La ricerca ha inteso ripercorrere la storia di “Pro Infantia” nel suo primo ventennio di vita (1913-1933). Il lavoro ha preso avvio dall’analisi dei dibattiti politico-scolastici e dall’approfondimento del contesto pedagogico e culturale in cui si collocava la decisione dei vertici dell’editrice La Scuola di avviare una rivista per educatrici d’asilo. Il lavoro è entrato nel vivo con la messa a fuoco delle prese di posizione del periodico in merito alle novità introdotte dai provvedimenti legislativi coevi, quali, ad esempio, i Programmi per le istituzioni infantili emanati da Credaro nel 1914, la Riforma Gentile e i Programmi elaborati da Giuseppe Lombardo Radice nel 1923. Sono state altresì analizzate le valutazioni espresse circa le prassi didattiche e i metodi pedagogici adottati nelle istituzioni infantili del tempo, l’associazionismo di categoria, i percorsi formativi per le educatrici. Lo spoglio della rivista ha consentito inoltre di fare ulteriore luce sulle scelte dell’editrice La Scuola e sull’atteggiamento assunto dal periodico di fronte ad alcuni momenti chiave non solo della storia dell’educazione infantile in Italia, ma anche di quella politica e sociale, come ad esempio il primo conflitto mondiale, il dopoguerra, l’ascesa del fascismo e la successiva affermazione della dittatura.<br>The aim of this research was to reconstruct the history of "Pro Infantia" over its initial twenty years of publication (1913-1933). The first step in the study was to analyse the political debate on education and the cultural and educational backdrop against which the publishing house, La Scuola, decided to set up a journal for infant school teachers. The core of the research work involved examining the journal’s positions on the legislation of the period – such as the programs for infant schools issued by Credaro in 1914, the Gentile reform and the programs drawn up by Giuseppe Lombardo Radice in 1923 – and its assessments of the teaching practices and educational methods adopted in contemporary infant schools, as well as of teachers’ associations, and infant teacher training courses. Finally, scrutiny of the journal’s content also shed light on the policies adopted by La Scuola and "Pro Infantia"’s stance concerning both key historical developments in Italian early childhood education and broader political and social events, such as World War One, the post-war period, the rise of fascism and the advent of the fascist dictatorship.

A partir d’una visió crítica emmarcada dins de l’hermenèutica d’arrel pareysoniana, hom aplica les consideracions heideggerianes al voltant de Hölderlin a la figura de Leopardi i es planteja, com a pressupòsit inicial, que aquest autor també forma part dels anomenats poetes-filòsofs: Giacomo Leopardi medita mentre canta. No es tracta, doncs, d’una investigació històrico-filosòfica o històrico-filològica, sinó d’una tesi de tipus documental amb una finalitat palesament exploratòria en què se subratlla la importància de l’activitat interpretativa a l’hora de constituir i transformar l’ésser. El principal objectiu d’aquest estudi consisteix a distingir el nulla possibilitador del niente anorreador i a equiparar, doncs, el nulla leopardià amb l’ésser. El nulla esdevé el veritable fonament sense fonament amb què es caracteritza el seu pensar poètico-filosòfic, així com l’element que origina la indissolubilitat entre les reflexions estètiques i ontològiques que saturen la totalitat de l’obra i el pensament leopardians. La principal aportació teorètica d’aquest estudi, el neologisme imitació-interpretant, i amb el qual hom situa la meditació de l’italià dins el solc de la mímesi aristotèlica, subratlla, de fet, l’originària i indissoluble duplicitat de tot acte imitatiu: tota imitació en imitar, interpreta i constitueix l’ésser. D’aquest nou terme en resulta, per tant, el segon gran objectiu de la tesi, és a dir, la distinció entre còpia i imitació, la qual permet de concloure que el seu és un pensar ultrametafísic. I és que mentre que la còpia classicista fa referència a un ésser-com-a-objecte i, per consegüent, a la mera còpia d’un contingut òntic —un model establert a priori—, la imitació-interpretant, en canvi, remet a un pensar ontològic segons el qual l’ésser es dóna en el mateix moment en què hom s’hi relaciona, és a dir, tan bon punt és imitat i, consegüentment, redescrit. A l’últim, tant el nulla com la imitació-interpretant permeten que la tesi, en tant que elements vertebradors del projecte ultrametafísic de Leopardi, reinterpreti tres dels conceptes poètico-filosòfics més importants del present autor: ultrafilosofia, mezza filosofia i poesia mista. I és que tots tres palesen la ultrametafisicitat del seu pensar poètic, en el sentit que es tracta d’una filosofia i d’una poesia que no cerca pas la transparent immediatesa, ja que això n’implicaria l’encongiment de l’ésser, sinó el nulla o el buit de sentit de la interposició interpretativa. La imitació, la traducció i la poesia esdevenen, doncs, els exemples amb els quals s’il·lustra la interposició com a obstacle ontològic, és a dir, com els elements que permeten de constituir la realitat.<br>A partir de una visión crítica enmarcada dentro de la hermenéutica de raíz pareysoniana, se aplican las consideraciones heideggerianas alrededor de Hölderlin a la figura de Leopardi y se plantea, como hipótesis inicial, que este autor también forma parte de los llamados poetas-filósofos: Giacomo Leopardi medita mientras canta. No se trata, pues, de una investigación histórico-filosófica o histórico-filológica, sino de una tesis de tipo documental con una finalidad manifiestamente exploratoria en la que se subraya la importancia de la actividad interpretativa a la hora de constituir y transformar el ser. El principal objetivo de este estudio consiste en distinguir el nulla posibilitador del niente agotador y equiparar, pues, el nulla leopardiano con el ser. El nulla se convierte en el verdadero fundamento sin fundamento con el que se caracteriza su pensar poético-filosófico, así como el elemento que origina la indisolubilidad entre las reflexiones estéticas y ontológicas que saturan la totalidad de la obra y el pensamiento leopardiano. La principal aportación teorética de este estudio, el neologismo imitación-interpretante, y con el cual se sitúa la meditación del italiano bajo la estela de la mímesis aristotélica, subraya, de hecho, la originaria e indisoluble duplicidad de todo acto imitativo: toda imitación en imitar, interpreta y constituye el ser. De este nuevo término resulta, por tanto, el segundo gran objetivo de la tesis, es decir, la distinción entre copia e imitación, la cual permite concluir que el suyo es un pensar ultrametafísico. Y es que mientras que la copia clasicista hace referencia a un ser-como-objeto y, por consiguiente, a la mera copia de un contenido óntico —un modelo establecido a priori—, la imitación-interpretante, en cambio, remite a un pensar ontológico según el cual el ser se da en el mismo momento en que uno se relaciona con él, es decir, en cuanto es imitado y, consiguientemente, redescrito. Por último, tanto el nulla como la imitación-interpretante permiten que la tesis, en calidad de elementos vertebradores del proyecto ultrametafísico de Leopardi, reinterprete tres de los conceptos poético-filosóficos más importantes del presente autor: ultrafilosofia, mezza filosofia y poesia mista. Y es que los tres manifiestan la ultrametafisicidad de su pensar poético, en el sentido de que se trata de una filosofía y de una poesía que no busca la transparente inmediatez, ya que esto implicaría el encogimiento del ser, sino el nulla o el vacío de sentido de la interposición interpretativa. La imitación, la traducción y la poesía se convierten, pues, en los ejemplos con los que se ilustra la interposición como obstáculo ontológico, es decir, como los elementos que permiten constituir la realidad.<br>Based on a critical vision framed within the hermeneutics of the Pareysonian root, Heideggerian considerations around Hölderlin are applied to the figure of Leopardi and it is considered, as an initial assumption, that this author also forms part of the so-called poets-philosophers: Giacomo Leopardi meditates while singing. Therefore, it is not a historical-philosophical or historical-philological investigation, but a thesis of a documentary type with a purpose that is fundamentally exploratory, underlining the importance of interpretative activity when it comes to constituting and transforming the Being. The main objective of this study is to distinguish the possibilitator nulla from the exhausting niente and to equate, thence, the leopardian nulla with the Being. The nulla becomes the true unfounded foundation with which its poetic-philosophical thinking is characterized, as well as the element that causes the inseparability between the aesthetic and ontological reflections, that saturate the totality of the work and the leopardian thought. The main theoretical contribution of this study, the neologism imitation-interpreting with which the Italian’s meditation is aligned with the Aristotelian Mimesis, underlines, in fact, the original and indissoluble duplicity of any imitative act: any imitation in imitating, interprets and constitutes the Being. This new term results, thus, as the second great objective of the thesis, that is, the distinction between copy and imitation, which allows us to conclude that his thinking is an ultrametaphysical one. And while the classicist refers to a Being-as-a-object and, as a result, to the mere copy of an ontic content —a model established a priori—, imitation-interpreting, on the other hand, refers to an ontological thinking according to which the Being occurs at the same moment in which one is related to it, that is to say, as soon as it is imitated and, consequently, redescribed. Ultimately, both the nulla and the imitation-interpreting allow the thesis, as the backbone of Leopardi's ultrametaphysical project, to reinterpret three of the most important poetic-philosophical concepts of the present author: ultrafilosofia, mezza filosofia and poesia mista. So, all three concepts point to the ultrametaphysicity of their poetical thinking, in the sense that it is a philosophy and a poetry that does not seek the transparent immediacy, which would imply the shrinkage of Being, but the nulla or the void of meaning of the interpretative interposition. Imitation, translation and poetry become, then, the examples with which interposition is illustrated as an ontological obstacle, that is, as the element that allow to create reality.

Intitulado de Moinhos de Água no Ardila. O património do sistema territorial, este trabalho monográfico visa o registo, estudo e interpretação de um conjunto de moinhos de água no Rio Ardila, afluente do Rio Guadiana. Com o intuito de contextualizar e compreender as componentes naturais e culturais da unidade de paisagem onde o objecto de estudo se insere desenvolveu-se um enquadramento histórico e geográfico, fazendo a leitura dos sistemas geológico, topográfico, hidrográfico, agrícola, viário e edificado. Desta forma executaram-se dois trabalhos de campo para selecção e caracterização de sete unidades e de entre estas, três foram objeto de registo gráfico e escrito pormenorizados. Confrontados com a realidade de abandono e ruína do património molinológico, esta dissertação resultou de uma investigação exaustiva com o objetivo de esta vir a ser uma base para futuros trabalhos propositivos, de modo a sensibilizar técnicos, políticos e arquitetos para a recuperação, salvaguarda e preservação do património que é parte integrante da paisagem do Alentejo; ABSTRACT : Titled Watermills in Ardila River. The heritage of the territorial system, this monograph aims to register, study and interpretate a group of water milIs in Ardila River, Guadiana River affluent. ln order to contextualize and understand the natural and cultural components of the landscape unit where the study object is inserted, it was developed a historical and geographical framework, making the reading of geological, topographical, hydrological, agricultural, road and buildings systems. ln this way they were carried out two field works for selection and characterization of seven units and between these, three were the subject of detailed written and graphic register. Faced with the reality of abandonment and ruin of molinological heritage, this work resulted from the research to a basis for future projects in order to sensitive technicians, politicians and architects for the recovery, protection and preservation of a heritage that is part of the Alentejo landscape.

