Abstract:

Intervjuer med kunniga på AIS-området har utförts parallellt med en enkät riktad till aktiva befäl ute till sjöss. Detta i syfte att ta reda på dels hur detta hjälpmedel bör användas vad avser antikollision, dels hur det faktiskt används i praktiken.

AIS, Automatic Identification System, är ett informationsdelningssystem som hjälper fartyg att navigera säkert genom att sända ut egen navigationsdata och samtidigt ta emot motsvarande information från andra fartyg. Systemet nyttjas även som informationssystem för landbaserade stationer såsom VTS-centraler och kustbevakning.

Undersökningen har engagerat och visat att expertisen anser att AIS, i många avseenden, är ett bra och användbart hjälpmedel som bör komplettera övrig utrustning vid bedömning om kollisionsrisk. Vidare visar enkäten att ARPA-systemet, trots vissa inneboende brister, tillmäts större förtroende än AIS och AIS-vektorer används därför sparsamt. Enkäten visar även att en stor del av de aktiva inte är främmande för att göra avsteg från sjövägsreglerna emedan expertisen i huvudsak avråder från sådant.

Interviews with experts, in the field of AIS, have been conducted alongside a quantitative survey aimed at certified seagoing officers. The purpose of the survey was to investigate how this navigational aid preferably should be used in collision avoidance, in contrast to how it is used in reality.

AIS, Automatic Identification System, is an information sharing system to assist in safe navigation by transmitting the own vessels navigational data while, at the same time, receiving corresponding information from other ships. The system is also utilized by land based stations such as VTS-centers and coast guards.

The investigation shows that experts conclude AIS to be, in many aspects, a good and useful aid which should be used in collaboration with other equipment when assessing risk of collision. Furthermore, the survey show that the ARPA-system, in spite of some internal shortcomings, is awarded more trust than AIS and AIS vectors are therefore rarely used. The survey also shows that a large number of officers are open to deviating from the COLREGs while the experts, in general, advise against it.

Abstract:

Questa analisi è volta a esaminare le perturbazioni che, in date diverse (5/6 Febbraio 2015, 19-22 dicembre 2009, 1 febbraio 2012), hanno portato piovosità diffusa e nevicate intense nell'Emilia-Romagna. Il primo caso preso in esame, ossia quello relativo al periodo suddetto, ha rivestito grande importanza nello scenario dell’inverno scorso, in quanto ha provocato numerosi disagi alla viabilità, interruzione della distribuzione di acqua potabile e black-out elettrici nelle zone interessate da precipitazioni nevose; in Romagna invece sono stati numerosi gli allagamenti soprattutto nel Cesenate e nel Riminese. Nel secondo caso analizzato, invece, assistiamo al fenomeno non troppo comune del gelicidio. Provoca, infatti, anch’esso disagi gravi alla viabilità stradale e autostradale, nonché il pericolo di interruzione della corrente nelle linee elettriche, dovuta al peso del ghiaccio depositato sui conduttori che fa crollare i cavi dell’alta tensione. L’ultimo caso, infine, è stato trattato poiché rientrante nella famosa ondata di gelo della prima metà del Febbraio 2012. Vedremo, infatti, un quadro generale dell’ondata, dopodiché un “focus” sulle importanti nevicate del primo Febbraio in regione.

