Full textLa epitranscriptomica es considerada una nueva rama de las ciencias biológicas que tiene como finalidad comprender la regulación de la expresión génica mediada por modificaciones químicas en el ARN. La N6-metiladenosina o m6A, es la modificación interna más abundante descrita en los ARNm. Los miembros de la familia de proteínas YTH han sido descritos como los principales encargados de reconocer la m6A presente en los ARNm y de ejecutar su función. Así, estas proteínas son conocidas como “lectoras” de m6A. En la familia de proteínas YTH existen 5 proteínas de las cuales 4 son citoplasmáticas, (YTHDF1, YTHDF2, YTHDF3 e YTHDC2) y una nuclear (YTHDC1). Mientras las proteínas citoplásmicas han sido asociadas con la regulación de la estabilidad, traducción o degradación de los ARNm que poseen m6A, el quinto miembro es nuclear y está involucrada con los procesos de corte y empalme alternativo y la exportación nuclear de ARNm metilados. Debido a que en los últimos años se ha descrito la presencia de m6A en ARNm virales, como es el caso de VIH-1, es que ha surgido el estudio de la epitranscriptómica viral, campo donde es necesario investigar las funciones de las proteínas involucradas. En este contexto, existen datos controversiales en donde se ha reportado que YTHDF1, YTHDF2 e YTHDF3 promueven la expresión génica de VIH-1 mediante el aumento de ARNm viral en las células infectadas e inhiben las etapas tempranas de la infección al inducir la degradación del genoma viral. Sin embargo, aún se desconoce si YTHDC1 o YTHDC2 regulan alguna etapa de la replicación de VIH-1. En este trabajo se planteó determinar la localización y proximidad entre las proteínas lectoras de m6A y el ARNm de VIH-1 a través de hibridación in situ acoplada al ensayo xiii de ligación proximal (ISH-PLA), de las cuales se obtuvo como resultado que YTHDC1 e YTHDF1 interactúan con el ARNm de VIH-1 en la periferia nuclear y región citoplasmática, respectivamente. Mientras que en el caso de YTHDF2 e YTHDF3, las interacciones observadas, no nos permitieron determinar la localización exacta de estas, mediante ISH-PLA. Una vez determinado esto, se escogió la proteína YTHDC1 como objeto de estudio. Se determinó el efecto de la sobreexpresión de esta lectora de m6A en la expresión génica viral analizando los niveles de la poliproteína Gag mediante Western blot y del ARN genómico mediante RT-qPCR. Aunque no se obtuvo consistencia en los resultados obtenidos, estos sugieren que YTHDC1 posiblemente promueve la exportación nuclear del ARN genómico de VIH-1.
The epitranscriptome is consider a new Branch of biology sciences that aims to understand the regulation of gene expression by chemical modifications on the RNA. The N6-methyladenosine or m6A, it is the most abundant inter modification described on the mRNA. The members of YTH proteins family have been described as the main responsible for recognizing m6A present on the mRNA and execute its function. So, these proteins are known as “readers” of m6A. In the YTH protein family exist 5 proteins of which 4 are cytoplasmic, (YTHDF1, YTHDF2, YTHDF3 e YTHDC2) and one nuclear (YTHDC1). Meanwhile cytoplasmic proteins have been associated with the stability regulation, translation or degradation of mRNA that m6A possesses, the fifth member is nuclear and is involved in the alternative splicing and the methylated mRNA nuclear export. xiv Because the presence of m6A on viral mRNA has been described in recent years, as in the case of HIV-1, the study of viral epitranscriptome has arisen, a field where it is necessary to investigate the functions of proteins involved. In this context, there are controversial data in where it has been reported that YTHDF1, YTHDF2 and YTHDF3 promote HIV-1 genetic expression by increasing viral mRNA in the infected cells and inhibit the early stage of infection by inducing the degradation of viral genome. However, it is still unknow if YTHDC1 or YTHDC2 regulate any stage of HIV-1 replication. In this work planned to determine the location and proximity among m6A reading proteins and HIV-1 mRNA by in situ hybridization coupled to the proximal ligation assay (ISH-PLA), from which YTHDC1 was obtained as a result and YTHDF1 interact with HIV-1 mRNA in the nuclear periphery and cytoplasmic region, respectively. While in the case of YTHDF2 and YTHDF3, the observed interactions did not allow us to determine the exact location of these, through ISH-PLA. Once it was determined, the YTHDC1 protein was chosen as the object of study. The effect of overexpression of this m6A reader in viral genetic expression was determined by analyzing the Gag polyprotein and genomic RNA by Western blot and RT-qPCR. Although no consistency was obtained in the results obtained, these suggest that YTHDC1 possibility promote the nuclear export of HIV-1 genomic RNA.

Full text-- Potato Leafroll Virus (PLRV) is responsible for severe losses in yield and quality of potato worldwide. There are native and wild potatoes with high levels of resistance to PLRV (Solanum brevidens, S. etuberosum, and S. chacoencse raphanifolium).The development of new varieties using these sources of resistance is one of the current challenges of genetic improvement crop, however the genetic nature of the potato in most cases is incompatible between varieties, moreover to obtain the desired resistance takes over 20 years. Therefore the direct insertion of a gene that confers resistance to a variety of commercial importance have a significant advantage. The present thesis evaluated ten events (Desiree variety) transformed with a hairpin-type construct, containing the RNA interference system (RNAi), it will keep active and constant the formation of dsRNA between the homology sequence of transgene and PLRV. The events were characterized by PCR and Southern blot; they had in their genome from one to two copies of the transgene. Serological tests of DAS-ELISA selected four events with high resistance (low or zero concentrations of PLRV) during primary, secondary and tertiary infection. Finally, it was determined by Northern Blot that the resistance to PLRV is related to the presence of fragments of small interference RNA (siRNA) formed from the mechanism of RNAi. -- KEY WORDS: Potato, transformation, PLRV, RNAi, siRNA.

Full text-- Proximal and lipidic analysis of lungs, heart, liver and kidney of adult, male guinea pigs: 10 from sea level ( Lima, 150 m. ) and 10 from altitude ( Huancayo, 3280 m. ) were perfomed. In the proximal analysis, moisture, ashes, proteins, lipids and carbohidrates content was determined, and in the lipidic analysis, phospholipides, cholesterol and triglycerides content in each organ was determined. The results were expressed in mg/g of moistured tissue. The corporal and organs weight were greater in the guinea pigs from altitude than those from sea level. In the lungs of guinea pigs from altitude the content of: moisture, lipides, phospholipides, cholesterol and triglycerides was smaller; and ashes and carbohidrates was greater. In the heart of guinea pigs from altitude the content of: ashes, lipides, phospholipides and triglycerides was smaller; and carbohidrates was greater. In the liver of guinea pigs from altitude the content of: moisture and proteins was smaller; and ashes, lipides, carbohidrates, phospholipides and cholesterol was greater. Full text--- With the purpose of standardizing a test for the determination of the sex in macaws we proceeded to extract of 25-50 ng. of genomic DNA from the blood of 31 birds previously sexed, belonging to 5 species of macaws (Ara ararauna, Ara chloropthera, Ara macao, Ara militaris and Propyrrhura couloni) coming from the Patronato del Parque de Las Leyendas Zoo and a private center, using an extraction kit of DNA (Wizard ® Promega). The used method was the technique of the PCR which amplified a fragment of the CHD gene of the female exclusive chromosome W (CHD-W), using the P2 (5´- TCTGCATCGCTAAATCCTTT - 3´) and P8 (5´- CTCCCAAGGATGAGRAAAYTG-3´ R same A / G, Y same T / C) primers, which are able to amplify not conserved regions (introns) of this gene. This differentiates it of its male homologous gene (CHD-Z). The amplification was carried by the optimization of the conditions and the thermal cycles. It was based on the conditions described by Griffiths et al. (1998). The obtained products were separated in agarosa gel to 3% (Promega) using a horizontal electrophoresis system (Hybaid) and visualized by fluorescence with etidio bromide through the ultraviolet light. Then we can see 2 fragments in female birds and only one in male birds, these fragments were between 300 - 400 bps. In this group 31(100 %) birds were sexing and we obtain 100% of compatibility between the conventional methods and DNA analysis. Finally, with the conditions previously described we sex 28 macaws with unkowned sex Key Words: CHD gene; P2 and P8 Primers; Macaws; Sex.

