Full textABB Power Grids Grid Integration, PGGI, levererar transformatorstationer till externa kunder, arbetet som utförs är i projektform. Alla projekt följs upp via nyckeltal, nyckeltalet OTD som ska undersökas i detta examensarbete är i form av 17 stycken standardiserade milstolpar satta från ABB för att stötta PGGI-projektens leveranssäkerhet. Problemet som ska redas ut är den nedåtgående trenden som upptäckts i leveranssäkerheten hos de 17 milstolparna. För att ta an detta gjordes en nulägesbeskrivning samt en börlägesbeskrivning, detta visade ett gap i arbetssättet gällande de 17 milstolparna. En analys av gapet mellan nuläge och börläge genomfördes, i analysen upptäcktes brister i kunskap, förståelse, rapportering samt opålitlig data gällande de 17 milstolparna. Därefter utvecklades en plan för att föreslå hur gapet mellan börläge och nuläge ska minska för att sedan elimineras. Lean filosofi, Lean verktyg samt Statistisk Process Styrning låg som grund för de framtagna förbättringsförslagen. Den framtagna planen är tänkt att användas som grund för vidare arbete med förbättringsförslagen som tagits fram.

nmez May 2010, 205 pages This thesis investigates how two loosely autobiographical works unveil the effects of colonization on their major characters in terms of their identities and senses of belonging. The Grass Is Singing by Doris Lessing, a second-generation member of the colonizer, and My Place by Sally Morgan, a third-generation hybrid Australian Aborigine, are selected because both novels essentially deal with colonial issues by depicting their major characters in a process of maturation within a colonial and post-colonial framework, the former using a semi-autobiographical narrative tone and the latter using an Aboriginal version of autobiography, which integrates oral tradition and storytelling. These two books reveal that a sense of identity is closely related to a sense of belonging and that both are fundamentally affected by the colonial situation. The effects of a sense of identity and a sense of belonging, which boil down to the demise or survival of the individual, interacts with family and society, physical environment, and race issues that the thesis investigates by dedicating a chapter to each. The method used in this point-by-point comparative analysis is to approach the issues of sense of belonging and identity in a colonial context with a close reading of the two works, to find out what the texts say for themselves regarding the effect of family and society, environment, and race as depicted in The Grass Is Singing and My Place. The theoretical background that is most relevant to this study is post-colonial literary theory, although here it is taken as secondary to the close reading that is the thesis&rsquo

FAPEMIG - Fundação de Amparo à Pesquisa do Estado de Minas Gerais
O objetivo desta dissertação é investigar as práticas discursivas de três professoras que se relacionaram direta e indiretamente com as Tecnologias da Informação e Comunicação (TICs) no processo de ensino-aprendizagem. Para cumprir com esta proposta, adotamos o modelo de Multiletramentos Digitais, de Selber (2004), que congrega as várias dimensões de Letramento(s) Digital(is) e tenta explicá-los razoavelmente através de três categorias principais: Letramento Digital Funcional; Letramento Digital Crítico e Letramento Digital Retórico. Além disso, para melhor investigar as práticas discursivas das professoras pesquisadas, encontramos suporte na Teoria do Posicionamento, de acordo com a qual nós disponibilizamos lugares discursivos para nós mesmos e para os outros dentro de uma ordem moral que estabelecemos durante uma troca simbólica, construindo pontos de vista específicos através de posicionamentos (cf. HARRÉ e VAN LANGENHOVE, 1999). Metodologicamente, esta pesquisa se estrutura como um estudo de caso etnográfico (ANDRÉ, 1995; YIN, 2001; VIÉGAS, 2007), associado a uma (micro) análise do discurso (GEE et al., 1992; SACKS et al., 1974, 2005; DAVIES e HARRÉ, 1990). O contexto investigado é uma escola pública da rede municipal de ensino de Juiz de Fora-MG, localizada em um bairro periférico da cidade. As participantes são três professoras que se relacionaram direta e indiretamente com as TICs no ensino, a saber: a professora de Informática Educacional, a professora regente de turma (4º ano do Ensino Fundamental) e a professora responsável pela biblioteca escolar. Para geração de dados, utilizamos questionários, notas de campo, observação e entrevistas semi-estruturadas. O corpus de pesquisa constitui-se principalmente das transcrições das entrevistas realizadas com as participantes. Os resultados do estudo indicam que, embora as professoras tenham um posicionamento favorável ao uso das TICs no ensino, suas experiências pessoais e profissionais com tais tecnologias acabam influenciando em diferentes níveis de engajamento quanto ao projeto de inserção das TICs no processo de ensino-aprendizagem. A análise dos dados também indica um predomínio do modelo de Letramento Digital Funcional na escola investigada, o que aponta para a necessidade de se adotar perspectivas mais críticas de Letramento(s) Digital(is) no contexto educacional.