A crise ambiental decorrente da lógica produtiva capitalista, que foi materializada nas atitudes destrutivas do homem em relação ao ambiente, implicou a emergência da discussão sobre a Tecnologia Social (TS) como uma alternativa sustentável de desenvolvimento, em contraste à Tecnologia Convencional (TC) que é funcional ao sistema capitalista. Assim, para que os alicerces da TS sejam fundamentados e que sua legitimidade seja alcançada perante a sociedade, a sua interlocução com a universidade, principalmente no âmbito da extensão, é indubitavelmente relevante. Para tanto, investigaram-se os impactos socioambientais do uso da TS na ação de extensão do Programa UNITRABALHO da Universidade Federal do Amazonas (UFAM) na Associação de Reciclagem e Proteção Ambiental (ARPA) e de ações de extensão de programas e projetos da Pró-Reitoria de Extensão e Interiorização/ UFAM. Portanto, a pesquisa teve como principais objetivos: desvelar o processo de institucionalização do desenvolvimento a partir das principais características, ações, resultados e/ou produtos; analisar os impactos socioambientais do uso da TS nas ações de extensão no locus da pesquisa, a ARPA; e verificar se os impactos do uso da TS contribuíram para a sustentabilidade da ARPA. No percurso metodológico da pesquisa utilizou-se a abordagem quanti-qualitativa para coleta de dados/informações, por meio da aplicação de questionário junto a 08 (oito) coordenadores de projetos e programas de extensão que participaram do I Fórum de Tecnologia Social da UFAM; e por meio da realização de entrevistas semi-estruturadas junto a 01 (uma) coordenadora da UNITRABALHO e aos 08 (oito) catadores de material reciclável. A análise dos resultados da pesquisa foi realizada mediante a técnica de triangulação e da categorização analítica dos dados/informações. Verificou-se que o espaço das TS‟s na UFAM ainda não está consolidado, mas a emergência das primeiras ações de extensão no âmbito da TS são significativas e qualificadas, abrindo um espaço de discussão como forma de ultrapassar as barreiras entre o conhecimento científico e o popular na interação direta com as demandas sociais. Os princípios da sustentabilidade fundamentaram a análise da implantação da Tecnologia Social de Incubação e de Autogestão na ARPA, que visava a Organização do Trabalho dos Catadores de Material Reciclável, que mediante os entraves como a baixa escolaridade dos catadores, obteve baixa efetividade, entretanto teve avanços organizacionais pontuais diretamente relacionados com o trabalho da incubadora

Abstract:

The pressing need for increased sustainability in the energy system combined with underinvestment in new energy technology demonstrates the need for government initiatives to drive innovation in the energy sector. In this thesis, we have investigated how design characteristics of a government agency intended to support market focused energy technology innovation affect the projects in its portfolio, and consequently suggested considerations that should be made in the design of such an agency on the basis of our findings.Our focus is on the level of individual projects being supported by such programs. An embedded single-case research design was chosen, where the units of analysis were projects supported by the U.S. agency ARPA E. The primary source of data was semi-structured, open-ended interviews with representatives from 13 projects in California. Following our research, the following findings emerge: The innovation process of projects within early-stage, transformative high-risk energy technology is affected by design characteristics related to the program design, project selection process and the support provided by an active management model in the agency. The organizational structure of the agency and the quality of its managing officials are important moderators of these factors.As guidance to policy makers, we establish the order of importance of design considerations to be made in the creation of such an agency. This thesis contributes to theory by presenting an integrative framework of interlinked factors describing these effects, as well as a cohesive portfolio of rationales for government intervention in energy sector innovation.

Abstract:

Replication Protein A (RPA), the eukaryotic single-stranded DNA-binding complex, is essential for multiple processes in cellular DNA metabolism including, but not limited to, DNA replication, DNA repair and recombination. The `canonical' RPA is composed of three subunits (RPA1, RPA2, and RPA3). In addition to the three canonical subunits, there is a human homolog to the RPA2 subunit, termed RPA4, which can substitute for RPA2 in complex formation. The resulting RPA complex has been termed `alternative' RPA (aRPA). The normal function of aRPA is not known; however, previous studies have shown that it does not support S-phase progression in vivo. The goal of this thesis was to characterize the function of aRPA in DNA replication, DNA repair and recombination and profile its expression in human tissues. The studies presented in this thesis show that the aRPA complex has solution and DNA binding properties indistinguishable from the canonical RPA complex as determined by gel mobility shift assays. However, aRPA was unable to support DNA replication and inhibited canonical RPA function in a cell-free simian virus 40 system. aRPA inhibited both initiation and elongation of DNA synthesis in the SV40 system. Two regions of RPA4, the putative L34 loop and the C-terminal winged helix domain, were responsible for inhibiting SV40 DNA replication. The mechanism of SV40 DNA replication inhibition during initiation and elongation was characterized using assays for DNA polymerase α and DNA polymerase δ. aRPA was shown to have reduced interaction with DNA polymerase α and was not able to efficiently stimulate DNA synthesis by DNA polymerase α on aRPA coated single-stranded DNA. However, aRPA stimulated DNA synthesis by DNA polymerase δ in the presence of PCNA and RFC even though a reduced interaction was observed between aRPA and polymerase δ. The role of aRPA in DNA repair was also investigated. aRPA interacted with both Rad52 and Rad51 but had a reduced interaction with Rad51. However, aRPA was still able to stimulate Rad51-dependent strand exchange. aRPA also supported the dual incision/excision reaction of nucleotide excision repair. aRPA was less efficient in nucleotide excision repair than canonical RPA and this reduction was attributed to reduced interactions with the repair factor XPA. In contrast, aRPA exhibited higher affinity for damaged DNA than canonical RPA. The expression of RPA4 and RPA2 was determined by quantitative PCR in established cell lines, human normal tissues and human tumor tissue. RPA4 was shown to be expressed in all normal tissues examined but the level of expression was tissue specific. Additionally, RPA4 expression was decreased in all tumor tissues examined and was at the limit of detection in established cell lines. Taken together, the results presented in this thesis suggest that aRPA is a `non-proliferative' form of RPA that functions to maintain the genomic stability of non-dividing cells.

Gli Open Data sono un'utile strumento che sta via via assumendo sempre più importanza nella società; in questa tesi vedremo la loro utilità attraverso la realizzazione di un'applicazione mobile, che utilizza questi dati per fornire informazioni circa lo stato ambientale dell'aria e dei pollini in Emilia Romagna, sfruttando i dataset forniti da un noto ente pubblico (Arpa Emilia Romagna). Tale applicazione mobile si basa su un Web Service che gestisce i vari passaggi dei dati e li immagazzina in un database Mongodb. Tale Web Service è stato creato per essere a sua volta messo a disposizione di programmatori, enti o persone comuni per studi e sviluppi futuri in tale ambito.

Abstract:

La méthode de conductancemétrie indirecte est évaluée comme une méthode de détection et de quantification de gaz produits lors de la croissance des micro-organismes. Après avoir souligné la place de l'impédancemétrie parmi les techniques rapides en microbiologie, le travail aborde dans un premier temps la définition des paramètres d'un nouvel appareil : (Analyse Rapide des Produits Appertisés). Dans un deuxième temps, la réponse impédancemétrique lors de la croissance bactérienne est étudiée pour différentes conditions et met de souligner les qualités de répétabilité et de linnéarité de la technique. L’appareil répond parfaitement aux critères des méthode alternatives ou rapides. La méthode est effectivement automatisable et l'appareil est apte à détecter la croissance des micro-organismes, et cela quel que soit le type de milieu considéré. L'appareil s'avère donc être un système de mesure d'activité globale aspécifique, dans le sens où il est indépendant des matrices et des micro-organismes recherchés, ce qui en fait son atout principal, et ce qui confère à la méthode une grande souplesse d'utilisation et une standardisation potentielle. A ce stade, nous avons réussi à concevoir un appareil capable de mesurer l'activité et/ou la croissance de micro-organismes. Les objectifs initiaux du contrat sont donc pleinement atteints d'une et mettent d'envisager la production de cet appareil dans un futur très proche. Il est raisonnable d'envisager que l'appareil ARPA pourra être dédié à des utilisations multiples, tant en routine (contrôle qualité des produits alimentaires, pharmaceutiques et cosmétiques, sans oublier le domaine de l'environnement) qu'au laboratoire (recherche fondamentale et développement). Sa simplicité d'utilisation et ses performances permettront d'élargir en permanence les champs d'application de cette technologie.

