Full textThis PhD work, mainly methodological, focused on the techniques of magnetic resonance imaging (MRI) to measure cerebral blood flow (CBF) and more particularly on arterial spin labeling (ASL), which uses water protons from the blood as markers. We implemented the pseudo-continuous ASL labeling sequence (pCASL) and evaluated its response to a hypercapnic stimulus. We evaluated different strategies to maximize the labeling inversion effeciency. For this, we implemented numerical simulation tools of ASL approaches. Our results demonstrated that the inversion efficiency is influenced by the homogeneity of the magnetic field in the labeling region, which becomes a problem at high magnetic field. The optimized pCASL protocol was then evaluated in rats at three magnetic fields (4.7, 7, and 11.7T) and compared with a conventional continuous ASL approach (CASL). This comparison showed excellent inter-animal and inter-field reproducibility of the developed method. In a second part, we evaluated the influence of the longitudinal relaxation time (T1) of brain tissue on the final CBF values. For this, we modified the tissue T1 by an intracerebral injection of manganese. This study demonstrated the difficulty of taking into account the change in T1 of the tissue. In a third part, we evaluated the contribution of a specific labeling coil for the CASL approach by comparing measurements of CBF obtained with that of pCASL. We found a good agreement between these two methods for all slices. Our results also illustrate the significant contribution of magnetization transfer effects in CASL sequence. The tools developed during this thesis were also used in several preclinical studies including brain tumors, stroke and head trauma models

Full textFunctional and perfusion imaging modalities are closely related since they both measure, directly or indirectly, blood flow in the brain. Functional Magnetic Resonance Imaging (fMRI) using the blood oxygen level dependent (BOLD) contrast exploits the magnetic properties of blood (oxy- and deoxyhemoglobin) to measure local changes in blood oxygen concentration in the brain. The neurovascular coupling allows us to infer brain function from fMRI images. Perfusion MRI images the cerebral vascular system by directly measuring blood flow. In particular, Arterial Spin Labeling (ASL) does not need contrast agents; it uses spins of endogenous water protons as a tracer instead. Usually ASL is used to probe the basal perfusion at rest. However, in the recent years, it has also been used as a functional imaging modality (as fASL) by tracking task-related perfusion changes. In contrast to the standard BOLD fMRI, results are quantitative, making this type of data attractive for use in clinical research.This thesis focuses on the investigation of the fASL modality and the development of new methods to analyze it. As previously done for BOLD data, a Bayesian framework is proposed for the analysis of fASL data. It provides a way of modeling activation values and both hemodynamic and perfusion response functions as probabilistic variables in the so-called joint detection estimation (JDE) framework. Bayesian models use a priori knowledge in the estimation of unknown parameters through the specification of probability distributions. In this work, we exploit this feature to incorporate physiological information to make the estimation more robust. In particular, we use physiological models based on the balloon model to derive a link between hemodynamic and perfusion responses and we turn this link into a prior distribution to regularize the estimation of the responses. A Markov Chain Monte Carlo solution with prior physiological knowledge has been first proposed for the estimation of the quantities contained in the fMRI signal. Since the computational cost of this algorithm is very high, we then reformulate the problem to use a variational expectation maximization approach that provides a much faster algorithm with similar results. The use of priors and constraints in this setting is also more straightforward.These methods have been evaluated on two different datasets using event-related and block designs with very simple experimental tasks. We show the performance of the methods investigated in comparison to standard methods at the subject and group levels. Experimental results show the utility of using physiological priors for improving the recovery of a perfusion response function. They also demonstrate that BOLD fMRI achieves better sensitivity to detect evoked brain activity as compared to fASL although fASL gives a more localized activation, which is in line with the existing literature. From the results, we discuss the impact of the modelling of spatial correlation, as well as the impact of the estimation of temporal responses.This work proposes new methodological contributions in the study of a relatively new fMRI modality that is functional ASL, and puts it into perspective with the existing techniques. Thus, we provide new tools for the neuroscientific community to study and understand brain function. These tools have been implemented in python in the PyHRF package