Mestrado em Engenharia do Ambiente<br>Nas últimas décadas, os fogos florestais têm sido a maior ameaça para a floresta mediterrânea e portuguesa, e apresentam ainda uma tendência para aumentar em frequência e severidade. Os inúmeros fogos florestais que ocorrem todos os anos em Portugal representam um risco elevado para o aumento do escoamento superficial e da erosão nas áreas ardidas e, ainda, nas zonas aquáticas a jusante.
Esta dissertação tem como principal objetivo o estudo de fenómenos de erosão hídrica à micro-escala e à escala de encosta, três anos após um incêndio ocorrido em Agosto de 2008. A área de estudo pertence à localidade de Colmeal (distrito de Coimbra) e incide em quatro encostas com usos de solo distintos: Pinhal e Eucaliptal não lavrados e dois Eucaliptais lavrados em diferentes sentidos de encosta (maior declive e curvas de nível). Para modelar o risco de erosão do solo foi selecionado o modelo Morgan-Morgan-Finney (MMF), que foi desenvolvido para prever as perdas anuais de solo. Pretende-se então estudar a eficácia do MMF para prever a erosão durante o terceiro ano após um incêndio florestal.
As observações mostraram que a erosão é superior à escala das micro-parcelas que á escala de encosta, i.e., a erosão média à micro-escala foi cerca de 10x superior à observada à escala de encosta. Esta diminuição de erosão com o aumento da escala pode ser explicada por um maior tempo de contacto, permitindo uma maior (re-) infiltração de escorrência superficial. À microescala foi a parcela ELH que mais se destacou em termos de taxa de erosão. Esta obteve uma erosão de 144,72g.m-2, cerca de 4x superior que a parcela mais baixa (ENL), com 31,91g.m-2. À escala de encosta, a mesma parcela (ELH), foi quem obteve menor taxa de erosão (2,47g.m-2), e a parcela com maior erosão obteve uma taxa cerca de 5x superior (12,18g.m-2), (PNL).
Observaram-se falhas no modelo pois sobrestimou os resultados medidos, i.e., os resultados estimados foram muito superiores aos medidos para a escala de encosta. Este fato pode ser justificado pela não calibração do modelo a esta área de estudo.<br>In recent decades, forest fires have been the biggest threat to Mediterranean and Portuguese forest, and still have a tendency to increase in frequency and severity, and still have a tendency to increase. The numerous forest fires that occur every year in Portugal represent a high risk to increase runoff and erosion in burned areas, and also downstream aquatic areas.
The aim object of this thesis was the study of the phenomena of erosion to the micro and slope scale, three years after a fire that occurred in August 2008. The study area belongs to Colmeal (district of Coimbra), and focuses on four slopes (one Pine and three Eucalyptus) with different forest management (not plowed and vertical and higher slope and contour lines). To model the risk of erosion was selected the Morgan-Morgan-Finney model (MMF), developed to predict annual soil losses. The aim is study the efficacy of the model to predict erosion during the third year after a forest fire.
The observations show that the erosion rates are bigger in microscale than slope scale, i.e., the erosion in microscale was 10xhigher than the erosion observed at the scale of slope. This reduction can be explained for longer periods of contact, allowing a bigger (re-) infiltration of runoff. In microscale was the ELH parcel that stood out in terms of rate erosion. This parcel got an erosion of 144,72g.m-2, 4xhigher than the lower parcel with 31,91g.m-2 (ENL). Perhaps, in slope scale, the same parcel got a lower erosion rate (2,47g.m-2), and the parcel PNL, with higher erosion rate, obtained an erosion about 5xhigher (12,18g.m-2).
Observed with the application of the model, that this overestimated the erosion rates, i.e., the estimated erosion rates is greater than the field measurement. This fact can be justified by not calibrating the model to this area of study.

Na legislação europeia a gestão da água é perspetivada à escala da Região Hidrográfica, bacia hidrográfica e da sub-bacia, permitindo uma visão alargada das pressões antropogénicas. Porém, a priorização e as ações de reabilitação fazem-se à escala da sub-bacia e do troço. Nesta tese estudou-se a bacia hidrográfica do Ardila, com vista à definição de um plano estratégico, com propostas de reabilitação para a melhoria da qualidade da água numa perspectiva transversal. Para o efeito, recorreu-se: 1) ao modelo SWAT, estimando o escoamento e cargas de nutrientes; 2) a inquérito sobre as perceções e participação dos atores sociais na gestão da bacia hidrográfica do rio Ardila; 3) a uma revisão das metodologias de reabilitação de linhas de água. Propõe-se a reabilitação de quatro troços da bacia do Ardila, prioritários em termos da carga de nutrientes. Dá-se assim, cumprimento à DQA (2015) numa perspetiva de sustentabilidade territorial (ambiental/social); ABSTRACT:
STRATEGIES FOR IMPROVING WATER QUALITY AT ARDILA WATERSHED
European legislation on water management is seen at a Hydrographic Region, watershed and sub-basin scale, allowing a wider view of the anthropogenic pressures. However, prioritization and rehabilitation actions are made on sub-basin and stretch scale. In this thesis it was studied the Ardila watershed to define a strategic plan with rehabilitation proposals for improvement of water quality using a transversal approach. For this purpose: 1) SWAT model was used to estimate discharges and nutrient loads; 2) a survey was performed to analyze stakeholder´s perception and participation at the Ardila watershed management; 3) an overview of stream rehabilitation methodology was made. It is proposed to rehabilitate four priority sections in the Ardila watershed due to their localization and high nutrient loads, thus fulfilling the Water Framework Directive (2015) in an environmental and social sustainability perspective of the territory.

Mestrado em Engenharia do Ambiente<br>Os incêndios florestais produzem elevadas quantidades de Hidrocarbonetos
Aromáticos policíclicos (HAPs), conhecidos pela sua potencial toxicidade e
persistência ambiental. Através de vários sistemas de transporte,
nomeadamente escorrências superficiais, os HAPs provenientes de zonas
ardidas entram nos sistemas aquáticos podendo comportar riscos para o
ecossistema e saúde humana. Neste sentido, a presente dissertação teve
como objectivo central a avaliação da potencial toxicidade das escorrências
superficiais provenientes de zonas ardidas em espécies aquáticas, através da
realização de ensaios ecotoxicológicos. Em colaboração com o projecto
EROSFIRE-II foi seleccionada como área de estudo uma zona ardida em
Setembro de 2009 localizada em Sever do Vouga. Foram seleccionadas duas
encostas (L e R) nas quais se recolheram amostras de escorrências
superficiais de água. Foi ainda recolhida uma amostra de água num curso de
água localizado entre as duas encostas (S). Em laboratório foi efectuada uma
caracterização físico-química das várias amostras, incluindo a medição de
vários parâmetros, tais como: pH, condutividade eléctrica, oxigénio dissolvido,
sólidos totais, nitratos, nitritos, orto-fosfatos, amónia, dureza e sulfatos.
Procedeu-se ainda à análise dos 16 HAPs prioritários, definidos pela US EPA.
No geral, os vários parâmetros básicos medidos, assim como os nutrientes
apresentaram-se relativamente semelhantes nas 3 amostras. Em relação aos
HAPs observou-se uma clara diferença entre as amostras L e R e a amostra
S, com valores superiores registados nas encostas. Deste modo, os
resultados obtidos mostram largamente a importância dos incêndios florestais
como fonte de HAPs para o ambiente aquático. Paralelamente foi efectuada
uma avaliação ecotoxicológica das amostras que envolveu uma bateria de
ensaios com diferentes organismos, incluindo: a bactéria Vibrio fischeri, a alga
Pseudokirchneriella subcapitata, a macrófita Lemna minor e o cladócero
Daphnia magna. Os resultados obtidos revelaram efeitos distintos nas várias
espécies. Salienta-se a elevada toxicidade que a amostra S apresentou para
V. fischeri com um EC50 de 8% (5min) e 12% (15min). As amostras L e R quer
filtradas quer não filtradas, ao contrário da amostra S, provocaram efeitos na
taxa de crescimento de P. subcapitata nas concentrações mais elevadas. A
espécie L. minor foi altamente sensível à amostra R não filtrada,
apresentando um LOEC correspondente à concentração de 12,5%. Por fim, o
organismo D. magna apresentou elevada mortalidade ao longo do ensaio nas
concentrações mais elevadas, tendo-se observado igualmente uma
diminuição significativa na taxa de crescimento intrínseco, em ambas as
amostras testadas (S e R) filtradas e não filtradas. De um modo geral, apesar
das limitações associadas, os ensaios ecotoxicológicos realizados revelaram
os riscos para o biota aquático associados à ocorrência de incêndios florestais
e consequente produção de substâncias tóxicas (e.g HAPs). Neste sentido,
este estudo salienta a importância de aprofundar os conhecimentos de
investigação nesta temática de forma a colmatar a falta de informação relativa
a um problema ambiental de elevada relevância.<br>Forest fires produce large amounts of Polycyclic aromatic
hydrocarbons (PAHs), known by its potential toxicity and
environmental persistence. Through various transport systems,
including runoff. Superficially, the PAHs from entering the burnt areas
aquatic systems can pose risks to the ecosystem and human health.
In this sense, this dissertation was central objective evaluation of the
potential toxicity of surface runoff from areas burned in aquatic
species by testing Ecotoxicology. In collaboration with the project
EROSFIRE-II was selected as study area an area burnt in September
2009 located in Sever do Vouga. We selected two slopes (L and R) in
which samples were collected surface water runoff. Was also collected
a sample water in a watercourse located between two slopes (S). In
laboratory characterization was carried out physical chemistry of the
various samples, including the measurement of various parameters
such as pH, electrical conductivity, dissolved oxygen, total solids,
nitrates, nitrites, ortho-phosphates, ammonia, hardness, and sulfates.
There has yet to Analysis of 16 priority PAHs, defined by U.S. EPA. In
generally, several basic parameters measured and the nutrients were
relatively similar in the three samples. Regarding PAHs observed a
clear difference between samples and sample L and R S, with higher
values recorded on the slopes. Thus, the results show broadly the
importance of forest fires as source of PAHs to the aquatic
environment. In a parallel carried out an ecotoxicological assessment
of the samples involved a battery of tests in different organisms,
including: the bacterium Vibrio fischeri, the green alga
Pseudokirchneriella subcapitata, the macrophyte Lemna minor and
cladoceran Daphnia magna. The results revealed distinct effects in
various species. We emphasize the high toxicity showed that the
sample S to V. fischeri with a EC50 of 8% (5min) and 12% (15min).
Samples L and R both filtered or unfiltered, unlike the sample S,
caused effects on the growth rate of P. subcapitata in higher
concentrations. The species L. minor was highly sensitive to sample R
unfiltered, with a LOEC a concentration of 12.5%. Finally, the body
D.magna showed high mortality over the test higher concentrations
and it was also observed a significant decrease in the intrinsic growth
rate in both samples tested (S and R) filtered and unfiltered. In
general, despite the constraints, the tests Ecotoxicological carried out
revealed the risks to the biota water associated with the occurrence of
forest fires and consequent production of toxic substances (eg PAHs).
In this sense, this study highlights the importance of deepening the
research knowledge in this subject in order to address the lack of
information on an environmental problem high relevance.

mapa em Sistemas de Informação Geográfica (SIG) com o programa
ArcGIS, que descreve o Risco de Erosão numa área sujeita a um
incêndio florestal na pequena localidade de Açores.
Para cartografar o risco de erosão do solo foram seleccionados dois
modelos. A Equação Universal de Perdas de Solo (Universal Soil Loss
Equation – USLE) pelo facto de ter sido escolhida pelo INAG na
elaboração do mapa “Perdas de Solo após incêndios Florestais do
Verão de 2003” e o modelo Morgan–Morgan–Finney (MMF) pelo facto
de apresentar um elevado potencial na determinação de perdas de solo
em áreas ardidas.
Devido ao facto de ambos os modelos terem sido desenvolvidos
especificamente para áreas agrícolas, pretende-se, neste estudo,
avaliar a adequação dos seus resultados para uma área florestal
recentemente ardida. Para avaliar os dois modelos, os seus resultados
foram comparados com dados anteriormente recolhidos pelo projecto
EROSFIRE através de Simulações de Chuva e de Parcelas Abertas, e
ainda no âmbito deste trabalho através de uma validação de campo que
consistiu numa inventariação de fenómenos erosivos no local.
Tal como era esperado, observam-se falhas na determinação das
taxas de erosão segundo os dois modelos e tal facto é justificado por
ambos dependerem de factores empíricos que dizem respeito ao
coberto vegetal e às práticas de conservação de solo utilizadas (Factor
C e P).
ABSTRACT: The main objective of this thesis is to obtain a Geographic
Information System map that describes the Erosion Hazard in an area
on the little village of Açores after a forest fire.
To follow up this proposed aim, two referenced erosion models
where selected. The Universal Soil Equation (USLE) because was
chosen by INAG to built the map “Soil losses after forest fires in 2003
summer” and the Morgan-Morgan-Finney model (MMF) because
presents a high potential in determination of soil losses in bunt areas.
Due to the fact that both models were develop specifically to be
applied in agricultural land fields, it is a purpose in this study to evaluate
the relation of its results in a recent burnt area. Through Rainfall
Simulation and Open Plot data that was collected by the EROSFIRE
project in 2005/2006 year, and also with a Field Validation based on a
survey for erosion features that was made in this study, the difference
between the reality and the applied models will be exposed.
As is was expected, the detected weakness of the obtained erosion
risk models results is justified due to, both models depend on empirical
factors (C and P) that correspond to cover and management and
support practice factor respectably.

