Abstract:
A new federal agency—approved last month by the United States Congress—is already off to a rocky start. The Advanced Research Projects Agency for Health (ARPA-H), proposed by President Biden in 2021, aims to tackle the most intractable biomedical problems by funding innovative, high-risk, high-reward research and swiftly turning discoveries into treatments and cures. But Congress gave the agency a much smaller budget than sought by the administration—$ 1 billion over 3 years, a fraction of the $6.5 billion requested. And as happens whenever there is new money and a new federal agency, a political scrum has erupted over who should control ARPA-H. It is now expected to answer to both the National Institutes of Health (NIH) and the Department of Health and Human Services (HHS). If it is to deliver on its mission, ARPA-H needs to be an autonomous entity that approaches biomedical research in a way never done before by the federal government. The stakes are high: If ARPA-H fails to produce new clinical advances relatively quickly, it will erode trust in US science. It’s time for clear thinking and action about what it will take to make ARPA-H successful.

Abstract:

Intervjuer med kunniga på AIS-området har utförts parallellt med en enkät riktad till aktiva befäl ute till sjöss. Detta i syfte att ta reda på dels hur detta hjälpmedel bör användas vad avser antikollision, dels hur det faktiskt används i praktiken.

AIS, Automatic Identification System, är ett informationsdelningssystem som hjälper fartyg att navigera säkert genom att sända ut egen navigationsdata och samtidigt ta emot motsvarande information från andra fartyg. Systemet nyttjas även som informationssystem för landbaserade stationer såsom VTS-centraler och kustbevakning.

Undersökningen har engagerat och visat att expertisen anser att AIS, i många avseenden, är ett bra och användbart hjälpmedel som bör komplettera övrig utrustning vid bedömning om kollisionsrisk. Vidare visar enkäten att ARPA-systemet, trots vissa inneboende brister, tillmäts större förtroende än AIS och AIS-vektorer används därför sparsamt. Enkäten visar även att en stor del av de aktiva inte är främmande för att göra avsteg från sjövägsreglerna emedan expertisen i huvudsak avråder från sådant.


Interviews with experts, in the field of AIS, have been conducted alongside a quantitative survey aimed at certified seagoing officers. The purpose of the survey was to investigate how this navigational aid preferably should be used in collision avoidance, in contrast to how it is used in reality.

AIS, Automatic Identification System, is an information sharing system to assist in safe navigation by transmitting the own vessels navigational data while, at the same time, receiving corresponding information from other ships. The system is also utilized by land based stations such as VTS-centers and coast guards.

The investigation shows that experts conclude AIS to be, in many aspects, a good and useful aid which should be used in collaboration with other equipment when assessing risk of collision. Furthermore, the survey show that the ARPA-system, in spite of some internal shortcomings, is awarded more trust than AIS and AIS vectors are therefore rarely used. The survey also shows that a large number of officers are open to deviating from the COLREGs while the experts, in general, advise against it.

Questa analisi è volta a esaminare le perturbazioni che, in date diverse (5/6 Febbraio 2015, 19-22 dicembre 2009, 1 febbraio 2012), hanno portato piovosità diffusa e nevicate intense nell'Emilia-Romagna. Il primo caso preso in esame, ossia quello relativo al periodo suddetto, ha rivestito grande importanza nello scenario dell’inverno scorso, in quanto ha provocato numerosi disagi alla viabilità, interruzione della distribuzione di acqua potabile e black-out elettrici nelle zone interessate da precipitazioni nevose; in Romagna invece sono stati numerosi gli allagamenti soprattutto nel Cesenate e nel Riminese. Nel secondo caso analizzato, invece, assistiamo al fenomeno non troppo comune del gelicidio. Provoca, infatti, anch’esso disagi gravi alla viabilità stradale e autostradale, nonché il pericolo di interruzione della corrente nelle linee elettriche, dovuta al peso del ghiaccio depositato sui conduttori che fa crollare i cavi dell’alta tensione. L’ultimo caso, infine, è stato trattato poiché rientrante nella famosa ondata di gelo della prima metà del Febbraio 2012. Vedremo, infatti, un quadro generale dell’ondata, dopodiché un “focus” sulle importanti nevicate del primo Febbraio in regione.