The aim of this thesis is to investigate the discursive practices of three teachers that have kept direct and indirect relationships with Information and Communication Technologies (ICT‟s) in the teaching-learning process. To accomplish with this purpose, we have adopted as theoretical framework the model of Digital Multiliteracies proposed by Selber (2004), which considers the multilayered dimensions of Digital Literacies and tries to explain them reasonably through three main levels of computer literacy: Functional Literacy, Critical Literacy, and Rhetorical Literacy. In addition, to better investigate the discursive practices of the teachers surveyed, we also have found support on Positioning Theory, according to which we provide discursive locations for ourselves and the others within a moral order that we establish during a symbolic exchange, building specific points of view inside the positions we take on (Harré and Van Langenhove, 1999). Methodologically, this research is structured as an ethnographic case study (André, 1995; Yin, 2001; Viégas, 2007) associated to a discourse (micro) analysis (Gee et al., 1992; Sacks et al., 1974, 2005; Davies and Harré, 1990). The context of this qualitative research is a public school pertaining to the municipal education system of Juiz de Fora – Minas Gerais. This school is located in a suburbian area of the city. The participants are three teachers that have kept direct and indirect relationships with ICT‟s in education, namely: the teacher of Educational Informatics, the teacher of a classroom (4th year of Brazilian elementary school) and the teacher responsible for the school library. To generate the data, we have used questionnaires, field notes, observation and semi-structured interviews. The corpus of this research consists mainly of the transcripts from interviews made with the participants. The results of this study point out that although the teachers have taken on favorable positions regarding the use of ICT‟s in education, their personal and professional experiences with such technologies tend to influence at different levels of commitment to the project of insertion of ICT‟s in the teaching-learning process. The data analysis also evinces a predominance of Functional Digital Literacy in the school investigated, which indicates the necessity for adoption of more critical perspectives regarding Digital Literacies in educational settings.

The text brings the analysis of aspects of the literary discourse as the processes of construction of the scenes and the magical universe, in which the narratives of Marina Colasanti are realized, having as it shows the tales of the works Doze reis e a moca no labirinto do vento (2006): "The woman ramada", Uma ideia toda azul (2006): "Beyond the frame", "Between the leaves of green ó" and "Yarn after yarn". The general and specific objectives are to highlight and distinguish the encompassing and generic scenes present in the narratives, to identify the nature of the verbal sign in its relation to the nonverbal sign, and to analyze intertext resources, paratext, among others, as an artistic procedure. The narrative plans are approached, in which the characters are realized mimically, starting from the initial assumption formulated by Dominique Maingueneau. Non-verbal language is an invitation to read verbal language and vice versa. Both are associated with the signs that are constructed through the textual writing: loom and fabric. They can be seen now either explicitly or implicitly, and put in the service of a power that is realized by the act of reading. Thus, in the narrative text, there are traces of a speech in which the text is staged.
O texto traz a análise de aspectos do discurso literário como os processos de construção das cenas e o universo mágico, em que se realizam as narrativas de Marina Colasanti, tendo como mostra os contos das obras Doze reis e a moca no labirinto do vento (2006): “A mulher ramada”, Uma ideia toda azul (2006): “Além do bastidor”, “Entre as folhas do verde ó” e “Fio após fio”. Os objetivos geral e específicos são destacar e distinguir as cenas englobante e genérica presentes nas narrativas, identificar a natureza do signo verbal na sua relação com o signo não verbal e analisar recursos de intertexto, paratexto, entre outros, como procedimento artístico. Abordam-se os planos narrativos, nos quais se dá a realização dos personagens mimeticamente, partindo do pressuposto inicial formulado por Dominique Maingueneau. A linguagem não verbal é um convite à leitura da linguagem verbal e vice-versa. Ambas se associam aos signos que se constroem por meio da escritura textual: tear e tecido. Elas podem ser vistas ora de modo explícito, ora implícito, e se colocam a serviço de um poder que se realiza pelo ato de leitura. Assim, no texto narrativo, há rastros de um discurso em que o texto é encenado.

