Abstract:
La formation de nouveaux vaisseaux sanguins ou angiogenèse soutient la croissance tumorale en fournissant l'oxygène et les nutriments qui lui sont nécessaires. Le rôle clé du facteur de croissance de l'endothélium vasculaire VEGF dans ce processus a suscité le développement de stratégies anti-angiogéniques pour le traitement du cancer. Cependant, des travaux précliniques et des données cliniques suggèrent l'émergence de résistances aux anti-angiogéniques, en raison notamment de la redondance des facteurs de croissance pro-angiogéniques. Il est donc nécessaire de développer des stratégies alternatives plus efficaces. En 2010, notre laboratoire a apporté la preuve de concept d'une thérapie anti-tumorale et anti-angiogénique innovante basée la dégradation des ARNm à demi-vie courte par la protéine à doigts de zinc TIS11b. Néanmoins, l'instabilité de la protéine thérapeutique a entravé la caractérisation plus détaillée de cette stratégie. Dans ce contexte, l'objectif majeur de ma thèse était l'optimisation de la stabilité et de l'activité de TIS11b et l'évaluation de son efficacité thérapeutique. Pour cela, nous avons généré une nouvelle protéine TIS11b génétiquement modifiée sur la base d'études biochimiques et moléculaires. Notamment, nous avons observé que la phosphorylation de la sérine 334 située dans le domaine C-terminal de TIS11b augmente de façon très significative la stabilité de la protéine et potentialise son activité déstabilisatrice de l'ARNm du VEGF. De plus, la délétion du domaine N-terminal augmente également la stabilité de TIS11b sans altérer son activité. Nous avons alors généré deux nouvelles protéines thérapeutiques, la protéine ZnC et la protéine ZnC334D pour laquelle la troncation du domaine N-terminal et la substitution de la sérine S334 par un aspartate mimant une phosphorylation ont été combinées. Les nouvelles protéines ont été fusionnées à une étiquette polyarginine R9 leur permettant de traverser les membranes cellulaires (R9-ZnC et R9-ZnCS334D). Nous avons montré que R9-ZnC et R9-ZnCS334D inhibent l'expression de VEGF in vitro dans la lignée de cancer du sein murin 4T1. De plus, R9-ZnCS334D exerce une activité anti-proliférative, anti-migratoire et anti-invasive dans ces cellules. In vivo, l'injection intra-tumorale de R9-ZnCS334D dans des tumeurs 4T1 préétablies inhibe significativement l'expression du VEGF, la croissance et la vascularisation tumorales. De façon remarquable, l'analyse des extraits tumoraux indique que le traitement diminue l'expression de chimiokines clés dans les processus d'angiogenèse, d'inflammation et d'invasion (Fractalkine, MCP-1, NOV, SDF-1, Pentraxin…). Enfin, R9-ZnC et R9-ZnCS334D inhibent l'expression de marqueurs de la transition épithélio-mésenchymateuse, un processus impliqué dans la dissémination métastatique. L'ensemble de ces travaux indique que R9-ZnC et R9-ZnCS334D sont des molécules anti-tumorales multi-cibles, qui inhibent plusieurs étapes clés de la progression tumorale. Cette étude confirme que le ciblage de la stabilité des ARNm est une stratégie prometteuse et novatrice pour le développement de nouvelles thérapies anti-cancéreuses
One of the innovative aspects of anti-cancer therapies is the possibility of preventing tumor growth by blocking blood supply. Cancer cells induce the formation of their own blood vessels from pre-existing vasculature, a process called angiogenesis. One of the most important proangiogenic factors is vascular endothelial growth factor (VEGF). The success of bevacizumab (a humanized anti-VEGF monoclonal antibody) combined to chemotherapy for the treatment of human metastatic cancers has validated VEGF as an efficient target. However, despite the initial enthusiasm, resistance to these anti-angiogenic treatments resulting from compensatory mechanisms occurs upon time. For this reason, there is a real need for new anti-angiogenic drugs that will target the angiogenic process through distinct mechanisms. In 2010, our laboratory has successfully developed an anti-angiogenic and anti-tumoral therapy based on destabilization of short-lived mRNAs by the zinc finger protein TIS11b. However, the therapeutic protein was highly unstable, thus making it difficult to further characterize the experimental therapy. In this context, the main task of my thesis was the optimization of TIS11b stability and activity followed by the evaluation of the multi-target action of our novel protein on tumor development. In a first part of this work, biochemical and molecular approaches allowed us to demonstrate that phosphorylation of the C-terminal serine S334 in TIS11b protein markedly increases its stability. In addition, deletion of the N-terminal domain of TIS11b highly increases its protein stability without affecting its activity. Therefore, we integrated N-terminal truncation (ZnC) and C-terminal substitution of S334 by an aspartate to mimic a permanent phosphorylation at S334 (ZnCS334D) as a novel TIS11b engineering strategy. Both proteins were fused subsequently to a cell-penetrating peptide polyarginine (R9). In vitro studies revealed that R9-ZnC and R9-ZnCS334D inhibit VEGF expression in the murine breast cancer cells 4T1. In addition, R9-ZnCS334D impaired proliferation, migration, invasion and anchorage-independent growth of 4T1 cells. In vivo, intra-tumoral injection of either protein significantly reduced VEGF expression and tumor vascularization. Strikingly, antibody array analyses of tumor extracts demonstrated a reduced expression of several chemokines such as Fractalkine, MCP-1, NOV, SDF-1 and Pentraxin upon R9-ZnC or R9-ZnCS334D treatment. These factors, which are produced by several cell types within tumor tissue, are key drivers of tumor angiogenesis, tumor-promoting inflammation and invasion. Furthermore, the expression of markers of the epithelial-to-mesenchymal transition was also significantly reduced, suggesting an anti-metastatic effect of R9-ZnC and R9-ZnCS334D. Thus, we provide R9-ZnC and R9-ZnCS334D as potential novel multi-target agents which inhibit key hallmarks of cancer progression. This work supports the emerging link between mRNA stability and cancer and proposes novel concepts for the development of innovative anti-cancer therapies

