Abstract:
ATP accumulates in the tumour microenvironment (TME) and via its receptor P2X7 is involved in immune response, cancer proliferation and metastasis formation. In this study we investigated ATP release, P2X7 expression and its function in acute myeloid leukemia (AML) and melanoma. In an in vivo AML model, treatment with daunorubicin causes the release of ATP in TME that is associated to tumour eradicating immune response. When we analysed the two functional P2X7 isoforms (P2X7A and P2X7B) in an AML population we observed that they are both overexpressed in patients affected by AML as compared to

Two neurotransmitters, ACh and ATP, initiate nerve-mediated contractions in detrusor smooth muscle from guinea-pigs and humans with bladder over-activity whereas only ACh has a role in muscle from stable human bladders. Both Acetylcholine and ATP are rapidly broken down, by acetylcholinesterases and ectonucleotidases respectively. The activity of ectonucleotidase has been shown to be reduced in the unstable human detrusor compared to stable. The reduced activity may contribute to the development of the purinergic component of the unstable human detrusor contraction. In vitro muscle strip exper

The main obstacle to the success of the immunotherapy is the well established tumor-induced tolerance. In 2007 a new cell population, Myeloid Derived Suppressor Cells (MDSCs), that accumulate under inflammatory conditions, especially in cancer, was identified. These cells are potent inhibitors of tumor immunity and are now considered a major contributor to the failure of the immunotherapy. Understanding the exact mechanism of immunosuppressive activity of MDSC is a crucial point in order to find new ways to improve anticancer therapies. ABCA1 is a key plasma membrane protein required for the efflux of cellular cholesterol to extracellular acceptors, particularly to apoA-I. This process is essential to maintain cholesterol homeostasis in the body. The detailed molecular mechanisms, however, are still insufficiently understood. Also, the molecular identity of ABCA1, i.e. channel, pump or flippase, remains unknown. In this study we analyzed the extracellular ATP levels in the medium of ABCA1-expressing BHK cells and RAW macrophages and compared them to the medium of relevant non-expressing cells. Abstract:
Glomerular epithelial cells (GECs) are intrinsic components of the kidney glomerulus and are in contact with extracellular matrix (ECM). Under normal conditions, there is little turnover of GECs, but in glomerular injury, apoptosis and proliferation of GECs, and expansion of ECM may lead to sclerosis and impaired glomerular function and/or permselectivity. In order to better understand how adhesion to ECM modulates GEC survival and proliferation, we monitored the activation of the intracellular signaling pathways activated by adhesion to ECM (collagen), along with their coordinate modul