Full textThe Human Immunodeficiency Virus (HIV) is the etiologic agent of Acquired Immunodeficiency Syndrome (AIDS). AIDS is a disease which has a global distribution, and it is estimated that there are currently at least 36.9 million people infected with the virus. During the replication cycle, HIV promotes several changes in the physiology of the host cell to promote their survival and enhance replication. The fast progression of HIV-1 in humans and animal models is closely linked to the function of an accessory protein Nef. Among several actions of Nef, one is the most important is the down-regulation of proteins from the immune response, such as the CD4 receptor. It is known that this action causes CD4 degradation in lysosome, but the molecular mechanisms are still incompletely understood. Nef forms a tripartite complex with the cytosolic tail of the CD4 and adapter protein 2 (AP-2) in clathrin-coated vesicles, inducing CD4 internalization and lysosome degradation. Previous research has demonstrated that CD4 target to lysosomes by Nef involves targeting of this receptor to multivesicular bodies (MVBs) pathway by an atypical mechanism because, although not need charging ubiquitination, depends on the proteins from ESCRTs (Endosomal Sorting Complexes Required for Transport) machinery and the action of Alix, an accessory protein ESCRT machinery. It has been reported that Nef interacts with subunits of AP- 1, AP-2, AP-3 complexes and Nef does not appear to interact with AP-4 and AP-5 subunits. However, the role of Nef interaction with AP-1 or AP-3 in CD4 down-regulation is poorly understood. Furthermore, AP-1, AP-2 and AP-3 are potentially heterogeneous due to the existence of multiple subunits isoforms encoded by different genes. However, there are few studies to demonstrate if the different combinations of APs isoforms are form and if they have distinct functional properties. This study aim to identify and characterize cellular factors involved on CD4 down-modulation induced by Nef from HIV-1. More specifically, this study aimed to characterize the involvement of AP-1 complex in the down-regulation of CD4 by Nef HIV-1 through the functional study of the two isoforms of ?-adaptins, AP-1 subunits. By pull-down technique, we showed that Nef is able to interact with ?2. In addition, our data from immunoblots indicated that ?2- adaptin, not ?1-adaptin, is required in Nef-mediated targeting of CD4 to lysosomes and the ?2 participation in this process is conserved by Nef from different viral strains. Furthermore, by flow cytometry assay, ?2 depletion, but not ?1 depletion, compromises the reduction of surface CD4 levels induced by Nef. Immunofluorescence microscopy analysis also revealed that ?2 depletion impairs the redistribution of CD4 by Nef to juxtanuclear region, resulting in CD4 accumulation in primary endosomes. Knockdown of ?1A, another subunit of AP-1, resulted in decreased cellular levels of ?1 and ?2 and, compromising the efficient CD4 degradation by Nef. Moreover, upon artificially stabilizing ESCRT-I in early endosomes, via overexpression of HRS, internalized CD4 accumulates in enlarged HRS-GFP positive endosomes, where co-localize with ?2. Together, the results indicate that ?2-adaptin is a molecule that is essential for CD4 targeting by Nef to ESCRT/MVB pathway, being an important protein in the endo-lysosomal system. Furthermore, the results indicate that ?-adaptins isoforms not only have different functions, but also seem to compose AP-1 complex with distinct cell functions, and only the AP-1 variant comprising ?2, but not ?1, acts in the CD4 down-regulation induced by Nef. These studies contribute to a better understanding on the molecular mechanisms involved in Nef activities, which may also help to improve the understanding of the HIV pathogenesis and the related syndrome. In addition, this work contributes with the understanding of primordial process regulation on intracellular trafficking of transmembrane proteins.

Full textNeoadjuvant chemotherapy (NAC i.e. chemotherapy before surgery) is increasingly being used for aggressive or locally advanced breast cancer (BCs). Beyond clinical benefits, it represents an opportunity to monitor in vivo sensitivity to treatment. Based on the analysis of datasets of BCs patients treated with NAC, we aimed at identifying mechanisms associated with resistance or sensitivity to treatment.In the first part, we evaluated biological, clinical, pathological and transcriptomic patterns. We demonstrated that unexplored pathological features such as post-NAC lymphovascular invasion may carried an important prognostic information.In a second part, we analyzed impact of imune infiltration in BC and we described extensively the changes of tumor infiltrating lymphocytes (TILs) between pre and post-NAC samples. We showed that the prognostic impact of TILs was different before and after NAC, and was opposite in TNBC and HER2-positive BCs. Finally, we investigated the impact of comedications use during NAC. We found both positive effects - while enhancing immune infiltration and response to treatment - and negative effects with deleterisous oncologic outcomes in specific patients subgroups. In conclusion, the neoadjuvant setting represents a platform to both generate and potentially validate research hypotheses aiming at increasing the efficacy of treatment. The public release of real-life datasets of BC patients treated with NAC would represent a major resource to accelerate BC research

Esta dissertação tem por objetivo compreender como as transformações no contexto da defesa sanitária e inspeção agropecuária entre 1996 e 2014 se relacionam com as configurações identitárias do Idaf manifestadas por gestores nesse contexto. Trata-se de uma pesquisa qualitativa que descreve os processos identitários da organização por um olhar interpretativista, pelo entendimento das relações simbólicas que permeiam essa organização, observados em discursos dos sujeitos. Para coleta dos dados foram utilizadas pesquisa documental e entrevistas semiestruturadas individuais. As entrevistas foram realizadas com cinco sujeitos da organização nos meses de setembro e outubro de 2014. O método de análise foi a Análise de Conteúdo (KRIPPENDORFF, 1990) que busca investigar dados textuais através de técnicas que permitam compreendê-los com seus significados, referências, valorações e intenções. Pretendeu-se contribuir, com esse estudo, para a compreensão da identidade organizacional, em uma organização pública, no contexto da defesa sanitária e inspeção agropecuária. Os resultados permitiram identificar o Idaf como único e distinto de outras organizações e como essa identificação está relacionada às transformações do contexto agropecuário.
This research aims to understand how changes in the context of health protection and agriculture inspection between 1996 and 2014 relate to the Idaf identity configurations expressed by managers in this context. It is a qualitative research that seek to describe the identity processes of the organization by an interpretive look at understanding the symbolic relations that permeate this organization, observed in speeches of the subjects. For data collection were used desk research and semi-structured individual interviews. Interviews were conducted with five subjects of the organization in the months of September and October 2014. The analysis method was content analysis (KRIPPENDORFF, 1990) that investigates textual data through techniques to understand them with their meanings, references , valuations and intentions. It was intended to contribute to this study, for understanding the organizational identity in a public organization in the context of health protection and agricultural inspection. The results showed the Idaf as unique and distinct from other organizations and how this identification is related to change in agricultural context.