Full textThe zona pellucida is a extracellular matriz that surrounds vertebrate oocytes, and plays important roles in the recognition and interaction of gametes specie- specific, induction of Acrosome Reaction (AR) of the spermatozoa, block to polyspermy, keeps th integrity of the early embryo through its transicion by the oviduct; It is composed of three glycoproteins: ZP3 that induces RA; ZP1, structural, crosslink ZP3 and ZP2. ZP2 acts as a secondary sperm receptor that is necessary for the maintenance of sperm binding to the egg, its proteolytical modification after fertilization permits the block to polyspermy ZP2 participates also in the organization, development, maturation of the oocyte beacuse it keeps consistently the matrix and the interaction between peripherical cells with the germ cell. We isolated and analized in silico a partial coding secquence of the glycoprotein of type 2 (aZP2) in alpacas, we determined that this protein is express exclusively in the ovaries. Also this amalized partial secquence is conserved, constituting a monophyletic group between Cetarteodactyla. This thesis work provides basic knowledge on the glycoprotein a ZP2 in alpacas, a protein implicitly involved in fertilization, to know it benefits the improvement of existing reproductive biotechnology techniques such as the follicular-oocyte maturation, cryopreservation of gametes and embryos in vitro fertilization and injection of intracytoplasmic sperm, techniques that are trying to be implemented with many difficulties in camelids.

Full textIn vivo DNA molecules are narrowly folded within chromatin fibers and self-interacting chromatin domains. Therefore, intra-molecular DNA entanglements (knots) might occur via DNA strand passage activity of topoisomerase II. In this thesis, we found that small steady state fractions of DNA knots are common in intracellular chromatin. These knots occur irrespectively of DNA replication and cell proliferation, though their abundance is reduced during DNA transcription. We also found that in vivo DNA knotting probability does not scale proportionately with chromatin length: it reaches a value of 0.025 in domains of 20 nucleosomes, but tends to level off in longer chromatin fibers. We postulate that regulation of topoisomerase II activity and the architecture of chromatin might be crucial to prevent a potentially massive and harmful self entanglement of DNA molecules in vivo. Also, we show the effects of(-) and(+) supercoiling of DNA that naturally builds up in front and behind of DNA transcribing complexes, respectively. Whereas(-) supercoiling barely affects basal fractions of DNA knots, (+) supercoiling produces a burst of DNA knot formation and complexity. Using computing knot formation in chromatin models, we show that large compaction of the nucleosomal fiber reproduces the increase of knot probability and complexity generated by (+) supercoiling of intracellular DNA. These findings clarify why topo II is required for proper synthesis of long transcripts. Although topo I can relax supercoils, only topo II can remove the knots that arise ahead of transcribing complexes. Finally, in this thesis we have developed a high-resolution 2D-gel electrophoresis procedure that quantitatively discerns the fractions of positive- and negative-noded trefoil knots formed in vitro and in vivo systems. Results with minichromosomes in yeast discard a chiral and regular folding of nucleosomal fibers in vivo.

Full textThe present study consists of using nuclear tools aiming to establish an alternative procedure to perform biochemistry analyses in whole blood to help the diagnosis of diverses pathologies. The aim is to determine the íons and metals concentrations in whole blood of human beings (specifically: Br, Cl, K e Na), using neutron activation analysis, providing the limits of normality, as well as, the matrix of the correlation for these elements. To perform this study, 283 samples of whole blood had been analyzed (of healthy volunteers selected from blood banks), resulting in the limits of normality for Br (0.0067 - 0.0263 gl-1), Cl (2.54 - 3.50 gl-1), K (1.33 - 1.89 gl-1) and Na (1.48 - 2.06 gl-1). These data are the first estimates for reference values in whole blood of the Brazilian population. These limits were evaluated in function of the sex and age for checking the biological differences. The behavior of these limits was also evaluated for different populations, i.e., in two distinct regions: Southeast (blood collection carried out in São Paulo city) and Northeast (blood collection carried out in Recife city). These places were chosen in function of the similarities (cities with high concentration people and industrialized). Futhermorer, a systematic study of these limits was also evaluated, in the period of 4 (four) years, in São Paulo city. This analysis was elaborated in function of time due the necessity to update these data, therefore they act as monitorial ambient. The estimation for Ca and Fe were also proposal for a set of 22 samples of whole blood.

Full textTen microsatellites for alpacas and llamas were used to evaluate paternity in 47 alpacas registered at IVITA Research Station Maranganí, Canchis Province (Cusco – Perú). Analysis was carried out using both methodologies: Automatic Sequencer (ABI 377 DNA sequencersâ) and silver staining techniques. Microsatellites were amplified in three multiplex reactions and ten single PCR reactions. They were polymorphic for all alpaca samples using both methodologies. The number of alleles varied between 4 and 20, the allelic frequencies and the exclusion probability were calculated using Cervus 2.0. All loci, except for two, were within the range published by Lang et al. (1996) and Penedo et al. (1998). The accumulated exclusion probability for the ten loci was 0.9999. For each multiplex reaction the accumulated exclusion probability was more than 0.90. Both methodologies yielded the same results. It was possible obtain the same results using both methodologies. The results confirmed paternity in 17 cases of parent-offspring pairs, however in a further 22% of cases alternative adults were identified as parents compared with the register.

Full text--- The use of microsatellites, as molecular markers of human DNA, are important in the studies of affiliation and identification in forensic practices. These studies are based on allelic frequencies in certain populations. For example, in Peru the frequencies reported are often for Hispano-Americans. This research attempts to determine if significant differences among allelic frequencies of the microsatellite D16S539 exist between two populations: a Peruvian Ancash sample and a general sample of Hispano-Americans. Genetic populations studies were carried out in a sample of 33 norelated individuals using PCR followed by 8% polyacrilamide gel electrophoresis and silver staining. The population presented 5 alleles of the 9 found for the D16S539 microsatellite, with the respective frequencies: 9 (0.242), 10 (0.258), 11 (0.288), 12 (0.106) and 13 (0.106). The population in study was found to be in Hardy-Weinberg equilibrium with regard to this marker and there were no significant differences between the allelic frequencies of the population of Ancash origin, and general sample of Hispano-Americans.

Full textPeruvian western slopes of the Andes are an arid ecosystem rich in endemic flora currently threatened by industrial and urban expansion. It is characterized by a flora of cacti and deciduous shrubs. Orthopterygium huaucui (A. Gray) Hemsley is an endemic monotypic genus of Anacardiaceae family which inhabits this ecosystem as scattered thickets on rocky slopes along 500 km of the central coast and whose closest species belonging to the genus Amphipterygium (found in Mexico and Central America). Between those two genera there is a disjunction of more than 3500 km. In order to determine 1) the population structure of O. huaucui, 2) the phylogenetic position of O. huaucui within the tribe Rhoeae of the Anacardiaceae family and 3) the divergence time of O. huaucui within Anacardiaceae, we performed a Bayesian phylochronology calibrated with five fossil records and determined the genetic diversity. To get these aims we sequenced TrnL, Rps16 (plastid introns) and ITS (nuclear) markers broadly used in plant phylogenetic and phylogeographic studies. We extracted DNA from 51 individuals of O. huaucui, we amplified the three regions by PCR, we sequenced each one and edited manually. We downloaded sequences of other anacardiacean species available in GenBank. The disjunction between Orthopterygium and Amphipterygium resulted in 16,52 and 14,82 million years and a genetic distance between these two species was similar to lineages with greater diversification; whereas no intraspecific mutations were detected in Orthopterygium. This result would respond to a low dispersion or low mutation rate in view of ecological and molecular studies. In spite of the fact that the absence of mutations in nuclear markers is a common pattern in recent species and the Miocenic origin of Orthopterygium, we propose that this species was affected by a severe genetic bottleneck. Moreover, by considering as the single representative of the genus, Orthopterygium huaucui would be the result of an ancestral species migration from the northern hemisphere which has got the western slopes, an arid ecosystem for more than 15 million years, through a well-established Panama isthmus.

Full textThe State ability to store genetic information raises important challenges for criminal sciences, as DNA Databases exacerbate the tension among the principles of freedom, safety and equality. This tension is particularly strong when the States seek to preserve genetic identifiers of individuals that have been arrested by the police without any formal accusation or of those who have been acquitted of all penal responsibilities. This issue has been analyzed, inter alia, by the European Court of Human Rights and the Supreme Court of the United States, reaching, in both cases, different conclusions. In this Thesis, we have made a juridical analysis of the state conservation of genetic profiles for forensic purposes and it is pretended to define an equitable registration model that guarantees the fundamental rights of individuals.