Abstract:

Ce travail de thèse s'inscrit dans le cadre des recherches du Centre de Recherches sur les Identités Nationales et l'Interculturalité de Nantes (CRINI) et propose une analyse de deux extraits de romans de l'auteur cubain Alejo Carpentier à deux époques différentes de sa production littéraire (1953 et 1979). Il s'agit de déceler les différents mécanismes discursifs mis en œuvre dans la construction de l'image de l'Europe dans le cadre des théories actuelles de l'Analyse de Discours. Cette recherche est élargie par l'examen des mécanismes pragmatico- discursifs qui participent également à la construction de cette image en fonction des théories de l'Analyse Linguistique du Discours et des recherches menées au sein du Groupe d'Analyse Sémantique et pragmatique de Nantes (GRASP-CALD) depuis 1999. Pour cela nous proposons l'analyse de deux termes de notre corpus : la « creacion dans Los pasos perdidos et le mot « oro » dans El arpa y lasombra. Nous espérons pouvoir mettre nos conclusions au service d'une approche sociolinguistique de la littérature hispano-américaine.

Abstract:

I detta examensarbete har jag undersökt förmågan hos nautiker att ta fram och tillgodogöra sig information från radar och ARPA för sitt beslutsunderlag i trafiksituationer. Syftet var att få förståelse i om fartygsbefäl haft svårigheter att få fram denna mycket relevanta information. Detta gjordes med hjälp av förstudier i form av intervjuer med erfarna fartygsbefäl. Dessa intervjuer gav kriterier som möjliggjorde analyseringen av 13 olycksfallsrapporter, vilket utgjorde litteraturstudien. Dessa rapporter var författade av engelska Department of Transport, Marine Investigation Branch(MAIB), Statenshaverikommission(SHK) och Sjöfartsverkets rapportserie(SjöfR). Dessutom användes både sjö- och luftfartsrelaterade artiklar för framtagning av fakta. Slutsatserna visade att man i hälften av fallen använt sig av väldigt låg nivå på funktionalitet för sitt beslutsunderlag, men att även i de fall man använt mer avancerad funktionalitet fann man brister i tolkning av informationen för att i tid ta beslut för att undvika olyckor.
This is a study about the Navigating Mariners ability to locate and use information from the Radar and ARPA, before taking important decisions in Trafic Situations. The purpose was to acquire an understanding if there are some difficulties in receiving this important information. This purpose was accomplished by a pre-investigation using two interviews with experienced deck officers. This pre-study produced criterias, that made it possible to compare 13 Accident Reports. These reports were used as a litterature for the investigation. The Department of Transport, Marine Investigation Branch(MAIB), The Swedish Accident Investigation Board(SHK) and The Swedish Maritime Safety Inspectorate(SjöfR) had produced these reports. In addition to these reports, the author used both maritime- and aeronautical based articles in order to find more facts about the issues around the investigation. The findings showed that in half of the investigated accidents the mariners involved had used a low level of functionality before making the final decisions. It was also shown, that the ones that had used a very high level of functionality, before the accident, misinterpreted the information and failed to take the right decision in ample time before the accident.

Abstract:

GOCE was launched in 2009 at 250 km altitude to recover Earth’s static gravity field. As part of the GOCE-Italy project, we carried out the precise modeling for the radiation pressure and the aerodynamic effects on this satellite. This analysis has been performed to reduce the mismodeling of the non-gravitational forces, in order to be able to estimate the ocean tides parameters from the LEO satellites orbital perturbation. A new software ARPA (Aerodynamics and Radiation Pressure Analysis), which takes advantage of the raytracing technique, has been designed and developed to accurately model the non-gravitational perturbations. ARPA can compute the Solar Radiation Pressure (SRP), Earth Radiation Pressure (ERP), the spacecraft Thermal Re-Radiation (TRR) and the aerodynamic forces and torques acting on any satellite with a high level of accuracy. The adopted methodologies and procedure are presented in this thesis, and the results of the tests on GOCE are illustrated and discussed. The NAPEOS (NAvigation Package for Earth Observation Satellites) software, developed and maintained at ESA/ESOC, was upgraded to make use of the new ARPA inputs and adopted to perform the tests on GOCE. The tests were performed on 30 consecutive daily arcs, starting at the beginning of the GOCE science phase on 1st November 2009. The results for the radiation test cases show a significant reduction of the empirical accelerations, especially in the cross-track direction, of about the 20% for the SRP, 12% for the ERP albedo, 13% for the ERP infrared and 20% for the TRR with respect to the standard NAPEOS force modeling (cannon-ball). For the aerodynamics, an important reduction of the post-fit RMS from 7.6 to 7.3 mm has been observed with the new ARPA model, and the a reduction from 4.6 to 4.2 cm of the distance of the orbits computed with ARPA from the official reduced-dynamics GOCE orbits (Precise Science Orbit) has been computed. The obtained results confirm the goodness of the modeling and techniques of ARPA for all the non-gravitational perturbations computed for GOCE. Even though the results are presented for the GOCE satellite, the new technique and software are adaptable to satellite of any shape, whether in Earth-bound orbit, or orbiting another planet, or cruising in interplanetary space.
GOCE è stato lanciato nel 2009, ad un'altezza di 250 km per recuperare il campo gravitazionale statico terrestre. Nell'ambito del progetto GOCE-Italy, abbiamo effettuato la modellazione precisa per la pressione di radiazione e gli effetti aerodinamici su questo satellite. Questa analisi è stata compiuta per ridurre il mismodeling delle forze non gravitazionali, per poter stimare i parametri di marea tramite perturbazioni orbitali dei satelliti LEO. Un nuovo software ARPA (Analisi Aerodinamica e di Pressione di Radiazione), che sfrutta la tecnica del raytracing, è stato progettato e sviluppato per modellare accuratamente le forze e le coppie che agiscono su un satellite, dovute alle perturbazioni non gravitazionali. ARPA può calcolare la pressione di radiazione solare (SRP), la pressione di radiazione terrestre, la pressione di radiazione termica emessa dal satellite (TRR ) con un alto livello di precisione. Le metodologie e le procedure adottate sono presentate in questa tesi, ed i risultati dei test su GOCE sono illustrati e discussi. Il software NAPEOS (NAvigation Package for Earth Observation Satellites), sviluppato e mantenuto a ESA/ESOC, è stato aggiornato per poter usufruire dei nuovi input ARPA ed è stato adottato per eseguire i test su GOCE. I test sono stati effettuati su 30 archi giornalieri consecutivi, a partire dall'inizio della fase scientifica di GOCE, il 1 Novembre 2009. I risultati per i test di radiazione mostrano una significativa riduzione delle accelerazioni empiriche, soprattutto nella direzione trasversale (cross-track), di circa il 20% per la componente di SRP, 12% per l' albedo ERP, 13% per l' infrarosso ERP e 20% per la TRR, rispetto ai modelli standard di NAPEOS (modello a palla di cannone). Per l'aerodinamica, è stata osservata una riduzione significative del post-fit RMS da 7.6 7.3 mm con il nuovo modello ARPA, e una riduzione da 4.6 a 4.2 cm della distanza delle orbite calcolate con ARPA dalle orbite ufficiali a dinamica ridotta di GOCE (Precise Science Orbit). I risultati ottenuti confermano la bontà della modellazione e delle tecniche di ARPA per tutte le perturbazioni non gravitazionali calcolate per GOCE. Anche se i risultati sono presentati per il satellite GOCE, la nuova tecnica e il nuovo software sono adattabili a satelliti di qualsiasi forma, sia in orbita attorno alla Terra, o in orbita intorno a un altro pianeta, o in crociera nello spazio interplanetario.