This thesis is a study of some of the processes which operate at mid crustal levels (10- 20km) during the emplacement of granitoid magmas, with particular reference to the process of in-situ expansion and the association between magma emplacement and orogen evolution. A number of plutons were studied to assess this the Ardara pluton from Donegal, NW Ireland, together with the Atibaia, Morro Azul, Imbiricu and Itapeti plutons from the Rio Paraiba do Sul shear belt (RPSSB), Sao Paulo state, SE Brazil. The emplacement of the Ardara pluton has been the subject of studies by many authors since the 1950's. It has been interpreted as a diapir ascending along a thrust, a granite balloon which forcefully created more than 60% of its space and, most recently, as a set of nested diapirs which forcefully created only 30% of their own space. Field mapping together with the determination of finite strain within the pluton, shortening estimates, shear sense determinations, petrographic deformation fabrics and computer modelling indicates that: i) the pluton displays a concentrically increasing finite strain and it expanded in-situ from a central 'injection point', having possibly ascended along a shear zone-related conduit; ii) the country rocks partitioned intense strains into the 500m closest to the pluton, a deformation feature which is shown to be consistent with a power-law wallrock theology; Hi) earlier granite pulses and the country rock were 'shouldered aside' to the east and west, expansion of the pluton, which was preferentially in a northwards direction; iv) that the intrusion related shortening preserved within the wallrocks was approximately equivalent to the strain preserved within pluton and the space requirements for the pluton are essentially met by preserved forceful emplacement-related strains; and v) if the pluton magma is considered as having ascended through dyke-like conduits emplacement could occur in a minimum of ~4000yrs, whereas magma ascent as a Hot Stokes diapir would require hundreds of thousands to millions of years for complete emplacement. Finally, the Ardara pluton does not conform to any of the established criteria for diapir-like ascent of a magma body and the data demonstrate it to be a testable example of a pluton which expanded entirely in-situ. The Rio Paraiba do Sul shear belt (RPSSB) is a dextral transpressional segment of the Late Precambrian Brasiliano-Pan African orogenic belt. The studied plutons were emplaced in the latter stages of this orogen in close spatial association with one of the many northeast-southwest trending, sub-vertical, continental-scale shear zones. All the granites show a similar emplacement and deformation history despite their different petrographic features and isotopic ages. This history consists of an early shallowly dipping, low angle fabric, in the country rocks, associated with crustal thickening, the later development of dominantly dextral sub-vertical shear zones, followed by the emplacement of the studied plutons and finally overprinting by a pervasive dextral plane strain and late-stage discrete mylonitic shears. Field examination demonstrates that: i) each of the plutons preserves an internally homogeneous emplacement-related finite strain, weak magmatic fabric, magma sheets and weakly deformed wallrocks; ii) magmatic shear sense determinations and wallrock deformation fabrics indicate that, during emplacement, the RPSSB was extending (at least at the emplacement level) in an east-west direction creating sinistral dilatational pull-aparts along the major shear zones, into which granitic magma was preferentially emplaced; and Hi) within this generally extensional context there was a component of long axis perpendicular in-situ forceful expansion, this created no more than 20% of the width of any one granite. Applying a simple pull-apart extension model suggests that pluton emplacement was associated with approximately 40% regional extension sub-parallel to the former orogenic convergence direction. This emplacement, as a result of sheeting through dyke-like conduits, requires thousands of years, rather than the millions of years required for ascent and emplacement of a Hot Stokes diapir. Interpreting these results suggests that the intruded granitoids are associated with a mid-crustal component of orogen perpendicular extension I collapse during the latter stages of the Brasiliano orogeny. Such extension I collapse could have been initiated by: a cessation in continental convergence or; by the delamination of a thickened thermal boundary layer, a process which has been suggested to be the driving force behind orogenic collapse in younger orogenic belts. These examples demonstrate that: i) 'forceful' balloon-like emplacement of granitic magma can occur; ii) there are very close inter-relationships between granite emplacement and orogenic dynamics; and Hi) that 'space' for granitoid magma can be created by a combination of forceful and dilatational and fault-related mechanisms.

Os ardeídeos têm sido utilizados para avaliar a contaminação ambiental em virtude principalmente dos seus hábitos alimentares. Estas espécies são consumidores secundários e terciários e tendem a acumular altas concentrações de elementos tóxicos nos seus tecidos. Por esta razão, estas espécies estão mais sujeitas aos efeitos deletérios da contaminação, o que requer uma atenção especial para a sua conservação. Com o objetivo de verificar a possibilidade do uso da Ardea alba (Garça-Branca-Grande) na avaliação da contaminação ambiental, fígados desta ave adulta encontrada ferida ou enferma na Região Metropolitana de São Paulo (RMSP) e outras localidades do estado de São Paulo, foram analisadas pelas técnicas de análise por ativação com nêutrons (NAA) e espectrometria de absorção atômica (AAS). Para obtenção das amostras, foi obtida a licença junto ao Instituto Brasileiro do Meio Ambiente e dos Recursos Renováveis (IBAMA). As amostras de fígado foram moídas, liofilizadas e homogeneizadas para as análises. Para avaliar a qualidade analítica dos resultados, foram analisados os materiais de referência certificados (MRCs) NIST 1577b Bovine Liver, INCT-TL-1 Tea Leaves e NIST SRM 2976 Mussel tissue (Mytilus edulis) e os resultados apresentaram exatidão satisfatória. Os elementos Br, Cd, Co, Cs, Cu, Fe, Hg, K, Mg, Mn, Na, Rb, Se e Zn foram determinados em 26 amostras de fígado das garças da RMSP e foi verificada uma ampla variabilidade nas suas concentrações. As comparações dos resultados obtidos com os valores da literatura indicaram que as concentrações da maioria dos elementos analisados nos fígados das garças da RMSP são mais altas ou da mesma grandeza que as de áreas poluídas. Comparações feitas entre as concentrações de elementos obtidas em relação ao gênero das garças por meio da aplicação do teste não paramétrico de Mann-Whitney U (p < 0,05), indicaram que as fêmeas apresentam concentrações mais baixas significativas de Cs, Hg, Se e Zn. Esses resultados podem estar associados às diferenças fisiológicas e ecológicas entre os gêneros. Utilizando a matriz de Pearson (r = 0,7, α <0,05, p <0,01 e p <0,05) foi feito um estudo da correlação entre os teores de elementos. As correlações positivas obtidas para os elementos Cd, Cu, Hg, Mn, Se e Zn, indicaram fontes comuns de origem destes elementos, principalmente associadas a atividades antrópicas na RMSP. Os resultados demonstraram a aplicabilidade de fígados de garças para avaliação da contaminação da RMSP e corroboram que o monitoramento de elementos químicos em ardeídeos pode ser fonte importante de informações para estudos de conservação dessas espécies.<br>Ardeids have been used to assess environmental contamination especially due to their eating habits. Since these species are secondary and tertiary consumers they tend to accumulate high concentrations of toxic elements in their tissues. Moreover, these species are more susceptible to the deleterious effects of environmental contamination, which brings up the need for their conservation. Aiming to verify the possibility of using Ardea alba (Great Egret) in the evaluation of environmental contamination, livers of adult birds found injured or sick in the Metropolitan Region of São Paulo (RMSP) and other locations of the state of São Paulo, were analyzed by the methods of neutron activation analysis (NAA) and atomic absorption spectrometry (AAS). Permission to collect samples was given by the Brazilian Institute of Environment and Renewable Resources (IBAMA). The liver samples were ground, lyophilized and homogenized for the analyses. To assess the quality of the analytical results the certified reference materials NIST 1577b Bovine Liver, INCT-TL-1 Tea Leaves, NIST SRM 2976 and Mussel (Mytilus edulis) tissue were analyzed and these results showed good accuracy. The elements Br, Cd, Co, Cs, Cu, Fe, Hg, K, Mg, Mn, Na, Rb, Se and Zn were determined in 26 liver samples of birds found in RMSP and a wide variability in their concentrations was verified. Comparisons of the results with reported literature values indicate that concentrations of most elements determined in egret livers from the RMSP are higher or of the same order of magnitude as those of polluted areas. Comparisons made between element concentrations for different genders of egrets by applying the nonparametric Mann-Whitney U test, (p < 0.05) indicated that females present significant lower concentrations of Cs, Hg, Se and Zn. These results may be associated with physiological and ecological differences between the genders. By using the matrix of Pearson (r = 0.7, α <0.05 and p <0.01, p <0.05) correlation between element concentrations were evaluated. The positive correlations obtained between the elements Cd, Cu, Hg, Mn, Se and Zn have indicated common sources of these elements, mainly associated with anthropogenic activities in the RMSP. The findings demonstrated the applicability of using livers of egrets to study the contamination in the RMSP and corroborate which monitoring of chemical elements in the ardeids can be an important source of information for studying conservation of these species.