Full textEl presente estudio, tiene por objetivo realizar una revisión y análisis del Sistema Nacional de Registros de ADN, creado por la Ley 19.970 del año 2004.Para el correcto tratamiento de nuestro tema de estudio, se ha dividido este trabajo en cuatro partes: En una primera, a modo de introducción, se hace una explicación somera del ADN y su función vital en la identificación de las personas. Una segunda parte se ha dedicado al estudio de la Historia Fidedigna de la Creación de la ley. En una Tercera, se realiza un análisis artículo por artículo de las disposiciones de la Ley 19.970. En una cuarta parte se realiza un análisis de constitucionalidad formal y de fondo de los preceptos de la Ley 19.970. Y por último, se realiza un alcance acerca del valor probatorio que revisten estas pruebas de ADN. Se realiza en un primer capítulo una breve descripción de que es lo que es el ADN, Capítulo que no pretende en forma alguna convertirse en un trabajo científico acerca de esta materia, sino que simplemente se limita a exponer, basados en bibliografía especializada, ciertos parámetros y conceptos que son necesario manejar para poder entender las palabras técnicas que utilizará luego, tanto la Ley 19.970, como su Reglamento

Full textTaenia solium es un céstodo de vida parásita, cosmopolita y tiene como hospedero definitivo al hombre. Estos helmintos aplanados son los responsables de infestaciones frecuentes en zonas endémicas en América Latina, Asia y África, así como en naciones desarrolladas con flujo masivo de inmigrantes provenientes de áreas endémicas. La teniosis se produce cuando el hombre consume las larvas (cisticercos) de Taenia solium, mientras que la cisticercosis es causada por el consumo de los huevos. Estos últimos se desarrollan hasta cisticerco en diferentes tejidos, principalmente en el sistema nervioso central, causando la neurocisticercosis, la que se caracteriza por lesiones y diferencias en la respuesta inmunológica del huésped frente al parásito. Las manifestaciones clínicas más frecuentes son epilepsia, signos neurológicos de focalización, hipertensión endocraneal y deterioro cognitivo. En estudios realizados a partir de fluido de cisticerco se reportaron proteínas antigénicas importantes como la cisteín proteasa homóloga a catepsina L, la cual interviene en la invasividad del cisticerco, desde el tracto digestivo del hospedero hasta el músculo y sistema nervioso, a través del torrente sanguíneo. En el presente trabajo, se ha identificado el gen de una proteína similar a catepsina L de cisticerco a partir del genoma de T. solium, secuenciado en el laboratorio de Bioinformática y Biología Molecular (UPCH). La región codificante madura de la secuencia homóloga a Catepsina L fue de 633 nt, esta secuencia fue clonada y expresada en el vector pET28a; la proteína expresada de 22.3 kDa se encontró en el sedimento del lisado celular de Escherichia coli BL21 (DE3), sistema procariótico de expresión, lo cual indica que fue insoluble. Con el propósito de obtener la proteína soluble, se estandarizó la temperatura de crecimiento bacteriano, el tiempo de inducción y la concentración de IPTG, pero no fue posible lograrlo, posiblemente por las glicosilaciones postraduccionales que presenta esta proteína, y que que las bacterias no pueden realizar.

La Ley N.° 19.970 establece el sistema nacional de Registros de Huellas de ADN de interés criminal. A través de la presente tesis de grado buscamos analizar las condiciones de legitimidad del tratamiento de las muestras biológicas y los resultados de los análisis que se realicen sobre ellas. Para ello nos serviremos del método científico aplicable a las investigaciones jurídicas, y en consecuencia descompondremos el problema del tratamiento de datos de huellas de ADN, identificando y estableciendo cuáles son las deficiencias y faltas de claridad existentes en nuestra normativa examinada a la luz de la doctrina y experiencia internacional y propondremos algunas soluciones. Los métodos de investigación que se utilizarán, entendiendo por tales aquellos que se refieren a los procedimientos que se usan con el propósito de llegar a demostrar una hipótesis, cumplir con los objetivos de un proyecto y dar una respuesta concreta al problema que se identificó, son de distintos tipos o especies, dentro de ellos el inductivo y el documental. Será inductivo, pues a partir de los fenómenos particulares, cuales son los registros de ADN previstos en la Ley N.° 19.970 se arribará a las conclusiones y premisas que servirán de base para demostrar la hipótesis, y será documental, pues se efectuará un examen de las fuentes formales del Derecho, como son la doctrina nacional y extranjera, las normas legales internas y de Derecho comparado, además de las resoluciones judiciales y administrativas que digan relación con la materia que se investiga. De acuerdo al núcleo central de la hipótesis de trabajo, sostenemos que la Ley N.° 19.970, Crea el Sistema Nacional de Registros de ADN, contraviene los principios y normativas de protección de datos, por lo que muchas de las actuaciones procesales establecidas para el Ministerio Público y los Tribunales adolecen al menos de falta de legitimidad, lo que pone en riesgo el sistema de derechos fundamentales y por ello es necesario esclarecer las condiciones de legitimidad en el uso de bases de datos de perfiles de ADN con fines de investigación criminal. Para los efectos de desarrollar la investigación, realizamos un análisis general, desde los principios básicos de la biología, el concepto de ADN y de huellas genéticas o huellas de ADN y la tecnología asociada, así como su habilidad a los efectos de determinar la identidad de una persona. Contrastada la información que hemos analizado con la Historia del Establecimiento de la Ley N.° 19.970 encontramos un alto grado de coincidencia en cuanto a la eficacia de la huella de ADN para estos efectos, con porcentajes de asertividad que superan el 99,6% de acuerdo a los distintos estudios realizados. A continuación efectuamos un examen de la huella de ADN y la muestra biológica a partir de la cual se determina a la luz de la legislación sobre protección de datos personales, en concreto la Ley N.° 19.628 de protección de la vida privada frente al tratamiento automatizado de datos personales y las normas de derecho comparado que se han pronunciado y que nos resultan relevantes por su cercanía a nuestra cultura jurídica. La principal conclusión que hemos obtenido a este respecto es que tanto las muestras biológicas como las huellas de ADN deben ser consideradas y tratadas como datos sensibles y por tanto estar sujetas a altos estándares de seguridad. A estos efectos comprobamos que en la historia de la ley como en el texto que en definitiva se aprobó se recoge esta idea, estableciéndose diversos resguardos de seguridad tanto en la obtención, como conservación, registro y comunicación de esta información. Una clara manifestación de esto es la necesidad de acreditación de los laboratorios que participan del análisis de las huellas de ADN. En un tercer enfoque de estudio, nos ha interesado revisar la Ley N.° 19.970 desde la óptica de la historia de su establecimiento, elemento que hemos considerado esencial a los efectos de la posterior aplicación de la misma. Especialmente, nos ha interesado desvelar la intención legislativa a la hora de establecer los distintos tipos de registros y las condiciones para que la información de una persona sea ingresada a los mismos y las condiciones de legitimidad del uso de esta información por los distintos agentes del Estado. No hemos realizado un análisis crítico de los tipos de delitos que se han incluido en el registro, pues consideramos que se aleja del objeto de nuestro estudio. De igual forma, no hemos efectuado un análisis sobre la naturaleza jurídica del registro, en cuanto a las finalidades de política criminal que pudieran estar asociadas, pues ello también dista del objeto de nuestro estudio. Ahora bien, para ilustrar como está funcionando el sistema, hemos detectado una sentencia que se refiere a la materia, relativa a la pertinencia de ingresar en el registro los datos de un adolescente infractor, que resulta interesante por la consideración general de que los datos de menores de 18 años deben ser tratados como datos sensibles. Finalmente, hemos realizado un estudio de la Ley N.° 19.970 a la luz de los principios del debido proceso, que se recogen a nivel internacional y nacional. Luego de analizar la legislación vigente, hemos arribado a que la ley adopta criterios adecuados respecto del resguardo de estos principios en los procedimientos de obtención, conservación y análisis de las muestras, sin embargo no queda claro que se dé plena satisfacción a los mismos en las normas relativas a los tipos de registros, las atribuciones de las policías y del Ministerio Público, a la hora de decidir sobre la toma de muestras y análisis. Creemos que esta materia debe ser seguida con atención en su experiencia práctica pues puede terminar afectando gravemente los principios de inocencia, derecho a la defensa, in dubio pro reo, etc. Asimismo, en la lista de delitos que habilitan la recogida y tratamiento de muestras, advertimos que la legislación ha adoptado un criterio expansionista, llegando incluso, en algunos casos, a prever la toma y análisis de muestras para delitos en los que estas pruebas no tienen relevancia, tales como el delito de violencia psicológica y las amenazas, a vía ejemplar. En consecuencia, concluimos que no sólo la novedad de la investigación, sino su pertinencia queda demostrada por cuanto las pruebas de ADN van adquiriendo cada vez mayor importancia, en todos los ámbitos de interés criminal