Une étape clé dans la maturation des pré-ARNms est le clivage et la polyadénylation que ceux-ci subissent sur leur extrémité 3’. Chez les métazoaires, le complexe CstF (Cleavage stimulation Factor) reconnaît une région de l’ARNm riche en U et U/G et stabilise le complexe CPSF (Cleavage Polyadenylation Stimulating Factor) sur le site de polyadénylation. Nous avons déterminé la structure cristallographique du domaine N-terminal d’une des trois sous-unités de CstF, CstF-50. Ce domaine forme un homodimère compact et présente deux surfaces identiques conservées dérivées de la formation du dimère. La structure dimérique de CstF-50 est en accord avec le modèle hexamèrique du complexe. L’homologue de CstF chez la levure, CF IA (Cleavage/polyadenylation Factor IA), est impliqué dans les réactions de clivage et polyadénylation de la maturation 3’. Nous avons reconstitué le complexe entier ‘in vitro’ et résolu la structure en solution par RMN des régions minimales impliquées dans l’interaction des sous-unités Rna14p et Rna15p. Pour la formation de l’hétérodimère, la région C-terminale de Rna14p, que nous avons appelé domaine « monkeytail », s’entrelace intimement avec la région « hinge » de Rna15p. La présence de ces deux domaines chez leurs homologues de mammifères, CstF-77 et CstF-64, suggère la conservation de ce type d’organisation entre ces deux sous-unités à travers les espèces
The removal of the 3’ region of pre-mRNA followed by polyadenylation is a key step in mRNA maturation. In metazoa, Cleavage stimulation Factor (CstF) recognizes U and G/U rich cis-acting RNA sequence elements through its 64kDa subunit and helps stabilize the Cleavage Polyadenylation Stimulating Factor (CPSF) complex at the polyadenylation site. We describe the crystal structure of the N-terminal domain of the CstF-50 subunit. Through highly conserved residues, CstF-50 forms a compact homodimer that exposes two geometrically opposite and identical conserved surfaces. Together with prior data, the structure of the CstF-50 homodimerization domain supports a hexameric model of CstF. The yeast homologue of CstF is the Cleavage/polyadenylation Factor IA (CF IA) complex and is involved in both the cleavage and polyadenylation of pre-mRNA. We have reconstituted ‘in vitro’ the overall complex and also solved the solution structure of one of the inter-subunit regions, specifically the heterodimer involving peptides from Rna14p and Rna15p. Upon binding, a short C-terminal region from Rna14p wraps intimately within the central hinge domain from Rna15p. Conservation of residues reveals that the structural tethering is preserved in the homologous mammalian proteins
La maduración 3’ del pre-ARNm es un proceso clave de la expresión génica que incluye el corte y la poliadenilación del extremo 3’ libre del pre-ARNm. En metazoos, el complejo CstF (Cleavage stimulation Factor) reconoce una secuencia del pre-ARNm rica en U y G/U y permite la estabilización del complejo CPSF (Cleavage Polyadenylation Stimulating Factor) en el sitio de poliadenilación. Hemos descrito la estructura cristalina del dominio N-terminal de una de las tres subunidades de CstF, CstF-50. La estructura ha revelado la organización de la proteína en un dímero compacto y conservado entre las especies. Dos zonas idénticas conservadas se encuentran expuestas a ambos lados de la superficie estructural. Nuestros resultados corroboran así la hipótesis sobre el modelo hexamérico del complejo CstF. CF IA (Cleavage/ polyadenylation Factor IA), el homólogo de CstF en levaduras, interviene en las dos etapas de la maduración 3’. Las bases para la reconstitución del factor CF IA ‘in vitro’ han sido establecidas. Al mismo tiempo, hemos resuelto la estructura del subcomplejo formado por las regiones de interacción de Rna14p y de Rna15p en solución mediante RMN. En el heterodímero, las dos proteínas forman una entidad única a través de la región C-terminal de Rna14p, dominio “monkeytail”, y el dominio “hinge” de Rna15p, quedando las hélices de la dos proteínas entrelazadas. Abstract:
Arms export research has largely focused on the behaviour of superpowers or major powers, while arms export behaviour of small democracies has received little attention. By using a quantitative method, this paper explored whether democracy had any impact on the arms export behaviour of small democracies. The argument was that a good international social reputation, as responsible global actors, would cause indirect pressure on small democratic states to follow democratic norms, and therefore make them avoid arms export to autocracies. The findings did not support the argument of democratic norms to have a negative relation to arms export to autocratic regimes. Additionally, the findings indicate that economic wealth is positively correlated to arms export to autocratic regimes.