Mestrado em Engenharia do Ambiente<br>Os solos ardidos são mais suscetíveis à erosão, consequência do
desaparecimento do coberto do solo provocado pelo incêndio e ainda devido
às alterações das características físicas e químicas do próprio solo,
nomeadamente a nível da repelência, porosidade e estrutura. Portugal,
devido a características como a aridez dos solos e precipitação irregular e
intensa, encontra-se inserido no contexto da problemática da degradação dos
solos, dado o elevado risco de incêndio associado aos países do Mediterrâneo.
Neste trabalho, enquadrado no projeto FIRECNUTS (PTDC/AGRCFL/
104559/2008), analisou-se a exportação hídrica da matéria orgânica do
solo como consequência do incêndio ocorrido em 26 de Julho de 2010, na
localidade de Ermida, pertencente ao concelho de Sever do Vouga, distrito de
Aveiro, na região centro-norte de Portugal. A área ardida tinha
aproximadamente 295 ha. Dentro desta área foram selecionadas três
encostas, dois eucaliptais com geologia diferente (granito e xisto) e um pinhal
(xisto) para o estudo do efeito sobre diferentes geologias e povoamentos
florestais. Fora da área ardida foi ainda selecionado um eucaliptal não
queimado (xisto), como base na comparação, para a avaliação do efeito do
incêndio.
Em cada uma das encostas foram instalados tanques de recolha de
escorrência, quatro à escala de encosta e três micro-parcelas, à exceção da
área não ardida que apenas apresentava as parcelas à escala de encosta.
Além desta instalação foram ainda caracterizados os solos ao nível da matéria
orgânica, textura e densidade aparente. A monitorização semanal iniciou-se
aproximadamente 1 mês após o incêndio e teve a duração de 6 meses. Os
resultados indicam, à microescala, maior risco de erosão em solos sobre xisto
(259g.m-2) que em solos sobre granito (63 g.m-2), e menor risco de erosão no
pinhal (27 g.m-2). Os resultados à escala de encosta sugerem um
comportamento contrário na comparação de eucaliptais (485 vs 36g.m-2), e
similares na comparação de eucaliptal com pinhal(36 vs 6 g.m-2). A encosta
não queimada permite verificar que o incêndio aumenta a erosão, da mesma
forma que o aumento da escala parece diminuir o risco de erosão, resultado
de maiores interferências, maior tempo de contacto e perda de escorrência.
Pode ainda verificar-se que os teores de matéria orgânica permanecem
constantes ao longo do tempo, em todas as encostas e às duas escalas (cerca
de 60%).<br>Burnt soils are more susceptible to erosion, as a consequence of the
disappearance of the topsoil caused by wildfires and also due to changes in
physical and chemical characteristics of the soil itself, namely, in repellence,
porosity and structure. Portugal, due to features as the aridity of soil and
irregular and intense rainfall, is included in the context of soil degradation, given
the high risk of fire associated to Mediterranean countries. In this study, framed
in the project FIRECNUTS (PTDC/AGR-CFL/104559/2008), hydric exportation of
soil’s organic matter was analysed, as a consequence of the fire that occurred in
the 26th of July in 2010, in Ermida, district of Aveiro, in the Central-North region
in Portugal. The burnt area had about 295 ha. Inside this area 3 slopes were
selected, 2 eucalypt plantations with different geology (granite and schist) and a
pine plantation (schist) for the study of the effects over different types of
geology and woodlands. Outside the burnt area, was also selected an unburned
eucalypt plantation (schist), as a basis of comparison, to evaluate the effect of
the fire. In each of the slopes were installed tanks to collect run-off, four at
slope-scale and three at micro-plot scale, with the exception of the unburned
area that only had slope-scale plots. Besides this installation, soils were
characterized at the level of organic matter, texture and bulk density. The
weekly monitoring began approximately one month after the fire and lasted 6
months. The results indicate, at micro-scale, increased risk of erosion on soils
upon schist (259g.m-2) than in soils upon granite (63 gm-2), and less risk of
erosion in the pine forest (27 gm-2). The results, at slope-scale, suggest an
opposite behaviour in the comparison between eucalypt plantations (granite and
schist) (485 vs 36 g.m-2), and similar behaviour when comparing eucalypt
plantations (schist) with pine forest (schist) (36 vs 6 g.m-2). The unburned slope
shows that fire increases erosion, just as higher scales seem to reduce the risk
of erosion, as a result of bigger interference, more contact time and run-off loss.
It can be also verified that the organic matter levels remain constant over the
time, in all slopes and at both scales (about 60%).

La investigación responde al problema ¿Cómo Influye el motor de CA en la Pala Eléctrica 4100XPC en la mina Cerro Verde - Arequipa?, como objetivo pretende, determinar la influencia del motor eléctrico de CA en la Pala Eléctrica 4100XPC en la mina Cerro Verde – Arequipa y como hipótesis se asume que el motor de CA influye positivamente en la Pala Eléctrica 4100XPC en la mina Cerro Verde - Arequipa. El diseño fue el descriptivo correlacional, en la muestra de los resultados del funcionamiento dela pala eléctrica 4100XPC mediante el motor eléctrico en CA, a quien se aplicó la experimentación, basado en el análisis de criticidad que permitió mejorar la confiabilidad operacional los equipos en la minera Cerro Verde. Concluyendo que el procedimiento del el motor de CA Influye positivamente en la Pala Eléctrica 4100XPC en la mina Cerro Verde – Arequipa la cual permite obtener niveles seguros de funcionamiento y productividad dentro de cada área de trabajo de la Pala eléctrica 4100XPC, también el estudio correlacional permitió confirmar una influencia positiva entre el motor de corriente alterna y la Pala eléctrica 4100XPC en la mina Cerro Verde Arequipa, lo que nos indica que a mejor funcionalidad de la corriente alterna mejor rendimiento de la Pala eléctrica 4100XPC.

In this context, the aim of this work was to study the diversity of endophytic bacteria obtained from soils in western Paraná, using plants such as wheat and corn baits and evaluate the potential of isolates to promote plant growth (PCV). A total of 136 endophytic bacteria isolates were obtaines from roots in three different locations: area 1 (Argissolo vermelho), area 2 (Latossolo vermelho) and area 3 (Nitossolo vermelho). A total of 136 endophytic bacteria strains were isolated from roots in three different locations: area 1 (Argissolo vermelho), area 2 (Latossolo vermelho) and area 3 (Nitossolo vermelho). The isolates were evaluated for their ability to solubilize insoluble phosphate in NBRIP medium and also their ability to synthesize auxin in DYGS medium. Eighty-four bacterial strains were analyzed by molecular analysis of polymorphism by ARDRA and 41 of these fragments were sequenced 16S rDNA. Six isolates randomly chosen were evaluated according to their ability in promoting the plant growth in vitro, where from these, two contrasting isolates were evaluated for their performances in pots. As results for biochemical characterization, from 136 isolates, 51 % showed no phosphate solubilizing capacity, 14 % had a low level of phosphate solubilization index (ISF) low (IS<2), 9.5% medium (IS<4) and 25.5% higher (IS≥4), the higher ISF was verified in UFPRPALM 3-66 (7.66). IAA (indole-3-acetic) values ranged from 19.94 (UFPRPALM1 -134) to 1170.98 mg IAA mg protein-1 (UFPRPALM 2-32) without tryptophan addition and adding this aminoacid , only 42 strains (31%) were able to produce IAA in higher levels ranging from 362.18 (UFPRPALM 3-65) to 5599.57 (UFPRPALM 3-80) mg IAA mg protein -1. The polymorphism analysis revealed by ARDRA bacterial 8 groups with 70% dissimilarity and only 8 isolates showed clonal profile. The sequences of the 16S rDNA allowed the grouping of 41 isolates in eight different phylogenetic groups, among these the bacteria of the genus: Agrobacterium, Shigella, Stenotrophomonas, Enterobacter, Pantoea, Pseudomonas, Ensifer, Brevibacillus, Escherichia, Acinetobacter, Bacillus and Burkholderia. Of these, six isolates producing IAA and P-solubilization ability wich were evaluated for in vitro growth capacity were chosen. Isolated in this experiment, two contrasting were characterized: UFPRPALM 3-87 (Burkholderia ambifaria), more and UFPRPALT 1-14 (Pantoea ananatis), with lower ability to promote the growth of wheat seedlings in vitro, respectively. In the pot experiment, the isolates showed no responses of plant growth promotion in high fertility soils, but at low fertility condition, the genus Pantoea (UFPRPALT 1-14) showed the best performance in terms of increases in nitrogen concentration of shoots in wheat. There was a differential response of the isolates in terms of epiphytic and endophytic microbial population and between plant species and conditions of fertilization and/or inoculation<br>As bactérias promotoras de crescimento vegetal constituem um amplo grupo de microrganismos, que associados às plantas representam um enorme potencial a ser explorado, por contribuírem beneficamente com o crescimento de várias espécies vegetais. Neste contexto, o objetivo do presente trabalho foi estudar a diversidade de bactérias endofíticas obtidas de solos do oeste do Paraná, usando plantas de milho e trigo como iscas e avaliar o potencial dos isolados para a promoção do crescimento vegetal (PCV). Foram obtidos 136 isolados bacterianos provenientes de raízes de trigo e milho semeadas em três diferentes locais: área 1 (Argissolo vermelho), área 2 (Latossolo vermelho) e área 3 (Nitossolo vermelho). Os isolados foram avaliados quanto à capacidade de solubilizar fosfato insolúveis em meio NBRIP e quanto à capacidade de sintetizar auxina em meio DYGS. Oitenta e quatro estirpes bacterianas foram analisadas molecularmente através da análise de polimorfismo por ARDRA e 41 destas tiveram fragmentos do gene 16S rDNA sequenciados. A avaliação da capacidade de promoção de crescimento de seis dos isolados foi realizada in vitro, onde destes, dois isolados contrastantes foram avaliados quanto as suas performances em vaso. Como resultados obtidos, dos 136 isolados, 51% não apresentaram capacidade solubilizadora de fosfato, 14% apresentaram índice de solubilização de fosfato (ISF) baixo (IS<2), 9,5% médio (IS<4) e 25,5% alto (IS≥4), destacando-se o isolado UFPRPALM 3-66 (ISF 7,66). Os valores de AIA (ácido indol-3-acético) variaram entre 19,94 μg de AIA mg-1 de proteína (UFPRPALM1-134) a 1170,98 μg de AIA mg-1 de proteína (UFPRPALM 2-32), sem a adição de triptofano, e com adição de triptofano, apenas 42 isolados (31%) foram capazes de produzir AIA, sendo os níveis maiores variando de 362,18 (UFPRPALM 3-65) a 5599,57 (UFPRPALM 3-80) μg de AIA mg-1 de proteína. A análise de polimorfismo por ARDRA revelou 8 grupos bacterianos com 70% de dissimilaridade e apenas 8 isolados apresentaram perfil clonal. As sequencias do gene 16S rDNA permitiu o agrupamento dos 41 isolados em oito grupos filogenéticos diferentes, entre estes estão as bactérias do gênero: Agrobacterium, Shigella, Stenotrophomonas, Enterobacter, Pantoea, Pseudomonas, Ensifer, Brevibacillus, Escherichia, Acinetobacter, Bacillus e Burkholderia. Destes, foram escolhidos seis isolados produtores de AIA e solubilizadores de P que foram avaliados in vitro quanto a capacidade de crescimento. Neste experimento, dois isolados contrastantes foram caracterizados: UFPRPALM 3-87 (Burkholderia ambifaria), com maior e o UFPRPALT 1-14 (Pantoea ananatis), de menor capacidade de promoção de crescimento das plântulas de trigo in vitro, respectivamente. Em vaso, os isolados não apresentaram respostas de promoção de crescimento vegetal em solos de alta fertilidade, mas em condições de baixa fertilidade, o gênero Pantoea (UFPRPALT 1-14) foi o que apresentou a melhor performance em termos de aumentos dos teores de nitrogênio da parte aérea em trigo. Previous issue date: 2009-09-24<br>Universidade Federal de Minas Gerais<br>The present work studied populations of Ardea alba egretta (Great Egret) family Ardeidae (Aves), sampled from four Brazilian regions situated at different latitudes (Rio Grande do Sul, Pantanal, São Paulo and Amapá). Species-specific primers developed in this study allowed the sequencing of 194 individuals in the Domain I of mitochondrial DNA control region (fragment of 586 bp). Fifty-eight polymorphic sites were found, defining 74 haplotypes. Haplotype Hap9 was the most frequent and the majority of the remaining haplotypes occurred in low frequencies. Average nucleotide diversity was 0.006 and average haplotype diversity 0.908. Distribution of nucleotide diversity followed a descending order: Amapá> Pantanal> São Paulo> Rio Grande do Sul, and the differences between Amapá and the other populations were statistically significant. Results of AMOVA analysis indicated that there is genetic differentiation among populations of the four regions (Fct = 0.02145, p = 0.03324). Values of the pairwise F-statistics showed low genetic structuring among the colonies within each region, but significant structuring among the four regions. It was evident by the analysis of structuring that the genetic composition of Amapá is different from other regions. Significant differences revealed by the tests of Fu's Fs, Tajima's D, R2 and analyses of mismatch distribution, together with star-shaped haplotype networks, indicated signals of demographic expansion in the populations of Rio Grande do Sul and Pantanal. For the Amapá population only biases of Fu's Fs were significant and the curves of "mismatch distribution" can be explained by the unimodal standard distribution. The estimated time of expansion (τ) for Rio Grande do Sul population was 7,195 years before present, for Pantanal population was 32,009 and for Amapá population was 68,674. Obtained results are consistent with the hypothesis that the equatorial region likely was a refuge for populations of this species during the Pleistocene glaciations.<br>Foram estudadas populações de Ardea alba egretta (garça-branca-grande) da família Ardeidae (Aves), amostradas em quatro regiões brasileiras, com latitudes diferentes (Rio Grande do Sul, Pantanal, São Paulo e Amapá). Oligonucleotídeos espécie-específicos desenvolvidos nesse estudo possibilitaram o seqüenciamento de um fragmento de 586 pb do Domínio I da região controladora do DNA mitocondrial em 194 individuos. Cinqüenta e oito sítios polimórficos foram encontrados, definindo 74 haplótipos. O haplótipo de maior freqüência foi o Hap9 e a maioria dos demais haplótipos apresentaram freqüências baixas. A diversidade nucleotídica média foi de 0,006 e a diversidade haplotípica média 0,908. A distribuição da diversidade nucleotídica seguiu ordem decrescente do Amapá > Pantanal > São Paulo > Rio Grande do Sul e as diferenças entre Amapá e as demais populações foram estatisticamente significativas. Ficou evidente pelas análises de estruturação que a composição genética do Amapá difere das demais regiões estudadas. Significantes desvios nos testes de Fs de Fu, D de Tajima, R2 e as análises da distribuição das diferenças par-a-par ( mismatch distribution ) e redes haplotípicas em formato de estrela apontam para sinais de expansão demográfica nas populações do Rio Grande do Sul e do Pantanal. Para o Amapá apenas os desvios de Fs de Fu foram significativos e as curvas de mismatch distribution podem ser explicadas pelo padrão unimodal. A estimativa do tempo de expansão (τ) para o Rio Grande do Sul foi de 7.195 anos antes do presente, para o Pantanal foi de 32.009 e para o Amapá foi de 68.674, anos antes do presente. Os resultados obtidos foram concordantes com a hipótese de que a região equatorial possivelmente foi o refúgio para as populações dessa espécie durante glaciações ocorridas no Pleistoceno.