Full textPulmonary rehabilitation (PR) is recommended in the management of subjects with ventilatory impairment to improve their quality of life. Although a large body of evidence support its use, only few subjects benefit from it and the optimal training modality has not been determined yet. In this context, the use of new and existing tools to optimize access as well as the effects of the program are major developments that deserve to be studied. As part of this thesis, we sought to explore these two major issues (1) by considering a rehabilitation model relocated outside the PR centres while assessing the obstacles to this model and (2) exploring the effectiveness of different add-on to PR in further optimizing the benefits of the program. In the first part, we have shown, through several retrospective studies and an original prospective multicentre contribution, that the six-minute stepper test can be used to prescribe endurance training, particularly for those patients with a mild to moderate chronic obstructive pulmonary disease (COPD), but not to prescribe muscle strengthening. Furthermore, we have shown in a cohort of 105 subjects referred for PR that the use of a remote tele monitoring device was feasible, valid and widely accepted. Finally, we explored the prevalence of cognitive dysfunction, another systemic impairment of COPD that could compromise the relocation of the program, and showed that it was a very common condition (around 75% of the subjects) but that it could improve following PR and did not seem to influence the use of a remote tele monitoring device. In the second part, we evaluated the effects of different add-on used to potentiate the benefits of the PR program. In a cross-over study of 21 COPD patients, we showed that non-invasive ventilation did not improve endurance exercise capacity due to technological limitation of the ventilator. Through a multicentre randomized controlled study carried out in 73 patients with severe to very severe COPD, we have shown that neuromuscular electrical stimulation at home, performed in addition to a PR program, did not provide further benefits on quality of life or exercise capacity. Finally, through a randomized cross-over double-blind study carried out in 10 patients, we were unable to show the effectiveness of transcutaneous nerve electrical stimulation in improving their endurance exercise capacity. Finally, in a last part, we highlighted the research currently carried out in our laboratory following the original contributions described during this thesis, as well as new area of research in order to pursue the themes explored. Thus, two systematic reviews and meta-analysis (the first about nasal high flow therapy in subjects with stable COPD and the second about the use of advanced telehealth technologies to deliver PR) will serve as a basis for future research

Full textOur goal was to optimize the management of severe acute pain in emergency medicine. Intravenous morphine titration, which is currently the referent method, must be modernised to meet the new challenges of practice while at the same time, we must keep guaranteeing an unchanged risk-benefit balance in the fight against oligoanalgesia. Our work, described in this thesis, has been a cornerstone for the evaluation of a nebulized alternative solution to emergencies through 3 clinical studies, (AEROMORPH1, CLIN-AEROMORPH, EPIMORPH), and study of its contextualisation in literature. Work in healthy volunteers allowed us to establish a simple and accessible procedure for aerosol, of short duration (5 min), which can be repeated in titration procedures (every 10 min). Its feasibility is likely to be confirmed on our multicentre clinical study at a large scale and its efficacy, by a non-inferiority design of study is being evaluated. Pharmacological data in healthy volunteers and in patients confirm a blood morphine concentration close to the effective blood concentrations observed by intravenous administration (CLIN-AEROMORPH: 1-20 ng/ml), which is already a positive result demonstrated by our work. Moreover, regarding eligibility, our observational study, combined to a experiment about decision mechanisms, combined with the analysis of medico-economic data in literature, suggest the need to base its indication on more than just the simple VAS/NRS assessment at triage. In this work we showed that emergency practitioners’ practice today underlines their lack of adherence to the systematic initiation of intravenous morphine titration by patient self-assessment of severe pain (compliance with SFMU criteria 6 to 20%, re-assessment of VAS/NRS scores 61%). If nebulized morphine titration is only proposed as a starter for morphine titration without modernising the algorithms for opiate prescription decision in emergency protocols, it is likely that this new therapeutic proposal will only solve part of the current problem. Targeted pharmacological management, as individualised as possible, is more relevant than ever, with prescription of morphine titration according to the patient's typology, in addition to prioritisation by pain typology. By developing an original pain model in healthy volunteers, we have also highlighted profiles of "pain sensitive" and "enduring" patients, according to neurophysiological and biochemical data, that are certainly represented in daily clinical practice. Therefore, all these components should be taken into account to improve pain management in emergency medicine, with a more systemic vision and more dedicated studies using innovative evaluation methods, combining robust quantitative criteria with comprehensive qualitative criteria