Full textAmong the great variety of species from the Peruvian Amazon forest are the land snails of the family Megalobulimidae (Mollusca, Gastropoda), among them are Megalobulimus popelairianus, Megalobulimus huascari y Megalobulimus capillaceus, that have economic importance for both the nutritional properties of their meat and cosmetic properties of their slime. The main goal of this research was to develop DNA profiles of the Megalobulimidae land snails from San Martin, to allow their recognition at specific level, and to study their population genetic health through the genetic diversity, in order to guarantee their sustainable use and eventually to certify these species in Peruvian biotrade. The biological samples were collected in September of 2007 and January, February and March of 2008. The total DNA extraction was carried out by a CTAB protocol, with chloroform-isoamílic alcohol, from foot muscle tissue of 65 snails. Fragments of the genes rRNA and Cytochrome C oxidase subunit I (COI) of the mitochondrial genome were amplified and sequenced, using internal and universal primers, respectively. DNA sequences were aligned, characterized, determined the data quality, genetic distances, genetic diversity; the phylogenetic reconstruction was performed with NJ, MP, ML and BI; DNA profiles were analyzed. COI amplification rate was lower than the rRNA one. Surprisingly, the 31 DNA sequences of M. capillaceus did not show any genetic variant. Low values of genetic divergence (lower than 2%) between M. huascari and Megalobulimus spp. in both genes were found, besides, they were in a monophyletic group, with robust statistical support for the COI marker. 16S rRNA phylogenies were inconclusive and varied according to the methodologies. The combined data showed unresolved phylograms. M. capillaceus did not show genetic diversity, probably as a result of over hunting, which could threaten its survival as a species.

Full textEl objetivo de este trabajo fue identificar genes expresados diferencialmente en un organismo cuyo genoma no ha sido secuenciado. Investigamos los patrones globales de expresión en respuesta a cambios en la concentración de Mn2+ en el medio de cultivo, con el fin de identificar genes involucrados en el metabolismo y homeostasis de este catión. El organismo modelo usado fue Ceriporiopsis subvermispora, un hongo ligninolítico altamente selectivo, el cual posee una maquinaria enzimática compuesta por peroxidasas dependientes de manganeso y lacasas principalmente. Para lograr el objetivo propuesto desarrollamos un protocolo de cDNA-AFLP (cDNA- Amplified Fragment Length Polymorphism) basado en la amplificación de fragmentos derivados de transcritos, para analizar los patrones de expresión génica en cultivos con diferente concentración de Mn2+. Al analizar los patrones generados obtenidos del hongo creciendo en diferentes concentraciones de manganeso, identificamos 34 fragmentos derivados de transcritos diferencialmente expresados en respuesta a manganeso. Veintidós de ellos mostraron homología con genes de función conocida o putativa, mientras que catorce de ellos no mostraron coincidencias considerables. El resultado más interesante fue identificar dos fragmentos derivados de transcritos que correspondían a la familia de transportadores tipo ABC (ATP Binding Cassette) altamente relacionados con la homeostasis de Mn2+ y Ca2+
The aim of this work was to identify differentially expressed genes in an organism whose genome has not been sequenced. We investigated the global gene expression pattern regulated by manganese, in order to identify genes involved in its metabolism and homeostasis. The model organism used was Ceriporiopsis subvermispora, a highly selective ligninolytic fungus, which possesses an enzymatic machinery composed of manganesedependent peroxidases and laccase. In order to achieve this objective we developed a cDNA-AFLP (cDNA- Amplified Fragment Length Polymorphism) protocol based on the amplification of transcript derived fragments (TDFs) to analyze the gene expression patterns on different Mn2+ concentration cultures. When analyzing cDNA-AFLP generated patterns obtained from the fungus grown on different concentrations of manganese, we identified 34 manganesedifferentially expressed transcript derived fragments. Twenty of them showed significant homology to genes with known or putative function, while fourteen fragments did not show significant matches. Interestingly we identified two TDFs that correspond to members of the ABC (ATP Binding Cassette) transporter family closely related with Mn2+/Ca2+homeostasis

Full textLa enfermedad neurológica conocida como Paraparesia Espástica Tropical, mielopatía asociada al HTLV-I (TSP/HAM) se caracteriza por la degeneración axonal de los haces córtico-espinales. Aunque no se conoce mucho como el HTLV-I afecta selectivamente a los axones más largos del sistema nervioso central (SNC), es la proteína viral Tax secretada desde los linfocitos T-CD4+ el principal candidato por su actividad estimuladora de factores transcripcionales a través de varias vías de señalización. Esta proteína, se encuentra presente en forma crónica en el líquido cefalorraquídeo (LCR), pudiendo alterar el transporte axonal actuando extracelularmente, siendo afectados principalmente los axones más largos por su mayor vulnerabilidad. Estudios in vitro de la proteína Tax muestran que se une a varias proteínas celulares que regulan vías relacionadas con la transcripción y el citoesqueleto, incluyendo fosfatasas y quinasas, por lo que se sugiere que en esta patogenia podría haber un desbalance en el nivel de fosforilación/desfosforilación de las proteínas del citoesqueleto. Se han propuesto como biomarcadores de varias enfermedades neurodegenerativas los niveles de Tau total y/o fosfo-Tau en el LCR. Por lo tanto, los objetivos de esta investigación fueron comparar la fosforilación de Tau presente en el LCR de pacientes con TSP/HAM y sujetos controles, a fin de aclarar si estas proteínas están involucradas en la patogenia de esta enfermedad, y aislar la proteína Tau desde el LCR para posteriores estudios de proteómica de sus fosfoformas. Los niveles de Tau en el LCR y la evaluación del patrón molecular de fosforilación de residuos específicos como Tre181, Ser199, Tre205, Tre231, Ser262, Ser356, Ser396, Ser404 y Ser422 fue realizada por “immunowestern blot” utilizando anticuerpos policlonales monoespecíficos. Con todos los anticuerpos ensayados se observó la presencia de una banda principal de 52 kDa. Los niveles de Tau en el LCR de los TSP/HAM no fueron significativamente distintos de los de controles, y aunque no fue significativamente diferente (p = 0,06), se podría sugerir sólo una anormal hiperfosforilación de Tre181. Si se confirmara este mayor nivel fosforilación en Tre181 trabajando con un número mayor de pacientes o utilizando otros métodos más confiables, sugeriría un aumento en la actividad o una sobre-expresión de dos quinasas dirigidas a Ser/Tre-Pro, GSK3-α y β, Cdk5. Una caracterización completa de la fosforilación de los distintos residuos de Tau en el LCR de los controles y pacientes con TSP/HAM requirió la aplicación adicional de la técnica analítica de Espectrometría de Masas (MS). Por lo tanto, se trató de aislar Tau desde LCR utilizando una columna de afinidad por fosfopéptidos de Ga(III), y dos métodos de inmunoprecipitación, uno usando proteínas A/G acopladas a agarosa y una segunda con unión covalente de los anticuerpos (“Sepharose” activada con CNBr). Ni la columna de Ga(III) ni el primer método de inmunoprecipitación fueron adecuados para posterior análisis por MS. Después de demostrar por SDS/PAGE que el método que usaba los anticuerpos unidos covalentemente mostraba una banda principal de 52 kDa, se eluyó la proteína del gel, tripsinizó y analizó por MS MALDI-TOF. Se observó la posible presencia de proteínas asociadas al citoesqueleto que podrían haber coinmunoprecipitados con Tau. Estos resultados no fueron confirmatorios por el bajo “score” observado. En conclusión, dos de los resultados muestran diferencias entre TSP/HAM y varias enfermedades neurodegenerativas: el no aumento en los niveles de Tau total, y la posible hiperfosforilación de sólo Tre181. Fue importante lograr aislar Tau desde el LCR para futuros estudios de MS
The neurological disorder known as tropical spastic paraparesis / HTLV-I-associated myelopathy (TSP/HAM) is characterized by axonal degeneration of the cortico-spinal tracts. Although the selectivity of HTLV-I towards the longest axons of the central nervous system (CNS) is not fully understood, the main candidate is the secreted viral protein Tax from lymphocytes T-CD4+ that shows a stimulatory activity of transcriptional factors acting through several signaling pathways is the main candidate. This protein, chronically present in CSF, could extracellularly alter axonal transport. The longest axons are mainly affected because they are more vulnerable. In vitro studies on Tax protein have shown the binding of this protein to many cellular proteins that regulate transcription and cytoskeleton related pathways including phosphatases and kinases, therefore an imbalance in the level of cytoskeleton phosphorylated/unphosphorylated proteins could be involved in this pathogeny. Levels of total Tau and/or phospho-Tau in the CSF have been proposed as biomarkers of various neurodegenerative diseases. Hence, the aims of this investigation were to compare Tau phosphorylation present in CSF of TSP/HAM patients and control subjects, to elucidate if these proteins are involved in the pathogeny of this disease, and to isolate Tau protein from CSF for further proteomic studies of its phosphoforms. Levels of Tau in CSF and evaluation of the molecular pattern of phosphorylation of selected residues such as Thr181, Ser199, Thr205, Thr231, Ser262, Ser356, Ser396, Ser404 and Ser422 were performed by immunowestern blot, using monospecific polyclonal antibodies. With all the antibodies tested we observed a main band with 52 kDa. The CSF levels of Tau in TSP/HAM were not significantly different from those of controls. Only an abnormal hyperphosphorylation on Thr181, although it was not significantly different (p = 0.06), could be suggested in TSP/HAM patients. If a high phosphorylation level in Thr181 is confirmed by working with a larger number of patients or other more reliable methods, it would suggest an increase in activity or over-expression of two Ser/Thr-pro-directed kinases GSK3-α and β, and Cdk5. A complete characterization of Tau phosphorylation sites in CSF from control and TSP/HAM patients required the use of the additional analytical technique of Mass Spectrometry (MS). Therefore, we tried to isolate Tau from CSF samples using the phosphopeptide affinity column with Ga(III), and two immunoprecipitation methods, one using protein A/G-coupled agarose beads and a second one with covalently bound antibodies (Sepharose activated with CNBr). Neither the Ga(III) column nor the first immunoprecipitation method were adequated for MS analysis. After demonstrating by SDS/PAGE that the method that used covalently bound antibodies showed a main band of 52 kDa, the sample was eluted, trypsinized and analyzed in a Mass Spectrometer MALDITOF. This preliminary study suggested the presence of Tau and of some other cytoskeleton associated proteins that could have been coimmunoprecipited with Tau. The low “score” obtained did not allow reach confirmatory results. In conclusions, two of the results showed differences with various neurodegenerative diseases: lack of changes in Tau levels in CSF, and possible hyperphosphorylation of only threonine 181. It was important to have been able to isolate a Tau form from CSF for further studies of MS