Previous issue date: 2014-02-20<br>CAPEs<br>Zymomonas mobilis vem despertando um grande interesse no meio científico, industrial e biotecnológico devido ao seu alto potencial fermentativo. Do ponto de vista taxonômico, Z. mobilis é a única espécie do gênero Zymomonas, e é subdivida em três subespécies: Z. mobilis subsp. mobilis, Z. mobilis subsp. pomaceae e Z. mobilis subsp. francensis. A diferenciação destas subespécies é baseada em testes fisiológicos. Estes testes consomem tempo e são frequentemente duvidosos. Por isso, técnicas moleculares são propostas como uma alternativa rápida e confiável para diferenciação destas bactérias. O presente estudo teve como objetivo realizar a caracterização molecular das 32 linhagens de Zymomonas mobilis depositadas na Coleção de Microrganismos UFPEDA, através da análise das sequências do gene 16S rDNA e ARDRA. As linhagens foram cultivadas em meio SSDL por 24 horas à 30º, seguida de centrifugação e extração de DNA cromossômico. As reações de PCR foram realizadas com iniciadores e condições específicas para a amplificação do gene 16S rDNA. Os produtos do gene 16S rDNA amplificados foram purificados, sequenciados e clivados com as enzimas de restrição Hae III, NdeII e StuI. Os dados obtidos pelo sequenciamento do gene 16S rDNA foram analisados, comparados e alinhados, pelo programas BLASTn e MultiAlin, com sequências de linhagens de Z. mobilis previamente depositadas no banco de dados GenBank. Um dendograma foi construido através do programa ClustalW pelo método de neighbor-joining Os perfis de restrição teórico das enzimas de restrição Hae III, NdeII e StuI foram gerados a partir do WebCutter 2.0. Dendogramas foram construídos a partir da matriz de similaridade genética de Jaccard, calculada pela análise dos perfis de restrição teóricos de cada enzima. A análise das sequências obtidas no presente estudo revelou o elevado grau de conservação no gene 16SrDNA, confirmando a relação de proximidade das linhagens de Zymomonas mobilis depositadas na Coleção de Micro-organismos UFPEDA e a aproximidade com a Z. mobilis subsp. mobilis LMG445, sugerindo que as linhagens desta coleção pertencem a esta subespécie.Além disso, conclui-se que a análise do perfil restrição teórico do gene 16S rDNA possibilita a diferenciação de Z. mobilis,a nível de subespécie, mas não é eficaz para analisar a variabilidade genética entre as linhagens de Z. mobilis UFPEDA. Baseados nestes resultados, outros marcadores filogenéticos devem ser empregados para analisar a variabilidade genética destas linhagens, possibilitando um melhor conhecimento da diversidade desta bactéria.<br>Zymomonas mobilis has attracted great interest in the scientific, industrial and biotechnological medium due to its high fermentation potential. The taxonomic viewpoint, Z. mobilis is the only species of the genus Zymomonas , and is subdivided into three subspecies : Z. mobilis subsp. mobilis, Z. mobilis subsp. pomaceae and Z. mobilis subsp. francensis. The differentiation of these subspecies is based on physiological tests. These tests are time consuming and often unreliable. Therefore, molecular techniques are proposed as a fast and reliable alternative to differentiation of these bacteria. This study aims to perform molecular characterization of 32 strains of Zymomonas mobilis deposited in the Collection of Microorganisms UFPEDA by sequence analysis of 16S rDNA gene and the theoretical restriction profile of this gene. The strains were grown in SSDL for 24 hours at 30 ° , followed by centrifugation and extraction of chromosomal DNA . PCR reactions were performed with primers and specific conditions for amplification of the 16S rDNA gene. The amplified products of 16S rDNA were purified, sequenced, and cleaved with restriction enzymes Hae III, NdeII and StuI . The data obtained by 16S rDNA gene were analyzed, compared and aligned by BLASTn and MultiAlin programs with sequences of strains of Z. mobilis previously deposited in the GenBank databas . A dendogram was constructed using the program ClustalW method by neighbor-joining. Profiles theoretical restriction of restriction enzyme Hae III, NdeII and StuI were generated from WebCutter 2.0. Dendrograms were constructed from the genetic Jaccard similarity matrix, calculated by analyzing the theoretical restriction profiles of each enzyme. The analysis of the sequences obtained in this study revealed the high degree of conservation in 16SrDNA gene, confirming the close relationship of strains of Zymomonas mobilis deposited in the Collection of Micro-organisms UFPEDA and closeness with Z. mobilis subsp. mobilis LMG445, suggesting that the strains in this collection belong to this subespécie. In addition, it is concluded that the theoretical restriction profile analysis of the 16S rDNA gene allows differentiation of Z. mobilis , the level of subspecies , but it is not effective to analyze the genetic variability between strains of Z. mobilis UFPEDA . Based on these results , other phylogenetic markers should be employed to analyze the genetic variability of these strains , allowing a better understanding of the diversity of this bacteria.

Appinites are enigmatic rocks commonly associated with major I-type plutons in the Caledonian orogen, a number of different models have been proposed for the origin of their parental magmas and unusual textural characteristics. The appinite cluster associated with the Ardara pluton has been studied using petrological techniques with the aim of understanding these features as well as their relationships to the main pluton. The approach of this study has been to select a few appinite bodies considered to be representative of the whole Ardara appinite suite, and to study each in detail in parallel with a limited study of the main pluton. The techniques adopted include field mapping, petrography, mineral chemistry, whole rock major and trace element geochemistry. The detailed aims were to constrain the origin of the magma, its emplacement history and differentiation, for each selected body. In one case (Portnoo) the role of fluids was investigated in detail using stable isotope geochemistry. The Ardara appinite suite comprises a wide variety of generally amphibole-rich rocks typically including hornblendite, meladiorite, diorite, granodiorite and granite. There is one case of cortlandtite. Most have evidence of a significant volatile phase at crystallisation. The intrusions display a wide range in form and emplacement mechanism and most were emplaced prior to, or coeval with, the diapiric granite pluton of Ardara. They have a history of tectonic control on emplacement and are associated with major shear zones acting as potential magma pathways. Mineral geobarometry and geothermometry indicate that the appinites crystallised at 5-8 kb, and selected mineral pairs equilibrated between 800 and 976°C. G1 and G2 of the Ardara pluton crystallised at 5 kb and amphibole-plagioclase equilibrated at around 800°C. These pressures are high if the appinites are high-level diatremes, and this apparent inconsistency is discussed. Variations in bulk geochemistry are considerable and attempts are made to model these as the products of crystal-liquid processes using mass balance major oxide and Rayleigh trace element techniques. The results tend to be dominated by solutions involving amphibole, other mafic minerals and plagioclase, although in many cases no satisfactory result was obtained which suggests that crystal fractionation is not always the sole cause of differentiation. Stable isotope studies of delta18O, delta13C and 5D from the appinites and country rocks at Portnoo indicate that the dominant fluid in this appinite was magmatic in origin, although crustal contamination had occurred, while 6D data also indicate role for meteoric fluids. The data show that fluids from the appinite penetrated the limestone aureole to about 40m. The compositional characteristics of the Ardara appinite suite support a model of derivation of a hydrous basic magma from a mantle source. These magmas undergo differentiation by multiple processes, including crystal-liquid fractionation, as well as contamination by crustal assimilation and possibly by other magmas. The source of G1 & G2 of the main pluton is relatively primitive and could be immature lower crust. It is possible to derive G1 from G2 by crystal fractionation, but probably not in situ. The central G3 granite was almost certainly derived from a distinct, rather more mature crustal source.

O presente trabalho busca entender a dinâmica de comunidades bacterianas cultiváveis e não cultiváveis do ecossistema do manguezal e prospectar nesse ambiente ainda inexplorado, um possível potencial biotecnológico. As amostras de sedimentos foram retiradas de duas profundidades no inverno e no verão. Bactérias caracterizadas por meio da técnica de ARDRA pertencem as ordens Vibrionales, Bacillales e Actinomycetales. As espécies bacterianas cultiváveis e as não cultiváveis foram também avaliadas por meio da técnica de PCR-DGGE, onde utilizou-se iniciadores seletivos para Actinobacterias, a e b Proteobacteria, Pseudomonas spp. e Paenibacillus spp., além do universal para bacteria. A análise multivariada de redundância (RDA) permitiu verificar a relação dos perfis obtidos das amostras com os fatores ambientais. Verificou-se uma diferente distribuição dos grupos de a e b Proteobacteria em relação à sazonalidade, enquanto que a profundidade de amostragem mostrou ser essencial no perfil das comunidades totais, a e b-Proteobacteria e Actinobacterias.<br>The objective of the present work is the understanding of culturable and non-culturable bacterial community dynamic present in the mangrove ecosystem also to explore the biotechnological potential of bacteria present in this environmental. Sediment samples were obtained from two depths winter and summer seasons and further analyzed. The Bacteria were characterized by ARDRA and identified the orders Vibrionales, Bacillales e Actinomycetales. Culturable and non-culturable bacteria were also assessed by PCR-DGGE using specific primers for Actinobacteria, a-Proteobacteria, b-proteobacteria, Pseudomonas spp., Paenibacillus spp. and bacteria universal primers. Multivariate redundancy analysis allowed the verification of main factors determining the bacterial communities patterns found on samples. It was verified a seasonal distribution of a and b-Proteobacteria groups, while the sampled depth was determinant for the total bacterial community composition, and also influenced the a and b-Proteobacteria and Actinobacteria profiles.