Abstract:
How are arms export choices made within a state? In this dissertation I use a foreign policy analysis framework to examine this question. I focus on examining each of the three primary levels of analysis in international relations as it relates to the main question. I begin with a typical international relations level and examine the characteristics of the two states that dominate the world arms trade: The United States and Russia. I then examine the full network of relations among all states in the international system that are involved in the sale or purchase of arms. To do this I use an Exponential Random Graph Model (ERGM) to examine these relations, which I derived from data on arms sales from the Stockholm International Peace Research Institute (SIPRI). I examine the arms sales in each decade from 1950 through 2010. In order to answer the question of how arms decisions are made within the state, I focus my inquiry on the United States and Russia. It is these states that have the practical capability to use arms transfers as a foreign policy tool. I examine the foreign policy making mechanisms in each of these states to determine how arms transfers can be used as a foreign policy tool. I examine and the bureaucratic institutions within each state and come up with a state ordering preference for how arms decisions are evaluated in each state. Finally, I use case studies to examine arms relations between the both the U.S. and Russia and three other states in each case. The other states were selected based on the pattern of sales between the two countries. I examine these sales to determine the impact of bureaucratic maneuvering and interest politics on the decision-making process within Russia and the United States. I find in my network analysis that the traditional measures of state power - military spending, regime type, and military alliances - do not account for the overall structure of the arms sale network. The most important factors in the formation of the arms sale network in each of the six decades that I study are specific configurations of triadic relations that suggest a continued hierarchy in the arms sale network. I find in my case study chapters that a simple model of state interest as a decision-making rule accounts for the decisions made by the different bureaucratic actors in the U.S. Russian arms sales are driven by a state imperative to increase Russia's market share, and there is high-level involvement in making different arms deals with other countries.