Full textEste trabalho propõe analisar o fenômeno da terceirização desenvolvido ao longo das duas últimas décadas em Bio-Manguinhos. Trata-se de um estudo de caso realizado na unidade de produção de vacinas e reagentes para diagnóstico da Fundação Oswaldo Cruz. Inicialmente foi realizado um estudo de cenário, analisando a política de gestão do trabalho desenvolvida pelo Estado brasileiro para as instituições públicas. A seguir, a análise passou a verificar a inserção da Fiocruz nessas políticas, desde a sua criação e no decorrer de vigência de diferentes formas de contratação, alternando da extrema rigidez para a total flexibilidade sempre com foco na gestão do trabalho, e, como essas políticas se refletiram no desempenho de Bio-Manguinhos. Baseado no contexto da conformação do Estado e na análise do desempenho da unidade, o trabalho critica a falta de planejamento e de prospecção dessas políticas, que favoreça a inovação de processos, produtos e procedimentos e o desempenho pleno de uma unidade de produção de insumos para a saúde, tão necessários ao atendimento das necessidades da população brasileira. Critica também, a ausência de um modelo de Estado consolidado que sirva de arcabouço à formulação dessas políticas. Devido às fortes críticas dos órgãos de controle acerca da extrapolação dos limites da terceirização praticada nas últimas duas décadas e à falta de definição clara desses marcos legais, o trabalho apresenta uma metodologia baseada nas atribuições dos cargos do plano de carreiras da Fiocruz e culmina com a apresentação de uma matriz de atribuições passíveis de serem realizadas por contratação indireta. Conclui pela necessidade de convivência de dois quadros de trabalhadores: um permanente formado por servidores, estáveis e de carreira e outro flexível, executado por contratação indireta, de caráter eventual, temporário ou de apoio às atividades relacionadas à missão de Bio-Manguinhos. O trabalho ainda sugere a aplicação da mesma metodologia às demais unidades da Fiocruz e que a instituição afirme, frente aos órgãos de controle, quais atividades que ela precisa manter no quadro de servidores permanente e quais ela quer delegar a terceiros sem contudo, ferir a legislação vigente. Por fim, constata-se que a metodologia apresentada ameniza, mas não resolve o problema, dessa forma, recomenda-se à Fiocruz buscar mecanismos que altere o modelo de gestão pelo qual está submetida a fim de viabilizar as duas formas de incorporação de mão-de-obra.

Full textOs polissacarídeos são os componentes maioritários dos grãos de café verde e torrado e da bebida de café. Os mais abundantes são as galactomananas, seguindo-se as arabinogalactanas. Durante o processo de torra, as galactomananas e arabinogalactanas sofrem modificações estruturais, as quais estão longe de estar completamente elucidadas devido à sua diversidade e à complexidade estrutural dos compostos formados. Durante o processo de torra, as galactomananas e arabinogalactanas reagem com proteínas, ácidos clorogénicos e sacarose, originando compostos castanhos de alto peso molecular contendo nitrogénio, designados de melanoidinas. As melanoidinas do café apresentam diversas atividades biológicas e efeitos benéficos para a saúde. No entanto, a sua estrutura exata e os mecanismos envolvidos na sua formação permanecem desconhecidos, bem como a relação estrutura-atividade biológica. A utilização de sistemas modelo e a análise por espectrometria de massa permitem obter uma visão global e, simultaneamente, detalhada das modificações estruturais nos polissacarídeos do café promovidas pela torra, contribuindo para a elucidação das estruturas e mecanismos de formação das melanoidinas. Com base nesta tese, oligossacarídeos estruturalmente relacionados com a cadeia principal das galactomananas, (β1→4)-Dmanotriose (Man3), e as cadeias laterais das arabinogalactanas, (α1→5)-Larabinotriose (Ara3), isoladamente ou em misturas com ácido 5-Ocafeoilquínico (5-CQA), o ácido clorogénico mais abundante nos grãos de café verde, e péptidos compostos por tirosina e leucina, usados como modelos das proteínas, foram sujeitos a tratamento térmico a seco, mimetizando o processo de torra. A oxidação induzida por radicais hidroxilo (HO•) foi também estudada, uma vez que estes radicais parecem estar envolvidos na modificação dos polissacarídeos durante a torra. A identificação das modificações estruturais induzidas por tratamento térmico e oxidativo dos compostos modelo foi feita por estratégias analíticas baseadas principalmente em espectrometria de massa, mas também em cromatografia líquida. A cromatografia de gás foi usada na análise de açúcares neutros e ligações glicosídicas. Para validar as conclusões obtidas com os compostos modelo, foram também analisadas amostras de polissacarídeos do café obtidas a partir de resíduo de café e café instantâneo. Os resultados obtidos a partir dos oligossacarídeos modelo quando submetidos a tratamento térmico (seco), assim como à oxidação induzida por HO• (em solução), indicam a ocorrência de despolimerização, o que está de acordo com estudos anteriores que reportam a despolimerização das galactomananas e arabinogalactanas do café durante a torra. Foram ainda identificados outros compostos resultantes da quebra do anel de açúcares formados durante o tratamento térmico e oxidativo da Ara3. Por outro lado, o tratamento térmico a seco dos oligossacarídeos modelo (individualmente ou quando misturados) promoveu a formação de oligossacarídeos com um maior grau de polimerização, e também polissacarídeos com novos tipos de ligações glicosídicas, evidenciando a ocorrência de polimerização através reações de transglicosilação não enzimática induzidas por tratamento térmico a seco. As reações de transglicosilação induzidas por tratamento térmico a seco podem ocorrer entre resíduos de açúcares provenientes da mesma origem, mas também de origens diferentes com formação de estruturas híbridas, contendo arabinose e manose como observado nos casos dos compostos modelo usados. Os resultados obtidos a partir de amostras do resíduo de café e de café instantâneo sugerem a presença de polissacarídeos híbridos nestas amostras de café processado, corroborando a ocorrência de transglicosilação durante o processo de torra. Além disso, o estudo de misturas contendo diferentes proporções de cada oligossacarídeo modelo, mimetizando regiões do grão de café com composição distinta em polissacarídeos, sujeitos a diferentes períodos de tratamento térmico, permitiu inferir que diferentes estruturas híbridas e não híbridas podem ser formadas a partir das arabinogalactanas e galactomananas, dependendo da sua distribuição nas paredes celulares do grão e das condições de torra. Estes resultados podem explicar a heterogeneidade de estruturas de melanoidinas formadas durante a torra do café. Os resultados obtidos a partir de misturas modelo contendo um oligossacarídeo (Ara3 ou Man3) e 5-CQA sujeitas a tratamento térmico a seco, assim como de amostras provenientes do resíduo de café, mostraram a formação de compostos híbridos compostos por moléculas de CQA ligadas covalentemente a um número variável de resíduos de açúcar. Além disso, os resultados