the end price that speaks.Program: Textilekonomutbildningen

The current study aims to investigate the feasibility of international apparel and footwear retailers to expand their business in Pakistan and compete with other brands. It also highlights the business opportunities in Pakistan apparel retail sector and the motivations of international brands behind expansions. This study was conducted with the help of interviews based on diamond model of Porter, and Hofsetede cultural dimensions. The open ended questions were delivered to the professionals electronically while interviews were conducted by telephone. Seven companies from Pakistan were selected for study purpose. Data was analyzed and assessed manually. The study revealed that there is a significant opportunity for international apparel retailers to launch their retail outlets in Pakistan along with some risks. International brands which are financially strong can tackle these risks. However it was concluded that the companies with less financial strength may find it difficult to go in a new market within 5 years. UK brands are already there and brands from other countries are also opening. Overall Pakistan retail sector is growing and people are becoming fashion conscious. This study provides information to International apparel brands which they can take into consideration while entering Pakistan's apparel retail market. It also gives an opportunity for assessment of market in the light of theoretical modules and shows a direction of getting better market share by launching.

In questo lavoro di tesi sono state studiate le caratteristiche di una macchina per tomosintesi Fujifilm AMULET Innovality in uso presso l'Istituto Scientifico Romagnolo per lo Studio e la Cura dei Tumori (I.R.S.T.) di Meldola.
Le valutazioni sono state fatte utilizzando diversi fantocci, uno dei quali costruito durante il lavoro di tesi. Per la valutazione delle immagini di mammografia digitale e di tomosintesi sono state seguite le linee guida della International Electrotechnical Commission (IEC) e della European Reference Organisation for Quality Assured Breast Screening and Diagnostic Services (EUREF).
Per lo studio delle mammografie digitali sono stati valutati, utilizzando i software COQ e ImageJ, i parametri di NPS, MTF e DQE.
Per lo studio delle immagini di tomosintesi sono stati appositamente sviluppati degli algoritmi in linguaggio Java, integrati poi all'interno del software COQ. Il programma sviluppato ha permesso di valutare ASF, MTF, NPS e omogeneità delle immagini ricostruite.