Abstract:
This paper constructs a graphical and mathematical model of dyadic interstate security competition. It does so by integrating arms racing and war initiation in to a single rational choice framework. The model is constructed from rigorously-defined concepts and all assumptions are made explicit. Equilibrium values for security-based arms racing are derived under the model and compared to conquest-seeking arms races. Comparative static results are provided for several shocks to the basic system. The model is informally extended in to probabilistic war scenarios. Finally, a number of testable predictions generated by the model are presented.
Ce mémoire présente un modèle graphique et mathématique du conflit dyadique dans le domaine de la sécurité internationale. Le modèle permet d'interpréter la course aux armements et le déclenchement des guerres dans un cadre formel. Le modèle est construit a partir de concepts rigoureusement définies et nos postulats ont été présenté de manières explicites. Les valeurs d'équilibre pour une course aux armements voulant assurer la sécurité nationale sont prédits à partir du modèle et comparé aux valeurs produites pour une course aux armements avec des objectifs de conquête. Différents résultats d'analyse statique sont comparés pour différentes perturbations du modèle de base. Le modèle est étendu intuitivement pour présenter des scénarios probabilistes de guerre. Finalement, de nombreuses propositions réfutables sont dérivées du modèle.

Abstract:
This dissertation examInes whether the arms industry is globalising. Although it is a broadly accepted view that the arms industry is glob ali sing, it is not always clear what this involves and there is little empirical evidence available to support this view. More importantly, the view that the arms industry is glob ali sing is often based on a meaning of globalisation that is not distinctively different from the meaning of internationalisation. This study is based on the view that the question whether the arms industry is globalising is meaningful only if the concept' globalisation' has a distinctly different meaning from the concept 'internationalisation'. Based on an extensive review of the globalisation literature, and especially of the literature on economic globalisation, three main features of globalisation are identified: (i) increased pace and global scope of economic cross-border activities (i.e. increased internationalisation); (ii) transformation of multinational companies into truly transnational companies; and (iii) a reduced government capacity to regulate and control cross-border activities. If these features are true for the arms industry, then globalisation of the arms industry would involve a radical change from during the cold war, when there was little internationalisation of the arms industry apart from government-to-government international armaments collaboration; when arms-producing companies were not transnational; and when governments were able to exercise control over their respective arms industries. This study examInes the intensity and global scope of international transfers of major weapons and the intensity and scope of cross-border mergers and acquisitions (M&As) in the Euro-Atlantic arms industry and analyses the impact of these trends on government control over the arms industry. It also discusses the extent of transnational companies in the arms industry and other trends in the arms industry that may be considered evidence of globalisation. The main focus of the study is on cross-border M&As in the Euro-Atlantic arms industry. The reason for this is that this trend, which has enlerged during the postcold war period, is one of the factors most often referred to as evidence of arms industry globalisation. However, so far there has been no systematic mapping of this trend, primarily due to the lack of comprehensive data. This study is based on a new data set on cross-border M&As. It covers the Euro-Atlantic area, since this is the area where most of the global arms industry is located. This is true in particular for the most advanced private arms industry, which is the most likely to be affected by the economic forces underlying globalisation.

