Abstract:

Abstract:
In eucaryotic cell, nuclear pore complexes (NPCs) allow traffic between nuclear and cytoplasmic compartments. NPCs are large assemblies of proteins, named nucleoporins, that are anchored within the nuclear envelope. During my PhD, I have analyzed the function of a NPC sub-complex, the Nup107-160 complex. The study of this complex using an RNA interference approach in human cells confirmed that this complex has an important role in messengers RNA export in interphase, a feature that might be linked to its appearance in intranuclear foci. In parallel, I have studied the function of the Nup107-160 complex during vertebrate mitosis. I observed that depletion of some components of the Nup107-160 complex by RNA interference induces a decreased staining at the nuclear envelope of numerous nucleoporins and a drastic decrease of NPC number. These data together with in vitro experiments done in collaboration with Dr Mattaj’s laboratory have demonstrated that the Nup107-160 complex plays a critical role during NPC post-mitotic reassembly. Furthermore, the Nup107-160 complex also localizes at kinetochores. We found a potential partner of this complex within these structures. In addition, analysis of the phenotypes induced by Nup107-160 complex depletion at kinetochores show that this complex might be important for proper chromosome segregation during mitosis. The data included in this manuscript have therefore demonstrated that the Nup107-160 complex plays an important role in NPC formation and open new perspectives for the understanding of the mechanisms underlying its involvement in RNA export and chromosome segregation

Abstract:

Azizi, Hiva. "Mechanistic insights of SIDER2 retroposon-mediated mRNA decay in Leishmania." Doctoral thesis, Université Laval, 2017. http://hdl.handle.net/20.500.11794/27725.

Abstract:
Leishmania spp. are important human pathogens which put lives of over 350 million people at risk, worldwide. Apart from being an important human pathogen, Leishmania has unique features in terms of gene regulation, rendering it an excellent model organism to study gene regulation mechanisms. Notably, Leishmania and other trypanosomatids lack control at the level of transcription initiation and therefore most of the regulation of gene expression takes place post-transcriptionally. Cis-acting elements in 3ʹ-untranslated regions (3ʹUTRs) of Leishmania messenger RNAs (mRNAs) are central to the regulation of mRNA decay or translation efficiency. We have identified a novel class of cis-acting retroposons, termed SIDERs (Short Interspersed DEgenerate Retroposons) that are widely distributed in the parasite genome (>2000 copies of SIDER1 and SIDER2), mostly within 3ʹUTRs. Transcripts bearing SIDER2 in their 3ʹUTR are degraded via a deadenylation-independent pathway involving endonucleolytic cleavage within the conserved signature II (SII) sequence of SIDER2 elements. My research project aimed at determining the sequence requirements for endonucleolytic cleavage and identifying the trans-acting factor(s) contributing to SIDER2-mediated mRNA decay. We employed a tethering approach using the MS2 system to capture the trans-acting proteins in vivo. Amongst the proteins specifically tethered to SIDER2, the Pumilio protein PUF6 was shown to downregulate the SIDER2-harboring reporter transcript. Furthermore, inactivation of the PUF6 gene resulted in upregulation and increased transcript stability, indicating that PUF6 contributes to SIDER2-mediated decay. Mutational analysis within the conserved SII region, known to regulate decay, highlighted the importance of the previously mapped putative cleavage sites in mediating degradation of SIDER2-containing transcripts. Furthermore, two additional regions closer to the end of the SIDER2 sequence were found to contribute to mRNA destabilization. Finally, we addressed the requirement of translation for SIDER2 mediated decay and showed that degradation of SIDER2 transcripts is linked to ongoing translation, underscoring significance of the translation apparatus in global regulation of SIDER2-containing transcripts.