This thesis examines the causes and consequences of habitat selection and timing of breeding of the Great Blue Heron (Ardea herodias). My general hypothesis was that the duration of low tides and seasonal abundance of prey strongly influenced the location of colony-sites; timing of the breeding season; habitat shifts; and the use of space by foraging herons of different age- and sex-classes.
I studied Great Blue Herons along the Pacific coast of Canada for five breeding seasons and four winters. Breeding herons were studied at a colony of 85 to 100 pairs on Sidney Island near the town of Sidney, and periodic visits were made to about 40 other colonies around the Strait of Georgia, British Columbia. At Sidney, I studied the foraging behaviour, food availability, habitat use and reproductive success in detail. At other colonies, I recorded the reproductive success of herons, located their main feeding areas and searched for nests of a predator, the Bald Eagle. In the non-breeding season, I investigated the foraging behaviour, dispersion pattern and habitat shifts of juvenile and post-breeding adult herons in the Fraser River delta.
I hypothesized that heron colony-sites were located near food supplies or away from predators. Twenty-nine of 33 colony-sites were located within 6 km of their main feeding site. The number of heron pairs was slightly greater where eagles nested in high abundance than where eagle abundance was low, contrary to the hypothesis that breeding herons avoid areas with active eagle nests.
I hypothesized that herons began breeding in spring shortly after females acquired enough food energy to make eggs, or so chicks were in nests when food was most plentiful to their parents. Egg-laying began about 9 days after a
female's daily food intake crossed an energy threshold of 1715 kJ/day, whereas the peak availability of food energy to adults occurred about 35 days before the peak food demands of their chicks. Food intake rates by adults increased gradually in March and April with the increasing duration of low tides and the inshore movement of fishes. Adult food intake rates reached a peak in May when sea perch were most abundant, and diminished through June and July.
Most juvenile and adult female herons foraged on beaches from February to October and in marshlands and grasslands from November to January. Some males returned to territories along riverbanks in August and remained there until the start of the next breeding season in March.
I tested the hypothesis that herons leave foraging habitats in autumn when they can no longer catch enough food or when interference from conspecifics reduced foraging intake rates below a threshold required to maintain their energy balance. In October and November adults moved to marshlands and juveniles moved to grasslands when they could no longer maintain daily energy balance on beaches as a result of declining duration of low tides and food intake rates. As a result, there is a growing need to integrate ML models and data curation into a homogeneous system such that the model informs the choice and extent of data curation. The bottleneck in designing such a system is to efficiently discern what additional information would result in improving the generalization of the ML models. We design an end-to-end system that takes as input a data set, a ML model and access to unstructured data, and outputs an augmented data set such that training the model on this dataset results in better generalization error. Our system has two distinct components: 1) a framework to search and join unstructured data with the input data, based on various attributes of the input and 2) an efficient feature selection algorithm that prunes our noisy or irrelevant features from the resulting join. Termites are known to harbor within their gut a diverse assemblage of symbiotic microorganisms. Little work has been done, however, to describe the diversity and function of the bacteria in the economically important eastern subterranean termite, Reticulitermes flavipes.
The first object of this study was to characterize the bacterial diversity in the gut of R. flavipes using amplified rDNA restriction analysis (ARDRA) and 16S rRNA gene sequencing. It was determined that ARDRA was an effective technique for characterizing the diversity of the termite gut microbiota. Of the 512 clones analyzed in the ARDRA study, 261 different ARDRA profiles were found. Forty-two 16S rRNA gene sequences were also analyzed, resulting in 33 different ribotypes. Representatives from six major bacterial phyla, Proteobacteria, Spirochaetes, Bacteroidetes, Firmicutes, Actinobacteria, and the newly proposed â Endomicrobia,â were discovered. Further analysis indicated that the gut of R. flavipes may harbor as many as 1,318 ribotypes per termite.
The second objective was to determine if the gut bacterial diversity could be manipulated by changing the termiteâ s food source. Using ARDRA analysis, I found no evidence that changing the food source affected the termite gut bacterial diversity. In addition, changing the food source did not induce aggression in nestmates fed on different food sources.
The third objective was to search for patterns of coevolution between termites and their gut symbiotic bacteria. Using rRNA gene sequences from this study and sequences from public databases (1,450 sequences total), a neighbor-joining tree demonstrated strong evidence for coevolution of termites and their symbiotic bacteria, especially in the phyla Bacteroidetes, Actinobacteria, Spirochaetes, and â Endomicrobia.â Many monophyletic clusters were entirely composed of phylotypes specific to Isoptera.<br>Ph. The current trend in computer game design is toward larger and richer virtual worlds, providing interesting and abundant game content for players to explore. The creation and continuous expansion of detailed virtual environments, however, is a time and resource-consuming task for game developers. Procedural content generation potentially solves this problem; textures, landscapes, and more recently the creation of entire cities and their constituent roads and buildings can be performed in an auto mated fashion, potentially offering considerable resource savings for developers. This thesis develops a comprehensive system for content generation, centering on a hierarchical world design. The Arda tool is presented as a modular system for supporting automatic content generation in a game context. Arda is composed of a framework for internally managing environmental data and content-generation algorithms, in addition to a graphical tool providing a simplified interface for generating video game content. The tool supports extensive customization, which is demonstrated through a novel algorithm for efficient and realistic city (road-plan) generation. Experimental results show the design is practical, producing qualitatively realistic results. It is also quite fast; large city plans, extending to the size of major real-world cities can be generated in a few seconds. The Arda design demonstrates the feasibility of automatic content generation, providing a practical and flexible system for quick prototyping, development and testing of game assets.<br>La nouvelle tendance dans la conception de jeux vidéo vise des environnements vastes et détaillés, pour mettre en valeur l’exploration de l’univers virtuel. Cependant, la création de ce contenu impose au développeur un investissement majeur au niveau des ressources et du temps requis pour la production. L’emploi du contenu procédural pourrait résoudre le problème: la création de textures, de paysages, et finalement de villes entières—incluant des réseaux routiers et des bâtiments complexes—se fait de façon automatisée, ce qui représente une économie considérable pour les développeurs. Cette thèse présente un système de génération de contenu procédural axé sur une conception hiérarchique du monde virtuel: Arda, un outil modulaire qui facilite la création automatique de contenu pour les jeux vidéo. Arda est composé d'un cadre d'applications pour la gestion de données environnementales et d'algorithmes procéduraux, ainsi que d'une interface d'utilisation graphique qui simplifie le processus de création de contenu. L'outil est fortement paramétrable, ce qui est démontré grâce à un nouvel algorithme, conçu ci-après, pour la création efficace et réaliste de réseaux routiers. Les essais prouvent que le concept est pratique et produit des résultats très réalistes. De plus, le système est rapide: des villes virtuelles, de la même dimension que des villes actuelles, sont construites en quelques secondes. Le système Arda démontre la faisabilité de la génération de contenu procédural, fournissant une méthode pratique et modulaire pour le dévelopement et la validation rapide de contenu pour les jeux vidéo.

The present study had how objective to determine the genetic diversity existing between the isolates of F. oxysporum f. sp. cubense collected in heliconia and banana through the ITS region of rDNA using ARDRA (Amplified Ribossomal DNA Restriction Analysis), markers like RAPD (Random Amplified Polymorphic DNA). In the ARDRA analysis products amplified by ITS1 and ITS4 primers were digested by restriction enzymes Hae III, Xho I, Hind III and Eco IV by 37ºC. But only enzyme Hae III showed to be efficient a molecular marker, to analysis genetic diversity of F. oxysporum f. sp. cubense to RAPD marker was realized the Polymerase Chain Reaction (PCR) using tenoligonucleotides follow arbitrary up, only seven oligonucleotides generated a total 44 bands polymorphic with fragments varying from 100 pb to 700 pb. The two techniques used in the study genetic diversity showed high genetic variability of fungal, don’t related to with geographical region where were colleted the isolates.<br>As helicônias são as flores mais cultivadas dentro da floricultura tropical no Nordeste brasileiro, devido a características como beleza, exoticidade, cores exuberantes e rusticidade, porém a produção de várias espécies vem sendo afetada pela murcha de fusário, causada pelo fungo de Fusarium oxysporum f. sp. cubense. O presente trabalho teve como objetivo caracterizar a diversidade genética existente entre os isolados de F.oxysporum f. sp. cubense, coletados em helicônias e bananeiras através da análise da região ITS do rDNA utilizando ARDRA (Amplified Ribossomal DNA Restriction Analysis) e marcadores do tipo RAPD (Random Amplified Polymorphic DNA). Para a amplificação da região do rDNA, foram utilizados os primers ITS1 e ITS4 que foram digeridos pelas enzimas de restrição Hae III, Xho I, Hind III e Eco IV a 37ºC. Porém apenas a enzima Hae III, neste trabalho mostrou ser um marcador molecular eficiente,utilizada na técnica de ARDRA para estudos de diversidade genética dos isolados. Para o marcador RAPD realizou-se a Polymerase Chain Reaction (PCR) utilizando-se dez oligonucleotídeos de seqüência arbitrária, onde apenas sete oligonucleotídeos geraram um total de 44 bandas polimórficas com fragmentos que variaram de 100pb a 700pb. As duas técnicas utilizadas no estudo de diversidade genética evidenciaram alta variabilidade genética do fungo, não relacionada com as regiões geográficas onde foram coletados os isolados.