Full textAutism Spectrum Disorders (ASD) are neurodevelopmental disorders characterized by abnormal social interactions. Eye-tracking studies have objectively reported social perception abnormalities in ASD, characterized by a decrease of gaze towards social stimuli. Brain imaging studies, using PET and SPECT methods, have revealed a decrease in cerebral blood flow (CSF) at rest in the temporal regions, particularly in the superior temporal sulcus (STS), in children with ASD. Nowadays, it is possible to measure rest CBF with MRI using arterial spin labeling (ASL) sequence. In this thesis, we confirmed the decreased in rest CBF within the STS in children with ASD using MRI-ASL, which could allow its use as a biomarker in ASD. We also suggest that rest CBF could be a more relevant index for studying basic brain function in ASD. In addition, we performed a cross-sectional eye-tracking study using the same stimuli over a wide age-range and showed the impact of age on social perception in ASD and in typical development. Finally, we showed social perception abnormalities, using eye-tracking, and cortical functioning abnormalities within the STS using MRI-ASL, in children with posterior fossa arachnoid cyst. A better understanding of the social difficulties underlying this disorder could have a major impact on patient outcome

Full textThe magnetic resonance imaging (MRI) allows the observation of various kind of tissues with always increasing resolution. The arterial and venous vascular trees can be explored, and the flows can be characterized in a noninvasive way. As an example, the arterial part of the tree can be obtained using so-called “Time Of Flight” MRI, and the venous part with phase contrast techniques. The development of quantitative susceptibility maps (QSM) improves the level of details achievable regarding veins; furthermore, it provides a new way to estimate physiological parameters such as venous saturation in oxygen. Eventually the implementation of dedicated algorithms and tools allows the in-silico reconstruction of a subject-specific coherent architecture. Moreover, due to the use of dynamic imaging sequences such as the dynamic phase contrast imaging and the arterial spin labeling, the arterial, venous and cerebral blood flow are measurable. All of these sequences are noninvasive and so usable on every subjects. Based on these anatomical and dynamics data, a full subject-specific model of the brain hydrodynamics is proposed here. The blood and cerebrospinal flow are described using basic balance equations of the hydrodynamics: continuity and momentum. To take into account of the adaptation of vessel diameter to the pressure, a wall elasticity parameter is added for each compartment together with the corresponding equation. Thanks to the MRI data, all the blood compartments, from arteries to vein, the cerebral parenchyma and the ventricular system are included. The model is able to simulate the flow and pressure repartition in all compartments of the subjects as well as show the impact of a located occlusion on the whole architecture

Full textAbstract The purpose of the study is through a qualitative method to investigate how teachers, relative to their educational background and experience of school practice, describe the balance between handwriting and computer writing in early writing instruction, and thereby contribute to existing research in the field. Three research questions have been formulated and to answer them, I have used semi-structured individual interviews with trained professional teachers in the data collection. In the data processing, I have used a phenomenographic analysis model within an action theoretical framework. The results show that teachers, with the support of their education but even more through proven experience, balance the use of handwriting and computer writing in their classrooms very differently, but that they justify their choice of methods on the same grounds. Teachers plan their teaching with support in research and state that the pupils have good goal fulfillment for writing at the end of year three, both in terms of handwriting and computer writing. The teachers' own descriptions of their writing instruction are that the pupils are encouraged to feel the joy of writing through chosen methods, and even if the learning takes place in different ways, the process leads to each individual pupil succeeding and feeling confident in their language ability. The results also show that teachers believe that all methods are really good, that the prerequisite for pupils to succeed in achieving the stated curriculum goals is that the teacher is committed, believes in his method and succeeds in enthusing the pupils.

Full textTranscranial direct current stimulation (tDCS) is used to modulate neuronal activity in the brain. It consists in applying a small constant current between two electrodes placed over the scalp. Two frontal tDCS montages have shown promises in modulating cognitive abilities and/or helping to alleviate clinical symptoms. However, the effects of tDCS on brain physiology are still poorly understood. The aim of this thesis work was to clarify brain mechanisms underlying frontal tDCS in healthy subjects, specifically in relation to the dopaminergic system. Using a double blind sham-controlled design, we combined a single session of tDCS online with several imaging techniques (PET or simultaneous PET-MRI) with the subject at rest. A first study (n=32, 2mA, 20min) showed that bifrontal tDCS induced an increase in extracellular dopamine in the ventral striatum, involved in the reward-motivation network, after the stimulation period. A second study (n=30, 1mA, 30min) showed that fronto-temporal tDCS induced an increase in extracellular dopamine in the executive part of striatum as well as a decrease in perfusion in a region part of the default mode network (DMN), after the stimulation period. The data analysis of this study is still ongoing. Overall, the present work provides evidence that a single session of frontal tDCS impacts the dopaminergic system in regions connected to the stimulated cortical areas. Therefore, levels of dopamine activity and reactivity should be new elements to consider for a general hypothesis of brain modulation by frontal tDCS