Full textIn the present study Neutron Activation Analysis technique has been used to determine, simultaneously, some element concentrations of clinical relevance in whole blood samples of Golden Hamster. The normal range for Br, Ca, Cl K, Mg, Na and S considering 2 (Two Standard deviations) was 0.011 0.047 gL-1 (Br); 0.11 0.35 gL-1 (Ca); 2.11 3.75 gL-1 (Cl); 1.35 2.79 gL-1 (K), 0.026 0.090 gL-1 (Mg), 1.03 2.51 gL-1 (Na) e 0.97 2.01 gL-1 (S). The knowledge of these limits became possible to perform clinical investigation in this animal model using whole blood. The comparison with the results from human being whole blood estimation (Hamster & human) became possible to check the similarities or physiologic differences, an important data for animal experimentation .

Full text[CAT] Els gràfics de control per atributs són ferramentes estadístiques àmpliament utilitzades tant en la indústria de béns com en la de serveis i serveixen per a monitoritzar processos per mitjà d'atributs de qualitat. Els gràfics per atributs uni- variants estan entre els més coneguts i són el tema central d'esta tesi. Existeixen dos tipus: els gràfics basats en la distribució binomial (Gràfics np i p) i els gràfics basats en la distribució de Poisson (Gràfics c i u). Els gràfics per atributs Shewhart són sens dubte els més populars i tenen la particularitat que els seus límits de control es basen en l'aproximació normal a les distribucions binomial i de Poisson. Quan s'utilitzen estos gràfics allò més habitual és assumir que, sempre que l'aproximació normal siga adequada, la seua capacitat de monitorització serà idònia, és a dir que podran detectar de la mateixa manera tant millores com deterioraments del procés. En aquesta tesi es demostra que, a causa de la asimetria de les distribucions binomial i de Poisson, l'ajust de l'aproximació normal és imprecís en les cues d'eixes distribucions i que això afecta negativament la potència de detecció dels gràfics Shewhart. Per a poder establir la magnitud de l'afectació es van desenrotllar diversos paràmetres nous que servixen per a avaluar i caracteritzar la capacitat de monitorització de qualsevol gràfic per atributs del tipus univariant. A través d'ells es va establir que els gràfics Shewhart, al contrari del que es presumeix, estan lluny de ser idonis. Els nous paràmetres mencionats en el paràgraf anterior també van servir per a analitzar gràfics de control plantejats com a alternatives superiors als Shewhart. Els resultats de les anàlisis van demostrar que tampoc tenen una capacitat de monitorització del tot satisfactòria. Dos nous gràfics de control, el p Kmod i el u Kmod, són proposats. Estos gràfics tenen una capacitat de monitorització superior a qualsevol altre gràfic de control (p i u respectivament) inclòs en esta tesi, a més de comptar amb un mètode de fàcil ús, per mitjà del qual és possible establir si eixa capacitat és òptima o no. Els resultats de la investigació han sigut publicats en actes de congressos i en revistes científiques internacionals.
[EN] Attribute control charts are statistical tools that are widely used in the goods and services industries, they serve to monitor processes by means of product quality attributes. The uni-variant attribute charts are amongst the most well-known and are the main topic of this thesis. Two types of such charts exist, namely: the ones based on the binomial distribution (np and p Charts) and the ones based on the Poisson distribution (c and u Charts). The Shewhart attribute charts are without doubt the most popular and have the peculiarity that their control limits are based on the normal approximation to the binomial and Poisson distributions. When these charts are used it is commonly assumed that, as long as the normal approximation is adequate, their monitoring capability will be ideal, or in other words, that they will be able to detect with equal capacity, either process improvements or deteriorations. In this thesis we show that due to asymmetry of the binomial and Poisson distributions, the adjustment of the normal approximation is inaccurate on their tail sides and that this affects on a detrimental way the detection power of the Shewhart charts. In order to be able to establish the magnitude of the affectation, various novel parameters that serve to assess and characterised the monitoring capability of any uni-attribute type chart were developed. Through them it was established that the Shewhart charts, contrary to what is commonly assumed, are far from being ideal. The aforementioned novel parameters, also served to analyse other control charts posed as superior alternatives to the Shewhart¿s. The analysis results demonstrated that those charts, although superior to the Shewhart¿s, also have a far from satisfactory monitoring capability. Two new control charts, the p Kmod and the u Kmod, are proposed. These charts have a superior monitoring capability compared to any other chart (p and u respectively) included in this thesis, in addition they have an easy to use method that makes it possible to establish if their monitoring capability is, or is not, ideal. The results of the research have been published in congress proceedings and international scientific journals.

Full textEmplea los marcadores moleculares rbcL y psbA en un amplio estudio florístico de algas rojas de la costa del Perú para investigar la taxonomía y evaluar las relaciones filogenéticas de 20 especies. Un total de 51 secuencias rbcL y 4 secuencias psbA fueron generadas, de las cuales 32 fueron incluidas por primera vez en un análisis molecular. Los resultados revelaron 21 especies agrupadas en 11 familias y la presencia de cuatro nuevos reportes para el Perú: Corallina caespitosa, Nothogenia chilensis, Porphyra mumfordii y Schizymenia dubyi. Adicionalmente, el análisis también reveló cinco taxones Haraldiophyllum sp., Hypnea sp., Phymatolithon sp., Pyropia sp.1 y Pyropia sp.2 que podrían corresponder a nuevas especies por lo que posteriores colectas y análisis morfológicos y moleculares son requeridos para develar su taxonomía. Los resultados obtenidos permiten actualizar la lista taxonómica de especies y resalta el uso del secuenciamiento de marcadores moleculares para establecer perfiles básicos de diversidad molecular y evaluar de manera objetiva las relaciones evolutivas de las algas marinas en el Perú.