De entre estes, o número de mini-hídricas aumentou consideravelmente, em Portugal, a partir de 1988, aquando da abertura do sector da produção de energia eléctrica à iniciativa privada. Neste âmbito, surgiram vários empreendimentos mini-hídricos, em particular nos pequenos rios do Norte de Portugal. Face a este crescimento, e tendo em conta a insuficiência de estudos relacionados com as alterações provocadas pelo regime de funcionamento das mini-hídricas, neste trabalho pretendeu-se atingir os seguintes objectivos: Analisar as dinâmicas espaciais e temporais, dos principais factores abióticos e da comunidade piscícola do rio Ardena, ao longo de toda a zona do sistema sujeita à influência da central mini-hídrica aí existente; avaliar a existência de diferenças nas comunidades piscícolas encontradas a jusante e a montante da mini-hídrica.O estudo foi efectuado, no rio Ardena, que desagua no rio Paiva, afluente da margem esquerda do rio Douro, onde se encontra implementada, aproximadamente, há 10 anos, uma central mini-hídrica. Dissertação para obtenção do Grau de Mestre em Biotecnologia<br>A diversidade de leveduras encontradas em uvas da casta Touriga Nacional (TN) foi avaliada em três vinhas de duas regiões do país. Os isolados obtidos às 0 e 48 horas após o esmagamento das uvas e após a perda de peso de 70 g/l do mosto, foram caracterizados por análise dos perfis de restrição de uma região do rDNA 26S, com as enzimas HaeIII, CfoI, MseI, HinfI e ApaI. Dos 570 isolados analisados, identificaram-se 21 espécies de leveduras, tendo a espécie H. uvarum sido a mais detectada em todas as vinhas. Observou-se uma boa correspondência entre a identificação obtida pelos perfis de restrição e os resultados da sequenciação.
Nas vinhas de Dois Portos, Palhacana e Montemor-o-Novo a diversidade de espécies detectadas foi diferente, observando-se maior biodiversidade para a vinha de Dois Portos. Embora para a vinha de Palhacana tenha sido encontrado um maior número de espécies, a grande maioria dos isolados pertencia apenas a uma espécie, reflectindo-se numa menor biodiversidade. Para a vinha de Montemor-o-Novo a diversidade de espécies foi reduzida.
Realizou-se a caracterização enológica de isolados das espécies encontradas às 70 g/l de perda de peso, sendo avaliados 23 isolados. Para os isolados em estudo observou-se um perfil de fermentação em massas vínicas de TN, mais lento do que para uma levedura comercial, utilizada como referência. Os isolados da espécie I. terricola revelaram-se os maiores produtores de sulfureto de hidrogénio, detectado química e sensorialmente. Verificou-se um efeito altamente significativo da espécie para a maioria dos parâmetros químicos avaliados. Efeito da espécie também foi observado para os descritores sensoriais do aroma ortonasal, aroma retronasal e caracterização do sabor. Sensorialmente os fermentados obtidos pelos isolados de C. zemplinina foram os que apresentaram intensidade significativamente maior dos aromas característicos da casta Touriga Nacional, designadamente esteva, bergamota e violeta e melhor apreciação global.

portuguese novels of the 60s Maina Mendes (1969), by Maria Velho da Costa, and A noite
e o riso (1969), by Nuno Bragança. Produced in a dictatorial period, these works point to this
context, although not mentioning it directly, acting as a delicate instrument of combat. To this
end, these novels utilize resources that rely on different processes of signification, through
which they reveal, in a sinuous and not rarely cynical way, a reality that could not be referred
to and much less criticized explicitly. The analysis carried out focuses on these processes in
order to observe how they act as triggers of a subtextual level of political nuance, helping to
configure what we call tactical discourse . The proposed analysis is supported by literary
and linguistic studies, as well as some theoretical formulations form the social science.<br>Neste estudo examinamos formas de produção e disseminação de sentidos em dois romances
portugueses da década de 60 Maina Mendes (1969), de Maria Velho da Costa, e A noite e
o riso (1969), de Nuno Bragança. Produzidas num período ditatorial, as obras selecionadas
apontam para esse contexto, ainda que sem referi-lo diretamente, atuando como um delicado
instrumento de combate. Para tanto, esses romances se utilizam de recursos que se apoiam em
variados processos de significação, por meio dos quais revelam, de forma sinuosa e não raro
cínica, uma realidade que não poderia ser referida e muito menos criticada explicitamente. A
análise que realizamos focaliza esses processos, de modo a observar de que maneira eles
atuam como deflagradores de um nível subtextual de matiz político, contribuindo para a
configuração do que chamamos discurso tático . A análise proposta busca apoio em estudos
literários e linguísticos, como também em algumas formulações teóricas das ciências sociais.

Foraging patterns of breeding Grey Herons were studied in the Camargue. Most individual birds used several different and widely dispersed feeding sites during the season. Some birds abandoned certain foraging sites but no seasonal trends were detected. There was evidence of both flock and territorial feeding at different sites. Birds were observed feeding in a number of different food patches (discrete feeding sites). There was evidence that the biomass intake rate influenced how long a bird would remain in a patch. When this was high, birds remained in patches, conversely when the rate was low birds left to feed elsewhere. Departures from a breeding colony to the feeding grounds were clumped. Birds leaving the colony together were likely to go to the same feeding site more often than birds leaving successively, but not together. I t is argued that the colony may have been used as an information centre. The diet of chicks in different colonies was compared. The proportions of the important prey types from different colonies and from different broods were different. There was evidence that adults sometimes pre-digest large prey so that their small chicks can consume prey that would normally be too large for them to eat. An experiment on prey selection provided evidence that chicks could select between two prey types which differed only in the irrelative profitability. The chicks consumed the most profitable prey type first. When the relative profit ability of the prey types was reversed, the chicks reversed their selection. This may be an adaption to maximise prey intake rate in the face of sibling competition. The results are discussed in relation to the success of the birds at different stages of their life cycle. The implications for the conservation of the species are considered.

A utilização de novos isolados de Lactobacillus com características probióticas impõe uma seqüência de testes bioquímicos que visam à verificação do potencial biotecnológico destas linhagens. Testes bioquímicos permitiram selecionar 08 linhagens com características probióticas isoladas de fezes de crianças saudáveis da creche da Universidade Estadual de Londrina. No presente trabalho, essas linhagens foram identificadas molecularmente através da técnica de ARDRA (Amplified Ribosomal DNA Restriction Analysis) e um isolado foi escolhido para a obtenção de um produto probiótico. Amplificou-se a região intergênica 16S - 23S do DNA ribossomal extraído dos isolados por PCR e os produtos foram submetidos à hidrólise com 11 enzimas de restrição. Os produtos desta reação foram verificados em eletroforese em gel de agarose 2,0% corado com brometo de etídeo 20mg/ml sob incidência de luz ultravioleta e fotodocumentado. As linhagens foram então, identificadas quanto as suas espécies como Enterococcus faecalis (2), Enterococcus faecium, Lactobacillus plantarum (04) e Bifidobacterium lactis e uma das linhagens de Lactobacillus plantarum foi submetida a estudo em leite desnatado reconstituído a 9,0%, no qual foram definidos os parâmetros de obtenção de um produto probiótico, o qual foi avaliado quanto às condições de crescimento, concentração e viabilidade dos microrganismos durante 60 dias de armazenamento a 5°C, bem como teste microbiológico para controle de qualidade. O leite probiótico atingiu uma acidez titulável máxima de 0,31%, não sendo considerado um leite fermentado por não ter atingido o mínimo de acidez preconizado pela legislação brasileira que é de 0,60%. A concentração de células viáveis manteve-se acima de 107 UFC/ml durante o período de armazenamento, acima do mínimo recomendado pela legislação que é de 106 UFC/ml. Seis marcas de leites fermentados comerciais foram avaliadas quanto ao pH, a acidez titulável, a concentração e a viabilidade dos microrganismos presentes. Todas as marcas se apresentaram dentro das normas exigidas pela legislação brasileira vigente quanto aos padrões para leite fermentado. A linhagem identificada como Lactobacillus plantarum apresentou características favoráveis à produção de um leite probiótico, atingindo uma concentração de células viáveis acima de 107 UFC/ml e mantendo essas contagens por no mínimo 60 dias durante a estocagem a 5°C.<br>The use of new isolates of characteristic probiotic Lactobacillus requires a sequence of biochemical tests aiming to verify the biotechnological potential of these strains. Biochemical tests allowed selecting 08 probiotic strains isolated from healthy children's feces in a day care center at the State University of Londrina. In this research, these strains have been molecularly identified through the technique of ARDRA (Amplified Ribosomal DNA Restriction Analysis) and an isolated was chosen to obtain a probiotic product. The 16S?23S Ribosomal DNA intergenic spacers regions were extracted from the isolated by PCR and the products were submitted to hydrolysis with 11 restriction enzymes. The products of this reaction were electrophoresed in a 2.0% agarose gel, stained with ethidium bromide 20mg/ml, verified by ultraviolet light transillumination and photo documented. The strains were then identified as belonging to Enterococcus faecalis (2), Enterococcus faecium, Lactobacillus plantarum (04) and Bifidobacterium lactis species and one of the strains of Lactobacillus plantarum was used in this study. The strain was applied in a 9.0% w/w solids reconstituted skimmed milk to define the parameters for obtaining a probiotic product, which was evaluated for the conditions of culture growth, microorganisms? concentration and viability for 60 storage days at 5°C, and microbiological testing for quality control. The probiotic milk reached a maximum titratable acidity of 0.31% and is not considered fermented milk according to the Brazilian legislation that is 0.60%. The concentration of cells remained above of 107 CFU/ ml during the storage period, which is above the minimum recommended by the legislation, which is 106 CFU/ ml. Six commercial fermented milk were assessed for pH, titratable acidity, concentration and viability of probiotic microorganisms present. All of them were found within the standards required by Brazilian law for fermented milk. The strain identified as Lactobacillus plantarum presented favorable characteristics for a probiotic milk development, reaching a concentration of cells above 107 CFU/ ml and maintaining these counts by at least 60 storage days at 5°C.

Gene orf18, which is situated in the conjugative transposon Tn916 from the bacterial pathogen Enterococcus faecalis, encodes a putative ArdA (alleviation of restriction of DNA) protein. ArdA from Tn916 may be responsible for the apparent immunity of the transposon to DNA restriction and modification (R/M) systems and for ensuring that the transposon has a broad host range. The orf18 gene was engineered for overexpression in Escherichia coli and the recombinant ArdA protein was purified to homogeneity. Biophysical characterisation of ArdA demonstrated tight association between ArdA and the M.EcoKI. Also, ArdA was shown to efficiently inhibit restriction and modification by all four major classes of Type I R/M enzymes in vivo. Thus, ArdA can overcome the restriction barrier following conjugation and so helps to increase the spread of antibiotic resistance genes by horizontal gene transfer. The amino acid sequence of KlcA, from the incompatibility plasmid pBP136 from Bordetella pertussis, showed a high degree of similarity with the antirestriction protein ArdB from the IncN plasmid pKM101. In this study the solution structure of KlcA was solved with high-resolution NMR and its antirestriction function demonstrated. The structure of KlcA showed a rigid globular molecule with a novel fold. No antimodification function was observed for KlcA in vivo and the antirestriction function of KlcA has been successfully shown in vivo but not in vitro. Because no direct binding of KlcA to EcoKI was observed in vitro, the mechanism of the endonuclease blocking was assumed to be different from that of ArdA. Preliminary experiments including coimmunoprecipitation assays were conducted in order to elucidate the antirestriction mechanism of KlcA.

Atividade microbiana foi monitorada através de técnicas clássicas na presença do organoclorado na faixa de 2,0 a 13 mg/L de PCP. O reator apresentou eficiência de 97% na remoção de DQO e completo desaparecimento do composto de PCP em todas as concentrações testadas. A fração orgânica foi consumida totalmente na primeira terça parte do reator de acordo com os valores determinados de ácidos voláteis, DQD e PCP. Não foi verificada inibição da atividade de culturas microbianas. Os exames microscópicos, fluorescência e varredura, permitiram verificar o predomínio de microrganismos pertencentes ao Domínio Archea. As técnicas moleculares PCR, ARDRA e hibridação slot-blot confirmaram o predomínio do Domínio Archaea e possibilitaram a verificação de alterações na diversidade das populações após adição de 2 mg PCP/L. Conclui-se que o reator sem prévia adaptação do inóculo foi eficiente para o tratamento do PCP, e os microrganismos relacionados às Archaea metanogênicas acetocláticas podem estar envolvidas na degradação deste composto.<br>The degradation of pentachlorophenol (PCP) was studied in a 2000 mL. Horizontal Anaerobic Immobilized System (HAIS). The reactor was inoculated with microorganisms obtained from an anaerobic reactor without previous adaptation to the PCP. The microbial activity was evaluated by using classic techniques in order to . monitor its behavior during the HAIS fed with a range of PCP between 2.0 to 13 mg/L. The reactor presented 97% of efficiency in the removal of COD and complete decrease of PCP in alI concentrations tested. The total consumption of organic fraction took place mainly in the first third part of the reactor according the values of volatile fatty acids, COD and PCP obtained. Microbial inhibition was not verified in during HAIS operation. Microscopic examinations allowed certifying the Archaea Domain predominance according the morphologies observed. The molecular techniques polimerase chain reaction (PCR), ARDRA and slot-blot hibridation confirmed the predominance of Archaea Domain and alIowed verifying some changes in the population\'s diversity under additions of 2mg PCP/L. The efficiency of PCP decreased in the anaerobic reactor was related to the presence of Archaea Domain, especially the acetoclastic methanogens, whose where probably involved with the organochlorine compound degradation.