Abstract:
AbstractThis short paper takes initial steps towards developing a novel approach, called log slicing, that aims to answer a practical question in the field of log analysis: Can we automatically identify log messages related to a specific message (e.g., an error message)? The basic idea behind log slicing is that we can consider how different log messages are “computationally related” to each other by looking at the corresponding logging statements in the source code. These logging statements are identified by 1) computing a backwards program slice, using as criterion the logging statement that generated a problematic log message; and 2) extending that slice to include relevant logging statements. The paper presents a problem definition of log slicing, describes an initial approach for log slicing, and discusses a key open issue that can lead towards new research directions.

Abstract:
AbstractA message is defined as any piece of information that a person communicates to another individual or group. On the other hand, a secure messaging system protects and secures individuals’ and organizations’ communication infrastructure. End-to-end encryption uses encryption and decryption keys to ensure the privacy of messages and the authenticity of the sender and recipient. Technological advancements, such as cloud-based and blockchain-based platforms, drive growth in the secure messaging market. However, risks like phishing and cyberattacks remain persistent and are projected to continue targeting messages in the future.

Abstract:
Smishing attack is generally performed by sending a fake short message service (SMS) that contains a link of the malicious webpage or application. Smishing messages are the subclass of spam SMS and these are more harmful compared to spam messages. There are various solutions available to detect the spam messages. However, no existing solution, filters the smishing message from the spam message. Therefore, this article presents a novel method to filter smishing message from spam message. The proposed approach is divided into two phases. The first phase filters the spam messages and ham messages. The second phase filters smishing messages from spam messages. The performance of the proposed method is evaluated on various machine learning classifiers using the dataset of ham and spam messages. The simulation results indicate that the proposed approach can detect spam messages with the accuracy of 94.9% and it can filter smishing messages with the accuracy of 96% on neural network classifier.

Abstract:
The aim of this paper is to provide a solution to the growing need for fresh music to use in media, as adding music can greatly enhance the media’s atmosphere and the viewers’ experience [6]. Our solution to this issue was the creation of a mobile application named MFly that can output music using the sentiment from an inputted message. To test the effectiveness of this new music-generating method, an experiment was conducted in which twenty-three participants inputted a message with a positive and negative sentiment each and recorded whether each outputted musical piece accurately represented the sentiment from the message [7]. A post-experiment survey was also provided to each of the participants to gauge the convenience and practicality of the application. The results indicated that MFly was largely successful at conveying messages into appropriately fitting music. However, the practicality of the application could use some work, as generating music based on the sentiment does not always seem to match up with the original inputted message's sentiment, especially with messages that have a negative sentiment. Furthermore, feedback from participants indicated that the application could still improve with the addition of more features, such as the ability to save the generated music for later use.

The term steganography is usually associated with hiding and concealing information and messages. People, and even IT professionals, very rarely come into contact with steganography and steganalysis. Only messages are protected by cryptographic protection, while steganography can be said to protect both messages and parties participating in the communication. Steganography also means hiding messages inside computer files and data streams. This paper provides an overview of the implementation of embedded messages using steganography in the PHP software package. Emphasis is placed on hiding information, i.e. messages in JPEG images. As well as decoding or reading a hidden message. The field of steganography is naturally linked to the field of steganalysis, the primary goal of which is the detection of a hidden message, and then its extraction from the object of the message carrier. The most commonly used method for hiding messages is LSB, it is a method that changes the least significant bits to match the secret message. Then, by passing the steganographic key, the message is encrypted using the RC4 algorithm.

Abstract:
Synthesis-based steganography without embedding (SWE) methods transform secret messages to container images synthesised by generative networks, which eliminates distortions of container images and thus can fundamentally resist typical steganalysis tools. However, existing methods suffer from weak message recovery robustness, synthesis fidelity, and the risk of message leakage. To address these problems, we propose a novel robust steganography without embedding method in this paper. In particular, we design a secure weight modulation-based generator by introducing secure factors to hide secret messages in synthesised container images. In this manner, the synthesised results are modulated by secure factors and thus the secret messages are inaccessible when using fake factors, thus reducing the risk of message leakage. Furthermore, we design a difference predictor via the reconstruction of tampered container images together with an adversarial training strategy to iteratively update the estimation of hidden messages. This ensures robustness of recovering hidden messages, while degradation of synthesis fidelity is reduced since the generator is not included in the adversarial training. Extensive experimental results convincingly demonstrate that our proposed method is effective in avoiding message leakage and superior to other existing methods in terms of recovery robustness and synthesis fidelity.