Full textIn this work the determination of chemistry elements in biological tissues (whole blood, bones and organs) of dystrophic mice, used as animal model of Duchenne Muscular Dystrophy (DMD), was performed using analytical nuclear technique. The aim of this work was to determine reference values of elements of clinical (Ca, Cl, K, Mg, Na) and nutritional (Br and S) relevance in whole blood, tibia, quadriceps and hearts from Dmdmdx/J (10 males and 10 females) dystrophic mice and C57BL/6J (10 males) control group mice, using Neutron Activation Analysis technique (NAA). To show in more details the alterations that this disease may cause in these biological tissues, correlations matrixes of the DMDmdx/J mouse strain were generated and compared with C57BL/6J control group. For this study 119 samples of biological tissue were irradiated in the IEA-R1 nuclear reactor at IPEN (São Paulo, Brazil). The concentrations of these elements in biological tissues of Dmdmdx/J and C57B/6J mice are the first indicative interval for reference values. Moreover, the alteration in some correlation coefficients data among the elements in the health status and in the diseased status indicates a connection between these elements in whole blood, tibia, quadriceps and heart. These results may help the researchers to evaluate the efficiency of new treatments and to compare the advantages of different treatment approaches before performing tests in patients with muscular dystrophy.

Ditassa R. Br. é um gênero neotropical composto por 100 espécies de lianas volúveis, distribuídas por toda a América Latina, exceto o Chile, e excepcionalmente no México. São 60 espécies brasileiras, 49 destas endêmicas. Ditassa burchellii Hook. & Arn. é restrita ao Distrito Federal e aos Estados do Sul e Sudeste. Na ocasião da sua floração, é facilmente localizada devido ao forte odor de urina liberado pelas suas pequenas flores, com cerca de 3 mm de diâmetro. Foi objetivo analisar as estratégias reprodutivas sexuadas dessa espécie, no maior remanescente de Floresta Atlântica do município de Viçosa, sudeste brasileiro. Para tanto, sete indivíduos foram acompanhados no período entre março de 2013 e dezembro de 2014. Neles, foi registrada a fenologia reprodutiva e, adicionalmente, foi verificada a sazonalidade das fenofases e sua correlação com as variáveis ambientais. A morfologia floral e o mecanismo de polinização foram analisados com auxílio da microscopia de luz e microscopia eletrônica de varredura, através de procedimentos usuais. Para verificar a atividade dos visitantes florais, foi quantificada a remoção de polinários em 450 flores senescentes. Os visitantes florais foram identificados e a frequência de visitação foi realizada no pico da floração. A composição específica dos voláteis florais foi quantificada utilizando Cromatografia Gasosa acoplada à Espectrometria de Massas. A floração e frutificação ocorreram, respectivamente, na transição da estação chuvosa para seca e na estação seca, com sobreposição entre elas e picos distintos e sequenciais. Os ciclos fenológicos foram ajustados à sazonalidade climática, influenciados principalmente pela precipitação, temperatura e comprimento do dia. A morfologia floral é semelhante à morfologia de outras espécies de Asclepiadoideae. Entretanto, a presença de tecidos secretores nos segmentos estaminais da corona é inédita. O néctar é o recurso procurado pelos visitantes florais e acumula-se em cavidades nectaríferas, localizadas abaixo das fendas anterais. Ao acessar o recurso, o aparelho bucal dos polinizadores é capturado pela fenda, que a percorre de baixo para cima, resultando em remoção ou inserção de polínias. Este mecanismo de polinização também foi observado em outras Asclepiadoideae. As flores apresentaram a tendência de um ou dois polinários removidos. Foram observados 53 espécies de visitantes florais, distribuídas entre as ordens Hymenoptera (23 espécies) e Diptera (30 espécies). Apesar dessa diversidade, D. burchellii foi caracterizada como sapromiiófila. Os principais polinizadores foram as micro-moscas (cerca de 4-5 mm) da família Milichiidae, que realizaram 23,49% das visitas. Os compostos mais abundantes encontrados nos voláteis florais foram (E)- -ocimeno, (E)- cariofileno, α-copaeno, tiglato de (Z)-hex-3-enila e p-1,3,8-mentatrieno, que provavelmente são os responsáveis pela atratividade dos insetos visitantes.
The neotropical genus Ditassa R. Br. consists of about 100 species of lianas, distributed in Latin America, except Chile, and exceptionally in Mexico. There are 60 Brazilian species, of which 49 are endemic. Ditassa burchellii Hook. & Arn. is restricted to the Distrito Federal and the states of the South and Southeast. On occasion of flowering, it is easily located because of the strong odor of urine released by its small flowers, with about 3 mm diameter. Our goal were to analyse the reproductive strategies of this specie, in the largest fragment of Atlantic Forest located in the municipality of Viçosa, southeastern Brazil. For this purpose, seven individuous were monitored from March/2013 to December/2014. The reproductive phenology was recorded in these individuous and, additionally, were verified seasonality of the fenofases and their correlation with environmental variables. The floral morphology and pollination mechanism were analysed with the aid of studies in light microscopy and scanning electron microscopy. For that, we applied the usual methods. Removal of pollinaria was measured at 450 senescent flowers to check the activities of floral visitors. The floral visitors were identified and the visitation frequency was assessed during flowering peak. The specific composition of floral volatiles was quantified using gas chromatography coupled to mass spectrometry. The flowering and fruiting occurred respectively in the transition from the rainy to dry and dry seasons, with overlap between them and distinct and sequential peaks. The phenological cycles were adjusted to climate seasonality, influenced mainly by rainfall, temperature and daylength. The flower morphology resembles the morphology of other species of Asclepiadoideae. However, the presence of secretory tissues in staminal corona is unprecedented. Nectar is the resource used by floral visitors and it accumulates inside the nectar cavity, located below the anther slit. To access the resource, the pollinator mouthparts is “caught” by the anther slit, that runs upwards, resulting in removal or insertion of pollinia. This pollination mechanism was also observed in other species of Asclepiadoideae. The flowers showed one or two pollinia removed per flower. The floral visitors of D. burchellii were 53 insects species, distributed among the orders Hymenoptera (23 species) and Diptera (30 species). Despite this diversity, D. burchellii was characterized as sapromyiophilous. The main pollinators were small flies from Milichiidae family (about 4-5 mm), accounted for 23,49% of the visits. The most abundant compounds found in the floral volatiles were (E)- -ocimene, (E)- caryophyllene, α-copaene, ethyl tiglate e p-1,3,8-menthatriene, which probably are responsible for the attractiveness of visiting insects.

Full textAspidosperma polyneuron Muell. Arg. and Aspidosperma cylindrocarpon Muell. Arg. belong to Apocynaceae and naturally occur in São Paulo State (Brazil). Both species were chemically investigated and in vitro cultures established. As there were no previous studies concerning the alkaloid composition from the leaves, our studies were focused on this plant part. Moreover, the volatile oil composition was determined for both species. As several biological activities have been described for indole alkaloids, the antimicrobial, antitumor and antimalarial activities were assayed for the total alkaloids. The leaf alkaloids were extracted by acid/base partitioning with organic solvents and isolated by adsorption chromatography techniques. The isolated alkaloids were analysed by GC-MS. A. polyneuron leaves afforded 5 aspidospermatane alkaloids: aspidospermine, demethoxy-aspidospermine, demethylaspidospermine cylindrocarine and pyrifolidine. In A. cylindrocarpon leaves different indole alkaloid skeletons were identified, 4 aspidospermatanes (aspidospermine, N-benzoyl-cylindrocarine, N-benzoyl-20-hydroxy-cylindrocarine, N-cynamoyl-20-hydroxy-cylindrocarine), 1 eburnane (16-epi-vincamine) and 1 heteroyohimbane (tetrahydroalstonine). The volatile oils were extracted from the leaves by steam distillation. A. polyneuron oil consisted of one single major compound, the diterpene kaurene (73,7%), and several long chain aldehydes. On the other hand, from the A. cylindrocarpon oil 25 constituents could be identified corresponding to 96.1 % of the crude oil. The major components were sesquiterpene hydrocarbons (germacreno-D, β-caryophyllene), oxygenated sesquiterpenes (spathulenol, epi-globulol, globulol), a monoterpene hydrocarbon (α-pinene), an oxygenated monoterpene (linalool) and also several long chain aldehydes. For A. polyneuron, callus cultures were induced, while for A. cylindrocarpon was also possible the establishment of cell suspension cultures. A. cylindrocarpon cell suspension cultures showed a lag phase of growth for approximately 4 days. The lag phase was followed by a growth phase for 15 days. After 20 days of culture the cells showed a stationary growth. Alkaloid extract from both species showed a antifungal activity in the bioautography assay with Clasdosporium cladosporiodes and C. sphaerospermum. The antitumor activity was evaluated with mutant Saccharomyces cerevisiae strains deficient in the DNA repair system (RS321 and Rad 52Y). In this assay, only the leaf alkaloids of A. cylindrocarpon presented activity. In vitro antimalarial assay were performed with Plasmodium falciparum chloroquine sensitive (K1) and resistant (Palo Alto). As such an activity was previously reported for A. polyneuron, only A. cylindrocarpon alkaloid extracts from leaves and stems were tested. Both alkaloid extracts showed strong inhibition of the parasite development. The IC50 for the K1 isolate were 9.0 µg/ml for the leaves and 8,0 µg/ml for the stems. The Palo Alto isolated presented a higher IC50, 13,3 µg/ml stems and 10,5 µg/ml.