Analisamos esses documentos como construções discursivas da identidade escrava realizada conforme as convicções dos senhores, dentro de um quadro de expectativas do que se pretendia em termos de qualidades, virtudes e defeitos dos cativos. Apesar dessa documentação se caracterizar pelo olhar de quem as escreve, ou seja, os donos desses escravos, uma análise mais detalhada nos permitiu perceber aspectos das lutas diárias, hábitos, valores, formas de vestir, de comemorar e de reivindicar melhores condições de vida dos cativos. No desenvolvimento da análise percebemos que os periódicos tinham uma importância fundamental para a sociedade do Espírito Santo do século XIX. Esses jornais cumpriram um papel social que foi além das questões políticas partidárias defendidas pelos grupos e associações que os mantiveram e também funcionaram como uma espécie de justiça informal, atuando como uma espécie de poder coercitivo, alertando os infratores da lei de que a persistência no acoitamento acarretaria querelas jurídicas, bem como a sua exposição pública. Essa constatação nos direcionou para a hipótese de que ao comunicar um anúncio de fuga de escravo nos jornais, o anunciante buscava além do retorno do cativo, comunicar um problema, qual seja, o acoitamento de sua propriedade privada por outros indivíduos da sociedade, que muitas vezes foram denominados nas fontes como sedutores. Esses sedutores puderam ser melhor identificados neste trabalho com o auxilio de alguns inquéritos encontrados que envolviam essas contendas de fugas de escravos, e que revelaram que esses acobertadores não precisaram de muitos artifícios para seduzir os cativos, ou ainda que em alguns casos eles foram os seduzidos.

We analyze these documents as discursive constructions of slave identity made according to master s convictions, within a patterns of expectations of what is intended in terms of qualities, virtues and defects of the captives. Although this documentation is characterized by the look of the person making, that is, the owners of slaves, a more detailed analysis allowed us to perceive aspects of their daily struggles, habits, values, ways of dressing, to celebrate and to demand better living conditions. In developing the analysis we perceive that the journals had a fundamental importance to society of the Province of the Espírito Santo of the nineteenth century. These newspapers have a social function that went beyond the issues advocated by partisan political groups and associations that remained: they also functioned as a kind of informal justice, acting as a kind of coercive power, warning violators of the law of the persistence in flogging entail legal disputes, as well as their public exposure. This finding directed us to the hypothesis that by communicating a notice of escape of a slave in the newspapers, the advertiser also sought the return of the captive, report a problem, namely, the flogging of private property by other individuals in the society, that many sometimes the sources were named as seductive. These seductive could be better seen in this study with the help of some investigations found these contentions involving leakage of slaves, which revealed that these harbourer did not have many tricks to entice captives, or, in some cases, they were seduced

In Karpfen sächsischer Karpfenteichwirtschaften spielen u.a. die Zestoden Neogryporhynchus cheilancristrotus und Paradilepis scolecina und der Trematode Diplostomum pseudospathaceum eine Rolle (KAPPE 2004, PIETROCK et al. 2006, KAPPE & SEIFERT unveröff.). Weitere Fischarten (Schleie, Plötze, Regenbogenforelle) sind mit den Trematoden Diplostomum sp., Hysteromorpha triloba, Posthodiplostomum cuticola infiziert (PIETROCK et al. 2006). Mit dieser Arbeit sollte der Versuch unternommen werden, die Bedeutung Fisch fressender Vögel als Vektor Fisch pathogener Helminthen in Sachsen aufzuklären. Dafür wurden im Zeitraum von Juni 2003 bis Dezember 2004 97 Kormorane (Phalacrocorax carbo) und 46 Graureiher (Ardea cinerea) aus dem Einzugsgebiet sächsischer Teichwirtschaften auf ihren Befall mit Endohelminthen untersucht. Im Winter 2004 konnte zusätzlich ein Silberreiher (Casmerodius albus) und eine Silbermöwe (Larus argentatus) in die Untersuchungen einbezogen werden. Während des Untersuchungszeitraumes konnten so insgesamt 29 Parasitengattungen bzw. -arten nachgewiesen werden, denen z.T. kommerziell genutzte Süßwasserfische als Zwischenwirte oder paratenische Wirte dienen. Die Parasiten des Kormorans umfassten 15 verschiedene Parasitenarten. Dabei handelte es sich um die beiden Zestoden Ligula intestinalis und Paradilepis scolecina, die sechs Nematoden Baruscapillaria spp., Contracaecum rudolphii, Cyathostoma microspiculum, Desmidocercella incognita, Eucoleus contortus und Syncuaria squamata und die sieben Trematoden Clinostomum complanatum, Holostephanus dubinini, Hysteromorpha triloba, Metorchis xanthosomus, Paryphostomum radiatum, Petasiger exaeretus und P. phalacrocoracis. Außerdem wurden die zwei Kratzer Corynosoma strumosum und Polymorphus sp. nachgewiesen. Im Graureiher konnten dagegen nur neun Helminthenarten nachgewiesen werden. So der Zestode Neogryporhynchus cheilancristrotus, die sechs Trematoden Apharyngostrigea cornu, Bilharziella polonica, Diplostomum sp., Echinoasmus belocephalus, Posthodiplostomum cuticola und Tylodelphis excavata und die drei Nematoden Avioserpens galliardi, Desmidocercella incognita und Desmidocercella numidica. In der Silbermöwe konnten der Zestode Echinocotyle verschureni, die vier Trematoden Apophallus mühlingi, Diplostomum pseudospathaceum, Renicola lari und Metorchis xanthosomus sowie die drei Nematoden Desmidocercella incognita, Cosmocephalus obvelatus und Paracuaria tridentata nachgewiesen werden. Die parasitologischen Befunde des Silberreihers beschränkten sich auf Apharyngostrigea sp., Desmidocercella incognita und Desmidocercella numidica. Eine Beeinflussung der Helminthenfauna von Karpfen durch Kormoran und Reiher als auch umgekehrt konnte anhand der eigenen Untersuchungen in sächsischen Teichwirtschaften nicht sicher nachgewiesen werden<br>In Saxonian carp ponds the cestodes Neogryporhynchus cheilancristrotus and Paradilepis scolecina and the trematode Diplostomum pseudospathaceum occur amongst others in carps (KAPPE 2004, PIETROCK et al. 2006, KAPPE & SEIFERT unpublished). Further fish species (tench, roach, rainbow trout) are infected with the trematodes Diplostomum sp., Hysteromorpha triloba and Posthodiplostomum cuticola (PIETROCK et al. 2006). This work was performed to analyse the importance of fish eating birds as vectors of fish pathogenic helminths. For this purpose 97 common cormorants (Phalacrocorax carbo) and 46 grey herons (Ardea cinerea) were examined on their infestation with endohelminths during the term of June 2003 to December 2004. All these birds came from the vicinity of Saxonian fish ponds. In winter 2004 additionally an egret (Casmerodius albus) and an hering gull (Larus argentatus) were included into the investigations. Altogether 29 parasite species and genus respectively could be detected. Some of these endohelminths use fresh water fishes as intermediate or paratenic hosts. The cormorants harboured 15 different parasite species and genus respectively: the two cestodes Ligula intestinalis and Paradilepis scolecina, the six nematodes Baruscapillaria carbonis, Contracaecum rudolphii, Cyathostoma microspiculum, Desmidocercella incognita, Eucoleus contortus and Syncuaria squamata and seven trematodes the Clinostomum complanatum, Holostephanus dubinini, Hysteromorpha triloba, Metorchis xanthosomus, Paryphostomum radiatum, Petasiger exaeretus and P. phalacrocoracis. In addition the two acanthocephales Corynosoma strumosum and Polymorphus sp. were detected. In the grey heron nine helminth species/genus could be detected: the cestode Neogryporhynchus cheilancristrotus, the six trematodes Apharyngostrigea cornu, Bilharziella polonica, Diplostomum sp., Echinoasmus belocephalus, Posthodiplostomum cuticola and Tylodelphis excavata and the three nematodes Avioserpens galliardi, Desmidocercella incognita and Desmidocercella numidica. In the hering gull the cestode Echinocotyle verschureni, the four trematodes Apophallus muehlingi, Diplostomum pseudospathaceum, Renicola lari and Metorchis xanthosomus as well as the three nematodes Desmidocercella incognita, Cosmocephalus obvelatus and Paracuaria tridentata were detected. Merely three endohelminths could be demonstrated in the investigated egret: Apharyngostrigea sp., Desmidocercella incognita and D. numidica. Six of the 29 endohelminth species and genus respectively were found in this examination to infest both fish and birds in Saxonian fish ponds. Nevertheless the helminth fauna of the inestigated birds, in particular of cormorants and herons did not make a definite impact on the carps´ infestation with endohelminths. A colony of Great Egrets (Ardea alba) and Nankeen Night Herons (Nycticorax caledonicus) has existed at the Perth Zoo in Metropolitan Perth for over 25 years. The colony is particularly significant for the conservation and management of Great Egrets in Western Australia as it is the only colony located in the Metropolitan area. Baseline information of their breeding biology was needed to facilitate the development of management guidelines for the zoo colony. Foraging behaviour was used to highlight specific adaptations in hunting strategies and diet. However, it was not possible to observe foraging Nankeen Night Herons as they forage at night. Therefore, another species, the Little Egret, was selected to highlight specific adaptations. From 1997 to 1999 the reproductive biology of the Great Egret and Nankeen Night Heron was assessed. The number of Nankeen Night Herons nesting at the Perth Zoo from 1996 to 1998 increased, while the number of Great Egrets declined. Both species nested in tall trees but only Great Egrets were specific in their choice of nesting tree species. Horizontal nest placement appeared to be influenced by body size. Great Egrets had a larger clutch size than the Nankeen Night Heron, and a slightly higher offspring mortality rate. There was some indication that Great Egrets may use the colony as an information centre about productive feeding grounds. Foraging behaviour of Great Egrets and Little Egrets was recorded at six wetlands in the Perth Metropolitan area. Great Egrets were found to be mainly searchers using ‘stand and wait’ and 'walk slowly’ foraging behaviour, while the Little Egret was a ‘pursuer’ hunting by 'walking slowly', 'walking quickly' and 'pursuing prey'. Great Egrets caught a greater number of prey per attempt at capture, feeding on larger sized prey, mostly fish. Little Egrets fed on smaller sized prey, mostly invertebrates. Habitat type and wind speed had a significant effect on striking success of Great Egrets. Cloud cover, wind speed and direction had a significant effect on striking success of Little Egrets. The larger body size of the Great Egret allowed them to forage in deeper water than the Little Egret. Baseline information provided by this study has assisted in the development of management recommendations for the zoo colony and for Great Egrets and Little Egrets in the Perth Metropolitan area. To provide long-term information on overall population trends for the colony, regular counting and population distribution mapping of Great Egrets and Nankeen Night Herons is needed. To prevent Nankeen Night Heron numbers elevating and possibly encroaching on the nesting habitat of the Great Egret, food available in the zoo grounds should be reduced by covering caged animals food. Planting of nesting trees within the existing colony may be required to enable the number of Great Egrets nesting in the Perth Zoo to increase. To prevent disturbances to birds when foraging within Perth wetlands, sites that are reachable by humans should be fenced off, or access restricted.