This thesis explores how the newly adopted Arms Trade Treaty (ATT), the first internationaltreaty to regulate the trade in conventional arms, can address the issue of the illegal andirresponsible proliferation of small arms and lights weapons (SALW) and improve peace andsecurity. By far the most commonly used weapons in modern conflicts, SALW and theireffects mainly on intrastate conflicts, I argue, are the most important issues for the ATT toaddress. Being one of the prime sources of fuel for, and even cause of, the new trend ofincreasingly deadly and destructive intrastate conflicts today, controlling the largely illegaland internationally unregulated SALW market would be a crucial step to improving peaceand security.The thesis uses Security Dilemma theory to describe how arms and their proliferation posethreats to peace and security, while international law and regime theory is used to identifyhow legal action and structures of cooperation (regimes) can offer solutions to theseproblems. To determine to what extent the ATT can address these issues, the thesis uses acase study approach together with a content analysis of the ATT text to identify the relevantlegal provisions and how they can operate in the context of the theoretical framework.The findings of this study suggest that the most important aspect of the ATT is that itimposes prohibitions on any arms transfer that risks being used to commit acts of genocide,human rights abuses and other violations of international law, or that risk leading todiversion. These provisions could be used to stop the irresponsible kinds of arms transfersthat facilitate these crimes. The effects of the ATT are however largely dependent on the willof states, which will determine the effectiveness of the treaty. Nonetheless, as this thesisshows, the ATT provides tools and a legal platform that could, if utilized, have a substantialimpact on these issues.

Clear changes occurred in the field of conventional arms control in the last two decades. States adopted a multitude of norms on especially small arms control in various multilateral control instruments. In addition, non-governmental advocacy actors often established themselves as active participants in control debates with governments. The changes are surprising because they took place in the security sphere and therewith in an area traditionally understood to be the exclusive domain of governments. This research project investigates the significance of the changes for the traditional understanding of security governance. Specifically, it investigates the emergence of control norms and the role and policy impact of non-governmental actors in the promotion of the norms. It asks whether the normative changes and significance of nongovernmental actors therein challenge the understanding of security governance that underpins many established approaches to international relations theory.

This thesis examines the impact of illegal U.S. arms transfers upon recipient nations' war fighting capabilities and upon the American national security. Data were gathered primarily from U.S. District Court records and interviews with U.S. governmental officials from intelligence services and the Departments of Commerce, Justice and State. An investigation of the illicit arms transfers to Iran formed the basis of conclusions reached. Additionally, policy recommendations are provided to enhance the governmental detection and investigation of illegal export violations. The viability of utilizing court documents as intelligence tools for measuring military capabilities is assessed.

Museums have always had and displayed weapons, including firearms. As museums have evolved, so too has exhibit design and practice. However, many weapons displays have not kept up with changing practices, and many of them are now irrelevant, have limited audiences, or are unhelpful to the broader public. Simply displaying weapons by type or as art is not enough anymore, and keeping them in storage does not take advantage of their potential. Also, many museums are increasingly trying to become places for public discourse about current issues. They often create exhibits meant to be relevant to today and promote discussions about controversial topics. Many museums are also trying to make their collections and objects more accessible to the public. Innovative displays of firearms could help them accomplish both these tasks. The battle over gun control and gun rights is often more of a shouting match than reasoned discourse. Museums could use historic firearms as an opportunity to help facilitate a more responsible conversation about the issue. These firearms are typically not as emotionally charged as modern guns, and could be used as a pathway into the gun debate if displayed creatively. Guns, historic or not, are often not very approachable objects for many people. This can be for a variety of reasons, including their associations with masculinity, power, and nationality. Museums should experiment with new ways to display firearms that can make them more approachable and accessible to broader audiences, and ideally to the entire public.