Full textThis dissertation implements a monitoring system that can be used in three different contexts: monitoring a single student, monitoring a classroom or monitoring a group of people. In order to build this system, we based the development on the use of algorithms for face detection, face recognition and facial features extraction. During this work it was also implemented an eye tracker, a face tracker, an head estimation pose and emotion detection. For each context, different approaches were developed. For the single user monitoring, there was the need of recognizing the face. For this context, the most convenient algorithm was based on Local Binary Patterns Histograms. After a successful recognition, the system assigns an ID to the face and starts tracking it while retrieving useful data from the facial features. For the classroom monitoring, there was no need of recognition, only face tracking. For each face detected, an ID is assigned and the face tracker starts. For the monitoring of a group of people, there was the need of making a face recognition each time a new face appears in the frame and, after a successful recognition, the face tracker starts. The main contributions of this thesis are an automatic calibration for the digital camera used in the system for a better face recognition, a modular solution separated in three components that can be used to monitor three different contexts, retrieving relevant information during a certain period of time that an individual was in front of the camera. The developed software was integrated in a graphical user interface software provided by the camera manufacturer.
Esta dissertação implementa um sistema de monitorização que pode ser usado em três contextos diferentes: monitorização de um estudante, monitorização de uma sala de aula ou monitorização de um grupo de pessoas. Por forma a construir este sistema, baseamo-nos no uso de algoritmos para deteção facial, reconhecimento facial e extração de características faciais. Durante este trabalho também foi implementado um tracking do olhar, tracking facial, estimativa da pose da cabeça e deteção de emoções. Para cada contexto, várias abordagens foram desenvolvidas. Para a monitorização de um estudante, foi preciso reconhecer a face. Para este contexto, o algoritmo mais conveniente foi baseado no Local Binary Patterns Histograms. Depois de um reconhecimento bem sucedido, o sistema atribui um ID à face e começa a dar tracking à mesma enquanto recolhe informação útil das características faciais. Para a monitorização de uma sala de aula, não foi preciso reconhecer faces, apenas tracking facial. Para a monitorização de um grupo de pessoas, foi preciso fazer reconhecimento facial de cada vez que aparecesse uma nova face em cena, e depois de um reconhecimento bem sucedido, o tracking facial começa. As principais contribuições desta tese foram uma calibração automática para a câmera digital usada no sistema para um melhor reconhecimento facial, uma solução modular separada em três componentes que podem ser usados para monitorizar três contextos diferentes, recolhendo informação relevante durante um certo período de tempo que certo indivíduo esteve em frente da câmera. O software desenvolvido foi integrado num software gráfico de user interface fornecido pelo fabricante da câmera.

Full text--- In order to obtain new crop varieties in shorter time, plant breeding programs require versatile and inexpensive tools. For cacao crop, there are available a number of molecular techniques that support the breeder’s job. Nonetheless, work objectives and laboratory conditions determine the utility and convenience of those techniques. In this research, the aim was to test the discriminatory power of ISSR molecular markers in a cacao collection in Tingo Maria, and to determine its genetic diversity. Fifty cacao accessions from different geographical and genetic origin constituted the sampled collection. The ISSR markers consist in DNA fragments amplified by PCR located between two adjacent microsatellite (SSR) loci with the same but inverted sequence. A single primer complementary to these microsatellite sequences is used. The leaf samples were collected from two locations at the Leoncio Prado province (Huanuco region): from the cacao germplasm bank of the Universidad Nacional Agraria de la Selva, and from the Agricultural Experimental Station “Tulumayo”. A hierarchical grouping into 2 populations and 11 subpopulations of 44 accesions was performed taking into consideration the accession geographical origin. The remaining six accessions were excluded due to their hybrid origin. Leaves in good condition were sampled from one or two clonal trees. Then, they were kept and dried in silicagel. As a preliminary assay, DNA extraction and PCR amplification were carried out with four accessions: ICS1, CAT4, U70 and EET223. A group of 59 available ISSR primers were tested with these genotypes and 13 of them gave adequate amplification profiles. Then, five primers were selected and used to amplify the 50 accessions. Multivariate analyses were carried out separating 2 groups of data: the total 50 accessions constituted one group and the second group constituted by six Bioversity/UNAS hybrids and eleven parentals (total: 17 individuals). Three software packages were utilized: NTSYSpc v2.1p, Arlequin v3.1 and POPGENE v1.32. Grouping and ordination analysis, and internal validations were done with the first software.

Full textAnaliza las secuencias nucleotídicas parciales del gen citocromo oxidasa c subunidad I de larvas de Prodiplosis sp. colectadas en el cultivo del espárrago, en la provincia de Virú, departamento de la Libertad , para determinar su ubicación taxonómica a nivel de familia. Se logró amplificar una secuencia de 439 pb, para la región parcial del gen de citocromo oxidasa c sub. I, esta sección coincidió con las posiciones 1752 a 2190 del genoma mitocondrial de Drosophila yakuba. En el análisis del DNA de Prodiplosis sp. se obtuvo tres tipos de secuencias que se diferenciaron en 13 pb (439) determinadas como Haplotipos. La construcción del árbol filogenético por el método de Neighbor-Joining (NJ) confirmó que Prodiplosis sp. esta estrechamente relacionada con las otras especies de la familia Cecidomyiidae, por lo que se concluye de este estudio, que utilizando los iniciadores C1-J1751 y C1-N219 es posible amplificar la secuencia parcial del gen de citocromo oxidasa c sub. I en Prodiplosis sp. y constituye una herramienta muy útil para conocer su relación filogenética.

Full textEvalúa el factor masculino de los pacientes con historial de infertilidad, estandarizando y llevando a cabo dos pruebas que son de interés diagnóstico como la fragmentación del ADN en espermatozoides, conocida como SCD, la cual se basa en la dispersión de la cromatina ante agentes denaturantes ácidos y ell FISH en espermatozoides, que busca conocer el riesgo de un paciente a formar aneuploidías cromosómicas en el proceso de espermatogénesis, este diagnóstico se correlacionó con los resultados en embriones cuando se realizó el Diagnóstico Genético Preimplantacional (PGD). La prueba de SCD fue estandarizada tomando solo dos tipos de comportamientos de dispersión que son: la dispersión y la no dispersión de la cromatina. La inducción a la descompactación se consiguió con tratamientos denaturantes a pH ácido y se visualizó por microscopía de fluorescencia. Para la evaluación cromosómica de los espermatozoides por FISH, las muestras fueron hibridadas para los cromosomas 13, 18, 21, X e Y. En el análisis del PGD, las muestras tomadas por las clínicas de fertilidad fueron recibidas en el laboratorio para el análisis de hibridización fluorescente in situ de una sola célula. Los resultados del SCD indican que el 30,51% de los pacientes presentaron índices de fragmentación del ADN (DFI) elevados. Se observó una relación directamente proporcional entre la edad y el DFI; se estableció un nuevo punto de corte de 29%. El 34,26% de los pacientes que se sometieron a la prueba de FISH en espermatozoides resultaron afectados por presentar aneuploidías cromosómicas. Con el PGD se muestra un riesgo incrementado de segregación de aneuploidías por vía paterna pasando desde un 30,71% de normalidad hacia un 19,14%. Nuestros resultados indican que la fragmentación de ADN en espermatozoides no se relaciona con las aneuploidías cromosómicas por ser eventos independientes y que las aneuploidías cromosómicas detectadas por FISH en espermatozoides conllevan a un riesgo embrionario, recomendándose el PGD.