Abstract:
Deuterium is a protocol for instant messaging that allows users to join a channel, securely exchange messages, and rotate the group key for security purposes. When a user wants to join a channel, they must first send their public key, wallet address, and a digital signature to verify their identity. If the user's identity is successfully verified, the channel’s creator will perform an elliptic Elliptic Curve Diffie-Hellman key exchange with the user using curve25519,generating a group key for encrypting messages in the channel. The group key is periodically rotated for security purposes. Users can send messages to the channel by encrypting them with the X25519-XSalsa-Poly1305 algorithm, including a Galois Message Authentication Code instead of an index after keys are exchanged, and attaching a digital signature to verify the authenticity of the message. The protocol also includes a "Termination event" for handling errors or exceptions that may occur during key exchange or message exchange.

Abstract:
The mechanisms we find in a communication process reveal to us a field of empathy, fundamental in a structured system of reading and interpreting concepts. In this way, homeostasis regulates our organism in a structured context and tries to maintain an emotional balance in the face of a world of prolific content. Thus, the conditions of environment and language reveal levels of interpretation to the messages consumed daily and in various conditions. And, in tune with the phonemes and graphemes, we find an association of meaning for the writing of information, poetry and, other media. However, the difficulty of interpreting and decoding the concepts implies a field of communicational skills knowledge levels, whose impact self-reveals trust in the receivers of these messages. How can these messages cause empathy in their receivers and enhance learning and knowledge levels to change attitudes and behaviors in a sophisticated environment? It is certain that the way of seduction, through careful communication, in whose approach of form, structure, and color, easily manipulates the presence of the word in a game of attraction and manipulation in the field such as information design. It is in the field of empathy that we witness a significant reduction in the reading of the contents and the understanding of the associated concepts. A clear approach to the world of abstract ideas and a certain egotism is demonstrated, at certain moments even with some aggressiveness, in whose lack of empathy no levels of generosity or collaboration are seen between the words spoken and the words that include said content. The broadening of the media and its general dispersal potentiates a varied manipulation of information and interests. However, the creation of empathy in the face of what is suggested and pronounced shows an artificiality in the way messages are associated with emotions, that is, they reveal themselves to be devoid of meaning. In this way, an approach is intended, reflecting the contemporary world, through the literacy of emotion in the context of verbal and non-verbal communication. In an effective communication process, we have emotions in our favor. And, in this context, we are faced with a set of tools with which nature has endowed us, in order to, create bonds of trust and thus achieve group harmony. Thus, considering the human evolutionary process, we find effective communication based on a message, whose origin is a common communication channel, a common language, and a genuine message, creating empathy in the receiver of the message, and provoking a reaction of complicity and connection to its content and context. If the message's origin has an emotional bond with the sender, it will certainly have a generous and collaborative impact on its receiver. On the contrary, a message whose emotion is reduced to thought, logic and strategy, aims to achieve in the receivers only information and a reaction of caution, selfishness and individualism, causing a social distancing from the group, enhancing the lack of empathy with the other.

Abstract:
This work builds an effective AI-based message generation system for diabetes prevention in rural areas, where the diabetes rate has been increasing at an alarming rate. The messages contain information about diabetes causes and complications and the impact of nutrition and fitness on preventing diabetes. We propose to apply reinforcement learning (RL) to optimize our message selection policy over time, tailoring our messages to align with each individual participant's needs and preferences. We conduct an extensive field study in a large country in Asia which involves more than 1000 participants who are local villagers and they receive messages generated by our system, over a period of six months. Our analysis shows that with the use of AI, we can deliver significant improvements in the participants' diabetes-related knowledge, physical activity levels, and high-fat food avoidance, when compared to a static message set. Furthermore, we build a new neural network based behavior model to predict behavior changes of participants, trained on data collected during our study. By exploiting underlying characteristics of health-related behavior, we manage to significantly improve the prediction accuracy of our model compared to baselines.