obtidos a partir da mistura contendo Man3 e 5-CQA mostraram que o CQA atua como catalisador das reações de transglicosilação. Por outro lado, nas misturas modelo contendo um péptido, mesmo contendo também 5-CQA e sujeitas ao mesmo tratamento, observou-se uma diminuição na extensão das reações transglicosilação. Este resultado pode explicar a baixa extensão das reações de transglicosilação não enzimáticas durante a torra nas regiões do grão de café mais ricas em proteínas, apesar dos polissacarídeos serem os componentes maioritários dos grãos de café. A diminuição das reações de transglicosilação na presença de péptidos/proteínas pode dever-se ao facto de os resíduos de açúcares redutores reagirem preferencialmente com os grupos amina de péptidos/proteínas por reação de Maillard, diminuindo o número de resíduos de açúcares redutores disponíveis para as reações de transglicosilação. Além dos compostos já descritos, uma diversidade de outros compostos foram formados a partir dos sistemas modelo, nomeadamente derivados de desidratação formados durante o tratamento térmico a seco. Em conclusão, a tipificação das modificações estruturais promovidas pela torra nos polissacarídeos do café abre o caminho para a compreensão dos mecanismos de formação das melanoidinas e da relação estrutura-atividade destes compostos.
Polysaccharides are the major components of green and roasted coffee beans, and coffee brew. The most abundant ones are galactomannans, followed by arabinogalactans. During the roasting process, galactomannans and arabinogalactans undergo structural modifications that are far to be completely elucidated due to their diversity and complexity of the compounds formed. During the roasting process, galactomannans and arabinogalactans react with proteins, chlorogenic acids, and sucrose, originating high molecular weight brown compounds containing nitrogen, known as melanoidins. Several biological activities and beneficial health effects have been attributed to coffee melanoidins. However, their exact structures and the mechanisms involved in their formation remain unknown, as well as the structure-biological activity relationship. The use of model systems and mass spectrometry analysis allow to obtain an overall view and, simultaneously, detailed, of the structural modifications in coffee polysaccharides promoted by roasting, contributing to the elucidation of the structures and formation mechanisms of melanoidins. Based on this thesis, oligosaccharides structurally related to the backbone of galactomannans, (β1→4)-D-mannotriose, and the side chains of arabinogalactans, (α1→5)-Larabinotriose, alone or in mixtures with 5-O-caffeoylquinic acid, the most abundant chlorogenic acid in green coffee beans, and dipeptides composed by tyrosine and leucine, used as models of proteins, were submitted to dry thermal treatments, mimicking the coffee roasting process. The oxidation induced by hydroxyl radicals (HO•) was also studied, since these radicals seem to be involved in the modification of the polysaccharides during roasting. The identification of the structural modifications induced by thermal and oxidative treatment of the model compounds was performed mostly by mass spectrometry-based analytical strategies, but also using liquid chromatography. Gas chromatography was used in the analysis of neutral sugars and glycosidic linkages. To validate the conclusions achieved with the model compounds, coffee polysaccharide samples obtained from spent coffee grounds and instant coffee were also analysed. The results obtained from the model oligosaccharides when submitted to thermal treatment (dry) or oxidation induced by HO• (in solution) indicate the occurrence of depolymerization, which is in line with previous studies reporting the depolymerization of coffee galactomannans and arabinogalactans during roasting. Compounds resulting from sugar ring cleavage were also formed during thermal treatment and oxidative treatment of Ara3. On the other hand, the dry thermal treatment of the model oligosaccharides (alone or when mixed) promoted the formation of oligosaccharides with a higher degree of polymerization, and also polysaccharides with new type of glycosidic linkages, evidencing the occurrence of polymerization via non-enzymatic transglycosylation reactions induced by dry thermal treatment. The transglycosylation reactions induced by dry thermal treatment can occur between sugar residues from the same origin, but also of different origins, with formation of hybrid structures, containing arabinose and mannose in the case of the model compounds used. The results obtained from spent coffee grounds and instant coffee samples suggest the presence of hybrid polysaccharides in these processed coffee samples, corroborating the occurrence of transglycosylation during the roasting process. Furthermore, the study of mixtures containing different proportions of each model oligosaccharide, mimicking coffee bean regions with distinct polysaccharide composition, subjected to different periods of thermal treatment, allowed to infer that different hybrid and non-hybrid structures may be formed from arabinogalactans and galactomannans, depending on their distribution in the bean cell walls and on roasting conditions. These results may explain the heterogeneity of melanoidins structures formed during coffee roasting. The results obtained from model mixtures containing an oligosaccharide (Ara3 or Man3) and 5-CQA and subjected to dry thermal treatment, as well as samples derived from spent coffee grounds, showed the formation of hybrid compounds composed by CQA molecules covalently linked to a variable number of sugar residues. Moreover, the results obtained from the mixture containing Man3 and 5-CQA showed that CQA acts as catalyst of transglycosylation reactions. On the other hand, in the model mixtures containing a peptide, even if containing 5-CQA and subjected to the same treatment, it was observed a decrease in the extent of transglycosylation reactions. This outcome can explain the low extent of non-enzymatic transglycosylation reactions during roasting in coffee bean regions enriched in proteins, although polysaccharides are the major components of the coffee beans. The decrease of transglycosylation reactions in the presence of peptides/proteins can be related with the preferential reactivity of reducing residues with the amino groups of peptides/proteins by Maillard reaction, decreasing the number of reducing residues available to be directly involved in the transglycosylation reactions. In addition to the compounds already described, a diversity of other compounds were formed from model systems, namely dehydrated derivatives formed during dry thermal treatment. In conclusion, the identification of the structural modifications in coffee polysaccharides promoted by roasting pave the way to the understanding of the mechanisms of formation of melanoidins and structure-activity relationship of these compounds.