A crise ambiental decorrente da lógica produtiva capitalista, que foi materializada nas atitudes destrutivas do homem em relação ao ambiente, implicou a emergência da discussão sobre a Tecnologia Social (TS) como uma alternativa sustentável de desenvolvimento, em contraste à Tecnologia Convencional (TC) que é funcional ao sistema capitalista. Assim, para que os alicerces da TS sejam fundamentados e que sua legitimidade seja alcançada perante a sociedade, a sua interlocução com a universidade, principalmente no âmbito da extensão, é indubitavelmente relevante. Para tanto, investigaram-se os impactos socioambientais do uso da TS na ação de extensão do Programa UNITRABALHO da Universidade Federal do Amazonas (UFAM) na Associação de Reciclagem e Proteção Ambiental (ARPA) e de ações de extensão de programas e projetos da Pró-Reitoria de Extensão e Interiorização/ UFAM. Portanto, a pesquisa teve como principais objetivos: desvelar o processo de institucionalização do desenvolvimento a partir das principais características, ações, resultados e/ou produtos; analisar os impactos socioambientais do uso da TS nas ações de extensão no locus da pesquisa, a ARPA; e verificar se os impactos do uso da TS contribuíram para a sustentabilidade da ARPA. No percurso metodológico da pesquisa utilizou-se a abordagem quanti-qualitativa para coleta de dados/informações, por meio da aplicação de questionário junto a 08 (oito) coordenadores de projetos e programas de extensão que participaram do I Fórum de Tecnologia Social da UFAM; e por meio da realização de entrevistas semi-estruturadas junto a 01 (uma) coordenadora da UNITRABALHO e aos 08 (oito) catadores de material reciclável. A análise dos resultados da pesquisa foi realizada mediante a técnica de triangulação e da categorização analítica dos dados/informações. Verificou-se que o espaço das TS‟s na UFAM ainda não está consolidado, mas a emergência das primeiras ações de extensão no âmbito da TS são significativas e qualificadas, abrindo um espaço de discussão como forma de ultrapassar as barreiras entre o conhecimento científico e o popular na interação direta com as demandas sociais. Os princípios da sustentabilidade fundamentaram a análise da implantação da Tecnologia Social de Incubação e de Autogestão na ARPA, que visava a Organização do Trabalho dos Catadores de Material Reciclável, que mediante os entraves como a baixa escolaridade dos catadores, obteve baixa efetividade, entretanto teve avanços organizacionais pontuais diretamente relacionados com o trabalho da incubadora

Abstract:
The pressing need for increased sustainability in the energy system combined with underinvestment in new energy technology demonstrates the need for government initiatives to drive innovation in the energy sector. In this thesis, we have investigated how design characteristics of a government agency intended to support market focused energy technology innovation affect the projects in its portfolio, and consequently suggested considerations that should be made in the design of such an agency on the basis of our findings.Our focus is on the level of individual projects being supported by such programs. An embedded single-case research design was chosen, where the units of analysis were projects supported by the U.S. agency ARPA E. The primary source of data was semi-structured, open-ended interviews with representatives from 13 projects in California. Following our research, the following findings emerge: The innovation process of projects within early-stage, transformative high-risk energy technology is affected by design characteristics related to the program design, project selection process and the support provided by an active management model in the agency. The organizational structure of the agency and the quality of its managing officials are important moderators of these factors.As guidance to policy makers, we establish the order of importance of design considerations to be made in the creation of such an agency. This thesis contributes to theory by presenting an integrative framework of interlinked factors describing these effects, as well as a cohesive portfolio of rationales for government intervention in energy sector innovation.

Abstract:
Replication Protein A (RPA), the eukaryotic single-stranded DNA-binding complex, is essential for multiple processes in cellular DNA metabolism including, but not limited to, DNA replication, DNA repair and recombination. The `canonical' RPA is composed of three subunits (RPA1, RPA2, and RPA3). In addition to the three canonical subunits, there is a human homolog to the RPA2 subunit, termed RPA4, which can substitute for RPA2 in complex formation. The resulting RPA complex has been termed `alternative' RPA (aRPA). The normal function of aRPA is not known; however, previous studies have shown that it does not support S-phase progression in vivo. The goal of this thesis was to characterize the function of aRPA in DNA replication, DNA repair and recombination and profile its expression in human tissues. The studies presented in this thesis show that the aRPA complex has solution and DNA binding properties indistinguishable from the canonical RPA complex as determined by gel mobility shift assays. However, aRPA was unable to support DNA replication and inhibited canonical RPA function in a cell-free simian virus 40 system. aRPA inhibited both initiation and elongation of DNA synthesis in the SV40 system. Two regions of RPA4, the putative L34 loop and the C-terminal winged helix domain, were responsible for inhibiting SV40 DNA replication. The mechanism of SV40 DNA replication inhibition during initiation and elongation was characterized using assays for DNA polymerase α and DNA polymerase δ. aRPA was shown to have reduced interaction with DNA polymerase α and was not able to efficiently stimulate DNA synthesis by DNA polymerase α on aRPA coated single-stranded DNA. However, aRPA stimulated DNA synthesis by DNA polymerase δ in the presence of PCNA and RFC even though a reduced interaction was observed between aRPA and polymerase δ. The role of aRPA in DNA repair was also investigated. aRPA interacted with both Rad52 and Rad51 but had a reduced interaction with Rad51. However, aRPA was still able to stimulate Rad51-dependent strand exchange. aRPA also supported the dual incision/excision reaction of nucleotide excision repair. aRPA was less efficient in nucleotide excision repair than canonical RPA and this reduction was attributed to reduced interactions with the repair factor XPA. In contrast, aRPA exhibited higher affinity for damaged DNA than canonical RPA. The expression of RPA4 and RPA2 was determined by quantitative PCR in established cell lines, human normal tissues and human tumor tissue. RPA4 was shown to be expressed in all normal tissues examined but the level of expression was tissue specific. Additionally, RPA4 expression was decreased in all tumor tissues examined and was at the limit of detection in established cell lines. Taken together, the results presented in this thesis suggest that aRPA is a `non-proliferative' form of RPA that functions to maintain the genomic stability of non-dividing cells.