Abstract:
The present work arose from problems occurred during the revamp of a pipeline SCADA (Supervisory Control and Data Acquisition) system at the beginning of 2012, when occurred some unexpected system crashes that could interrupt the operation of the second major Brazilian pipeline maritime terminal. Before a system breakdown, we observed some signs, like fail-overs in the event log files. If the development and maintenance crews were aware of these events not only the problem causes could be better understood, but also the imminent crash could have been avoided. A faster and autonomous way for the system communicates its problems was necessary. ACARS (Aircraft Communications Addressing and Reporting System) — a part of an autonomous communication system, which reports aircraft condition for a system on the ground, through satellite links and short messages — inspired us to develop an Internet of Things (IoT) system, using text messages (SMS, short message service) of the Global System for Mobile Communications (GSM). Autonomous and short text messages are the keywords that drove our work, and the solution came through a text message gateway — the solution to get information in advance. This presentation will discuss the idea, hardware and software components, message format, applications and future perspectives.

Abstract:
Dialogue disentanglement aims to separate intermingled messages into detached sessions. The existing research focuses on two-step architectures, in which a model first retrieves the relationships between two messages and then divides the message stream into separate clusters. Almost all existing work puts significant efforts on selecting features for message-pair classification and clustering, while ignoring the semantic coherence within each session. In this paper, we introduce the first end-to- end transition-based model for online dialogue disentanglement. Our model captures the sequential information of each session as the online algorithm proceeds on processing a dialogue. The coherence in a session is hence modeled when messages are sequentially added into their best-matching sessions. Meanwhile, the research field still lacks data for studying end-to-end dialogue disentanglement, so we construct a large-scale dataset by extracting coherent dialogues from online movie scripts. We evaluate our model on both the dataset we developed and the publicly available Ubuntu IRC dataset [Kummerfeld et al., 2019]. The results show that our model significantly outperforms the existing algorithms. Further experiments demonstrate that our model better captures the sequential semantics and obtains more coherent disentangled sessions.

Abstract:
This article presents an approach for inter-agent communication and inter-platform agent migration based on IEEE FIPA (Foundation for Intelligent Physical Agents) Agent Communication Language (ACL) messages. The promising features of IEEE FIPA standards that support open, flexible, and interoperable agent communication and migration are discussed in the paper. Messages for agent communication and migration, platform architecture that deliver and manage these messages, and the main procedure of processing a mobile agent message are also introduced. An application example, in which a mobile agent visits multiple hosts and travels via mobile agent messages, simulates a mobile agent for remote data processing. This example shows the feasibility, simplicity, and openness of the presented approach for mobile agent migration. Using FIPA ACL messages for agent migration in FIPA compliant agent systems simplifies agent platform, reduces development effort, and easily achieves inter-platform migration through well-designed communication mechanisms provided in the agent platform.

Abstract:
The results of a field experiment in Argentina indicate that taxpayers who received a deterrent message (describing the penalties for non-compliance) are more likely to comply with payment of taxes than taxpayers in the control group. After receiving reciprocity and peer effects messages, the probability of compliance increased for some contributors but decreased for others according to their underlying distribution of beliefs. The use of messages on tax bills influences taxpayers depending on their prior beliefs, the location of their residence, and whether or not they live in the city where they have to pay the tax.