This thesis, by means of SCP (Structure—Conduct—Performance) system of industrial organization theory, analyzes the industrial structure, how the structure to is influence market conduct, and how the market conduct to decide performance. In the thesis, China’s banking sector is taken as the object of analysis. It is learned that the current situation of China’s banking market is under a market structure of lose oligopoly. Such a market structure will render different directions for Chinese-founded banks, foreign-founded banks and Taiwan-invested banks in future layout in China. In this thesis, it by means of case study regarding current strategies and operation modes of China-founded, foreign-founded and Taiwan-invested banks, incisively analyzes the pros and cons of the current operation and improved ways. Finally, it raises concrete suggestions according to research results, hoping to have a reference value for sustainable development of China’s banking sector, the business layout of foreign-founded banks in Mainland China, and the westward strategies of Taiwan-invested banks.

Full textDisability happens to be one of the stigmatized discourses in rural Swaziland where myths and superstitions on just about any discourse abound (SINTEF, 2011). My research coincided with the 2018 national elections in Swaziland which is generally a period that is often marked by an alarming rate of ritual murders of people with disabilities. At the heart of ritual murders are ludicrous cultural and social constructions about people with disabilities. Socio-cultural constructions about people with disabilities tend to reinforce and reproduce existing stigma and social inequalities prevalent in rural communities and seldom are these cultural constructions publicly let alone privately challenged, dispelled or interrogated despite their harm. The media (print) being one of the key knowledge producers in Swaziland has often reproduced ableistic notions about people with disabilities thus reinforcing ableism and disability oppression. Entman and Paletz (1980, p. 154) assert that “however unsubstantial or fanciful, the media’s depiction of reality influence the way people think and act politically, the messages affect power”. I therefore opted for this research because disability is amongst the most unpopular, less understood/misunderstood and least researched discourses in Swaziland which is evident in the little, scarce and scanty available resources on disability research. It is also still a less talked about taboo discourse or rather spoken about in hush tones due to misperceptions about it being ‘dirty-linen that ought to be swept under the straw-mat’. Personally, being a rural resident, I have also been indirectly affected by disability through kinship relations whereby I have observed how everyday taken-for-granted language and clichés have propagated and reinforced ableism and disability oppression as is invisible naturalized and legitimated at the expense of people with disabilities. The research also centres on rural communities because that is where about 86% of people with disabilities reside in Swaziland (MoHSW, 2000). Rural communities are generally characterized by a higher level of illiteracy and limited access to knowledge hence myths, misconceptions and superstitions easily breed and are perceived as ‘gospel’ truth (Dlamini, 2017). Socio-cultural constructions become powerful influences that often ‘spread like gangrene’ but when timeously ‘nipped in the bud’ significantly prevent or undo harm on those to whom they are directed or coined which is often marginalized minority groups. It is against this background then that in this research I critically identify and analyse the socio-cultural constructions of people with disabilities as portrayed in mainstream media. This work is divided into five (5) Chapters namely; Chapter 1 contains the Introduction, Research problem, Rationale and Objectives of the study, Chapter 2 is the Literature Review, Chapter 3 is about the Methodology and Theoretical Framework, Chapter 4 is the Data Analysis and lastly Chapter 5 is the Summary Conclusion and the Bibliography. This research is about the possibility of water turning to steam which is used in a continuous production process of Activated Carbon Fabric. This production process use the regenerated cellulose fiber fabric as the raw material, such as ModalTM, Polynosic and Regular Rayon, and it is has been prepared by pretreatment of refractory reagent [Ammonium Sulfate˙(NH4)2SO4] and [Ammonium Phosphate Dibasic˙(NH4)2HPO4] (mixture ratio: 95/5, concentration:40wt.%). This process is used water into high temperature activation equipment become to steam as the activator gas. In our experimental equipment, used ModalTM as the raw material, the production rate, water flow and the highest treatment temperature of the activated process is used as parameters. We hope knowing the possibility about water into the high temperature activation equipment directly. Then we used same method in the high temperature activation equipment is bigger than old one to understand the possibility of industrialization. The activated carbon fabric (ACF) material is used in the gas adsorption field to understand its properties. When we examined the results, it revealed that it is sure in industrialization when water used in the high temperature activation equipment on a continuous production process of ACF will turn to steam as the activator gas. In this method, we can save energy, equipment cost and so on. The other hand, we also get some characteristic which can control the surface pore''s radius and other activator gas cannot acquired. In the gas adsorption field, our ACF have more pore volume''s adsorption amount because the surface pore''s radius is the biggest among any other activator gas products of ACF.