A work of creative fiction in the form of a short novel, Asleep in the Arms of God is a limited-omniscient and omniscient narrative describing the experiences of a man named Wafer Roberts, born in Jack County, Texas, in 1900. The novel spans the years from 1900 to 1925, and moves from the Keechi Valley of North Texas, to Fort Worth and then France during World War One, and back again to the Keechi Valley. The dissertation opens with a preface, which examines the form of the novel, and regional and other aspects of this particular work, especially as they relate to the postmodern concern with fragmentation and conditional identity. Wafer confronts in the novel aspects of his own questionable history, which echo the larger concern with exploitative practices including racism, patriarchy, overplanting and overgrazing, and pollution, which contribute to and climax in the postmodern fragmentation. The novel attempts to make a critique of the exploitative rage of Western civilization.

Abstract:
This thesis is intended to enhance our knowledge of the processes behind the relinquishing of Canada's nuclear roles. As such, the underlying factors which helped bring about this change in Canadian defence policy are to be isolated and assessed. The process of retiring Canada's nuclear roles was long and complex, involving many actors and influences. The factors examined in this thesis are looked upon in the greater context of the 1960s and 1970s. The global and domestic political climates, the strategic environment and Canada's power within the global community as a whole during this time period, are considered. This study has revealed several interesting conclusions which one may draw concerning the relinquishing of Canada's nuclear roles. First of all, the Trudeau government's position on nuclear weapons coincided with growing opposition to nuclear weapons within the Canadian public. Furthermore, it is evident that public opinion on the nuclear issue was closely related to an individual's perception of the United States and his or her position on defence spending. Secondly, the process of retiring the nuclear weapon systems was led largely by Pierre Trudeau and some of his close associates, in particular Ivan Head and Donald MacDonald. This was done in the face of intense bureaucratic resistance. Thirdly, abandoning the nuclear roles was strongly related to Canada's declining position in the global community and also to the growth of detente. In addition, it was also partly the result of a rapidly changing strategic environemnt in which weapon systems were quickly made obsolete. Perhaps most importantly, this thesis shows that relinquishing Canada's nuclear roles was an extremely important part of the 1971 defence review. The issue of nuclear weapons is one which has been largely overlooked by defence and foreign policy analysts in the past, such as Thordarson as well as Granatstein and Bothwell, and therefore merits our attention. For this thesis, telephone interviews had to be conducted because very little written material is available to the public. Regrettably, these interviews must remain confidential for the time-being. Several books, articles and public opinion surveys also were very helpful in conducting this analysis.

Les ARNs H/ACA remplissent des fonctions variées dans la cellule. Ils servent d'ARNs
guides pour les conversions d'uridines en pseudouridines des ARNs ribosomiques mais
également des snARNs du spliceosome. Les ARNs H/ACA nucléolaires, les snoARNs,
guident les modifications des ARNr alors que ce sont des ARNs H/ACA qui s'accumulent
dans les Cajal bodies, les scaARNs, qui guident les modifications des snARNs du
spliceosome transcrits par l'ARN polymérase II. Nous avons montré par une large analyse
mutationnelle d'un ARN chimérique artificiel que la localisation des scaARNs dans les Cajal
bodies ne dépendait que d'un court motif de quatre nucléotides situé au niveau des boucles
terminales 5' et 3' du domaine H/ACA et appelé « boîte CAB ». La boîte CAB est conservée
chez de nombreux organismes et est nécessaire et suffisante à la localisation des scaARNs
dans les Cajal bodies. De façon plus inattendue, l'ARN de la télomérase, hTR, responsable de
l'élongation des télomères lorsqu'il est associé à la reverse transcriptase hTERT, comporte
également un domaine scaARN dans sa partie 3'. Ce domaine H/ACA possède en effet une
boîte CAB, située au niveau de la tige boucle 3' du domaine H/ACA, qui est responsable de la
localisation de hTR dans les Cajal bodies. Nous avons voulu comprendre la signification
biologique de cette accumulation dans les Cajal bodies tout au long du cycle cellulaire par une
approche d'hybridation in situ et d'immunofluorescence. L'étude de la dynamique
intranucléaire de hTR par microscopie à fluorescence nous a permis de mettre en évidence un
rôle de hTR, associé aux Cajal bodies, dans la régulation de l'élongation des télomères. Les
Cajal bodies pourraient en effet délivrer hTR, potentiellement associé à hTERT, directement
au niveau des télomères.
Nous nous sommes intéressés dans un deuxième temps, à l'expression et à la maturation des
sno/scaARNs H/ACA introniques. Alors que plusieurs travaux mettent en évidence
l'existence d'une synergie entre la machinerie d'épissage et la machinerie d'assemblage des
snoRNPs C/D, nos résultats montrent qu'au contraire, l'épissage et l'assemblage de la
particule H/ACA chez l'homme sont deux évènements indépendants. Nous apportons
également l'évidence que l'expression correcte des ARNs H/ACA nécessite une transcription
par l'ARN polymérase II et que la particule H/ACA s'associe très précocement au niveau du
pré-ARNm.