Full textThis work presents the strategies for quantitative and semi quantitative analyses by Energy Dispersive X-Ray Fluorescence (EDXRF) in three typical situations: a) homogeneous materials whose components are mainly within the field of detection of the technique (quantitative analysis), b) homogenous materials whose components are mostly outside the field of detection of the technique (quantitative analysis of low-content detected elements), and c) heterogeneous materials in superposed layers (semi-quantitative analysis of detected elements). The work describes the two spectrometers used for measurement, developed in the laboratory. For the determination of peak areas, it was used mainly PyMCA software developed by CERN. For the determination of compositions, two quantitative Fundamental- Parameters based (FP) softwares were used. The first is PyMCA, that performs simultaneously the fitting of peaks and the determination the contents. The second belongs to the Ara-Lihuen package, developed in the laboratory, and obtains contents from peak areas. The work presents the development of Ara-Lihuen routines that allow verification of intermediate calculation steps.

Várias dietas de diferentes composições estão disponíveis para o uso em experimentação animal e têm sido utilizadas como padrão, mas podem induzir efeitos metabólicos distintos, comprometendo a comparação entre os resultados dos diversos estudos. A literatura registra inúmeros relatos de alterações relacionadas ao uso dessas dietas, porém, há carência de estudos que comparem os efeitos metabólicos do consumo das diferentes dietas padrão utilizadas em experimentos animais. Assim sendo, o objetivo da presente dissertação foi avaliar as repercussões metabólicas do consumo de dietas consideradas padrão, amplamente utilizadas na pesquisa animal, sendo apresentada na forma de dois artigos: uma revisão da literatura, que reúne evidências ainda pouco debatidas pela comunidade científica relativas à alimentação de animais de laboratório, e um segundo artigo, que se refere a um estudo experimental com ratos Wistar recémdesmamados, que receberam dois tipos de dieta: uma comercial à base de cereais, Nuvilab®, e outra purificada proposta pelo American Institute of Nutrition, a AIN-93. Nas condições experimentais estabelecidas, coeficientes de eficiências protéica e alimentar apresentaram-se significativamente maiores no grupo AIN-93 que no grupo Nuvilab®. A AIN-93 apresentou digestibilidades lipídica e protéica significativamente maiores que a Nuvilab®. As diferentes dietas não causaram diferença na evolução ponderal dos animais e a análise histológica ao microscópio óptico dos rins, coração, baço, estômago e intestinos não evidenciou alterações nas estruturas desses órgãos, apesar dos diferentes tratamentos. Os animais alimentados com a dieta AIN-93, independente da idade, apresentaram esteatose hepática em uma frequência significativamente maior que os animais que receberam a comercial Nuvilab®. As diferentes dietas não exerceram influência sobre os pesos absoluto e relativo dos órgãos dos animais, com exceção do peso absoluto do fígado, entre os animais mais jovens, e do peso relativo do intestino, entre os animais mais velhos. Não se observou influência das diferentes dietas sobre os parâmetros bioquímicos avaliados, sendo as diferenças detectadas possivelmente resultantes da interação entre a idade e o tempo de exposição dos animais às dietas. Os marcadores de lesão e função hepática e renal foram similares e a creatinina sérica variou em função da idade. Demonstrou-se que ambas as dietas, AIN-93 e Nuvilab®, são capazes de promover o crescimento de ratos Wistar por um período de estudo considerado subcrônico. Porém, a ocorrência de esteatose hepática nos animais alimentados com a dieta AIN-93 peletizada, reforça a importância de monitoramento de protocolos padrão de experimentação e é indicativa de inadequações nutricionais, impondo a necessidade de investigações adicionais para esclarecer que componentes ou características dessa dieta, amplamente utilizada em experimentação animal, podem ter contribuído para tal resultado. Por hora, sugere-se que a dieta AIN-93 seja oferecida preferencialmente na forma de farinha, particularmente em protocolos que investiguem efeitos metabólicos.

Full textIn this work, we have treated and analyzed data cubes of 8 nearby active galactic nuclei, in the optical and in the infrared. All data cube treatment and analysis were performed using a series of specific methodologies, many of which were developed in the last few years by the author\'s working group. The analysis of all data cubes was performed with four main objectives: analysis of the general phenomenology of the AGN environment, using PCA Tomography; analysis of the stellar populations, of the thermal emission from dust and of the featureless continuum in the vicinity of the AGN, with a spectral synthesis performed with the Starlight software; analysis of the stellar and gas kinematics around the central black hole and, in some cases, simulations with the purpose of determining the mass of the black hole; analysis of the excitation and ionization mechanisms of the gas or of the emission from molecular hydrogen.

Full textDe una panorámica jurídico-histórica del sector eléctrico hasta la década del 70’ en Chile, se profundiza en el nuevo marco regulatorio que introduce la Ley General de Servicios Eléctricos de 1982. Luego, se describen los principales cambios normativos del sector, en particular aquellos que propenden a la inclusión de generadores de ERNC y la reciente promulgación de la Ley N°20.936. En seguida, se explica el concepto de licitación y sus diversos mecanismos, para luego reseñar el contexto en que surge la Ley N°20.018, sus objetivos, características y críticas a sus resultados. A continuación, se señalan los fundamentos y objetivos que informan a la Ley N°20.805, así como se indican y desarrollan los principales cambios que la norma introduce en el sistema de licitación de suministro eléctrico a clientes regulados, al mismo tiempo que se mencionan las críticas surgidas desde el debate público en torno a las referidas modificaciones. Finalmente, se exponen los resultados de los dos procesos licitatorios bajo el nuevo esquema de la Ley Nº 20.805 y los cuestionamientos de que ha sido objeto.

Full textCroton is a large genus of Euphorbiaceae comprising around 1.300 species, widespread in tropical and subtropical regions of America, Africa, Asia and Australia. Brazil is one of the main hot spot in the world, with more than 350 species described, many of them endemic. Although several species are used in tradicional medicine for the treatement of diseases like cancer, diabetes, fever, high cholesterol and high blood pressure. Most species have no phytochemical studies concerning their biological activities. Literature shows terpenoids as predominant secondary metabolite constituents in Croton. The present study was carried outwith the analysis of essential oils and crude methanolic extracts obtained from leaves and bark of 10 samples of Croton sect. Lamprocroton. All species used on this study were submitted to their first chemical description. Forty four compounds were identified on essential oil by GC/MS, 14 monoterpenes and 30 sesquiterpenes. Some oxygenated compounds with important biological activity were detectaed in high concentrations: 1,8-cineol (C. ericoides: 24.1%; C.linearifolius: 26.9%; C. muellerianus: 23.9%), linalol (C. dusenii: 18.2%), bisabolol (C. ceanothifolius: 49.9%), 1-isopropil-7-metil-4-metileno-1,3,4,6,8-hexa-hidro-2H-naftalen-4-ol (C. linearifolius: 25.2%; C. pallidulus var. pallidulus: 23.6%). The metanolic extract analysis reveled by GC/MS15 diterpenes, 9 triterpenes e 13 steroids. Diterpenes are describe as the main secondary metabolite in Croton. Besides diterpenes with labdan, cauran, clerodan and podocarpan skeletal types, three opened ring diterpenes (fitol, hexadecatetraenol e furanona) and one alcoholic diterpene (retinol) were detected. Two clerodans were identified with trans-desidrocrotonin estructure-like, a compound with high pharmacological use. Podocarpans that have only been registered for two Croton species, were commonly found on Croton sect. Lamprocroton (podocarp-7-en-3-one 13β-metil-13-vinil; podocarp-7,8-diien-3-one-13-acetoxi; podocarpa-7,13-dien-15-oic acid; and metil-13(2-metoxi-2-oxoetildeno)-14-metil-7-oxopodocarpan-15-oate). Triterpenes and steroids shown high diversity and were detected at all studied species, with a highlight for compounds with high pharmacological potencial like triterpens α-amirine, lupeol and lupenone; and the steroids beta and gamma-sitosterol. The results obtained led to a list of compounds and suggestions for future pharmacological tests and developments.