Full textIntroduction Given the increasing prevalence of non-typhoidal salmonella in humans, especially as an opportunistic illness associated with HIV, enhanced surveillance for non-typhoidal salmonella (NTS), including screening for antibiotic resistance, is conducted annually in South Africa. We aimed to determine whether there is an association between trimethoprim-sulfamethoxazole (TMP-SMX) prophylaxis and multi-drug resistant NTS infection, to establish whether various factors modify the relationship between TMP-SMX resistance and invasive NTS infection, to examine whether these associations vary by province, and to quantify the resistance rates of NTS to a range of antibiotics. Methods This study was a secondary analysis of enhanced surveillance data on NTS collected between 2003 and 2005. We used descriptive methods to assess the prevalence of NTS by year, province and serotype, and to determine the prevalence of four MDR patterns. Univariate and multivariate regression models were used to investigate the relationships between TMP-SMX prophylaxis and MDR NTS. Univariate logistic regression was used to assess the relationship between invasive NTS and TMP-SMX resistance. Results TMP-SMX prophylaxis is associated with the ACKSSuT pattern (OR 1.91, 95% CI 1.14 – 3.19, p=0.0080) and the AKSSuT MDR pattern (OR 2.00, 95% CI 1.26 – 3.15, p=0.0015). Being on TMP-SMX prophylaxis is associated with an increased odds of having at least one of the four MDR patterns investigated (OR 1.43, 95% CI 1.00 – 2.04, p=0.0388). We also found high rates of resistance to all antibiotics tested except for ciprofloxacin and imipenem. The highest resistance rate was observed for sulfamethoxazole (>75.85%). S. enterica Isangi isolates showed the highest levels of resistance, with 94.43% having at least one MDR pattern. Other factors significantly associated with MDR NTS were ESBL production, prior treatment with antibiotics, HIV status and resistance to TMP-SMX. Discussion and conclusions Isolates from patients on TMP-SMX prophylaxis were associated with an increased odds of having the ACKSSuT and AKSSuT MDR patterns, not taking into account other explanatory factors. These associations did not remain significant when possible confounders were taken into account. Despite the threat of increased multi-drug resistance, TMP-SMX prophylaxis remains important in certain clinical settings.

Full textUsing a text-immanent multi-dimensional methodology that combines impulses from both synchronic and diachronic reading of a text, this study focuses on understanding the spirituality embedded in ‘seeing him as he is’ in 1 John 3:2. Discourse analysis has provided the structure of the entire research by identifying the different semantic networks that enhance better understanding and dynamic interaction between text and reader.it has also helped in determining the argument and rhetoric of the Elder, assisting in constructing the bigger picture by means of semantic networks that create coherent mind maps and also relating what has been read with what is still to be read. The environs of the pericope under investigation have been used as a backdrop in order to arrive at an understanding of this envisaged eschatological phenomenon. These environs include the window provided by Judaism through the Old Testament, Hellenistic and Palestinian Judaism, extra-biblical literature, the Graeco-Roman world, mystery religions, philosophies, and the New Testament. These environs have pointed to the use of intermediaries in the visio-Dei. While ‘seeing him as he is’ in 1 John 3:2 involves both the Father and the Son, this eschatological expectation is weaved into a matrix of discourse that the Elder used to cushion the adherents in view of the pending apostasy. The adherents’ status as ‘children of God’ is the axis from which the Elder builds his entire discourse. They will experience love (1 Jn 4:16), his purity (1 Jn 3:3), his righteousness (1 Jn 2:1), his truth (1 Jn 5:20), and his glory (Jn 17:24). Although the adherents were already experiencing all these, it would be experienced completely after the Parousia, when they ‘see him as he is’. This study contributes towards a Johannine understanding of perceiving the divine, and reveals the climactic involvement of the Son in both the past and future perceptions including ‘seeing him as he is’ in 1 John 3:2. This study has identified the object of the Visio-Dei as Christ. It is He through whom believers will see the Father.