Industrial robots have been used as a key factor to improve productivity, quality and safety in manufacturing. Many tasks can be done by industrial robots and they usually play an important role in the system they are used, a robot stop or malfunction can compromise the whole plant as well as cause personal damages. The reliability of the system is therefore very important. Nevertheless, the tools available for maintenance of industrial robots are usually based on periodical inspection or a life time table, and do not consider the robot’s actual conditions. The use of condition monitoring and fault detection would then improve diagnosis. The main objective of this thesis is to define a parameter based diagnosis method for industrial robots. In the approach presented here, the friction phenomena is monitored and used to estimate relevant parameters that relate faults in the system. To achieve the task, the work first presents robot and friction models suitable to use in the diagnosis. The models are then identified with several different identification methods, considering the most suitable for the application sought. In order to gather knowledge about how disturbances and faults affect the friction phenomena, several experiments have been done revealing the main influences and their behavior. Finally, considering the effects caused by faults and disturbances, the models and estimation methods proposed, a fault detection scheme is built in order to detect three kind of behavioral modes of a robot (normal operation, increased friction and high increased friction), which is validated within some real scenarios.

Abstract:
In this work, I studied the in vivo functions of Kidins220 (Kinase D-interacting substrate of 220kDa, also known as ARMS; ankyrin-repeat rich membrane spanning), by phenotypic analyses of full and conditional Kidins220 knockout mice. Kidins220 is a large transmembrane protein involved in neurotrophin and Eph/ephrin signalling, with roles in neurite outgrowth, neuroprotection, and synaptic modulation. I demonstrate pleiotropic functions of Kidins220 during development of the nervous and cardiovascular systems, and a novel function downstream of vascular endothelial growth factor (VEGF)/neuropilin-1 (Nrp1) pathway. Kidins220-/- embryos develop smaller than wildtype, and die around birth. Analysis of Kidins220-/- brains revealed increased apoptosis in specific brain regions. Increased apoptosis was also observed in the peripheral nervous system, suggesting that it mediates the survival of specific cell populations. Kidins220 also regulates proliferation in the cortex. Enlargement of the heart and outflow tract defects are observed in Kidins220-/- mice, suggesting its function in neural crest cells. In vitro experiments confirmed Kidins220 as mediating neurotrophic response. Vascular defects are also observed in Kidins220-/- brains. Glomeruloid vascular tufts formed in the Kidins220-/- brains are similar to that of Nrp1-/- mice. Nrp1 is a coreceptor to VEGF receptors and I observed that Kidins220 coimmunoprecipitates with VEGFR2 and VEGFR3. The presence of angiogenesis defects specifically in the brain point to the spatially and temporally restricted functions of Kidins220. In addition, I have also identified the presence of alternative splice isoforms of Kidins220 only found in the adult. The splicing will yield a shorter Kidins220 protein lacking many of its protein-protein interacting domains, suggesting that it plays a different function from that of the conventional Kidins220 isoform. This work therefore reports novel functions of Kidins220 during embryonic development, likely through its involvement in the neurotrophin, ephrin and VEGF receptor pathways, and novel splice isoforms of Kidins220.