Abstract:
The purpose of this study was to map the on-vessel receipt of message 21 broadcasts from shoreside Automatic Identification System (AIS) sites. Message 21 is one of 27 different AIS messages, and it is used to provide information about real and virtual aids to navigation (AtoNs). Virtual AtoNs are broadcast to warn mariners of hazards like temporary construction zones or submerged debris that may not be marked with a physical buoy. In this study, message 21 was broadcast from different shore-based AIS transceiver sites along the river. Equipment onboard the patrol vessel Pathfinder was monitored for receipt of message 21 during patrols on the Mississippi River that ranged from Lock and Dam (L&D) 22 to Cairo, Illinois, with the confluence of the Ohio River. The Pathfinder is owned by the US Army Corps of Engineers (USACE) and is based out of the St. Louis District (MVS). Understanding where vessels receive, or do not receive, message 21 has important implications for maritime safety in this heavily traveled portion of the inland waterway system.

Abstract:
Despite increased efforts to improve safety in recent years (e.g., the Focus Cities Program in California), California continues to have a high rate of pedestrian and bicyclist fatalities. Currently, the state currently lacks a cohesive messaging strategy to improve behaviors related to pedestrian and cyclist traffic safety practices. To fulfill this need, this research showcases the differential effect of message framing on attitudes and intended behaviors related to pedestrian and cyclists traffic safety practices. This project investigated factors & risky behaviors contributing to accidents involving vulnerable road users, preventive measures to decrease accidents involving vulnerable road users, and more. The qualitative analysis presented a significant lack of coherent, long-term, evidence-based communication strategies that aimed at enhancing the safety of vulnerable road users in California. Quantitatively, this research also experimentally investigated various messages, employing different time horizons and regulatory focus message framings. Findings indicate that the messages with a limited time horizon tend to be associated with better safety perceptions and attitudes than messages with an expansive time horizon. California transportation authorities, professionals, and advocacy groups will be able to use this information to effectively allocate the communication effort and spending to induce attitudinal and behavioral change that can impact the safety of active transportation modes.

Abstract:
This paper develops a semiotic-inferential model of verbal communication for incomplete information games: a language is seen as a set of conventional signs that point to types, and the credibility of a message depends on the strategic context. Formally, there is an encoding-decoding step where the receiver can understand the sender's message if and only if a common language is used, and an inferential step where the receiver may either trust the message's literal meaning or disregard it when updating priors. The epistemic requirement that information be transmitted through the literal meaning of the message uttered leads to an equilibrium concept distinct from a Perfect Bayesian Equilibrium, ruling out informative equilibria where language is not used in its ordinary sense. The paper also proposes a refinement by which the sender selects among equilibria if all sender types are willing to play the same equilibrium.

This paper presents novel evidence of an intervention to foster preschool students cognitive skills during COVID-19 pandemic. We conducted a policy experiment that provided preschool student parents with a SMS text message program to support student learning at home. Taking advantage of existing parent networks, we study the direct effect of being selected to receive the SMS text messages, and the spillovers of being part of a parent network. We show that after 15 weeks of intervention, SMS text messages increase student cognitive skills by 0.11 to 0.12 standard deviations. The effect is driven by an increase of parental involvement through the proposed activities. We find no evidence that information is transferred within parent networks.

Abstract:
Key messages • We found that there are limited studies analysing the effectiveness of e-cigarette cessation interventions in a clinical setting and of those that do exist the sample sizes are small, and the studies are underpowered to make any confident assessment of their effectiveness. • Clinical interventions appropriate for young people included nonpharmacologic interventions such as contingency management and behavioural counselling while NRT may be an effective pharmacologic intervention. • There was limited evidence to demonstrate the effectiveness of behavioural counselling as a stand-alone cessation strategy, but it may be effective in conjunction with other approaches. • Emerging evidence suggests that digital cessation interventions (i.e. text message or app-based delivery) may be the preferred mode of delivery for young people, however, their effectiveness in maintaining abstinence is yet to be confirmed. • Evidence suggests there is a need to quantify and create a consistent measure of nicotine intake to appropriately inform clinical treatment decisions. • Studies are generally very low quality, and it is not possible nor is it appropriate to make any definitive conclusions.