This thesis recovers a parliamentary struggle over English taxation which erupted at the 1660 Convention parliament that welcomed Charles II back from exile. It lasted until the eve of the second Anglo-Dutch War of 1665-67. At its centre lay a complex and shifting legislative initiative for preserving England’s naval transformation of the 1650s: the ‘Supply Acts’ for granting statutory taxes to support a strong and durable royal navy. Sponsored by King Charles and his parliamentary servants, this legislation met a hostile response in scribal and print publications that advanced a rival military agenda. The most prolific author of this offensive was the lawyer and intellectual Fabian Philipps. Caricatured by modern historians as an eccentric on a mad quest to restore England’s equestrian caste to its ancient glory and prestige, he has been dismissed as a feudal anachronism. This thesis situates his views within a fresh interpretive framework and a newly reconstructed historical setting, permitting us to appreciate that the final outcome of the contest over taxation had the long-term consequence of proportionately reducing the traditional discretionary violence of the caste. Through close attention to Philipps’ work in these contexts, a case study of state monopolization of violence is furnished.

Cette thèse traite de l’augmentation de la productivité des robots manufacturiers, lors de l’exécution de tâches référencées capteurs. De telles tâches peuvent provenir de la cible non positionnée de manière absolue ou d’un environnement mal connu. Les commandes par asservissement visuel sont bien connues pour leur robustesse et leur précision, mais nécessitent généralement de longs temps d’exécution en raison de différents facteurs. La commande est généralement formulée uniquement à un niveau cinématique et caractérisée par des vitesses décroissantes de façon exponentielle. De plus, l’application non linéaire de l’espace opérationnel à l’espace des capteurs peut conduire à des chemins sous-optimaux et plus longs. Pour augmenter les performances de commande et réduire le temps nécessaire à la réalisation d’une tâche, cette thèse étudie l’utilisation de modèles d’interaction de second ordre. Leur utilisation dans une commande au niveau dynamique est étudiée et comparée aux approches classiques. Ils sont ensuite utilisés dans des schémas de commande par modèle prédictif, permettant d’obtenir des vitesses plus élevées tout en générant de meilleures trajectoires. Cependant, un inconvénient des techniques prédictives est leur charge de calcul. Afin d’obtenir de pallier ce défaut, un nouveau type de commande prédictive est étudié. Il conduit à une réduction du nombre de variables impliquées dans les problèmes d’optimisation grâce à l’utilisation d’un paramétrage des séquences d’entrée
This thesis deals with increasing the productivity in manufacturing robots, when performing sensor-based tasks. Such tasks may be coming from the target not being absolutely positioned. Visual servoing control schemes are well known for their robustness and precision, but generally require long execution times due to differentfactors.Control laws are generally formulated only at a kinematic level and characterized by exponentially decreasing velocities. Moreover, the nonlinear map from the operational space to the sensor space can lead to sub-optimal and longer paths. To increase control performances and reduce the time required to complete a task, this thesis investigates the use of second-order interaction models. Their use in dynamic feedback control laws is investigated and compared to classical controllers. They are then employed in Model Predictive Control (MPC) schemes, allowing to obtain higher velocities and better sensor trajectories. However, a drawback of MPC techniques is their computational load. In order to obtain even better results, a new type of predictive control is thus investigated, leading to a reduced number of variables involved in MPC optimization problems thanks to the use of a parameterization of the control input sequences
